Abstract
Τα ευκαρυωτικά γονιδιώματα διπλασιάζονται από την ενεργοποίηση πολλαπλών αμφίδρομων σημείων έναρξης του αναδιπλασιασμού. Τα προγράμματα αναδιπλασιασμού αυτών των κυττάρων εξαρτώνται από τη περιστασιακή και χωροταξική οργάνωση των σημείων έναρξης του αναδιπλασιασμού σε όλη την έκταση του γονιδιώματος. Για να ερευνηθεί το πρόγραμμα του αναδιπλασιασμού σε ένα ανώτερο ευκαρυωτικό, χρησιμοποιήσαμε μια τεχνική που ονομάζεται μοριακό χτένισμα. Αυτή η τεχνική επιτρέπει μια ποσοτική ανάλυση του αναδιπλασιασμού του DNA ενός γονιδιώματος ευρείας βάσης. Σαν σύστημα μοντέλο, η χρωματίνη του σπέρματος του Xenopus laevis σημάνθηκε διαφορικά σε διαδοχικά χρονικά σημεία μετά την έναρξη της σύνθεσης του DNA.
