Abstract

本实用新型的肝细胞大规模培养和冷冻的装置,包括培养盒和冷冻盒,所述培养盒包括盒体、至少2层平板、壳聚糖纳米纤维膜、平板小孔、上塞盖、支架管、支架侧孔和下塞盖,所述冷冻盒包括冷冻盒体、冷冻剂和冷冻盒盖,所述培养盒置于冷冻盒内的冷冻液中。能将一次分离所获肝细胞按要求直接接种于培养盒中的壳聚糖多层平板上进行扩增,实现大规模培养,培养盒内的多层平板表面上的壳聚糖纳米纤维充分模拟体内微环境能促进肝细胞粘附,并能提高解冻后肝细胞的细胞活力和功能,还具有结构设计合理,操作使用简单方便易行等优点,适合需要为肝衰患者提供大量肝细胞的医院使用。

肝细胞大规模培养和冻存的装置
技术领域
本实用新型涉及一种装置,特别是一种主要用于进行肝细胞大规模培养和冷冻的装置,属于专用于医学目的的容器。
背景技术
慢性肝炎肝硬化、中晚期肝癌的病人常常死于终末期肝功能衰竭,应用生物人工肝以及肝脏移植等疗法来治疗肝脏衰竭最近几年来取得了显著的成就。生物人工肝支持系统借助体外循环装置清除体内各种有害物质并且补充体内必需的蛋白质和凝血因子,为患者提供短暂的肝功能支持;肝移植疗法则是借助同种或异种肝脏移植到患者体内发挥功能来达到治疗目的。但上述两种治疗方法都面临着缺乏理想肝细胞源的问题,而且并不是每次获取的肝细胞量都能达到治疗的要求,因此探索一种符合实际情况的肝细胞大规模培养、冻存及复苏方法是提高肝衰患者治疗效果的重要前提。
目前细胞的大规模培养常使用生物反应器,常见的有搅拌式、旋转式、灌注式等。但生物反应器构造复杂、价格昂贵、使用不方便等缺点使其在生物医学领域应用发展缓慢。
在本发明之前,常规使用的细胞冷冻降温方法大致分为三种:常规冷冻、程序降温仪冷冻、冷冻盒冷冻。其中使用常规方法(即:4℃放置30min,-20℃冷冻30min,最后-80℃过夜,次日投液氮)对原代肝细胞冷冻效果较差;而采用计算机程序降温仪冷冻操作复杂、费时、费力;采用冷冻盒进行冷冻操作简便、经济、省时,冷冻效果与前者相近。上述几种冷冻方法均是将细胞混于冷冻保护液并置于冷冻小管中冷冻保存,这种方法对一般分裂传代细胞进行少量冷冻,效果较好且方便,细胞复苏后可传代培养并用于细胞学或分子生物学研究,但针对以治疗为目的的肝细胞却存在一定缺陷。首先,肝细胞用来治疗所需量大,常规采用冷冻管冷冻的方法数量少、操作过程繁琐,无法达到大规模冷冻的要求;其次,常规采用冷冻方法无法满足一次性急需大量肝细胞的需求,更不能达到成品化输出的目的。
发明内容
本实用新型的目的就在于克服上述现有装置的不足之处,为满足肝衰患者急需大量肝细胞的要求,提供一种适合医院使用的结构简单合理,操作使用简单方便的肝细胞大规模培养和冷冻的装置。
本实用新型的肝细胞大规模培养和冷冻的装置,主要由培养盒和冷冻盒组成,所述培养盒包括盒体、至少2层平板、壳聚糖纳米纤维膜、平板小孔、上塞盖、支架管、支架侧孔和下塞盖,其中,支架管装在盒体的上口与下口之间,平板经中心孔分别安装在支架管上,壳聚糖纳米纤维膜贴在平板上,平板小孔分布在平板上,支架侧孔分布在支架管壁上,上塞盖和下塞盖分别装在盒体的上口与下口上;所述冷冻盒包括冷冻盒体、冷冻剂和冷冻盒盖,冷冻剂装在冷冻盒体内,冷冻盒盖盖装在冷冻盒体的盒口上;所述培养盒置于冷冻盒内的冷冻液中。
为了便于将装有培养盒的冷冻盒吊于液氮罐中冷冻,可在冷冻盒盖上设有吊耳和吊绳,吊耳安装在冷冻盒盖上,吊绳系扣在吊耳上,为了提高密封性能,可在冷冻盒盖与冷冻盒体之间装有耐低温密封圈。
上述培养盒体选用耐低温的PIR聚异氰脲酸酯泡沫盒,冷冻液选用异丙醇或丙二醇,耐低温密封圈选用硅橡胶密封圈。
本实用新型的优点和效果在于:能将一次分离所获肝细胞按要求直接接种于培养盒中的壳聚糖多层平板上进行扩增,实现大规模培养,大幅度减少了每次分离肝细胞的浪费,而且培养盒直接放入盛有异丙醇或丙二醇的冷冻盒中冻存,培养盒内的多层平板表面上的壳聚糖纳米纤维充分模拟体内微环境能促进肝细胞粘附,并能提高解冻后肝细胞的细胞活力和功能,解决了常规冷冻肝细胞装置使细胞贴壁不稳定而容易脱落和难以提供大量肝细胞等问题。还具有结构设计合理,操作使用简单方便易行等优点,适合需要为肝衰患者提供大量肝细胞的医院使用。

具体实施方式
参见附图1和2,本实用新型的肝细胞大规模培养和冷冻的装置,主要由培养盒和冷冻盒组成,所述培养盒包括盒体1、至少2层平板2、壳聚糖纳米纤维膜3、平板小孔4、上塞盖5、支架管6、支架侧孔7和下塞盖8,其中,支架管6装在盒体1的上口与下口之间,平板2经中心孔分别安装在支架管6上,壳聚糖纳米纤维膜3贴在平板2上,平板小孔4分布在平板2上,支架侧孔7分布在支架管6壁上,上塞盖5和下塞盖8分别装在盒体1的上口与下口上;所述冷冻盒包括冷冻盒体11、冷冻剂10和冷冻盒盖9,冷冻剂10装在冷冻盒体11内,冷冻盒盖9盖装在冷冻盒体11的盒口上;所述培养盒置于冷冻盒内的冷冻液中。
为了便于将装有培养盒的冷冻盒吊于液氮罐中冷冻,可在冷冻盒盖9上设有吊耳12(如图2所示)和吊绳(图中未画),吊耳12安装在冷冻盒盖9上,吊绳系在吊耳12上,为了提高密封性能,可在冷冻盒盖9与冷冻盒体11之间装有耐低温密封圈13,其结构如图2所示。
上述培养盒体1选用耐低温的PIR聚异氰脲酸酯泡沫盒,冷冻液10选用异丙醇或丙二醇,耐低温密封圈13选用硅橡胶密封圈。培养盒和冷冻盒的形状选用方形或圆形。
操作时,将体外分离的肝细胞经纯化离心后,调节细胞至合适密度,一般为1*107/ml左右,取若干毫升该浓度的细胞,将其从培养盒上口接种至多层平板上的壳聚糖纳米纤维膜上,将接种肝细胞的培养盒置37℃培养箱预培养一段时间,待细胞贴壁后从培养盒上口添加新鲜培养液使其没过支架管,此时即可将培养液换成冷冻保存液,并将培养盒置入盛有异丙醇或丙二醇的冷冻盒内后再放入-80℃过夜,次日再将装有培养盒的冷冻盒吊于液氮罐中冷冻待用。需要使用时,可将冷冻盒从液氮中取出,置于37℃水浴复温后,将培养盒从冷冻盒中取出,从培养盒底部出口打开下塞盖,放出冷冻培养液,换成新鲜培养液孵育一段时间,待细胞活力恢复后,即直接用于治疗使用或加入胰酶消化即可收集所需细胞。

1.一种肝细胞大规模培养和冷冻的装置,其特征在于包括培养盒和冷冻盒,所述培养盒包括盒体、至少2层平板、壳聚糖纳米纤维膜、平板小孔、上塞盖、支架管、支架侧孔和下塞盖,其中,支架管装在盒体的上口与下口之间,平板经中心孔分别安装在支架管上,壳聚糖纳米纤维膜贴在平板上,平板小孔分布在平板上,支架侧孔分布在支架管壁上,上塞盖和下塞盖分别装在盒体的上口与下口上;所述冷冻盒包括冷冻盒体、冷冻剂和冷冻盒盖,冷冻剂装在冷冻盒体内,冷冻盒盖盖装在冷冻盒体的盒口上;所述培养盒置于冷冻盒内的冷冻液中。
2.如权利要求1所述的肝细胞大规模培养和冷冻的装置,其特征在于所述冷冻盒盖上设有吊耳和吊绳,吊耳安装在冷冻盒盖上,吊绳系扣在吊耳上,冷冻盒盖与冷冻盒体之间装有耐低温密封圈。
3.如权利要求1或2所述的肝细胞大规模培养和冷冻的装置,其特征在于所述培养盒体选用耐低温的PIR聚异氰脲酸酯泡沫盒,冷冻液选用异丙醇或丙二醇,耐低温密封圈选用硅橡胶密封圈。
