Description
技术领域
本发明涉及一种新的基因送递系统和重组腺病毒,具体而言,本发明涉及一种新的基因送递系统和包括松弛素编码序列的重组腺病毒、一种包括该重组腺病毒的药学抗肿瘤组合物、一种特征为改善组织穿透能力的药物组合物和一种用于治疗与过量细胞外基质累积相关的疾病或病症的药物组合物。
背景技术
基因治疗旨在通过将外源基因引入细胞或组织治疗病理状况。对于如镰形细胞贫血、α1抗胰蛋白酶缺乏、苯丙酮尿症、血友病和囊性纤维化的遗传性疾病,基因治疗的目的是为了使细胞或组织发挥正常功能,置换缺失或缺陷基因。此外,基因治疗用于除去异常细胞。通过送递能够引起靶异常细胞死亡的基因,基因治疗允许治疗如癌症、发炎和自身免疫疾病的多种疾病。
虽然基因治疗有前途,但是向细胞或组织无效的基因送递是发展成功的基因治疗的主要障碍。例如,许多使用反转录病毒、腺病毒或腺伴随病毒(adeno-associated viruses,AAV)的基因送递的研究已表明当用于个体或组织(例如,肿瘤组织)时,基因送递效率不足阻碍了基因治疗的应用。
因此,表现改善的基因送递效率的新的基因送递策略对于实现成功的基因治疗仍是必需的。
早期基于腺病毒的基因治疗通常使用缺失对于腺病毒复制必需的 E1基因的携带治疗基因的复制缺陷型腺病毒。然而,这些重组腺病毒仅在受感染细胞和很少量的周围细胞中诱导抗肿瘤活性,在临床应用中表现出严重的问题。
为了克服此类问题,已开发在肿瘤细胞中选择性复制的溶瘤腺病毒,ONYX-015(dl1520)。缺失E1B 55kDa基因的腺病毒在缺乏功能性p53的肿瘤细胞中选择性复制。当重组腺病毒感染正常细胞时,因为没有诱导p53失活,所以其增殖受到抑制,导致溶瘤作用失败,而其在具有失活的p53的肿瘤细胞中活跃地增殖并最终引起肿瘤细胞的选择性死亡(Chang,F.,等,J Clin Oncol 13:1009-22(1995))。
根据脑癌的II/III期临床试验的最近报道,肿瘤特异的溶瘤腺病毒表现出可观的治疗效能(Kirn,D.,等,Nat Med4:1341-2(1998);Nemunaitis,J.等,Cancer Res 60:6359-66(2000);和Ganly,I.等,ClinCancer Res6:798-806(2000))。虽然施用重组腺病毒诱导肿瘤生长的部分抑制作用,没有发现肿瘤完全根除,且经过一段时间后快速发生肿瘤的再生长。这些结果可能是因为局部注射到肿瘤的重组腺病毒在有限的周围部分扩散以引起限制性的抗肿瘤活性,以使没有感染病毒的肿瘤细胞快速生长。根据最近的研究报道,施用到裸鼠中的人肿瘤中的重组腺病毒在晚至最初的病毒注射后100天持续复制并不保证完全根除肿瘤,但是从肿瘤组织可获得有活力病毒(Sauthoff,H.等,HumanGene Therapy 14:425-433(2003))。
因此,可以理解理想的肿瘤特异溶瘤腺病毒具有诱导更大的溶瘤活性和在遍布肿瘤组织扩散以及感染周围肿瘤细胞的能力。

发明内容
本发明已进行深入研究以改善基因送递系统的转导效率,具体而言,以增强组织中基因送递系统的转导(或扩散)效率,结果,发现松弛素可以显著地改善基因送递系统的转导效率。
因此,本发明的一个目的是提供一种具有改善的转导效率的基因送递系统。
本发明的另一目的是提供一种向细胞中递送具有改善的转导效率的基因的方法。
本发明的又一目的是提供一种具有改善的肿瘤组织穿透性和肿瘤特异性凋亡能力的重组腺病毒。
本发明的另一目的是提供一种用于治疗癌症的抗肿瘤药物组合物。
本发明的又一目的是提供一种通过使用所述抗肿瘤药物组合物治疗癌症的方法。
本发明的另一目的是提供一种用于改善药物进入组织的穿透能力的药物组合物。
本发明又另一目的是提供一种用于治疗与过量细胞外基质累积相关的疾病或病症的药物组合物。

在本发明的一个技术方案中,提供一种包括待递送到细胞中的目的核苷酸序列的基因送递系统,其改善包含编码松弛素的核苷酸序列以增强目的所述核苷酸序列进入细胞的转导效率。
本发明人已进行深入研究以改善基因送递系统的转导效率,具体而言,以增强组织中的基因送递系统的转导(或扩散)效率。此类研 究基于我们的通过促进降解合成的或抑制细胞外基质组分的合成降低细胞外基质组分的水平可增强组织内基因送递系统的扩散的假设。令人惊奇地,本发明人已发现松弛素可以显著地提高基因送递系统的转导效率。
在此所用的术语“松弛素”包含实施例中示出和举例说明的松弛素以其任意同源物以提高为本发明目的的转导效率。
在本发明中作为提高转导效率的增强子起关键作用的松弛素是6kDa的肽激素,与胰岛素和胰岛素样生长因子(insulin-like growthfactors,IGFs)结构相关。其主要在黄体和子宫内膜中产生并且其血清水平在怀孕期间大大提高(Sherwood,O.D.等,Dynamic changes ofmultiple forms of serum immunoactive relaxin during pregnancy in the rat.Endocrinology 114:806-13(1984))。虽然基于报道松弛素仅在怀孕期间的性器官有活性的早期研究,最初将松弛素分类为“怀孕激素”,但是因为在除性器官外的其它器官和组织中发现其活性,所以最近认为其为“万能激素”(Hisaw,F.L.,等,Effects of relaxin on the endotheliumof endometrial blood vessels in monkeys(Macaca mulatta).Endocrinology81:375-85(1967))。
已知松弛素有助于胎盘和子宫的生长和再生并在分娩期间松弛子宫颈以加宽产道。其促进产道组织中如MMP2、MMP3和MMP9的多种MMP的表达以降解胶原,以使结缔组织和基底膜降解以导致产道的细胞外基质破坏。除此之外,在松弛素作为抑制剂起作用以防止胶原过表达的肺、心脏、皮肤、肠道、乳腺、血管和精管中也观察到由松弛素促进的MMP1和MMP3的表达(Qin,X.,等,Biol Reprod 56:800-11(1997);Qin,X.,等,Biol Reprod 56:812-20(1997);和Palejwala,S.等,Endocrinology 142:3405-13(2001))。
根据本发明的基因送递系统,表达的松弛素蛋白质诱导细胞外基质周围细胞的主要组分胶原的降解,以破坏结缔组织和基底膜,因此导致细胞外基质的降解。这种连续作用是在下述实施例中清楚地证实的通过松弛素提高转导效率的一种机制。
因此,参考松弛素的上述作用,本基因送递系统的优点集中在包括通过细胞外基质互相连接的细胞的组织内的细胞。具体而言,当用于由结缔组织紧密包围的肿瘤组织,与任何常规送递系统相比,本基因送递系统表现出提高的转导效率。
在此所用的术语“基因送递”是指将基因转移到细胞中并与基因转导具有相同的含义。在组织水平,基因送递与基因扩散成为同义。因此,本发明的基因送递系统也表示为基因转导系统或基因扩散系统。
为了构建本发明的基因送递系统,优选在合适的表达构建体中包含编码松弛素的核苷酸序列。根据所述的构建体,优选编码松弛素的核苷酸序列与启动子可操作地连接。术语“可操作地连接”是指核酸表达控制序列(如启动子、信号序列或转录因子结合位点的阵列)与第二个核酸序列之间的功能性连接,其中所述表达控制序列影响对应于第二序列的核酸的转录和/或翻译。根据本发明,与松弛素基因连接的启动子优选在动物细胞中、更优选在哺乳动物细胞中是可操作的,以控制松弛素基因的转录,所述启动子包括来源于哺乳动物细胞基因组或哺乳动物病毒的启动子,例如,CMV(巨细胞病毒)启动子、腺病毒晚期启动子、痘苗病毒7.5K启动子、SV40启动子、HSV tk启动子、RSV启动子、EF1α启动子、金属硫蛋白启动子、β-肌动蛋白启动子、人IL-2基因启动子、人IFN基因启动子、人IL-4基因启动子、人淋巴毒素基因启动子和人GM-CSF基因启动子。最优选地,所述启动子为CMV启动子。

根据本发明的优选实施方式,用于编码松弛素的核苷酸序列的表达构建体具有“启动子-编码松弛素的核苷酸序列-多腺苷酸化序列”的结构。
在本基因送递系统中,待送递到细胞中的目的核苷酸序列可包含在与用于编码松弛素的核苷酸序列具有相同结构的表达构建体内。
例如,待送递到细胞中的目的核苷酸序列可为包括编码具有抗肿瘤活性并最终变性肿瘤细胞的蛋白质的癌治疗基因的任一序列,所述癌治疗基因如肿瘤抑制基因、免疫调节基因[例如,细胞因子基因、趋化因子基因和共刺激因子基因(对于T细胞活性如B7.1和B7.2)]、抗原基因、自杀基因、细胞毒性基因、细胞抑制基因、前凋亡基因和抗血管生成基因,但不局限于此。
所述自杀基因编码能够赋予肿瘤细胞对化学治疗剂的敏感性的蛋白质,或能够诱导肿瘤细胞中毒性状况的蛋白质。最公知的自杀基因是单纯疱疹病毒-胸苷激酶(herpes simplex virus-thymidine kinase,HSV-TK)基因(美国专利号5,631,236和5,601,818)。表达HSV-TK的细胞易于通过更昔洛韦(gancyclovir)选择性细胞死亡。所述肿瘤抑制基因编码多肽以抑制肿瘤发生。所述肿瘤抑制基因是哺乳动物细胞中固有的并且认为它们的缺失或失活是肿瘤发生的前提。肿瘤抑制基因的实例包括以APC、DPC4、NF-1、NF-2、MTS1、WT1、BRCA1、BRCA2、VHL、p53、Rb、MMAC-1、MMSC-2、成视网膜细胞瘤基因为例的肿瘤抑制基因INK4家族的成员(Lee等,Nature,329:642(1987))、腺瘤性结肠息肉病蛋白基因(Albertsen等,美国专利号5783,666)、在染色体3p21.3上作图的鼻咽癌肿瘤抑制基因(Cheng等,Proc.Na tl. Acad.Sci.,95:3042-3047(1998))、缺失结肠癌(colon carcinoma,OCC)基因、MTS1、CDK4、VHL、p100Rb、p16和p21、和其治疗有效片段(例如,p56Rb、p94Rb)。本领域普通技术人员应理解可使用其它已知的抗肿瘤基因。
在此所用的术语“抗原基因”是指编码通过免疫系统识别的抗原细胞表面蛋白的核苷酸序列。所述抗原基因的实例包括癌胚抗原(carcinoembryonic antigen,CEA)、前列腺特异抗原(prostate specificantigen,PSA)、甲胎蛋白(α-feto protein,AFP)和p53(WO94/02167)。为了促进免疫识别,所述抗原基因可与MHCI型抗原融合。
在此所用的术语“细胞毒性基因”是指其在细胞中的表达引起毒性作用的核苷酸序列。细胞毒性基因的实例包括编码假单胞菌外毒素、蓖麻毒素、白喉毒素等的核苷酸序列。
这里所用的术语“细胞抑制基因”是指其在细胞中的表达诱导细胞周期的停滞的核苷酸序列。所述细胞抑制基因的实例包括但不限于p21、成视网膜细胞瘤基因、E2F-Rb融合蛋白基因、如p16、p15、p18和p19的编码细胞周期蛋白依赖性激酶抑制剂的基因、生长停滞特异性同源异型框(growth arrest specific homeobox,GAX)基因(WO97/16459和WO96/30385)。
此外,在本发明的基因送递系统中可以携带用于治疗多种疾病的各种治疗基因。所述治疗基因的非限制实例包括编码细胞因子(例如,干扰素-α、干扰素-β、干扰素-δ、干扰素-γ)、白细胞介素(例如,IL-1、IL-2、IL-4、IL-6、IL-7、IL-10、IL-12、IL-19和IL-20)、集落刺激因子(例如,GM-CSF和G-CSF)、趋化因子基因[单核细胞趋化蛋白1(MCP-1)、单核细胞趋化蛋白2(MCP-2)、单核细胞趋化蛋白3(MCP-3)、单核细胞趋化蛋白4(MCP-4)、巨噬细胞炎性蛋白1α (MIP-1α)、巨噬细胞炎性蛋白1β(MIP-1β)、巨噬细胞炎性蛋白1γ(MIP-1γ)、巨噬细胞炎性蛋白3α(MIP-3α)、巨噬细胞炎性蛋白3β(MIP-3β)、趋化因子(ELC)、巨噬细胞炎性蛋白4(MIP-4)、巨噬细胞炎性蛋白5(MIP-5)、LD78β、RANTES、SIS-ε(p500)、胸腺和激活可调节趋化因子(thymus and activation-regulated chemokine,TARC)、嗜酸细胞活化趋化因子(eotaxin)、I-309、人蛋白HCC-1/NCC-2、人蛋白HCC-3和鼠蛋白C10]的基因。此外,所述治疗基因包括编码组织型纤溶酶原激活物(tissue-type plasminogen activator,tPA)或尿激酶型纤溶酶原激活物的基因和LAL-产生基因以提供用于防止高胆固醇血症的持续的血栓溶解。此外,已知用于治疗包括囊性纤维化、腺苷脱胺酶缺陷、AIDS和其它感染性疾病以及恶性和炎性疾病的多种疾病的核苷酸序列可用作治疗基因。
这里所用的术语“前凋亡基因”是指其表达导致凋亡的核苷酸序列。所述前凋亡基因的实例包括p53、腺病毒E3-11.6K(来源于Ad2和Ad5)或腺病毒E3-10.5K(来源于Ad)、腺病毒E4基因、Fas配体、TNF-α、TRAIL、p53途径基因和编码一系列胱天蛋白酶(caspase)的基因。
这里所用的术语“抗血管生成基因”是指其表达导致抗血管生成因子的细胞外分泌的核苷酸序列。抗血管生成因子包括血管他丁(angiostatin)、如Tie2的血管内皮生长因子(VEGF)的抑制剂(PNAS,1998,95,8795-8800)、和内皮他丁(endostatin)。
从如GenBank和EMBL的DNA序列数据库可获得上述目的核苷酸序列。
本发明的基因送递系统以多种形式构建,优选地,(i)裸露的重组DNA分子,(ii)质粒,(iii)病毒载体,或(iv)含有裸露的重组DNA 分子和质粒的脂质体或新核蛋白体。
编码松弛素的核苷酸序列可应用于许多用于基因治疗的基因送递系统,优选质粒、腺病毒(Lockett LJ,等,Clin.Cancer Res.3:2075-2080(1997))、腺伴随病毒(AAV,Lashford LS.,等,Gene TherapyTechnologies,Applications and Regulations Ed.A.Meager,1999)、反转录病毒(Gunzburg WH,等,Retroviral vectors.Gene Therapy Technologies,Applications and Regulations Ed.A.Meager,1999)、慢病毒(Wang G.等,J. Clin.Invest.104(11):R55-62(1999))、单纯疱疹病毒(ChamberR.,等,Proc.Natl.Acad.Sci USA 92:1411-1415(1995))、痘苗病毒(Puhlmann M.等人,Human Gene Therapy 10:649-657(1999))、脂质体(Methods in Molecular Biology,Vol 199,S.C.Basu和M.Basu(编者),Human Press 2002)或新核蛋白体。最优选地,通过将编码松弛素的核苷酸序列整合到腺病毒构建本发明的基因送递系统。
(i)腺病毒
因为腺病毒基因组中等大小、易于操作、高效价、宽靶细胞范围和高感染性,所以腺病毒已通常用作基因送递载体。基因组的两端均含有100-200bp对于病毒DNA复制和包装必需的顺式元件ITR(反向末端重复序列)。E1区(E1A和E1B)编码负责调节病毒基因组和几种细胞基因的转录的蛋白质)。E2区(E2A和E2B)的表达引起用于病毒DNA复制的蛋白质的合成。
在至今开发的腺病毒载体中,通常使用具有缺失E1区的复制缺陷型腺病毒。腺病毒载体中缺失E3区可提供用于转基因的插入位点(Thimmappaya,B.等,Cell,31:543-551(1982);和Riordan,J.R.等,Science,245:1066-1073(1989))。因此,优选将编码松弛素的核苷酸序列插入到缺失的E1区(E1A区和/或E1B区,优选E1B区)或缺失的 E3区,更优选地,缺失的E3区。优选将待送递的目的核苷酸序列插入到缺失的E1区(E1A区和/或E1B区,优选E1B区)或缺失的E3区,更优选缺失的E1区。此外,插入序列可整合到缺失的E4区。参考病毒基因组的术语“缺失”包含全部缺失以及部分缺失。
根据最优选的实施方案,本发明的腺病毒基因送递载体包括“启动子-目的核苷酸序列-poly A序列”和“启动子-松弛素基因-poly A序列”。所述启动子-目的核苷酸序列-poly A序列优选存在于缺失的E1区(E1A区和/或E1B区,优选E1B区)或缺失的E3区,更优选缺失的E1区。所述启动子-松弛素基因-poly A序列优选存在于缺失的E1区(E1A区和/或E1B区,优选E1B区)或缺失的E3区,更优选缺失的E3区。此外,所述腺病毒基因送递系统可包括双顺反子表达系统,其中,目的核苷酸序列和编码松弛素的核苷酸序列通过IRES(internalribosome entry site,内部核糖体进入位点)互相连接以形成“启动子-目的核苷酸序列-poly A序列-松弛素基因-poly A序列”。
在自然界,腺病毒可包装约105%的野生型基因组,提供约额外2kbDNA的能力(Ghosh-Choudhury等,EMBO J.,6:1733-1739(1987))。在这方面,插入到腺病毒中的上述外源序列还可插入到腺病毒野生型基因组。
所述腺病毒可为42种不同的已知血清型或A-F亚类中的任一种。C亚类的5型腺病毒是用于构建本发明的腺病毒基因送递系统的最优选的原料。关于5型腺病毒的很多生物化学和遗传学的信息是已知的。
通过本腺病毒基因送递系统送递的外源基因是游离基因,所以,对宿主细胞具有低的遗传毒性。因此使用本发明的腺病毒基因送递系统的基因治疗可相当安全。
(ii)反转录病毒
由于反转录病毒将其基因组整合到宿主基因组并具有宽宿主范围,能够携带相对大的外源基因的反转录病毒已用作病毒送递载体。
为了构建反转录病毒载体,在一定病毒序列的位置将编码松弛素的核苷酸序列和待转移的目的核苷酸序列插入到病毒基因组中以产生复制缺陷型病毒。为了产生毒粒,构建了含有gag、pol和env基因但没有LTR(long terminal repeat,长末端重复)和ψ组分的包装细胞系(Mann等,Cell,33:153-159(1983))。当将含有编码松弛素的序列、目的核苷酸序列、LTR和ψ的重组质粒引入到该细胞系中时,ψ序列使重组质粒的RNA转录本包装进毒粒中,然后将毒粒分泌到培养基中(Nicolas和Rubinstein“Retroviral vectors,”In:Vectors:A survey ofmolecular cloning vectors and their uses,Rodriguez和Denhardt(编者),Stoneham:Butterworth,494-513(1988))。然后收集含有重组反转录病毒的培养基,选择性地进行浓缩并用于基因送递。
已报道使用第二代反转录病毒载体的成功的基因转移。Kasahara等(Science,266:1373-1376(1994))制备了莫洛尼小鼠白血病毒变体,其中,将EPO(红细胞生成素)序列插入包膜区的位置,因此产生具有新结合性质的嵌合蛋白质。同样,根据用于第二代反转录病毒载体的构建策略,可以构建本基因送递系统。
(iii)AAV载体
腺伴随病毒能够感染不分裂细胞和多种类型的细胞,使其可用于构建本发明的基因送递系统。美国专利号5,139,941和4,797,368中可以找到AAV载体的用途和制备的详细说明。
在LaFace等,Viology,162:483486(1988),Zhou等,Exp.Hematol.(NY),21:928-933(1993),Walsh等,J. Clin.Invest.,94:1440-1448(1994)和Flotte等,Gene Therapy,2:29-37(1995)中公开了AAV作为 基因送递系统的研究结果。最近,已批准AAV载体用于治疗囊性纤维化的I期人体试验。
一般地,通过共转染含有侧接两个AAV末端重复的目的基因(即,松弛素基因和待送递的目的核苷酸序列)的质粒(McLaughlin等,1988;Samulski等,1989)和含有野生型AAV编码序列而没有末端重度的表达质粒(McCarty等,J.Virol.,65:2936-2945(1991))制备重组AAV病毒。
(iv)其它病毒载体
其它病毒载体可以用作本发明的基因送递载体。在本发明的基因送递系统中可以使用来源于如痘苗病毒(Puhlmann M.等,Human GeneTherapy 10:649-657(1999);Ridgeway,″Mammalian expression vectors,″In:Vectors:A survey of molecular cloning vectors and their uses.Rodriguez和Denhardt,编者,Stoneham:Butterworth,467-492(1988);Baichwal和Sugden,″Vectors for gene transfer derived from animal DNAviruses:Transient and stable expression of transferred genes,″In:Kucherlapati R,编者Gene transfer.纽约Plenum Press,117-148(1986)和Coupar等,Gene,68:1-10(1988))、慢病毒(Wang G.等,J.Clin.Invest.104(11):R55-62(1999))和单纯疱疹病毒(Chamber R.,等,Proc.Natl.Acad.Sci USA 92:1411-1415(1995))的病毒的载体可用于将松弛素基因和目的核苷酸序列转移到细胞中的本送递系统。
(v)脂质体
当磷脂在过量的含水培养基中悬浮时自然形成脂质体。如在Nicolau和Sene,Biochim.Biophys.Acta,721:185-190(1982)和Nicolau等,Methods Enzymol.,149:157-176(1987)中所述,脂质体介导的核酸送递已非常成功。用于用脂质体转染动物细胞的可商业获得的试剂的 实例包括脂转染胺(Lipofectamine)(Gibco BRL)。捕捉松弛素基因和目的核苷酸序列的脂质体与细胞通过如内吞、吸收和融合的机制相互作用并然后将所述序列转移到细胞中。
在本发明的另一技术方案中,提供一种用于送递基因的方法,所述方法包括将上述本发明的基因送递系统与含有细胞的生物样品接触。
由于在病毒载体构建的基础上构建本基因送递系统,所以如本领域已知的常规感染方法进行接触。在上面引用的出版物中很好地描述了使用病毒载体感染宿主。
由于基因送递系统是裸露的重组DNA分子或质粒,通过微注射(Capecchi,M.R.,Cell,22:479(1980)以及Harland和Weintraub,J.Cell Biol.101:1094-1099(1985))、磷酸钙共沉淀(Graham,F.L.等,Virology,52:456(1973)以及Chen和Okayama,Mol.Cell.Biol.7:2745-2752(1987))、电穿孔(Neumann,E.等,EMBO J.,1:841(1982)以及Tur-Kaspa等,MoI.Cell Biol.,6:716-718(1986))、脂质体介导的转染(Wong,T.K.等,Gene,10:87(1980)以及Nicolau和Sene,Biochim.Biophys.Acta,721:185-190(1982);和Nicolau等,Methods Enzymol.,149:157-176(1987))、DEAE-葡聚糖处理(Gopal,Mol.Cell Biol.,5:1188-1190(1985))和粒子高速撞击(Yang等,Proc.Natl.Acad.Sci.,87:9568-9572(1990))将编码松弛素的序列和待送递的核苷酸序列引入到细胞中。
在本发明的又另一技术方案中,提供包括腺病毒ITR(反向末端重复序列)核苷酸序列和编码松弛素的核苷酸序列的重组腺病毒;其中表达的松弛素蛋白增强了重组腺病毒进入肿瘤组织的穿透能力和用重组腺病毒感染的肿瘤细胞凋亡。
在本发明的重组腺病毒中,表达的松弛素蛋白显著增强了重组腺病毒进入肿瘤组织的穿透能力和用重组腺病毒感染的肿瘤细胞的编程性细胞死亡,使腺病毒的治疗效能大大提高。
已知小部分腺病毒基因组作为顺式元件是必需的(Tooza,J.Molecular biology of DNA Tumor viruses,第2版,冷泉港实验室,冷泉港,纽约(1981)),尤其是与如293的合适的细胞系一起使用,允许用外源序列替代大片段的腺病毒DNA。在本文中,所述重组腺病毒包括作为必需序列的腺病毒ITR序列以及编码松弛素的核苷酸序列。
优选将编码松弛素的核苷酸序列插入到缺失的E1区(E1A区和/或E1B区,优选E1B区)或缺失的E3区,更优选缺失的E3区。将待送递的目的核苷酸序列(例如,细胞因子基因、免疫共刺激因子基因、凋亡基因和肿瘤抑制基因)插入到缺失的E1区(E1A区和/或E1B区,优选E1B区)或缺失的E3区,更优选缺失的E1区(E1A区和/或E1B区),最优选E1B区。此外,插入序列可整合到缺失的E4区。

根据本发明的优选实施方案,本发明的重组腺病毒包含失活的E1B19基因、失活的E1B55基因或失活的E1B19/E1B55基因。这里所用的与基因连接的术语“失活”是指使基因的转录和/或翻译无功能发生,因此不能产生基因编码的蛋白质的正确功能的条件。例如,失活的E1B19基因是通过突变(替代、添加、以及部分或全部缺失)不能产生功能性E1B19kDa蛋白的基因。E1B19缺失引起凋亡发生率增加并且缺失E1B55使重组腺病毒具有肿瘤特异性(参见韩国专利号 10-2002-0023760)。
根据优选的实施方式,本发明的重组腺病毒包括有活性的E1A基因。携带有活性的E1A基因的腺病毒是增殖型的。更优选,本发明的重组腺病毒包括失活的E1B19/E1B55基因和有活性的E1A基因。最优选,所述重组腺病毒包括灭活的E1B19/E1B55基因、有活性的E1A基因和缺失E3区的位置的编码松弛素的序列。
根据最优选的实施方式,本发明的重组腺病毒包括“ITR-E1A-ΔE1B-启动子-松弛素基因-poly A序列”结构,其中,启动子-松弛素基因-poly A序列存在于缺失的E3区。本发明的示例性腺病毒具有由图1的Ad-ΔE1B-RLX表示的遗传图。
与常规抗肿瘤腺病毒相比,本发明的重组腺病毒显示进入肿瘤的高度改善的转导(穿透)效率以及凋亡能力。这些改善的功效主要归于松弛素有效地降解细胞外基质和提高凋亡能力。因此,本发明的重组腺病毒表现出显著增强的溶瘤作用。
肿瘤组织不是单独由肿瘤细胞组成的聚团而是还包含血管和正常细胞的复杂结构。尤其是,肿瘤组织中的结缔组织通常是坚硬的并形成围绕肿瘤细胞的紧密的细胞外基质。因此,抗肿瘤药物以及病毒不能有效穿透到肿瘤中,从而它们通常显示有限的抗肿瘤作用。用含有所述松弛素基因的本发明的重组腺病毒可以克服此类障碍。
如在下述实施例所示,具有插入的松弛素基因的本发明的腺病毒活跃地扩散甚至进入到肿瘤球状体的中央及其表面。对于体内肿瘤组织,本发明的表达松弛素的腺病毒从注射位点(针迹)广泛并远距离地扩散到远端位点。通过表达松弛素的腺病毒实现的转导效率的提高是明显的,甚至用肉眼可以容易地区分。应当理解与如胶原酶/分散酶或胰蛋白酶、弹性蛋白酶的蛋白酶预处理以降解弹性蛋白或透明质酸 酶预处理以降解细胞外基质的转导效率提高约2~3倍相比,提高的转导效率是非常显著的。
通过松弛素在组织中增强的扩散作用可以大大地提高肿瘤特异溶瘤腺病毒的抗肿瘤效力。这种提高的抗肿瘤效力可以在复制缺陷型腺病毒以及增殖型腺病毒中显示。通过松弛素增强腺病毒诱导凋亡的能力是令人惊奇的和没有预料到的。
在本发明的另一技术方案中,提供一种用于治疗癌症的抗肿瘤药物组合物,所述抗肿瘤药物组合物包含(a)治疗有效量的上述重组腺病毒;和(b)可药用载体。
本发明的又一技术方案中,提供一种用于治疗癌症的方法,所述治疗癌症的方法包括向动物施用上述抗肿瘤药物组合物。
在药物组合物中作为活性成分的重组腺病毒为上文描述的本发明的腺病毒并且因此以上描述可以适用于药物组合物的重组腺病毒。因此,为了避免导致本说明书复杂性的过度冗余,省略了它们之间的共有描述。
为了通过重组腺病毒有效地引发抗肿瘤作用,需要病毒增殖并扩散到邻近细胞快于癌细胞的生长率以诱导溶瘤作用。此外,使用腺病毒的成功的癌基因治疗需要增强安全性以及高治疗益处。本发明的表达松弛素的腺病毒提高了病毒扩散和凋亡以显示显著提高的抗肿瘤作用。尤其是,具有缺失E1B55基因的本发明的重组腺病毒显示细胞毒性中的优异的肿瘤特异性。因此,本发明的表达松弛素的腺病毒使用于癌症治疗的剂量降低,显著降低对正常细胞的毒性和不希望的体内免疫反应。
由于本发明的重组腺病毒对许多肿瘤细胞具有溶瘤作用,所以本发明的药物组合物可用于治疗肿瘤相关疾病,包括胃癌、肺癌、乳腺 癌、卵巢癌、肝癌、气管原癌、鼻咽癌、喉癌、胰腺癌、膀胱癌、结肠癌和子宫颈癌。这里所用的术语“治疗”是指(i)预防肿瘤发生;(ii)通过根除肿瘤细胞抑制和治疗肿瘤相关疾病或病症;和(iii)通过根除肿瘤细胞减轻肿瘤相关疾病或病症。因此,这里所用的术语“治疗有效量”表示足够达到上述药学作用的量。
在药物配方中通常采用,但不受限制的本发明的药物组合物中含有的可药用载体包括乳糖、葡萄糖、蔗糖、山梨醇、甘露醇、淀粉、阿拉伯橡胶(rubber arable)、磷酸钾、精氨酸盐(arginate)、明胶、硅酸钾、微晶纤维素、聚乙烯吡咯烷酮、纤维素、水、糖浆、甲基纤维素、羟基苯甲酸甲酯、羟基苯甲酸丙酯、滑石、硬脂酸镁和矿物油。根据本发明的药物组合物还可以包括润滑剂、湿润剂、增甜剂、调味剂、乳化剂、悬浮剂和防腐剂。
根据本发明的药物组合物可以通过在基因治疗中常用的途径施用,并且优选肠胃外施用,即通过静脉内、腹膜内、肌内、皮下施用或局部施用。例如,药物组合物可腹膜内施用以治疗卵巢癌和静脉内施用以治疗肝癌、直接注射到可见肿瘤块以治疗乳腺癌、直接灌肠注射以治疗结肠癌、和直接注射到导尿管以治疗膀胱癌。
本发明的药物组合物的合适剂量可以取决于药物制备方法、施用方法、患者的年龄、体重、性别、致病状态、饮食、施用时间、施用途径、排泄率和对所用药物组合物的敏感性而变化,并且本领域普通医师可以确定对于理想治疗的药物组合物的有效量。一般而言,本发明的药物组合物包括1×105~1×1015pfu/ml的重组腺病毒,并且一般两周隔日注射1×1010pfu的重组腺病毒。
根据本领域技术人员已知的常规技术,包含根据本发明的重组腺病毒的药物组合物可以与可药用载体和/或上述载体进行配制,最终提 供几种形式,一种单元药剂形式和一种多元药剂形式。剂型的非限制性实例包括但不限于溶液、在油或水介质中的悬浊液或乳浊液、提取物、酏剂、粉剂、颗粒剂、片剂和胶囊,并且还可包含分散剂或稳定剂。

实施例
材料和方法
细胞系和细胞培养
用于实验的细胞系为从美国标准培养物保藏中心(ATCC)可获得的人脑癌细胞系(U343、U87MG)、子宫颈癌细胞系(C33A)、肝癌细胞系(Hep3B)、肺癌细胞系(A549)和携带腺病毒早期基因E1区的293细胞系。所有细胞系在添加10%胎牛血清(Gibco BRL)、青霉素和链霉素的杜尔贝科变法伊格尔培养基(Dulbecco’s modified Eagle’medium,DMEM;Gibco BRL)中培养并保持在37℃5%CO2气氛中。
重组腺病毒的产生和滴定
为了产生表达松弛素和作为报道基因的lacZ的缺失E1/E3-基因的复制缺陷型的腺病毒,我们首先用vmdl324Bst(由瑞士Fribourgh大学Verca博士赠予;Heider,H.等,Biotechniques,28(2):260-265,268-270(2000)构建了在缺失E1区具有lacZ基因的pd1-LacZ病毒载体。为了制备该载体,用HindIII和NaeI消化pcDNA-hygro-LacZ质粒(Invitrogen,Carlsbad,加利福尼亚州,美国)以分离CMV启动子、lacZ基因和polA并将分离的三个序列插入到E1腺病毒穿梭载体pΔElsplA以制备pΔElsplA/CMV-LacZ穿梭载体。用XmnI消化制备的pΔElsplA/CMV-LacZ穿梭载体并与用BstBI线性化的vmdl324Bst腺病毒共转化到大肠杆菌(E.coli)BJ5183(瑞士Fribourgh大学Verca博士)中以诱导同源重组,获得pd1-LacZ腺病毒。
为了构建表达松弛素的腺病毒,用SalI-HindIII消化pDNR-LIB-RLX(ATCC,#MGC-14599)以获得1kb DNA片段,将所该片段亚克隆到pCA14载体(Microbix,安大略省,加拿大),产生pCA14-RLX。在实施例中所用的松弛素的核苷酸序列在GenBank登录号BC005956下公布。然后,用BglII将CMV-RLX-polA表达盒从pCA14-RLX切下并随后插入到腺病毒E3穿梭载体pSP72ΔE3(Promega,Madison,威斯康星州,美国)以构建pSP72ΔE3-cRLX E3 穿梭载体。用XmnI线性化构建的pSP72ΔE3-cRLX E3穿梭载体并然后与pdl-LacZ共转化到大肠杆菌BJ5183(瑞士Fribourgh大学Verca博士)以诱导同源重组,产生dl-LacZ-RLX(或dl-Z-RLX)腺病毒载体(图1)。2004年3月19日将dl-Z-RLX腺病毒保藏在韩国微生物培养中心(KCCM)登录号为KCCM-10567。
为构建表达松弛素基因的能复制的腺病毒,将以上制备的pSP7ΔE3-cRLX E3穿梭载体用XmnI线性化并与用SpeI线性化的缺失E1B19kDa/E1B55kDa的pAdΔE1B19/55腺病毒载体(KFCC11288)一起共转化到大肠杆菌BJ5183中用于同源重组,产生Ad-ΔE1B-RLX腺病毒载体(图1)。2004年3月19日将Ad-ΔE1B-RLX腺病毒保藏在韩国微生物培养中心(KCCM)登录号为KCCM-10566。
在图1中,ψ表示包括ITR(反向末端重复序列)和包装信号的序列,Ad表示腺病毒,CMV表示CMV启动子,Pol A为poly A序列,IX表示编码IX蛋白的基因。
为了证实各自的同源重组,用HindIII消化质粒DNA并分析消化图样。将适当的同源重组的腺病毒质粒DNA用PacI消化并转染到293细胞中以产生dl-lacZ-RLX和Ad-ΔE1B-RLX腺病毒。所有病毒在293细胞中增殖并根据有限稀释或空斑试验(Hitt,M.等,Construction andpropagation of human adenovirus vectors.Cell biology:a laboratoryhandbook.纽约:Academic Press Inc,479-490(1994))进行其滴定,接着用CsCl梯度浓缩和纯化。
根据前述步骤,用pCA14作为E1穿梭载体构建并制备Ad-ΔE1B(缺失E1B区)作为对照病毒。
dl-LacZ-RLX和Ad-ΔE1B19/55-RLX中松弛素表达模式的检验
用dl-LacZ-RLX或Ad-ΔEIB-RLX或dl-LacZ或Ad-ΔE1B19腺
病毒(KFCC-11288)以MOI(multiplicity of infection,感染复数)1~50感染人子宫颈癌细胞系C33A并在24小时后回收所用的培养基。根据制造商的方法用ELISA试剂盒(Immune diognostik,Benshem,德国)分析松弛素的表达。
肿瘤球形体中dl-LacZ-RLX的扩散和穿透性的评估
通过将细胞注射到6~8周龄裸鼠的腹部皮下建立U343、U87MG、C33A和A549异体移植物并且一旦肿瘤体积达到150~200mm3时,将新鲜的肿瘤组织用手术除去。将肿瘤组织切成1~2mm碎片。将这些移植物各个置于涂布0.75%琼脂糖的板上并在添加5%FBS(Gibco BRL)和青霉素/链霉素(Gibco BRL)的DMEM(Gibco BRL)中5%CO2 气氛下37℃培养。培养基每周更新一次。用腺病毒感染前,将直径2mm的球形体转移到涂布0.75%琼脂糖的48-孔板上并加入150μl DMEM(含有5%FBS),然后加入1×106、1×107或1×108PFU的病毒。48小时后,将培养基吸出并将球形体在固定液中固定用于X-gal染色。在体视镜下观察X-gal染色的球形体的表面。为了观察腺病毒穿透肿瘤球形体,将X-gal染色的肿瘤球形体包埋在O.C.T.化合物(Sakura Finetec,托伦斯市,加利福尼亚州)中并快速冷冻。然后将8μm冷冻切片置于涂布明胶的载玻片上。
体内dl-LacZ-RLX的扩散和穿透性的评估
通过将细胞注射到6-~8周龄裸鼠的腹部皮下建立了U343、U87MG、C33A、Hep3B和A549异体移植物并且一旦肿瘤体积达到150~200mm3时,将小鼠随机分为两组并3次以50μl中5×107~1×108 PFU的dl-LacZ-RLX腺病毒肿瘤内注射到肿瘤中。最后注射后3天,将动物处死并取肿瘤用于病毒分布,然后将它们在4℃的4%低聚甲醛中固定4~8小时并在30%的蔗糖溶液中脱水12小时。将脱水的肿瘤 组织在O.C.T.化合物中冷冻并切成厚度为8μm的切片,然后将其置于涂布明胶的载玻片上用于X-gal染色。
Ad-ΔE1B-RLX病毒的致细胞病变效应(CPE)
为了评估表达松弛素的腺病毒的溶瘤活性,将人肿瘤细胞系(U343、U87MG、C33A、Hep3B和A549)置于24-孔板上并然后用MOI为0.1~10的Ad-ΔE1、Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染。当MOI为0.1~0.5用任一所述病毒感染的细胞显示完全的细胞溶解时,将死细胞洗去并将板上的细胞用50%甲醇中的0.5%结晶紫染色。
空斑发生试验
为了观察松弛素表达的空斑大小的变化,将3×105个Hep3B细胞置于6-孔板并在细胞生长1天后以3MOI的Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染。培养4小时后,将感染细胞用37℃的2×DMEM(含有10%FBS和青霉素/链霉素)和42℃的1.4%琼脂糖的琼脂糖-DMEM混合物覆盖然后在37℃的5%CO2培养箱中培养。培养约10天后,以10%TCA(三氯乙酸)浸泡30分钟后,将琼脂糖覆盖层除去,并将剩余细胞用50%甲醇中的0.5%结晶紫染色。
凋亡能力的流式细胞术分析
为了检验由松弛素诱导的凋亡,将人肿瘤细胞系U343、U87MG、C33A、Hep3B和A549引入到25T培养瓶,24小时后,用MOI为0.5~5的Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染。用0.1~1μM CPT-11(camptothecin,喜树碱)处理的细胞作为正对照并且用PBS处理的细胞作为负对照。感染48小时、72小时和96小时后,收集感染细胞并用70%乙醇在4℃固定24小时。固定后,将细胞与PI(碘化丙锭,50μg/μl)和RNA酶的混合物培养15分钟并进行流式细胞术分析。此外,为了检验由松弛素诱导的早期凋亡,如上述用Ad-ΔE1B或Ad- ΔE1B-RLX腺病毒感染几种人肿瘤细胞系。收集感染细胞然后根据ApoAlert V-FITC凋亡试剂盒(Clontech,Palo Alto,加利福尼亚州)中制造商说明,进行膜连蛋白V/PI双重染色,然后进行流式细胞术分析。
TUNNEL试验
将U343(5×104)、U87MG(5×104),C33A(5×105)、Hep3B(4×105)和A549(5×104)细胞置于腔室玻片上然后用MOI为0.2~20的腺病毒感染。感染24小时和48小时后,除去培养基并根据ApopTag试剂盒(Intergen,Purchase,纽约)的制造商说明书进行TUNNEL测定。为了显色,将细胞与过氧化物酶结合的抗生物素蛋白链菌素培养然后与二氨基联苯胺(DAKO,Carpinteria,加利福尼亚州)培养。细胞的颜色变成棕色时,将细胞用0.5%的甲基绿复染10分钟并在显微镜下观察4个以上选定的视野。计算染色细胞与总细胞的比率。
表达松弛素的腺病毒的体内抗肿瘤作用
为了评估AdΔE1B-RLX腺病毒对裸鼠中形成的人肿瘤球形体生长的作用,通过皮下注射1×107个癌细胞(U343、U87MG、C33A、Hep3B和A549)将肿瘤植入到6~8-周龄裸鼠腹部。当肿瘤达到50~80mm3范围时,隔日3次肿瘤内施用将PBS中的5×107~1×108PFU的Ad-ΔE1B或Ad-ΔE1B-RLX,并观察肿瘤的生长模式。用游标卡尺测量长轴和短轴计算肿瘤的体积:体积=(短轴mm)2×(长轴mm)×0.523。
由施用表达松弛素的增殖型腺病毒诱导的肿瘤特征变化的观察
当在裸鼠腹部形成的C33A肿瘤达到50~80mm3时,将PBS中的5×107~1×108PFU的Ad-ΔE1B或Ad-ΔE1B-RLX肿瘤内施用3次。注射3天后,取出肿瘤组织并制备其石蜡块。将石蜡块切成4μm切片并在二甲苯中然后在分级乙醇(100%、95%、80%和70%)中脱蜡,然后用苏木精和伊红染色。为了观察胶原的分布,将结缔组织组分,4μm 石蜡包埋的切片用boulin、苏木精和biebrich′s深红酸性品红染色。
此外,进行腺病毒六联体区的免疫组织化学染色。将切片如上所述脱蜡并与原发抗腺病毒六联体抗体AB1056F(Chemicon,Temecula,CA)培养,然后与次级山羊抗-大鼠IgG-HRP(Santa Cruz Biotechnology,Inc.,Santa Cruz,CA)培养。用DAB(DAKO,Carpinteria,加利福尼亚州)进行显色。
为了观察肿瘤中凋亡的发生,根据ApopTag试剂盒(Intergen,Purchase,NY)制造商说明书进行TUNNEL试验。为了显色,将细胞与过氧化物酶结合的抗生物素蛋白链菌素培养然后与二氨基联苯胺(DAKO,Carpinteria,CA)培养。当细胞的颜色变为棕色时,将细胞用0.5%的甲基绿复染10分钟并在显微镜下观察。
鼠B16BL6自发肺转移模型
自发转移模型用于检验施用松弛素基因对肿瘤转移的作用。更具体而言,将B16BL6细胞(2×105/鼠)皮下施用到6-周龄雄性C57BL/6鼠(Charles River Korea,首尔,韩国)的右后足垫。一旦肿瘤体积达到100~200mm3,将动物随机分为3组(PBS、Ad-ΔE1B、Ad-ΔE1B-RLX)每组5只,开始处理。处理的第1天指定为1日,在1、3和5日,将腺病毒或PBS直接注射到肿瘤(50μl PBS中每个肿瘤5×108PFU)。在7日,在温和麻醉下,将原发性肿瘤通过膝盖以下切断手术去除。在去除原发性肿瘤后25日,评估鼠肺中的转移肿瘤病灶的重量。
对瘢痕疙瘩细胞、细胞球形体和组织球形体的转导效率的评估
为了评价表达松弛素的腺病毒对瘢痕疙瘩的治疗功效,我们检验了对瘢痕疙瘩细胞、细胞球形体和组织球形体的转导效率和病毒对组织的穿透效率。
对瘢痕疙瘩细胞的基因转导效率评估如下:将从瘢痕疙瘩患者获得的第二代的原代瘢痕疙瘩细胞系转移到24-孔板然后以MOI为10或50的Ad-ΔE1B或Ad-ΔE1B-RLX病毒感染,然后在病毒感染2天后用X-gal染色。
对瘢痕疙瘩细胞球形体的基因转导效率评估如下:将瘢痕疙瘩组织从瘢痕疙瘩患者取出。将从取出的瘢痕组织获得的1×105/ml的第二代原代细胞转移到培养管并500×g离心5分钟,然后将其在37℃培养。当1~5天培养圆颗粒从管底脱落显示为细胞球形体时,将球形体转移到涂布0.75%琼脂糖的48-孔板并加入150μl DMEM(含有5%FBS),然后用1×107PFU的腺病毒感染球形体。病毒感染3天后,将瘢痕疙瘩球形体用固定液固定并用X-gal染色。在体视显微镜下观察X-gal染色的球形体表面。对于腺病毒进入球形体的穿透的观察,将X-gal染色的瘢痕疙瘩球形体包埋在O.C.T.化合物中并快速冷冻。然后将10μm冷冻切片置于涂布明胶的载玻片上。
病毒进入瘢痕疙瘩组织球形体的穿透性分析如下:将瘢痕疙瘩组织从瘢痕患者取出并切成1~2mm的切片。将切片在涂布0.75%琼脂糖的培养箱中在含有5%FBS和青霉素/链霉素的DMEM中培养。每周一次或两次更换所用的培养基并将瘢痕疙瘩组织培养一周以上。将直径为2mm的瘢痕疙瘩组织球形体转移到涂布0.75%琼脂糖的48-孔板上并加入150μl DMEM(含有5%FBS),然后用1×108PFU的腺病毒感染。病毒感染3天后,将瘢痕疙瘩撒组织用固定液固定并用X-gal染色。在体视显微镜下观察X-gal染色的球形体的表面。为了观察腺病毒进入球形体的穿透性,将X-gal染色的瘢痕疙瘩球形体包埋在O.C.T.化合物中并快速冷冻。然后将10μm冷冻切片置于涂布明胶的载玻片上。
统计学分析
数据表示为平均值±平均标准误(SEM)。用Stat View软件(Abacus Concepts,Inc.,伯克利,加利福尼亚州)和Mann-Whitney试验(nonparametric rank sum test,非参量型秩总检验)进行统计学比较。统计学显著性的标准为P<0.05。
结果
表达松弛素的腺病毒的构建和松弛素的表达模式
为了真实评估依赖于松弛素表达的进入组织的穿透效率的变化,构建表达作为报道基因的LacZ的复制缺陷型的dl-LacZ-RLX腺病毒。此外,构建了肿瘤特异性溶瘤Ad-ΔE1B-RLX腺病毒以增强增殖型腺病毒进入组织的转导效率(图1)。为了评估构建的腺病毒的松弛素表达模式,以多种MOI的dl-LacZ、dl-LacZ-RLX、Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染子宫颈癌细胞系C33A并将培养基回收用于ELISA(图2)。作为复制缺陷型的腺病毒的负对照的dl-LacZ和作为肿瘤特异性溶瘤腺病毒的负对照的Ad-ΔE1B感染的细胞显示不表达松弛素,而用dl-LacZ-RLX和Ad-ΔE1B-RLX感染的细胞显示取决于施用腺病毒的滴度的松弛素的剂量依赖性表达。
使用肿瘤球形体的dl-LacZ-RLX腺病毒进入体外肿瘤组织的转导效率的评估
为了评估dl-LacZ-RLX进入肿瘤球形体的转导效率和组织穿透性,将多种人肿瘤细胞系皮下注射到裸鼠中并且一旦肿瘤体积达到150~200mm3,将新鲜的肿瘤组织取出。将取出的肿瘤组织切成1~2mm碎片并用1×106、1×107或1×108PFU的腺病毒感染。感染48小时后用X-gal染色。与用1×106PFU的dl-LacZ处理相比,相同剂量的dl-LacZ-RLX在肿瘤球形体表面显示更强的X-gal染色。1×107或1×108 PFU的dl-LacZ-RLX在肿瘤球形体的所有表面显示更深的X-gal染色。为了准确研究腺病毒进入肿瘤球形体的穿透性,将X-gal染色的肿瘤球形体切开用于观察。1×106、1×107或1×108PFU的dl-LacZ在肿瘤组织中显示弱LacZ表达,且其扩散局限于肿瘤球形体的表面。相反,相同剂量的dl-LacZ-RLX显示高很多的LacZ表达水平且其扩散延伸到肿瘤球形体的内部(图3)。该结果清楚地表明在肿瘤球形体的三维结构中,表达松弛素的dl-LacZ-RLX腺病毒以比对照载体dl-LacZ高的效率转导和扩散到球形体的核心。
体内肿瘤块中dl-LacZ-RLX腺病毒转导效率的评估
为了研究在体外肿瘤球形体中观察到的dl-LacZ-RLX的增强的转导效率和病毒扩散是否引起体内lacZ基因送递到肿瘤块提高,使用了肿瘤异种移植物模型。将5×107或1×108PFU的dl-LacZ或dl-LacZ-RLX腺病毒肿瘤内注射到在裸鼠腹部形成的肿瘤块中。3天后,取出肿瘤并进行切片用于X-gal染色。dl-LacZ显示低水平的LacZ表达并且染色区集中在病毒注射部位,而dl-LacZ-RLX显示高很多的LacZ表达并且发现染色区广泛分布在病毒注射部位之外的其它区域(图4)。尤其是,由于在所有肿瘤组织强烈的LacZ表达,所以用dl-LacZ-RLX转导的U87MG和C33A肿瘤块显示深蓝色。这些表达图式说明与没有表达松弛素的dl-LacZ对照病毒相比,在体内肿瘤块中dl-LacZ-RLX的穿透性和扩散增强。
表达松弛素的溶瘤腺病毒的肿瘤细胞杀伤作用的评估
为了揭示表达松弛素的腺病毒的穿透性和扩散提高有助于提高肿瘤特异性溶瘤腺病毒的肿瘤细胞杀伤作用,进行了CPE试验。用MOI为0.1~10的dl-LacZ(负对照)、Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染各人肿瘤细胞系(U343、U87MG、C33A、Hep3B和A549)并 对产生的肿瘤细胞杀伤作用进行分析。如图5所示,负对照dl-LacZ在各种肿瘤细胞系中引起很小的或不引起细胞杀伤作用,而Ad-ΔE1B-RLX显示比不表达松弛素的Ad-ΔE1B的肿瘤杀伤作用高约2~10倍。尤其是,在Hep3B细胞系中,Ad-ΔE1B-RLX腺病毒显示比Ad-ΔE1B高约10倍的杀肿瘤作用,并且在U87MG、C33A和A549细胞系中,Ad-ΔE1B-RLX显示比Ad-ΔE1B高约5倍的杀肿瘤作用。根据结果,可理解松弛素的表达没有减弱腺病毒的复制能力并且有助于显著提高腺病毒的杀肿瘤作用。
表达松弛素的溶瘤腺病毒的空斑形成
为了显现松弛素表达对致细胞病变能力和病毒扩散到周围细胞的作用,比较了含有琼脂糖的固体培养基中的空斑形成。用MOI为3的Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染Hep3B细胞然后分析空斑形成。如图6所示,对用Ad-ΔE1B-RLX感染的Hep3B细胞形成空斑的时间短于用Ad-ΔE1B感染的细胞。此外,在用Ad-ΔE1B-RLX感染的Hep3B细胞中形成的空斑的大小大于用Ad-ΔE1B感染的Hep3B细胞。更具体而言,用Ad-ΔE1B,感染后16天观察到空斑,而对于Ad-ΔE1B-RLX感染后早到4天就形成空斑。这些结果表明,由于增强溶瘤活性和病毒向周围细胞扩散,表达松弛素的腺病毒引起较短时间内形成大很多的空斑。由表达松弛素的腺病毒诱导的凋亡
显示复制缺陷型的腺病毒dl-LacZ-RLX诱导从培养板底部脱落的细胞的死亡。因此,我们检验松弛素的表达是否是致细胞病变作用的原因。首先,为了确定松弛素是否诱导凋亡,在PI染色后进行了流式细胞术测定以分析由于凋亡含有随机片段DNA的subG1细胞群的增加率。
将人肿瘤细胞系代表用Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒感染 并在感染后48~96小时收获用于测定subG1细胞群的增加(图7)。CPT-11用作诱导凋亡的正对照。对于A549细胞,Ad-ΔE1B诱导约3.11%的subG1细胞群而Ad-ΔE1B-RLX引起subG1细胞群显著增加22.90%。在其它细胞系(U343、U87MG、C33A和Hep3B)中也观察到此种增加的subG1细胞群。
此外,为了准确检验松弛素表达对细胞杀伤效力的作用,通过膜连蛋白-V和PI双重染色评估通过Ad-ΔE1B-RLX诱导的凋亡的进程。膜连蛋白-V作为凋亡的早期标记用于检测磷脂酰丝氨酸(PS)到外膜小叶的易位,和PI作为凋亡的晚期标记用于通过与核染色质结合鉴定坏死。因此,膜连蛋白-V-/PI-膜连蛋白-V+/PI-和PI+分别表示健康、凋亡和坏死细胞。
在CPT-处理的C33A细胞中,57.10%(膜连蛋白-V+/PI-)的细胞是凋亡的,而用Ad-ΔE1B或Ad-ΔE1B-RLX感染的细胞分别显示21.99%和33.03%的凋亡率,表明与Ad-ΔE1B相比,Ad-ΔE1B-RLX腺病毒诱导更强的凋亡率(图8)。对于包括U343、U87MG、Hep3B和A549的其它细胞系,表达松弛素的腺病毒较Ad-ΔE1B腺病毒显示更高的凋亡率。此外,说明Ad-ΔE1B-RLX反映全部细胞死亡的凋亡和坏死(膜连蛋白-V+/PI-和PI+)的总量比Ad-ΔE1B高得多。
总而言之,这些结果促使我们推论Ad-ΔE1B-RLX腺病毒引起比Ad-ΔE1B更高的凋亡率,从而Ad-ΔE1B-RLX比Ad-ΔE1B更经常发生细胞死亡。
进行了TUNNEL试验以鉴定作为早期凋亡特征的DNA片段。图9中显示几乎所有用CPT处理作为正对照的细胞染色为深棕色,表明发生了活跃的凋亡。32.5±12.5%的感染Ad-ΔE1B的U343细胞显示浅棕色和69.7±5.40%的感染Ad-ΔE1B-RLX的细胞显示深棕色,表明 Ad-ΔE1B-RLX比Ad-ΔE1B诱导凋亡的效力高(表1)。在其它肿瘤细胞系中也发现此种增加的凋亡。

表达松弛素的溶瘤腺病毒体内抗肿瘤作用的评估
为了研究表达松弛素的Ad-ΔE1B-RLX的体内抗肿瘤作用,隔日3次用5×107~1×108PFU的Ad-ΔE1B或Ad-ΔE1B-RLX感染裸鼠形成肿瘤异种移植物并且观察肿瘤的生长模式。对于人脑肿瘤U87MG,负对照PBS引起肿瘤的显著生长至1089±167.22mm3,而Ad-ΔE1B和Ad-ΔE1B-RLX分别引起肿瘤生长的显著抑制至115.70±119.60mm3和55.63±28.42mm3(图10)。
换言之,用Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒处理的肿瘤显示比用PBS处理的肿瘤高10~30倍的抗肿瘤作用。处理后25天后,用PBS处理的9只小鼠全部死亡。感染后33天后,Ad-ΔE1B和Ad-ΔE1B-RLX腺病毒引起肿瘤的体积分别为399.68±96.95mm3和64.51±36.73mm3,推论表达松弛素的腺病毒比Ad-ΔE1B具有更强的抗肿瘤效力。令人惊奇地,在病毒感染后19天,Ad-ΔE1B-RLX腺病毒完全根除7只小鼠中的2只的肿瘤并且在感染后41天去除5只小鼠的肿瘤。并且,甚至在感染后60天没有观察到肿瘤的再生长。
为了检验Ad-ΔE1B-RLX的这种优异的抗肿瘤作用是否在其它人肿瘤细胞中也是真实的,对C33A、A549、Hep3B和U343异体移植物进行了抗肿瘤作用分析。如图10所示,施用肿瘤特异性溶瘤Ad-ΔE1B或Ad-ΔE1B-RLX腺病毒的组比用PBS处理的组显示更显著的抗肿瘤作用。用表达松弛素的腺病毒处理的肿瘤的肿瘤块的体积比用Ad-ΔE1B处理小得多。这些结果表明松弛素的表达显著提高抗肿瘤作用。尤其是,处理后32天,与用Ad-ΔE1B和Ad-ΔE1B-RLX处理同期分别达到平均肿瘤体积917±354mm3和77±27mm3相比,用PBS对照处理的具有C33A的小鼠显示平均肿瘤体积为2252±1392mm3。在消退方面,处理后45天,与用Ad-ΔE1B-RLX处理的小鼠中看到的50%完全消退相比,25%的小鼠显示用Ad-ΔE1B处理的肿瘤完全消退。
检验了表达松弛素的腺病毒处理的荷瘤小鼠的存活率(图11)。对于C33A荷瘤小鼠,处理开始后80天,用Ad-ΔE1B-RLX处理的动物100%仍有活力,而同期用Ad-ΔE1B处理的动物只有50%有活力。此外,甚至在处理后6周,50%的用Ad-ΔE1B-RLX处理的动物仍完全无肿瘤。类似地,在检验的所有其它异种移植物模型(U343、U87MG、Hep3B和A549)中获得了Ad-ΔE1B-RLX诱导的存活益处。相对于彼此,在所有检测的异种移植物模型中,用Ad-ΔE1B-RLX处理的荷瘤小鼠比那些用Ad-ΔE1B处理的荷瘤小鼠存活长得多。在研究过程中,没有观察到如腹泻、体重减轻或恶病质的系统毒性。这些结果表明Ad-ΔE1B-RLX可赋予显著的存活益处和体内肿瘤减少。
由表达松弛素的增殖型腺病毒诱导的肿瘤特征的变化
3次用Ad-ΔE1B或Ad-ΔE1B-RLX感染在裸鼠腹部形成的人子宫颈肿瘤细胞系C33A。注射3天后,取出肿瘤组织并用苏木精和伊红染色用于组织学鉴定(图12)。Ad-ΔE1B-RLX处理的肿瘤中的坏死灶主 要在肿瘤块的外周发现,而Ad-ΔE1B处理的肿瘤的坏死灶几乎不能检测到,如果存在,在肿瘤块的中央发现。
然后通过使用腺病毒六联体蛋白特异的抗体的免疫组织化学证实肿瘤块内病毒的持续性和分布。如图12所示,主要在遭遇坏死的肿瘤的外周检测到Ad-ΔE1B-RLX腺病毒。TUNNEL试验显示在与坏死相同的区域活跃地发生凋亡。相反,如果可检测到,Ad-ΔE1B在肿瘤中央诱导坏死。
总而言之,可认识到Ad-ΔE1B-RLX在腺病毒注射位点活跃地复制,有助于诱导凋亡和坏死。
采用Masson三色染色的肿瘤块中胶原分布研究
3次用Ad-ΔE1B或Ad-ΔE1B-RLX感染裸鼠腹部形成的人脑肿瘤细胞系U343,注射3天后,取出肿瘤组织并用Masson三色染色以分析细胞外基质的主要组分胶原的分布(染成蓝色)。用PBS或Ad-ΔE1B处理的肿瘤由高含量的胶原(蓝色染色)组成。显著地对比,用Ad-ΔE1B-RLX处理的肿瘤显示无胶原,表明松弛素的表达显著降低肿瘤块内胶原含量。有趣的是,用Ad-ΔE1B-RLX处理的肿瘤被结缔组织包裹,并且只在肿瘤和正常组织之间的边界发现蓝色染色(图13)。
通过表达松弛素的溶瘤腺病毒抑制肿瘤转移
已知抑制松弛素诱导可能增强转移能力的MMP的表达。为了证实这个假说,我们用自发性肿瘤转移模型评估了表达松弛素的溶瘤腺病毒对肿瘤转移的作用,其中,在C57BL/6小鼠的足垫将B16BL16黑色素瘤细胞皮下植入以形成局部原发性肿瘤。一旦肿瘤体积达到100~200mm3,将动物隔日3次用PBS、Ad-ΔE1B或Ad-ΔE1B-RLX处理。第5天,在温和麻醉下通过切除膝盖以下手术去除原发性肿瘤。原发性肿瘤除去后第20天,测量小鼠肺中转移肿瘤灶的重量以评估向远缘 器官的转移。如图14所示,与PBS-处理对照组(268±0.27mg)相比,来自用Ad-ΔE1B或Ad-ΔE1B-RLX处理的小鼠的平均相关肿瘤负荷分别为48±0.05mg和10±0.01mg。此外,在6只处理小鼠的4只中,Ad-ΔE1B或Ad-ΔE1B-RLX均完全抑制转移性病灶的形成。这些数据表明,在原发性肿瘤部位肿瘤内注射表达松弛素的溶瘤腺病毒可以极大地防止并且不增强远缘部位转移性病灶的形成。
表达松弛素的腺病毒进入瘢痕疙瘩的穿透性和转导
以上描述的数据(尤其是,来自Masson’s三色染色的肿瘤块的胶原分布的数据)清楚地表明松弛素的表达显著降低肿瘤块中的胶原含量。瘢痕疙瘩是由细胞外基质广泛形成引起的一种病症。为了评价表达松弛素的腺病毒对瘢痕疙瘩的治疗效力,将从瘢痕疙瘩患者获得的原代瘢痕疙瘩细胞系用MOI为10或50的dl-LacZ或dl-LacZ-RLX腺病毒感染并在2天后进行X-gal染色。结果,用dl-LacZ-RLX感染比用dl-LacZ感染显示更强的LacZ表达,表明松弛素的表达负责进入瘢痕疙瘩细胞的转导效率提高(图15)。
此外,我们用瘢痕疙瘩细胞球形体检验表达松弛素的腺病毒的组织穿透性。为了证实表达松弛素的腺病毒进入瘢痕疙瘩组织的提高的转导效率,将用来自瘢痕疙瘩患者的原代瘢痕疙瘩细胞制备的瘢痕疙瘩细胞球形体用1×107PFU的dl-LacZ或dl-LacZ-RLX腺病毒感染并进行X-gal染色用于显微镜观察。在瘢痕细胞球形体的表面用dl-LacZ-RLX比用dl-LacZ显示更强的染色(图15)。
为了检验用瘢痕疙瘩细胞球形体显示的dl-LacZ-RLX的增强的传导效率是否在来自患者的瘢痕疙瘩组织中也能重复,将来自患者的瘢痕组织用1×108PFU的dl-LacZ或dl-LacZ-RLX感染并进行X-gal染色用于显微镜观察(图15)。dl-LacZ处理的瘢痕组织显示弱LacZ 表达,而dl-LacZ-RLX处理的瘢痕疙瘩组织为强烈的X-gal染色。此外,为了评价腺病毒进入瘢痕疙瘩组织的穿透性,将感染的组织包埋在O.C.T.化合物中并快速冷冻。用dl-LacZ感染的瘢痕疙瘩组织的内部与它们的表面为非常弱的染色。极大的相反,对于用dl-LacZ-RLX感染的组织,LacZ表达的分布广泛得多并在不局限于在注射部位的整个球形体观察到(图15)。
这些数据清楚地显示诱导细胞外基质破坏的表达松弛素的腺病毒的转导效率在瘢痕疙瘩组织中显著增强,表明表达松弛素的腺病毒是治疗瘢痕疙瘩病症的有前途的治疗剂。
工业适用性
本发明提供一种新的基因送递系统和包括松弛素编码序列的重组腺病毒、一种使用所述基因送递系统的基因送递方法、一种包括所述重组腺病毒的抗肿瘤药物组合物、一种特征为改善的组织穿透效力的药物组合物和一种用于治疗与过量细胞外基质累积相关的疾病或病症的药物组合物。根据本发明,松弛素负责改善传导效力和凋亡能力以显著增强肿瘤细胞杀伤能力。
已描述本发明的优选实施方案,应理解落入本发明的精神中的其变化和修饰对于本领域技术人员可为显而易见的,并且通过所附权利要求书及其等同物确定本发明的范围。
Claims (3)
1.编码松弛素的核苷酸序列在制备用于增强目的核苷酸序列进入细胞的转导效率的基因送递系统的用途,其中,所述基因送递系统包括待投送到细胞中的目的核苷酸序列和编码松弛素的核苷酸序列,其中,所述基因送递系统为重组腺病毒,所述重组腺病毒包括缺失的E3区并将编码松弛素的核苷酸序列插入到所述重组腺病毒的缺失的E3区。
2.根据权利要求1的编码松弛素的核苷酸序列的用途,其中所述细胞为通过细胞外基质互相连接的细胞构成的组织中的细胞。
3.根据权利要求2的编码松弛素的核苷酸序列的用途,其中所述组织为肿瘤组织。
