Abstract
本发明涉及从多核苷酸和多糖生产缀合物的方法,其包括下述步骤:a)提供多糖或其衍生物的醛糖酸;b)使该醛糖酸与醇衍生物反应,优选地与醇的碳酸酯衍生物反应,形成醛糖酸酯,优选地形成活化的醛糖酸酯;和c)使该醛糖酸酯与多核苷酸反应,其中所述多核苷酸表现出功能氨基,其中所述步骤b)中醛糖酸和醇衍生物的反应在无水的非质子极性溶剂中进行。
Description
本发明涉及从多核苷酸和多糖生产缀合物(conjugate)的方法,和可根据这类方法得到的缀合物。
近年来,随着来自生物技术研究的药物蛋白的增加,药物活性成分(尤其是蛋白)与聚乙二醇衍生物的缀合(″PEG化″)或与多糖例如右旋糖酐或尤其是羟乙基淀粉的缀合(″HES化″)已经变得重要。
药物活性的化合物(例如,蛋白)的PEG化或HES化的作用主要在于,通过将蛋白偶联到上述聚合物例如聚乙二醇(PEG)或羟乙基淀粉(HES)上,可以特异性地延长它们对于开发完全的药物潜能而言太低的短生物半衰期。但是,通过偶联,也可以正向地影响蛋白的抗原性质。在其它药物活性成分的情况下,通过偶联,可以显著增加水溶性。药物活性成分的HES化的实例记载在例如国际专利申请WO 02/080979 A2或国际专利申请WO 03/000738 A2中。
在高亲和力结合寡核苷酸(例如称作适体的D-寡核苷酸或称作镜像体(Spiegelmer)的L-寡核苷酸)的生物靶分子领域的最新进展,也使用与聚合物例如聚乙二醇缀合的可能性(B.Wlotzka等,PNAS 13,vol.99(2002)第8898-8902页),以有利的方式改变药物动力学特征和生物利用度。
HES是蜡质玉米淀粉中占95%以上的葡萄糖聚合物支链淀粉的羟乙基化衍生物。支链淀粉由葡萄糖单元组成,所述葡萄糖单元以α-1,4-糖苷键存在,且表现出α-1,6-糖苷分支。HES表现出有利的流变性质,目前在临床上用作容量代用品和用于血稀释疗法(Sommermeyer等,Krankenhauspharmazie,vol.8(1987)第271-278页和Weidler等,Arzneimittelforschung/Drug Res.,41(1991)第494-498页)。
在DE 196 287 05和DE 101 29 369中,为血红蛋白或两性霉素B特别描述了如何通过羟乙基淀粉的对应醛糖酸内酯与血红蛋白或两性霉素B的游离氨基,在无水二甲基亚砜(DMSO)中进行与羟乙基淀粉的偶联的方法。
因为,尤其在蛋白的情况下,经常不可能与无水的非质子溶剂一起工作,原因可能是溶解度,也可能是蛋白的变性,HES在含水介质中的偶联方法也记载在文献中。因而,例如国际专利申请PCT/EP02/02928公开了借助于水溶性的碳化二亚胺EDC(1-乙基-3-(3-二甲基氨基丙基)-碳化二亚胺),在链的还原端选择性地氧化成醛糖酸的羟乙基淀粉的偶联。但是,碳化二亚胺的应用经常伴有缺点,因为碳化二亚胺非常容易造成蛋白的分子间或分子内的交联反应,作为副反应。
本发明是基于下述问题,即提供从多核苷酸和多糖生产缀合物的方法。
根据本发明,通过从多核苷酸和多糖生产缀合物的方法,在第一方面解决了该问题,该方法包括下述步骤:
a)提供多糖或其衍生物的醛糖酸;
b)使该醛糖酸与醇衍生物反应,优选地与醇的碳酸酯衍生物反应,形成醛糖酸酯,优选地形成活化的醛糖酸酯;和
c)使该醛糖酸酯与多核苷酸反应,其中所述多核苷酸表现出功能氨基,
其特征在于,
步骤b)中醛糖酸和醇衍生物的反应在无水的非质子极性溶剂中进行。
在一个实施方案中,所述溶剂选自二甲基亚砜,二甲基甲酰胺和二甲基乙酰胺。
在一个实施方案中,纯化醛糖酸酯,然后用于步骤c)。
在一个替代实施方案中,来自步骤b)的含醛糖酸酯的反应料直接用于步骤c)。
在一个实施方案中,在7-9、优选7.5-9和更优选8.0-8.8的pH范围,进行步骤c)。

在第二个方面,通过可根据本发明的第一方面的方法得到的多糖与多核苷酸的缀合物,按本发明解决了该问题。
本发明是基于惊人的认识,即从羟乙基淀粉醛糖酸和其它多糖(例如,蜡质玉米淀粉降解级分)的醛糖酸,在无水的非质子极性溶剂(例如,二甲基乙酰胺(DMA),二甲基亚砜(DMSO)或二甲基甲酰胺(DMF))中,与醇尤其是与醇的碳酸酯(从而二氧化碳与醇的二酯,例如,N-羟基-琥珀酰亚胺),可以生成对应的醛糖酸酯,其可以在含水介质中有利地与来自多核苷酸的亲核氨基反应成为更稳定的酰胺。作为副反应,发生醛糖酸酯与水的皂化,产生游离的醛糖酸和游离的醇。
令人惊奇地,没有由此活化脱水葡萄糖单元的羟基,但是取而代之,特异性地活化了醛糖酸的羧基,条件是反应物的摩尔比设定在大约1∶1左右。
在这方面,本发明背离现有技术迄今为止的教导,或者是基于下述知识,即现有技术中描述的不同方法不适用于有效率地生产多核苷酸和多糖的缀合物。
因而,本发明人还意外地发现,L-5′-氨基-官能化的寡核苷酸不能通过碳化二亚胺(EDC)-介导的与5′氨基形成酰胺键而与HES醛糖酸反应,不论可能的反应参数和反应物比率的变化如何。
实际上,已知含有磷酸酯和磷酸基团的化合物会增加碳化二亚胺的损失,经常是非常显著的,在该情况下,甚至极大过量的EDC也不能产生可测量的反应产物(S.S.Wong,Chemistry of ProteinConjugation and Cross-Linking,CRC-Press,Boca Raton,London,New York,Washington D.C.(1993)第199页)。
而且,已知EDC可以在含水介质中用于将含有氨基官能团的分子偶联到寡核苷酸的末端磷酸基团,形成氨基磷酸酯键。在进行的反应条件下,内部的磷酸基团不反应。这样,可以特异性地修饰尤其是5′磷酸基团(Bioconjugate Techniques,Greg T.Hermanson,AcademicPress,San Diego,New York,Boston,London,Sydney,Tokyo,Toronto(1996)第52页)。
而且,还令人惊奇地发现,文献中描述的羟乙基淀粉衍生物的其它确立的偶联方法,也不能成功地用于5-氨基-官能化的L-寡核苷酸。因而,普通方法是,从酸形成反应性的酸imidazolide,以生产酸酰胺,其含义是作为中间阶段形成酰胺键,然后,利用该有活性的酰化剂,进行胺的反应,产生对应的酰胺,并释放咪唑。
在HES醛糖酸的情况下,反应性的HES imidazolide的生产是成功的。但是,在检查的所有pH值和反应物比率,在水溶液中进行反应的过程中,这分解为咪唑和HES酸,不涉及与实际上更亲核的5-氨基-官能化的多核苷酸的偶联。
作为另一种可能性,通过HES酸的有活性的羟基琥珀酰亚胺酯,进行偶联,所述酯之前在无水介质中根据文献说明生产,通过EDC活化或通过HES内酯与羟基琥珀酰亚胺在无水介质中的反应。但是,两种方法都不成功。
尽管非常长的反应时间,还原胺化意义的HES通过唯一的还原性端基与氨基-官能化的多核苷酸的反应也是不成功的。
根据本发明的从多核苷酸和多糖生产缀合物的反应方案显示在图1,其中图1A显示了多糖醛糖酸的醛糖酸基团的结构,图1B解释了反应进程。图2中的反应方程式是实施例4-14的主题,它们总结了从多核苷酸和多糖生产缀合物的不成功的尝试。
尽管根据本发明的方法原则上不限于某些多糖,羟乙基淀粉是特别优选的多糖。但是,使用其它淀粉衍生物也在本发明的范围内,例如羟基脯氨酰基淀粉。同样地,在本发明的范围内,可以使用德国专利申请102 17 994中描述的超分支的淀粉级分,尤其是具有大于10mol%、优选地大于10mol%且小于16mol%的分支程度的超分支的淀粉级分。
HES主要通过重均平均分子量Mw,数均平均分子量Mn,分子量分布和取代度来表征。在醚键中的羟乙基取代由此可能是在脱水葡萄糖单元的碳原子2,3和6上。取代样品因而描述为C2与C6取代之比(C2/C6比率)。取代度因而可以描述为DS(″取代度″的英文),它指所有葡萄糖单元中取代的葡萄糖分子的含量,或者描述为MS(″摩尔取代″的英文),它指每个葡萄糖单元的羟乙基平均数。
在科学文献中,也如在本文中,以kDa为单位的分子量Mw连同取代度MS是对羟乙基淀粉给出的缩写。因而HES 10/0.4指分子量Mw为10,000且取代度MS为0.4的羟乙基淀粉。
通过使多糖或它的衍生物的醛糖酸在无水的非质子溶剂例如二甲基甲酰胺(DMF)、二甲基亚砜(DMSO)或二甲基乙酰胺(DMA)中与醇组分的碳酸酯反应,生产根据本发明使用的醛糖酸酯。本文所述的醛糖酸是现有技术已知的,且可以根据例如德国专利申请DE 196 28 705的公开内容生产。
在醛糖酸与醇衍生物、尤其是分别使用的醇的碳酸酯的反应中,摩尔比是大约0.9-1.1,优选地大约1.0,因为使用过量的碳酸酯,如醇衍生物提供的,会选择性地活化多糖的OH基团,而当碳酸酯较少时,过度的酸官能团不反应。
在本发明的范围内特别优选的醇是N-羟基-琥珀酰亚胺,磺化的N-羟基-琥珀酰亚胺,苯酚衍生物和N-羟基-苯并三唑。合适的苯酚衍生物包含,尤其是,氯化的、氟化的或硝化的化合物,其中它们可以被活化一次或几次,尤其是被前述的亲电子基团活化。相应地,在本发明的范围内可使用单-或多氯化的苯酚,单-或多氟化的苯酚或单-或多硝化的苯酚。
使用无水的乙醇、异丙醇或丙酮,可以从在DMF中的溶液中沉淀出根据本发明使用的醛糖酸酯,并通过重复该方法数次将其纯化或富集。然后,可以将这样的醛糖酸酯实质上分离地用于与多糖偶联。但是,也可以直接重复使用反应产物在惰性非极性溶剂中的溶液,无需分离用于与多糖偶联的有活性的醛糖酸酯。
原则上,在本发明的范围内,任何类型的多核苷酸都能与多糖缀合。因而,可以从L-核苷或D-核苷或其混合物生产多核苷酸,其中它们可以各自地或一起表现出其它修饰,例如增加在生物系统中的稳定性的修饰。这类修饰是例如在核苷酸或核苷的糖组分的2′位置的氟化。因此,形成多核苷酸的核苷酸的糖组分的至少一部分可以表现出除核糖或脱氧核糖以外的糖,这也在本发明的范围内。这类糖可以是,例如,其它的戊糖,例如,阿拉伯糖,但也可以是己糖或丁糖。这类糖还可以含有氮原子或硫原子,例如在氮杂-或硫代糖中,和/或多核苷酸的糖内容(sugar content)可以至少部分地被吗啉代环替代。而且,多核苷酸可至少部分地开发成锁定的(locked)核酸(LNA)或肽核酸(PNA)。形成多核苷酸的骨架的分子组分的OH基团可以被合适的NH2,SH,醛,羧酸,磷酸,碘,溴或氯基团化学修饰。
此外,在本发明的范围内的是,多核苷酸是核糖核酸或脱氧核糖核酸或其组合,即个别核苷酸或一组核苷酸作为RNA存在,而形成核酸的其它核苷酸作为DNA存在,和反过来的情况。术语L-核酸在本文中与术语L-寡核苷酸或L-多核苷酸同义地使用,指尤其是L-脱氧核糖核酸和L-核糖核酸和其组合,即个别核苷酸或一组核苷酸作为RNA存在,而形成核酸的其它核苷酸作为DNA存在,反之亦然。因而也可以预期,替代脱氧核糖或核糖,其它糖形成核苷酸的糖组分。而且包含在2′位置具有其它修饰例如NH2,OMe,OEt,O烷基,NH烷基的核苷酸的应用,和天然的或非天然的核碱基例如异胞苷和异鸟苷的应用。因而,也在本发明的范围内的是,L-核酸表现出所谓的无碱基位置,即其中不存在核碱基的核苷酸。这类无碱基位置可以排列在L-核酸的核苷酸序列内,也可排列在一端或两端,即5′和/或3′端。
而且,在本发明的范围内的是,多核苷酸以单链存在,但是,也在本发明的范围内的是,它以双链存在。典型地,根据本发明使用的多核苷酸是单链L-核酸,但是,作为它的一级序列的结果,它可以形成确定的二级结构和三级结构。在二级结构中,由于L-核酸多重性,也会存在双链部分。
本文所述的缀合的核酸优选地是所谓的镜像体。如在开始已经提及的,镜像体是功能L-核酸或L-多核苷酸,即这类核酸,它们结合靶分子或其部分,并且是使核酸文库、尤其是统计学核酸文库接触靶分子的结果。
组合DNA文库是为用于开发功能核酸的选择方法首先生成的。通常,这是DNA寡核苷酸的合成,其中央含有一系列10-100个随机化的核苷酸,后者在5′和3′末端侧接2个引物结合区域。这类组合文库的产生记载在例如,Conrad,R.C.,Giver,L.,Tian,Y.和Ellington,A.D.,1996,Methods Enzymol.,vol.267,336-367。通过聚合酶链式反应,这样的化学合成的单链DNA文库可以转化成双链文库,其自身实际上可以用于选择。但是,通常,可用合适的方法进行各链的分离,以便可以再次得到单独链文库,如果是DNA选择,则其可以用于体外选择方法(Bock,L.C.,Griffin,L.C.,Latham,J.A.,Vermaas,E.H.和Toole,J.J.,1992,Nature,vol.355,564-566)。但是,也可能在体外选择中直接包括化学合成的DNA文库。另外,原则上,如果提前导入了T7启动子,从而通过合适的DNA-依赖性的聚合酶,例如T7 RNA聚合酶,RNA文库可以从双链DNA生成。使用所述的方法,可能生产1015和更多的DNA或RNA分子的文库。来自该文库的每个分子具有不同的序列,从而具有不同的三维结构。
通过体外选择方法,然后可能经过几个循环的选择和扩增和任选的突变,从所述文库分离一个或几个DNA分子,其表现出显著的对给定靶物的结合性质。靶物可以是,例如,病毒,蛋白,肽,核酸,小分子例如代谢产物,药物活性成分或其代谢产物或其它化学的、生化的或生物的组分,例如记载在例如Gold,L.,Polisky,B.,Uhlenbeck,O.和Yarus,1995,Annu.Rev.Biochem.vol.64.763-797和Lorsch,J.R.和Szostak,J.W.,1996,CombinatorialLibraries,Synthesis,Screening and application potential,ed.Riccardo Cortese,Walter de Gruyter,Berlin。进行该方法,以便从原始使用的文库分离结合DNA或RNA分子,并在选择步骤后,通过聚合酶链式反应进行扩增。在RNA选择中,反转录应当连接到聚合酶链式反应的扩增步骤之前。然后,第一轮选择后富集的文库可以用于更新一轮的选择,以便在第一轮选择中富集的分子具有通过选择和扩增再次遗留下来的机会,并与更多的子代分子一起进入另一轮选择。同时,聚合酶链式反应步骤打开了在扩增过程中导入新突变的可能性,例如通过盐浓度的变化。足够数目的选择和扩增轮数后,结合分子已经遗留下来。从而生产了富集的库,通过克隆可以分离其中的代表物,然后通过DNA序列测定的普通方法确定它的一级结构。然后,检查得到的序列对靶物的结合性质。因此,生产这类适体的方法称作SELEX方法,且记载在例如EP 0 533 838,其内容在本文引作参考。
通过将一级序列缩短至它们的基本结合域,可以缩短最好的结合分子,并通过化学或酶合成再次呈现。
特定形式的可在这样程度上生产的适体是所谓的镜像体,其基本特征在于,它们至少部分地、优选完全地由非天然L-核苷酸构成。生产这类镜像体的方法记载在PCT/EP97/04726,其内容在本文引作参考。其中描述的方法的独特之处在于对映体核酸分子的生产,即能结合天然靶物(即以这类靶物结构的天然形式或构型存在)的L-核酸分子的生产。上述的体外选择方法首先用于选择针对对映体的结合核酸或序列,所述对映体即天然存在的靶物的非天然存在的结构,例如在靶物分子是蛋白的情况下,针对D蛋白。检测这样得到的结合分子(D-DNA,D-RNA或相应的D-衍生物)的序列,然后用镜像核苷酸构件(L-核苷酸或L-核苷酸衍生物)合成相同的序列。这样得到的镜像对映体核酸(L-DNA,L-RNA或相应的L-衍生物),即所谓的镜像体,由于对称的原因,具有镜像三级结构,并因而具有对以天然形式或构型存在的靶物的结合性质。
如在本文所述的选择和缩短方法中得到的多核苷酸,尤其是功能核酸例如适体或镜像体,具有大约300Da-50,000Da的分子量。优选地,它们表现出4,000Da-25,000Da、更优选地7,000-16,000Da的分子量。
上述的靶分子,也称作靶物,可以是分子或结构,因而,例如是病毒,类病毒,细菌,细胞表面,细胞器,蛋白,肽,核酸,小分子例如代谢产物,药物活性成分或其代谢产物或其它化学的、生化的或生物的组分。
根据本发明的方法,多核苷酸优选地在多核苷酸的磷酸基团上呈现亲核基团,醛糖酸酯与其反应形成缀合物。因此在本发明的范围内特别优选地,该亲核基团是功能氨基,优选伯氨基(NH2基团)。因而,在本发明的范围内,与醛糖酸酯反应的多核苷酸含有功能仲氨基,亚氨基基团。
但是,特别优选地,在本发明的范围内,亲核基团是伯氨基,其优选地存在结合在多核苷酸的磷酸基团上。氨基优选地存在于5′或3′端磷酸基团,即多核苷酸的末端磷酸基团。在一个实施方案中,氨基可以从而直接地结合到磷酸基团,或通过接头结合到磷酸基团。这类接头是现有技术已知的。优选的接头是烷基基团,其具有1-8、优选2-6个C原子的长度。更具体地,当使用N-羟基-琥珀酰亚胺的醛糖酸酯时,也存在于多核苷酸中的亲核基团(例如核酸中的嘌呤或嘧啶碱基)不反应。
在本发明的范围内,使用寡核苷酸替代多核苷酸。在一个实施方案中,如本文使用的多核苷酸是寡核苷酸。

实施例1:从镜像体和羟乙基淀粉生产缀合物
使用的HES化物
使用具有分子参数Mw 11092 D,MS 0.4和C2/C6>8的HES 10/0.4,其在还原链端被氧化成羧酸。对HES酸的生产的描述记载在例如德国专利申请DE 196 28 705中。
NHS酯的生产
如下生产HES化物的N-羟基-琥珀酰亚胺酯:
将0.2g(0.05mMol)无水的HES酸10/0.4溶解于1ml无水的二甲基甲酰胺中,并与等摩尔量的N,N′-二琥珀酰亚胺基碳酸酯(12.8mg)在室温反应1.5小时。
镜像体HES化物的生产
将5mg(相当于1.3μmol)根据_Seq.ID.no.1的5′-氨基己基官能化的RNA-镜像体溶解于0.7ml 0.3 M二碳酸酯溶液(pH 8.4)。将如上所述生产的活性酯直接加入该溶液,并在室温反应2小时。
RNA-镜像体具有下述序列:
5′-氨基己基-UGAGUGACUGAC-3′(SEQ.ID.NO.1)
对以这种方式生产的缀合物的分析
通过低压GPC,检测了缀合物。其中使用的分析条件如下,分析结果显示在图3:
柱: Superose 12 HR 10/30,300mm x 10 mm i.d.
(Pharmacia,art.no.17-0538-01)
流动溶剂:磷酸盐缓冲液pH 7.0
(27.38mM Na2HPO4,12.62mM NaH2PO4,
0.2M NaCl,0.005%NaN3在Milli-Q水中)

检测: UV 280nm
运行时间:70min
注射体积:20μl原始装料
反应料产生了62%(垂直于色谱图)或77%(拖尾峰评价)的产率。
用具有下述分子参数的另一种形式的羟乙基淀粉(50/0.7)进行上述反应:Mw:54110D,MS:0.7和C2/C 6:~5。
用除此之外其余等同的反应方法,得到53%的产率。反应产物的对应的GPC色谱图显示在图4中。
实施例2:增加缀合物的产率
基于实施例1所述的方法,将逐份添加的活化的醛糖酸酯与实验RNA-镜像体的比率增加到2∶1或3∶1。在比率倍增的情况下,得到了超过95%的产率,用三倍过量的活化的醛糖酸酯,得到了实际上定量的产率(>98-99%)。
实施例3:对比结合Ghrelin的HES化的和非-HES化的镜像体对Ghrelin-诱导的Ca释放的抑制
以5-7x104/孔的数量,将表达Ghrelin的人受体(GHS-Rla)的稳定地转染的CHO细胞(获自Euroscreen,Gosselies,Belgium)接种到透明底的黑色96-孔微量滴定板(Greiner)中,并在37℃和5%CO2在UltraCHO培养基(Cambrex)中培养过夜,该培养基另外含有100单位/ml青霉素,100μg/ml链霉素,400μg/ml遗传霉素和2.5μg/ml两性霉素B。
将非-HES化的和根据实施例1生产的5′-HES化形式的结合Ghrelin的镜像体(根据SEQ.ID no.2,内部参考SOT-B11),与人或大鼠Ghrelin(Bachem)一起在已经添加了5mM丙磺舒和20mMHEPES的UltraCHO培养基(CHO-U+)中,在RT或37℃,在0.2ml″隐蔽96-管″平板中温育15-60min。将这些刺激溶液制备成在CHO-U+中的十倍浓缩溶液。
SOT-B11的序列:5′-CGU GUG AGG CAA UAA AAC UUA AGU CCG AAGGUA ACC AAU CCU ACA CG-3′(seq.ID.no.2)
在添加Ca指示剂染料Fluo-4之前,分别用200μl CHO-U+洗涤细胞1x。然后添加50μl指示剂染料溶液(10μM Fluo-4(molecularprobes)),在CHO-U+中的0.08%Pluronic 127(molecular probes),并在37℃温育60min。然后,分别用180μl CHO-U+洗涤细胞3x。再每孔加入90μl CHO-U+。
在Fluostar Optima多检测读板器(BMG)中,在485nm的激发波长和520nm的发射波长,进行荧光信号的检测。
将刺激溶液加给细胞,以准确分析Ghrelin造成的Ca浓度变化的时间进程。连带测量96孔平板的垂直系列的孔,以平行测量几个样品。为此,首先以4秒的间隔记录3个测量值,以确定基线。然后,中断测量,从读板器取出平板,用多道吸管,将来自进行预温育的″隐蔽96-管″平板的10μl刺激溶液加入待测量系列的孔中。然后,再将平板插入机器,继续测量(共20次测量,间隔4秒)。
从得到的测量曲线,确定每个孔的最大荧光信号和刺激前的荧光信号之间的差异,并对Ghrelin的浓度作图,或者,在镜像体抑制Ca释放的实验中,针对镜像体的浓度作图。
为了显示HES化的镜像体的有效性,用5nM Ghrelin或已经与不同量的HES化的或非-HES化的镜像体一起预温育的Ghrelin刺激表达Ghrelin受体的细胞。将测量的荧光信号相对于在没有镜像体时得到的信号标准化。HES化的镜像体抑制Ghrelin-诱导的Ca++释放,其IC50为约6.5nM,而非-HES化的镜像体以约5nM的IC50抑制。结果显示在图5。
实施例4:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
在搅拌下,将0.25g HES 10/0.4醛糖酸(62.5μmol)溶解于10mL水中。在室温,将9.95mg(2.5μmol)根据SEQ.ID.no.1的RNA-镜像体加入溶液。然后,经2小时,在室温,在搅拌下,逐份添加溶解在1mL水中的50mg N-乙基-N′-(3-二甲基氨基丙基)-碳化二亚胺氢氯化物(261μmol)。通过加入盐酸或氢氧化钠溶液,使pH5保持恒定。当反应结束时,将反应物在室温再搅拌另外2小时。通过低压GPC检查反应料,使用的镜像体的反应转化率小于1%。
实施例5:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
经3小时,在室温和在搅拌下,将150mg EDC加入如实施例4所述的HES 10/0.4醛糖酸和根据seq.ID.no.1的RNA-镜像体的混合物中。通过分析低压GPC,没有检测到反应转化。
实施例6:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
在搅拌下,在加热下,将1.0g HES 10/0.4醛糖酸(240μmol)溶解于10mL水中。冷却至室温后,将10mg根据seq.ID.no.1的RNA-镜像体加入反应料。然后,在搅拌下,经2小时,在室温,逐份添加溶解在1mL水中的50mg EDC(260μmol),用盐酸或氢氧化钠溶液,将pH保持恒定在5。
另外2小时反应时间后,通过低压GPC分析反应料。没有检测到反应产物。
实施例7:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
重复根据实施例6的反应料,其中在本例中,经3小时添加100mgEDC。
当反应结束时,没有检测到反应产物。
实施例8:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
在4.0和6.0的pH值,重复实施例6和7。
通过低压GPC,在2种反应料中没有检测到反应产物。
实施例9:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
在4℃和37℃的反应温度,重复实施例4。
在两种情况下,都没有检测到反应产物。
实施例10:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
在室温,将303.7mg(2.6mmol)琥珀酰亚胺和0.502g HES10/0.4醛糖酸(0.125mmol)溶解于10mL无水的二甲基亚砜(DMSO)。
然后,添加50mg EDC(0.25mmol),并搅拌反应料过夜。
将5mg(相当于1.3μmol)根据seq.ID.no.1的RNA-镜像体溶解于10mL水,用氢氧化钠溶液将pH设定在8.5,或溶解于10mLpH 8.4的0.3 M碳酸氢盐缓冲液。
分别将5mL上述的二甲基亚砜溶液加入2种部分反应料中,通过添加氢氧化钠溶液,将第一种部分反应料的水溶液的pH保持恒定在pH 8.5。
在室温,搅拌反应料过夜。在2种部分反应料中,分析低压GPC没有产生任何反应产物。
实施例11:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
将300mg(2.6mmol)琥珀酰亚胺溶解于10mL无水的二甲基亚砜(DMSO),并在80℃添加0.5g(0.125mmol)干燥的HES 10/0.4醛糖酸过夜,形成对应的内酯。在70℃,反应料反应过夜。
然后,在室温,将溶液加入5mL 5mg根据seq.ID.no.1的RNA-镜像体在10mL pH 8.4的0.3M碳酸氢盐缓冲液中形成的溶液,并在室温搅拌4小时。通过分析低压GPC,没有发现反应产物。
实施例12:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
将5.0g HES 10/0.4醛糖酸(1.2mmol)溶解于30mL无水的二甲基甲酰胺(DMF)。将195mg(1.2mmol)羰基二咪唑(CDI)加入溶液,并在室温搅拌2小时。
将5mg根据seq.ID.no.1的RNA-镜像体溶解于5mL水。将10mL上述咪唑基-HES醛糖酸10/0.4溶液加入该溶液,并用氢氧化钠溶液将pH设定在7.5。在室温搅拌过夜后,通过低压GPC检查反应料中的反应产物。仅仅检测到微量的反应产物。
实施例13:使用根据现有技术的方法,从多核苷酸和HES醛糖酸生产缀合物
将5mg根据seq.ID.no.1的RNA-镜像体溶解于12.5mL pH 8.4的0.3M碳酸氢盐缓冲液。用冰水将反应料冷却到0℃,并与8.5mL实施例12所述的HES 10/0.4醛糖酸-咪唑基在DMF中的溶液混合。在0℃2小时和在室温另外2小时后,检查反应料中的反应产物。没有检测到产物。
实施例14:使用根据现有技术的方法,从多核苷酸和HES生产缀合物
在加热下,将1g HES 10/0.4(0.25mmol)溶解于5mL H2O。将10mg(相当于2.5μmol)根据seq.ID.no.1的RNA-镜像体加入冷却后的溶液,并用氢氧化钠溶液将pH设定在7.5。然后,添加200μl硼烷-吡啶复合物(Sigma-Aldrich),并在室温,在黑暗中搅拌反应料10天。然后,通过低压GPC,检查反应料中的任何反应产物。仅仅检测到基于使用的镜像体<3%的转化率。
可以基本上单独地和以任意的组合,以前面说明书、权利要求书和附图中公开的本发明的特征,以不同实施方案实施本发明。
Claims (16)
1.从多核苷酸和多糖生产缀合物的方法,其包括下述步骤:
a)提供多糖的醛糖酸,其中所述多糖选自右旋糖酐,羟乙基淀粉,羟基丙基淀粉和分支的淀粉级分;
b)使该醛糖酸与醇的碳酸酯反应,形成醛糖酸酯;和
c)使该醛糖酸酯与多核苷酸反应,其中所述多核苷酸具有伯或仲氨基,
其特征在于,
步骤b)中醛糖酸和醇的碳酸酯的反应在无水的非质子极性溶剂中进行,且醛糖酸与醇的碳酸酯的摩尔比是0.9-1.1,所述多核苷酸是适体或镜像体以及所述醇的碳酸酯是N,N′-二琥珀酰亚胺基碳酸酯。
2.根据权利要求1的方法,其特征在于,所述溶剂选自二甲基亚砜,二甲基甲酰胺和二甲基乙酰胺。
3.根据权利要求1的方法,其特征在于,纯化醛糖酸酯,然后用于步骤c)中。
4.根据权利要求1的方法,其特征在于,来自步骤b)的含醛糖酸酯的反应料直接用于步骤c)。
5.根据权利要求1的方法,其特征在于,在7-9的pH范围进行步骤c)。
6.根据权利要求5的方法,其特征在于,在8.4的pH进行步骤c)。
7.根据权利要求1的方法,其特征在于,所述多糖是羟乙基淀粉。
8.根据权利要求7的方法,其特征在于,所述羟乙基淀粉表现出3,000-100,000道尔顿的重均平均分子量。
9.根据权利要求7的方法,其特征在于,所述羟乙基淀粉表现出2,000-50,000道尔顿的数均平均分子量。
10.根据权利要求7的方法,其特征在于,所述羟乙基淀粉表现出1.05-1.20的重均分子量与数均平均分子量之比。
11.根据权利要求7的方法,其特征在于,所述羟乙基淀粉表现出0.1-0.8的摩尔取代。
12.根据权利要求7的方法,其特征在于,所述羟乙基淀粉表现出2-12的表示为C2/C6比率的取代样品。
13.根据权利要求1的方法,其特征在于,所述多核苷酸表现出300-50,000Da的分子量。
14.根据权利要求1的方法,其特征在于,所述伯或仲氨基结合到多核苷酸的末端磷酸。
15.根据权利要求14的方法,其特征在于,所述伯或仲氨基通过接头结合到磷酸基团上,其中所述的接头是具有1-8个C原子的长度的烷基基团。
16.根据权利要求1的方法,其特征在于,所述伯氨基是5-氨基己基。
