Abstract

Description
发明背景
本发明涉及到来自谷类(小麦族(Triticeae))的第4组主要致敏原的DNA序列的提供,本发明也包括具有降低过敏作用的片段、部分序列的新组合及含有点突变的突变体。重组的DNA分子和其衍生而来的多肽、片段、部分序列的新组合和变异体可以被用于花粉过敏性疾病的治疗。由重组方法制备的蛋白可以用于花粉过敏的体外和体内诊断。
I型过敏反应是全球性的,在工业国家中有多至20%的人口患有如过敏性鼻炎、结膜炎或支气管哮喘。这些过敏由从各种来源如植物花粉、螨类、猫和狗释放到空气中的致敏原(空气致敏原)引起,多至40%的I型过敏患者有对禾本花粉致敏原的特异性IgE反应,对谷类花粉致敏原尤甚(Freidhoff et al.,1986,J.Allergy Clin.Immunol.78,1190-2001),在谷类花粉致敏原中,又以黑麦致敏原为最重要。
触发I型过敏反应的物质为蛋白、糖蛋白或多肽,通过粘膜被摄取后,在已被致敏的个体体内这些致敏原与结合于肥大细胞表面的I,分子反应,如果两个IgE分子通过致敏原相互交叉连接,即会导致效应细胞释放介质(如组胺、前列腺素)和细胞因子,并引起相应的临床症状。
根据各种致敏原与过敏患者的IgE反应的相对频率,将致敏原区分为主要致敏原和次要致敏原。
禾本科(Poaceae)来源的各种花粉被划分为不同组,各组之间相互同源。
特别值得一提的是主要致敏原第4组中的分子相互间具有很强的交叉免疫反应,对鼠单克隆抗体和人IgE抗体均如此(Fahlbusch et1993 Clin.Exp.Allergy 23:51-60;Leduc-Brodard et 1996,J.AllergyClin.Immunol.98:1065-1072;Su et 1996,J.Allergy Clin.Immunol.97:210;et 1998,Clin.Exp.Allergy 28:799-807;Gavrovic-et 2000,10(6):361-367;Stumvoll et al.2002,Biol.Chem.383:1383-1396;Grote et2002,Chem.383:1441-1445;Andersson und Lidholm,2003,Arch.Allergy Immunol.130:87-107;Mari,Clin.Exp.Allergy,33(1):43-51)。
迄今为止,尚没有一个第4组主要致敏原的完整DNA序列被弄清楚。
迄今第4组致敏原中的鸭茅(Dactylus glomerata)致敏原只可能通过酶消化获得多肽并进行序列测定:DIYNYMEPYVSK(SEQ ID NO 13),VDPTDYFGNEQ(SEQ ID NO 14),ARTAWVDSGAQLGELSY(SEQ ID NO 15)和GVLFNIQYVNYWFAP(SEQ NO 16,Leduc-Brodard et J.AllergyClin.Immunol.98:1065-1072)。

对于黑麦草(Lolium perenne),作为第4组基本致敏原的具有以下序列的肽片段已有描述:FLEPVLGLIFPAGV(SEQ ID NO 28)和GLIEFPAGV(SEQ ID NO 29 Jaggi et 1989,Int.Arch.342-348)。
尚未发表的来自梯牧草(Phleum pratense)的PhI p 4序列(SEQ IDNO 11),作为第一个第4组致敏原序列,本专利申请的发明者已在国际申请WO 04/000881中对其进行了阐述。
迄今为止还不知道来自谷类(小麦族(Triceae))的第4组致敏原的任何序列。
因此,本发明的目的是要提供来自谷类的第4组致敏原的DNA序列,特别是来自黑麦(Secale cerale)的Sec c 4(SEQ ID NO 1、3)、来自大麦(Hordeum vulgare)的Hor v 4(SEQ ID NO 5)和来自小麦(Triticum aestivum)的Tri a 4(SEQ ID NO 7、9)以及相应的重组DNA分子,以这些重组DNA分子为基础,致敏原能以无修饰的或修饰过的蛋白形式表达进而得到,用于有意义的药物开发。PhI p 4序列(SEQID NO 11)是本发明的起始点。

发明描述
本发明目前第一次提供谷类花粉主要致敏原Sec c 4、Hor v 4和Tria 4的DNA序列,对应的序列号为SEQ ID NO1、3、5、7和9。本发明因此涉及到选自对应于SEQ ID NO1、3、5、7和9的核酸序列的DNA分子。
本发明进一步涉及到与本发明的DNA序列同源的序列和来源于其他禾本科(poaceae)的第4组致敏原的相应的DNA分子,这类植物有黑麦草(Lolium perenne)、鸭茅(Dactylis glomerata)、草地早熟禾(Poapratensis)、狗牙根(Cynodon dactylon)和绒毛草(Holcus Lanatus),由于存在同源性,它们能在严格条件下与本发明的DNA序列杂交或与本发明的致敏原发生交叉免疫反应。
本发明也包括具有降低过敏反应作用的片段、部分序列的新组合和含有点突变的突变体。
本发明因此进一步涉及到编码来自禾本科(poaoaceae)的第4组致敏原的免疫调节、T细胞反应片段的对应的部分序列、部分序列的组合,或含有取代、删除或添加的突变体。
随着对自然发生的致敏原DNA序列的了解,目前,已经有可能制备能够用于过敏性疾病的诊断和治疗的这些致敏原的重组蛋白(Scheiner and Kraft,1995,Allergy 50:384-391)。
经典的有效治疗过敏的方法为特异性免疫治疗或脱敏治疗(Fiebig,1995,J.4(6):336-339,Bousquet et 1998,J.Alllergy Clin.Immunol.102(4):558-562)。在此方法中,自然致敏原的提取物以不断提高的剂量给予病人皮下注射。然而这种方法有引起过敏,甚至过敏性休克的危险,为了最大限度地降低这种危险,使用了新型的类致敏原制剂,它们是经过化学修饰的致敏原提取物,其IgE反应性已明显降低,但T细胞反应与未处理的提取物相同(Fiebig,1995,Allergo J.4(7):377-382)。
用重组方法制备的致敏原使更有实质意义的优化治疗成为了可能。通过重组方法获得的且任选性地与病人个体的过敏模式匹配的高纯度的致敏原“鸡尾酒”可以取代自然致敏原来源的提取物,因为除了各种致敏原外,它们含有相对大量的具有免疫原性但无过敏性的二级蛋白。
用表达产物引起可靠的脱敏反应的现实前景因使用经过特异性突变删除了IgE表位,但不损害治疗作用所必需的T细胞表位的重组致敏原而产生(Schramm等,1999,J.Immunol.162:2406-2414)。
治疗过敏患者体内TH细胞失衡的另一个可能方法是免疫治疗性DNA疫苗接种,它涉及到用编码相关致敏原的表达性DNA进行治疗。通过给啮齿动物注射致敏原编码DNA已得到致敏原特异性免疫应答影响的初步实验证据(Schramm et 1999,J.Immunol.162:2406-2414)。
本发明因此涉及到以上或以下描述的作为药物的DNA分子和作为药物的相应的重组表达载体。
通过重组方法制备的相应的蛋白可以被用于花粉过敏的治疗及体外及体内诊断。
为了制备重组致敏原,克隆的核酸被连接到表达载体上,该构建体在一个适合的宿主生物体内表达,经过生物化学纯化,即可通过已建立的方法用重组致敏原检测IgE抗体。
本发明进一步涉及到含有以上或以下描述的功能性连接到表达控制序列的DNA分子的重组表达载体和被所述DNA分子或所述表达载体转化的宿主生物。
本发明还涉及到至少一种以上描述的DNA分子或至少一种以上描述的表达载体在制备用于禾本科(poaceae)第4组致敏原,优选是小麦族(Triticeae),尤其是Sec c 4、Hor v 4、Tri a 4参与触发的过敏病人的免疫治疗性DNA药物和/或预防这些过敏的药物中的用途。
正如已经陈述的那样,本发明可以作为重组致敏原或含核酸制剂的基本成分用于特异性免疫治疗,许多种可能在这里呈现,一方面,制剂的组成物可以是一级结构无改变的蛋白;另一方面,根据本发明,可以通过整体特异性删除分子上的IgE表位或产生编码T细胞表位的单一片段产生低致敏(类致敏原)形式来进行治疗以避免不必要的副作用;最后,核酸本身如果被连接到真核表达载体而制成制剂直接使用,能够治疗性地调节过敏免疫状态。
本发明进一步涉及到由以上描述的一个或多个DNA分子编码的多肽,特别是它们作为药物的效用。
它们是具有与SEQ ID NO 2、4、6、8或10一致的氨基酸序列的蛋白,特别值得一提的是它们是成熟蛋白(无信号序列成分),起始于第23位氨基酸(SEQ ID NO 2、4和6)和第22位氨基酸(SEQ ID NO 8和10)。本发明进一步涉及到含有这些氨基酸序列或这些序列的组成部分的蛋白。
本发明因此也涉及到通过培养宿主生物制备这样的多肽和从培养物中分离相应多肽的方法。
本发明同样涉及到至少一种上述多肽在制备用于诊断和/或治疗禾本科(poaceae),特别是小麦族(Triticeae)的第4组致敏原,尤其是Secc4、Hor v 4、Tri a 4参与触发的过敏反应的药物和预防这些致敏原的药物中的用途。
在确定本发明的蛋白和DNA序列时,应用了以下步骤:
来自黑麦的Sec c 4
1.从PhI p 4 DNA序列(SEQ ID NO 12,WO04/000881)入手,合成来自PhI p 4序列的特异性引物(表1),通过PCR用引物#87和#83从黑麦花粉DNA上获得5个克隆,扩增出的对应这些克隆的Sec c 4基因片段1编码与PhI p 4(SEQ ID NO 12)上第68-401位氨基酸序列对应的多肽。
2.用Sec c 4的部分序列在EST数据库进行了检索,在专门的黑麦EST数据库中没有发现与之同源的序列,然而,发现单个的、同源的、非重叠的EST片段在专门的大麦和小麦的EST数据中存在。单个的EST片段延伸至对应基因的5’-UTR区,其他延伸至3’-UTR区(UTR=非翻译区)。
3.然而,无法从EST数据库中的EST序列拼接出小麦和大麦的完整的第4组基因,因为这些序列不重叠,而且不知道第4组基因的同源序列。但是,参考PhI p 4序列(SEQ ID NO 11)和在步骤1获得的Sec c 4片段可以定位这些EST序列,这些序列可用作制备PCR引物的模板。
4.有了通过这种途径制备的引物#195和#189的帮助,用PCR得到3个克隆,引物#195来自大麦EST序列,引物#189为PhI p 4特异性引物,并与PhI p 4的终止密码子和PhI p 4的C末端的10个氨基酸序列重叠。通过这种途径扩增的Sec c 4基因片段2编码一个起始于信号序列,终止于与PhI p 4对应的490位的多肽。该多肽覆盖Sec c 4的N末端。
5a.用引物#195和#202通过PCR获得了另外3个克隆。这两个引物均来自大麦的EST序列,扩增的Sec c 4基因片段3编码起始于Sec c 4的信号序列,终止于Sec c 4的C末端的氨基酸。由此成熟Secc 4的完整序列以确定的序列被呈现出来。
以下5b和5c两个步骤用于对步骤5a获得的结果的“双保险”确定。5b.通过PCR用引物#195和#203获得了另外3个克隆,引物#195来自大麦的EST序列,引物#203来自小麦EST序列。该扩增的Sec c 4基因编码起始于Sec c 4的信号序列,终止于Sec c 4的C末端的对应氨基酸,由此成熟Sec c 4的完整序列以确定的序列被呈现出来。
5c.用引物#195和#198通过PCR获得了另外1个克隆,(alsoprimer#198),该扩增的Sec c 4基因编码起始于Sec c 4的信号序列,终止于Sec c 4的C末端的对应氨基酸,由此成熟Sec c 4的完整序列以确定的序列被呈现出来。
发现有Sec c 4.01和Sec c 4.02两种同工型,成熟致敏原起始于对应于SEQ ID NO 2、4和6序列的第23位氨基酸。来自大麦的Hor v 4
有通过以上描述获得的Sec c 4序列的帮助,发现同源的EST片段在大麦(Hordeum vulgare)的EST数据库中存在。这些片段重叠、但从中得不出完整基因。参考已发现的EST片段,有可能设计出Horv 4特异性引物,用于从染色体DNA中扩增Hor v 4基因。

有通过以上描述获得的Sec c 4序列的帮助,发现同源的EST片段在小麦(Triticum aestivum)的EST数据库中存在。这些片段重叠,但从中得不出完整的基因。参考已发现的EST片段,有可能设计出Tria 4特异性引物#199、#203、#204和#206,用于从染色体DNA中扩增Tri a 4基因。

其衍生的蛋白序列起始于Tri a 4的信号序列并延伸至该蛋白的C末端。发现有Tri a 4.01(从SEQ ID NO 8的第22位氨基酸起)和Tri a4.02(从SEQ ID NO 10的第22位氨基酸起)两个变异体。
为了制备本发明的重组致敏原,对应于SEQ ID NO 1、3、5、7和9的DNA序列被重组进表达载体(例如pProEx、pSE 380),应用了E.coli优化密码子来编码从该蛋白序列测定所知的N端氨基酸。
在转化进E.coli后,通过各种分离技术对本发明的重组致敏原进行表达和纯化,得到的蛋白质进行重新折叠。两种致敏原均可用于禾本花粉过敏的高特异性诊断,诊断可以是在体外通过检测特异性抗体(IgE、IgG1-4、IgA)并与携带IgE效应细胞(如血液中的嗜碱性白细胞)反应来进行,也可以是在体内通过皮试反应和在反应器官进行诱发来进行。
通过致敏原特异性刺激T淋巴细胞引起的增殖和细胞因子合成可以检测到禾本花粉过敏患者的T淋巴细胞与本发明的致敏原的反应,细胞可以是新鲜制备的血淋巴细胞中的T细胞,也可以是已经建立的n Sec c 4、nHor v 4或nTri a 4反应性T细胞系和克隆。
对编码半胱氨酸的三联子通过特异性点突变技术进行修饰,使其编码其他氨基酸,优选丝氨酸,制备出了单个半胱氨酸被替代的变异体和那些各种组合的两个半胱氨酸残基或所有半胱氨酸被修饰的变异体,这些半胱氨酸点突变体表达的蛋白与过敏患者的IgE抗体的反应被大大降低或与IgE抗体无反应,但能与这些病人的T淋巴细胞发生反应。
本发明因此涉及到以上或以下描述的DNA分子,这些DNA分子对应的多肽中的一个、多个或所有半胱氨酸残基通过特异性点突变技术被另一个氨基酸所取代。
含有T细胞表位和有低致敏性点突变(例如半胱氨酸取代)的多肽对应的低致敏性片段的免疫调节活性可以通过它们与禾本花粉过敏患者的T细胞进行反应来检测。
这样的低致敏片段或半胱氨酸点突变体可以用作过敏患者的脱敏制剂,因为它们与T细胞的反应效力不变,但由于与IgE的反应性降低或完全缺失所以它们引起的IgE介导的副作用会很低。
如果本发明的编码低致敏性致敏原变异体的核酸或本发明的未经修饰的DNA分子被连接到人体表达载体,这些构建体同样可用作免疫治疗(DNA疫苗接种)制剂。
最后,本发明涉及到用于禾本科(poaceae),特别是小麦族(Triticeae)的第4组致敏原,尤其是Sec c 4、Hor v 4、Tri a 4参与触发的过敏反应的免疫治疗或预防这些过敏反应的药用组合物,该药用组合物包括至少一种上述DNA分子或至少一种上述表达载体和可选的其他活性成分和/或佐剂。
本发明的另一组适用于所述过敏反应的诊断和/或治疗的药用组合物包含至少一种上述多肽,而不是DNA。
本发明意义上的药用组合物包含作为活性成分的本发明的多肽或表达载体和/或它们各自的药学上适用的衍生物,包括以各种比率混合的它们的混合物。本发明的活性成分可以与至少一种固体、液体和/或半液体赋形剂或佐剂做成合适的剂量形式,并可选择性地与一种或多种其他活性成分联用。
特别合适的佐剂为免疫刺激DNA或含有CpG基序的寡核苷酸。
这些组合物可以用作人类或兽类医疗方面的治疗药物或诊断试剂。合适的赋形剂为适合于肠外给药且对本发明的活性成分的功效没有负面影响的有机或无机物质。适合于肠外使用的,具体有溶液剂,优选油性或水性溶液剂,此外有混悬剂、乳剂或植入剂。本发明的活性成分也可以被冷冻干燥,形成的冻干物用于制备注射制剂。所指组合物可以是灭菌的和/或含有佐剂,如防腐剂、稳定剂和/或润湿剂、乳化剂、调节渗透压的盐、缓冲物质和/或多种其他活性成分。
此外,可以通过相应配制本发明活性成分,例如将其吸附于氢氧化铝而获得缓释制剂。

Claims (18)
1.一种DNA分子,其与选自SEQ ID NO 1、3、5、7和9的序列之一的谷类花粉主要致敏原的核苷酸序列相对应。
2.一种DNA分子,其在严格条件下与权利要求1中的DNA分子杂交并来源于禾本科物种的DNA序列。
3.一种编码多肽的DNA分子,其与来自黑麦(Secale cerale)、大麦(Hordeum vulgare)或小麦(Triticum aestivum)的主要致敏原Sec c4、Hor v4或Tri a4有免疫交叉反应并来源于禾本科物种的DNA序列。
4.一种DNA分子,其与权利要求1到3中的一项或多项部分序列或部分序列的组合相对应并且编码禾本科的第4组致敏原的免疫调节、T细胞反应片段。
5.一种与权利要求1到4中的一项或多项编码免疫调节性T细胞反应片段的核苷酸序列相对应的DNA分子,其特征是通过对单个密码子的特异性突变、删除或添加而特异性修饰所述核苷酸序列。
6.权利要求5中的DNA分子,其特征是所述突变导致了相应多肽的一个、多个或所有半胱氨酸被其他氨基酸替代。
7.一种重组DNA表达载体或克隆系统,其包含功能性连接到表达控制序列的权利要求1到6中一项或多项的DNA分子。
8.一种宿主生物,其用权利要求1到6中一项或多项的DNA分子或权利要求7中的表达载体转化。
9.一种制备由权利要求1到6中一项或多项的DNA序列编码的多肽的方法,所述方法为对权利要求8中的宿主生物进行培养并从培养物中分离相应的多肽。
10.一种与SEQ ID NO 2、4、6、8和10中氨基酸序列之一对应的多肽,其由权利要求1到6中一项或多项的DNA序列编码。
11.一种与权利要求10中的氨基酸序列的成熟致敏原相应的多肽,选自以下氨基酸序列:
-一段SEQ ID NO 2、4或6的氨基酸序列,起始于第23位氨基酸;
-一段SEQ ID NO 8或10的氨基酸序列,起始于第22位氨基酸。
12.权利要求10或11中的多肽,其用作药物。
13.一种包含至少一种权利要求12中的多肽并任选包含其他活性物质和/或佐剂的药用组合物,其用于诊断和/或治疗来自禾本科的第4组致敏原参与触发的过敏反应。
14.至少一种权利要求12中的多肽在制备用于诊断和/或治疗来自禾本科的第4组致敏原参与触发的过敏反应的药物中的用途。
15.权利要求1到6中一项或多项的DNA分子,其用作药物。
16.权利要求7中的重组表达载体,其用作药物。
17.一种包含至少一种权利要求15中的DNA分子或至少一种权利要求16中的表达载体并任选包含其他活性成分和/或佐剂的药用组合物,其用于免疫治疗性DNA疫苗接种来自禾本科的第4组致敏原参与触发的过敏反应病人和/或预防这样的过敏反应。
18.至少一种权利要求15中的DNA分子或至少一种权利要求16中的表达载体在制备用于免疫治疗性DNA疫苗接种来自禾本科的第4组致敏原参与触发的过敏反应病人和/或预防这样的过敏反应的药物中的用途。
