Abstract
本发明涉及含有一种或多种酶的组合物,即一种或多种水解酶和一种或多种抗氧化剂,所述抗氧化剂选自泛醌、类胡萝卜素和抗氧化性维生素。本发明的组合物作为生产食物产品或药物的组分形式使用,尤其作为饮食食物补充物使用,特别用于补充饮食方案中,以及用于治疗免疫系统病症。
Description
本发明涉及含酶的组合物和食物产品领域,特别是饮食性食物补充物,其尤其用作补充平衡膳食,且涉及包括该含酶组合物或由该含酶组合物组成的药物。本发明更确切的涉及包括水解酶和另外的药物活性成分的含酶组合物、食物产品和药物,且涉及其尤其在通过内在和外在影响,治疗由免疫系统调节异常导致的所有疾病中的用途。
对于本发明来说,平衡和补充平衡膳食是指,病人在为了治疗疾病的特定医药目的的医疗监督下所摄入的食物产品。
食物补充物是以片剂、胶囊或粉剂形式提供的食物产品。其通过特定物质用于补充膳食,例如:维生素、矿物质、痕量元素和填充剂。为了实现这个目的,这些物质通常都以增加的浓度存在。饮食食物补充物是指那些本身仅含有较小平均热值和营养值,且从而也可作为膳食的一部分被消耗的产品。
食物补充物有助于提高健康人的营养,上述人,例如,由于其正常的日常饮食习惯,在正常饮食中,没有机会或没有时间常规的消耗足够量的所需的全部营养。
而且,食物补充物还可用于补偿由疾病或治疗疾病导致的缺陷状况。在各类疾病的常规膳食中通常不可能存在的,对(例如,饮食的)食物补充物具有保护效果的必需微量营养物和/或食物成分缺乏的针对性补偿,可改善治疗效果和病人的生活质量。因此,食物补充物通常可改善使用者和患病者的健康状况,特别是有益于疾病的进程和疗效。
食物补充物一般而言不是药物。但是根据其使用的领域、选择的成分及其剂量,其也可作为药物。
本发明涉及含有用于食物补充物的活性成分的特定组合物,以及由其获得的食物产品和药物,所述的食物产品和药物还可另外包含赋形剂,例如用于药物制剂的。采用这些组合物、食物产品或药物可改善健康人和患病者的必需营养物供应,此外在患病者的情况下,可增强治疗的效果。
含有酶的产品是本领域已知的。WOBE-Mugos包括:胰蛋白酶、胰凝乳蛋白酶、木瓜蛋白酶和胸腺提取物。其尤其用于恶性肿瘤的支持性长期治疗。Wobenzym包含动物胰腺酶和来源于凤梨(Ananascomosus)和番木瓜(Caica papaya)的植物酶的混合物加上芸香苷。其用于炎症和血管病症的支持性治疗。
从现有技术出发,本发明的目的是指出具有修饰的化学组成的组合物、食物产品和药物,其作为促进健康的食物产品具有改善的效果,例如,以食物补充物的形式,以及作为医疗部门的药物具有改善的效果,并且适合于新的用途和适应症。
在第一方面,本发明涉及包含一种或多种水解酶和一种或多种选自具有抗氧化活性的维生素、类胡萝卜素、含硒物质、泛醌的抗氧化剂的组合物。
水解酶是能水解切割酯、醚、肽、糖苷键等的酶。其包括酯酶、磷酸酶、糖苷酶和蛋白酶,所述的酶可根据本发明来使用。除了从天然产物中获得的天然存在的水解酶外,还可能采用人工,例如通过遗传操作生产的水解酶。水解酶可以从植物、动物器官、细菌(例如,来自于扩展短杆菌(Brevibacterium linens)的蛋白酶)和真菌产业化中获得。细菌或其他微生物通过遗传操作用于获得上述水解酶。水解酶可以单独使用,或者与较多的水解酶联合使用。根据本发明可以采用的水解酶的例子将在下文中示出。
即使在较低浓度下,抗氧化剂也可阻止或推迟活性物质的氧化分解作用。在人体中,它们可以保护细胞不受损伤并且具有抗癌作用。抗氧化剂可以单独使用,或者与较多的抗氧化剂联合使用。根据本发明可以采用的来自具有抗氧化作用的维生素族、类胡萝卜素、含硒物质、泛醌的抗氧化剂的例子将在下文中示出。
将一种或多种水解酶与一种或多种抗氧化剂联合可得到一种组合物,其对健康人和患病者,特别是患有免疫系统调节异常的人来说,具有比单独一种物质效果更好的促进健康效果。在此包括的疾病是慢性或急性免疫缺陷、机体防御的损伤、炎症-风湿性病症和其他炎症,免疫和肿瘤病症。为了治疗,所述组合物可以以饮食性食物产品的形式使用,用于补充平衡膳食。增加的营养物可增强患病者的健康。所述的患病者可以是风湿病患者或免疫缺陷患者,和特别是,正接受控制肿瘤的治疗的肿瘤患者。上述治疗可包括手术过程,化学疗法和/或放射疗法,在这些过程中,病人对必需微量营养物和具有保护效果的食物成分具有更大的需求。

此外,本发明的上述组合物还可包括一种或多种多酚,一种或多种类黄酮和/或一种或多种含类黄酮的物质。
此外,上述列出的本发明的组合物还可包括一种或多种氨基酸和/或一种或多种多糖。
在第二方面,本发明涉及包含一种或多种水解酶和一种或多种氨基酸的组合物。该组合物还具有有益的免疫调制特性。
在第三方面,本发明涉及包含一种或多种水解酶和一种或多种多糖的组合物,所述组合物与水解酶和氨基酸的组合物一样,尤其可通过食物产品或药物的形式来施用,以用于改善健康状况,特别是改善免疫状况。在两种情况下,令人惊讶的发现,即使在不存在抗氧化剂的情况下,也可获得这些有益特性。
本发明进一步涉及包含水解酶与氨基酸和多糖联合的组合物。
在第四方面,本发明还涉及仅包括水解酶作为活性组分的组合物,其可与药学上可接受的或食物产品中允许的赋形剂适当地联合使用。

(VII)组合物(IV)+多酚和/或类黄酮和/或含类黄酮物质;
(VIII)组合物(V)+多酚和/或类黄酮和/或含类黄酮物质;
(IX)组合物(VI)+多酚和/或类黄酮和/或含类黄酮物质;
(X)水解酶;
其中,上述叙述,例如,“氨基酸”应当理解为一族物质的含义,其中的一个或多个代表存在于本发明的组合物中。因此,“氨基酸”是指“一个或多个氨基酸”,例如“L-精氨酸或“L-精氨酸+L-半胱氨酸”等,并且这对于全部其他的所述物质族来说也一样适用。
所述水解酶优选为植物蛋白酶和/或动物蛋白酶,其来源于天然产物,表示天然产物本身,或者由合成或遗传操作产生。
所述植物蛋白酶优选地由纯菠萝蛋白酶、凤梨提取物、纯木瓜蛋白酶和番木瓜提取物组成,因为其可以单独使用或联合使用。菠萝蛋白酶由许多具有蛋白水解活性的酶,更确切的说是内肽酶组成,其存在于成熟和不成熟的凤梨中,并且取自种子头部的蒂中。菠萝蛋白酶尤其具有抗炎作用。木瓜蛋白酶是一种取自不成熟番木瓜果的乳液中的蛋白酶。特别优选木瓜蛋白酶和菠萝蛋白酶联合使用。上述酶可以纯化的形式使用;或者以植物提取物的形式使用,只要其含有所述植物蛋白酶;尤其以凤梨提取物和番木瓜提取物的形式使用。还可优选的采用来源于无花果,或无花果提取物的无花果蛋白酶,它是一种与木瓜蛋白酶和菠萝蛋白酶情况相似的半胱氨酸蛋白酶。
本发明所采用的动物蛋白酶优选的由胰蛋白酶和/或胰凝乳蛋白酶和/或胃蛋白酶(肽基-肽水解酶)组成。胰蛋白酶来源于,例如猪的酸性胰腺液经在含水介质中分段沉淀和在弱碱性pH值范围中活化胰蛋白酶原得到。这样获得的是α-,β-,和γ-胰蛋白酶的混合物,其可以水解肽,酰胺和酯键,如果在这些键中包含了碱性氨基酸精氨酸和赖氨酸的话。通过在含水介质中提取和随后的分段沉淀可从牛酸性胰腺液中制备得到胰凝乳蛋白酶。胰凝乳蛋白酶在弱碱性介质中转化成活性形式。通过进一步的化学处理步骤(超滤,色谱纯化)可获得结晶形式的胰凝乳蛋白酶,并且可随后使用。获得的胰凝乳蛋白酶A和B混合物可水解肽,酰胺和酯键,如果在这些键中包括了芳族氨基酸酪氨酸、苯丙氨酸和色氨酸,以及亮氨酸、甲硫氨酸和谷氨酸的话。作为蛋白水解酶,胃蛋白酶也可切割IgG型循环免疫球蛋白,从而将IgG分子降解成Fc片段和二价F(ab)2片段。
菠萝蛋白酶具有有效的抗水肿作用,从而比其他蛋白酶具有更快的消肿效果。胰蛋白酶同样具有抗水肿活性。胰蛋白酶和胰凝乳蛋白酶对血液凝固和从而对血液的流动特性有很强的作用。将木瓜蛋白酶和胰蛋白酶联合使用,可能实现特别明显的C1Q结合减少(免疫系统的补体区域)。这两个蛋白酶以不能形成结合的方式改变了该结合区域。从而抑制了炎症反应。
菠萝蛋白酶和木瓜蛋白酶可以有效的切割免疫复合体,所述免疫复合体被认为是免疫系统调节异常的标志。高免疫复合体浓度可在,特别是与自身免疫病和肿瘤病症(癌症)相关的病症中反复测量到。将其除去是不损害小噬细胞活性的前提。
所有的蛋白酶都作用于所谓的细胞表面粘附分子的表达。细胞间的联系尤其是通过这些粘附分子实现的。通过蛋白酶的联合作用,粘附分子的调节被显著改善了。
单酶产品的施用(例如,单独采用波罗蛋白酶)到目前为止不具备酶联合施用那样的治疗效果。将菠萝蛋白酶和胰蛋白酶联合施用可能发现其在降低炎症方面有增加的效果。这一点可以通过酶联合使用比在单酶产品的情况下显著缩短恢复阶段来体现。
植物与动物蛋白酶的联合可活化静息单核细胞和NK细胞,从而刺激免疫系统。另一方面,本发明的酶的联合可下调过度刺激、过度活化的免疫细胞。
当所述蛋白酶与抗氧化剂,特别是含硒物质,尤其是亚硒酸钠,含类黄酮物质等,氨基酸和任选的多糖联合使用时,这种效果特别明显。
本发明优选的组合物(X)的例子为由菠萝蛋白酶和/或木瓜蛋白酶和/或胰蛋白酶和/或胰凝乳蛋白酶组成的组合物,其不包括其他的活性成分,并且任选地与药学上可接受或食物产品中允许的赋形剂联合使用,例如菠萝蛋白酶+木瓜蛋白酶+胰蛋白酶+胰凝乳蛋白酶。
根据本发明可以采用的其他水解酶的离子为:α-淀粉酶,β-淀粉酶,α-D-半乳糖苷酶,葡糖淀粉酶,脂肪酶,蛋白酶,鞣酸酶,转化酶,溶菌酶,支链淀粉酶,硫葡糖苷酶,乳糖酶,pectinolase,α-L-鼠李糖苷酶,其可以单独,或者以混合物的形式使用,也可以与上述植物和动物蛋白酶联合使用。
所述水解酶可在一较宽的浓度范围内使用。其下限值相应的浓度为低于该浓度则所需的健康效果或治疗效果将不存在。上限值可被定义为超过其浓度则活性不再增加,从而仅仅导致产品制造的费用上升的浓度。
水解酶例如菠萝蛋白酶和木瓜蛋白酶优选的以20重量%至60重量%,更优选30重量%至50重量%的总浓度使用,所述比例基于组合物中的活性成分总量。如果所述组合物包含两种或多于两种的水解酶,则优选的以类似大小的量使用,例如25重量%(或15重量%)的木瓜蛋白酶和25重量%(或15重量%)的菠萝蛋白酶。不过,所述水解酶也可能采用不同的浓度使用,例如15重量%的动物蛋白酶和35重量%的植物蛋白酶,反之亦可,或者15重量%的第一种植物蛋白酶,例如菠萝蛋白酶,和35重量%的第二种植物蛋白酶,例如木瓜蛋白酶,或者反之亦可。
在本发明一优选实施方案中,抗氧化剂由选自维生素A,C和E的维生素,和维生素A和E的酯组成,其可在食物产品和药物中使用。这类酯的例子为醋酸酯,天冬氨酸酯和酒石酸酯。
还可能支持性地采用维生素K,维生素B1(硫胺素),B2(核黄素),B6(吡哆醇),B12(氰-钴胺素),烟酸,泛酸,生物素,叶酸,肌醇,但是,上述物质本身不具有抗氧化作用。B族维生素都具有辅酶的功能,从而将其口服施用,特别是与氨基酸和/或多糖联合施用具有生理上的优点。
所述维生素在本发明的组合物中单独或混合使用。其总浓度为,例如30重量%至60重量%,更优选40重量%至50重量%,所述比例基于组合物中的活性成分总量,如果不采用其他抗氧化剂的话。如果存在其他抗氧化剂,则具有抗氧化作用的维生素可以以较低浓度使用。
类胡萝卜素也表现出抗氧化的作用,它是次级植物成分,例如,番茄红素——一种具有多烯结构的黄红色植物色素,其存在于野蔷薇果实、番茄和其他水果中。番茄红素是一种多不饱和多烯,其被证实是一种非常有效的天然自由基清除剂。从而它可以特别有利地与上述抗氧化剂维生素联合使用。与番茄红素的联合使用可导致在风湿病治疗和癌症治疗的框架范围内的伴随治疗中现察到显著的改善效果。作为抗氧化剂总量的组份之一的类胡萝卜素,例如番茄红素的适当的量为,例如5重量%至20重量%,所述比例基于活性成分总重量,其更优选9重量%至16重量%。
此外,可在本发明组合物中作为抗氧化剂使用的类胡萝卜素,例如番茄红素为α-,β-,γ-胡萝卜素,链孢红素(neurosporin),六氢番茄红素,来源于胡萝卜素族的八氢番茄红素,和叶黄素,堇莱黄素,番红花酸,虾青素,来源于叶黄素族的辣椒红。
本发明的组合物还可包含一种或多种类黄酮,其以纯类黄酮的形式或以包括类黄酮的天然产物的形式掺合。类黄酮属于多酚族或植物酚族(PHP=多羟基苯酚)。
根据本发明,所述类黄酮可选自:黄酮,黄酮醇,黄烷醇,异黄烷醇(isoflavanol)。除此之外,这些还包括:槲皮黄酮,槲皮黄酮糖苷,例如芸香苷,黄烷酮圣草酚,桔皮素,甘草素,柚皮素,生松素,黄烷酮醇黄杉素(flavanolol taxifolin),黄酮芹菜素,白杨素,香叶木素,半齿泽兰素,木犀草素,黄芩素,黄芩素四甲基醚,橙黄酮和黄酮醇美苍耳素,猫眼草黄素,chrysoplenol,eupaletin,eupatoletin,黄颜木素,高良姜素,棉黄素,异鼠李黄素,棕鳞矢车菊素,山奈素,杨梅树皮素,藤菊黄素,六羟黄酮,鼠李黄素,刺槐亭,菠菜素。
根据本发明优选的类黄酮为:槲皮黄酮,桔皮素,生松素,黄杉素,白杨素,木犀草素,山奈素,鼠李黄素。
合适的含类黄酮的物质为洋葱粉末,荞麦提取物,葡萄籽类黄酮和柑橘类黄酮。葡萄籽和柑橘类黄酮包括属于黄酮醇类的寡聚矢车菊苷配质(OPC)。标准的OPC包含至少50%或更多的松树树皮OPC(海岸松(Pinus maritima))和葡萄籽OPC(葡萄(Vitis vinifera))。葡萄籽OPC包括,例如最高达20%的儿茶素,表儿茶精和表没食子儿茶精,松树OPC包括最高达10%的儿茶素和最高达10%的黄杉素。由于其抗氧化作用,这些寡聚矢车菊苷配质也可被用于本发明的组合物。
槲皮黄酮和槲皮黄酮糖苷,例如芸香苷(rutoside)是特别优选的。芸香苷通常用作止痛剂,但是与其他类黄酮相似,其在本发明的治疗方法中也显示出令人惊讶的好特性,即作为饮食食物产品中的平衡微量营养物来应用,从而加强免疫系统,增强机体防御,用于治疗炎性风湿性病症和在身体和/或精神压力的情况下调节免疫系统。
类黄酮和含类黄酮的天然产物可以单独使用或以两种或更多种该物质的混合物形式使用。其有利地在活性成分总重量的10重量%至50重量%的浓度下使用。不过,更高和更低的浓度同样的也是可以预期的。葡萄籽类黄酮和松树皮类黄酮应用的每日剂量有利地为150mg。
如果本发明的组合物另外包含一种或多种具有抗氧化作用的含硒物质,例如硒酵母,和/或碱金属亚硒酸盐,例如亚硒酸钠,碱土金属亚硒酸盐,例如亚硒酸镁,亚硒酸铵,及其相应的硒酸盐或硒代氨基酸(selenamino acid)(Se-甲硫氨酸,Se-半胱氨酸),则可在癌症和风湿病的伴随治疗中,以及在加强机体免疫防御过程中获得很好的效果。硒是一种重要的的痕量元素。其以谷胱苷肽过氧化物酶、硫氧还蛋白还原酶和甲状腺素5-deiodase的组份天然存在。硒以胞内抗氧化剂的方式其作用。它作为自由基清除剂用于对抗自由基形成物或脂质氢过氧化物造成的细胞损伤。例如,硒可以调节淋巴细胞的功能,并且可以增强天然杀伤细胞的活性。硒可对DNA修复机制起作用,并且参与细胞凋亡的引发。在本发明的组合物中,它可以导致所述组合物的免疫调节特性的协同增加。硒在上述组合物中是一种具有抗氧化作用的重要痕量元素,并且根据本发明它是抗氧化剂维生素E和维生素C的理想联合配偶体。
特别优选亚硒酸钠。亚硒酸钠(以及其他碱金属和碱土金属亚硒酸盐),由于其化学结构,在施用后吸收非常良好,尤其是在口服施用本发明的组合物时,并且它对靶细胞显示出理想的直接催化活性。因此亚硒酸钠具有具有特别好的生物可利用率和特别高的生物学活性。例如,当本发明的含有亚硒酸钠的组合物被用于保护化疗或放疗过程中癌症病人的健康机体细胞不受化疗/放疗造成的细胞损伤的影响时,其作用没有时间延迟并且效果很好,这是非常有利的。亚硒酸钠的快速的生物可利用率和高度的生物学活性保证了本发明的组合物在癌症治疗当天的摄取在很短时间内导致了所需的细胞保护作用。相反地,硒酵母中存在的硒代甲硫氨酸首先必须转化成为无机前体,随后可被活化成可利用的硒库。因此,在硒酵母的情况中,上述效果出现时间延迟现象。此外,亚硒酸钠长期施用没有危险,这是因为硒以非活化且有时甚至有害于健康的形式聚集。
对受试者口服施用亚硒酸钠后,可能测量到血清中白细胞介素4(IL-4)显著减少,且细胞因子INF-γ显著增加。口服施用亚硒酸钠后,还可能发现免疫状况发生显著变化:CD8+T细胞显著减少。亚硒酸钠可将慢性病患者中常见的免疫平衡紊乱调节至免疫体内稳态的方向。可实现1型T辅助细胞(Th1),2型T辅助细胞(Th2),细胞毒性T细胞和CD8+细胞毒性T细胞间明显的平衡。
水解酶,更确切的说是蛋白酶,特别是蛋白的组合物,例如菠萝蛋白酶+木瓜蛋白酶或菠萝蛋白酶+木瓜蛋白酶+胰蛋白酶和/或胰凝乳蛋白酶,通过其免疫调节作用可改善亚硒酸钠的免疫调节特性。通过亚硒酸钠和蛋白酶的共同施用可防止免疫系统单方面高度刺激及产生的各种免疫系统参数的不平衡现象。这种正向协同效应可用于所有的慢性疾病和肿瘤病症。
亚硒酸盐可采用活性成分总量0.001重量%至0.3重量%的浓度,优选0.01重量%至0.1重量%。50至300μg的每日剂量被证实特别有效。
根据本发明采用的与水解酶联合使用或作为优选添加物加入本发明的其他组合物中的另一类物质为一类氨基酸。甘氨酸,L-丙氨酸,L-丝氨酸,L-苏氨酸,L-缬氨酸,L-亮氨酸,L-异亮氨酸,L-天冬氨酸,L-天冬酰胺,L-谷氨酸,L-谷氨酰胺,L-精氨酸,L-赖氨酸,L-羟赖氨酸,L-鸟氨酸,L-瓜氨酸,L-半胱氨酸,L-胱氨酸,L-甲硫氨酸,L-酪氨酸,L-苯丙氨酸,L-色氨酸,L-组氨酸,L-脯氨酸,L-羟脯氨酸可单独使用或组合使用,并且只要上述氨基酸能形成酯和盐,则其药学上可接受的和可用于食物产品的酯和盐也可使用。
甘氨酸,L-精氨酸,L-谷氨酰胺和L-甲硫氨酸被证实特别有效。L-精氨酸最优选。
氨基酸在调节免疫系统中也起作用。其通过细胞表面和细胞因子调节,并通过影响系统的氧化还原潜能来影响免疫系统的信号传递(信号转导)。精氨酸参与多胺的合成。这些依次非常重要地参与细胞分裂,DNA合成和细胞周期调节。通过这些机制,精氨酸也对淋巴细胞的增殖,巨噬细胞和NK细胞的细胞溶解能力,以及细胞因子平衡(IL1)产生作用。精氨酸是氧化氮(NO)的主要内源初级代谢物,此外,其对血流动力学状况有重要影响。细胞因子TGF-β的超表达在慢性和急性病,包括癌症中都有出现。氨基酸精氨酸可在炎症位点局部减少细胞因子TGF-β的超表达。根据本发明的与蛋白酶的组合物也确保具有这种效果。
基于活性组分的总量,氨基酸的浓度优选为5重量%至15重量%,更优选7重量%至12重量%。
由于其良好的免疫调制特性,本发明的组合物还由水解酶和多糖组成,或者优选地包括多糖与抗氧化剂的联合,所述多糖选自:直链淀粉,石耳素,右旋糖酐,支链淀粉,纤维素,阿拉伯聚糖,阿拉伯半乳聚糖,果聚糖,葡聚糖,例如:β-葡聚糖,甘露聚糖,糖原,几丁质,果胶,刺槐豆胶,罗望子胶,瓜尔胶,其可单独使用或以混合物的方式使用,并且β-葡聚糖具有特别有益的效果。
基于活性组分总量,其优选的在5重量%至10重量%的浓度范围内使用。
β-葡聚糖刺激巨噬细胞活性,并导致NK细胞和T淋巴细胞数目增加。作为免疫调制物质,它是蛋白酶的一种最优的联合配偶体,特别是在上述组合物(III),(V),(VI),(VIII)和(IX)中。
为了改善免疫系统细胞的功能,本发明的组合物还可与矿物质混合,例如钾,钙,镁,铁,锌,锰或锗,其呈无机盐,有机酸盐,或在复合体形式中包括了该金属的有机化合物的形式。锌和锰化合物作为蛋白酶的辅酶起作用,因此有助于细胞的生物化学和生理学过程。锌是蛋白质和核酸新陈代谢的必需痕量元素。它可将细胞中的维生素A转化成活化的形式。锌与维生素C显示出显著的协同效果。锗在免疫系统中具有重要作用,它可刺激干扰素合成并且在所有的免疫触发的疾病中都很重要。因此,锗在所有的肿瘤病症和风湿病症中都非常重要。
因为其有益的效果范围,从医学角度看,根据本发明的如下活性物质特别优选:
根据物质种类
●水解酶:菠萝蛋白酶,木瓜蛋白酶,胰蛋白酶,胰凝乳蛋白酶,其单独的或作为两种至四种水解酶的组合物使用;
●维生素:维生素A,E和C,单独或作为这些维生素的混合物使用;
●类胡萝卜素:番茄红素,α-,β-,γ-胡萝卜素,叶黄素,单独或作为这些类胡萝卜素的混合物使用;
●含硒物质:亚硒酸钠,;
●类黄酮(多酚-PHP):桔皮素,柚皮素,生松素,黄杉素,白杨素,半齿泽兰素,木犀草素,猫眼草黄素,chrysoplenol,eupaletin,高良姜素,棉黄素,山奈素,槲皮黄酮,其单独或以混合物的形式使用;
●氨基酸:L-精氨酸,L-甲硫氨酸,L-甘氨酸,L-谷胺酰胺,其单独或以混合物的形式使用;
●多糖:阿拉伯聚糖,果聚糖,葡聚糖,甘露聚糖,支链淀粉,单独或以多种多糖的混合物的形式使用;
●矿物/痕量元素:钾,钙,镁,铁,锌,锰或锗,其单独或联合使用。
蛋白酶,维生素,氨基酸和痕量元素的这些组合物对免疫系统的作用比单独物质的预期作用要好。在免疫系统对急性或慢性紊乱因子产生应答后,在本发明的组合物的辅助下,当免疫系统,细胞因子和细胞表面粘着分子的执行机构从各方受到调节和影响时,可更快的恢复作为免疫系统平衡先决条件的免疫系统体内稳态。仅采用上述各种免疫调制物质的组合物就可在可接受的时间内有效的实现这种效果。
具有特定有益特性的本发明的组合物如下文所述。根据活性组分,它们是具有如下性质的组合物:
●木瓜蛋白酶,菠萝蛋白酶,番茄红素,维生素E醋酸盐,维生素A醋酸盐和亚硒酸钠(参见实施例1);
●木瓜蛋白酶,菠萝蛋白酶,番茄红素,维生素E醋酸盐,β-胡萝卜素,葡萄籽类黄酮和亚硒酸钠(参见实施例2);
●木瓜蛋白酶,菠萝蛋白酶,β-胡萝卜素,柑橘类黄酮,L-精氨酸和β-葡聚糖(参见实施例3);
●木瓜蛋白酶,菠萝蛋白酶,维生素E醋酸盐,含芸香苷的荞麦提取物,含槲皮黄酮的洋葱粉末和L-精氨酸(参见实施例4)。
本发明的组合物还可包括一种和多种动物蛋白酶,例如:胰蛋白酶或胰凝乳蛋白酶或胃蛋白酶;胰蛋白酶+胰凝乳蛋白酶;胰蛋白酶+胰凝乳蛋白酶+胃蛋白酶;胰凝乳蛋白酶+胃蛋白酶;胰蛋白酶+胃蛋白酶。
定量的数据参照涉及相同组合物的实施例1至4。如前文水解酶中所提到的那样,组合物中的这些活性组分的量不是临界值。所述下限值为在该量以下将不再观察到所需作用的值。所述上限值被认为是在该各活性组份量之上将不能观察到作用的进一步增加的值。
可通过简单混合活性组分以及随后使其呈粉末形式来制备本发明的组合物。其可仅由所述的活性成分组成,或者包括后来添加的常规赋形剂。不过,为了制备特定的剂量形式,例如片剂,具有肠溶衣的膜涂覆的片剂,胶囊,糖衣片剂,泡腾片,栓剂,灌肠剂,它们可与常规的药物赋形剂混合。其中用于该目的的是:微晶纤维素,滑石,胶体二氧化硅,硬脂酸,硬脂酸镁,氧化镁,鱼明胶,交联聚乙烯吡咯酮,硅酸钙,羟丙基纤维素,羟甲基纤维素和二氧化钛。各种剂型的药物制剂不是临界值,并且是本领域已知的。
本发明进一步涉及食物产品,特别是补充平衡膳食,其用于加强免疫防御或机体防御,用于治疗炎症和炎性风湿性关节炎,用于调节身体和/或精神压力状况下的免疫系统,用于在肿瘤消解治疗之前,过程中和/或之后加强免疫系统,或用于减少肿瘤复发和继发的风险,所述食物产品包含上文所述的本发明的组合物中的一种,或者如果不加入添加剂,则由该组合物组成。
本发明还涉及包含本发明的组合物或由本发明组合物组成的药物,其用于加强免疫防御或机体防御,用于治疗炎症和炎性风湿性关节炎,用于调节身体和/或精神压力状况下的免疫系统,用于在肿瘤消解治疗之前,过程中和/或之后加强免疫系统,或用于减少肿瘤复发和继发的风险。
本发明还涉及本发明的组合物在饮食治疗中的用途,尤其是作为补充平衡膳食,用于加强免疫防御或机体防御,用于治疗炎症和炎性风湿性关节炎,用于调节身体和/或精神压力状况下的免疫系统,用于在肿瘤消解治疗之前,过程中和/或之后加强免疫系统,或用于减少肿瘤复发和继发的风险。
用于说明上述用途的实例为在癌治疗过程中的伴随治疗,其中所采用的组合物为实施例1所示的组合物。采用所述组合物和由其生产的食物产品可保证健康和完全的膳食。这适用于肿瘤破坏性治疗,例如,手术,化疗和/或放疗之前,过程中和之后的确切时间间隔的时间段。由恶心,食物摄入减少,呕吐,腹泻和出汗引起的微量营养物(例如,维生素、痕量元素)摄入的减少得到了补偿。这改善了病人对治疗的耐受性,改善了对治疗的应答并且提高了病人的生活质量。所述蛋白酶减少了治疗的副作用,所述番茄红素显示出预防和治疗作用。例如可能发现前列腺癌的发病率和进攻性减轻了。总而言之,肿瘤破坏性治疗的效果改善了。
限定的平衡微量营养物,例如维生素A,维生素E,亚硒酸钠,番茄红素,菠萝蛋白酶和木瓜蛋白酶的施用可被分成两部分:第一部分表示了采用组合物的增加剂量的积极治疗时期,随后是第二部分,其中在四周的时间内施用维持剂量。所述剂量通常是积极治疗时期剂量的50%。

Claims (27)
1.一种组合物,其包含一种或多种水解酶和一种或多种抗氧化剂,所述抗氧化剂选自具有抗氧化活性的维生素,类胡萝卜素,含硒物质,泛醌。
2.一种组合物,其包括一种或多种水解酶和一种或多种氨基酸。
3.一种组合物,其包括一种或多种水解酶和一种或多种多糖。
4.一种组合物,其包括一种或多种水解酶,一种或多种氨基酸和一种或多种多糖。
5.如权利要求1所述的组合物,其特征在于其包括一种或多种多酚,一种或多种类黄酮和/或一种或多种含类黄酮的物质。
6.如权利要求1或5所述的组合物,其特征在于其包括一种或多种氨基酸和/或一种或多种多糖。
7.如权利要求2,3和4中任意一项所述的组合物,其包括一种或多种类黄酮和/或一种或多种含类黄酮的物质。
8.如权利要求1至7中任意一项所要求保护的组合物,其特征在于所涉及的水解酶为植物蛋白酶和/或动物蛋白酶。
9.如权利要求8中所要求保护的组合物,其特征在于所述植物蛋白酶选自菠萝蛋白酶,含菠萝蛋白酶的凤梨提取物,木瓜蛋白酶,含木瓜蛋白酶的番木瓜提取物,并且所述动物蛋白酶选自胰蛋白酶和胰凝乳蛋白酶。
10.如权利要求1和5至9中任意一项所要求保护的组合物,其特征在于所述具有抗氧化作用的维生素选自维生素A,C和E以及维生素A和E的酯。
11.如权利要求1和5至10中任意一项所要求保护的组合物,其特征在于所述具有抗氧化作用的含硒物质选自硒酵母,碱金属亚硒酸盐例如亚硒酸钠,碱土金属亚硒酸盐例如亚硒酸镁,或亚硒酸铵,及其相应的硒酸盐。
12.如权利要求5至11中任意一项所要求保护的组合物,其特征在于所述类黄酮选自黄酮,黄酮醇,黄烷醇,异黄烷醇。
13.如权利要求12所要求保护的组合物,其中所述类黄酮包括:槲皮黄酮,芸香苷,桔皮素,生松素,黄杉素,白杨素,木犀草素,山奈素,鼠李黄素。
14.如权利要求5至11中任意一项所要求保护的组合物,其中含类黄酮物质选自洋葱粉末,荞麦提取物,葡萄籽类黄酮和柑橘类黄酮。
15.如权利要求1,5,6和8至14中任意一项所要求保护的组合物,其中泛醌为辅酶Q10。
16.如权利要求1,5,6和8至15中任意一项所要求保护的组合物,其中类胡萝卜素包括胡萝卜素,番茄红素,链孢红素,六氢番茄红素,八氢番茄红素,叶黄素,堇莱黄素,番红花酸,虾青素,辣椒红。
17.如权利要求2,4,6至16中任意一项所要求保护的组合物,其特征在于所述氨基酸选自L-精氨酸和其酸加成盐,甘氨酸,L-谷氨酰胺和其酰胺,L-甲硫氨酸。
18.如权利要求3,4,6至17中任意一项所要求保护的组合物,其特征在于所述多糖选自直链淀粉,石耳素,右旋糖酐,支链淀粉,纤维素,阿拉伯聚糖,果聚糖,葡聚糖,例如:β-葡聚糖,甘露聚糖,糖原,几丁质,果胶。
19.如权利要求1中所要求保护的组合物,其包含木瓜蛋白酶,菠萝蛋白酶,番茄红素,维生素E醋酸盐,维生素A醋酸盐和亚硒酸钠作为活性成分。
20.如权利要求5中所要求保护的组合物,其包含木瓜蛋白酶,菠萝蛋白酶,番茄红素,维生素E醋酸盐,β-胡萝卜素,葡萄籽类黄酮和亚硒酸钠作为活性成分。
21.如权利要求5中所要求保护的组合物,其包含木瓜蛋白酶,菠萝蛋白酶,维生素E醋酸盐,含芸香苷的荞麦提取物,含槲皮黄酮的洋葱粉末和L-精氨酸作为活性成分。
22.如权利要求6中所要求保护的组合物,其包含木瓜蛋白酶,菠萝蛋白酶,β-胡萝卜素,柑橘类黄酮,β-葡聚糖和L-精氨酸作为活性成分。
23.如权利要求19至22中任意一项所要求保护的组合物,其包括:胰蛋白酶或胰凝乳蛋白酶或胃蛋白酶;胰蛋白酶+胰凝乳蛋白酶;胰蛋白酶+胰凝乳蛋白酶+胃蛋白酶;胰凝乳蛋白酶+胃蛋白酶;胰蛋白酶+胃蛋白酶。
24.一种用于食物补充,特别是补充平衡膳食的食物产品,其用于加强免疫防御或机体防御,用于治疗炎症和炎性风湿性关节炎,用于调节身体和/或精神压力状况下的免疫系统,用于在肿瘤消解治疗之前,过程中和/或之后加强免疫系统,或用于减少肿瘤复发和继发的风险,其包括如权利要求1至23中任意一项要求所保护的组合物,或由该组合物组成。
25.一种包括如权利要求1至23中任意一项所要求保护的组合物,或由该组合物组成的药物,其用于加强免疫防御或机体防御,用于治疗炎症和炎性风湿性关节炎,用于调节身体和/或精神压力状况下的免疫系统,用于在肿瘤消解治疗之前,过程中和/或之后加强免疫系统,或用于减少肿瘤复发和继发的风险。
