Abstract

本发明公开了一种鞣酸苦参碱分散片及其制备工艺,它是以鞣酸苦参碱为原料加辅料聚乙烯吡咯烷酮、聚丙烯酸树脂、交联聚维酮、阿斯巴甜等制成。本发明在保持原剂型对治疗小儿腹泻,小儿细菌性痢疾及肠炎有显著的疗效的同时,可在水中迅速崩解分散,以益于小儿服用,并特别采用掩味技术,从而大大提高了小儿服用的依从性。

鞣酸苦参碱分散片及其制备工艺
技术领域:
本发明涉及一种药物分散片及其制备工艺。
技术背景:
鞣酸苦参碱片主要成分为鞣酸苦参碱,是豆科植物苦参(Sophora flavescens Ait.)的根及根茎中提取的苦参总碱与五倍子(漆树科植物盐肤木Rhus chinensis Mill.、青麸杨Rhus potaninii Maxim.、或红麸杨Rhus punjabensis Stew.Var.sinica(Diels)Rehd.etWils.叶上的虫瘿,主要由五倍子蚜Melaphis chinensis(Bell)Baker寄生而形成)提取的鞣质经反应合成而得的一种有机鞣酸弱碱盐,经口服后,在胃液的酸性条件下溶解,游离出苦参总生物碱和鞣质而共同发挥药理作用。原剂型为普通片剂,临床上用于细菌性痢疾及肠炎等,疗效确切,特别是对小儿腹泻有独特的疗效。但在临床应用过程中,因直径约为7mm的药片难以吞咽,婴幼儿服用时依从性差,在一定程度上限制本品的广泛应用。另外,因鞣酸苦参碱味苦,若直接制备成分散片分散后服用,同样存在婴幼儿服用困难的问题。
发明内容:
本发明的目的在于:在原剂型(片剂)的基础上,不改变原药品的药理作用及疗效,通过改变其辅料的成份及制备工艺,研制出鞣酸苦参碱分散片,并采用掩味技术,以解决婴幼儿服用时的依从性问题。
本发明是这样实现的:它是以鞣酸苦参碱为原料加辅料制成,以制取1000片,每片重约130mg~140mg(含苦参碱计10mg),各原料用量为:
鞣酸苦参碱   55%~65%
辅料:
1)聚乙烯吡咯烷酮(PVP)    0.7%~0.8%      (粘合剂)
2)微粉硅胶               0.6%~0.7%      (助流剂)
3)聚丙烯酸树脂(IV号)     6%~7%          (成膜材料)
4)邻苯二甲酸二乙酯       0.6%~0.8%      (增塑剂)
5)滑石粉                 0.6%~0.8%      (抗粘剂)
6)交联聚维酮(PVPP)       7%~8%          (崩解剂)
7)微晶纤维素(MCC)        7%~8%          (溶胀材料、崩解剂)
8)低取代羟丙基纤维素       3%~4%       (溶胀材料、崩解剂)
(LS-HPC)
9)阿斯巴甜(200倍)          2%~3%       (甜味剂)
10)硬脂酸镁                0.1%~0.3%   (润滑剂)
11)淀粉                    加至足量       (填充剂)
薄膜包衣剂                 4%~5.6%
其制备工艺为:取鞣酸苦参碱,以3%PVP乙醇溶液制成100目颗粒,干燥,加微粉硅胶,混匀,备用;另取聚丙烯酸树脂(IV号)溶于150ml~170ml乙醇中,加邻苯二甲酸二乙酯、滑石粉,搅匀,包衣过程中持续搅拌,以流化床包衣机包薄膜衣;包衣颗粒与交联聚维酮、微晶纤维素、低取代羟丙基纤维素、阿斯巴甜、硬脂酸镁、淀粉混匀,压片,包薄膜衣,包装,即得。
本发明由于采用了上述技术方案,在保持原剂型对治疗小儿腹泻,小儿细菌性痢疾及肠炎有显著疗效的同时,可在水中迅速崩解分散,以益于小儿服用,并特别采用掩味技术,从而大大提高了小儿服用的依从性。
具体实施方式:
本发明的实施例:以制取1000片,每片重约130mg~140mg(含苦参碱计10mg)为例:
1、药粉的前处理:取鞣酸苦参碱原料药85g,以PVP乙醇溶液制粒,用量为40ml/100g药粉左右为宜,制成100目微粒。
2、颗粒的处理:为减少包衣过程中的粘壁现象,增加药粉抗静电能力,在包衣前加入微粉硅胶0.85g作为助流剂。
3、包衣液的选择:根据鞣酸苦参碱口服后,在胃液的酸性条件下溶解,游离出苦参总生物碱和鞣质而共同发挥药理作用的特点,本发明主要采用胃溶型包衣材料,选择聚丙烯酸树脂(IV号)8.6g为包衣材料,将其溶于160ml乙醇中。
由于增塑剂可提高薄膜的柔韧性,有助于喷雾液滴在粉粒表面铺展和相互结合,有利于完整薄膜的形成。本发明用邻苯二甲酸二乙酯0.86g为增塑剂。
在包衣过程中,喷雾在微粒表面的包衣液随溶剂蒸发而变粘,包衣液中加入适量的抗粘剂可减少粘性,避免微粒之间粘连。本发明用滑石粉0.86g作为抗粘剂。
4、流化床类型的选择:本发明包衣工艺采用底喷或切喷流化床进行包衣。
5、包衣条件的选择:参照底喷流化床特点,同时考虑鞣酸苦参碱高温易变性的特性,拟定包衣条件为:主风温度45℃,物料温度25℃,辅风温度35℃,喷液雾化压力0.15Mpa,喷液速度70g/ml,包衣预热时间5min,包衣后干燥时间30min。
在上述工艺及处方条件下,制备三批鞣酸苦参碱包衣药粉,结果衣膜牢固、连续、均匀,分散性好,口尝无味。
6、崩解剂的选择:
本发明选用交联聚维酮(PVPP)10.4g、微晶纤维素(MCC)10.4g、低取代羟丙基纤维素(LS-HPC)3.9g作为崩解剂。采用外加法,以淀粉为填充剂(加至130g),硬脂酸镁0.26g为润滑剂,压片压力控制在(4.5±0.5)Kg·cm-2左右。
为了验证上述优选条件,取100片量鞣酸苦参碱包衣后药粉9.717g,按交联聚维酮(PVPP)8%、微晶纤维素(MCC)8%、低取代羟丙基纤维素(LS-HPC)3%比例分别加PVPP1.04g、MCC1.04g、LS-HPC0.39g,加淀粉至13g,压片压力控制在(4.5±0.5)Kg·cm-2左右,压制分散片,按《中国药典》2000年版二部附录6页测定崩解时间。三次实验结果分别为11s、10s、13s,说明该处方合理,结果稳定。
7、甜味剂的选择:选用阿斯巴甜为甜味剂。通过调味试验,确定阿斯巴甜(200倍)用量为3.25g。
8、分散片包衣工艺:选用天津爱勒易公司包衣液,型号为胃溶GAIBJ27G。
参照厂家提供的资料,并结合生产工艺暂定工艺参数如下:
进风温度:50±2℃    雾化压力:0.2-0.25MPaS
片床温度:34±2℃    转速:8-10rpm
片剂增重:3-5%
此条件下包衣产品外观光滑,细腻,遮盖完全,崩解符合规定。

1.一种鞣酸苦参碱分散片,它是以鞣酸苦参碱为原料加辅料制成,其特征在于:以制取1000片,每片重130mg~140mg,各原料用量为:
鞣酸苦参碱                  55%~65%
辅    料:
1)聚乙烯吡咯烷酮(PVP)       0.7%~0.8%  (粘合剂)
2)微粉硅胶                  0.6%~0.7%  (助流剂)
3)聚丙烯酸树脂(IV号)        6%~7%      (成膜材料)
4)邻苯二甲酸二乙酯          0.6%~0.8%  (增塑剂)
5)滑石粉                    0.6%~0.8%  (抗粘剂)
6)交联聚维酮(PVPP)          7%~8%      (崩解剂)
7)微晶纤维素(MCC)           7%~8%      (溶胀材料、崩解剂)
8)低取代羟丙基纤维素        3%~4%      (溶胀材料、崩解剂)
(LS-HPC)
9)阿斯巴甜(200倍)           2%~3%      (甜味剂)
10)硬脂酸镁                 0.1%~0.3%  (润滑剂)

2.一种如权利要求1所述的鞣酸苦参碱分散片的制备工艺,其特征在于:取鞣酸苦参碱,以3%PVP乙醇溶液制成100目颗粒,干燥,加微粉硅胶,混匀,备用;另取聚丙烯酸树脂(IV号)溶于150ml~170ml乙醇中,加邻苯二甲酸二乙酯、滑石粉,搅匀,包衣过程中持续搅拌,以流化床包衣机包薄膜衣;包衣颗粒与交联聚维酮、微晶纤维素、低取代羟丙基纤维素、阿斯巴甜、硬脂酸镁、淀粉混匀,压片,包薄膜衣,包装,即得。
