Abstract
本发明涉及在蜱中鉴定出的新型蛋白酶抑制剂蛋白。这些蛋白可用作疫苗组分、肥大细胞类胰蛋白酶抑制剂、用于检测肥大细胞以及分离纯化肥大细胞类胰蛋白酶。本发明还涉及控制动物和人体内由寄生物引起的疾病和损伤,而且涉及本发明蛋白在治疗某些疾病和变态反应方面的用途。
Description
本发明涉及在蜱中鉴定出的新型蛋白。这些蛋白可用作疫苗组分、肥大细胞类胰蛋白酶(此后称为MCT)抑制剂、用于检测肥大细胞以及分离纯化MCT。本发明还涉及控制寄生物引起的动物和人类疾病和损伤,以及涉及本发明蛋白在治疗疾病和变态反应中的应用。

人MCT是一种存储在肥大细胞分泌颗粒中的内切蛋白酶,一旦激活则作为肝素稳定化四聚体由肥大细胞释放。去除肝素导致类胰蛋白酶复合物解离为酶失活单体(Schwartz,1994)。
类蛋白酶是人肥大细胞颗粒的主要蛋白介导物成分,占总细胞蛋白的20%以上(Schwartz,1994)。MCT是肥大细胞的特异性标志,使肥大细胞与嗜碱性粒细胞区分开来。
肥大细胞见于许多组织,但沿着机体上皮层(如皮肤、呼吸道和胃肠道)存在的数目较多。肥大细胞通过位于小血管附近。它们参与各种生理和病理生理状态,包括急性炎症、即发型超敏反应、迟发型超敏反应、细胞生长调节、防御瘤形成以及痛觉和发痒(Liang等,1998)。肥大细胞还影响慢性炎症状态和参与神经免疫作用(Leon等,1994)。
肥大细胞类胰蛋白酶是一种重要的炎症介质。实验(主要在体外进行)表明,它在诸如哮喘、牛皮癣、间质性肺炎、类风湿性关节炎、齿龈炎和牙周炎的疾病中起重要作用。肥大细胞类胰蛋白酶还由于其激活尿激酶原和基质金属蛋白酶溶基质素原的潜能而参与肿瘤发生和血管发生。还描述了类胰蛋白酶样酶参与激活和内化致病病毒,如流感病毒、副流感病毒和人免疫缺陷病毒(Pohlig等,1996)。
小分子量物质(例如亮抑蛋白酶肽和二异丙基氟磷酸)抑制人类胰蛋白酶。二价阳离子(如钙离子)和苄脒及其衍生物是人肥大细胞类胰蛋白酶的竞争性抑制剂(Schwartz,1994)。然而,与大多数其它丝氨酸酯酶不同,人类胰蛋白酶不受典型的丝氨酸蛋白酶抑制剂(如抑蛋白酶肽和大豆胰蛋白酶抑制剂)抑制。还报道了针对肥大细胞类胰蛋白酶催化部位的内源性抑制剂。乳铁蛋白和髓过氧化物酶(均来源于嗜中性粒细胞)以及抗凝血酶III抑制人类胰蛋白酶活性,所有这些抑制剂都拮抗稳定MCT四聚体的粘多糖(肝素或硫酸软骨素)(Alter等,1990;Cregar等,1999;Elrod等,1997)。
先前已经描述了水蛭来源的人类胰蛋白酶抑制剂(LDPI)。已发现重组形式的Kazal型蛋白有效抑制四聚体类胰蛋白酶4个催化部位中的2个部位(Stubbs等,1997;Auerswald等,1994;Mühlhahn等,1994;Sommerhoff等,1994)。
由于已知MCT在哺乳动物疾病和变态反应中的重要性,因此确实需要针对MCT的高度特异性的有效抑制剂。现在在蜱物种中发现了一种能够抑制人肥大细胞类胰蛋白酶活性的新型蛋白。
发明概述
按照本发明的第一方面,提供与图1给出的蜱源性蛋白酶抑制剂蛋白(TdPI)序列具有显著序列同源性的重组蛋白、所述蛋白的活性片段或所述蛋白的功能等同物。
本文使用的术语“显著序列同源性”是指包括所有与TdPI具有共同功能并具有共同的序列同源性或在多肽序列中存在的基序之间具有共同同源性的蛋白。“显著的”整体同源性是指如果将所述同源蛋白与TdPI序列进行序列对比,则在完全保守的序列中有50%或50%以上的氨基酸为相同残基。最好使用GCG最佳拟合命令(空位生成罚分=2.5;空位延长罚分=0.5)(Genetics-Computer-Group,1994)进行序列对比。
所述蛋白完整长度上的同源性程度优选为至少60%。更优选同源性程度为至少70%,甚至更优选为75%,最优选为80%或更高。
本发明在该方面包括提供含本文鉴定为蜱源性蛋白酶抑制剂蛋白(TdPI)的序列的蛋白、其活性片段或其功能等同物。附图1给出了该序列。经鉴定,该蛋白由蜱唾液腺文库的cDNA编码。该蛋白的分子量约13.5kDa,看来属于Kunitz型蛋白酶抑制剂家族。所述序列与与该家族其它成员如抑蛋白酶肽和间α-胰蛋白酶(inter-alpha-trypsin)抑制剂的相似性低,但推定的反应中心和半胱氨酸位置在一定程度上保守。
术语“功能等同物”在本文用于描述与TdPI蛋白在抑制类胰蛋白酶或具有一个或多个可用于开发疫苗的表位方面具有相似功能的蛋白,所述疫苗针对与TdPI具有显著序列同源性的蛋白。术语“功能等同物”还指在结构上与本文鉴定的TdPI蛋白相似或含有相似或相同三级结构的分子。该术语还包括保留抑制类胰蛋白酶(优选人肥大细胞类胰蛋白酶)能力的蛋白片段。
抑制类胰蛋白酶的相似功能最好针对类胰蛋白酶(优选肥大细胞类胰蛋白酶,更优选人肥大细胞类胰蛋白酶)的催化活性,其特征在于Ki小于1μM,更优选小于100nM,甚至更优选小于20nM,甚至更优选小于10nM,最优选小于1nM,可使用如本文描述的任何标准类胰蛋白酶抑制测定评价(参见以下实施例中标题为“蛋白酶抑制测定”的部分)。
或者,或者除了具有抑制类胰蛋白酶活性之外,“功能等同物”在本文用于描述含有表位的蛋白,所述表位可用于开发针对本发明蛋白的疫苗。这样的功能等同物以及含有合适表位的片段,可用于开发针对吸血寄生物的疫苗,该疫苗针对TdPI蛋白家族成员。当然,可制备免疫原性高于或低于对应的野生型蛋白或蛋白片段的功能等同物,以便适于需要的用途。所述“野生型”是指大多数种成员特征性天然存在的基因型。如果所述蛋白用于诱导针对寄生物蛋白的宿主抗性的免疫接种方案,则可以修饰所述分子,以便增强它们的免疫原性。因此,它们更可能在接种的宿主中激发免疫应答。
本发明蛋白的功能等同物相对于野生型蛋白序列存在取代、添加、插入和/或缺失一个或多个氨基酸以及化学修饰氨基酸的取代,而不以相反的方式影响所述蛋白的功能或活性。该术语还包括天然生物变异体(例如所有产生野生型蛋白的不同物种的等位基因变异体或地理变异)。
“活性”片段是或者抑制类胰蛋白酶(优选人肥大细胞胰蛋白酶)和/或包含一个或多个可用于开发针对本发明蛋白的疫苗的表位的片段。这些生物特性如上所述。
本发明这一方面的蛋白优选得自吸血外寄生物(如蚊或水蛭)或有毒动物(如蜘蛛、蝎子或蛇)。所述蛋白更优选得自蜱,最优选得自硬蜱(Ixodid tick),如非洲扇头蜱(Rhipicephalus appendiculatus)。
按照本发明的第二方面,提供抑制类胰蛋白酶的得自吸血节肢动物外寄生物的重组蛋白、其活性片段或其功能等同物。所述重组蛋白优选得自蜱,最优选得自硬蜱(Ixodid tick),如非洲扇头蜱。这些分子在抑制类胰蛋白酶(优选肥大细胞类胰蛋白酶,更优选人肥大细胞类胰蛋白酶)催化活性方面具有活性,其特征在于Ki小于1μM,更优选小于100nM,更优选小于20nM,甚至更优选小于10nM,最优选小于1nM或更小。

可使用已知的分子生物学和蛋白质化学技术制备本发明的蛋白。可通过化学合成(一种特别用于产生衍生自全长蛋白序列的短肽的技术)制备用作免疫原的蛋白片段。
可通过在宿主细胞中表达制备重组形式的本发明的蛋白。这样的表达方法是本发明技术人员众所周知的,其中许多方法在Sambrook等,1989和Fernandez & Hoeffler,1998中有详细描述。
本发明的第三方面提供本发明的蛋白、蛋白片段和功能等同物在哺乳动物中抑制类胰蛋白酶(如肥大细胞胰蛋白酶),由此调节其活动和控制其病理作用的用途。这样的分子还可用于抑制胰蛋白酶、纤溶酶和较低程度的抑制组织激肽释放酶。
本发明还包括将上述蛋白、蛋白片段和功能等同物用作抗炎药物。这些分子优选以包含惰性载体的药用组合物提供。所述蛋白、蛋白片段或功能等同物可构成所述组合物的唯一活性组分,或者可构成完整治疗的组成部分,如局部用于昆虫、蛇或蝎子咬伤部位或皮炎皮肤的乳膏剂组分。它还可以用作乳膏剂、油剂、粉剂或丸剂中的类胰蛋白酶和类胰蛋白酶相关化合物的载体分子,以缓释结合组分。
本发明还包含本发明的蛋白、蛋白片段和功能等同物用于定量例如血液、鼻灌洗液、组织或食品中的类胰蛋白酶(优选人肥大细胞类胰蛋白酶)水平的用途。这可以是含有一种或多种本发明的蛋白、蛋白片段或功能等同物以及允许精确定量所测试样品中的类胰蛋白酶的检测材料(例如放射性标记类胰蛋白酶、抗体、酶,如碱性磷酸酶、过氧化物酶和荧光素酶)的试剂盒的一部分。该试剂盒类似于放射免疫测定试剂盒或ELISA试剂盒,其中本发明的蛋白起结合分子的作用,而不是起抗类胰蛋白酶或类胰蛋白酶相关分子的抗体的作用。本发明的一个方面包括加入本发明分子的这种试剂盒。
本发明的蛋白、蛋白片段和功能等同物还可以用于检测携带类胰蛋白酶的细胞,尤其是用于检测肥大细胞。在此检测方法中可使用本领域通用的任何技术,可包括免疫细胞化学和组织学技术,其中所述蛋白、蛋白片段或功能等同物与抗血清(如抗-TdPI抗血清)联合使用或所述分子与标记物或染料(如FITC)直接偶合。可使用完整蛋白,或者可只使用活性结合片段,以便检测底物。在另一个实施方案中,所述野生型蛋白可遗传性或合成性与另一种蛋白(如碱性磷酸酶、荧光素酶或过氧化物酶)融合,以便促进其检测。其它检测含类胰蛋白酶细胞或样品的方法可包括印迹技术(Towbin等,1979)、凝胶阻滞、亲和层析或本领域使用的任何其它合适方法。
本发明还包括使用与支持物结合的本发明蛋白、蛋白片段和功能等同物,以从例如机体组织、血液或食品中去除、纯化、分离或提取类胰蛋白酶。所述支持物包括任何合适的惰性物质,包括凝胶、磁珠和其它珠、微球、结合柱和树脂。
本发明还包括使用本发明的蛋白、蛋白片段和功能等同物作为研究涉及类胰蛋白酶的炎症、炎症相关过程或其它生理过程的工具。这些分子还可以用作进一步研究MCT自身特征和功能的工具。例如,所述分子可用于抑制或消除细胞培养物或发炎的动物组织中的类胰蛋白酶,以便研究类胰蛋白酶在这些体系中的重要性。
后生动物寄生物,尤其是节肢动物和蠕虫,也是人和兽医学中具有重要影响的感染性疾病和其它有害作用的来源。目前,控制节肢动物和蠕虫寄生物主要依赖于使用化学物质,如杀螨剂和抗蠕虫剂。已经通过使用疫苗技术在免疫控制方法应用方面进行了尝试。已经在鉴定某些为潜在疫苗侯选物的保护性抗原方面取得了一定的成功,但迄今对最引人注目的牛肺线虫胎生网尾线虫(Dictyocaulusviviparous)和牛蜱微小牛蜱(Boophilus microplus)仅有少量产生了经济效益。不管发展如何,仍然需要后生动物寄生物疫苗,尤其需要可跨越大范围的节肢动物和/或蠕虫属使用的疫苗。
因此本发明还提供本发明的蛋白、蛋白片段和功能等同物作为免疫原的用途,所述免疫原用作后生动物寄生物疫苗,特别是用作控制由节肢动物和其它后生动物寄生物引起的疾病的保护性免疫原。合适的疫苗接种侯选者包括家畜(如牛、山羊、绵羊、犬、猫)和其它需要抗后生动物寄生物(尤其是蜱)保护的动物。所述疫苗可以包含某些用作佐剂的化合物。合适的佐剂是本领域众所周知的,包括水包油乳剂、皂苷佐剂、弗氏完全佐剂(CFA)和弗氏不完全佐剂(IFA)、细胞因子和起增强所述组合物功效的免疫刺激剂作用的其它物质。
按照本发明的再一个方面,提供针对疾病或病症接种哺乳动物的方法,包括给予哺乳动物其表达与所述疾病或病症相关的本发明上述方面的蛋白、蛋白片段或功能等同物。
本发明的再一个方面提供治疗罹患疾病或病症(例如哮喘、牛皮癣、间质性肺病、类风湿性关节炎、齿龈炎、牙周炎、变态反应、癌症或任何其它类胰蛋白酶介导性疾病)的哺乳动物的方法,包括给予所述哺乳动物治疗有效量的本发明上述方面的蛋白、蛋白片段或功能等同物,任选与药学上可接受的载体联合给予。
按照本发明的再一方面,提供含本发明上述方面的蛋白、蛋白片段或功能等同物和药学上可接受的载体的免疫原性组合物。
药学上可接受的载体包括其自身不诱导产生对接受所述组合物的个体有害的抗体的任何载体。合适的载体通常为较大、缓慢代谢的大分子,如蛋白质、多糖、聚乳酸、聚羟基乙酸、多聚氨基酸、氨基酸共聚物、脂质聚合物(如油滴或脂质体)和失活病毒颗粒。这样的载体是本领域技术人员众所周知的。所述组合物可用作疫苗,并因此可选地含有免疫刺激剂(佐剂),例如以上列举的佐剂。按照本发明的再一方面,提供配制疫苗组合物的方法,包括将按照本发明上述方面的蛋白、蛋白片段或功能等同物与药学上可接受的载体、任选与佐剂组合在一起。
按照本发明的再一方面,提供含编码本发明上述方面的蛋白、蛋白片段或功能等同物的核苷酸序列的核酸分子。这样的分子包括单链或双链DNA、cDNA和RNA,以及合成核酸类别。所述核酸序列最好包含DNA。
本文利用实例公开了编码TdPI的cDNA,图1显示了其序列以及其编码的氨基酸序列(核苷酸和氨基酸以其标准的单字母缩写给出)。
本发明的核酸分子优选含有与示于图1的序列相同或互补的核苷酸序列,或者与其简并或大致同源的序列,或者在非严格条件(例如6×SSC/50%甲酰胺,室温)下与其杂交并在低严格条件(例如2×SSC、室温或2×SSC、42℃)下清洗或者更优选在高严格条件(例如2×SSC、65℃)下结合的序列。(SSC=0.15M NaCl、0.015M柠檬酸钠、pH7.2)。
在序列对比时,最好使用GCG最佳拟合命令(空位生成罚分=2.5;空位延长罚分=0.5)(Genetics-Computer-Group,1994)测定,所述核酸序列优选与编码TdPI的cDNA或者与TdPI序列具有至少60%或更高的同一性的翻译产物(或部分节段或整个翻译产物)的DNA序列具有至少60%的同一性。
本发明还包括含本发明此方面DNA序列的克隆和表达载体。这样的表达载体可加入合适的转录和翻译控制序列,例如按照读框与本发明的核酸分子连接的增强子元件、启动子-操纵子区、末端终止序列、mRNA稳定序列、起始或终止密码子或核糖体结合位点。
另外,它们可能便于使某些宿主分泌所述重组蛋白。因此,所述载体的其它元件可包括编码分泌信号序列和加工序列的核酸序列。
本发明的载体包括质粒和病毒(包括噬菌体病毒和真核生物病毒)以及其它线性或环状DNA载体,如使用转座因子或同源重组技术的载体。许多这样的载体和表达系统是众所周知的,并在本领域中有文献报道(Fernandez & Hoeffler,1998)。特别合适的病毒载体包括杆状病毒型载体、腺病毒型载体和痘苗病毒型载体。
用于重组表达的合适宿主包括通常使用的原核物种(如大肠杆菌)或可制备来高水平表达重组蛋白并可易于大量培养的真核酵母。体外培养的哺乳动物细胞系也是合适的,特别是在使用病毒驱动型表达系统时。另一个合适的表达系统是使用昆虫细胞作为宿主的杆状病毒表达系统。表达系统还可以构成其基因组中加入编码DNA的宿主细胞。蛋白或蛋白片段还可以体内表达,例如在昆虫幼虫或在哺乳动物组织中表达。
可以使用各种已知技术将本发明的载体引入到原核或真核细胞中。合适的转化或转染技术在文献(Sambrook等,1989;Ausubel等,1991;Spector,Goldman & Leinwald等,1998)中有详细介绍。在真核细胞中,表达系统可根据该系统的需要或者为瞬时(例如附加型)表达系统或者为持久(染色体整合型)表达系统。
本发明的核酸分子还可以用于产生转基因动物,尤其是啮齿动物。这可局部性通过改变体细胞或通过生殖系治疗(germ line therapy)加入可遗传改变来完成。
本发明还包括转化或转染的原核或真核宿主细胞,或含有以上定义的核酸分子的转基因生物。
本发明的再一方面提供制备如上定义的本发明蛋白、功能等同蛋白片段的方法,包括在表达所述蛋白的条件下培养含本发明核酸分子的宿主细胞并回收由此产生的所述蛋白。
现在将通过实施例,特别是参考分离自非洲扇头蜱的蛋白的实施例,更详细地描述本发明的各个方面和实施方案。当然,需要指出的是,可以对细节进行修改,而不背离本发明的范围。

实施例蜱
按照Jones等,1988所述饲养蜱。所有三个发育阶段的非洲扇头蜱都饲养在Dunkin Hartley豚鼠上。当不再喂饲时,将所有的蜱保持在21-26℃和85%的相对湿度。cDNA
含TdPI cDNA的克隆76是由λZap II(Stratagene)的非洲扇头蜱唾液腺表达文库随机挑出的几个克隆中的一个,所述文库由在DunkinHartley豚鼠上喂饲2天的蜱的mRNA构建(Paesen & Nuttall,1996)。体内切除噬菌粒,并用于在XL1-Blue细胞中产生双链pBluescript SK(-)质粒(Short等,1988)。由过夜培养物中纯化质粒(Goode & Feinstein,1992),碱变性(Mierendorf & Pfeffer,1987),然后按照Sanger &Coulson,1975测序。
测定76-3插入片段的正链和负链的完整序列。图1显示了正向引物(S1→)(相当于核苷酸209-224)、反向引物(←S2)(退火至核苷酸255-271)和质粒特异性T3(T3→)和T7(←T7)引物(以下划线表示插入片段特异性引物序列或它们的退火部位)。P1→和P2←表示RT-PCR实验中使用的引物部位。

在测序克隆76之后,通过影印噬菌斑的DNA杂交再筛选所述文库中的其它克隆(Sambrook,Fritsch & Maniatis,1989)。通过用洋地黄毒苷(Boehringer Mannheim)随机引物标记原cDNA(用EcoRI和Eco0109I由纯化质粒切除),构建使用的探针。分离并测序三个阳性克隆。重组蛋白表达

在感染Sf21细胞后60小时收集培养基,加入(NH4)2SO4(30g/100ml培养基)沉淀rTdPI。在含300mM NaCl和10%甘油的50mM磷酸钠缓冲溶液(pH8)中再溶解所述沉淀物。主要按照Janknecht等,1991所述使用Ni-NTA琼脂糖(Qiagen)柱纯化rTdPI。含300mM NaCl和10%甘油的50mM磷酸钠缓冲溶液(pH6.5)用于流洗该柱。使用200mM咪唑的75mM NaH2PO溶液洗脱组氨酸标记蛋白。使用配有HiTrap SP阳离子交换柱(Pharmacia Biotech)的BioLogic系统(Bio-Rad)经低压层析获得进一步的纯化。层析缓冲液为50mM Hepes、pH8,1小时内0-250mM NaCl线性梯度;流速1ml/分钟。Centricon 3浓缩仪(Amicon)用于浓缩洗脱液和交换缓冲液。纯化的蛋白在使用前于PBS中储存于-20℃。使用Bio-Rad蛋白测定仪和Micro BCA蛋白测定仪(Pierce)检测蛋白浓度。蛋白质电泳
按照Laemmli,1970进行十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)。
图2显示了15%SDS-聚丙烯酰胺凝胶电泳显现的、利用金属亲和层析和阳离子交换纯化的rTdPI。A道的蛋白在电泳前用PNGaseF(在凝胶上约35kDa的蛋白)处理。B道含未处理的rTdPI。分子量以kDa给出。C道含未还原的rTdPI(在上样缓冲液中不含还原剂)。未还原rTdPI道上的分子量较高一般提示通过分子间二硫键二聚化,但质谱分析法确认分子量约13,500Da,与形成二聚物相矛盾。
通过用N-糖苷酶F(PNGase F;New England BioLabs)处理rTdPI,然后进行SDS-PAGE,研究天冬酰胺连接的糖基化。PNGase F水解所有普通类型的糖基化蛋白的Asn-多糖链(Maley等,1989)。
图3显示TdPI和牛初乳胰蛋白酶抑制剂(BovCol;Cechova,1976)、(牛)抑蛋白酶肽(Creighton & Charles,1987)和大鼠组织因子途径抑制剂(TFPI-2;仅显示了第二个因子Xa-抑制性结构域;Enjyoji等,1992)的Kunitz结构域的序列对比。还包括蜱抗凝肽TAP(Waxman等,1990)的Kunitz结构域和ornithodorin中的两个结构域(ornithl和ornith2;Van de Locht等,1996)。使用GCG的“堆积(pileup)”和“prettyplot”命令,选择较低的空位和长度权重(分别为1和0.03),对TdPI和脊椎动物Kunitz结构域进行序列对比。然后主要通过引入额外空位修改序列对比,使得可以将TAP和ornithodorin结构域包括在内。如Van de Locht等,1996所报道,所述修改很大程度上是基于后一结构域与抑蛋白酶肽的序列对比。箭头表示抑蛋白酶肽结合环的P1残基。星号表示参与传统Kunitz结构域中二硫键形成的半胱氨酸。N端测序

由Sigma购买弹性蛋白酶(I型,得自猪胰腺)、α-胰凝乳蛋白酶、胰蛋白酶、凝血酶、纤溶酶、组织激肽释放酶、血浆激肽释放酶、尿激酶、抑蛋白酶肽、n-琥珀酰-Ala-Ala-Ala-对硝基苯胺(nitroanilide)、Gly-Arg-对硝基苯胺、n-α-苯甲酰基-DL-Arg-对硝基苯胺和n-苯甲酰基-Pro-Phe-Arg-对硝基苯胺。Xa因子和重组人类胰蛋白酶得自Promege,而试卤灵标记酪蛋白、大豆胰蛋白酶抑制剂、Chromozym TH和Chromozym X得自Boehringer Mannheim。
使用n-α-苯甲酰基-DL-Arg-对硝基苯胺作为显色底物以及使用含120mM NaCl的50mM HEPES pH7.6作为反应缓冲液,在96孔微量板中检测类胰蛋白酶活性。将50μl含1μl类胰蛋白酶母液(200μg/ml)的缓冲液与50μl抑制剂溶液(各种浓度)混合。于37℃温育45分钟后,加入50μl的3mM底物溶液,使用Titertek Multiskan PlusMKII平板读数仪(ICN)检测405nm吸光度的增加。
将其它蛋白酶与在总体积100μl的蛋白酶缓冲液中的各种量的蛋白酶抑制剂预温育(20分钟;37℃)。通过加入合适底物(在900μl蛋白酶缓冲液中)并检测消化程度测定残余的蛋白酶活性。如Nakamura等,1987所述,在蛋白酶缓冲液A(0.1M Tris.HCl、10%甘油、10mM CaCl2、pH8)中检测胰蛋白酶、α-胰凝乳蛋白酶和弹性蛋白酶活性;在蛋白酶缓冲液B(50mM Tris.HCl、0.1mg/ml牛血清白蛋白、150mM NaCl、1mM CaCl2、pH8)中测定纤溶酶、尿激酶、激肽释放酶、α-凝血酶和Xa因子活性。试卤灵标记的酪蛋白用作胰蛋白酶、α-胰凝乳蛋白酶和纤溶酶的底物,并检测释放的肽的量,以测定蛋白酶活性(Twining,1984)。对硝基苯胺(pNA)-底物用于弹性蛋白酶、激肽释放酶、尿激酶、α-凝血酶和Xa因子活性(分别为n-琥珀酰-Ala-Ala-Ala-pNA、n-苯甲酰基-Pro-Phe-Arg-pNA、Gly-Arg-pNA、Chromozym TH和Chromozym X);通过测定410nm吸光度的增加检测蛋白酶活性。
图4显示了一幅说明rTdPI对组织激肽释放酶的较弱抑制活性的图。显示在向激肽释放酶/抗蛋白酶样品中加入30μg底物(n-苯甲酰基-Pro-Phe-Arg-pNA)后不同时间点的410nm吸光度。每个样品(1ml终体积)使用0.5u/ml的组织激肽释放酶。实线(◆-◆)表示没有蛋白酶抑制剂时的激肽释放酶活性。使用0.75μM浓度的抑蛋白酶肽完全抑制激肽释放酶活性(●----●)。10倍高浓度的rTdPI[7.5μM(○----○)]刚刚抑制50%的激肽释放酶活性。在该实验中使用的其它rTdPI浓度为3.75μM(△----△)和0.75μM(□----□)。
图5显示了在存在逐渐增加量的rTdPI情况下,经检测试卤灵标记的酪蛋白释放的肽测定出的纤溶酶(左)和胰蛋白酶(右)活性。不存在抑制剂时的肽释放设为100%,0%活性为没有蛋白酶时的水解。空心圆表示rTdPI值。为了根据蛋白测定获得的mg/ml数据计算rTdPI单体的μmol浓度,使用12kDa的计算分子量(○----○;假定考马斯蓝未结合糖蛋白的糖组分)和质谱分析测定的(平均)分子量(13.5kDa;○-○)。对应于抑制50%纤溶酶的浓度为0.097μM(抑蛋白酶肽,△-△)、0.23μM(大豆胰蛋白酶抑制剂,■-■)、0.32μM(rTdPI单体,○-○)和0.43μM(rTdPI单体,○----○)。50%胰蛋白酶抑制值为0.024μM(△-△)、0.026μM(■-■)、0.033μM(○-○)和0.044μM(○----○)。
图6显示TdPI对重组人类胰蛋白酶(Promega)的抑制作用。将重组人类胰蛋白酶与逐渐增加量的rTdPI预温育迅速将催化活性降低至没有抑制剂时活性的约33%(V0:不存在类胰蛋白酶时检测的底物转化速率;Vi:加入抑制剂的底物转化速率)。反转录酶-聚合酶链反应(RT-PCR)
从未喂饲的成虫蜱和已在豚鼠上喂饲2、4和6天的成虫蜱切取唾液腺。每个组织样品由15对腺体组成。使用RNAce Total Pure提取试剂盒(Bioline Ltd)由这些腺体中分离总RNA,并将所获得量的1/30(相当于1个腺体)用作使用Titan单管RT-PCR系统(BoehringerMannheim)的RT-PCR(35个循环)模板。还对由喂饲2天的成虫蜱取用的肠、性腺、副性腺和马尔皮基氏管的合并RNA进行RT-PCR。使喂饲3天的幼虫和喂饲3天的若虫的整体匀浆进行相同处理步骤;每个PCR反应使用的RNA量相当于1个若虫或2个幼虫的提取物。图1下划线标示引物序列(P1和P2)。为检查RT-PCR产物是否由TdPImRNA特异性衍生,将它们的大小与使用相同引物由PCR扩增原质粒DNA获得的标记物的大小进行比较。
图7显示1.5%琼脂糖凝胶显现的由幼虫(L)和若虫(N)的整体提取物、雄性和雌性非洲扇头蜱成虫唾液腺提取物获得的RT-PCR产物。数字对应于不同时间点的成虫喂饲期;0表示取自未喂饲蜱的样品;2、4和6分别表示喂饲2、4和6天的蜱。M道显示用作分子量标记的利用相同引物组(图1)但使用TdPI cDNA代替RNA作为模板获得的目标(tee)PCR产物。

Claims (33)
1.与图1所示的蜱源性蛋白酶抑制剂蛋白(TdPI)序列具有显著序列同源性的重组蛋白、该蛋白的活性片段或该蛋白的功能等同物。
2.权利要求1的重组蛋白、蛋白片段或功能等同物,它起类胰蛋白酶、优选人肥大细胞类胰蛋白酶的抑制剂的作用。
3.权利要求1-2任一项的重组蛋白、蛋白片段或功能等同物,它含有一个或多个可用于开发疫苗的表位,所述疫苗针对与TdPI具有显著序列同源性的蛋白。
4.权利要求1的重组蛋白或蛋白片段,其中所述序列同源性的定义为:所述蛋白与图1的序列进行对比时,在完全保守的序列中有50%或50%以上的氨基酸为相同残基,序列对比结果使用GCG最佳拟合命令(空位生成罚分=2.5;空位延长罚分=0.5)获得。
