Abstract

Description
技术领域
本发明属生物工程类,主要涉及跨膜型金黄色葡萄球菌肠毒素A融合蛋白的制备及其在肿瘤治疗方面的应用。
背景技术
1989年,White等[1]提出了超抗原(Superantigen,SAg)的概念,它是具有激活大量T细胞能力的一组结构上相关的细菌和病毒的蛋白。细菌SAg作为一个完整的蛋白与主要组织相容性复合物(MHC)II类分子结合,随后与表达特异T细胞受体(TCR)Vβ序列的T细胞相互作用,导致T细胞增殖和细胞因子释放[1,2]。与普通抗原不同的是:SAg无需抗原提呈细胞(APC)的加工即可直接与细胞膜上的MHCII类分子结合,激活所有携带特异TCR Vβ片段的T细胞,激活T细胞的克隆数大约是普通抗原的1000倍,产生大量细胞因子,杀伤表达MHCII类分子的靶细胞,称之为SAg依赖的细胞介导的细胞毒作用(Superantigen-Dependent Cell-MediatedCytotoxicity,SDCC)[3,4]。
超抗原强烈激活T细胞的特性启发人们将其用于肿瘤的治疗。目前对细菌SAg的抗肿瘤作用研究较多的主要是金黄色葡萄球菌肠毒素(SEA、SEB和TSST1等)。但是,将超抗原直接用于肿瘤治疗尚存在许多限制,因为大多数肿瘤细胞并不表达MHCII类抗原;同时,表达MHCII类抗原的正常细胞如APC也可能成为超抗原的靶细胞而受到攻击。
为了充分发挥超抗原的特异性抗肿瘤作用,Dohlsten[5]等通过基因工程的方法,制备了人结肠癌单克隆抗体C215的Fab段与SEA的融合蛋白,此融合蛋白在体外将反应性的细胞毒T淋巴细胞(CTL)导向表达C215抗原的肿瘤细胞,并降低与MHCII类分子的结合亲和力。用C215Fab-SEA融合蛋白治疗B16-C215黑色素瘤的肺转移,结果能消除95%的转移灶。Rosendahl等[6]用C215Fab-SEA融合蛋白治疗荷C215转染的同基因B16黑色素瘤小鼠的肺转移,也证明了能够产生强烈的抗肿瘤作用。Sakurai[7]等构建了SEA双功能抗体的融合蛋白SEA-scFv,结果表明:该融合蛋白在肿瘤的免疫治疗中发挥了非常重要的作用。
在近年来的研究中,尽管单克隆抗体(McAb)靶向的超抗原在一系列实验肿瘤的治疗中显示出是一个极具有潜力的肿瘤生物治疗制剂,由于其本身尚存在一些不足之处,以致于限制了它的实际应用,这些缺点包括:(1)McAb靶向超抗原的抗肿瘤谱很窄,一种McAb只能靶向表达特异性抗原的肿瘤组织,这种制剂缺乏广谱性;(2)融合蛋白活化的T细胞是SEA反应性的,并非肿瘤特异性T细胞。
如何增强融合蛋白的抗肿瘤疗效,具有重要的现实意义。除了McAb靶向超抗原的抗肿瘤作用之外,另有学者用化学偶联的方法使超抗原结合在肿瘤细胞表面。Shimizu等[8]使用Sulfo-GMBS交联剂能够使SEB结合在Meth A肿瘤细胞表面,表面赋有SEB的Meth A肿瘤细胞在体外能有效地刺激Vβ8+T细胞增殖,抑制肿瘤细胞生长,用表面赋有SEB的肿瘤细胞灭活后免疫小鼠,对肿瘤细胞的攻击具有很强的保护作用。但是,化学偶联的稳定性欠佳,严重影响了其应用价值。Shrayer等[9]将SEA基因转染B16肿瘤细胞作为肿瘤疫苗,虽然作者报道取得了较好的疗效,但是SEA本身是分泌型的,直接用SEA转染的肿瘤疫苗用于治疗,将无法避免分泌型SEA所带来的全身性毒副作用。在这些背景研究的基础上,本发明克服了现阶段对超抗原用于肿瘤治疗方面的不足。
发明内容
本发明的目的是为了拓展SEA的抗肿瘤范围,构建跨膜型金黄色葡萄球菌肠毒素A(Transmembrane-SEA,TM-SEA)的原核表达系统(pET-28a-TM-SEA),制备TM-SEA融合蛋白,使SEA能够更加广泛地应用于多种肿瘤的治疗。
本发明构建的TM-SEA融合蛋白具有跨膜功能,可以将超抗原SEA锚定在肿瘤细胞膜上,并具有相当的稳定性,SEQ ID NO:1,显示本发明TM-SEA融合蛋白的核苷酸序列。
本发明的另一个目的是制备TM-SEA融合蛋白的方法:包括将分泌型的SEA蛋白与c-erb-B2基因中的一段编码TM区的序列融合,构建TM-SEA的融合蛋白。其中,TM序列来自表达c-erb-B2原癌基因的卵巢癌HO-8910细胞系;SEA基因来源于金黄色葡萄球菌(ATCC 13565)。首先从HO-8910卵巢癌细胞系中提取其总RNA,使用一对针对TM区序列特异的引物,采用RT-PCR的方法扩增出编码TM的一段序列;然后从金黄色葡萄球菌(ATCC 13565)中提取DNA,PCR扩增出SEA基因;用剪接重叠延伸法(splicing overlap extension,SOE)将TM与SEA连接,获得TM-SEA融合基因。然后将TM-SEA融合基因定向克隆到pGEM-T载体中,经过DNA限制性内切酶酶切鉴定,筛选出阳性克隆。经核苷酸序列分析证实获得了正确的TM-SEA融合基因。亚克隆构建TM-SEA的表达载体pET-28a-TM-SEA,再次测序证实了表达载体pET-28a中目的基因TM-SEA是正确的。将包含目的基因的重组表达载体pET-28a-TM-SEA的质粒转化用于融合蛋白表达的大肠杆菌BL21(DE3)pLysS,卡那霉素和氯霉素抗性LB平板筛选阳性克隆,阳性克隆扩大培养后,经过IPTG诱导5h,收获工程菌,用融合蛋白纯化试剂盒纯化获得TM-SEA融合蛋白;并通过一系列的体外和体内实验证实了TM-SEA融合蛋白的跨膜功能以及刺激T细胞增殖的功能。
本发明构建的TM-SEA融合蛋白可制备肿瘤疫苗,可有效激发机体的特异性和非特异性抗肿瘤免疫,具有对荷黑色素瘤小鼠的免疫治疗作用,以及具有对亲本肿瘤细胞攻击的免疫保护作用。可用于肿瘤复发和微转移的治疗,及广泛地应用于多种肿瘤的治疗。
本发明的特点是(1)TM-SEA融合蛋白具有跨膜功能,可以将超抗原SEA锚定在肿瘤细胞膜上,并具有相当的稳定性;同时保持了SEA的生活学活性,具有刺激人外周血淋巴细胞和小鼠脾细胞增殖的功能。(2)体内研究表明TM-SEA融合蛋白制备的肿瘤疫苗可有效激发机体的特异性和非特异性抗肿瘤免疫,能显著抑制荷瘤小鼠肿瘤生长,延长其生存期,其脾细胞的NK和CTL活性显著增强。(3)TM-SEA融合蛋白制备的肿瘤疫苗可诱导特异性抗肿瘤反应,对同种肿瘤细胞攻击可产生较强的免疫保护作用:动物成瘤时间延迟,肿瘤生长缓慢,生存期显著延长,部分动物无瘤生存,其脾细胞的CTL活性增强。该肿瘤疫苗有可能作为一种有效的方法用于肿瘤复发和微转移的治疗。

下面结合实施例和附图对本发明作进一步的说明,而非限定本发明。
本实验中所有相关数据均采用SPSS软件包进行统计学分析,生存率比较采用Log-rank检验,其它计量资料采用方差分析(F检验)和两两比较q检验。实施例一 TM-SEA融合基因的克隆与表达载体的构建(一)超抗原SEA基因的克隆与表达载体的构建1.扩增SEA基因产物的电泳分析
扩增的SEA基因大小为795bp,扩增产物的电泳结果见附图1。2.重组pGEM-T-SEA质粒DNA的酶切鉴定
重组pGEM-T-SEA质粒DNA用Nhe I和Hind III双酶切,酶切产物用1.7%的琼脂糖凝胶电泳,目的基因大小为784bp,结果见附图1。3.扩增SEA基因的序列分析
除扩增片段两端的限制性内切酶识别序列及保护碱基序列外,测序结果与Genbank中的标准序列完全一致[10],见附图4。4.重组pET-28b-SEA质粒DNA的酶切鉴定
将亚克隆后的重组pET-28b-SEA质粒用Nhe I和Hind III内切酶进行双酶切,酶切产物的凝胶电泳结果见附图1。目的基因大小为784bp。

(二)跨膜型超抗原TM-SEA基因的克隆与表达载体的构建
1.扩增SEA基因产物的电泳分析
以金黄色葡萄球菌(ATCC 13565)的DNA为模板,用引物P1和P2,PCR扩增出用于和TM片段连接的SEA基因片段,大小为796bp,见附图3。
2.扩增编码TM序列片段产物的电泳分析
以HO-8910细胞系RNA的逆转录产物cDNA为模板,用引物P3和P4扩增出编码TM序列的片段,大小为155bp,电泳结果见附图2。
3.扩增TM-SEA融合基因PCR产物的电泳鉴定
以扩增SEA和编码TM序列的PCR产物为模板,用引物P1和P4扩增出跨膜型超抗原TM-SEA融合基因,其大小为922bp,电泳结果见附图3。同时,对融合基因TM-SEA进行克隆和亚克隆后,TM-SEA重组质粒的双酶切产物也一并电泳,结果见附图3。

6.3本研究克隆的TM-SEA融合基因的序列
通过ABI PRISM 377全自动核酸测序仪(PE,USA)测序,本研究克隆的TM-SEA融合基因的序列与Genbank[10,11]中的标准序列完全一致(见测序图:附图4)。
实施例二:跨膜型超抗原SEA融合蛋白的表达与纯化
(一)PTG诱导TM-SEA融合蛋白表达的时效Western Blot分析
制备诱导前及诱导后1h、3h、5h和7h的蛋白样本,其Western Blot分析结果见图5。将图片扫描后,经密度分析确定,诱导后5h时,目的蛋白的表达产量最高。从图5中可以看出:空载体转化的工程菌在诱导后无目的蛋白表达,目的蛋白的产量在IPTG诱导后1h,3h,5h逐渐增加;而没有诱导的工程菌呈现微量的蛋白表达。图5中,泳道1,阳性对照ppSEA;泳道2,空载体转化的工程菌;泳道3,诱导前样本对照;泳道4,诱导后1h的样本;泳道5,诱导后3h的样本;泳道6,诱导后5h的样本;泳道7,诱导后7h的样本。
(二)PTG诱导超抗原SEA蛋白表达的Western Blot分析
同样地,取诱导后1h、3h、5h的蛋白样本。Western Blot分析的结果见图6。从图6中可以看出:目的蛋白的产量在IPTG诱导后1h,3h,5h逐渐增加;将图片扫描后,经密度分析确定:在诱导后3h,目的蛋白的表达产量最高。图6中,泳道1,阳性对照(ppSEA);泳道2,诱导后1h;泳道3,诱导后3h;泳道4,诱导后5h。
(三)目的蛋白的纯化
IPTG诱导5h后的工程菌,按照纯化试剂盒的说明书,对目的蛋白进行分离纯化,纯化后的目的蛋白的纯度在95%以上,可直接用于体内、外实验研究。纯化蛋白样本的Western Blot分析(参见图7)。图7中,泳道1,阳性对照ppSEA;泳道2,工程菌裂解后的上清;泳道3,纯化过程中未结合的蛋白;泳道4,纯化后的TM-SEA融合蛋白。
实施例三:跨膜型超抗原SEA融合蛋白的功能研究
(一)TM-SEA融合蛋白的体外功能研究(In vitro)
1.TM-SEA融合蛋白对肿瘤细胞膜的锚定作用
(1)免疫组化法(Immunohistochemistry)
纯化的TM-SEA融合蛋白与肿瘤细胞共同孵育4h后,用免疫组化方法可以检测到肿瘤细胞膜上SEA蛋白的染色(见图8A);在对照组样本(图8B)中,用纯化的SEA蛋白同样与肿瘤细胞Colo-205共同孵育4h后,免疫组化结果未见细胞膜上有棕黄色蛋白抗原颗粒存在。图8中,A是TM-SEA融合蛋白锚定在肿瘤细胞膜上的阳性染色结果,B是相关对照。
(2)免疫荧光法(Immune Fluorescence)
0.3μM的TM-SEA融合蛋白与B16细胞共同孵育4h。分别加入一抗和二抗作用后,在倒置荧光显微镜下观察到细胞膜表面存在显示细胞轮廓的荧光(见图9)。锚定阳性细胞的百分率达92%以上。同时,检测了此融合蛋白对肿瘤细胞膜锚定的稳定性,结果表明锚定(共同孵育)结束后4h,阳性细胞的百分率为83%,并未显著减少。
(3)流式细胞术(Flow Cytometric Analysis)
流式细胞仪对TM-SEA融合蛋白对肿瘤细胞膜的锚定作用的检测结果见图10a、10b、10c。对照样本(图10a)细胞的平均荧光强度为2.16;阳性细胞数(本底)的百分率为1.6%(图10b)的平均荧光强度为13.1;阳性细胞数百分率为94.9%;实验样本2(图10c)的平均荧光强度为12.0;阳性细胞数为87%。锚定后4h样本的荧光强度(12.0)较正常制备的阳性样本的荧光强度(13.1)略低,表明TM-SEA融合蛋白与B16肿瘤细胞孵育4h后,能够锚定在肿瘤细胞膜上,而且这种锚定具有相当的稳定性。
(二)TM-SEA融合蛋白对人PBLs及C57BL/6小鼠脾细胞的活化作用
1.TM-SEA融合蛋白对人PBLs的增殖诱导活性
PHA-P(10μg/ml)在作用48h时,活化人PBLs的PI值为1.13;TM-SEA蛋白在3×10-5nM浓度时,已经表现出对人PBLs的增殖诱导能力(PI为1.19);在3×10-5nM~3×10-1nM浓度范围内,TM-SEA蛋白刺激细胞增殖的作用呈剂量依赖性;其中,3×10-1nM浓度的TM-SEA蛋白,在作用48h时的细胞增殖能力最强(PI为1.74),而且显著高于最佳剂量的阳性对照PHA-P(P<0.05)。纯化的SEA蛋白对人PBLs的刺激作用的结果表明,TM-SEA融合蛋白具有与SEA蛋白类似的刺激作用。然而,当两种蛋白的终浓度达到3nM以上时,刺激PBLs增殖的作用开始降低。结果见图11。同时,也比较了作用24h、72h的刺激增殖作用的结果(未显示),但以作用48h时的效果最佳。
2.TM-SEA融合蛋白对C57BL/6小鼠脾细胞的增殖诱导活性
Con A(20U/ml)在作用48h时,活化C57BL/6小鼠脾细胞的PI值为1.95;TM-SEA融合蛋白在4.5×10-4nM浓度时,已经表现出对脾细胞的增殖诱导能力(PI为1.52);在4.5×10-4nM-4.5nM浓度范围内,TM-SEA融合蛋白刺激小鼠脾细胞增殖的作用呈剂量依赖性正相关;其中,4.5nM浓度的TM-SEA融合蛋白,在作用48h时的细胞增殖能力最强(PI为3.28),而且显著高于最佳剂量的阳性对照Con A(P<0.05)。纯化的SEA蛋白对小鼠脾细胞刺激作用的结果表明:SEA蛋白具有与TM-SEA融合蛋白类似的刺激作用。当融合蛋白的终浓度达到4.5nM以上时,刺激小鼠脾细胞增殖的作用开始减弱。结果见图12。同时,本实验也比较了作用24h、72h的刺激小鼠脾细胞增殖作用的结果(未显示),但以作用48h的效果最为显著。
3.锚定在肿瘤细胞膜上的TM-SEA融合蛋白对人PBLs及C57BL/6小鼠脾细胞的活化作用
3.1锚定在Colo-205细胞膜上的TM-SEA融合蛋白对人PBLs的活化作用
与不用融合蛋白孵育的结果(Colo-205+PBLs)比较,Colo-205细胞膜上锚定的TM-SEA融合蛋白能显著性地诱导人PBLs增殖(P<0.01),Colo-205肿瘤细胞膜上结合的TM-SEA融合蛋白对人PBLs的活化作用(见图13)。
3.2锚定在B16细胞膜上的TM-SEA融合蛋白对C57BL/6小鼠脾细胞的活化作用
与对照组(无TM-SEA融合蛋白孵育的B16 cells+Splenocytes)比较,B16细胞膜上结合的TM-SEA能够显著性地刺激小鼠脾细胞的增殖(P<0.01)。结果表明B16肿瘤细胞膜上结合的TM-SEA融合蛋白在体外能诱导人小鼠脾细胞增殖(见图14)。
(三)TM-SEA融合蛋白的体内功能研究(In vivo)
1.肿瘤疫苗对荷瘤小鼠的治疗作用
1.1肿瘤疫苗对荷瘤小鼠肿瘤生长的抑制作用
于C57BL/6小鼠的右后肢皮下注射1×105个野生型B16细胞。5~7天后,发现有可触及的肿瘤。分别给予PBS、B16肿瘤疫苗、B16-SEA肿瘤疫苗、B16-TM-SEA肿瘤疫苗瘤体内注射,三天一次,共4次。24天时,B16-TM-SEA肿瘤疫苗治疗组小鼠的平均肿瘤直径显著性小于PBS组、B16和B16-SEA肿瘤疫苗组(见图15,P<0.01);B16瘤苗组和B16-SEA瘤苗组小鼠的平均肿瘤大小较PBS组小,但其间差异无显著性(见图15)。
1.2荷瘤小鼠脾细胞中的NK活性
采用LDH释放法对各种不同治疗组的(荷瘤)小鼠的脾细胞进行NK活性检测。靶细胞为YAC-1,效应细胞与靶细胞的比例分别为25∶1,50∶1和100∶1。图16表明:与对照组(PBS)相比,B16-TM-SEA肿瘤疫苗组小鼠的NK活性显著性增高(P<0.05);而B16和B16-SEA肿瘤疫苗组小鼠的NK活性无显著性改变(P>0.05)。1.3荷瘤组小鼠脾细胞的CTL活性
治疗结束后5天,分别选取PBS组、B16肿瘤疫苗组、B16-SEA肿瘤疫苗组、B16-TM-SEA肿瘤疫苗组的小鼠各3只,常规制备其脾细胞,在重组小鼠IL-2存在的情况下,与B16细胞共同孵育7天,以诱导CTL产生,LDH释放法测定CTL的活性。
与PBS组、B16疫苗组及B16-SEA疫苗组比较,B16-TM-SEA疫苗组小鼠脾细胞的CTL活性显著升高(见图17,P<0.05);而且当E∶T=50∶1时,CTL活性升高最为显著。而B16疫苗组和B16-SEA疫苗组小鼠脾细胞的CTL活性较PBS组略高,但差异无显著性(P>0.05)。
1.4荷瘤小鼠的生存期
除了用于检测NK和CTL活性的3只小鼠之外,对剩余的5只小鼠,观察其生存期。结果表明:B16-TM-SEA肿瘤疫苗治疗组小鼠较B16、B16-SEA肿瘤疫苗治疗组和PBS组小鼠的生存期显著性延长(见图18,P<0.01)。荷瘤35天后,B16-TM-SEA肿瘤疫苗组小鼠的生存率为80%;而其它组小鼠的生存率为0。
2.肿瘤疫苗对肿瘤生长的免疫保护作用
2.1各免疫组小鼠对肿瘤细胞攻击后的肿瘤生长曲线
用野生型B16细胞攻击的B16-TM-SEA疫苗免疫组小鼠与B16疫苗免疫组、B16-SEA疫苗免疫组、PBS组小鼠,以及与EL4细胞攻击的B16-TM-SEA肿瘤疫苗免疫组比较,其肿瘤显著性缩小(结果见图19,P<0.01)。与PBS对照组和用EL4攻击的B6-TM-SEA肿瘤疫苗免疫组小鼠相比,B16肿瘤疫苗免疫组和B16-SEA肿瘤疫苗免疫组的肿瘤存在不同程度的缩小(P<0.05)。结果表明,使用B16-TM-SEA肿瘤疫苗免疫的C57BL/6小鼠,可以免受同种肿瘤细胞的攻击,获得较强的特异性免疫保护作用(结果见图19)。
2.2免疫小鼠脾细胞的CTL活性
免疫结束后5天,分别选取PBS组,B16肿瘤疫苗组,B16-SEA肿瘤疫苗组和B16-TM-SEA肿瘤疫苗组的小鼠各3只,常规制备其脾细胞,在重组小鼠IL-2存在的情况下,与B16细胞共同孵育7天,以诱导CTL产生,LDH释放法测定CTL的活性。
与其它三组比较,B16-TM-SEA肿瘤疫苗免疫组小鼠脾细胞的CTL活性显著升高(见图20,P<0.01=;而且当E∶T=50∶1时,CTL活性升高最为显著。B16肿瘤疫苗组和B16-SEA肿瘤疫苗组小鼠脾细胞的CTL活性与PBS组比较,也有明显升高(见图20,P<0.05)。
2.3免疫组小鼠的生存期
每组剩余的5只小鼠,用于观察其生存期。结果表明:用B16-TM-SEA肿瘤疫苗免疫的小鼠,当用野生型B16细胞攻击时,此组小鼠的生存期较PBS组和用EL4肿瘤细胞攻击组的小鼠的生存期显著性地延长(见图21,P<0.01)有40%的小鼠无瘤生存。

Claims (4)
1.跨膜型金黄色葡萄球菌肠毒素A融合蛋白的制备及其应用,其特征是:构建了跨膜型金黄色葡萄球菌肠毒素A(Transmembrane-SEA,TM-SEA)的原核表达系统(pET-28a-TM-SEA),制备TM-SEA融合蛋白,为了拓展SEA的抗肿瘤范围,使SEA能够更加广泛地应用于多种肿瘤的治疗,SEQ ID NO:1,显示本发明TM-SEA融合蛋白的核苷酸序列。
2.如权利要求1所述的制备本发明TM-SEA融合蛋白的方法,其特征是:本发明中TM序列来自表达c-erb-B2原癌基因的卵巢癌HO-8910细胞系,SEA基因来源于金黄色葡萄球菌(ATCC 13565),剪接重叠延伸法(splicing overlapextension,SOE)将TM片段与SEA基因连接后,分别进行克隆和亚克隆,经酶切鉴定及核苷酸测序证实,获得重组的表达载体pET-28a-TM-SEA,经过诱导产生TM-SEA融合蛋白。
3.如权利要求1所述的TM-SEA融合蛋白,其特征是:用此融合蛋白可制备成肿瘤疫苗,可有效激发机体的特异性和非特异性抗肿瘤免疫,用于荷黑色素瘤小鼠的免疫治疗,及对亲本肿瘤细胞攻击的免疫保护作用等。
