Abstract
本发明公开了与人肺癌变相关的基因。与人肺癌变相关的基因,由序列表中序列1-12的抑癌基因群及序列表中序列13-26的促癌基因群组成。本发明利用临床低分化肺鳞癌组织以及癌旁支气管上皮组织,构建了低分化肺鳞癌差别基因表达的抑制性消减杂交cDNA文库,并从中筛选出与肺癌病变相关的新基因,对于寻找参与肺癌恶性转化、肺癌诊断及其它肿瘤相关基因的发掘具有重要意义,同时也为肺癌发病机理研究及药物靶标的寻找奠定了基础。

技术领域
本发明涉及与人类脏器癌变相关的基因,特别是与人肺癌变相关的基因。
背景技术
人肺癌变是一个涉及多种基因及其产物相互作用的复杂过程。寻求人肺癌发生发展过程的关键基因,将有助于探讨其癌变的内在分子生物学机制,并将对肺癌的预防、诊断和治疗产生重要的意义。人肺癌变关键基因的寻求不仅依赖于对现有已知基因功能研究的不断深化,还依赖于对其相关未知新基因的探索。人类基因组草图表明,人体大约有3-4万个基因。就现有肺癌相关基因的数量来说,对于科研和应用还是不够的,探索更多人肺癌变相关基因仍是我们目前亟需解决的问题,将为我们最终获得肺癌变相关的关键基因提供基础。
发明内容
本发明的目的是提供与人肺癌病变相关的基因。
本发明提供的与人肺癌病变相关的基因,是序列表中序列2的核苷酸序列。
本发明利用临床低分化肺鳞癌组织以及癌旁支气管上皮组织,构建了低分化肺鳞癌差别基因表达的抑制性消减杂交cDNA文库,并从中筛选出与肺癌病变相关的新基因,对于寻找参与肺癌恶性转化、肺癌诊断及其它肿瘤相关基因的发掘具有重要意义,同时也为肺癌发病机理研究及药物靶标的寻找奠定了基础。
具体实施方式
实施例1、细胞总RNA的提取
细胞总RNA的提取包括以下步骤:
1、低分化肺鳞癌肿瘤组织,通过微切割方法,取肿瘤纯度大于85%的标本部分进行总RNA提取;
2、从癌旁支气管上皮刮取鳞状上皮细胞,取细胞纯度大于85%的标本进行总RNA的提取;
3、总RNA提取方法用Trizol(Cat#155096-026,美国Gibico公司)试剂,按试剂说明书进行。
实施例2、抑制性消减杂交
按SMARTTM cDNA Synthesis Kit(Cat# k1052-1,美国CLONTECH公司)和ClontechPCR-SelectTM cDNA Subtractive Kit(Cat# k1804-1,美国CLONTECH公司)说明书进行操作。
获得抑癌基因克隆的消减方案为:以低分化肺鳞癌组织标本的cDNA为驱动方,癌旁支气管上皮细胞的cDNA为检测方,进行消减杂交。
获得促癌基因的消减方案为:以癌旁支气管上皮细胞的cDNA为驱动方,低分化肺鳞癌组织标本的cDNA为检测方,进行消减杂交。
实施例3、消减cDNA文库的构建(T/A载体克隆法)
经过常规PCR扩增的差减基因产物进行纯化后,分别取出3μl,与1μlpGEM-T载体连接,4℃过夜。从连接载体中取4μl转化DH5α受体细菌,接种于X-gal/IPTG/Ampcilin琼脂平板上培养过夜,挑取单菌落于加有1mlLB培养基的96孔板中,37℃振摇培养过夜。吸取80μl菌液,加20μl甘油,混匀,-70℃保存。其余用于测序。
实施例4、文库的测序
文库的克隆经扩增后提取质粒,采用pUC/M13下游引物、Megabase测序仪对插入片段进行单向测序,共获得883个核苷酸片段的序列。
实施例5、新基因的获得
将测序所得序列采用BLAST(http://www.ncbi.nlm.nih.giv/blast/)方法与美国国家生物技术信息中心(National Centre for Biotechnology Institute,NCBI)已知功能核酸库(Nr库)进行比较,确定新基因序列,它们是序列表中序列1-12的抑癌基因群及序列表中序列13-26的促癌基因群
实施例6、本发明抑癌基因群在肺癌变中的作用
将抑癌基因群的每个基因分别构建表达载体并转染肺癌细胞系,并以转染空表达载体的肺癌细胞系做对照进行研究,绘制生长曲线表明,这些基因的表达都具有抑制生长的特性;进行软琼脂实验,计算克隆形成率,这些基因的表达能够明显不同程度地降低克隆形成率;将转染抑癌基因群基因的肺癌细胞分别接种裸鼠,与转染空载体的肺癌细胞相比,这些基因抑瘤效应显著。
实施例7、本发明促癌基因群在肺癌变中的作用
将促癌基因群的每个基因分别构建表达载体并转染永生化支气管上皮细胞系,并以转染空表达载体的永生化支气管上皮细胞系做对照进行研究,绘制生长曲线表明,每个促癌基因群的基因都具有促进生长的特性;软琼脂实验表明,克隆形成率明显增高;将转染抑癌基因群基因的肺癌细胞分别接种裸鼠,与转染空载体的肺癌细胞相比,这些基因的致瘤效应显著,而相应的转染空载体的肺癌细胞不致瘤。
