Abstract

Description
本发明涉及用于蛋白或多肽的细胞内转运及某些表位的膜呈递的物质及其应用。
逆向转运可定义为细胞膜分子向内质网(RE)的行进,有时经过高尔基体。在几类羧基末端带有四肽KDEL(或在酵母中为HDEL)的内质网蛋白中证明了这种机制。生化和形态学证据都表明这些蛋白离开内质网、到达高尔基体,在这里进行糖链的修饰,然后再转向内质网。四肽KDEL是一种保留信号,它能捕捉在内质网中与其结合的肽或蛋白,如Lewis M.J.等在《自然》(Nature),348,(6297):162:3,1990年11月8日中所述,这种捕捉是通过与基元KEDL的受体蛋白相互作用而发生的。
证明存在细胞内逆向转运的另一些证据来自对某些细菌毒素的研究,它们在通过内质网后进入真核细胞的胞质中(Pelham等,1992,细胞生物学趋势(Trends cell.Biol.),2:183-185)。经研究的一个具体实例是痢疾志贺氏菌的志贺菌毒素(Shiga toxin)以及大肠杆菌的志贺菌型毒素。这些毒素是两条多肽链的复合物,一条(A片段)是毒性片段并具有脱腺苷酶活性,它通过对28S核糖体RNA的作用抑制蛋白质的合成,另一个亚单位(B片段)使毒素与其靶标偶合(O’Brien等(1992),Curr.Top.Microbiol.Immunol.180:65-94)。电子显微镜研究证明在A431细胞、Vero细胞、特别是Dandi细胞中可检测到志贺菌毒素(Sandvig等,1992和1994;KHINE,1994)。另外,用一种真菌代谢物(其可造成高尔基体结构的丧失)处理细胞可保护细胞抗志贺菌毒素,这也提示这些毒素在到达内质网之前通过了高尔基体。Kim等(1996)最后证实这种毒素的B片段定位于高尔基体中。
在下列文献中给出了对逆向转运、特别是志贺菌毒素B片段在内质网中转运的认识状态:Sandvig等(1992),自然,358:510-512,Sandvig等(1994),细胞生物学杂志(J.Cell.Biol.)126:53-64;Kim等(1996),细胞生物学杂志134:1387-1399。
现将细胞内转运定义为细胞不同区室间交换的总和。
本申请的作者发现,B片段不但向内质网行进,而且向造血细胞系、特别是树状细胞和巨噬细胞的细胞核中行进。
作者证明,将这些细胞在2微摩尔B片段-gly-KDEL如下文所述保温3小时、然后固定,在这些细胞的核中、甚至在核仁中呈现抗该毒素的特异性抗体(结果未公开),这清楚地表明存在该片段的细胞内转运。
本发明源自对志贺菌毒素B片段(B片段)细胞内转运的这些观察结果,并利用其行进特性构建含有下列元件的嵌合多肽:
-与该片段或其任何功能等价物连接的目的治疗肽或多肽,或
-带有欲表达序列的多核苷酸序列。B片段和多核苷酸序列的连接通过本领域技术人员已知的任何技术、特别是Allinquant B.等在《细胞生物学杂志(Journal of Cell Biology),1288(5):919-27,(1995)中描述的技术进行。
除DNA分子或其他分子与B片段的共价连接外,这种连接可以借助于强的非共价相互作用来实现。例如,B片段的cDNA与链亲和素或亲和素的任何衍生物的cDNA融合,按已公开的方法进行(Johannes等(1987),生物化学杂志(J.Biol.Chem),272:19544-19561)。
可将融合产生的蛋白(B-链亲和素)与利用生物素化引物经PCR或用任何其他生物素化物质获得的生物素化DNA反应。形成的复合物存在于靶细胞中并将如细胞内B片段那样被转运。
另一种连接方法求助于B片段的位点特异性生物素化步骤。为此,将B片段的cDNA与编码酶BirA识别位点的cDNA融合(Boer等(1995),细菌学杂志(J.Bacterial),177:2572-2575;Saou等(1996),基因(Gene),169:59-64)。体外生素化后,通过链亲和素或亲和素的任何其他四价衍生物的作用B片段与生物素化的其他分子连接(如cDNA,见上文)。
功能等价物应理解为通过突变、缺失或添加而衍生自B片段并具有B片段的行进特性的任何序列。
广义地讲,功能等价物可由任何具有如对B片段所述的逆向转运特性、甚至到达细胞核的细胞内转运特性的片段构成。例如,可举出美国国家科学院院报(Proceedings of the National Academy of Science ofthe United States of America),84(13);4364-8(1987年7月)中描述的Vero细菌毒素的B片段,或Lamb F.I.等在《欧洲生物化学杂志》(European Journal of Biochemistry),148(2):265-70(1995)中描述的蓖麻蛋白的B片段。在描述了这些片段转运的具体特性后,本领域技术人员将会选择最好的这种片段作为将任何序列引入任何细胞腔室的载体。
因此,本发明包括以下片段的应用:志贺菌毒素的B片段、或具有相似活性的细菌毒素的任何其他亚单位,特别是具有与B片段类似的行进特性的那些,其中包括模拟志贺菌毒素B片段的多肽。这些多肽、更广地讲这些功能类似物可通过筛选方法鉴定,筛选方法的共同原则是检测随机多肽序列和受体Gb3或该受体可溶类似物间的相互作用。例如,可利用表达随机多肽序列的噬菌体文库,用于在载有Gb3的亲和柱上筛选,或在与Gb3的可溶性放射活性类似物杂交后进行筛选。糖脂Gb3被鉴定为志贺菌毒素的细胞受体(Lingwood(1993),Adv.Lipid Res.25:189-211)。Gb3被对所述毒素敏感的细胞表达,该毒素的内在化借助于与Gb3的相互作用进行。本申请的发明人证明,在抑制了受体Gb3表达的Hela细胞中(图1)A),内在化的B片段不转运至高尔基体,而在胞浆中泡囊结构水平积累,主要由溶酶体代表。在对照细胞中,B片段被转运至高尔基体(图1)B)。
该假说已通过生化实验证实(图2),根据该假说,无Gb3时,B片段不再向生物合成或分泌的系统转运。
发明人已证明,在受体Gb3的合成抑制剂PPNP存在下(+PPNP),内在化B片段的多达50%被以可溶性TCA物质的形式降解,这与向外部降解腔室如内体或溶酶体的转运活性一致。由于受体Gb3的合成不被抑制(-PNPP),内在化B片段变为可溶性TCA的比例要小得多。由此可以得出,受体Gb3对于将B片段投送至特定腔室是必需的,这支持这样的观点:B片段活性的优势方面是与受体Gb3结合。
本发明的主题是嵌合的多肽序列,这些序列至少含有:志贺菌毒素B片段或其功能性等价物,和与其羧基末端连接的一个或多个多肽X,相应于下式:

本发明的嵌合分子另外可包含:

结构B-X或B-X’的所有嵌合序列的共同点是含有B片段或其功能等价物。
本发明的嵌合分子使得可以将序列X或X’的表达产物引入内质网中。当X与B片段连接时,逆向转运中经过高尔基体并可能经过内体。另外,在某些情况下,本发明的分子可进行成熟化,导致含于嵌合多肽序列中的某些表位的膜呈递。
成熟化应理解为从给定的多肽产生其本身可在某细胞腔室(包括胞浆)中存在的肽的任何过程。可通过在内质网中的酶切或通过在胞浆中的转运进行这种成熟化,在所述转运中多肽被切割,然后所得肽被转运到内质网中。
在这种切割后,I类主要组织相容性复合物分子(MHC cl I)可装载目标多肽分子X或X’,并呈递在细胞膜上。
当本发明的嵌合分子由B片段或其等价物与多核苷酸分子或含有欲表达序列的表达载体偶联所构成时,在细胞核中转录、然后在胞浆中翻译后所合成的多肽可经历上述嵌合多肽序列同样的切割、成熟化和细胞内转运的阶段。
本发明的嵌合多肽分子可构成用于免疫治疗的药物组合物的活性成分,其机制在适于产生免疫反应之抗原的呈递方面与生物过程相似。片段X因而代表寻求细胞表面膜呈递的一个或多个表位。片段X的长度只受相关嵌合分子的细胞内转运能力的限制。
这一途径可设想用作抗感染或抗癌的免疫治疗,也可在某些自身免疫病中用作抗原诱导物。

在本发明的结构中,X或X’的表达产物还可以代表可使由某种原因扰乱的细胞内转运得以恢复的多肽序列。例如,由于突变、缺失或添加一种序列的修饰,可将一种生物分子捕获在内质网中,具有阻断该分子的成熟或转移的作用。例如对于突变体CFTR(ΔF508)就是如此,其与伴侣分子(如钙联接蛋白)的结合被修饰而阻碍或延迟其扩增(relargage),从而阻碍其细胞内转运。这种突变是粘稠物阻塞症的病因。在内质网中引入未突变的复本可使与钙联接蛋白作用位点的糖蛋白CFTR(ΔF508)的N-糖基化链移动,具有使该蛋白重新转运至浆膜的效果,并使肺的上皮细胞有正常功能。
本发明还涉及含有编码嵌合蛋白之核酸序列的核酸构建物,特别是DNA或cDNA,其中嵌合蛋白的结构和不同变化形式定义如上。更具体地讲,本发明涉及带有如上构建物并适于在细菌培养物中表达的表达载体或质粒。例如,表达载体可以是G.F.Su等(1992),Infect.Immum.,60:3345-59中描述的质粒pSU108。
更具体地讲,本发明涉及一种构建物,其含有:
-编码B片段的序列,
-编码一种或多种欲表达多肽的序列。这可以是在细胞表面寻求膜表达的表位;这还可以是可将蛋白保留在高尔基体中的多肽;最后这还可以是可恢复细胞内受扰的转运功能的多肽。
该构建物另外可含有编码在待用本发明分子处理之目标细胞中存在时允许正确细胞内转运的多肽的任何核酸序列。这特别可为:
-编码N-糖基化信号的序列,
-编码KDEL保留信号的序列。
本发明的多核苷酸结构处于启动子的控制之下,优选使得在所转染细菌中有适当表达率的强启动子。
本发明还涉及含有这些构建物并适于产生本发明嵌合蛋白或多肽的转染的细菌。
用本发明的分子处理的宿主细胞也构成本发明的一部分;这可以是任何类型的细胞,特别是:
-可被离体处理的细胞,如细胞免疫激发中的活性免疫系统细胞,例如树状细胞、巨噬细胞、或B淋巴细胞;
-可被原地处理的细胞,如用于恢复已变异功能(由于遗传缺陷或由于代谢紊乱)的上皮细胞;
-癌细胞。
一般地讲,本发明的嵌合分子使得接近一种新的治疗方法,其解决了与用于在动物细胞中整合或表达外源分子的常规病毒或逆转录病毒载体有关的问题。这种治疗方法,如源自本发明分子的方法,包括患者细胞的直接处理,可以在离体条件下进行,也可以是嵌合多肽序列的立体定位型直接应用,或按粘膜治疗的常规方法进行,如气雾剂。
本发明涉及本发明的嵌合多肽或编码本发明多肽的多核苷酸序列在制备治疗性组合物中的用途,其中特定的多肽在靶细胞的膜水平表达。这些多肽最好是这样的表位:希望产生对抗它的免疫反应,并且它被呈递在免疫系统细胞、特别是树状细胞、巨噬细胞或B淋巴细胞的表面。志贺菌毒素的B片段作为可被抗原呈递细胞呈递之表位的载体。
本发明涉及一种免疫治疗方法,即当不希望的抗原出现在机体中后提高细胞免疫力,该方法包括使细胞免疫的关键细胞如树状细胞和巨噬细胞表达特定的表位。本发明的治疗方法旨在通过目的表位限于靶细胞中后载于MHC cl I或cl II分子,从而激发细胞和体液的调节性免疫。这导致在遇到欲消除的抗原时细胞毒性T细胞的激活。
由本发明构建物呈递的表位来自欲消除的病毒、寄生虫、细菌、或任何细胞、细胞单元或微生物抗原,如可以是癌细胞或被感染细胞。这些表位还可以是使得在自身免疫病中被认作外来抗原的“自身”分子被本发明表位替代的诱导物,从而导致免疫反应延缓或减弱。
这些表位的实例已在上文描述嵌合多肽序列时举出。
本发明还涉及本发明的嵌合分子在制备治疗性组合物中的用途,其中希望表达的特定多肽可恢复结构改变导致被捕获在内质网中并且表达缺陷的蛋白质的细胞内转运。经历细胞内成熟化(包括糖基化、硫酸化、折叠等)的膜表达蛋白就是如此。
一个具体的实例是突变体CFTR(ΔF508)的表位,由于蛋白的修饰其与伴侣分子如钙联接蛋白的结合被改变;这导致该分子被捕捉(这是粘稠物阻塞症的原因),表现为外分泌腺的分泌普遍不足,特别是在胰腺和肺中。
本发明涉及由于缺乏蛋白质的分泌所致疾病的治疗方法,该方法包括直接施用嵌合多肽或在患者细胞中施加质粒形式的遗传信息,所述质粒带有编码可恢复细胞缺陷功能的肽或多肽的外源序列。
这种恢复可以是由多肽X补充缺陷功能的结果,或突变蛋白和从外源序列合成的多肽间竞争结合细胞器的特异分子或受体的结果。一个具体的实例是通过施予携带编码蛋白CFTR与其伴侣分子结合位点之序列的载体、或通过直接施予嵌合多肽,治疗上述的突变,即粘稠物阻塞症的病因。
本发明的构建物为人类或动物医疗界提供了一种治疗病因在于细胞内转运缺陷之疾病、提高或诱导分子、多肽或目的表位的膜呈递的新治疗方法。
在下列实施例和附图的启示下,本发明的其它特征将会显而易见。

I-重组嵌合多核苷酸的构建和相应多肽的产生
I-1)质粒的构建
选择的表位X是表位MAGE,它存在于患黑素瘤之患者的癌细胞中。所用的质粒为Su等,1992,Infact.Immun.,60,33-45,3359中描述的质粒pSU108。
所用的PCR引物如下:

产生的片段被克隆在质粒pUS108的限制性位点SphI和SaII处。
含有糖基化位点和序列KDEL的适配片段,由寡核苷酸硫酸酯1:(5’-磷酸化;5’-GGCCGCCATCCTAATTCTACTTCT-3’)和硫酸酯2(5’-CTCAGAAGTAGAATTAGGATGGC-3’)或硫酸酯3(5’-GAGTCTGAAAAAGATGAACTTTGATGAG-3’)组成,在16℃下连接过夜。
将生成的片段克隆在pSU108的限制位点NotI和EcoRI中,并含有编码B-Glyc-KDEL的cDNA。
I-2)蛋白的纯化
重组片段的纯化也按Su等,1991(上文所引述)中描述的技术进行。简言之,将含有从pSU108所得重组表达质粒的大肠杆菌细胞在30℃培养过夜。然后将培养物在补有50mg/ml氨苄青霉素的LB中50℃下稀释5倍。在42℃下保温4小时后,细胞用大量10mM Tris/HCl(pH8)洗涤,在10mM Tris/HCl,pH8;25%蔗糖,1mM EDTA中保温10分钟,最后迅速重悬在含1mM PMSF和蛋白酶抑制剂混合物(亮抑蛋白酶肽、胰凝乳蛋白酶抑制剂、胃蛋白酶抑制剂、抗蛋白酶和抑蛋白酶肽)的冰水混合物中。这最后一步导致周质破裂。澄清后将上清液载于QFF柱(Pharmacia)上,用20mM Tris/HCl,pH7.5中的NaCl线性梯度洗脱。根据其结构,B片段在120mM和400mM之间被洗脱。然后将含有B片段的流份对20mM Tris/HCl,pH7.5透析,再载于monoQ柱(Pharmacia)上,如前述相同的方式洗脱。得到的蛋白于-80℃保存到使用时,经SDS-聚丙烯酰胺凝胶电泳估测其纯度为95%。
I-3)体外CTL应答的诱导
按已建立的方法(Romani等,1994)培养树状细胞。简言之,将PBMC悬浮在Iscove培养基中,在6孔板中37℃保温2小时。洗去没有粘附的细胞,剩余的细胞在GM-CSF(800U/ml)和IL-4(500U/ml)存在于37℃保温。培养5天后,分别加入浓度为50U/ml和150U/ml的IL-1α和IFN-γ,在30℃下继续保温24小时。然后将树状细胞悬浮在Iscove培养基中,其中存在递增浓度的与MAGE表位偶联的B片段和3μg/ml人β2微球蛋白,以便改善细胞的膜表位呈递能力。将该混合物在30℃下保温4小时。将内在化了与该表位偶联的B片段的树状细胞以5000拉德照射,离心收集,重悬,与CD8+淋巴细胞(从PBMC制得)混合。然后将用抗原致敏的树状细胞与CD8+在5ng/ml IL-7存在下共培养。
10天后用新制备的照射后的树状细胞再刺激应答性CD8+淋巴细胞,并且在递增浓度的与同样表位偶联的B片段存在下保温。在10U/ml和5ng/ml的IL-2和IL-7存在下进行树状细胞与应答性CD8+淋巴细胞的共培养。重复该再刺激过程3次。
为了测定对CTL应答的诱导,将如上述预刺激的应答性CD8+淋巴细胞在癌细胞或被病毒感染的细胞存在下保混。将表达所选择表位的细胞用Na2 51CrO4标记,然后与应答性CD8+接触5小时(Bakker等,1994)。然后测定释放在培养液中的放射活性,可对预刺激的应答性CD8+淋巴细胞的细胞毒性活性进行定量。
结果
II用带有抗原MAGE 1的B片段刺激的PenaEBV细胞和树状细胞对该抗原的呈递
II-1)携带MAGE-1表位之B片段细胞内转运的形态学研究。
我们已证明,可以在志贺菌毒素B片段的羧基末端融合肽序列而保持该蛋白向内质网细胞内行进的特性。在此证明过程中,构建了含有B片段、N-糖基化位点和保留信号KDEL的嵌合多肽。作为对照,使用了保留信号KDELGL,它是肽KDEL的无活性形式,Misendock和Rothman,1995,细胞生物学杂志(J.Cell.Biol.),129,309-319。通过形态学和生化研究表明,修饰的B片段从浆膜通过内体和高尔基体向内质网中转运。这种转运被BFA(真菌代谢物布菲尔德菌素(Brefeldine)A)抑制,被诺考达唑(微管的解聚剂)减弱。
这些实验清楚地表明,保留了融合蛋白向内质网细胞内行进的特性。为了体外评价B片段作为表位的载体用于抗肿瘤免疫接种的潜力,在上文所述试验条件下将表位MAGE-1加入到片段B-Glyc-KDEL上,该新蛋白称为B-MAGE-Glyc-KDEL。将蛋白B-MAGE-Glyc-KDEL与荧光团DTAF偶联,以便通过聚焦显微镜跟踪其细胞内转运。在其内在化以后,在HeLa细胞、Pena-EBV细胞(用EB病毒永生化的B淋巴细胞系)的高尔基体中和内质网中可检测到该蛋白。这些结果证实了关于羧基末端修饰(如上所述)的B片段细胞内转运的原始观察结果,并可以确认造血细胞系的某些呈递细胞能够内在化B片段并将该蛋白转运至内质网中。
我们现明确提出测定蛋白B-MAGE-Glyc-KDEL的N-糖基化的研究。N-糖基化是一种在内质网中特异地进行的修饰。我们以前已证明带有N-糖基化识别位点的B片段若被转运至内质网确实可被糖基化。
II-2)用蛋白B-MAGE-Gly-KDEL刺激的Pena-EBV细胞和树状细胞对抗原MAGE-1呈递的研究
为了评价B片段作为表位载体的能力,我们利用了一个特异性识别与单倍型HLA-A1呈递细胞的I类MHC结合的MAGE-1表位的细胞毒性T淋巴细胞(CTL 82/30)。在用蛋白B-MAGE-Glyc-KEDL刺激的Pena-EBV细胞或树状细胞存在下保持这些CTL。如果表位MAGE-1被这些呈递细胞呈递,CTL将被活化并分泌γ干扰素(IFN-γ),随后定量测定。
所分泌INF-γ的量与呈递细胞对CTL的刺激程度成正比。
20000个呈递细胞(Pena-EBV和树状细胞)(每个圆底微孔中的细胞数)用4%PBS-多聚甲醛固定1小时,或不固定。然后用OptiMEM洗2次,与蛋白B-MAGE-Glyc-KDEL稀释液保温15小时。以4个稀释度试验该蛋白,从10μM终浓度开始,在OptiMEM培养液(无血清培养液)中5倍稀释。15小时后,低速离心洗板。将CTL(CTL 82/30)以5000 CTL/孔的比例再加到100μl培养液(ID-HS-AAG+25U/ml IL-2)中。作为阳性对照,将CTL 82/30在G43细胞系(被MAGE-1表达质粒转染的B淋巴细胞系)存在下保持。培养24小时后,收集上清,定量测定IFN-γ产物。
所得结果示于下表I中。

结果以每个条件下产生的IFNγ单位表示(均值±标准差;n=3)。
我们注意到,用蛋白B-MAGE-Glyc-KEDL刺激的树状细胞和Pena-EBB细胞可被CTL很好地识别,甚至在低蛋白浓度下也是如此。相反,被预先固定的树状细胞和Pena-EBV细胞不被识别,看来发生了蛋白B-MAGE-Glyc-KDEL的胞吞作用和加工。这些令人鼓舞的结果现可得到体外接种实验的支持。
III.小鼠体内的抗肿瘤和/或抗病毒活性试验
准备小鼠的树状细胞并用衍生自蛋白P21RAS、P53或PE2/NER的抗原标记(以测定抗肿瘤活性)或用衍生自HBV、EBV或HPV的抗原标记(以测定抗病毒活性)。按上文I-4)中所述方法准备树状细胞。
然后将这些树状细胞引入小鼠体内。
通过对移植了表达该抗原的肿瘤细胞或病毒的小鼠进行后续处理,观察到了抗病毒或抗肿瘤效果。
结论
当所述病毒或所述癌细胞的特定表位被整合在重组核苷酸序列中、导致合成被限于I类MHC并在免疫系统细胞膜表面表达时,本发明的多肽序列或多核苷酸序列可有利地构成用于治疗某些癌症或某些病毒或细菌感染的药物组合物的活性成分。
IV:借助于志贺菌毒素B片段恢复突变蛋白CFTR(ΔF508)的细胞内转运。
蛋白CFTR(囊性纤维变性跨膜调节蛋白)是浆膜的氯通道。在绝大部分患粘稠物阻塞症的病人中,CFTR基因带有突变。最常见的突变体(ΔF508)的通道活性仍有效,只是被阻滞在内质网中,而不是被转运至浆膜上。我们借助于志贺菌毒素的B片段向内质网中引入已知与钙联接蛋白(内质网的“伴侣分子”)相互作用的CFTR蛋白结构域。该结构域与B片段的羧基端融合。检测发现,该嵌合蛋白可将糖蛋白CFTR(ΔF508)与钙联接蛋白作用位点的N-糖基化链转移,这使得蛋白CFTR(ΔF508)不再被保留在内质网中,而可被转运至浆膜上,发挥正常功能。
我们第一次构建了由B片段和衍生自蛋白CFTR的相互作用结构域组成的嵌合蛋白。在该蛋白的羧基端加上了再循环信号(肽KEDL),以增加其在内质网中的保留。首先证实了这种新蛋白也被转运到靶细胞的内质网中。在用CFTR(ΔF508)的cDNA转染的LLCPK1稳定细胞系中研究了CFTR(ΔF508)的迁移。该细胞系由Mlle.M.A.Costa deBeauregard和M.D.Louvard(巴黎居里研究所,CNRS UMR 144)建立。在这些细胞中表达的蛋白CFTR(ΔF508)另外装有一个表位标签。因而可能通过免疫荧光检测蛋白CFTR(ΔF508)是否到达浆膜。在没有这种处理时,LLCPK1细胞系的浆膜中没有CFTR(ΔF508)的特异性标志。如果这些初步试验的结果是积极的,我们将着眼于粘稠物阻塞症的治疗开发这一途径。
结论
上述试验证明,其中X代表蛋白CFTR和钙联接蛋白间相互作用结构域的合成多肽可有利地构成用于治疗粘稠物阻塞症的药物组合物的活性成分。事实上,在突变者中存在的突变作用结构域与合成多肽片段间与钙联接蛋白相互作用的竞争可使得突变蛋白在支气管中恢复分泌。
抗原呈递实验
将抗原呈递细胞(PBMC,B-EBV细胞,T细胞,树状细胞)在96孔微板中以105个细胞/孔的密度在37℃保温、并用溶于100μl Iscove培养液的抗原敏(刺)4小时或15小时。保温结束后,除去培养液,在每孔的100μl含25U/ml IL2的CTL培养基中加入20000个CTL细胞。24小时后,收集50μl上清液,通过ELISA(Diaclone)试验测定γ干扰素。在某些试验中,细胞用1%多聚甲醛在室温下固定10分钟,在转入微孔板中之前充分洗涤。

其中B代表B片段,X代表其长度上限与逆向转运相适应的一个或多个多肽。
