Abstract
本发明公开了一种存在于植物中的与细胞周期相关的基因CCR及所编码的蛋白质。它参予细胞周期中的G1期至S期。转基因实验证明该基因的超量表达促使植物的生长和发育,转基因植株的长势明显好于对照且比对照提前开花和结实。转反义基因的植株则矮小且发育缓慢。与对照相比,超量表达该基因的转基因植株还有持久的耐盐性。本发明还提供了一种利用CCR基因进行植物品种改良的方法。
Description
技术领域
本发明属于基因工程领域,具体地讲涉及植物中的一类新的细胞周期相关基因及其所编码的蛋白质。
技术背景
细胞分裂是生命的最重要且普遍存在的特征。细胞周期过程是一个基础而复杂的过程。在真核和原核生物中存在着一些相当保守的元件控制着细胞的周期。在酵母和人类中,细胞周期(S,G2,M和G1)的完成与一系列丝/苏氨酸激酶相关。这些激酶包括催化亚基,称为依赖细胞周期的激酶,cyclin-dependent kinase(CDK)和一个活化亚基,cyclin。七十年代,首次运用酵母突变体研究细胞周期调控的分子机制,鉴定了调控复合物的主要分子细胞周期蛋白CDC2和CDC28,与此同时,在海胆中也鉴定了另一个重要分子依赖于细胞周期蛋白的激酶。在过去的30年中,酵母和哺乳类的细胞周期调控研究得到了长足的进展。1989年,John等人用免疫学方法在植物中发现了类似CDK的存在。之后,1990年,从豌豆中克隆了类CDK蛋白的基因。随之,又有一些CDK和cyclin被相继发现。十年来已从各种植物中克隆了cyclin,CDK,CDK-活化激酶CAK和CDK抑制物CKI基因,并且勾画了简单的植物细胞周期调控的遗传机制。
在细胞周期调控中,G1到S相是最重要的。在G1相至细胞分裂中已发现了许多基因涉及调控作用。在酵母和动物中,这些基因的突变引起表型变化甚至死亡。拟南芥中,cyclinD在G1到S相时表达并起作用。
发明内容
本发明人从盐生植物山菠菜Atriplex hortensis中克隆了一个cDNA,与上述cyclinD基因不同源。经在数据库中比较它的编码蛋白与人类中编码G1至S的细胞周期蛋白p38-2G4(AAB91536)有50%相似性,因此命名为AhCCR(Cell Cycle Related)基因。AhCCR cDNA全长1460bp,含有1194bp的ORF,编码含397个氨基酸的蛋白质,5’-UTR为82bp,3’-UTR为184bp。
以AhCCR基因序列从拟南芥基因组序列中查出其同源基因序列,据此设计引物,从拟南芥cDNA中克隆了AhCCR的同源基因AtCCR基因。AtCCR cDNA全长1472bp,含有1179bp的ORF,编码392个氨基酸,5’-UTR为96bp,3’-UTR为197bp。
转基因研究表明,无论将AhCCR基因转入烟草还是将AtCCR基因转入拟南芥,转基因植株在正常条件下的长势明显优于对照且提前开花结实。而以反义AtCCR转化的拟南芥生长缓慢。AtCCR基因过量表达的转基因植株比对照有明显高的耐逆性。该基因可作为目的基因导入植物,提高植物的产量及耐逆性,以进行植物品种改良。
综上所述,本发明人提供的CCR基因是植物中的一类新基因,其参予植物细胞周期的调控。
因而,本发明的一个目的是提供一种新的参予植物细胞周期调控的基因的核苷酸序列,此类基因中的AhCCR、AtCCR分别如序列表1,2所示。
本发明所述的一种新的参予植物细胞周期调控的蛋白质(基因)可以是来自所有植物包括山菠菜、烟草、拟南芥、草坪草、杨树、大豆、小麦、水稻、棉花或蕃茄等。这种蛋白质的氨基酸序列中AhCCR、AtCCR分别如序列表1,2所示。
本发明的再一个目的是提供一种培育耐逆植物品种的方法,包括将本发明所述的植物细胞周期相关基因导入植物细胞并使其表达的步骤。
在本发明的方法中,可利用CCR基因作为目的基因构建植物表达载体,其中可用任何一种启动子例如花椰菜花叶病毒(CAMV)35S启动子、Ubiquitin启动子或其它启动子,该表达载体中必要时可包括增强子,不论是转录增强子或翻译增强子。为了简化转化细胞的鉴定可使用选择性标记包括对抗生素抗性的酶,也可利用颜色变化(例如B-葡糖醛酸糖苷酶GUS)或发光(例如荧光酶)来识别的化合物的酶类,也可用无标记选择。所用的表达载体可使用Ti质粒,Ri质粒,植物病毒载体等。转化方法可用经农杆菌介导法、基因枪法、花粉管通道法或其它方法转化植物。
本发明的CCR基因的受体植物包括单子叶和双子叶植物, 例如水稻、小麦、玉米、大豆、棉花、蔬菜、树木和草坪草等。

具体实施方式
实施例1
山菠菜种于温室中(22℃),6星期后以400mM NaCl浇灌,4天后取叶子,分离mRNA,按常规方法构建cDNA文库。以AhDREB(已申请发明专利,申请号:01123827.5)的240bp片段为探针从1.5×106噬斑中以低严谨条件筛选到46个噬斑。经过3轮筛选后取插入片段大于1.0Kb的进行序列分析。其中一个片段约1.5Kb,包括1194bp ORF,82bp 5’-UTR及184bp 3’-UTR(序列表1)。GeneBack blast比较发现该片段与人类中G1至S细胞周期蛋白p38-2G4基因相似性为51%。(图1)。
实施例2
根据AhCCR基因序列,在GeneBack数据库中从拟南芥基因组全序列中检索到相应的基因AtCCR的序列,据此,用PCR方法从拟南芥cDNA中扩增并克隆了AtCCR基因。其氨基酸和核苷酸序列列于序列表2。根据序列检索表明AtCCR基因在拟南芥基因组中也仅有一个拷贝。
实施例3
用实例1中的山菠菜材料提取的总DNA进行Southern分析表明在山菠菜基因组中仅有一个拷贝AhCCR基因(图2)。以AhCCR基因序列检索GeneBack表明在拟南芥基因组中也仅有一个拷贝AtCCR基因。
实施例4
用实验例1中所得到的AhCCR基因构建植物表达载体(图3),以农杆菌介导法转化烟草。转基因烟草的生长明显优于对照,发育(开花)也比对照提前(图4)。
实施例5
以实验例2中得到的AtCCR基因,按实验例4的方法构建AtCCR基因正义和反义的植物表达载体并分别转化拟南芥,转正义基因也即AtCCR基因超表达的转基因植株生长和发育均优于对照,而转反义基因的植株也即AtCCR基因低表达的植株生长与发育均明显劣于对照(图5)。
上述实施例表明本发明中克隆的这一类新的植物细胞基因CCR具有与细胞周期相关的特征,与人类中的同类蛋白p38-2G4的相似性为51%,此类基因在植物中的功能迄今未经描述,为一类新基因。实验例4、5表明该基因与植物的生长、发育以及对逆境的耐性相关。此类基因可作为目的基因用以改良植物的品种。
