Abstract

Description
发明背景
本发明涉及用于持续释放生物活性蛋白质的新组合物。更具体地说,本发明提供了牛生长激素释放因子的优良的水性持续释放可注射配剂,也提供了使用这些新的组合物进行持续或延长释放牛生长激素释放因子的方法。
随着遗传工程技术的发展,许多生物活性肽和蛋白质已可大量获得。然而,这些重组产生的肽和蛋白质的施用存在一些独特的问题。在许多情况下,这些蛋白质生物效应的维持需要长期施用。由于这些药剂的日常施用不很方便,最好进行持续或延长释放。
由于多种原因,长期以来现有技术集中于使用生物相容性油剂作为载体来达到许多药物的持续释放,包括蛋白质尤其是生长激素。针对这一技术的专利有Mitchell的美国专利5,013,713和Ferguson等人的美国专利4,977,140。Mitchell报道了使用含有至少约10%(重量)的生物活性生长激素和作为组合物连续相的生物相容性油如玉米油的基本上非水性的组合物来达到牛生长激素(bSt;BGH)以所需效应水平的延长的胃肠外释放。Ferguson等人报道了注射含有bSt,蜡和一种油的持续释放配剂在一段长的时间内提高了奶牛的每日牛奶产量。

发明概述

该生长激素释放因子优选以约200mg的剂量和约180mg/ml的浓度存在于一种水性液体中。在这一剂量和浓度下,该水性生长激素释放因子配方提供了将生长激素释放因子在大于约35天的时间内持续释放入动物循环系统的技术。
本发明因而提供了具有高效能并易于制备的延长释放组合物。而且这一效能可以可注射组合物的形式获得,不需要将生长激素释放因子与其它物质进行强行压缩以形成固体植入物。再者,本发明的处理方法的实施不需要切入,而切入在例如使用固体延长释放植入物时是必需的。本发明另外的实施方案、范围和优点将通过下面的描述和后附的权利要求看出。

发明详述
为便于理解本发明的原理,现将提及某些实施方案并将使用特定的语言讲行描述。不过,应该理解这不是为了限定本发明的范围,本文所描述的本发明原理的这种改变,进一步的变化和应用对于本发明所属技术领域的普通技术人员来说是容易想到的。
如上所述,本发明的一个优选实施方案以物质的组合物形式提供了一种可注射延长释放配剂,它包括一种在水性载体中的生长激素释放因子。注射之后的该配剂提供了效应剂量的激素或释放因子在长时间内对动物的释放。该动物可以是可产生内源性生长激素的任何一种,包括脊椎动物如牛、绵羊、猪、山羊、马、鸟、鱼和人类。

配剂/施用
在试验开始前7天将12头Holstein小公牛(250-300公斤)进行称重。根据重量将其从最重的至最轻的进行排序。根据重量将阉牛分成两组,各有6头。在组内随机指定进行处理。

水性配剂的制备是通过将研细的冷冻干燥的GRFA分散于注射用无菌水(SWFI,Vedco,Inc.)中而进行的。由于在加入过程中粉末的快速水化作用,粉末的加入是在50mL烧杯中用刮刀手工调拌而进行的。将1g的这种粘糊立即装入6个预先灭菌的3mL注射器中。
将肋骨上面的距肩胛骨腋窝边缘的尾侧约4英寸处的区域剪毛,并在该位点进行皮下注射(SC)。用14号,1.5英寸的针头施用该水性糊状配剂。
至少在用药前一天将一根套管插入颈静脉。在皮下注射该配剂后的第1,3,7,14,21,28和35天,在从喂食前7小时至喂食前2小时的5小时时间内,每隔30分钟取血(8mL)。在第1天相对于配剂注射-40和-20分钟抽取另外的样品。
测试/统计
该12头Holstein小公牛在The Upjohn Farms被关闭于室内分隔栏内(18-20℃),光照为16小时光照:8小时黑暗(光照:0600)。1500小时后小公牛被训练成在2小时内消费掉每日一次为其提供的全天的饲料。每日饲料由41.1%的谷物青贮饲料和58.9%的谷物添加物/浓缩物(B-382)组成,以干重计,以维持每头每天0.8-1.0公斤的增重水平。随意饮水。研究工作表明,内源性血清ST曲线可受饲喂影响(Moseley等(1988)J.Endocr.117:253-259)。因而,血液采样方式的安排考虑到与饲喂的关系,从而优化表征时间性ST分布型的能力。通过放射免疫测定(Moseley等人(1982)J.Anim.Sci.55:1062-1070)检测所得血清的ST浓度。
通过由不规则四边形求和法计算的每一采样日的ST响应曲线下的面积(AUC),每日ST-AUC总和,在全部采样期间的最大ST浓度,最大ST发生时间和每一采样日平均ST-AUC回复到平均对照ST-AUC的时间来评估该配剂的有效性。演绎假设检测的显著水平为0.05。最大总ST浓度和AUC的分析是使用一个随机化完全区组设计的方差分析进行的,将区组(重量组)作为随机效应(Steel,R.G.D和J.H.Torrie(1980)统计学原理和程序(Principles and Procedures of Statistics),Mc Graw-Hill Book Co.,New York)。每日曲线下面积的分析是通过方差的混合模型分析(SAS Procedure MIXED)进行的。如果处理与采样时期的相互作用显著,则在每一采样时期检测到处理差异(Milliken,G.A.和D.E.Johnson(1984)凌乱数据的分析(Analysis of MessyData),Van Nostrand Reinhold,New York,pp.19-22)。使用处理的加权估值X区组和残数方差和自由度的Satterthwaite估值计算最低位差(LSD),以比较给定时间的处理平均值,如Milliken和Johnson上述文献所述。使用LSD在每一时间在对照平均值周围建立置信区间(95%)。在所有方差分析前使用方差同质性的Levene氏测验。将具有方差同质性的数据用log10转换。所有分析均使用SAS(1989)SAS使用者指南:统计学,版本6。(SAS User’s Guide:Statistics,Version6.)SASInstitute,Cary,NC进行。
结果A.血清ST浓度
在第1天和第35天在接受该水性配剂或没有接受配剂的小公牛中观察到的血清ST平均浓度分别示于图1和图2。这些数据与在第3,7,14,21和28天得到的浓度值一起被用于计算每日ST-AUC。对接受了该水性配剂的动物,最大平均血清ST(121.4ng/mL)和该最大值出现的时间(6.5天)示于表1。该最大浓度大于对照的38.4ng/mL(P<0.05),并且该最大值出现的时间与对照不同。B.ST曲线下面积
计算第1,3,7,14,21,28和35天的日ST-AUC。观察到了方差的同质性(Levene’s测验),因而将数值转换成log10。对于水性配剂处理组和对照组的每日血清ST-AUC的最小平方平均值示于表2。全部对照平均值为0.299单位,是通过将所有取样日对照小公牛的曲线下面积进行平均而确定的。将该对照平均值与水性配剂处理组进行比较,以确定每日ST-AUC回复至基线的时间。比较结果示于图3。在第35天本研究结束时,在对照和WAT-18组之间仍有差异。
在接受WAT-18或没有U-90699F后的35天中Holstein小公牛的总血清ST-AUC示于表1。总ST-AUC的最小平方平均值为14.9(CONT)和36.2(WAT-18)单位。接受了该水性配剂的小公牛比对照小公牛具有较大的总ST-AUC(P<0.05)。
本实验得出的主要结果是处理ST-AUC比对照提高的时间。用水性配剂达到了长期效应。将处理ST-AUC除以每日平均ST-AUC(0.299单位)可以看到,在第1天该水性配剂ST-AUC比平均对照ST-AUC大3.55倍。到第7天降至大1.81倍。在第7-35天,该ST-AUC大于对照的倍数保持在1.74至1.83之间,表明为零级释放。

1.一种在动物的循环系统中达到在长于7天的时间内具有提高了的内源性生长激素浓度的方法,它包括给该动物胃肠外施用基本上水性的牛生长激素释放因子组合物,该组合物含有在水中的以至少约20mg/mL的浓度存在的至少约50mg生物活性牛生长激素释放因子。
2.一种药物组合物,它含有在水中的以至少约180mg/mL的浓度存在的至少约200mg生物活性牛生长激素释放因子,以形成可注射用的水性牛生长激素释放因子溶液,所说溶液基本上不含有吸收改性剂和缓冲剂。
3.一种在动物的循环系统中达到在长于35天的时间内具有提高了的内源性生长激素浓度的方法,它包括给该动物胃肠外施用基本上水性的牛生长激素释放因子组合物,该组合物含有在水中的以至少约80mg/mL的浓度存在的至少约200mg的生物活性牛生长激素释放因子。
