Abstract

Background
The vaginal structure of women has natural defensive function of preventing invasion of external pathogens, but is very easy to cause invasion of pathogens in life to cause gynecological inflammation. Researches show that the gynecological inflammation has complex etiology, is related to physical and chemical stimulus and injury, bacteria, trichomonas, virus infection and other factors, and clinically common gynecological inflammation mainly comprises female pruritus vulvae, vulval ulcer, vulval scabies, vulval furuncle and internal vaginal inflammation. The invention creates a traditional Chinese medicine fermented hydrogel for treating gynecological inflammation, which is a traditional Chinese medicine composition consisting of raw materials of macleaya cordata, abelmoschus manihot, long pepper, dysosma versipellis, frankincense, clove, dahurian rhododendron leaf and star anise, and is prepared by grinding, crushing and leaching with solvent to obtain leaching liquor; or concentrating the leaching solution under negative pressure to obtain leaching concentrated solution; and (3) performing biological fermentation and concentration on the leaching solution to obtain a fermented paste of the leaching solution of the traditional Chinese medicine composition, and compounding the fermented paste with a hydrogel material to obtain the traditional Chinese medicine fermented hydrogel. The traditional Chinese medicine fermented hydrogel is applied to the vulva or pushed into the vagina, can quickly heal and treat pruritus vulvae, vulval ulcer, vulval scabies, pustule of the vulva, furunculosis of the vulva, trichomonas vaginitis, colpitis mycotica, bacterial vaginitis, senile vaginitis, cervicitis or cervical erosion, has quick response, high cure rate, easy coating, no irritation, good compliance, no foreign matter sense, long retention time, full efficacy, and convenient use, storage and carrying.

The traditional Chinese medicine composition comprises, by weight, 6-12 parts of macleaya cordata, 6-12 parts of abelmoschus manihot, 6-12 parts of long pepper, 6-12 parts of dysosma versipellis, 2-12 parts of frankincense, 2-8 parts of dahurian rhododendron leaf, 2-8 parts of clove and 2-8 parts of star anise.
The gynecological inflammation refers to female pruritus vulvae, vulval ulcer, vulval scabies, vulval furuncle, vulval furunculosis, trichomonas vaginitis, colpitis mycotica, bacterial vaginitis, senile vaginitis, cervicitis or cervical erosion.
The preparation method of the traditional Chinese medicine hydrogel comprises the following steps:
the first step: preparation of Chinese medicine leaching liquor
6-12 parts of macleaya cordata, 6-12 parts of abelmoschus manihot, 6-12 parts of long pepper, 6-12 parts of dysosma versipellis, 2-12 parts of frankincense, 2-8 parts of dahurian rhododendron leaf, 2-8 parts of clove and 2-8 parts of star anise are sequentially weighed, mixed together and ground to obtain traditional Chinese medicine composition powder. Immersing the traditional Chinese medicine composition powder into ethanol which is 5-10 times of the traditional Chinese medicine composition powder in weight, controlling the temperature to be 15-80 ℃ and stirring and leaching for 2-200 hours, and filtering to obtain traditional Chinese medicine ethanol leaching liquid for later use; and then pouring the filtered Chinese medicine residues into water with the weight being 1-5 times of that of the Chinese medicine residues, refluxing and boiling for 0.5-5 hours under stirring, cooling and filtering to obtain Chinese medicine water leaching solution. Mixing the Chinese medicinal water extract and the Chinese medicinal ethanol extract together to obtain Chinese medicinal composition extract, which is called Chinese medicinal extract for short.
Wherein the preferable raw material medicines comprise 12 parts of macleaya cordata, 12 parts of abelmoschus manihot, 12 parts of long pepper, 12 parts of dysosma versipellis, 6 parts of dahurian rhododendron leaf, 6 parts of frankincense, 2 parts of clove and 2 parts of star anise.
The preferred raw material medicaments comprise 8 parts of dahurian rhododendron leaf, 8 parts of dysosma versipellis, 6 parts of macleaya cordata, 6 parts of abelmoschus manihot, 6 parts of long pepper, 2 parts of frankincense, 2 parts of clove and 2 parts of star anise.
The present inventors have selected a process and method for the segmental leaching of a Chinese medicine with water and ethanol, respectively, in order to further utilize the active ingredients in the Chinese medicine. Experiments show that the traditional Chinese medicine residues obtained by leaching the traditional Chinese medicine powder with ethanol can be further leached out by leaching with water, and the obtained traditional Chinese medicine leaching solution has higher efficacy.
Wherein macleaya cordata is warm in nature, pungent in flavor, bitter in flavor and toxic. The main bioactive substances of macleaya cordata comprise isoquinoline alkaloids such as sanguinarine, chelerythrine, protopine, allocryptopine and the like, and have various biological activities such as antimicrobial, insecticidal, antiviral, anti-inflammatory, antitumor, liver function improving, immunity enhancing and the like. Dysosma versipellis is cool in nature, bitter and pungent in flavor and toxic. The bioactive components of Dysosma versipellis include lignans, flavonoids, alkaloids and sterols. Wherein the medicinal active ingredients are mainly podophyllotoxin and its derivatives, podophyllotoxin ketone, its glycoside, dysosmallpox alcohol, kaempferol, quercetin, kaempferol glycoside. Modern pharmacological tests show that podophyllotoxin of dysosma versipellis has remarkable inhibiting effect on herpesvirus, wherein kaempferol has inhibiting effect on staphylococcus aureus, typhoid fever, pseudomonas aeruginosa and the like. The dysosma versipellis has the functions of clearing heat and detoxicating, drawing out toxin and promoting tissue regeneration, removing blood stasis and relieving pain, and resolving phlegm and resolving masses.
The mastic is pungent, bitter and warm, wherein the resin content in the chemical components is high, the gum accounts for only 27-35%, and the volatile oil accounts for 3-8%. Example 4 biological assay
1. Bacteriostasis test: taking 2mL of the traditional Chinese medicine leaching solution, the traditional Chinese medicine fermentation liquid and the traditional Chinese medicine fermentation hydrogel of the examples 1-3 according to the proportion of 1:2, 1:5, 1:10, 1:20, 1:50 and 1:100, diluting the traditional Chinese medicine leaching solution, the traditional Chinese medicine fermentation liquid and the traditional Chinese medicine fermentation hydrogel into a sample to be tested with the same concentration by using the culture solution of the step three of the example 1, adding 2 drops of pathogenic bacterial strain liquid at the same time, fully and uniformly mixing, and culturing in a 37 ℃ incubator for 24 hours; the growth of pathogenic bacteria was observed, the Minimum Inhibitory Concentration (MIC) was calculated, and the test results are shown in table 1.

5. The traditional Chinese medicine fermented hydrogel for rapidly healing gynecological inflammation according to claim 1, wherein the hydrogel material is prepared by compounding gel matrix, purified water, cosolvent and pH value regulator in a weight ratio of 1:1-100:0-10:0-10;
