Description
Technical Field
The invention relates to the technical field of laboratory sample preservation, cleaning and recovery, in particular to a device suitable for zooplankton preservation, cleaning and recovery.
Background
Zooplankton refers to a group of animals living in a water body, is various in species and extremely large in quantity, mainly comprises lower protozoa, coelenterates, comb plate animals, rotifers, crustaceans, mollusks, jawberries, higher tail cord animals and the like, is sensitive to environmental changes due to community structures, plays a role in supporting and starting in a food chain, and is generally used for evaluating important content of water environment at home and abroad. Laboratory qualification identification requires that samples of each voyage zooplankton in the laboratory be fixedly stored after detection is finished so as to meet the requirements of laboratory examination such as flight examination and retest of the samples; meanwhile, the target species are also often required to be fixedly stored in daily inspection and monitoring activities.
At present, the method for fixedly preserving zooplankton mainly comprises freezing at-20 ℃, liquid nitrogen, 1% of Lu Ge reagent and 5% of formaldehyde, wherein 5% of formaldehyde is most used in standard specification, and the effect is optimal. However, the fixation of 5% formaldehyde has certain defects, such as strong reducibility of formaldehyde, long time of fixing the formaldehyde sample is easy to oxidize into formic acid, the pH of the fixing solution is reduced, and the long-time fixation and preservation of zooplankton are affected; therefore, to ensure the integrity of the preserved samples, the formaldehyde fixing solution needs to be replaced periodically. In addition, it is often necessary to recover the stored sample, to retest the retained sample, or to re-identify the species.
The preservation tube of the traditional technical sample is generally a cylinder, the surface of the cylinder is smooth, the preservation tube is easy to slide off, and the sample name and preservation time are not easy to label when the sample name and preservation time are labeled on the preservation tube; the traditional formaldehyde fixing solution replacement needs to pour a sample into a funnel filled with bolting silk for suction filtration, then the bolting silk carrying zooplankton is reversely buckled in a culture dish filled with water for swing washing, finally the zooplankton is washed out of the bolting silk, the washed zooplankton is sucked by a suction pipe and is placed in a preservation pipe again, new fixing solution is added into the preservation pipe for fixation again, the process needs to suction-filter the zooplankton, and in addition, the suction filtration process, the swing washing process and the process of sucking the zooplankton sample to the preservation pipe are particularly easy for the zooplankton to be lost and damaged. Traditional zooplankton cleaning and recycling have a pumping filtration process as well, samples are easy to lose in the swing washing process, and the samples are easy to drop in the transportation process.
Therefore, how to provide a device suitable for zooplankton preservation, cleaning and recovery is a problem that needs to be solved by those skilled in the art.
