Abstract
本发明涉及GS‑9620在制备预防和/或治疗银屑病的药物中的应用,属于生物技术领域。本发明提供了GS‑9620在制备预防/或治疗银屑病的药物中的应用;研究发现,GS‑9620能够改善银屑病小鼠模型的皮肤损伤程度,降低银屑病小鼠模型皮肤组织中炎症细胞因子的表达,降低银屑病小鼠模型脾脏和淋巴结中T淋巴细胞的侵袭,增加银屑病小鼠模型皮肤组织中自噬相关蛋白的表达,并且,GS‑9620能够增加M5诱导的银屑病角质形成细胞模型中自噬相关蛋白的表达,抑制银屑病角质形成细胞模型中炎症小体的产生,抑制银屑病角质形成细胞模型中炎症细胞因子的表达,因此,GS‑9620在制备预防/或治疗银屑病的药物中极具应用前景。

技术领域
本发明涉及GS-9620在制备预防和/或治疗银屑病的药物中的应用,属于生物技术领域。
背景技术
银屑病(Psoriasis)俗称牛皮癣,是一种由环境因素刺激、多基因遗传控制、免疫介导的慢性炎症性皮肤病,其特点是角质形成细胞(Keratinocytes,KCs)过度增殖和分化异常,以及免疫细胞浸润。虽然银屑病的发病机制尚不完全清楚,但目前普遍认为角质形成细胞与免疫细胞的异常相互作用在银屑病的发展中起着非常重要的作用。活化的角质形成细胞表达过量的细胞因子、趋化因子和辅助因子,将这些信号传递给先天性免疫细胞和适应性免疫细胞,激活免疫系统,并不断分泌促炎因子,维持甚至放大炎症反应。
银屑病的典型表现为鳞屑性红斑或斑块,局限于一处或全身广泛分布,部分患者可同时有关节症状(如关节肿胀、疼痛)、指甲的异常,中重度患者患代谢综合征(以肥胖、血脂紊乱、高血压、血糖异常为主要表现)、心血管疾病的风险增加。可见,银屑病严重影响患者生活质量,亟需找到更多疗效更好的银屑病治疗药物。
GS-9620(维沙莫德,CAS:1228585-88-3)是一种口服的Toll样受体7(Toll-LikeReceptor-7,TLR-7)激动剂,其可以通过诱导一种称为干扰素的抗病毒细胞因子,而不是直接激活人干细胞中的抗病毒机制,延长HBV和HIV的抑制作用,因此,GS-9620被用于治疗HBV和HIV感染。目前,尚无关于使用GS-9620预防/或治疗银屑病的相关报道。
发明内容
为解决上述问题,本发明提供了GS-9620在制备预防/或治疗银屑病的药物中的应用。
在本发明的一种实施方式中,所述预防/或治疗银屑病包括通过促进角质形成细胞的自噬来减轻银屑病引起的皮肤炎症。
在本发明的一种实施方式中,所述药物的成分包含活性组分以及药学上可接受的载体;所述活性组分包含GS-9620。
在本发明的一种实施方式中,所述活性组分为GS-9620。
在本发明的一种实施方式中,所述药学上可接受的载体包括表面活性剂、赋形剂、稳定剂、助悬剂、等渗剂、防腐剂、填充剂、崩解剂、抗氧剂、乳化剂、包衣剂、黏合剂、润滑剂和/或矫味剂。
在本发明的一种实施方式中,所述药物的剂型为散剂、片剂、颗粒剂、胶囊剂、口服液体剂或注射剂;所述口服液体剂为溶液剂、糖浆剂、乳剂或混悬剂。
本发明还提供了一种预防/或治疗银屑病的药物,所述药物含有活性组分;所述活性组分包含GS-9620。

本发明提供了GS-9620在制备预防/或治疗银屑病的药物中的应用;研究发现,GS-9620能够显著改善咪喹莫特(imiquimod,IMQ)诱导的银屑病小鼠模型的皮肤损伤程度,降低银屑病小鼠模型皮肤组织中炎症细胞因子(IL-1β、IL-6、IL-18、HMGB和TNF-α)的表达,降低银屑病小鼠模型脾脏和淋巴结中T淋巴细胞(CD3+、CD4+CD25+、CD4+IL-17A+和CD4+INF-γ+)的侵袭,增加银屑病小鼠模型皮肤组织中自噬相关蛋白(ATG5、ATG12和ATG16L1)的表达,并且,GS-9620能够显著增加由M5(IL-1α、IL-17A、IL-22、抑瘤素M和肿瘤坏死因子-α)诱导的银屑病角质形成细胞模型中自噬相关蛋白的表达,抑制银屑病角质形成细胞模型中炎症小体(NLRP3)的产生,抑制银屑病角质形成细胞模型中炎症细胞因子的表达,可见,GS-9620能够通过调节角质形成细胞的自噬来减轻银屑病引起的皮肤炎症,因此,GS-9620在制备预防/或治疗银屑病的药物中极具应用前景。

下述实施例中未注明具体实验步骤或条件者,按照本领域内的文献所描述的常规实验步骤的操作或条件即可进行。所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规试剂产品。
实验例1:GS-9620作为制备预防/或治疗银屑病的药物的验证
1、材料与方法
1.1、银屑病患者、健康志愿者皮肤样本收集
本研究遵循《赫尔辛基宣言》的原则,经深圳大学附属第六医院伦理委员会(中国广东深圳)批准。在本研究之前,所有受试者都获得了书面知情同意。分别取银屑病患者和健康志愿者的皮肤活检标本,根据银屑病面积和严重程度指数(psoriasisareaandseverityindex,PASI)(参见文献:R.G.Langley and C.N.Ellis:“Evaluating psoriasis with Psoriasis Area and Severity Index,Psoriasis GlobalAssessment,and Lattice System Physician’s Global Assessment.”J.Am.Acad.Dermatol.vol.51,no.4,pp.563–569,2004.)对银屑病患者(n=8)和健康志愿者(n=8)进行评估。所有受试者都提供了参与研究的书面知情同意书。
1.2、人原代角质形成细胞
经供者同意,采用酶消化法从包皮环切术患者中分离原代表皮角质形成细胞(NHEKs)(参见文献:T.Aasen and J.C.Izpisúa Belmonte:“Isolation and cultivationof human keratinocytes from skin or plucked hair for the generation ofinducedpluripotent stem cells.”Nat.Protoc.vol.5,no.2,pp.371–382,2010.)。
1.3、银屑病小鼠模型的造模及治疗
雄性BALB/c小鼠(6~8周龄,18~22g)购自广东省实验动物中心(中国广东)。饲养条件:恒温24±2℃,湿度55%±15%,光照/暗循环12h,自由取水和食物。所有动物的使用均经深圳大学机构动物保护和使用委员会(IACUC)批准。小鼠在实验室适应2周后,在背部刮3×3cm的毛。刮毛24小时后开始实验。实验分为4组,4组分别为空白对照组(Control)、咪喹莫特造模组(IMQ)、GS-9620治疗组(IMQ+GS-9620)、MTX(甲氨蝶呤)治疗组(IMQ+MTX),其中,空白对照组小鼠不进行任何操作;咪喹莫特造模组为:使用含5%(w/v,g/100mL)咪喹莫特的Aldara乳膏(中国四川明鑫药业)涂抹小鼠刮毛处,每天涂抹一次,每次涂抹剂量为62.5mg,持续涂抹7天(将涂抹前的一天记为0d),以进行银屑病小鼠模型的造模,并且,从第1d开始,每日在涂抹乳膏后2h对小鼠腹腔注射生理盐水,每天注射一次,每次注射剂量为100mL/只,持续注射6天;GS-9620治疗组为在咪喹莫特造模组的基础上,从第1d开始,每日在涂抹乳膏后2h对小鼠腹腔注射GS-9620(MedChemExpress,USA),每天注射一次,每次注射剂量为4.5mg/kg(溶于与空白对照组等量生理盐水中后进行注射),持续注射6天;MTX治疗组为在咪喹莫特造模组的基础上,从第1d开始,每日在涂抹乳膏前2h对小鼠腹腔注射MTX(MedChemExpress,USA),每天注射一次,每次注射剂量为1mg/kg(溶于与空白对照组等量生理盐水中后进行注射),持续注射6天(干预策略如图1所示)。实验期间,每天对小鼠进行称重,称重结果如图2所示。
1.4、PASI和皮肤组织学
银屑病面积及严重程度指数(PASI)衡量背部临床炎症的严重程度(参见文献:M.Na Takuathung,A.Wongnoppavich,A.Panthong,P.Khonsung,N.Chiranthanut,N.Soonthornchareonnon,and S.Sireeratawong:“Antipsoriatic Effects ofWannachawee Recipe on Imiquimod-Induced Psoriasis-Like Dermatitis in BALB/cMice.”Evid.-Based Complement.Altern.Med.ECAM.vol.2018,pp.7931031,2018.)。分别对小鼠皮肤的红斑、鳞屑和厚度划痕进行评分。每个项目以4分制进行评分(0=无;1=轻度;2=中度;3=重度;4=极重度)。将单项得分相加,得到总分最高9分,评分结果如图3所示。
分别取小鼠皮肤用福尔马林固定,石蜡包埋,切片(厚度5μm),苏木精和伊红(H&E)染色,进行组织病理学检查。取载玻片使用MF53显微镜(Mshot,中国)捕获图像,并使用ImagePro Plus(6.0版本,美国)进行分析,分析结果如图4所示。
1.5、免疫组织化学
以健康人和银屑病患者的皮肤组织免疫组化结果为对照,将人皮肤和小鼠皮肤分别用福尔马林固定,石蜡包埋,切片(厚度5μm),将载玻片在室温(25℃)下于浓度为3%(w/v,g/100mL)的双氧水中浸泡5分钟,以消除内源性过氧化物酶活性。浸泡结束后,用PBS缓冲液(pH 7.4,浓度0.01M,Gibco)冲洗载玻片三次,每次2分钟。冲洗结束后,将载玻片在室温(25℃)下于封闭用正常羊血清(5%工作液,中杉金桥)中浸泡10分钟。浸泡结束后,将载玻片在37℃下与不同一抗分别孵育1小时,其中,一抗分别为:ATG5(Proteintech,10181-2-AP,使用一抗稀释液稀释1000倍后使用)、ATG12(Proteintech,11122-1-AP,使用一抗稀释液稀释1000倍后使用)和ATG16L1(Proteintech,29445-1-AP,使用一抗稀释液稀释1000倍后使用)。孵育结束后,用PBS缓冲液洗涤载玻片三次,每次2min。冲洗结束后,将载玻片先在37℃下用生物素偶联二抗(中杉金桥,ZB-2306)孵育30分钟,再在37℃下用辣根过氧化物酶亲和素(HRP)(Proteintech,SA00001-2,使用TBST缓冲液稀释5000倍后使用)孵育2小时。孵育结束后,用SuperSignal West Pico PLUS试剂盒(Thermofisher,34580,USA)冲洗载玻片,并使用MF53显微镜(Mshot,中国)拍摄图像,实验结果如图5~6和图31所示。
1.6、酶联免疫吸附试验(ELISA)
采用ELISA试剂盒定量小鼠皮肤组织中TNF-α(CHE0004,四正博生物科技公司,中国)、IL-1β(IBILB0801,IBL-America,美国)、IL-6(BE27768,IBL-America,美国)、IL-18(IBATGP0271,IBL-America,美国)和HMGB1(ARG81351,ariigo生物实验室,中国)的表达量,实验结果如图7~11所示。所有的实验都按照制造商的说明进行。
1.7、流式细胞术
收集小鼠脾脏和淋巴结,在小鼠脾脏和淋巴结中分别加入5mL PBS缓冲液(pH7.4,浓度0.01M,购自Gibco)研磨后,过滤去除残留碎片,得到滤液;将滤液离心,取沉淀。在淋巴结所得沉淀中加入5mL PBS缓冲液重悬,得到淋巴结细胞悬液。在脾脏所得沉淀中加入1mL 1×ACK红细胞裂解液(Servicebio Technology),于37℃孵育1min裂解红细胞,得到裂解液;在裂解液中加入10mLPBS缓冲液后离心,取沉淀;在沉淀中加入5mLPBS缓冲液重悬,得到脾细胞悬液。先从淋巴结细胞悬液和脾细胞悬液中分别取1×106细胞,然后在两种细胞中分别加入CD3、CD4和CD25抗体(BD Biosciences,三种抗体同时加入,每种抗体均加入0.5μg),于37℃下避光孵育15min对细胞表面标记染色。先从淋巴结细胞悬液和脾细胞悬液中分别取1×106细胞,然后在两种细胞中分别加入2μL细胞刺激混合物(为添加蛋白转运抑制剂的PMA、离子霉素、BrefeldinA和莫能菌素的混合物,500X,BD Biosciences,货号00-4975-93),于37℃、5%(v/v)CO2培养箱内培养6h刺激细胞使其活化;活化结束后,在两种细胞中分别加入0.5μg CD3、CD4和CD25抗体(BD Biosciences,三种抗体同时加入,每种抗体均加入0.5μg),于37℃下避光孵育15min对细胞表面标记染色;对细胞表面标记染色后,使用固定/破膜试剂盒(BD Biosciences)固定和破膜两种细胞;固定和渗透细胞后,在两种细胞中分别加入IFN-γ和IL-17A抗体(BD Biosciences,两种抗体同时加入,每种抗体均加入0.5μg),于4℃下孵育40min染色细胞内标记物。染色结束后,所有样品均采用流式细胞术检测,实验结果如图12~30所示。
1.8、免疫印迹
取小鼠背侧皮肤;在小鼠背侧皮肤中以1mL的添加量加入配制好的消化液(0.1mg/mL Liberase TM+0.1mg/mLDNaseI+20mM HEPEs,以无血清加双抗的1640培养基作为溶剂)后,于37℃避光孵育2h消化皮肤细胞,得到消化液;将消化液用100μm滤网研磨过滤后,500g离心5min,得到皮肤单细胞悬液;收集皮肤单细胞悬液中的蛋白;将蛋白经12.5%(w/v,g/100mL)SDS-PAGE电泳分离后,转移至PVDF膜(MerckMinipore,USA)。为了进行westernblotting检测,将转移至PVDF膜的蛋白先在37℃下与不同一抗分别孵育16小时,再在37℃下与二抗(GADPH,Proteintech,10494-1-AP,使用TBST缓冲液稀释5000倍后使用)孵育1小时;其中,一抗分别为:ATG5(Proteintech,10181-2-AP,使用一抗稀释液稀释1000倍后使用)、ATG12(Proteintech,11122-1-AP,使用一抗稀释液稀释1000倍后使用)、ATG16L1(Proteintech,29445-1-AP,使用一抗稀释液稀释1000倍后使用)和NLRP3(Proteintech,68102-1-Ig,使用一抗稀释液稀释1000倍后使用)。孵育结束后,使用ImageJ软件(美国国立卫生研究院)评估蛋白表达,进一步定量图像中的条带强度,实验结果如图32所示。
1.9、细胞培养及CCK-8测定
将角质形成细胞以每孔1×104个的接种量接种至每孔添加有0.1mL含10%(v/v)胎牛血清、100U/mL青霉素和100U/mL链霉素的DMEM培养基(购自Gibco公司)的96孔板中后,于37℃、5%(v/v)CO2的细胞培养箱中培养16小时。培养结束后,先将GS-9620(M2728,Abmole,美国)、3-MA(3-甲基腺嘌呤,HY-10219,MCE,美国)和雷帕霉素(HY-19312,MCE,美国)分别添加至96孔板中至GS-9620的浓度分别为0.005μM、0.01μM、0.05μM、0.1μM、0.5μM、1μM、5μM和10μM,3-MA的浓度分别为0.005mM、0.01mM、0.05mM、0.1mM、0.5mM、1mM和5mM,雷帕霉素的浓度分别为10mM、0.005μM、0.01μM、0.05μM、0.1μM、0.5μM、1μM、5μM和10μM,于37℃、5%(v/v)CO2的细胞培养箱中孵育24小时,再将CCK-8(购自MCE公司)以每孔10μL的添加量添加至96孔板中,于37℃、5%(v/v)CO2的细胞培养箱中孵育4小时。孵育结束后,使用680型微孔板读卡器(INFINIE E PLEX,TECAN,瑞士)记录每孔在450nm处的吸光度,并根据公式细胞活力=[(As-Ab)/(Ac-Ab)]×100%计算每组细胞的细胞活力,实验结果见图33;式中,As为实验孔吸光度(含细胞、培养基、CCK-8溶液和药物溶液),Ab为对照孔吸光度(含细胞、培养基、CCK-8溶液,不含药物),Ac为空白孔吸光度(含培养基、CCK-8溶液,不含细胞、药物)。
1.10、银屑病角质形成细胞模型的体外诱导及治疗
实验分为7组,7组分别为空白对照组(vehicle)、M5组、M5+GS-9620组、M5+GS-9620+雷帕霉素组、M5+3-MA组、M5+雷帕霉素组、M5+GS-9620+3-MA组,其中,M5组为:将角质形成细胞以每孔1.2×106个的接种量接种至每孔添加有2mL DMEM培养基的6孔板中后,在DMEM培养基中分别加入10ng/mL的重组IL-17A(Bioworld,USA,BK0234)、TNF-a(Bioworld,USA,BK0171)、IL-22(Bioworld,USA,BK0100)、IL-1a(Bioworld,USA,PR1053)和oncostatin-M(Bioworld,USA,PR1104)联合(M5)刺激角质形成细胞,并加入2%(v/v)胎牛血清,于37℃、5%(v/v)CO2的细胞培养箱中培养24小时,以进行银屑病角质形成细胞模型的体外诱导(参照文献:K.Guilloteau,I.Paris,N.Pedretti,K.Boniface,F.Juchaux,V.Huguier,G.Guillet,F.-X.Bernard,J.-C.Lecron,and F.Morel:“Skin Inflammation Induced bythe Synergistic Action ofIL-17A,IL-22,Oncostatin M,IL-1{alpha},and TNF-{alpha}Recapitulates Some Features ofPsoriasis.”J.Immunol.Baltim.Md1950.vol.184,no.9,pp.5263–5270,2010.和X.Teng,Z.Hu,X.Wei,Z.Wang,T.Guan,N.Liu,X.Liu,N.Ye,G.Deng,C.Luo,N.Huang,C.Sun,M.Xu,X.Zhou,H.Deng,C.K.Edwards,X.Chen,X.Wang,K.Cui,Y.Wei,and J.Li:“IL-37ameliorates the inflammatory process inpsoriasis by suppressing proinflammatory cytokine production.”J.Immunol.Baltim.Md 1950.vol.192,no.4,pp.1815–1823,2014.);空白对照组为在M5组的基础上,在DMEM培养基中不添加任何物质;M5+GS-9620组为在M5组的基础上,在DMEM培养基中额外添加0.01μM的GS-9620;M5+GS-9620+雷帕霉素组为在M5组的基础上,在DMEM培养基中额外添加0.01μM的GS-9620和0.05μM的雷帕霉素;M5+3-MA组为在M5组的基础上,在DMEM培养基中额外添加0.1mM的3-MA;M5+雷帕霉素组为在M5组的基础上,在DMEM培养基中0.05μM的雷帕霉素;M5+GS-9620+3-MA组为在M5组的基础上,在DMEM培养基中额外添加0.01μM的GS-9620、0.05μM的雷帕霉素和0.1mM的3-MA。培养24h后,收集细胞,通过westernblot检测各组细胞中ATG5、ATG12、ATG16L1、NLRP3蛋白的表达,同时,收集细胞培养上清,采用ELISA试剂盒检测细胞培养上清中TNF-α(CHE0019,中国四正博生物科技公司)、IL-1β(IBILB0701,美国IBL-America)、IL-6(BE69157,美国IBL-America)、IL-18(BE69171,美国IBL-America)和HMGB1(ARG81351,ariigo生物实验室,中国)的表达水平,实验结果见图34~43。
1.11、统计分析
所有统计分析采用SPSS21.0软件(美国IBM公司)和GraphPadPriam 7.0软件(美国GraphPad软件公司)进行可视化处理。结果以至少三个决定的均数±标准误差(SEM)表示,每对列的比较采用学生t检验,组间差异采用单因素方差分析和事后Tukey检验。以P<0.05为差异有统计学意义。
2、结果
2.1、GS-9620可减轻咪喹莫特诱导的银屑病小鼠模型皮肤炎症
为了验证GS-9620对银屑病的治疗作用,采用咪喹莫特诱导的银屑病小鼠模型,并给予GS-9620治疗。甲氨蝶呤是治疗牛皮癣的传统药物。用甲氨蝶呤作为阳性对照进行了测试。背部皮肤形态学观察如图2所示。可以看到,咪喹莫特组在试验第2天出现红斑、鳞屑、肿胀,临床症状持续恶化至第6天,并伴有体重下降(图3)。经GS-9620或MTX治疗后,临床症状明显减轻,体重增加(图3)。通过PASI总分也可以看出GS-9620或MTX可以减轻临床症状和皮肤炎症(图4)。
2.2、GS-9620能减轻咪喹莫特诱导的银屑病小鼠模型的皮肤炎症
银屑病患者表皮的特征是角化过度和真皮炎症细胞浸润。用H&E对四组实验小鼠的皮肤损伤部位进行染色,发现咪喹莫特组的皮肤与牛皮癣患者的皮肤特征相似。用GS-9620治疗后,与咪喹莫特组相比,组织病理学改变、表皮厚度和炎症细胞浸润明显减少(图5~6)。分析皮肤组织中IL-1β、IL-6、IL-18、HMGB1和TNF-α的水平。与咪喹莫特组比较,GS-9620处理显著降低皮肤组织中IL-1β、IL-6、IL-18、HMGB1、TNF-α水平(图7~11)。上述结果表明,GS-9620可显著减轻咪喹莫特诱导的银屑病小鼠模型的皮肤炎症。
2.3、GS-9620抑制咪喹莫特诱导的银屑病小鼠模型的全身免疫反应
脾脏和淋巴结是免疫器官。当机体的免疫反应增强时,免疫细胞的表达增多,脾脏和淋巴结的大小和重量也相应增大。研究发现,咪喹莫特诱导的银屑病样小鼠模型的脾脏大小、体重和淋巴结重量显著增加。与咪喹莫特组相比,接受GS-9620或MTX治疗的患者脾脏大小、重量和淋巴结重量均减少(图12~14)。推测GS-9620可以减轻咪喹莫特诱导的银屑病样小鼠模型的全身炎症。测量四组小鼠脾脏和淋巴结中CD3+、CD4+、CD8+和CD4+CD25+、CD4+IL-17A+和CD4+IFN-γ+细胞百分比。经GS-9620或MTX治疗后,与咪喹莫特组相比,CD3+、CD4+、CD8+和CD4+CD25+、CD4+IL-17A+和CD4+IFN-γ+细胞的百分比明显降低(图15~30)。这些数据表明GS-9620可以调节咪喹莫特诱导的牛皮癣样小鼠的全身免疫反应。
2.4、GS-9620促进自噬相关蛋白的表达
自噬是维持细胞和生物体稳态的核心分子途径。它在细胞生理中起着至关重要的作用,包括适应代谢应激,清除危险物质(如蛋白质聚集体、受损的细胞器和细胞内病原体),在分化和发育过程中更新,以及防止基因组损伤。使用免疫组织化学染色检测自噬相关蛋白ATG12、ATG5和ATG16L1在健康和银屑病患者皮肤样本中的表达。发现自噬相关蛋白在健康人皮肤表皮中高度表达,但在牛皮癣表皮中被抑制(图31)。上述实验在小鼠身上进行,正常小鼠表皮中自噬相关蛋白的表达较高,而咪喹莫特诱导的银屑病样小鼠中自噬相关蛋白的表达受到抑制,但经GS-9620处理后,自噬相关蛋白的表达增加(图32)。westernblot进行半定量验证,结果与免疫组化结果一致(图32)。推测GS-9620促进角质形成细胞自噬,具有抗炎作用。
2.5、GS-9620调节角质形成细胞的自噬和炎症体,具有抗炎作用
使用CCK-8测定不影响角质形成细胞细胞活力的GS-9620、GS-9620+3-MA和GS-9620+雷帕霉素的终浓度(图33)。利用IL-17A、TNF-a、IL-22、IL-1a和oncostatin-M(M5)刺激角质形成细胞体外构建银屑病样角质形成细胞模型。这五种炎症因子与银屑病的发病机制密切相关。将实验组设为:空白对照组(vehicle)、M5组、M5+GS-9620组、M5+GS-9620+雷帕霉素组、M5+3-MA组、M5+雷帕霉素组、M5+GS-9620+3-MA组。共培养24小时后检测到M5刺激角质形成细胞后,自噬相关蛋白ATG12、ATG5、ATG16L1的表达降低,炎性小体NLRP3的表达升高。而给GS-9620后,ATG12、ATG5、ATG16L1表达升高,NLRP3表达降低。接下来,研究GS-9620是否通过调节自噬来影响炎性小体NLRP3的表达。分别给予自噬抑制剂3-MA和自噬诱导剂雷帕霉素进行实验。发现M5+GS-9620+雷帕霉素对NLRP3的抑制作用更强,M5+3-MA对NLRP3表达的上调作用更强(图34~38)。检测细胞培养液中相关细胞因子IL-1β、IL-6、IL-18、HMGB、TNF-a,结果与NLRP3的变化趋势一致(图39~43)。以上结果表明,GS-9620通过促进角质形成细胞自噬发挥抗炎作用
综合图1~34的结果可知,综合图1~34的结果可知,GS-9620能够显著改善咪喹莫特(imiquimod,IMQ)诱导的银屑病小鼠模型的皮肤损伤程度,降低银屑病小鼠模型皮肤组织中炎症细胞因子(IL-1β、IL-6、IL-18、HMGB和TNF-α)的表达,降低银屑病小鼠模型脾脏和淋巴结中T淋巴细胞(CD3+、CD4+CD25+、CD4+IL-17A+和CD4+INF-γ+)的侵袭,增加银屑病小鼠模型皮肤组织中自噬相关蛋白(ATG5、ATG12和ATG16L1)的表达,同时,GS-9620能够显著增加M5(IL-1α、IL-17A、IL-22、抑瘤素M和肿瘤坏死因子-α)诱导的银屑病样角质形成细胞模型中自噬相关蛋白的表达,抑制银屑病角质形成细胞模型中炎症小体(NLRP3)的产生,抑制银屑病角质形成细胞模型中炎症细胞因子的表达,即GS-9620能够通过促进角质形成细胞的自噬,抑制角质形成细胞中NLRP3炎症小体的激活,从而抑制角质形成细胞中炎症细胞因子的表达,进而达到减轻银屑病引起的皮肤炎症的目的。
显然,上述实施例仅仅是为清楚地说明所作的举例,而并非对实施方式的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式的变化或变动。这里无需也无法对所有的实施方式予以穷举。而由此所引伸出的显而易见的变化或变动仍处于本发明创造的保护范围之中。
Claims (10)
1.GS-9620在制备预防/或治疗银屑病的药物中的应用。
2.如权利要求1所述的应用,其特征在于,所述预防/或治疗银屑病包括通过促进角质形成细胞的自噬来减轻银屑病引起的皮肤炎症。
3.如权利要求1或2所述的应用,其特征在于,所述药物的成分包含活性组分以及药学上可接受的载体;所述活性组分包含GS-9620。
4.如权利要求3所述的应用,其特征在于,所述药学上可接受的载体包括表面活性剂、赋形剂、稳定剂、助悬剂、等渗剂、防腐剂、填充剂、崩解剂、抗氧剂、乳化剂、包衣剂、黏合剂、润滑剂和/或矫味剂。
5.如权利要求1~4任一项所述的应用,其特征在于,所述药物的剂型为散剂、片剂、颗粒剂、胶囊剂、口服液体剂或注射剂;所述口服液体剂为溶液剂、糖浆剂、乳剂或混悬剂。
6.一种预防/或治疗银屑病的药物,其特征在于,所述药物含有活性组分;所述活性组分包含GS-9620。
7.如权利要求6所述的药物,其特征在于,所述预防/或治疗银屑病包括通过促进角质形成细胞的自噬来减轻银屑病引起的皮肤炎症。
8.如权利要求6或7所述的药物,其特征在于,所述药物的成分还包含药学上可接受的载体。
9.如权利要求8所述的药物,其特征在于,所述药学上可接受的载体包括表面活性剂、赋形剂、稳定剂、助悬剂、等渗剂、防腐剂、填充剂、崩解剂、抗氧剂、乳化剂、包衣剂、黏合剂、润滑剂和/或矫味剂。
10.如权利要求6~9任一项所述的药物,其特征在于,所述药物的剂型为散剂、片剂、颗粒剂、胶囊剂、口服液体剂或注射剂;所述口服液体剂为溶液剂、糖浆剂、乳剂或混悬剂。
