Abstract
The invention discloses a hair-growing and nourishing composition, and a preparation method and application thereof. The composition comprises an active ingredient and a solvent, wherein the mass ratio of the active ingredient to the solvent is 1 (15-30); the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate according to the mass ratio of (0.1-1) to (1-5); the solvent is a compound solvent and consists of deionized water, an alcohol solvent and a solubilizer according to the mass ratio of (40-60), 20-30 and 1-3. The invention discovers that three components of apigenin, polydatin and yeast/mother-of-pearl fermentation lysate filtrate have synergistic effect in the aspects of hair growth and hair benefiting. The composition stimulates hair growth by various biological means, thereby increasing hair thickness, and has remarkable hair growth promoting and hair nourishing effects, low irritation, and no side effects.
Description
Technical Field
The invention belongs to the technical field of hair care agents, and particularly relates to a hair-growing and hair-nourishing composition, and a preparation method and application thereof.
Background
Alopecia is a phenomenon of hair loss, which is one of the more common dermatological diseases. With the increasing pressure of modern people on work and life, the influence of environmental factors is increasing, the number of people suffering from alopecia is increasing in recent years, and the people suffering from alopecia are younger.
Alopecia is mainly classified into androgenetic alopecia, alopecia areata, anagen alopecia, telogen alopecia, and chemical/physical alopecia. Among them, androgenetic alopecia (Androgenic alopecia, AGA) is the most common type of alopecia in clinic, also called seborrheic alopecia, accounting for 90% of the population suffering from alopecia, which is common to men and women, and is common to men. AGA onset may be related to genetic factors, fungal infections, poor eating habits, high pressure of life and work, etc. In addition, hair follicles at the top of the head of the person suffering from AGA alopecia are sensitive to androgens, and 5 alpha-reductase activity is high, so that testosterone in the alopecia area is converted into Dihydrotestosterone (DHT) to be increased, and the DHT can shrink and degenerate when acting on the hair follicles, so that the growing period of the hair is shortened, and the proportion of hairs in the telogen period is increased.
In the alopecia treatment, the western medicines commonly used in clinic are minoxidil and finasteride, but the two medicines are easy to relapse after stopping taking, and can also cause side effects such as skin irritation, scalp itching, facial and hand hirsutism, dizziness, anorexia, tachycardia and the like. In addition, although hair transplantation has a certain therapeutic effect, the clinical application of the hair transplantation is limited due to the large surgical trauma and high cost and the need of multiple surgeries. The effect of non-drug treatment such as acupuncture is also not ideal. At present, research reports on alopecia treatment by using plant extracts such as traditional Chinese medicines are more and more, and plant source components are relatively safe and effective and are convenient to use. For example, the single or mixed use of astragalus root, fleece-flower root, glossy privet fruit, dodder seed and the like has the effects of stimulating the growth of hair follicles of mice cultivated in vitro, increasing the number of hair follicles, accelerating the hair regeneration speed and the like, however, the simple treatment of the traditional Chinese medicine extract has long treatment period and slow effect, and often causes physiological alopecia when the hair regeneration is carried out, and the effect of comprehensively improving the alopecia is also poor when the speed of promoting the hair regeneration of the traditional Chinese medicine extract is lower than that of the physiological alopecia.
Disclosure of Invention
Aiming at the problems of large side effect, poor effect and the like of the existing cosmetics with the functions of hair growth and hair loss prevention, the invention provides a hair growth and hair benefiting composition which stimulates hair growth through various biological ways, thereby increasing the thickness of hair, having remarkable hair growth and hair benefiting effects, low irritation and no side effect.
In order to achieve the above object, the present invention provides a hair growth and benefit composition comprising an active ingredient and a solvent; the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate according to the mass ratio of (0.1-1) to (1-5).
Preferably, the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate according to the mass ratio of (0.2-0.8) (0.5-1) (2-5).
More preferably, the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate according to the mass ratio of 0.6:1:4.
Preferably, the yeast fermentation product filtrate is yeast/mother-of-pearl fermentation lysate filtrate
In the invention, the yeast/mother-of-pearl fermentation lysate filtrate can be a commercial product, can be purchased from Shanghai Yu New Material science and technology Co-Ltd, and can also be prepared by self according to the method described in patent ZL 202110773653.3.
Preferably, the solvent is a compound solvent, and consists of deionized water, an alcohol solvent and a solubilizer according to the mass ratio of (40-60) (20-30) (1-3).
Preferably, the mass ratio of the active ingredient to the solvent is 1 (15-30).
Preferably, the alcohol solvent includes, but is not limited to, one or more of ethanol, propylene glycol, glycerol, 1, 3-butanediol, 1, 2-hexanediol, polyethylene glycol, and dipropylene glycol.
Preferably, the solubilizer comprises one or more of, but not limited to, jojoba wax PEG-120 esters, fatty alcohol polyoxyethylene ethers, POE castor oil or hydrogenated castor oil derivatives, fatty acid esters, sucrose fatty acid esters or alkyl glycosides.
The invention also aims to provide a preparation method of the hair-growing and hair-nourishing composition, which comprises the following steps: the active ingredients with the formula amount are respectively weighed, dissolved in a solvent and stirred for dissolution, and the composition is obtained.
It is another object of the present invention to provide the use of the hair tonic composition described above for the preparation of a formulation for promoting hair growth. The preparation comprises a medicine or a cosmetic.
Preferably, the cosmetic is a essence, shampoo, cream, emulsion or conditioner.
Preferably, the composition is added to the cosmetic in an amount of 5 to 15wt%.
In the present invention, apigenin (apigenin) is a poorly soluble flavonoid substance, which is a component known to be contained in various plants and to exhibit an antioxidant effect, or brain cell production and protective effect. It has been shown that apigenin has the effect of alleviating hair loss and stimulating hair growth via various biological pathways, thereby increasing hair thickness, reducing the appearance of white hair, and providing a full hair appearance. For example, apigenin can prolong the anagen phase of hair follicles, prevent alopecia and promote hair growth by inhibiting 5α -reductase.
In the invention, polydatin (PD) is a natural antioxidant with various pharmacological activities, has the beneficial effects of resisting oxidative stress and inflammation, and can promote the activity and proliferation capacity of hair follicle cells by eliminating inflammation caused by excessive free radicals, thereby ensuring the normal proliferation and differentiation of the cells. It also regulates the proliferation and development pattern of hair follicles by activating PI3K/Akt signaling pathway, up-regulating Shh protein expression, and promoting hair growth.
In the invention, the yeast/mother-of-pearl fermentation lysate filtrate has good effects of balancing grease, moisturizing and repairing, can effectively regulate excessive scalp sebum, relieve symptoms of seborrheic alopecia, and promote hair growth by repairing damaged hair follicle cells. Compared with the yeast fermentation lysate filtrate, the yeast/mother-of-pearl fermentation lysate filtrate is rich in microelements, amino acids, calcium phosphate and other substances decomposed by mother-of-pearl, can accelerate cell growth, promote protein synthesis and promote hair growth.
The invention has the following beneficial effects:
the invention discovers that three components of apigenin, polydatin and yeast/mother-of-pearl fermentation lysate filtrate have synergistic effect in the aspects of hair growth and hair benefiting. The composition stimulates hair growth by various biological means, thereby increasing hair thickness, and has remarkable hair growth promoting and hair nourishing effects, low irritation, and no side effects.
Detailed Description
The following examples are further illustrative of the invention and are not intended to be limiting thereof. The components of the formulations in the examples below, unless otherwise specified, are all conventional commercial products.
Example 1: preparation of hair-growing and nourishing composition
Weighing the raw materials according to the following weight ratio, adding apigenin, polydatin and yeast/mother-of-pearl fermentation lysate filtrate into a mixed solvent consisting of deionized water, ethanol, glycerol and water-soluble jojoba ester, and stirring for dissolution to obtain the composition.
Comparative example 1: preparation of hair-growing and nourishing composition
The following raw materials were weighed in the following weight ratio, and a composition was prepared in accordance with the method of example 1.
Comparative example 2: preparation of hair-growing and nourishing composition
(1) Taking 20g of ethanol, 10g of glycerol, 3g of water-soluble jojoba ester and 60g of deionized water, and stirring and uniformly mixing to obtain a mixed solvent;
(2) Adding 0.6g of apigenin, 1g of polydatin and 4g of yeast fermentation lysate filtrate into the mixed solvent in the step (1), and stirring for dissolution to obtain the composition.
The yeast fermentation lysate filtrate was purchased from Shanghai Bilai chemical engineering Co.
Example 2: allergy test
The samples a to E prepared in example 1 of the present invention were tested with reference to the requirements and measurement methods related to the "human skin patch test" in the 2015 edition "cosmetic hygiene Specification", issued by the Ministry of health, and skin reactions were observed at 0h, 6h, 12h, 24h, 48h, respectively, and all experimental results of volunteer subjects were negative results, with no skin adverse reaction. The hair-growing and hair-nourishing composition provided by the invention can not cause anaphylactic reaction, has low irritation and no side effect.
Example 3 evaluation of pilatory Effect
1. Test animals: c57BL/6 male mice with the weight of 18-22g for 6-8 weeks are selected, and 104 mice are taken.
2. The test method comprises the following steps: after all mice were adapted to 1 week of feeding, they were divided into a negative control group, a positive control group, sample a to E groups, comparative sample F to J groups and comparative example 2 group, 13 groups of each group, 8 mice of each group were lightly anesthetized, a 2×2cm area was selected as a test area on the back, and a rosin/paraffin mixture (mass ratio 1:1) was heated and melted and then applied to the test area when the temperature was appropriate, and after solidification, pulled down to induce growth of hair in mice from rest period to growth period. Starting on the next day, samples were uniformly applied to the dehairing areas of each group, the negative control group was given a mixed solvent (mixed solvent of sample C), the positive control group was given an equal volume of minoxidil solution (1.5 wt%) and the samples A to E were given an equal volume of sample A to E solutions, the comparative samples F to J were given an equal volume of comparative sample F to J solutions, the comparative example 2 was given an equal volume of the composition solution of comparative example 2, 2 times per day, 0.1 mL/time, and 28d continuously. The hair growth cycle of the plucked area was observed, the time required for the skin color at the plucked area to change from pink (resting period) to black (growing period) and gray (retrogradation period) was recorded, and the average value was taken, and the results are shown in table 1. Each group was treated with 10 new hairs from the plucking zone at random on the last day of the application of the sample, the lengths were measured under a magnifying glass with a vernier caliper and recorded, the farthest distances at both ends of the hairs were recorded as the hair lengths, and the average value was calculated, and the results are shown in table 1.

The results show that compared with the negative control group, the time for the hair follicle growth of the mice using the hair growth and hair benefiting composition (samples A to E) provided by the invention to enter the growing period is advanced, and the effect is equivalent to that of the positive control group. Compared with a negative control group, the time delay for the growth of the hair follicles of the mice, which are prepared by using the hair-growing and hair-nourishing composition (samples A to E) provided by the invention, to enter the withdrawal period is slightly better than that of the positive control group. In general, the duration of the growth period of the hair follicles of the mice using the hair-growing and hair-nourishing composition (samples A to E) provided by the invention is obviously better than that of a negative control group, and the result shows that the hair-growing and hair-nourishing composition provided by the invention has the effect of obviously prolonging the growth period of the hair follicles, and the effect is slightly better than that of a positive control group. As can be seen from comparative samples F-J, the effect of the hair growth promoting and hair nourishing composition provided by the invention for prolonging the growing period of hair follicles is obviously better than the effect of the composition used by a single component or two components, and the result shows that three components, namely apigenin, polydatin and yeast/mother-of-pearl fermentation lysate filtrate, have a synergistic effect in the aspect of prolonging the growing period of hair follicles. From comparative example 2, it is assumed that the yeast/mother-of-pearl fermentation lysate filtrate has better complexation with apigenin and polydatin than the yeast fermentation lysate filtrate, and this is because the yeast/mother-of-pearl fermentation lysate filtrate is rich in trace elements, amino acids, calcium phosphate and the like decomposed by mother-of-pearl, and the growth period of hair follicles is more effectively prolonged.
In addition, the average hair length of the hair-growing and hair-nourishing composition (samples A to E) provided by the invention is obviously better than that of a negative control group and slightly better than that of a positive control group, and the result shows that the hair-growing and hair-nourishing composition provided by the invention has an obvious effect of promoting hair growth. As can be seen from comparative samples F-J, the hair growth promoting and nourishing composition provided by the invention has a significantly better effect than the single component or the combined use of two components of the composition, and the results show that three components, namely apigenin, polydatin and yeast/mother-of-pearl fermentation lysate filtrate, have a synergistic effect in promoting hair growth. From comparative example 2, it is understood that the yeast/mother-of-pearl fermentation lysate filtrate has better complexation with apigenin and polydatin than the yeast fermentation lysate filtrate, presumably because the yeast/mother-of-pearl fermentation lysate filtrate is rich in trace elements, amino acids, calcium phosphate and the like decomposed by mother-of-pearl, which can accelerate cell growth, promote protein synthesis and promote hair growth.
The foregoing is merely a preferred embodiment of the present invention, and it should be noted that the above-mentioned preferred embodiment should not be construed as limiting the invention, and the scope of the invention should be defined by the appended claims. It will be apparent to those skilled in the art that various modifications and adaptations can be made without departing from the spirit and scope of the invention, and such modifications and adaptations are intended to be comprehended within the scope of the invention.
Claims (10)
1. A hair growth and benefit composition, characterized in that the composition comprises an active ingredient and a solvent; the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate according to the mass ratio of (0.1-1) to (1-5).
2. The hair-growing and hair-nourishing composition according to claim 1, wherein the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate in a mass ratio of (0.2-0.8) (0.5-1) (2-5).
3. The hair growth and benefit composition according to claim 2, wherein the active ingredients consist of apigenin, polydatin and yeast fermentation product filtrate in a mass ratio of 0.6:1:4.
4. A hair growth promoting and hair nourishing composition according to any one of claims 1 to 3 wherein the yeast fermentation product filtrate is a yeast/mother of pearl fermentation lysate filtrate.
5. The hair growth and benefit composition according to claim 1, wherein the solvent is a compound solvent and consists of deionized water, an alcohol solvent and a solubilizer according to the mass ratio of (40-60) (20-30) (1-3).
6. The hair-growing and benefit composition according to claim 1, wherein the mass ratio of the active ingredient to the solvent is 1 (15-30).
7. A method of preparing a hair tonic composition according to any one of claims 1 to 6, comprising the steps of:
the active ingredients with the formula amount are respectively weighed, dissolved in a solvent and stirred for dissolution, and the composition is obtained.
8. Use of a hair tonic composition according to any one of claims 1 to 6 in the preparation of a formulation for promoting hair growth.
9. The use according to claim 8, wherein the formulation comprises a pharmaceutical or cosmetic product.
10. The use according to claim 9, wherein the cosmetic is a concentrate, a shampoo, a cream, an emulsion or a conditioner.
