Description

Composition for preventing alopecia and promoting hair growth
Technical Field
The present application claims priority based on korean patent application No. 10-2020-0086878, filed on 7 months and 14 days in 2020, and the entire contents of the specification of the related application and the disclosure in the accompanying drawings are incorporated herein by reference.
The present invention relates to a composition for preventing hair loss and promoting hair growth.
Background
With the change of the age, hair occupies an important part in the process of increasing attention to beauty. Hair is a fine and keratinized structure produced on the surface of the skin. The multifunctional external shock absorber has the buffering effect on external shock, plays a role of protecting a human body from external stimuli such as direct light, cold, friction, danger and the like, and discharges heavy metals such as arsenic, mercury, zinc and the like harmful to the body to the outside, and has the beauty aspect of modern and emphasized decoration. However, there are increasing numbers of people complaining of hair loss for various reasons including changes in dietary life and increased internal and external stress. It is known that genetic factors are the most important cause of hair loss, but as described above, increase in social stress, environmental pollution, frequent permanent waving and dyeing, and erroneous scalp management are also becoming causes of hair loss.
Human hair is about 10-15 tens of thousands, each hair having a different growth cycle from the others. The formation of the hair follicle (hair follicle) occurs under the action of β -catenin to form a basal plate (placode) from basal epidermal cells (Sarah E.Miller, 2002). Dermal fibroblasts receiving signals from the formed substrate form skin aggregates (dermal condensate). At the same time, the substrate grows and sinks into the dermis and combines with the skin aggregate to form a primary blank (primary hair germ). The epidermal cells forming the hair embryo grow long in the direction of the dermis to form a long columnar structure composed of multiple layers, i.e., hair peg, and the skin aggregate is formed of a spherical hair papilla (dermal papilla). The distal end of the hair pin gradually becomes thicker to form hair bulb (hair bulb), and the hair bulb forms a form of wrapping the hair papilla, so that hair follicle is completed, and epidermal cells inside the hair follicle are differentiated to generate hair.
The initially generated hair circulates the following period: the hair enters the catagen stage and the telogen stage after the anagen stage, and then enters the anagen stage again after the hair falls off. The hair growth cycle consists of three phases, classified as the anagen phase (anagen stage) where hair growth is most active, the catagen phase (catagen stage) where hair begins to degenerate, and the telogen stage where hair growth is stopped or enters rest.
It is well known that in regulating the growth cycle of such hair, the activity of external root sheath cells (outer root sheath cell) including papilla cells and follicle stem cells constituting hair follicles, various cytokines and growth factors produced in these cells play an important role. For example, DKK-1 (Dickkopf-1) plays an important role in hair follicle growth inhibition and killing. On the other hand, it is well known that for periodic hair growth, regeneration of hair follicles and activation of stem cells must be achieved, which is achieved by Wnt/β -catenin production and activation. In addition, growth factors such as VEGF, IGF-1, FGF, etc. are known to promote the anagen phase of hair, and TGFb-1, EGF, etc. are known to induce the catagen phase of hair follicles.
In general, alopecia refers to an abnormal increase in the number of hair loss due to an increase in the number of hair loss in the catagen phase, which is caused by a decrease in the proportion of hair in the anagen phase and an increase in the telogen phase in such a cycle. Alopecia is a normal phenomenon, but in normal people, there are many hairs in anagen phase, whereas in people with ordinary alopecia, there are many hairs in telogen phase, so that macroscopic alopecia occurs. People suffering from alopecia are characterized by miniaturization of hair. With the progress of alopecia, the time of the growing period is shortened, the transition to the retrograde period and the resting period is promoted, and then the volume of the hair papilla is reduced, and the hair follicle is gradually miniaturized. Therefore, in order to treat alopecia, it is important to rapidly transition the telogen state hair follicle to anagen phase and lengthen the shortened anagen phase.
As described above, as causes of alopecia, there are proposed male hormonal effects, hypersecretion of sebum, poor blood circulation, scalp function decline due to peroxides, bacteria, etc., genetic factors, aging, stress, etc. However, it is in a state that the clear cause of hair loss has not been revealed so far, and it is known that interleaving has a very complex effect.
For such various and complicated causes of alopecia, known alopecia preventing products include, as active ingredients, ingredients for the purpose of promoting blood circulation, strengthening hair root function, moisturizing scalp effect, preventing dandruff effect, antioxidant effect, hair growth period prolonging effect, inhibiting male hormonal effect, etc., but the effects are in a state where no significant effect has been achieved so far, and there is also a problem of side effects.
On the other hand, adenosine is a constituent of amino acids produced in cells, and is a major component of cellular energy metabolism. Adenosine is a compound in which a base called adenine and a sugar called ribose are combined as one of the elements constituting the gene DNA. For adenosine, if three phosphates bind to a sugar, it becomes the main source of intracellular energy, namely ATP (adenosine-5' -triphosphate). Thus, it is known that adenosine is an important element that plays a decisive role in the smooth progress of all metabolism while constituting the body. Adenosine is an intracellular component, and thus has excellent stability and durability when penetrating the skin, and promotes synthesis of DNA and protein in the dermis layer, thereby acting in addition to improving skin wrinkles.
Nicotinamide (Niacinamide) is a vitamin B complex, which is present in the plant kingdom and widely distributed with niacin, and exists as a component of redox coenzyme NAD or NADP in the animal kingdom organism. Nicotinamide is a physiologically active amide of vitamin B3, also known as pyridine-3-carboxylic acid, which is also known to act as an anti-inflammatory material or antioxidant. Nicotinamide blocks migration of melanosomes, thereby blocking migration of synthesized melanin to keratinocytes, thereby reducing skin pigmentation, enhancing whitening effect. In addition, nicotinamide increases collagen synthesis and intercellular lipid synthesis, thereby contributing to skin moisturization and the like.
In addition, D-panthenol (dexpanthenol) is an alcohol form of vitamin B5 (Pantothenic acid), which may be referred to as provitamin (provitamin) B5. D-panthenol is immediately after being absorbed by the skin and converted into vitamin B5 for use. D-panthenol is widely used as a component of skin moisturizer, wrinkle improver, etc. in cosmetics and living goods, and is also used as an ointment for wound healing, skin burn, itch improvement, etc. In addition, D-panthenol is known as a humectant having permeability, and is known to have effects of conditioning skin and hair and increasing recovery of damaged hair.
In addition, polygonum multiflorum is a perennial herb belonging to the Polygonaceae family, which is a vine medicinal plant and its discipline names are varied as Pleuropterus multiflorus, polygonum multiflorum, fagopyrum multiflorum, reynoutria multiflora or Fallopia multiflora. It is well known that polygonum multiflorum extract is used as a tonic, a seminal agent, a menstrual flow-promoting agent, a blood-nourishing agent or an antitussive agent, etc., and is used for treating cold, hematemesis, neurasthenia or arthritis, etc.
In addition, houttuynia cordata (Houttuynia cordata) is known as a perennial herb of the family Saururaceae as a herb of the genus Fagopyrum, a heavy herb, houttuynia cordata or ten herbs. Herba Houttuyniae contains various essential oil components, and emits unique smell such as fishy smell, so it is called herba Houttuyniae. Herba Houttuyniae flowers mainly in 5-7 months, and is a herb for treating gonorrhea, enteritis, urinary tract infection, pneumonia or bronchitis.
In addition, zinc pyrithione (zinc pyrithione) has an antibacterial effect, thus preventing the proliferation of microorganisms on the scalp and skin. In particular, zinc pyrithione has a proliferation inhibiting effect on malassezia (malassezia) and other fungi, and thus plays a role in preventing seborrheic dermatitis and dandruff formation.
In addition, salicylic acid is a major component of aspirin, and has been widely used as an anti-inflammatory agent, an analgesic agent, or a antipyretic agent. Salicylic acid plays a role in removing skin cutin and softening cutin. Salicylic acid is mainly added to cosmetics such as cleansing foam (cleansing foam), and softens and exfoliates keratin, thereby softening pores. Thus, salicylic acid can help the removal of sebum from pores, keeping the skin clean. In addition, salicylic acid has antibacterial and anti-inflammatory effects, and can inhibit proliferation of scalp or skin microorganisms, thereby improving skin and scalp or preventing dandruff.
On the other hand, the current alopecia therapeutic agent of the coating agent approved by the U.S. Food and Drug Administration (FDA) is minoxidil (2, 4-diamino-6-piperidylpyrimidine-3-oxide, 2, 4-diamido-6-piperidinopyridine-3-oxide) only, but the effect thereof is not ideal at 32 to 48 weeks, 38% and 13% respectively, and has the disadvantage of poor hair treatment effect.
Therefore, there is a need for developing a composition excellent in preventing hair loss or promoting hair growth, but there is no research on what effect is produced in preventing hair loss or promoting hair growth when the above adenosine, D-panthenol, nicotinamide, polygonum multiflorum extract, houttuynia cordata extract, zinc pyrithione, salicylic acid and the like are combined.

Experimental example 2: promoting effect of mitochondrial Activity of papilla cells
Human hair papilla cells (dermal papilla cells, dermal papilla cells, DPCs) were purchased from PromoCell company. The DPCs were treated with 5% CO at 37℃in DMEM (Hyclone Inc, UT, USA) containing 5% fetal bovine serum (FBS; gibco, N.Y., USA), penicillin 100unit/mL and streptomycin 100. Mu.g/mL 2 Culturing under the condition. The cultured DPC s 3,000 cells/well were plated in 96-well plates and then cultured under 0.1% serum conditions for 24 hours. Thereafter, in cultured cells, DMSO (vehicle) diluted at 1:1000 in serum-free DMEM was used as a control group, minoxidil 2 μg/ml was used as a positive control group, and one or more of Polygoni Multiflori radix extract, herba Houttuyniae extract, zinc pyrithione, salicylic acid in a mixture of adenosine, D-panthenol, nicotinamide was treated for 1 day as shown in Table 5 belowAnd incubation was performed. After incubation, the extent of proliferation of cells was assessed using the JC-1 method. JC-1 was treated at 1:100 in culture and incubated for 1 hour. After 1 hour, fluorescence (fluorescence) was measured for each well at 590nm and 530 nm. All experiments were repeated 3 times and the mean of the fluorescence values was calculated. For the above results, the control group was set to 100, which is expressed as a percentage of the treatment group relative to the control group.
TABLE 5
Figure BDA0004113795020000121
From the results disclosed in table 5 above, it was confirmed that when one or more of polygonum multiflorum extract, houttuynia cordata extract, zinc pyrithione, salicylic acid was further contained in the mixture of adenosine, D-panthenol and nicotinamide and treated, a remarkable synergistic effect was produced in enhancing mitochondrial activity of hair papilla cells.
Experimental example 3: modulation of hair papilla cell Activity Signal (Wnt/beta-catenin)
In general, wnt/β -catenin signaling activated in the hair papilla occurs when the growth cycle of the hair transitions from the catagen phase to the active phase, i.e., during the entire active phase from the beginning of hair growth to the occurrence of hair growth. In telogen and catagen phases, wnt/β -catenin signaling decreases or disappears, and hair follicle degeneration and hair loss occur. Thus, it was confirmed by this experiment how much one or more treatment groups of polygonum multiflorum extract, houttuynia cordata extract, zinc pyrithione, salicylic acid in the mixture of adenosine, D-panthenol, nicotinamide of the present disclosure contributed to Wnt/β -catenin signal amplification.
Wnt3a protein was used as a positive control group for amplifying Wnt/β -catenin signal, and DMSO was used as a negative control group. Wnt Reporter HEK293A cells 3X10 in culture 96-well plates 4 Inoculating into each well, and then treating Polygoni Multiflori radix extract, herba Houttuyniae extract, zinc pyrithione, and salicylic acid in the mixture of adenosine, D-panthenol, and nicotinamide of the present disclosure as shown in Table 6 belowMore than one kind of acid. Then, report analysis (reporter assay) was performed using a luciferase assay kit (Luciferase assay kit) from Promega corporation (E1960). The Wnt/β -catenin promoter activity was measured according to the experimental method of manufacturing company and using photometer Victor (luminometer Victor) (perkin elmer, walthamm, ma, USA).
TABLE 6
Figure BDA0004113795020000131
From the results disclosed in table 6 above, it was confirmed that when one or more of polygonum multiflorum extract, houttuynia cordata extract, zinc pyrithione, salicylic acid was further contained in the mixture of adenosine, D-panthenol and nicotinamide and treated, the activity of Wnt/β -catenin signaling system was synergistically enhanced. Since Wnt/β -catenin signaling is a hair growth promoting signaling of hair follicle stem cells, it can be seen from the above experimental results that the composition of the present disclosure exhibits a remarkable ascending effect in promoting hair growth.
