Abstract
本发明公开了一种人工淋巴结原基的构建方法及应用。所述构建方法包含以下步骤:S1:制备永生化的脾原代基质细胞;S2:将永生化的脾原代基质细胞进行脱细胞处理得到脱细胞支架;S3:在脱细胞支架中添加间充质干细胞和淋巴组织诱导细胞进行混合培养后即形成人工淋巴结原基。本发明构建的人工淋巴结原基复刻了胚胎期淋巴结发生的生理过程,所得结构和功能更加完整,制备过程中不涉及异种蛋白,降低机体免疫和炎症风暴的风险。而且将人工淋巴结原基移植体内能较好地招募淋巴细胞,支持其增殖活化,并将活化的细胞再次输出至病灶局部发挥功能;可以达到恢复淋巴结缺陷的转基因小鼠乳腺癌荷瘤条件下的局部免疫清除功能的效果。

技术领域
本发明属于类器官培养技术领域,具体涉及一种人工淋巴结原基的构建方法及应用。
背景技术
现有的人工淋巴结构相关报道主要分为两种技术模型,肾包膜下移植的模型,一是利用慢病毒修饰基因表达的基质细胞系和骨髓来源的DC细胞混合用海绵胶原支架,进行肾包膜下移植;另一种是利用缓释蛋白包裹的趋化因子放入海绵胶原支架进行肾包膜下移植;皮下移植的模型是利用淋巴结原代分离的基质细胞和脱细胞支架进行体外打包构建。以上技术均需要用到异种蛋白支架结构;需要大量的原代分离和培养扩增工作;所制备的人工淋巴结构功能不成熟。
发明内容
本发明的目的在于提供一种人工淋巴结原基的构建方法及其应用。
本发明所采取的技术方案是:
本发明的第一方面,提供一种人工淋巴结原基的构建方法,包含以下步骤:
S1:制备永生化的脾原代基质细胞;
S2:将永生化的脾原代基质细胞进行脱细胞处理得到脱细胞支架;
S3:在脱细胞支架中添加间充质干细胞和淋巴组织诱导细胞进行混合培养后即形成人工淋巴结原基。
在本发明的一些实施方式中,所述步骤S1中的永生化的脾原代基质细胞的构建方法包括:将脾原代基质细胞进行慢病毒转染,获得转染后的脾原代基质细胞;将转染后的脾原代基质细胞通过嘌呤霉素筛选,对嘌呤产生耐药的细胞即为永生化的脾基质细胞。
在本发明的一些实施方式中,所述慢病毒包括SV40、hTERT。
优选地,包括使用慢病毒SV40中的T抗原进行转染;优选为SV40中的大T或小T抗原。
在本发明的一些实施方式中,所述病毒转染的MOI=15~20。
在本发明的一些实施方式中,所述慢病毒转染的时间为16~18h。
在本发明的一些实施方式中,所述嘌呤霉素的浓度为3~7ug/ml。
在本发明的一些实施方式中,本发明并不对永生化处理的方式做具体限定,常用的永生化处理方式都可以用于本发明,达到相似的效果。
在本发明的一些实施方式中,步骤S2的具体方法包括:
(1)在培养皿中分别添加细胞贴壁促进剂、交联剂、氨基酸溶液进行处理,然后加入完全培养基对培养皿进行预处理;
(2)消化永生化的脾原代基质细胞,然后种板至步骤(1)中所述的预处理后的培养皿;
(3)当细胞单层融合后,更换成包含L-抗坏血酸的培养基,培养14~21天;
(4)加入细胞裂解液清除多余细胞,然后加入DNaseI溶液去除残余的细胞DNA,即为脱细胞支架。
在本发明的一些实施方式中,所述细胞贴壁促进剂包括明胶。
在本发明的一些实施方式中,所述明胶溶液浓度为0.1~0.3%,处理条件为35~39℃,0.5~1.5h。

本发明提供了一种人工淋巴结原基的构建方法,所述构建方法包含以下步骤:S1:制备永生化的脾原代基质细胞;S2:将永生化的脾原代基质细胞进行脱细胞处理得到脱细胞支架;S3:在脱细胞支架中添加间充质干细胞和淋巴组织诱导细胞进行混合培养后即形成人工淋巴结原基。
本发明从诱导多能干细胞诱导淋巴组织诱导细胞制备人工淋巴结原基,其中诱导的淋巴组织诱导细胞(lymphoid tissue inducer cell,LTiC)是淋巴组织结构形成过程的启动者,通过其膜结合形式的淋巴毒素复合物(lymphotoxinα1β2,LTα1β2)与局部间充质细胞的淋巴毒受体(lymphotoxinβreceptor,LTβR)相互作用,激活基质细胞表型转化成熟,成为淋巴组织组织者细胞(lymphoid tissue inducer cell,LToC),进而启动其下游的NF-κB信号通路转导和级联放大反应形成淋巴结原基,而后源源不断招募大量自身的免疫细胞进入期内栖息、激活、增殖、分化。其中,LToC是淋巴结的建筑师,它是淋巴结发挥功能的关键,这些细胞不仅为免疫细胞的相互作用提供支架,产生一个适于免疫反应进行的微环境,而且还执行多种生理功能。其前体被认为是组织中的间充质细胞(mesenchymal stem cell,MSC),在LTiC的作用下成熟、活化,从而执行功能。因而,LTiC作用在淋巴结的胚胎发生中处于金字塔顶层,少量的LTiC可以调动大量的免疫细胞,协调免疫反应,不需要像目前的细胞治疗手段输入大量的细胞;克服了干细胞诱导分化产率较低的问题。
因此,本发明构建的人工淋巴结复刻了胚胎期淋巴结发生的生理过程,所得结构和功能更加完整,具有包含高密度的T、B淋巴细胞的组织学结构,和淋巴细胞进出人工淋巴结的血管、淋巴管两大“门户”结构。而且,制备过程中不涉及异种蛋白,降低机体免疫和炎症风暴的风险;而且细胞来源不受限制,不存在使用胚胎干细胞ES的伦理问题。而且本发明制备的人工淋巴结移植体内能较好地招募淋巴细胞,支持其增殖活化,并将活化的细胞再次输出至病灶局部发挥功能;此外,本发明制备的人工淋巴结原基需求的起始投入细胞量低,而传统过继性细胞治疗的细胞需求量大(107-1010细胞量),通过给予少量的LTiC细胞抗原调动大量的自体T、B等免疫细胞发挥功能;在淋巴结缺陷的转基因小鼠(Rorc(γt)-/-小鼠)体内植入本发明构建的人工淋巴结原基可以达到恢复乳腺癌荷瘤条件下的局部免疫清除功能的效果。

以下将结合实施例对本发明的构思及产生的技术效果进行清楚、完整地描述,以充分地理解本发明的目的、特征和效果。显然,所描述的实施例只是本发明的一部分实施例,而不是全部实施例,基于本发明的实施例,本领域的技术人员在不付出创造性劳动的前提下所获得的其他实施例,均属于本发明保护的范围。
本发明提供了一种人工淋巴结的构建方法,流程图见图1;其中淋巴结缺陷的转基因小鼠皮肤下移植人工淋巴结示意图见图2。
实施例1人工淋巴结的构建
1、从诱导多能干细胞(iPSC)诱导分化淋巴组织诱导细胞(LTiC)
1)、将过表达转录因子3TF(Runx1、转录因子Id2和转录因子Rorγt)的iPS细胞(iPSC-3TF)消化重悬在第一分化培养基(15%FBS+IMEM+1%谷氨酰胺+50ug/ml抗坏血酸+5ng/ml BMP4)中至1x105/ml细胞密度,以20ul/滴用悬滴培养方法培养2.5天制备拟胚体(embryonic body,EB)。
2)、收集D2.5的EB,转移至0.1%明胶预包被的孔板中,在第二分化培养基中(15%FBS+IMEM+1%谷氨酰胺+50ug/ml抗坏血酸+5ng/ml BMP4+5ng/ml VEGF),继续培养至第6天。
3)、将分化至D6的细胞加0.25%胰酶-EDTA消化,收集离心,加入至OP9-DL1饲养层细胞中共培养,在第三分化培养基(15%FBS+IMEM+1%谷氨酰胺+50ug/ml抗坏血酸+KL+FL+IL-3+IL-6)中培养,隔天半量换液;
4)、分化培养至第10-12天,可见造血祖干细胞样集落,将其消化吹打收集,离心后加入至OP9-DL1饲养层细胞中共培养,一周后共培养体系可见大量的悬浮或半贴壁细胞。
5)、将悬浮或半贴壁细胞离心收集,重新接种于新制备的OP9-DL1饲养层细胞中共培养,在第四分化培养基(15%胎牛血清+IMEM+1%谷氨酰胺+50ug/ml抗坏血酸+KL+FL+IL-7)中培养,每隔2-3天半量换液,每两周更换新鲜得OP9-DL1饲养层细胞。培养至Day45-50,将共培养体系的血细胞细胞进行磁珠富集或流式分选,所得细胞即为转化的淋巴组织诱导细胞LTiC,用于可进行进一步检测和功能实验。

2、制备脾原代基质细胞(spleen stromal cell,SSC)
将5-6周龄的C57/BL6小鼠颈椎脱臼处死,无菌操作取出脾脏,放置于40um的细胞过滤器上,用20ml注射器的针芯研磨,基质部分收集在含有2ml胶原酶IV提取缓冲液(2%胎牛血清,1mg/ml胶原酶IV和40ug/ml DNase I)的试管中,在37℃旋转下孵育20分钟,加入2ml胶原酶D提取缓冲液(2%胎牛血清,1mg/ml胶原酶D和40ug/ml DNase I)于37℃旋转孵育20分钟,再加入2ml胶原酶D提取缓冲液,继续孵育20分钟后,加入60uL的500mM EDTA,终浓度为5mM。用5ml PBS离心(300g,4℃,5分钟)洗涤消化的脾脏基质两次,按推荐加入流式抗体CD45,4度避光孵育30min,洗涤后流式分选CD45阴性的基质细胞成分,种板培养(DMEM高糖+10%FBS)。
3、脾原代基质细胞的永生化(immortalized spleen stromal cell,iSSC)
将分选后的基质细胞扩增至一定数目,消化细胞,计数离心,接种于24孔板,细胞密度为2.5x104/孔,细胞贴壁12小时后,按照MOI=150加入慢病毒SV40T-puro,病毒感染16-18hr后换液,PBS洗涤两次,病毒感染48hr后加入5ug/ml的嘌呤霉素连续筛选2周,对嘌呤霉素产生耐药的细胞即为永生化的脾基质细胞(iSSC),可支持体外多次传代。
4、iSSC制备脱细胞支架(cell derived matrix,CDM)
10cm的细胞培养皿加入10ml 0.2%gelatin溶液,在37度孵育1小时,吸走gelatin溶液并用PBS洗一次,加入8ml 1%戊二醛溶液室温孵育30分钟,吸走戊二醛溶液后PBS洗一次,加入10ml 1M甘氨酸溶液室温孵育20min后PBS洗一次,加入10ml完全培养基(DMEM+10%FBS)在37度预孵育1小时后,培养皿即可用于制备脱细胞支架。用0.25%胰酶-EDTA消化iSSC,细胞皱缩变圆后终止消化,离心计数接种至预处理的培养皿,种板密度控制在使其一两天内可以长成融合的一层。当细胞长成单层融合时,将培养基换成含50ug/ml L-抗坏血酸的培养基,隔天换液,持续14-21d天。达到处理时间后,吸走培养基,PBS洗一次。每个皿加入5ml 37度预温的无菌裂解缓冲液(1ml NH4OH+250μl Triton X-100+48.75ml PBS配制),镜下观察,细胞裂解后小心轻轻地吸走裂解液,小心加入5ml PBS洗两次。小心加入10ug/mlDNaseI溶液,37度孵育30min,去除残余的细胞DNA,PBS洗一遍,即为脱细胞支架。
通过检测发现所制备的脱细胞支架保存完整的细胞外基质三维结构;经过脱细胞处理的细胞外基质膜片完整地保留了I型胶原蛋白、纤连蛋白等主要基质成分,并且没有细胞核的DAPI着色,说明脱细胞处理充分且适当。
5、人工淋巴结原基(pre-artificial lymph node,pre-aLN)构建
在含CDM的大皿加入完全培养基在37度预孵育1小时,弃去培养基,按照10:1比例加入间充质干细胞和由iPSC经体外诱导获得的淋巴组织诱导细胞(lymphoid tissueinducer cell,LTiC),培养3小时贴壁后,在体外组装成三维结构,转移至低吸附孔板培养,即形成pre-aLN。
6、人工淋巴结原基体内移植及人工淋巴结(artificial lymph node,aLN)结构形成
对于体内移植应用,pre-aLN在低吸附培养板组装过夜,用1%戊巴比妥钠按照25mg/kg麻醉淋巴结缺陷的Rorc(γt)-/-纯合转基因小鼠(Jackson lab,Jax#007502),固定小鼠的四肢,腹部皮肤备皮后用75%酒精棉球擦拭消毒,用眼科剪在距第四对脂肪垫约0.5cm处将皮肤剪开2mm后用眼科镊钝性分离出距离切口0.5~0.8cm的空间,将人工淋巴结原基pre-aLN移植至皮下空间,缝合伤口,将小鼠放至保温垫上麻醉复苏,即完成移植手术。同时,以CDM包载相同数量文献报道过表达淋巴毒素α(lymphotoxinα,LTα)的间充质干细胞作为处理对照,以野生型C57小鼠的淋巴结作为阳性对照。
将上述Rorc(γt)-/-纯合转基因小鼠处理对照组(LTα)、处理组(aLN)以及野生型(WT)C57小鼠颈椎脱臼处死,分别取出移植物LTα/aLN,以及淋巴结LN,用眼科剪剪碎组织后使用胶原酶消化制备单细胞悬液,然后使用CD3、CD19、CD11c、PDPN、CD31流式抗体标记染色,染色完成后洗涤,用Attune NxT流式细胞仪检测各组的细胞构成。
结果发现,本实施例构建的人工淋巴结具有与本体淋巴结相似的主要细胞组分构成,包括CD19+B细胞、CD3+T细胞和CD11c+DC细胞等免疫细胞组分,以及PDPN+网状成纤维细胞(FRC)细胞、CD31+血管内皮细胞(BEC)等基质细胞组分(图3)。
此外,上述分组实验设计进行移植物植入,处死植入4周的小鼠,取出Rorc(γt)-/-转基因小鼠近第4对脂肪垫皮肤下的植入物aLN,进行腹股沟淋巴结位置大体照片拍摄分析和组织固定包埋切片、H&E染色分析。以野生型C57BL/6小鼠(WT)及未接受移植处理的Rorc(γt)-/-小鼠(UT)分别作为阳性和阴性对照,以CDM包载相同数量文献报道过表达淋巴毒素α(lymphotoxinα,LTα)的间充质干细胞作为处理对照(LTα)。通过H&E染色观察到本实施例构建的人工淋巴结与本体淋巴结相似,具有高密度细胞结构(图4)。
实施例2人工淋巴结原基(pre-aLN)体内移植和抗肿瘤应用
对于抗肿瘤应用目的,Rorc(γt)-/-纯合转基因小鼠接受移植pre-aLN一周后,给小鼠同侧第四对乳腺脂肪垫注射5x105过表达OVA的E0771细胞(命名为aLN组),同时以野生型C57小鼠(命名为WT组)和Rorc(γt)-/-纯合转基因小鼠(命名为UT组)接受第四对乳腺脂肪垫注射5x105过表达OVA的E0771细胞处理分别作为阳性对照和阴性对照,以荷瘤Rorc(γt)-/-纯合转基因小鼠接受CDM包载相同数量文过表达淋巴毒素α(lymphotoxinα,LTα)的间充质干细胞移植作为处理对照(命名为LTα组)。成瘤3周后分别取Rorc(γt)-/-转基因小鼠的肿瘤引流移植物LTα和aLN、野生型C57小鼠的肿瘤引流淋巴结和肿瘤进行下一步分析。
颈椎脱臼处死小鼠,无菌取出皮下移植的人工淋巴结移,对于荷瘤鼠则先取外周血再处死小鼠后同时取负荷肿瘤和皮下移植的人工淋巴结。根据分析目的进行原代细胞制备、流式抗体染色等。
在EO771-OVA荷瘤的Rorc(γt)-/-转基因小鼠的移植瘤模型,用OVA257-264(SIINFEKL)肽-H2kb抗体检测抗原提呈细胞DC针对肿瘤OVA的抗原提呈,可以检测到肿瘤引流的人工淋巴结内DC的有较高比例的OVA抗原提呈,与野生型C57小鼠的天然肿瘤引流淋巴结内DC细胞抗原提呈比例接近(图5)。
与上述分组相同,在EO771-OVA荷瘤的Rorc(γt)-/-转基因小鼠的移植瘤模型中,用T-Select H-2K b OVA tetramer-SIINFEKL四聚体抗体检测肿瘤抗原特异性的CD8+T细胞的比例,可以观察到接受人工淋巴结移植的转基因荷瘤小鼠(aLN组)的肿瘤引流人工淋巴结、外周血、肿瘤原位的肿瘤特异性CD8+T细胞比例均明显上升(图5)。
与上述分组相同,在EO771-OVA荷瘤的Rorc(γt)-/-转基因小鼠的移植瘤模型中,在EO771-OVA荷瘤的Rorc(γt)-/-转基因小鼠的移植瘤模型,申请人用游标卡尺每隔2天测量肿瘤的长径(mm)和宽径(mm),按照肿瘤体积计算公式:肿瘤体积(mm3)=[长径(mm)×宽径(mm)2]/2确定肿瘤体积,绘制肿瘤生长曲线。观察到接受LTα植入物组的小鼠肿瘤生长与未处理组无明显差别,而接受aLN植入物组的荷瘤小鼠肿瘤生长得到抑制,生长趋势较前两组缓慢,接近免疫功能完整的野生型C57BL/6荷瘤小鼠的肿瘤生长趋势。此外,aLN植入物组小鼠的生存期也得到了明显改善(图6)。
因此,本发明的人工淋巴结组织结构增强了抗肿瘤免疫反应能力,通过人工淋巴结的植入,淋巴结缺陷的Rorc(γt)-/-转基因小鼠DC细胞的肿瘤特异性抗原提呈、肿瘤特异性CD8+T细胞活化均得到明显改善,小鼠的肿瘤生长得到一定程度抑制。
上述具体实施方式对本发明作了详细说明,但是本发明不限于上述实施例,在所属技术领域普通技术人员所具备的知识范围内,还可以在不脱离本发明宗旨的前提下作出各种变化。此外,在不冲突的情况下,本发明的实施例及实施例中的特征可以相互组合。
Claims (10)
1.一种人工淋巴结原基的构建方法,其特征在于,包含以下步骤:
S1:制备永生化的脾原代基质细胞;
S2:将永生化的脾原代基质细胞进行脱细胞处理得到脱细胞支架;
S3:在脱细胞支架中添加间充质干细胞和淋巴组织诱导细胞进行混合培养后即形成人工淋巴结原基。
2.根据权利要求1所述的方法,其特征在于,步骤S1中的永生化的脾原代基质细胞的构建方法包括:将脾原代基质细胞进行慢病毒转染后通过嘌呤霉素筛选,对嘌呤产生耐药的细胞即为永生化的脾基质细胞。
3.根据权利要求1所述的方法,其特征在于,步骤S2的具体方法包括:
(1)在培养皿中分别添加细胞贴壁促进剂、交联剂、氨基酸溶液进行处理,然后加入完全培养基对培养皿进行预处理;
(2)消化永生化的脾原代基质细胞,然后种板至步骤(1)中所述的预处理后的培养皿;
(3)当细胞单层融合后,更换成包含L-抗坏血酸的培养基,培养14~21天;
(4)加入细胞裂解液清除多余细胞,然后加入DNaseI溶液去除残余的细胞DNA,即为脱细胞支架。
4.根据权利要求3所述的方法,其特征在于,所述细胞贴壁促进剂包括明胶;优选地,所述细胞贴壁促进剂的浓度为0.1~0.2%;优选地,所述交联剂包含醛类交联剂;优选地,所述醛类交联剂包含戊二醛、甲醛或葡萄糖醛;优选地,所述交联剂的浓度为0.5~1.5%。
5.根据权利要求3所述的方法,其特征在于,所述氨基酸包括甘氨酸;优选地,所述氨基酸浓度为0.5~1.5M。
6.根据权利要求1所述的方法,其特征在于,步骤S3中间充质干细胞和淋巴组织诱导细胞的数量比为(5~10):1。
7.根据权利要求1所述的方法,其特征在于,所述淋巴组织诱导细胞的制备方法包括:在诱导多能干细胞中过表达转录因子,然后在分化培养基中培养,收集分化获得的淋巴组织诱导细胞;优选地,所述转录因子包括Rorγt、Tcf1、Id2、Batf3、Runx1、Nfil3中的至少一种。
8.一种人工淋巴结原基,其特征在于,所述淋巴结原基由权利要求1~7任一项所述的方法制备。
9.权利要求9所述淋巴结原基在制备抗肿瘤产品和/或制备治疗淋巴缺陷产品和/或药物体外筛选和/或药物毒性检测中的应用;优选地,所述肿瘤包括乳腺癌。
10.一种产品,所述产品包含权利要求8所述的人工淋巴结原基;优选地,所述产品用于抗肿瘤和/或治疗淋巴缺陷和/或药物体外筛选和/或药物毒性检测,优选地,所述肿瘤包括乳腺癌。
