亲和沉淀的典型实例是使用弹性蛋白样蛋白(ELP)融合的Z结构域以通过温度和盐触发的ELP的溶解度转变来沉淀IgG。39-40然而,细菌表达的ELP的高质量、每个ELP融合的配体上有限的结合位点以及抗体在升高的温度的潜在变性促进了寻找呈现抗体结合配体的新的底物的兴趣。
受到自组装肽两亲物的巧妙的分子设计的启发,我们研究了将蛋白A模拟肽Z33并入自组装免疫两亲物(IA)的方式,并且探索了其在自组装状态下与靶抗体的结合能力。使用等温滴定量热法(ITC)研究了自组装的免疫纤维(IF)和治疗性IgG之间的结合亲和力,表明含有IF的Z33保持其高的IgG结合亲和力。
发明概述
许多一维(1D)纳米结构通过包含作为芯构建基序的短β-片层序列的肽或肽缀合物的自组装来构建,所述芯构建基序对于促进所得到的组装体的定向各向异性生长的分子间氢键合至关重要。虽然该分子设计策略已经导致成功地生产了大量用于与细胞接合的有生物活性的丝状β-片层组装体,但联想到淀粉样蛋白原纤维与潜在毒性相关的担忧已经促进了具有α-螺旋肽的其他超分子工艺策略。
迄今为止,文献中已有大量研究表明,生物活性肽可以成功地并入到超分子肽纳米结构中,同时保持其生物活性。然而,在表位必须保持α-螺旋构象以便有生物活性的情况下,在使用形成β-片层的序列和α-螺旋基序的呈现之间似乎存在间距不相容的问题。在此情况下,本发明人现在示出了蛋白A模拟肽Z33(含有两个α-螺旋的基序)与直链烃的直接缀合,以产生两种对单克隆抗体具有高结合亲和力的自组装免疫两亲物,并且首次证明了本发明的这些超分子免疫纤维(IF)可以用于沉淀和纯化单克隆抗体免疫球蛋白G(IgG)。

葡萄球菌蛋白A(SPA)是最初在金黄色葡萄球菌的细胞壁中发现的蛋白质。它包括折叠成三螺旋束的五个同源结构域。蛋白A由于其与来自大多数哺乳动物物种(包括人类)的免疫球蛋白G(又称IgG)的Fc部分特异性结合而在免疫学中发挥重要的作用。已经对蛋白A进行了广泛的结构和生化研究。第一个编码SPA的基因在1984年被克隆、测序和表达,随后许多基于蛋白A的合成的和最小化的IgG结合结构域被克隆、测序和表达。其中Z-58结构域是第一个且是最著名的广泛用于亲和色谱法和亲和沉淀的合成结构域。另一个最小化的结合结构域Z-33在1996年获得,分子的功能无显著改变。

实施例
材料。所有Fmoc氨基酸和树脂均购自Advanced Automated Peptide ProteinTechnologies(AAPPTEC,Louisville,KY,USA),并且Fmoc-Lys(Fmoc)从Novabiochem(SanDiego,CA,USA)获得。治疗性人IgG1(IgG1)从Bristol-Myers Squibb(Boston,MA,USA)获得,并且IgG洗脱缓冲液来源于Thermo Fisher Scientific(Rockford,IL,USA)。所有其他试剂均从VWR(Radnor,PA,USA)获得,并且在不经进一步纯化的情况下按原样使用。
分子合成。使用类似的方法合成C12-Z33和2C8-Z33免疫两亲物。简言之,Z33肽首先在Focus XC自动肽合成器(AAPPTEC,Louisville,KY)上使用标准的9-芴基甲氧羰基(Fmoc)固相合成方案来合成。然后将C12(或2C8)烷基链与相对于Z33肽为4(或8):4:6比率的月桂酸(或辛酸)/HBTU/DIEA手动地偶联在Z33肽的N-末端(在Fmoc除去之后),在室温震荡过夜。使用在DMF溶液中的20%4-甲基哌啶进行Fmoc脱保护持续10分钟,重复一次。在所有情况下,通过茚三酮测试(Anaspec Inc.,Fremont,CA)监测反应的游离胺。使用以92.5:5:2.5比率的TFA/TIS/H2O的混合物将完成的肽从固体支持物裂解2.5小时。通过旋转蒸发除去过量的TFA,并且添加冷的乙醚以沉淀粗肽。通过离心方法,以6000rpm分离沉淀的肽和乙醚持续3分钟。将肽用乙醚再洗涤2次,并且通过离心除去溶液。
在配备有级分收集器的Varian ProStar 325型制备型HPLC(AgilentTechnologies,Santa Clara,CA)上,在25℃使用Varian聚合柱(PLRP-S, 10μm,150×25mm)通过制备型RP-HPLC纯化IA。含有0.1%v/v TFA的水/乙腈梯度被用作洗脱剂,流速为20ml/min。监测Z33肽区段在220nm处的吸光度峰。将粗材料溶解在20ml的0.1%水性TFA中,并且每次纯化运行用10ml注射液进行。通过MALDI-ToF(BrukerAutoflex III MALDI-ToF仪器,Billerica,MA)分析收集的级分,并且将含有期望的产物的那些级分冻干(FreeZone-105℃4.5L冷冻干燥器,Labconco,Kansas City,MO),并且储存在-30℃。
免疫两亲物的自组装和TEM成像。将1mM浓度的免疫两亲物用HFIP预处理,并且然后溶解在1×PBS或去离子水中,并且在室温老化过夜;将10μl的10倍稀释的样品点在具有400平方目的碳膜铜载网(grid)(来自EMS:Electron Microscopy Sciences)上,并且将过量部分用滤纸除去以在载网上留下样品的薄膜。在让样品干燥5分钟之后,将10μl的2%乙酸铀酰添加至样品载网中,并且在30秒之后除去过量部分。在TEM成像之前将所有样品干燥至少3小时。
圆二色谱(CD)。两个自组装的样品的CD实验在Jasco J-710分光偏振计(JASCO,Easton,MD,USA)上使用1mm路径长度的石英UV-Vis吸收池(ThermoFisher Scientific,Pittsburgh,PA,USA)在25℃进行。在实验之前,将样品立即从1mM储备溶液稀释至1×PBS中的100μM。光谱在190nm-280nm的波长范围内作为三次扫描的平均值收集。获得溶剂的背景光谱,并且从样品光谱中减去。将收集的数据相对于样品浓度归一化。
ITC实验。等温滴定量热法实验使用高精度VP-ITC滴定量热系统(Microcal Inc.)进行。在15℃用在1×PBS(pH 7.4或2.8)中的免疫两亲物滴定IgG1溶液。对于0.1%(1mg/ml)IgG溶液,使用在280nm的1.4的质量消光系数(mass extinction coefficient)来计算IgG1浓度。免疫两亲物的浓度通过总氮测定来确定(Anal.Biochem.,61.2(1974):623-627)。每次注射之后释放的热量通过量热信号的积分获得。与免疫两亲物结合至IgG1相关的热量通过减去稀释的热量获得。数据的分析使用MicroCal OriginTM包进行。
实施例1
全长Z33免疫两亲物的分子设计。这种两亲性肽缀合物(诸如肽两亲物、肽-聚合物缀合物、肽-药物缀合物等)的构建已经被广泛地用于产生多种超分子纳米结构。由亲水性Z33肽序列(FNMQQQRRFYEALHDPNLNEEQRNAKIKSIRDD)(SEQ ID NO:1)和疏水性烷基链组成的IgG结合免疫两亲物被设计为用作免疫纤维(IF)的构建基序。Z33肽是来自蛋白A的双螺旋衍生物(图1A),其以高结合亲和力(Kd=43nM)特异性地结合至IgG的Fc部分。28,41-42
经由将月桂酸部分(C12)或两个辛酸部分(2C8)直接缀合至Z33肽的N-端合成了两种IA,C12-Z33和2C8-Z33(图1B)。如图1C所示,预期IA自组装成IF,并且特异性地结合来自抗体混合物溶液的IgG。还合成了纯的Z33肽,以比较Z33分子与含Z33的IF之间的生物活性。另一对照分子C12-Z33是通过将C12缀合至具有加扰序列(scrambled sequence)的Z33的N-末端设计的。所有分子均使用自动固相肽合成(SPPS)方法和RP-HPLC来合成和纯化。合成的化合物的纯度和预期的分子量使用分析型HPLC和质谱法证实。
实施例2
全长Z33免疫两亲物实施方案的分子自组装和表征。两种IA的自组装可以通过两步操作容易地实现。首先,将IA分别在六氟异丙醇(HFIP)中预处理,以消除任何可能影响其溶解度和自组装形态的均匀性的预先存在的纳米结构。第二,经由蒸发除去HFIP,接着后续添加去离子水或磷酸盐缓冲盐水(PBS)以达到1mM的最终浓度。IF由通过疏水相互作用被捕获在IF的芯中的烷基链段和在面向溶剂的壳中显示的生物活性Z33序列来形成(图2A)。在室温老化过夜之后,利用透射电子显微镜(TEM)和圆二色谱(CD)来表征组装的纳米结构的形态。
鉴于pH条件在本发明的IgG纯化方法中的重要作用,评估了C12-Z33响应于pH变化的自组装行为。通常,中性pH一般用作结合条件,而酸性pH用于从蛋白A亲和柱洗脱抗体。32,34为了研究在中性pH和低pH的自组装行为,PBS(pH 7.4)和IgG洗脱缓冲液(pH 2.8)被用作用于C12-Z33的自组装的水性环境。通过TEM(图2C至图2F)和CD(图2B)研究了C12-Z33IF在不同pH的形态。发现在上文提及的两种pH条件下,C12-Z33分子能够很好地溶解并且自组装成纳米纤维。来自100μM C12-Z33的溶液的代表性TEM图像揭示,C12-Z33在生理条件和酸性条件下都能自组装成纳米纤维结构,直径为16.0±1.7nm,该值小于完全延伸的肽分子的长度(β-片层构象为约22.5nm)。纳米纤维的长度以微米尺度示出并且不能被良好地控制。为了进一步理解自组装的结构内的分子堆积,使用圆二色谱(CD)研究肽二级结构。在C12-Z33中观察到在约222nm(n-π*)和208nm(π-π*)处的强阴性信号,表明处于自组装状态的Z33区段的α-螺旋二级结构的形成,如同纯的Z33肽所示。基于IF的CD光谱和测量的直径,合理地推断出肽在堆积成IF时保持其α-螺旋二级结构。值得注意的是,尽管在PBS溶液或IgG洗脱缓冲液中的C12-Z33的CD光谱仅保持部分α-螺旋信号,但与在相同缓冲液中的Z33肽相比,在约222nm和208nm处的两个负峰的椭圆率改变。CD光谱的位移可能是由于IF的形成,IF可以改变Z33区段的分子堆积使其脱离游离状态,并且随后可以由于结合位点所需的特定构象影响其对IgG的结合亲和力。
实施例3
用于测量IF的结合亲和力的ITC实验。鉴于Z33肽在并入IF之后二级结构的构象变化,有极大兴趣知道C12-Z33 IF的形成是否将影响原始Z33肽中存在的IgG结合能力。为了研究自组装的C12-Z33 IF的结合亲和力,通过等温滴定量热法(ITC)研究了与IgG1结合的热力学性质。ITC已经被广泛地用于监测大量蛋白质和配体之间的结合事件,43-45它是探索C12-Z33 IF和IgG1之间是否可以发生结合的优异的方法。46-47与结合反应相关的热量在逐步注射期间被记录,并且热力学参数,包括热力学解离常数(Kd)、摩尔焓变(ΔH°)和化学计量(N),可以直接获得。44

在典型的ITC实验中,将在PBS缓冲液中的100μM C12-Z33的溶液老化过夜,并且然后在15℃注射到在pH 7.4的相同缓冲液中的2μM IgG1的溶液中。典型的热谱图和结合等温线在图3A中示出,并且按照配体报告的热力学参数总结在表1中。C12-Z33 IF与IgG1结合的ITC结果揭示了焓驱动的结合事件,其特征在于650的Kd。为了进一步比较C12-Z33 IF的结合效率,我们合成了Z33肽,其被证明与IgG1紧密结合,通过表面等离子体共振测量的Kd为43nM。Z33肽与IgG1的结合性质在15℃在pH7.4的PBS中通过ITC测量,并且典型的热谱图和结合等温线在图3C中示出。除了提高100倍的亲和力之外,Z33的化学计量是2.3,而C12-Z33的表观化学计量是3.1,这指示IF中并非所有的C12-Z33都可用于与IgG1分子结合。C12-Z33分子能够与IgG1结合的效率可以通过将Z33的化学计量除以C12-Z33的化学计量评估为74.2%。
虽然按照配体的归一化允许确定结合的表观化学计量,但热力学参数的比较应在对每摩尔IgG归一化之后进行,如表2所示。Z33与IgG的结合的特征在于与大的不利熵变相对的大的有利焓变。尽管焓变和熵变的幅度较小,但C12-Z33的结合的热力学特征类似。尽管C12-Z33相比于Z33以较小的不利熵结合,但有利焓的损失甚至更大,这导致总体较低的结合亲和力。有利结合焓的总体损失可能是由与IF破坏相关的不利焓引起的。由于IF中的限制,存在与IgG1的有利相互作用受到限制的可能性。用C12-Z33滴定IgG1也在15℃在IgG洗脱缓冲液(pH 2.8)中进行(图3B),以便证明在适合于从IF中洗脱的该低pH下显著较低的结合亲和力。
为了排除IF和IgG1之间的非特异性结合,具有加扰Z33肽序列的C12-Z33被用作阴性对照。该C12-Z33 IA示出类似的自组装性质和用TEM和CD表征的二级结构(数据未示出)。ITC实验通过以下进行:将100μMC12-Z33 IA在15℃注射到在pH 7.4的PBS中的2μM IgG1溶液中,以测量它们的结合能力。图3D中的热谱图和结合等温线表明了IgG1和Z33肽之间的特异性相互作用。
实施例4
为了进一步证明IF功能的普遍性,还研究了双链烷基化的IA 2C8-Z33的从自组装性质到与IgG1的结合亲和力(图4A-图E)。在TEM图像中观察到具有均匀直径的纳米级IF,并且通过CD证实了α-螺旋二级结构。根据ITC结果,2C8-Z33和IgG1之间的结合在15℃在PBSpH 7.4中发生,而在洗脱缓冲液pH 2.8中没有可检测的结合发生。2C8-Z33的结合的表观化学计量是9.1,指示甚至更低的结合效率。尽管2C8-Z33相比于C12-Z33以较不有利的结合焓结合,但熵的贡献是没那么不利的,这导致稍好的结合亲和力(表2)。根据上文讨论的结果,我们证明了在表面显示高密度的结合位点的情况下,自组装的IF能够保持与IgG1的有利结合能力,如同原始的Z33肽所示。然而,观察到IF的总体结合亲和力的损失,这是由焓所致的。有利焓的损失可以通过由于IF中的限制导致的相互作用的损失和与颗粒的破坏相关的不利焓贡献来解释。IF内的分子水平堆积决定了它们的形态性质以及功能性质,这些性质可以极大地影响它们的生物活性性能。
实施例5
IgG分子的纯化的潜在应用。
组成氨基酸的多样性为自组装肽纳米纤维之间的非共价相互作用提供了广泛的基础,所述非共价相互作用包括氢键合、π–π堆叠、疏水塌陷(hydrophobic collapse)和静电相互作用。例如,酸性氨基酸和碱性氨基酸的溶解度由离子化程度决定,离子化程度是依赖pH和离子强度的性质。因此,可以通过调节pH或添加盐来减少静电排斥、促进聚集以及甚至沉淀来促进带电荷的肽的自组装过程。考虑到Z33肽中显示的大量带电荷的氨基酸残基,本发明的创造性免疫纤维系统的吸引人的优点在于简单可调的溶解度。一旦IgG与IF结合,通过添加具有高离子强度的盐,IgG-IF复合物具有被沉淀出来的高潜力(图5A)。
因为C12-Z33 IF的相对高的与IgG1的结合亲和力,选择C12-Z33 IF来研究沉淀IgG1的可能性。如图5B(i-ii)所示,5mM C12-Z33可以充分溶解在PBS溶液中,但在0.6MNa2SO4的PBS溶液中沉淀。C12-Z33在PBS溶液中的ζ电位是–7.61mV,并且Na2SO4的添加可以屏蔽IF表面上的电荷,并且由此诱导沉淀。对于IgG1,它充分溶解在5mM C12-Z33以及0.6MNa2SO4中。然而,在将20μM IgG1和5mM C12-Z33混合5分钟,随后添加0.6M Na2SO4之后,观察到沉淀。为了确定沉淀物的组成,进行两个平行实验。将在0.6M Na2SO4中的5mM C12-Z33离心,并且使用紫外-可见(UV-Vis)光谱监测添加Na2SO4之前和之后的上清液在280nm处的吸光度变化。对在0.6M Na2SO4中的5mM C12-Z33和20μM IgG1的混合物进行相同的程序。如图5C所示,大多数C12-Z33 IF能够被0.6M Na2SO4沉淀出来。对于IgG1-IF复合物系统,280nm处的吸光度降低至低于IgG1的初始吸光度的水平,指示IgG1从溶液中除去。更清楚地,经由从蓝线中减去绿线的值来绘制净IgG1的上清液的吸光度,表明多于60%的IgG1从上清液中除去。目前为止,我们的IF用作新型亲和沉淀剂的可能性得到了初步证明。
实施例6
IgG与本发明的IF结合的确认。
在TEM中鉴定IgG结构的困难限制了免疫纤维和IgG之间直接结合的可视化。为了证实IgG存在于免疫纤维的表面上,将预形成的C12-Z33和C12-Z33免疫纤维分别与10nmIgG标记的Au纳米颗粒孵育2h。将每种溶液的液滴放置在TEM载网上之后,将载网用滤纸吸干并且放置自然干燥。然后,我们用PBS缓冲液仔细洗涤载网3次,试图在用乙酸铀酰染色样品之前除去未结合的Au纳米颗粒。与IgG涂覆的Au纳米颗粒孵育的C12-Z33在致密和稀疏(图6A和图6C)的区域中的TEM图像证实了IgG与存在Z33的纳米纤维的表面的结合。观察到非常少的Au纳米颗粒附接到具有加扰Z33序列的对照纳米纤维(图6B和图6D)。该研究表明,C12-Z33免疫纤维和IgG涂覆的Au纳米颗粒的共定位确实是特异性结合的结果。应注意,Au纳米颗粒的密度在长的免疫纤维上是相对低的,这可能是由于Z33免疫两亲物在其自组装成纳米纤维之后紧密堆积的可及性有限。另一种可能性是,IgG涂覆的Au纳米颗粒可以结合至单体状态的C12-Z33,并且然后在洗涤步骤中被带走,因为IgG涂覆的Au纳米颗粒中的IgG浓度(0.33μM-0.66μM)低于C12-Z33的临界胶束浓度值(其为2μM-5μM)(图7)。
实施例7
IgG与本发明的IF相互作用的可视化。
为了更好地可视化C12-Z33免疫纤维和IgG之间的相互作用,用荧光染料(罗丹明B)标记C12-Z33允许荧光免疫纤维和FITC-IgG在共聚焦激光扫描显微镜下直接成像。罗丹明B已经被广泛地用于染色生物材料。罗丹明B标记的C12-Z33(RB-C12-Z33)通过将赖氨酸添加在Z33的N-末端来合成(图8A-图8B)。罗丹明B和C12分别缀合至新添加的赖氨酸的侧链和主链上的胺基团。因为染料在与C12一起自组装成免疫纤维之后将留在疏水性芯中并且它远离结合序列Z33被定位,因此假定荧光标记将不干扰C12-Z33的结合能力。使用TEM证实了PBS中的自组装的纤维的形态(图9C)。由1mM储备溶液稀释的100μM RB-C12-Z33和2μMFITC-IgG在PBS中预混合。在即将成像之前,将30μl溶液点在干净的显微镜载玻片上,并且用盖玻片覆盖以获得液体薄层。然后通过共聚焦激光扫描显微镜拍摄荧光图像(图10A-图10C),并且荧光图像示出了来自RB-C12-Z33(红色)和FITC-IgG(绿色)两者的荧光信号的共定位。发现FITC-IgG形成了在纯FITC-IgG溶液和在相同条件下与C12-Z33一起孵育的溶液中从未观察到的大的明亮的组装体(数据未示出)。相比于FITC-IgG与RB-C12-Z33孵育时的明亮的荧光,在PBS溶液中或在与C12-Z33孵育之后检测到低的荧光信号。这示出FITC-IgG在PBS缓冲液中或在C12-Z33的存在下分散良好,同时RB-C12-Z33和FITC-IgG之间的结合诱导FITC-IgG的聚集和强荧光。

Claims (6)
1.一种免疫两亲物,包含与直链烃链缀合的抗体结合肽。
2.一种免疫两亲物,包含与直链烃链缀合的抗体结合肽,并且其中所述肽当在生理pH的水性溶液中时具有α-螺旋构象。
3.一种免疫两亲物,包含与直链烃链缀合的抗体结合肽,其中所述抗体结合肽具有金黄色葡萄球菌(Staphylococcus aureus)的蛋白A的Z33肽的亲水性氨基酸序列、或其功能部分或片段或衍生物。
4.一种免疫两亲物,包含与直链烃链缀合的抗体结合肽,其中所述抗体结合肽具有氨基酸序列FNMQQQRRFYEALHDPNLNEEQRNAKIKSIRDD(SEQ ID NO:1)或其功能部分或片段或衍生物。
5.一种用于纯化抗体或Fc融合蛋白的方法,包括以下步骤:
a)将权利要求1至4中任一项所述的免疫两亲物溶解在生理pH的水性溶液中并且老化过夜,以使其自组装成免疫纤维(IF);
b)将含有抗体或Fc融合蛋白的样品与所述IF混合,并且允许所述IF结合所述抗体或所述Fc融合蛋白的Fc部分并且在溶液中形成免疫纤维-抗体复合物或免疫纤维-Fc融合蛋白复合物;
c)通过添加盐和离心从所述溶液中分离所述免疫纤维-抗体复合物或所述免疫纤维-Fc融合蛋白复合物;以及
d)从所述抗体或所述Fc融合蛋白中解离所述IF,并且收集未结合的抗体或Fc融合蛋白。
6.如权利要求5所述的方法,其中通过将pH降低至洗脱条件并且过滤或微滤来从所述抗体或所述Fc融合蛋白中分离所述IF。
