Abstract
本发明公开了可促进运动恢复的鼠李糖乳杆菌及其应用。该鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS‑6,其在中国微生物菌种保藏管理委员会普通微生物中心的保藏编号为CGMCC No.24390。本发明的鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS‑6CGMCC No.24390的应用包括:(1)在制备促进运动恢复的产品中的应用;(2)在制备具有抗氧化活性的产品中的应用;(3)在制备具有抗炎作用的产品中的应用。本发明使用出发菌株鼠李糖乳杆菌M6经航天搭载,筛选得到的鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS‑6CGMCC No.24390具有更高的生长速率、更强的抗氧化及抗炎活性,具有减缓小鼠运动时乳酸累积、降低血清尿素氮的产生及增加机体肝糖原储备的效果,可用于提高机体耐力及运动表现、促进运动后疲劳恢复、防止超负荷运动带来的机体损伤。

技术领域
本发明涉及生物技术领域,特别地涉及可促进运动恢复的鼠李糖乳杆菌及其应用。
背景技术
适度运动有益于身体健康,然而,长时间、大强度的剧烈运动,在一定程度上会带来因机体持续疲劳引发的多种潜在问题,如:肠粘膜屏障受损、免疫力降低、肌肉疲劳、供氧不足等。因此,常有运动人士在一次大强度跑步、比赛后,体力疲劳恢复缓慢,免疫力下降,且易患上感冒、呼吸道感染等疾病。
随着人体微生物组研究的进展,肠道菌群与机体健康的关系越来越受到关注,人体肠道中数以万亿计的细菌共同构成复杂的微生物群落,影响机体能量代谢、免疫及下丘脑-垂体-肾上腺等功能。有研究发现,长期运动的人群肠道微生物多样性更好,并且乳杆菌、韦荣氏球菌、粪杆菌、考拉杆菌等种属的微生物丰度显著高于无运动习惯的人群。部分研究显示,益生菌在提高运动表现和改善运动引起的机体指标异常方面存在一定的作用,其可能的机制包括:提供营养物质参与代谢、调节肠道菌群、免疫调节(抑炎)、抗氧化等。
航天太空环境是一种严酷的独特复合环境,具有真空、温度骤变、复杂的空间辐射、微重力等特点,对进入其中的生物体能诱导产生一系列生理生化反应。航天搭载诱导已为人类提供了许多有利用价值的微生物变异株,如我国第17颗返回式卫星搭载实验后,发现NIKKO霉素抗生素产量提高13%~18%的空间变异菌株-圈卷链霉菌;美国多次在航天飞机上开展生物制药研究,成功地在空间生产出产量大、纯度高的治疗心血管病的尿激酶药物。因此,利用太空特因环境资源诱导益生菌产生生理适应性变化,对菌种生长发育、形态、代谢产物积累等生物学特性进行改良,将为其在生物医药及功能食品工业中的应用提供更优异的种质资源。
现有技术中用于促进运动表现的益生菌,有的筛选途径及作用机制不明确,有的未列入安全的、药物或食品可用的菌种名单,目前无法使用。
发明内容
有鉴于此,本发明采用具有改善肠道健康作用的出发菌种鼠李糖乳杆菌,经航天搭载驯化筛选,获得一株优选的分离株鼠李糖乳杆菌。
本发明所提供的鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6,其在中国微生物菌种保藏管理委员会普通微生物中心的保藏编号为CGMCC No.24390。
鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6,已于2022年02月10日保藏在“中国微生物菌种保藏管理委员会普通微生物中心”,保藏编号为CGMCC No.24390;保藏中心的地址是:北京市朝阳区北辰西路1号院3号,邮编100101。
检测菌株MSIS-6的16S rDNA的序列,测得序列如SEQ ID NO.1所示。
本发明还提供了鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCCNo.24390的如下任一应用:
(1)鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390在制备促进运动恢复的产品中的应用;
(2)鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390在制备具有抗氧化活性的产品中的应用;
(3)鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390在制备具有抗炎作用的产品中的应用。
所述促进运动恢复包括如下一种或多种:降低运动者体内的血乳酸浓度、降低运动者体内的血尿素氮浓度、增加运动者体内肝糖原含量、或延长运动者力竭运动持续时间。
所述运动者为动物或人;所述动物为小鼠;所述小鼠为SPF级昆明种小鼠。
所述抗氧化活性包括如下一种或多种:DPPH自由基清除活性、羟基自由基清除活性或还原活性;
所述抗炎作用是:抑制人结肠癌细胞HT29在促炎性细胞因子刺激下分泌炎症因子IL8的活性;所述促炎性细胞因子为TNF-α和/或IL-1β和/或IL-6。
发酵鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390得到的发酵产物也属于本发明的保护范围。
所述发酵产物的如下任一应用也属于本发明的保护范围:
(1)所述发酵产物在制备促进运动恢复的产品中的应用;
(2)所述发酵产物在制备具有抗氧化活性的产品中的应用;
(3)所述发酵产物在制备具有抗炎作用的产品中的应用。
所述促进运动恢复包括如下一种或多种:降低运动者体内的血乳酸浓度、降低运动者体内的血尿素氮浓度、增加运动者体内肝糖原含量或延长运动者力竭运动持续时间;
所述抗氧化活性包括如下一种或多种:DPPH自由基清除活性、羟基自由基清除活性或还原活性;

本发明还提供了一种鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6冻干粉。
本发明所提供的鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6冻干粉,按照如下方法制备得到:
将鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390接种于MRS肉汤,厌氧培养,连续转接两代后,按体积百分比2‰~1%接种量接种于MRS肉汤,厌氧培养10h~22h,获得发酵培养液;去除所述发酵培养液的上清液,收集下层菌泥;将获得的菌泥与冻干保护剂按质量比为1:1~1:5混合,搅拌获得均匀的乳化菌悬液;将获得的乳化菌悬液进行真空冷冻干燥,即制得所述冻干粉;所述冻干保护剂包含如下质量百分比的成分:2~10%脱脂奶粉、0~5%玉米淀粉、1~5%麦芽糊精,余量为纯化水。
可选地,所述冻干保护剂由如下质量百分比的成分组成:2%脱脂奶粉、5%玉米淀粉、5%麦芽糊精,余量为纯化水。
可选地,所述冻干保护剂由含如下质量百分比的成分组成:10%脱脂奶粉、5%麦芽糊精,余量为纯化水。
优选地,所述冻干保护剂由如下质量百分比的成分组成:5%脱脂奶粉、5%玉米淀粉、2%麦芽糊精和88%纯化水。
优选地,所述冻干粉中活菌数为1×108~1×1012CFU/g。
冻干粉在促进运动恢复中的应用也属于本发明的保护范围;所述促进运动恢复包括如下一种或多种:降低运动者体内的血乳酸浓度、降低运动者体内的血尿素氮浓度、增加运动者体内肝糖原含量或延长运动者力竭运动持续时间。
本发明使用出发菌株鼠李糖乳杆菌M6经航天搭载,筛选得到的鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390具有更高的生长速率、更强的抗氧化及抗炎活性,具有减缓小鼠运动时乳酸累积、降低血清尿素氮的产生及增加机体肝糖原储备的效果,可用于提高机体耐力及运动表现、促进运动后疲劳恢复、防止超负荷运动带来的机体损伤。

具体实施方式
下面将结合实施例对本发明的实施方案进行详细描述,但是本领域技术人员应当理解,下列实施例仅用于说明本发明,而不应视为限制本发明的范围。实施例中所用的试剂或仪器,如果未注明来源,均为可通过商购获得的常规产品。
实施例1、菌种航天搭载驯化
本发明所用的出发菌株(地面出发株)分离自人体肠道,经鉴定为鼠李糖乳杆菌M6,属于卫生部《可用于食品的菌种名单》中规定可应用于食品的菌种。该出发菌株鼠李糖乳杆菌M6由上海明现生物科技有限公司保存,目前是商业化的菌株,公众可从上海明现生物科技有限公司购买获得该出发菌株。
将出发菌种制成干粉,严格密封后,利用太空特殊环境,将密封的干粉搭载神舟十二号载人飞船经运载火箭发射升空,历经92天的在轨飞行驯化返回地面后,开展高通量的菌株分离、筛选和性能测定。
最终筛选得到一株鼠李糖乳杆菌,分类命名为鼠李糖乳杆菌(Lactobacillusrhamnosus)MSIS-6,已于2022年02月10日保藏在“中国微生物菌种保藏管理委员会普通微生物中心”,保藏编号为CGMCC No.24390;保藏中心的地址是:北京市朝阳区北辰西路1号院3号,邮编100101。
检测菌株MSIS-6的16S rDNA的序列,测得序列如SEQ ID NO.1所示。
实施例2、菌种培养与制备方法
1、菌种培养与发酵液制备:
将鼠李糖乳杆菌MSIS-6菌株CGMCC No.24390接种入MRS肉汤(购自青岛海博),37℃厌氧培养16h,连续转接2代后,按体积百分比4‰接种量接种于MRS肉汤(购自青岛海博),37℃厌氧培养14h,获得MSIS-6发酵培养液。
2、制备冻干粉:包括以下依次进行的步骤:
1)取上述获得的MSIS-6发酵培养液;
2)将MSIS-6发酵培养液在14000转条件下离心分离30min,获得菌泥;
3)冻干保护:将获得的菌泥与冻干保护剂混合按质量比为1:1,搅拌获得均匀的乳化菌悬液,冻干保护剂由如下质量百分比的成分组成:5%脱脂奶粉、5%玉米淀粉、2%麦芽糊精和88%纯化水;
4)真空冷冻干燥:将获得乳化菌悬液进行真空冷冻干燥。真空冷冻干燥操作工艺参数如下:
冷冻:以2℃/min的降温速率降至-45~-50℃,保持约60min;
一步干燥:以1℃/min的升温速率升温至-25℃,维持此温度直至真空度达到约0.10MPa;
解析干燥:以4℃/min的升温速率至15℃,维持此温度直至真空度达到约0.02MPa;
完成上述操作后,用粉碎机粉碎过筛、包装(例如采用铝箔袋),置于10℃以下保存。
上述方法制得的冻干粉,活菌数在1×108~1×1012CFU/克。
本发明的冻干粉,用于动物实验灌胃时,根据需要,以0.85%生理盐水溶解并重悬浮,调整其菌悬液活菌含量为1×106~1×1010CFU/克。
3、制备地面出发株发酵培养液:
将地面出发株接种入MRS肉汤(购自青岛海博),37℃厌氧培养16h,连续转接2代后,按体积百分比4‰接种量接种于MRS肉汤(购自青岛海博),37℃厌氧培养14h,获得地面出发株发酵培养液。
实施例3、MSS-6表型特征、生长速率
菌体形态观察:分别取MSIS-6发酵培养液和地面出发株发酵培养液,经革兰氏染色后,显微镜油镜观察菌体细胞形态,结果如图1所示。由图1可见,与地面出发株相比,航天驯化株MSIS-6的菌体杆状更小、更短,且更倾向于聚集。另外,航天驯化株细胞表型与地面出发株相比,细胞壁更厚,菌体更粗壮,杆体更均匀。
生长速率:分别将鼠李糖乳杆菌MSIS-6菌种和地面出发株接种入MRS肉汤(购自青岛海博),37℃厌氧培养16h,连续转接2代后,按体积百分比2‰接种量接种至新鲜的MRS液体培养基,37±1℃厌氧培养,每1h测定OD600值,绘制生长曲线(图2)。MSIS-6在接种后2h开始进入生长指数期,10h达到指数期末期,OD600值从1.479增加至8.600,指数生长期时长为7h,OD值增加7.121。地面出发株在接种后5h开始进入指数生长期,12h达到指数期末期,OD600值从1.523增加至7.950,指数生长期时长为7h,OD值仅增加6.427(表1)。

经空间环境驯化筛选获得的本发明MSIS-6菌株,与地面出发株相比,菌体形态更小,生长速率与发酵培养性能显著提高,提示其具有更优的培养性能;同时,更倾向于聚集粘附、更优的生长能力,也提示其可能具有较地面出发株更优的益生性能。
实施例4、MSIS-6抗氧化活性
DPPH自由基清除活性:分别取MSIS-6发酵培养液和地面出发株发酵培养液各1mL,加入1mL DPPH甲醇溶液(0.2mol/L)中,对照组以等量MRS液体培养基代替无细胞上清液。室温下置黑暗环境中反应30min,用三氯甲烷抽提后,于517nm测吸光度(OD517),记为A。
DPPH自由基清除率(%)=(A对照组-A实验组)/A对照组×100
羟基自由基清除活性:分别取MSIS-6发酵培养液和地面出发株发酵培养液各0.5mL,加入1mL邻菲啰啉(购自Merck默克)、1mLPBS(pH7.4)、1mL 2.5mM硫酸亚铁、1mL 20mM过氧化氢,对照组以等量MRS液体培养基代替无细胞上清液。37℃反应1..5h后,在536nm处测定吸光度(OD536),记为B。
羟基自由基清除率(%)=(B对照组-B实验组)/B对照组×100
还原活性的测定:按照文献(Antioxidative ability of lactic acidbacteria.Journal of Agricultural and Food Chemistry,1999,47:1460-1466)所述方法分别测定地面出发株、MSIS-6细胞培养液的还原活性,计算还原力,表示相当于半胱氨酸盐酸盐的浓度。
DPPH自由基清除率测定是一种常见的筛选和评价抗氧化剂的有效方法,羟基自由基是活性、氧化性最强的自由基,对DNA、蛋白质和脂类有较强的结合能力,是引起体内氧化损伤的主要因素。还原活性主要指酶类(过氧化氢酶、NADH氧化酶等)和非酶复合物(维生素C、维生素E、谷胱甘肽等)具有减少氧自由基的能力,进而减少氧化反应的发生。测定结果见表2。

如表2结果所示,与地面对照地面出发株相比,经航天搭载驯化筛选获得的MSIS-6菌株,DDPH自由基清除率由22.15%增加到35.02%,增幅58.10%;羟基自由基清除率由13.80%增加至45.66%,增幅230.86%;同时,MSIS-6的还原力也显著提高,由107.53μmol/L提高到216.29μmol/L,增幅101.14%。本发明MSIS-6具有较强的抗氧化活性,可减少运动带来的应激损伤,帮助机体运动恢复。
实施例5、MSIS-6抗炎作用
体外培养人结肠癌细胞(HT29),培养方法如下:
将人结肠癌细胞(HT29)(购自中科院上海细胞库)细胞置于含10%热灭活的新生牛血清和双抗(青霉素、链霉素浓度各为100U/ml)的DMEM细胞培养基(Gibco)中,37℃,10%二氧化碳、90%空气条件下培养,每天换一次培养基,一周传代一次,15~20天后用于实验。
体外培养的人结肠癌细胞(HT29),分为4组:空白对照组(C)、促炎症对照组(T)、地面出发株组(RC)、MSIS-6组(MS)。分别取生长至稳定期末期的地面对照株和MSIS-6菌体细胞,PBS洗涤两次,分别用于RC组、MS组,与HT29细胞共培养2小时。随后,于T组、RC组、MS组分别加入等量添加了促炎细胞因子(50ng/mL的TNF-α、2.5ng/mL的IL-1β和7.5ng/mL的IFN-γ)的Cytomix缓冲液(购自北京百奥莱博),共培养6小时,促进H29细胞炎症因子的表达。最后,回收上清液,采用ELISA试剂盒法测定各组H29细胞分泌的IL-8浓度,结果见表3。本实施例采用IL-8ELISA检测试剂盒,购自武汉赛培生物,实验操作按照试剂盒说明书进行。

TNF-α、IL-1β和IL-6为促炎性细胞因子,本实施例以含有促炎性细胞因子的混合物Cytomix缓冲液模拟炎症攻击条件刺激体外培养的人结肠癌细胞(HT29),检测在不同组别和条件下HT29细胞分泌炎症因子IL-8的活性差异。由表3结果可见,正常非炎症生理条件下,HT29细胞仅有较少的IL-8分泌(125.6pg/ml),经促炎因子刺激后,T、RC、MS三组的HT29分泌IL-8均明显增多,炎症对照组(T)的I-L8分泌量为789.2pg/ml。与T组相比,地面对照株组(RC)和MSIS-6组(MS)HT29细胞的IL-8分泌量均显著下降,其中,与RC组(663.5pg/ml)相比,MS组(349.1pg/ml)IL-8分泌量下降尤为显著。
结果表明,与地面对照株相比,MSIS-6菌株强烈抑制HT29细胞的炎性应答,具有显著的抗炎和免疫调节作用,有利于促进运动后肌肉恢复,缓解运动疲劳。
实施例6、MSIS-6提高小鼠运动表现及促进小鼠运动后恢复
选用SPF级昆明种(4周龄,雄性)小鼠(购自上海南模生物),共分为6组:对照安静组(C0)、对照运动组(C1)、低剂量安静组(LM0)、低剂量运动组(LM1)、高剂量安静组(HM0)、高剂量运动组(HM1),每组8只小鼠,共48只。适应性饲养一周后,各组灌胃MSIS-6菌悬液:LM0、LM1组每只小鼠灌胃5×106CFU/天,HM0、HM1组每只小鼠灌胃5×107CFU/天,灌胃量0.2mL,C0、C1组以等量生理盐水灌胃,持续4周。灌胃期结束后,安静组小鼠保持静息,运动组进行耐力运动恢复测试;耐力测试完成后,安静组小鼠继续用于负重力竭运动分组及测试。
1.耐力运动恢复测试:游泳槽水深45~50cm、水温维持在28℃±2℃,运动组小鼠连续游泳90min,休息60min,安静组保持静息。运动组分别于游泳前、游泳后、休息后采血;安静组采血一次后,用于后续的负重力竭运动测试。检测实验小鼠耐力运动前、运动及休息后血乳酸、血尿素变化情况。
血乳酸浓度测定方法如下:
每只小鼠采血0.2ml,4℃条件下,1500转离心10min后,收集血清。采用日立7060全自动生化分析仪,检测样本血乳酸浓度(血尿素氮浓度同时检测)。
剧烈运动时机体对能量的需求增加,由此带来一系列与能量代谢相关的机体反应,包括血乳酸含量升高、血尿素浓度增加、糖原大量消耗等,引起运动后疲劳。长期超负荷运动后得不到及时有效恢复,甚至可能对机体造成不可逆的损伤。
高强度运动时,若有氧代谢能力不足,需通过无氧酵解提供能量,产生大量乳酸累积,使肌组织中H+离子浓度升高,pH值降低,引起一系列生化反应,导致运动疲劳的发生。本实施例结果表明,与对照组相比,使用本发明菌株MSIS-6的小鼠(LM1组、HM1组),耐力运动引起的血乳酸浓度累积量显著低于对照组(C1组),并且,经过短暂休息后,血乳酸清除与恢复程度显著优于对照组(表4)。

血尿素氮浓度测定方法如下:
每只小鼠采血0.2ml,4℃条件下,1500转离心10min后,收集血清。采用日立7060全自动生化分析仪,检测样本血尿素氮浓度。
血尿素是机体蛋白质代谢的产物。正常的情况下,蛋白质的分解代谢率较低,血尿素的含量维持在一个相对稳定的状态;而在高强度运动的情况下,机体能量代谢平衡遭到破坏,糖原消耗过多、供应不足,蛋白质、氨基酸的分解代谢率显著升高,蛋白质分解加剧加重肾脏负担,肾脏功能下降致使尿素的清除能力下降,导致血尿素氮含量升高。机体对运动负荷适应能力越差,血尿素氮增加越明显。本实施例结果表明,与对照组相比,使用本发明菌株MSIS-6的小鼠(LM1组、HM1组),耐力运动引起的血尿素氮浓度增加显著低于对照组(C1组),并且,经过休息后,LM1组、HM1组小鼠血尿素氮浓度很快恢复到接近运动前水平(表5)。

2.负重力竭运动测试:C0、LM0、HM0组小鼠参加完耐力运动恢复测试的采血后,恢复1天,于第2天进行负重力竭运动测试。用小动脉夹将铅丝夹在小鼠背部进行负重游泳(铅丝重量为小鼠平均体重的5%),让小鼠游泳直至力竭为止,力竭标准为达到下述之一:①游泳动作明显失调,不能再坚持;②沉入水下10s不能回到水面。记录小鼠负重游泳耐久时间,并在力竭测试结束后,取组织,检测肝糖原含量。
肝糖原含量测定方法如下:
采用小鼠肝糖原ELISA检测试剂盒(上海研谨),测试肝糖原含量。
糖原是运动中最重要的能量物质,体内糖原的水平与耐力密切相关,糖原储备明显影响运动能力,糖原储备的提高有利于机体耐力的提高。肝脏是储存糖原和调节血糖平衡的重要器官,超负荷运动可能对小鼠肝脏的超微结构造成损害,导致肝糖原、肌糖原的减少,并且随着超负荷运动次数的增多而加重其损害程度。经测试,使用本发明菌株MSIS-6后,LM1组、HM1组小鼠负重力竭运动后体内肝糖原储备量显著高于对照组。同时,肝糖原储备高的LM1组、HM1组小鼠,其负重力竭运动持续时间亦显著延长(表6)。

以上实验证明,本发明菌株MSIS-6经航天搭载筛选,具有更高的生长速率、更强的抗氧化及抗炎活性,具有减缓小鼠运动时乳酸累积、降低血清尿素氮的产生及增加机体肝糖原储备的效果,可用于提高机体耐力及运动表现、促进运动后疲劳恢复、防止超负荷运动带来的机体损伤。
上述具体实施方式,并不构成对本发明保护范围的限制。本领域技术人员应该明白的是,取决于设计要求和其他因素,可以发生各种各样的修改、组合、子组合和替代。任何在本发明的精神和原则之内所作的修改、等同替换和改进等,均应包含在本发明保护范围之内。
Claims (10)
1.鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6,其在中国微生物菌种保藏管理委员会普通微生物中心的保藏编号为CGMCC No.24390。
2.鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390的如下任一应用:
(1)鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390在制备促进运动恢复的产品中的应用;
(2)鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390在制备具有抗氧化活性的产品中的应用;
(3)鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390在制备具有抗炎作用的产品中的应用;所述抗炎作用是:抑制人结肠癌细胞HT29在促炎性细胞因子刺激下分泌炎症因子IL8的活性;所述促炎性细胞因子为TNF-α、IL-1β和IL-6。
3.根据权利要求2所述的应用,其特征在于:
所述促进运动恢复包括如下一种或多种:降低运动者体内的血乳酸浓度、降低运动者体内的血尿素氮浓度、增加运动者体内肝糖原含量、或延长运动者力竭运动持续时间。
4.根据权利要求2所述的应用,其特征在于:
所述抗氧化活性包括如下一种或多种:DPPH自由基清除活性、羟基自由基清除活性或还原活性。
5.发酵鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390得到的发酵产物。
6.权利要求5所述的发酵产物的如下任一应用:
(1)所述的发酵产物在制备促进运动恢复的产品中的应用;
(2)所述的发酵产物在制备具有抗氧化活性的产品中的应用;
(3)所述的发酵产物在制备具有抗炎作用的产品中的应用;所述抗炎作用是:抑制人结肠癌细胞HT29在促炎性细胞因子刺激下分泌炎症因子IL8的活性;所述促炎性细胞因子为TNF-α、IL-1β和IL-6。
7.根据权利要求6所述的应用,其特征在于:
所述促进运动恢复包括如下一种或多种:降低运动者体内的血乳酸浓度、降低运动者体内的血尿素氮浓度、增加运动者体内肝糖原含量或延长运动者力竭运动持续时间;
所述抗氧化活性包括如下一种或多种:DPPH自由基清除活性、羟基自由基清除活性或还原活性。
8.一种鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6冻干粉,按照如下方法制备得到:
将鼠李糖乳杆菌(Lactobacillus rhamnosus)MSIS-6CGMCC No.24390接种于MRS肉汤,厌氧培养,连续转接两代后,按体积百分比2‰~1%接种量接种于MRS肉汤,厌氧培养10h~22h,获得发酵培养液;
去除所述发酵培养液的上清液,收集下层菌泥;将获得的菌泥与冻干保护剂按质量比为1:1~1:5混合,搅拌获得均匀的乳化菌悬液;将获得的乳化菌悬液进行真空冷冻干燥,即制得所述冻干粉;
所述冻干保护剂包含如下质量百分比的成分:2~10%脱脂奶粉、0~5%玉米淀粉、1~5%麦芽糊精,余量为纯化水。
9.根据权利要求8所述的冻干粉,其特征在于:所述冻干粉中活菌数为1×108~1×1012CFU/g。
10.权利要求8或9所述的冻干粉在制备促进运动恢复的产品中的应用;所述促进运动恢复包括如下一种或多种:降低运动者体内的血乳酸浓度、降低运动者体内的血尿素氮浓度、增加运动者体内肝糖原含量或延长运动者力竭运动持续时间。
