Abstract
Wrinkle-improving composition comprising low-molecular collagen and rosmarinic acid. The present invention relates to a cosmetic composition for improving wrinkles, comprising low molecular collagen and rosmarinic acid; a pharmaceutical external composition; and food compositions. The composition for improving wrinkles comprising low molecular collagen and rosmarinic acid according to the present invention has excellent synergistic effects on increasing collagen synthesis while increasing collagen absorption, and thus can be usefully applied to cosmetics, medical external products, foods, etc. for enhancing skin elasticity or improving wrinkles.
Description
Technical Field
The present invention relates to a wrinkle-improving composition comprising low-molecular collagen and rosmarinic acid.
Background
Collagen exists in the extracellular matrix as a main matrix protein generated from fibroblasts (fibroblastist) of the skin, and is known as an important function, such as mechanical firmness of the skin, resistance of connective tissue and binding force of tissues, support of cell adhesion, induction of cell division and differentiation (growth of organisms or wound healing), and the like. Such collagen is reduced by age and photoaging caused by ultraviolet irradiation, and collagenase (collage) enzyme activity that breaks down collagen promotes collagen reduction. This is known to have a close association with the formation of wrinkles in the skin.
Collagen is classified into high molecular weight collagen and low molecular weight collagen according to molecular weight, and high molecular weight collagen generally means that molecular weight is 5,000da or more, but molecular weight is large, absorption rate in vivo is reduced, and it is difficult to exhibit functionality. In contrast, low molecular weight collagen has a molecular weight of 5,000Da or less, and there is a problem that it is not sufficient to increase collagen synthesis in the skin by this alone, although attempts to improve absorption by lowering the molecular weight of collagen have been studied with respect to conventional cosmetic compositions comprising collagen (Korean registered patent publication No. 10-1636680).
Under such circumstances, the present inventors have conducted intensive studies to solve these problems, and as a result, have confirmed that a cosmetic composition comprising low-molecular collagen and rosmarinic acid exerts a synergistic effect on increasing collagen synthesis while increasing collagen absorption, thereby completing the present invention.
Disclosure of Invention
Problems to be solved
An object of the present invention is to provide a cosmetic composition for improving wrinkles, which comprises low molecular collagen and rosmarinic acid.
Another object of the present invention is to provide a wrinkle-improving pharmaceutical external composition comprising low molecular collagen and rosmarinic acid.
It is still another object of the present invention to provide a wrinkle-improving food composition comprising low molecular collagen and rosmarinic acid.
It is a further object of the present invention to provide a use of a composition comprising low molecular collagen and rosmarinic acid for improving wrinkles.
Means for solving the problems
The specific description is as follows. On the other hand, the respective descriptions and embodiments disclosed in the present invention are applicable to the respective other descriptions and embodiments. That is, all combinations of the various elements disclosed in the invention are within the scope of the invention. The scope of the present invention is not to be considered as limited by the following detailed description.
An aspect of the present invention for achieving the above object provides a wrinkle-improving composition comprising low-molecular collagen and rosmarinic acid.
The composition may further comprise a black truffle extract.
The term "low molecular collagen (low molecular weight collagen)" in the present invention may refer to collagen of 5,000da or less which is reduced in molecular weight by a mechanism caused by any one or more of physical treatment, chemical treatment and biological treatment.
More specifically, the low molecular collagen may be collagen that is reduced in molecular weight by a hydrolysis treatment (hydrolyzed collagen ).
Specifically, the molecular weight of the low molecular collagen peptide may be 100 to 5,000Da, 100 to 3,000Da, 200 to 2,000Da, 300 to 1,000Da.
The content of the low molecular collagen may be 0.0001% to 10%, 0.001% to 10%, 0.01% to 5% relative to the total content of the composition.
The term "rosmarinic acid" as a polyphenol compound found in various plants (perilla leaf, lemon, rosemary, etc.) of the present invention may refer to a compound represented by the formula C 18 H 16 O 8 Biological compounds (bio-compounds) are shown.
The rosmarinic acid may be present in an amount of 0.01ug/ml to 10mg/ml, 0.1ug/ml to 1mg/ml, 1ug/ml to 500ug/ml, relative to the total composition.
The invention relates to a black truffle of the term black truffle (Tuber melanosporum) extract or black American truffle extract, which belongs to fungus of truffle genus (Tuber) of Tuberaceae, and is characterized in that fruiting bodies are formed underground (20-30 cm underground) and the fruiting bodies are closed. The origins are France and Italy, and are mainly cultivated by oak trees because Mycorrhizae (Mycorhizae) is used to live at the root of oak.
The black truffle may be present in an amount of 0.0001% to 10%, 0.001% to 10%, 0.01% to 5% relative to the total composition.
The term "extract" of the present invention refers to a product of a liquid component obtained by immersing a target substance in a plurality of solvents and then extracting at normal temperature, low temperature or in a heated state for a certain period of time, a solid component obtained by removing the solvent from the above-mentioned liquid component, or the like. Furthermore, in addition to the above-mentioned products, it can be construed broadly to include dilutions of all the above-mentioned products, concentrates thereof, crude refinements thereof, refined products, and the like.
In the present invention, the above extract may be black truffle extract (Tuber melanosporum extract) or American truffle extract. The above extract may be extracted from various organs of natural, hybrid, and variety plants of the subject plant, for example, not only spore (spore), hypha (hyphae), fruiting body (fruiting body), fruiting layer (hymenium), ascus (ascos), basidium (basidium), warts (warts), cap (pileus), shirre (latifolia), mushroom (mushroom), fungus curtain (stick), fungus stick (stick), fungus tray (volva), and the like, but may be appropriately selected according to the kind of the subject plant, but is not limited thereto. The above extract can be obtained by extracting with water or various organic solvents. At this time, the organic solvent used is not particularly limited as long as an extract can be obtained, but specifically, water, a polar solvent or a nonpolar solvent, more specifically, water, a lower alcohol having 1 to 6 carbon numbers (as an alcohol of C1 to C6, methanol, ethanol, propanol, butanol, or the like), a mixed solvent thereof may be used.
The method for obtaining the extract is not particularly limited as long as the extract can be obtained, but specifically, the fruit body of black truffle, its dried product, processed product, or the like may be immersed in the solvent, and a cold extraction method, a heat extraction method, an ultrasonic extraction method in which ultrasonic waves are applied to extract the fruit body of black truffle, a reflux extraction method using a reflux cooler, or the like may be used.
The term "fraction" in the present invention means a product obtained by performing fractionation (fractionation) to separate a specific component or a specific group of components from a mixture containing various constituent components. In the present invention, the fraction may be a fraction of black truffle extract.
In the present invention, the fractionation method for obtaining the above-mentioned fraction is not particularly limited as long as the above-mentioned fraction can exhibit the effect of enhancing skin beneficial bacteria, but may be carried out according to a method generally used in the art. For example, there may be mentioned a solvent fractionation method by treating a plurality of solvents, an ultrafiltration fractionation method by an ultrafiltration membrane having a certain molecular weight cut-off value, a chromatography fractionation method by performing a plurality of chromatography (for separation according to size, charge, hydrophobicity or affinity), a combination thereof, and the like.
In the present invention, the kind of the fractionation solvent used to obtain the above-mentioned fractions is not particularly limited, and any solvent known in the art may be used. As non-limiting examples of the above-mentioned fractionation solvent, there may be mentioned polar solvents such as water, distilled water, alcohols and the like; non-polar solvents such as hexane (Hexan), ethyl acetate (Ethyl acetate), chloroform (Chloroform), and methylene chloride (dichlormethane). These may be used singly or in combination of two or more. When the alcohol in the above-mentioned fractionation solvent is used, a lower alcohol having 1 to 4 carbon numbers (alcohol of C1 to C4) may be used.
The black truffle can be cultivated or sold in the market without limitation.
In one embodiment of the present invention, the collagen increasing ability was determined by treating the collagen with the molecular weight, and it was confirmed that the effect of low molecular collagen was excellent (table 2), and that the combination of collagen and rosmarinic acid, and the synergistic effect of collagen biosynthesis performance due to the combination of collagen, rosmarinic acid and black truffle extract, were confirmed. The effect of the combination was also confirmed to be excellent in the synergistic effect in combination with low molecular weight collagen by the data on the molecular weight of collagen.
As described above, the wrinkle-improving composition of the present invention has a remarkable effect of improving wrinkles, since it has an excellent collagen biosynthesis performance while increasing the collagen content by increasing the absorption of low-molecular collagen.
Another aspect of the present invention provides a cosmetic composition for improving wrinkles, comprising low molecular collagen and rosmarinic acid.
The cosmetic composition may further comprise a black truffle extract.
In the present invention, the above cosmetic composition may be prepared into a dosage form selected from the group consisting of a solution, a topical ointment, a cream, a foam, a nutritional lotion, a soft lotion, a mask, a skin lotion, an emulsion, a pre-makeup base, an essence, a soap, a liquid cleansing, a bathing agent, a sun cream, a sun oil, a suspension, an emulsion, a paste (paste), a gel, a lotion, a powder, a soap, a surfactant-containing cleansing agent, an oil, a powder foundation, an emulsion foundation, a wax foundation, a patch, and a spray, but is not limited thereto. In the present invention, the cosmetic composition may further contain one or more cosmetically acceptable carriers blended into a general skin cosmetic, and as a conventional ingredient, for example, oil, water, surfactant, moisturizer, lower alcohol, thickener, chelating agent, pigment, preservative, perfume, etc. may be suitably blended, but is not limited thereto.
The cosmetically acceptable carrier contained in the cosmetic composition of the present invention is various depending on the formulation of the cosmetic composition.
When the dosage form of the present invention is an ointment, paste, cream or gel, as a carrier component, animal oil, vegetable oil, wax, paraffin, starch, tragacanth, cellulose derivatives, polyethylene glycol, silicone, bentonite, silica, talc, zinc oxide or the like can be used, but is not limited thereto. These may be used singly or in combination of two or more.
When the dosage form of the present invention is a powder or spray, lactose, talc, silica, aluminum hydroxide, calcium silicate, polyamide powder, etc. may be used as a carrier component, and in particular, when it is a spray, a propellant such as chlorofluorocarbon, propane/butane or dimethyl ether may be additionally contained, but is not limited thereto. These may be used singly or in combination of two or more.
When the dosage form of the present invention is a solution or emulsion, as a carrier component, a solvent, a solubilizing agent, an opacifying agent, or the like can be used, and for example, water, ethanol, isopropyl alcohol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1, 3-butyl glycol oil, cotton seed oil, peanut oil, corn germ oil, olive oil, castor oil, sesame oil, glycerin fatty ester, polyethylene glycol, or fatty acid ester of sorbitan can be used, but is not limited thereto. These may be used singly or in combination of two or more.
When the dosage form of the present invention is a suspension, as the carrier component, a liquid diluent such as water, ethanol or propylene glycol, suspending agents such as ethoxylated isostearyl alcohols, polyoxyethylene sorbitol esters and polyoxyethylene sorbitan esters, microcrystalline cellulose, aluminum metahydroxide, bentonite, agar-agar or tragacanth, and the like can be used, but are not limited thereto. These may be used singly or in combination of two or more.
When the dosage form of the present invention is a soap, alkali metal salts of fatty acids, fatty acid half ester salts, fatty acid protein hydrolysates, isethionates, lanolin derivatives, fatty alcohols, vegetable oils, glycerin, sugar, etc., can be used as carrier components, but are not limited thereto. These may be used singly or in combination of two or more.
In yet another aspect, the present invention provides a wrinkle-improving pharmaceutical external (quad) composition comprising low molecular collagen and rosmarinic acid.
The pharmaceutical external composition may further comprise a black truffle extract.
In addition to the above-mentioned components, the pharmaceutical external composition of the present invention may further comprise a pharmaceutically acceptable carrier, excipient or diluent, as required. The pharmaceutically acceptable carrier, excipient or diluent mentioned above is not limited as long as it does not impair the effect of the present invention, and may include, for example, fillers, extenders, binders, wetting agents, disintegrants, surfactants, lubricants, sweeteners, fragrances, preservatives, and the like.
The pharmaceutical composition for external use of the present invention may be exemplified by, but not limited to, disinfectant cleaners, bath foam, ointment, wet tissues, coating agents, etc., and the method of preparation, amount, method of use, composition, etc. of the pharmaceutical composition for external use may be appropriately selected from conventional techniques known in the art.
In still another aspect of the present invention for achieving the above object, there is provided a food composition for wrinkle improvement comprising low molecular collagen and rosmarinic acid.
The food composition may further comprise a black truffle extract.
The above-mentioned food composition may be used in the form of a health functional food, but is not limited thereto. In addition, the food composition of the present invention may contain food auxiliary additives which are pharmaceutically acceptable in addition to the low molecular collagen and rosmarinic acid.
In the present invention, the "food auxiliary additive" means a component that can be added to food in an auxiliary manner, and is added when preparing health functional foods in various forms, and can be appropriately selected and used by a person of ordinary skill. As examples of the food auxiliary additives, various nutrients, vitamins, minerals (electrolytes), flavors such as synthetic flavors and natural flavors, colorants and fillers, pectic acids and salts thereof, alginic acid and salts thereof, organic acids, protective colloid thickeners, pH adjusters, stabilizers, preservatives, glycerin, alcohols, carbonating agents used in carbonated beverages, and the like are included, but not limited thereto.
The food composition of the present invention may include a health functional food. The above health food refers to a food manufactured and processed in the form of tablets, capsules, powders, granules, liquids, pills, etc. using raw materials or ingredients having a function useful for the human body. The term "functional" as used herein means an effect useful for health care such as regulating nutrients for the structure and function of the human body and obtaining physiological effects. The health functional food of the present invention can be prepared by a method commonly used in the art, and in the above preparation, can be prepared by adding raw materials and ingredients commonly added in the art. The formulation of the health functional food is not limited as long as it is a formulation recognized as a health functional food. The food composition of the present invention can be prepared in various forms of dosage forms, and unlike general medicines, has advantages of taking food as a raw material and thus not having side effects or the like that may occur due to long-term administration of the medicines, and has excellent portability, so that the health functional food of the present invention can be taken as an auxiliary agent for enhancing the effect of improving wrinkles and enhancing skin elasticity.
The form that the health functional food of the present invention can take is not limited, and may include all foods in the conventional sense, and may be used in combination with terms known in the art such as functional foods. The health functional food of the present invention can be prepared by mixing appropriate other auxiliary components, which can be contained in the food, with known additives according to the choice of those skilled in the art. Examples of foods that can be added are meats, sausages, breads, chocolates, candies, snacks, biscuits, pizzas, instant noodles, other noodles, chewing gums, dairy products including ice cream, various soups, beverages, teas, oral liquids, alcoholic beverages, vitamin compounding agents, and the like, and can be added to juices, teas, jellies, fruit juices, and the like prepared with the extract of the present invention as a main component. In addition, food products for use as animal feed are also included.
In yet another aspect, the present invention provides a method for improving skin wrinkles, comprising the step of administering a wrinkle-improving composition comprising low molecular collagen and rosmarinic acid to a subject.
The composition may further comprise a black truffle extract.
The term "administration" as used herein means that the composition of the present invention is introduced into a patient by any suitable method, and the route of administration of the above composition may be by any general route, as long as it reaches the target tissue. Based on the characteristic that the composition of the present invention has an effect of improving wrinkles, the administration route of the above composition may be administered by applying it to the skin.
The composition of the present invention may be administered daily or intermittently, and the number of administrations per day may be divided into 1 or 2 to 3. In addition, the composition of the present invention may be used alone or in combination with other drugs for enhancing skin elasticity or improving wrinkles. In view of all the above, it is important to administer an amount that can obtain the maximum effect in a minimum amount without side effects, and can be easily determined by one skilled in the art.
In the present invention, the term "individual" means all animals including mice, domestic animals, etc., including humans, which have or may have developed wrinkles or skin wrinkles. Specifically, it may be a mammal including a human.
In yet another aspect, the present invention provides a use of a composition comprising low molecular collagen and rosmarinic acid for improving wrinkles.
Effects of the invention
The composition for improving wrinkles comprising low molecular collagen and rosmarinic acid according to the present invention has excellent synergistic effects on increasing collagen synthesis while increasing collagen absorption, and thus can be usefully used in cosmetics, medical external products, foods, etc. for enhancing skin elasticity or improving wrinkles.
Detailed Description
Experimental example 1 evaluation of collagen biosynthesis
To evaluate collagen biosynthesis of preparations 1 to 6, skin fibroblasts were treated at 5x 10 4 Concentration of individual cells/wells (seed) was seeded in 24-well plates (24-well plates) and cultured for 24 hours. The growth medium was removed, a medium containing the sample (no FBS added) was added, and after culturing for 24 hours, the cell culture broth was taken and subjected to ELISA assay (ELISA assay). As a result of the above table 1, it was confirmed that the collagen biosynthesis amount was significantly increased for the case of the whole treatment of the low molecular collagen and rosmarinic acid of preparation example 5, compared with the result of the treatment of only the low molecular collagen of preparation example 2 and the result of the treatment of only rosmarinic acid of preparation example 3. This can be seen as a synergistic effect compared to the case of treating the respective individual substances.
In addition, compared with the results obtained by treating only the low molecular collagen of preparation example 2, rosmarinic acid of preparation example 3 and black truffle extract of preparation example 4 separately, the amount of collagen synthesis was significantly increased when all of the 3 substances of preparation example 6 were treated simultaneously, and the synergistic effect was confirmed as well.
From the above results, it was confirmed that, in the case of using low molecular collagen and rosmarinic acid or low molecular collagen, rosmarinic acid and black truffle extract, there was a synergistic effect on increasing collagen synthesis while increasing collagen absorption, thereby having an excellent effect on enhancing skin elasticity or improving wrinkles.
Experimental example 2 evaluation of collagen biosynthesis based on molecular weight of Low molecular collagen
To evaluate collagen biosynthesis based on molecular weight of low molecular collagen, human dermal cells (human dermal fibroblasts, human dermal fibroblast; HDF) were placed in a 24-well plate (24 well plate) to achieve 5X 10 4 Cells/well, cultured for 1 day. After 1 day, the cells were treated with 1% collagen, 10ug/ml rosmarinic acid and 0.01% black truffle, and then cultured for another 1 day, and then the concentration of collagen contained in the cells was measured by ELISA (enzyme-linked immunosorbent assay ).
The ELISA method was performed as follows.
1. The day before ELISA experiments were performed, procollagen Iα1capture antibody (pro-collagen Ialpha 1capture antibody) was diluted to 4ug/ml and placed in an immunoplate (immunoplate) at 100 ul/well for one day at ambient temperature.
2. After one day the capture antibody (capture antibody) was removed and washed (washing) 3 times with 1 Xwash buffer (1 Xwash buffer).
3. 5% BSA was placed at 200 ul/well and left at room temperature for 1 hour.
After 4.1 hours, 5% BSA was removed and washed (washing) 3 times with 1 Xwash buffer (1 Xwash buffer).
5. A solution of a pre-diluted collagen I.alpha.1 standard (Pro-collagen I.alpha.1 standard) and a cell culture broth were placed at 100 ul/well, respectively, and shaken (shaking) at 300rpm for 2 hours.
After 6.2 hours, the procollagen Iα1standard (Pro-collagen Ialpha 1 standard) solution and the cell culture broth were removed and washed (washing) 3 times with 1 Xwash buffer (1 Xwash buffer).
7. The procollagen Iα1detection antibody (pro-collagen Ialpha 1detection antibody) diluted to 0.1ug/ml was placed in an immunoplate (immunoplate) at 100 ul/well and shaken (shaking) at 300rpm for 2 hours.
After 8.2 hours, the procollagen Iα1detection antibody (pro-collagen Ialpha 1detection antibody) was removed and washed (washing) 3 times with 1 Xwash buffer (1 Xwash buffer).
9. streptavidin-HRP (streptavidin-HRP) diluted to 1X was placed into an immunoplate (immunoplate) at 100 ul/well and shaken (shaking) for 20 minutes after shading.
After 10.20 minutes, streptavidin-HRP (strepavidin-HRP) was removed and washed (washing) 3 times with 1 Xwash buffer (1 Xwash buffer).
11. The Substrate (Substrate) was placed in an immunoplate (immunoplate) at 100 ul/well, shaking (shaking) at 300rpm with light blocking was performed while confirming whether the color was changed, then if the color of the portion of the procollagen Iα1standard solution (Pro-collagen Ialpha 1standard solution) was changed, a stop solution (stop solution) was placed in the immunoplate (immunoplate) at 50 ul/well, and absorbance was measured at 450nm, 570 nm.
12. The relation between the concentration of the procollagen I alpha 1standard solution (Pro-collagen I alpha 1standard solution) and the absorbance was obtained, and then the absorbance of each culture solution was substituted into the relation to calculate the concentration of collagen (collagen) in the culture solution.
As a result, as shown in table 2 below, it was confirmed that collagen and rosmarinic acid were combined; synergistic effects of collagen synthesis from the combination of collagen, rosmarinic acid and black truffle extracts.
The results are expressed as% relative to the untreated control group, and the results show a value increased by about 20% or more for the combination of collagen and rosmarinic acid, so that the synergistic effect of collagen synthesis can be confirmed as a significant value compared with the sum of the mere rising values of the two substances.
In addition, in the case of treating the combination of collagen, rosmarinic acid and black truffle extract, the numerical value was increased by about 50 to 70%, and excellent collagen synthesis effect was exhibited, thereby confirming the synergistic effect.

Specifically, when rosmarinic acid and collagen were simultaneously treated, it was confirmed that the synergistic effect with rosmarinic acid was reduced as the molecular weight of collagen was increased, and it was confirmed that when the molecular weight of collagen was 1000Da or less, the combined treatment of rosmarinic acid and collagen showed a synergistic effect, but when the molecular weight of collagen was 90000Da, it did not show a synergistic effect with rosmarinic acid.
In addition, when rosmarinic acid/collagen complex and black truffle extract were simultaneously treated, the same excellent collagen synthesis effect was exhibited as compared with the effect caused by the above single component, and thus the synergistic effect by the combination could be confirmed.
From the results of the specific molecular weight, it was confirmed that the synergistic effect was reduced with the increase in molecular weight of collagen, and that rosmarinic acid/collagen complex and black truffle extract showed a synergistic effect when the molecular weight of collagen was 1000Da or less, but did not show a synergistic effect when the molecular weight of collagen was 90000 Da.
Therefore, as the above-mentioned results, it was confirmed that the combination of collagen and rosmarinic acid or the combination of collagen, rosmarinic acid and black truffle extract showed a synergistic effect of collagen biosynthesis, and in particular, it was confirmed that the synergistic effect was excellent when low molecular collagen was used.
In addition, collagen is an important factor for determining skin wrinkles, and an increase in collagen content is closely related to reduction of skin wrinkles, whereby an effect of improving skin wrinkles can be confirmed.
Further, the composition for improving wrinkles of the present invention has a remarkable effect of improving wrinkles, since it increases collagen content by increasing absorption of low molecular collagen and has excellent collagen biosynthesis performance.
Based on the above description, it will be appreciated by those skilled in the art that the present invention may be embodied in other specific forms without departing from the spirit or essential characteristics thereof. In this regard, it should be understood that the above-described embodiments are illustrative in all respects and not restrictive. The scope of the invention should be construed as including the meaning of the following claims and all changes and modifications that come within the meaning and range of equivalency thereof, and not just the detailed description.
Claims (9)
1. A cosmetic composition for improving wrinkles, comprising low molecular collagen and rosmarinic acid.
2. The cosmetic composition according to claim 1, wherein the cosmetic composition further comprises a black truffle extract.
3. The cosmetic composition according to claim 1, wherein the molecular weight of the low molecular collagen is 100 to 5000Da.
4. The cosmetic composition according to claim 1, wherein the cosmetic composition exhibits an effect of improving wrinkles by increasing collagen synthesis.
5. A pharmaceutical composition for external use for improving wrinkles comprises low molecular collagen and rosmarinic acid.
6. The pharmaceutical composition according to claim 5, wherein the pharmaceutical composition further comprises a black truffle extract.
7. A food composition for wrinkle improvement comprising low molecular collagen and rosmarinic acid.
8. The food composition of claim 7, wherein the food composition further comprises a black truffle extract.
9. Use of a cosmetic composition comprising low molecular collagen and rosmarinic acid for improving wrinkles.
