本发明属于中药组合物制备技术领域,具体涉及一种治疗高血糖的中药组合物及其制备方法。所述中药组合物,原料包括如下组分:知母、炙甘草、竹茹、枳实、防己、白术、茯苓、生地、石斛、天花粉、当归、郁金、女贞子和柴胡,该组合物能较好的减低血糖,从根本上解决高血糖患者的病症,同时,原料少、价格低、制备工艺简单,适宜于临床推广应用。

技术领域
本发明属于中药组合物制备技术领域,具体涉及一种治疗高血糖的中药组合物及其制备方法。
背景技术
当血糖值高于正常范围即为高血糖。高血糖包括糖尿病前期和糖尿病。长期高血糖未缓解,患者的食欲降低,会引起合并感染、酮症及其他并发症。治疗高血糖首先要去除血糖升高的诱因,如感染等诱发血糖升高的原因。糖尿病前期和糖尿病的治疗应该遵循综合治疗的原则,包括合理的饮食、适当的运动、对血糖和病情的监测以及药物的治疗。同时高血糖的患者还经常合并高血压、高血脂、高血黏度等问题,需要进行相应的治疗。
常用的降糖药物按作用的机理共分为八种,主要有胰岛素及其类似物、磺酰脲类促泌剂、二甲双胍类、α-葡萄糖苷酶抑制剂、噻唑烷二酮类衍生物促敏剂、苯茴酸类衍生物促泌剂、GLP-1受体激动剂、DPP-4酶抑制剂和中成药九大类多个品种。
但是现有药物多是治标不治本,且会产生一定的副作用。因此,中药治疗高血糖成为研究热点,但目前中药治疗高血糖的效果有待进一步提高,同时还存在一些其他问题。例如中国专利申请CN201510144549.2公开一种缓解高血糖的中药组合物,是由以下重量份的原料制成:当归30~50份、山楂30~50份、牛七30~50份、石斛30~50份、知母30~50份、冬虫夏草20~40份、何首乌20~40份、山楂20~40份、菊花20~40份、草决明20~40份、菟丝子20~40份、伸筋草1~20份、茯苓1~20份、杜仲1~20份、金银花1~20份、莲子1~20份、黄连1~20份、沙参1~20份、五味子1~20份。该发明的药物声称用于缓解餐后血糖异常和高血糖,具有疗效好、用药成本底、使用方便的优点。
中国专利申请CN201310281793.4公开了一种治疗高血脂合并高血糖症的中药组合物,涉及中医药领域。其由如下原料制成:牛蒡子10-20份,淮山药12-18份,决明子6-10份,泽泻10-15份,姜黄10-20份,茯苓12-16份,紫草6-9份,鸡血藤20-30份,山楂20-30份,柴胡10-16份,桑寄生12-18份,荷叶15-25份,虎杖10-20份,白芍6-12份,怀牛膝15-20份和甘草10-15份。该发明中药组合物声称在治疗高血糖和/或高血脂方面尤其是对高血脂合并高血糖的患者具有很好的治疗效果,具有显著的临床推广价值。
但以上现有技术中其原料众多,作用机理不明确,效果数据有待进一步验证。同时,过多的原料可能导致其它副作用。因此,目前还不能有效的为众多高血糖患者所用。
因此,本发明旨在提供一种具有较好的治疗高血糖的中药组合物,能较好的减低血糖,从根本上解决高血糖患者的病症,同时,原料少、价格低、制备工艺简单,适宜于临床推广应用。
发明内容
为克服以上技术问题,本发明提供了一种治疗高血糖的中药组合物及其制备方法,能较好的减低血糖,从根本上解决高血糖患者的病症,同时,原料少、价格低、制备工艺简单,适宜于临床推广应用。
为实现以上目的,本发明提供的技术方案如下:
一种中药组合物,原料包括如下组分:知母、炙甘草、竹茹、枳实、防己、白术、茯苓、生地、石斛、天花粉、当归、郁金、女贞子和柴胡。
优选地,按照重量份数计,所述中药组合物,原料包括如下组分:知母2-10份、炙甘草1-5份、竹茹1-5份、枳实1-8份、防己1-5份、白术2-7份、茯苓1-3份、生地2-8份、石斛1-10份、天花粉3-9份、当归1-4份、郁金1-6份、女贞子1-7份和柴胡1-8份。
优选地,按照重量份数计,所述中药组合物,原料包括如下组分:知母5-6份、炙甘草1.5-3份、竹茹1-2份、枳实2.2-4份、防己2-4份、白术3-5份、茯苓1.2-2份、生地2-3份、石斛2-3份、天花粉3.5-5份、当归1-3份、郁金1.5-3份、女贞子1-3份和柴胡2-3份。
优选地,按照重量份数计,所述中药组合物,原料包括如下组分:知母5.6份、炙甘草2份、竹茹1.4份、枳实2.2份、防己2.2份、白术3.4份、茯苓1.4份、生地2.2份、石斛2.2份、天花粉3.6份、当归2份、郁金2.2份、女贞子2.2份和柴胡2.2份。
本发明的另一目的在于提供一种中药组合物的制备方法,包括如下步骤:
(1)取防己、炙甘草、竹茹和郁金,混合后加入水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,用水加热提取,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合提取,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物。
优选地,步骤(1)中,所述加水量为防己、炙甘草、竹茹和郁金药材体积的2-3倍。
优选地,步骤(2)中,所述加热提取为先在40-50℃在提取30-60分钟,再按照1-5℃/min的速率升温至煮沸,煮沸后煎煮1-2h。
优选地,步骤(3)中,所述混合提取用滤渣1、滤渣2、知母、枳实、白术、天花粉、当归和柴胡混合物5-10倍质量的水提取。
优选地,步骤(3)中,所述提取过程中,先加入柠檬酸钠,使柠檬酸钠在水中的浓度为1-2%,提取30-60min后,加入L-苹果酸,使L-苹果酸在水中的浓度为0.05-0.1%,继续提取30-60min。
本发明的另一目的在于提供所述中药组合物在制备治疗高血糖药物中的应用。
一种治疗高血糖的药物,所述药物包括所述中药组合物和药学上常规的辅料。
与现有技术比,本发明的技术优势在于:
1、本发明提供了一种能够有效治疗高血糖中药组合物,该中药组合物能较好的减低血糖,从根本上解决高血糖患者的病症,同时,原料少、价格低、制备工艺简单,适宜于临床推广应用,各原料按照君臣佐使的原则进行科学配伍,本发明中知母味苦,性寒,有滋阴降火之功效。可治烦热消渴,是以重用为君药。天花粉性微寒,味甘、微苦,有清热生津、消肿排脓的功效,可助知母滋阴降火的功效,为臣药。白术、茯苓有健脾和胃;防己、茯苓功专利水消肿;当归、生地滋阴补血,兼有活血功效;柴胡、郁金、枳实可疏肝行气解郁;石斛、女贞子可滋养阴津、滋补肝肾;枳实破气化痰、竹茹除烦消痰;以上药味均为佐药,共凑疏肝、健脾、滋阴、补血之功效。炙甘草既可助白术、茯苓发回健脾和胃的功效,又能调和主药,功兼佐使。
2、本发明中防己、炙甘草、竹茹和郁金具有较好的协同增效作用,同时将四种药材一起混合提取能有效促进各原料中有效成分的提取并协调发挥药效,能有效降低高血糖症状。
3、本发明中石斛、女贞子、茯苓和生地混合提取能有效促进石斛、女贞子、茯苓和生地活性成分的提取;同时,步骤(3)在混合提取过程中先使用柠檬酸钠能够促进药材中黄酮的溶出,同时,提取后再加入L-苹果酸,一方面能够调节混合提取液的pH值,另一方面能够促进其它活性成分的溶出,促进中药组合物的降血糖功效,从根源上解决糖尿病前期和糖尿病的高血糖症状。

下面通过具体实施例对本发明进行说明,以使本发明技术方案更易于理解、掌握,但本发明并不局限于此。下述实施例中所述实验方法,如无特殊说明,均为常规方法;所述试剂和材料,如无特殊说明,均可从商业途径获得;且不同来源对产品性能不具有显著性影响。
实施例1
一种中药组合物,原料包括如下组分:知母5.6g、炙甘草2g、竹茹1.4g、枳实2.2g、防己2.2g、白术3.4g、茯苓1.4g、生地2.2g、石斛2.2g、天花粉3.6g、当归2g、郁金2.2g、女贞子2.2g和柴胡2.2g。
所述中药组合物的制备方法,包括如下步骤:
(1)取防己、炙甘草、竹茹和郁金,混合后加入3倍的纯净水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,加5倍的纯净水,在50℃在提取60分钟,再按照5℃/min的速率升温至煮沸,煮沸后煎煮2h,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合,加入5倍质量的纯净水,提取120min,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物。
实施例2
一种中药组合物,原料包括如下组分:知母5.6g、炙甘草2g、竹茹1.4g、枳实2.2g、防己2.2g、白术3.4g、茯苓1.4g、生地2.2g、石斛2.2g、天花粉3.6g、当归2g、郁金2.2g、女贞子2.2g和柴胡2.2g。

一种中药组合物,原料包括如下组分:知母6g、炙甘草1.5g、竹茹1g、枳实2.2g、防己2g、白术3g、茯苓1.2g、生地3g、石斛2g、天花粉5g、当归1g、郁金3g、女贞子3g和柴胡2g。
所述中药组合物的制备方法,包括如下步骤:
(1)取防己、炙甘草、竹茹和郁金,混合后加入3倍的纯净水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,加5倍的纯净水,在50℃在提取60分钟,再按照5℃/min的速率升温至煮沸,煮沸后煎煮2h,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合,加入5倍质量的纯净水,先加入柠檬酸钠,使柠檬酸钠在水中的浓度为1.5%,提取30min后,加入L-苹果酸,使L-苹果酸在水中的浓度为0.05%,继续提取60min,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物。
实施例4
一种中药组合物,原料包括如下组分:知母5g、炙甘草3g、竹茹2g、枳实4g、防己4g、白术5g、茯苓2g、生地2g、石斛3g、天花粉3.5g、当归1g、郁金1.5g、女贞子1g和柴胡3g。
(1)取防己、炙甘草、竹茹和郁金,混合后加入2倍的纯净水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,加3倍的纯净水,在40℃在提取30分钟,再按照1℃/min的速率升温至煮沸,煮沸后煎煮1h,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合,加入10倍质量的纯净水,先加入柠檬酸钠,使柠檬酸钠在水中的浓度为2%,提取60min后,加入L-苹果酸,使L-苹果酸在水中的浓度为0.1%,继续提取30min,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物。
实施例5
与实施例2的区别在于将不使用防己,增加炙甘草、竹茹和郁金的用量。
一种中药组合物,原料包括如下组分:知母5.6g、炙甘草2.2g、竹茹2.4g、枳实2.2g、白术3.4g、茯苓1.4g、生地2.2g、石斛2.2g、天花粉3.6g、当归2g、郁金3.2g、女贞子2.2g和柴胡2.2g。
所述中药组合物的制备方法,包括如下步骤:
(1)取炙甘草、竹茹和郁金,混合后加入3倍的纯净水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,加5倍的纯净水,在50℃在提取60分钟,再按照5℃/min的速率升温至煮沸,煮沸后煎煮2h,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合,加入5倍质量的纯净水,先加入柠檬酸钠,使柠檬酸钠在水中的浓度为1%,提取60min后,加入L-苹果酸,使L-苹果酸在水中的浓度为0.05%,继续提取60min,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物。
实施例6
与实施例2的区别在于将不使用炙甘草,增加防己、竹茹和郁金的用量。
一种中药组合物,原料包括如下组分:知母5.6g、竹茹2g、枳实2.2g、防己3.2g、白术3.4g、茯苓1.4g、生地2.2g、石斛2.2g、天花粉3.6g、当归2g、郁金2.6g、女贞子2.2g和柴胡2.2g。
所述中药组合物的制备方法,包括如下步骤:
(1)取防己、竹茹和郁金,混合后加入3倍的纯净水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,加5倍的纯净水,在50℃在提取60分钟,再按照5℃/min的速率升温至煮沸,煮沸后煎煮2h,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合,加入5倍质量的纯净水,先加入柠檬酸钠,使柠檬酸钠在水中的浓度为1%,提取60min后,加入L-苹果酸,使L-苹果酸在水中的浓度为0.05%,继续提取60min,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物。
1.高血糖治疗效果试验
(1)试验动物:4-6周龄SPF级健康雄性Wistar大鼠,体重在200±20g。
(2)试剂材料配制:
高糖高脂饲料配方:66.5%小鼠大鼠维持饲料(符合《GB 14924.3-2001》要求)+10%猪油+20%蔗糖+2.5%胆固醇+1%胆酸钠。
1%链脲佐菌素溶液的配制:使用pH值=4-5之间的柠檬酸-柠檬酸钠缓冲液配制1%浓度的链脲佐菌素溶液。
(3)造模分组:大鼠在环境温度(20±2)℃,相对湿度40-60%环境下使用维持饲料(符合GB 14924.3-2001要求)适应性喂养1周后分别进行如下操作:
将大鼠使用高糖高脂饲料喂养4周,禁食12h,用1%链脲佐菌素溶液的溶液,按照30mg/kg腹腔注射,制备大鼠糖尿病模型;注射后继续喂养高糖高脂饲料1周后,测定空腹血糖,血糖>11mmol/L者视为造模成功。取造模成功的大鼠80只,随机分为8组,分别为实施例1-6组、模型组、阳性药物对照组,每组10只大鼠。
另取10只未造模的大鼠使用维持饲料喂养4周,禁食12h,腹腔注射生理盐水0.375g/kg,作为正常对照组。
(4)药物干预治疗:实施例1-6组,给予各自实施例制备得到的中药组合物,灌胃给药量100mg/kg BW;阳性药物组,灌胃二甲双胍200mg/kg;模型组和正常对照组分别给予200mg/kg生理盐水。药物干预10天;干预期间,模型组、阳性药物组和实施例1-6组继续给予高糖高脂饲料,正常对照组继续使用维持饲料喂养。
(5)大鼠在灌胃前1天和灌胃第10天分别禁食不禁水12h后,剪尾取血,用血糖分析仪测量其空腹血糖值。各组大鼠药物干预前后血糖值变化,见下表1:

由以上数据可知,与模型组相比,本发明提供的中药组合物具有显著的降血糖的作用,同时,实施例1-6的效果优于阳性药物组,说明本发明提供的组合物药效比二甲双胍药效更好;本发明实施例2-4提供的组合物治疗效果优于实施例1和实施例5-6,说明本发明提供的中药组合物中组分组成和提取方法对药物起到较为重要的影响,合理的配方组成和药物提取方法能使得药物组合物起到更优的降糖效果。
2.临床效果试验
选取糖尿病前期(空腹血糖值在6.1mmol/L-7.0mmol/L之间;用餐后两小时血糖在7.8mmol/L-11.1mmol/L之间)未经药物干预的高血糖志愿者60人,志愿者满足高血糖症状3个月以上,年龄在45-60岁之间。将志愿者随机分为6组,每组10人,不改变原来的生活和饮食习惯,每日服用本发明实施例1-6制备的中药组合物1剂,连续服用1月,测量治疗前后血糖变化值,结果见表2。

由以上数据可知,本发明提供的中药组合物具有较好的治疗高血糖的疗效。
上述详细说明是针对本发明其中之一可行实施例的具体说明,该实施例并非用以限制本发明的专利范围,凡未脱离本发明所为的等效实施或变更,均应包含于本发明技术方案的范围内。
Claims (4)
1.一种治疗高血糖的中药组合物,其特征在于,按照重量份数计,原料由如下组分构成:知母2-10份、炙甘草1-5份、竹茹1-5份、枳实1-8份、防己1-5份、白术2-7份、茯苓1-3份、生地2-8份、石斛1-10份、天花粉3-9份、当归1-4份、郁金1-6份、女贞子1-7份和柴胡1-8份;
所述中药组合物的制备方法包括如下步骤:
(1)取防己、炙甘草、竹茹和郁金,混合后加入水,进行破壁粉碎处理,过滤,得到滤渣1和滤液1;
(2)将石斛、女贞子、茯苓和生地,用水加热提取,过滤,制得滤渣2和滤液2;
(3)合并滤渣1和滤渣2后,加入知母、枳实、白术、天花粉、当归和柴胡混合提取,制得混合提取液;
(4)将混合提取液和滤液1、滤液2混合,制得混合提取物,干燥即得所述中药组合物;其中,
步骤(1)中,加水量为防己、炙甘草、竹茹和郁金药材体积的2-3倍;
步骤(2)中,所述加热提取为先在40-50℃提取30-60分钟,再按照1-5℃/min的速率升温至煮沸,煮沸后煎煮1-2h;
步骤(3)中,所述提取过程中,先加入柠檬酸钠,使柠檬酸钠在水中的浓度为1-2%,提取30-60min后,加入L-苹果酸,使L-苹果酸在水中的浓度为0.05-0.1%,继续提取30-60min;所述混合提取用滤渣1、滤渣2、知母、枳实、白术、天花粉、当归和柴胡混合物5-10倍质量的水提取。
2.如权利要求1所述的中药组合物,其特征在于,按照重量份数计,原料由如下组分构成:知母5-6份、炙甘草1.5-3份、竹茹1-2份、枳实2.2-4份、防己2-4份、白术3-5份、茯苓1.2-2份、生地2-3份、石斛2-3份、天花粉3.5-5份、当归1-3份、郁金1.5-3份、女贞子1-3份和柴胡2-3份。
3.如权利要求1所述的中药组合物,其特征在于,按照重量份数计,原料由如下组分构成:知母5.6 份、炙甘草2份、竹茹1.4份、枳实2.2份、防己2.2份、白术3.4份、茯苓1.4份、生地2.2份、石斛2.2份、天花粉3.6份、当归2份、郁金2.2份、女贞子2.2份和柴胡2.2份。
4.如权利要求1-3任一所述中药组合物在制备治疗高血糖药物中的应用。
