Abstract
本发明涉及生物技术领域,尤其涉及一种兼具抗炎、抗氧化功能的植物乳酸杆菌胞外多糖的制备方法及应用。在EPS制备上,本发明中通过离子交换层析、琼脂糖凝胶层析获得纯度较高的中性EPS,并测定了EPS的分子量,单糖组成,官能团组成以及微观结构;在功能活性上,本发明中的EPS具有优秀的体外抗氧化活性、同时可以缓解LPS诱导的炎症反应,具有良好的抗炎作用;在EPS产量水平上,本发明通过单因素优化、响应曲面等方法优化了植物乳杆菌EPS的产量,提供了一套高产EPS的植物乳杆菌发酵培养基及发酵条件的配方,这也为其将来的生产奠定了基础。

背景技术
乳酸菌是可使葡萄糖等糖类分解为乳酸的各种细菌的总称,属革兰氏阳性菌。乳酸菌作为一种公认的食品级安全产品,已经广泛应用于食品、医药、工业等相关领域。畜牧生产中,乳酸菌作为微生态制剂的常用菌株在生产中应用非常广泛,近年来随着我国饲料无抗、替抗的进程加快,乳酸菌等益生菌的研究也越来越受到关注。
胞外多糖(exopolysaccharides,EPS)是微生物在生长代谢过程中分泌到细胞外的一类长链多糖。根据多糖存在的位置可以分为荚膜多糖和黏液多糖,而根据组成多糖的单糖种类又可以分为同型多糖和异型多糖。EPS作为一种天然的增稠剂,其在食品的加工和生产中应用十分广泛。而乳酸菌产EPS的能力十分出色,更因为其安全性被广泛的研究:乳酸菌的EPS除了可以被用作食品加工中的增稠剂,在抗氧化、抗肿瘤、降低胆固醇、增强免疫活性和改善肠道微生态环境等方面也有着重要作用。尤其是随着近些年大型分析仪器的发展和分子生物学的兴起,糖研究步入一个新的阶段。近年来关于多糖的活性和结构研究成为蛋白质和核酸之后生物化学的又一个巨大学术前沿领域。
在畜牧生产中,预防炎症疾病发生对于增强动物免疫力、提高生产力以及提高经济效益有着重要的意义。机体炎症的发生机理复杂,受到体内外多种因素的影响,乳酸菌所产的EPS作为一种潜在的抗氧化、抗炎大分子具有十分可观的应用前景,但乳酸菌的EPS目前在畜牧养殖中仍未得到推广,主要原因是目前报道的EPS功能活性并不强,大部分EPS功能单一且活性较弱,实际应用效果难以达到预期,因此开发一种具有高活性的乳酸菌EPS是其在畜牧领域应用的关键一环。

本发明提供一种兼具抗炎、抗氧化功能的植物乳酸杆菌胞外多糖的制备方法及应用,用以解决现有技术中大部分EPS功能单一且活性较弱的缺陷,获得一种具有高产量、高活性的乳酸菌EPS。
为了实现本发明目的,本发明首先提供了一株乳酸杆菌,所述菌株为从泡菜中分离,经生物学性能和抗逆性比较得到的一株乳酸杆菌,该菌株的菌体呈短杆状,单个或成对、短链、偶见长丝状菌体;菌落表面呈圆形,乳白色,边缘整齐,表面光滑湿润无褶皱,质地均匀,不透明,隆起,拉丝后较为粘稠,能闻到明显的酸味。经革兰氏染色鉴定为革兰氏阳性菌,并通过采用16S rRNA对该菌株进行鉴定,所得结果在NCBI数据库中进行BLAST比对,该菌株与植物乳杆菌16SrRNA的同源性为100%。结合以上鉴定结果,最终确定发菌株为植物乳杆菌,该菌株已于2023年3月7日保藏于中国微生物菌种保藏管理委员会普通微生物中心(简称CGMCC,地址:北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所,邮编100101),保藏号为CGMCC No.26768,编号为R301,分类命名:植物乳杆菌(Lactobacillusplantarum)。

本发明提供一种植物乳杆菌培养基,每100ml植物乳杆菌培养基中包括以下原料:1-8g葡萄糖,1-8g小肽,0.1-1g磷酸氢二钾,0.1-1g无水乙酸钠,0.1-1g柠檬酸三胺,0.01-0.5g七水合硫酸镁,0.01-0.5g无水硫酸锰和0.1-0.5ml Tween-80。
优选的,每100ml植物乳杆菌培养基中包括以下原料:1-4g葡萄糖,2-5g小肽,0.2-0.6g磷酸氢二钾,0.5-0.6g无水乙酸钠,0.2-0.4g柠檬酸三胺,0.01-0.05g七水合硫酸镁,0.04-0.1g无水硫酸锰和0.1-0.4ml Tween-80。
乳酸菌EPS产量普遍较低,对于养殖应用而言成本过高,本发明通过单因素优化、响应曲面优化将植物乳杆菌EPS的产量由初始的53.34mg/L提升至98.52mg/L,产量提升接近一倍。

本发明对植物乳杆菌的EPS产量水平进行了优化,主要采用单因素优化以及响应曲面优化的方法来提高EPS的产量,本发明优化得到了植物乳杆菌产EPS的培养基配方及最佳发酵条件:1-4g葡萄糖,2-5g小肽,0.2-0.6g磷酸氢二钾,0.5-0.6g无水乙酸钠,0.2-0.4g柠檬酸三胺,0.01-0.05g七水合硫酸镁,0.04-0.1g无水硫酸锰和0.1-0.4ml Tween-80,发酵温度20-35℃,培养基初始pH值6.5-7.5,种子液接种量2%-7%,最佳发酵条件下植物乳杆菌EPS的产量为98.52mg/L,与优化前相比EPS的产量提高了84.7%。
本发明提供一种胞外多糖的制备方法,包括利用所述植物乳杆菌(Lactobacillusplantarum)R301、所述菌剂、所述组合物、所述植物乳杆菌培养基、所述植物乳杆菌的培养方法中的一种或几种;
优选的,所述方法包括,培养植物乳杆菌(Lactobacillus plantarum)R301得到发酵液,从发酵液中提取胞外多糖。
优选的,得到发酵液后;对所述发酵液除菌体、乙醇沉淀、除蛋白、经离子交换柱、琼脂糖凝胶柱洗脱,得到胞外多糖,进一步优选得到分子量为3.1×106Da的胞外多糖;

本发明筛选得到的植物乳杆菌的EPS具有抗氧化功能:具体表现为体外抗氧化试验表明在4mg/mL、1mg/mL和2mg/mL时,EPS对DPPH自由基、ABTS自由基、羟基自由基的清除率均能达到100%,4mg/mL时还原力约为抗坏血酸的30%。
本发明筛选得到的植物乳杆菌的EPS具有抗炎功能:具体表现为20μg/mL的EPS即可缓解LPS诱导的小鼠巨噬细胞炎症反应,减少NO的产生,缓解NF-κB的应激反应,降低促炎因子IL-6的表达水平。
本发明筛选得到的植物乳杆菌可耐受低pH(pH=3.0)、较高浓度的胆盐环境(0.1~0.3%),能够在模拟消化道环境中生存,具有很强的消化道环境耐受性。
本发明提供所述植物乳杆菌(Lactobacillus plantarum)R301、所述菌剂、所述组合物、所述植物乳杆菌培养基、所述植物乳杆菌的培养方法中的一种或几种在制备胞外多糖中的应用。
本发明提供植物乳杆菌胞外多糖在抗氧、抗炎或抗逆中的应用;

本发明解决了现有技术中EPS功能活性不够强、产量不够高这两方面的问题,在功能活性上,本发明中的EPS具有优秀的体外抗氧化活性、同时可以缓解LPS诱导的炎症反应,具有良好的抗炎作用,而在畜牧集约化养殖中,动物因氧化应激、炎症等问题造成的经济损失巨大,本发明的EPS抗氧化、抗炎功能正适于解决这一问题;在EPS产量水平上,本发明通过单因素优化、响应曲面等方法优化了植物乳杆菌EPS的产量,提供了一套高产EPS的植物乳杆菌发酵培养基及发酵条件的配方,这也为其将来的生产奠定了基础。

为使本发明的目的、技术方案和优点更加清楚,下面将结合本发明中的附图,对本发明中的技术方案进行清楚、完整地描述,显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
本发明实施例中的“小肽”购自于北京澳龙港生物技术研究中心。
实施例1:
1.单因素优化
(1)最佳碳源筛选
选取五种碳源:蔗糖、葡萄糖、果糖、麦芽糖以及乳糖分别替代MRS培养基中的碳源,添加量均为2%(w/v)。挑取R301单菌落接种至种子液培养基培养12h,调整种子液600nm吸光光度值为5.0,以2%(v/v)的接种量接种至不同碳源的培养基中37℃静置培养24h,利用分光光度计测定菌液600nm吸光光度值,提取EPS并测定含量。每组做三个重复,种子液培养基为MRS液体培养基,EPS提取方法及含量测定参考以下操作:
1.将R301按3%(v/v)的种子液接种量接种至MRS改良培养基中,37℃培养24h;
2.发酵液100℃水浴10min;
3.取适量发酵液12000rpm离心2min去除菌体;
4.取上清加入80%(w/v)TCA溶液至终浓度为4%(w/v),混匀后4℃静置6h,12000rpm离心20min保留上清;
5.上清加入3倍体积的无水乙醇混匀4℃静置过夜,12000rpm离心20min。弃上清,加入适量去离子水于50℃水浴复溶粗多糖;
6.使用分子截流量为8000~14000Da的透析袋透析24h,换三次水(分别隔3h,6h,12h),EPS的含量测定采用苯酚硫酸法测定。
(2)碳源添加浓度优化
由前述试验得到最佳碳源,分别以质量浓度1%~8%的添加量替代MRS培养基中的碳源。挑取R301单菌落接种至种子液培养基中培养12h,以2%(w/v)的接种量接种至不同碳源浓度的培养基中37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液培养基为MRS液体培养基,EPS提取方法及含量测定参考上述。
(3)最佳氮源筛选
选取8种氮源:小肽、牛肉膏、胰蛋白胨、乳清蛋白粉、氨水,尿素,硫酸铵以及硝酸铵分别替代MRS培养基中的氮源,添加量均为2.5%(w/v)。挑取R301单菌落接种至种子液培养基中培养12h,以2%(v/v)的接种量接种至不同氮源的培养基中37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液培养基为MRS液体培养基,EPS提取方法及含量测定参考上述。
(4)氮源添加浓度优化
由前述试验得到最佳氮源,分别以质量浓度1%~8%的添加量替代MRS培养基中的氮源。挑取R301单菌落接种至种子液培养基中培养12h,以2%(v/v)的接种量接种至不同氮源浓度的培养基中37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液培养基为MRS液体培养基,EPS提取方法及含量测定参考上述。
(5)Mn2+添加浓度优化
分别以质量浓度0~0.5%添加量的一水合硫酸锰替代MRS培养基中的一水合硫酸锰。挑取R301单菌落接种至种子液培养基中培养12h,以2%(v/v)的接种量接种至不同Mn2+浓度的培养基中37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液培养基为MRS液体培养基,EPS提取方法及含量测定参考上述。
(6)Mg2+添加浓度优化
分别以质量浓度0~0.5%添加量的七水合硫酸镁替代MRS培养基中的七水合硫酸镁。挑取R301单菌落接种至种子液培养基中培养12h,以2%(v/v)的接种量接种至不同Mg2+浓度的培养基中37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液培养基为MRS液体培养基,EPS提取方法及含量测定参考上述。
(7)种子液接种量优化
挑取R301单菌落接种至种子液培养基中培养12h,分别以体积比1%~8%的种子液接种MRS培养基中37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液为MRS液体培养基,EPS提取方法及含量测定参考上述。
(8)培养温度优化
挑取R301单菌落接种至种子液培养基中培养12h,以2%(v/v)的接种量接种MRS培养基中分别于20℃~40℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液为MRS液体培养基,EPS提取方法及含量测定参考上述。
(9)培养基的初始pH值优化
分别调整培养基的pH值为5.5~8.0。挑取R301单菌落接种至种子液培养基中培养12h,以2%(v/v)的接种量接种MRS培养基中于37℃静置培养24h,利用分光光度计测定菌液在600nm处吸光光度值,提取EPS并测定含量。每组做三个重复,种子液为MRS液体培养基,EPS提取方法及含量测定参考上述。
2.响应曲面优化
(1)Plackett-Burman试验
使用Design-Expert 8.0设计Plackett-Burman试验,筛选影响因素包括:碳源浓度、氮源浓度、Mn2+浓度、Mg2+浓度、培养基的初始pH值、培养温度、种子液接种量。每种因素有两水平,以EPS产量为响应值,筛选影响EPS产量的显著因素。试验设计见表1。

(2)中心组合设计试验
根据Plackett-Burman试验结果,使用Design-Expert 8.0设计中心组合设计试验。以EPS产量为响应值,得出回归模型,确定R301生产EPS的最佳条件。试验设计见表2。

植物乳杆菌发酵产生EPS的优化结果如下:
Plackett-Burman试验结果及方差分析见表3、4:其中表4方差分析结果中模型P值为0.0232<0.05,差异显著,表明模型建立成功。R-square=0.9370表明模型拟合良好。在所有的八个因素中E和G差异显著(P<0.05),即培养基的初始pH值和种子液接种量为影响EPS产量的两个显著因素,此外选择C:Mg2+添加量作为第三种因素。因此最终选择培养基的初始pH值、种子液接种量以及Mg2+添加量作为响应曲面优化的因素。
依据Plackett-Burman试验结果设计CCD试验进一步优化EPS的产量,试验结果和方差分析见表5、6。利用Design-Expert 8.0对实验数据进行回归分析,得到EPS产生量(Y)对Mg2+添加量(A)、培养基的初始pH值(B)和种子液接种量(C)的二次多项回归方程模型:

对上述回归模型的方差分析结果见表5:回归模型差异极显著(P<0.01),一次项中种子液接种量表现显著(P<0.01),二次项均表现显著(P<0.01),回归模型R-square=0.9288,接近1,说明该函数可以较好的模拟试验过程。失拟项不显著,表明该函数符合实际情况,可以用于预测和分析R301产EPS的产量。通过P值可知对EPS合成影响最大的因素为种子液接种量,其次为培养基的初始pH值,最次为Mg2+添加量,这也与Plackett-Burman试验结果相一致。
为分析各个交互项对EPS产量的影响,利用Design-Expert得到各个交互项的等高线图和3D响应曲面图(图10),分别为A(图10中A)、B(图10中B)、C(图10中C)固定为零水平时另外两个因素对EPS产量的影响:当Mg2+添加量为0.01%(w/v),种子液接种量固定不变时,EPS的产量随培养基的初始pH值的增大先升高后降低,但整体影响较小。当固定培养基的初始pH值不变时,EPS的产量随种子液接种量的增大而增大,达到一定值后增加速度变缓或不再增加(图10中A);当培养基的初始pH值为7,种子液接种量固定不变时,EPS产量随Mg2+添加量的增大先升高后降低,但整体影响较小,当固定Mg2+添加量不变时,EPS产量随种子液接种量的增大而增大,达到一定值后增加速度变缓或不在增加(图10中B);当种子液接种量为6%(v/v)时,无论固定培养基的初始pH值或者Mg2+添加量对EPS的产量均呈现先升高后降低,且整体影响较小(图10中C)。响应曲面存在着R301产EPS的最大值,预测的最佳条件为:Mg2+添加量为0.01%(w/v),培养基的初始pH值为7.41,种子液接种量为6.4%(v/v),预测EPS最大产量为97.85mg/L。
为验证响应曲面预测值是否符合实际,按照软件预测的最佳发酵条件进行三次平行试验,测得EPS的平均产量为98.52mg/L,与软件预测值相接近,即回归方程能够较好的反映R301实际EPS产量。

3.植物乳杆菌R301胞外多糖的分离与提取
种子培养基配方为(以100mL培养基计):2g葡萄糖,0.5g酵母提取物,1g蛋白胨,1g牛肉膏,0.2g柠檬酸三胺,1mL的Tween-80,5mL的盐液甲(盐液甲由11.5g七水合硫酸镁和2.8g一水合硫酸锰定容至100mL并单独灭菌后无菌添加至灭菌的种子培养基)。
发酵培养基配方为(以100mL培养基计):1-4g葡萄糖,2-5g小肽,0.2-0.6g磷酸氢二钾,0.5-0.6g无水乙酸钠,0.2-0.4g柠檬酸三胺,0.01-0.05g七水合硫酸镁,0.04-0.1g无水硫酸锰和0.1-0.4ml Tween-80,发酵温度20-35℃,培养基初始pH值6.5-7.5。
采用Sevag法去除蛋白质,Sevag试剂为氯仿:正丁醇(5:1)(体积比)。
胞外多糖分离与提取流程:
1.将R301按3%(v/v)的种子液接种量接种至MRS改良培养基中,37℃培养24h;
2.发酵液100℃水浴10min;
3.取适量发酵液12000rpm离心2min去除菌体;
4.取上清加入Sevag试剂(粗多糖液:Sevag试剂=4:1,v/v),室温下涡旋混匀10min,离心,保留水相,反复操作,直至蛋白完全除去,将粗提的EPS冻干备用;
5.上清加入3倍体积的无水乙醇混匀4℃静置过夜,12000rpm离心20min。弃上清,加入适量去离子水于50℃水浴复溶粗多糖;
6.使用分子截流量为8000~14000Da的透析袋透析24h,换三次水(分别隔3h,6h,12h),EPS的含量测定采用苯酚硫酸法测定。
7.胞外多糖的纯化:
采用DEAE-52阴离子交换层析柱对上述得到的EPS进行纯化,分别使用不同浓度的氯化钠水溶液(0,0.3,0.6和0.9mol/L)洗脱150min(1mL/min),收集洗脱出峰管的洗脱液,使用苯酚硫酸法检测OD490并绘制检测峰曲线。如图11所示,粗EPS主要成分在使用蒸馏水洗脱时已经出峰,而在NaCl浓度为0.3mol/L进行洗脱时得到了一个微弱的峰,以上信息可知植物乳杆菌的胞外多糖主要是以中性多糖的形式存在的,因此后续的纯化以及功能验证主要针对中性多糖。
采用Sephadex G-100琼脂糖凝胶层析柱对上述所得的中性胞外多糖初级纯品进行分离纯化,使用0.9%的氯化钠水溶液作为洗脱液,速度为0.5mL/min,收集出峰管的洗脱液,使用苯酚硫酸法检测OD490并绘制检测峰曲线。如图12所示,经过琼脂糖凝胶层析柱纯化得到了纯度较高的R301中性多糖。
分子量测定:
分别采用SB-806HQ和SB-804HQ柱的凝胶渗透色谱法测定EPS的分子重量,在25℃下使用激光散射检测和折射率检测器来检测EPS。然后将EPS溶液送入系统,并使用超纯水进行洗脱。图13为凝胶渗透色谱分析的EPS分子量分布图,结果表明EPS有一个单峰,表明EPS是均一的,R301的EPS分子量为3.1×106Da。
胞外多糖单糖组成分析(甘露糖、鼠李糖、葡萄糖、半乳糖、阿拉伯糖):
标准品的配制:在离心管内加入甘露糖、核糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖、木糖以及岩藻糖各100mg,使用去离子水定容至10mL得到10mg/mL的母液,使用时稀释至100μg/mL的工作液浓度。
样品前处理:前述得到的中性EPS称取样品5mg,加入三氟乙酸溶液,121℃加热2h,吹干后使用甲醇清洗以除去三氟乙酸,重复3次,加入蒸馏水复溶得到胞外多糖水解液。将水解液加入1-苯基-3-甲基-5-吡唑啉酮及氢氧化钠在70℃衍生70min,加入盐酸中和,随后加入同体积氯仿进行萃取,重复3次后取水层转入色谱瓶待测。
HPLC条件分析:分离柱为Shim-pack GIST C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈-0.05mol/mL磷酸盐缓冲液(pH 6.8)(18∶82),流速0.8mL/min,柱温30℃,紫外检测波长245nm,进样量20μL。图14为R301 EPS色谱图,结果表明R301 EPS主要由甘露糖、鼠李糖、葡萄糖、半乳糖和阿拉伯糖组成的杂多糖,单糖组成比例为5.7:35.8:27.5:15.8:15.2。
胞外多糖红外光谱分析:
采用溴化钾压片法,取10mg胞外多糖,加入100mg的溴化钾粉末,使用压片机制成均匀的薄片,采用傅里叶变换红外光谱仪(Bruker VERTEX33型)在4000-5000cm-1范围内扫描红外光谱并记录数据。由图15可知在3420cm-1处的峰值是羟基的伸缩振动,在2938和2888cm-1处的峰值为烷基中C-H键的伸缩振动峰,表明存在糖等有机物,在1644cm-1处的峰值是羰基的伸缩振动峰,表明存在甘露糖或半乳糖,在1454cm-1处的峰值是烷基的变角振动峰,1426cm-1处的峰可能归属于羧基中羰基的对称伸缩振动峰,1359cm-1处的峰值是属于烷基的面外摇摆峰,1265、1161、1129cm-1处的峰值都属于C-O-C的伸缩振动峰,1065cm-1处的峰值表明R301 EPS中可能存在吡喃环。
胞外多糖扫描电镜表观形态分析:取充分干燥的胞外多糖组分,取少量涂抹于导电胶上,喷金后采用扫描电镜观察其表面形态。电镜结果如图16所示,植物乳杆菌R301 EPS呈现片状结果,随着放大倍数的增加可以看到其表面有细小颗粒。
实施例2:
本发明还对植物乳杆菌产的EPS功能活性进行了鉴定,本发明中的植物乳杆菌EPS具有抗氧化、抗炎活性,同时该植物乳杆菌具有很强的抗逆性,具体鉴定过程如下:
1.植物乳杆菌EPS体外抗氧化试验
(1)ABTS自由基清除试验
取粗EPS配制成不同浓度(0.5、1.0、2.0、4.0、6.0和8.0mg/mL)的溶液1mL,7mM的ABTS和2.4mM的过硫酸钾等体积混合,室温避光反应16个小时,用于生成ABTS自由基。ABTS用PBS(pH=7.4)稀释到734nm吸光光度值为0.7,然后将500μL的样品加到1mL的ABTS中避光反应10min,于734nm处测定吸光光度值。空白组以等体积无菌水代替ABTS自由基溶液,对照组以等体积无菌水代替EPS溶液,Vc做阳性对照。清除率计算公式:
清除率(%)=[1-(A1-A)/A0]×100%
式中:A为空白组的吸光光度值;A0为对照组的吸光光度值;A1为样品组的吸光光度值。
(2)DPPH自由基清除试验
取粗EPS配制成不同浓度(0.5、1.0、2.0、4.0、6.0和8.0mg/mL)的溶液1mL,与2mL0.2mM的DPPH无水乙醇溶液混合均匀,室温下避光反应30min,于517nm处测定吸光光度值。空白组以等体积乙醇代替DPPH溶液,对照组以等体积无菌水代替EPS溶液,Vc做阳性对照。清除率计算公式:
清除率(%)=[1-(A1-A)/A0]×100%。
式中:A为空白组的吸光光度值;0A0为对照组的吸光光度值;A1为样品组的吸光光度值。
3)羟基自由基清除试验
将1.5mL 1.8mM的水杨酸、2mL的1.8mM的硫酸亚铁、1mL 6mM的过氧化氢分别与1mL不同浓度的粗EPS(0.5、1.0、2.0、4.0、6.0、8.0mg/mL)混合均匀,在37℃下反应30min,3000rpm离心5min,取上清于510nm处测定吸光光度值。空白组以等体积的无菌水代替水杨酸,对照组为以等体积的无菌水替代EPS溶液,Vc作为阳性对照。清除率计算公式:
清除率(%)=[1-(A1-A)/A0]×100%。
式中:A为空白组的吸光光度值;A0为对照组的吸光光度值;A1为样品组的吸光光度值。
(4)还原力测定试验
取粗EPS配制成不同浓度(0.5、1.0、2.0、4.0、6.0和8.0mg/mL)的溶液1mL加入2.5mL的磷酸缓冲溶液(0.2M、pH=6.6)和2.5mL质量分数1%的铁氰化钾,混合均匀于50℃反应20min。随后加入2.5mL的三氯乙酸(10%,w/v)3000rpm离心10min。取2.5mL的上清加入等体积的蒸馏水和0.5mL的FeCl3(0.1%,w/v)混合均匀室温放置10min。于700nm处测定吸光光度值。样品还原力与吸光光度值成正比。
2.植物乳杆菌EPS细胞毒性试验
(1)细胞复苏
液氮取出的细胞冻存管置于37℃水浴锅水浴2min,待细胞完全融化后将冻存管内容物转移至15mL离心管并加入完全培养基10mL混匀,1000rpm离心4min。弃上清,加入5mL的完全培养基,吹打均匀后转移至25T细胞培养瓶,37℃5%二氧化碳静置培养,12~24h换一次液。
(2)细胞传代
正常培养的细胞贴壁至80%左右开始传代,倒掉培养瓶中的培养液,加入PBS清洗细胞,然后加入适量胰酶轻轻摇晃,培养箱培养0.5~4min,期间通过显微镜观察细胞状态,待大部分细胞变圆后立即加入10mL完全培养基以中和胰酶,将细胞悬浮液转移至离心管1000rpm离心4min。弃上清,加入适量完全培养基吹打均匀转移至细胞培养瓶传代培养。
(3)粗EPS细胞毒性试验
细胞毒性试验采用CCK-8试剂盒进行测定,细胞复苏后进行传代培养,待细胞状态良好时进行CCK-8试验:
1.利用完全培养基将细胞稀释至3×105个/mL,按照每孔100μL接种至96孔细胞培养板培养,阴性对照孔仅加入100μL完全培养基。将96孔细胞培养板于37℃,5%二氧化碳培养12h;
2.利用完全培养基将粗EPS稀释至不同浓度:0、20、40、60、80、100、500、1000μg/mL;分别吸取100μL粗EPS稀释液加入96孔细胞培养板,37℃,5%二氧化碳培养箱培养24h;
3.向每孔加入10μL CCK-8试剂,将细胞培养板于37℃,5%二氧化碳培养箱避光孵育2~4h;
4.用酶标仪测定其在450nm处的吸光光度值。
细胞存活率=[A(EPS)-A(空白)]/[A(对照)-A(空白)]×100%
A(EPS):具有细胞、CCK-8溶液和EPS溶液的孔的吸光光度值
A(空白):具有培养基和CCK-8溶液但没有细胞的孔的吸光光度值
A(对照):具有细胞、CCK-8溶液但没有EPS溶液的孔的吸光光度值
3.植物乳杆菌EPS抗炎活性试验
将RAW 264.7以每孔3.0×105个细胞的密度播种到96孔板中,培养12小时,在每孔中加入不同浓度的EPS,再培养12小时,然后在每孔中加入终浓度为1μg/mL的LPS,再培养24小时。按上述方法构建LPS诱导的炎症巨噬细胞模型。用Griess试剂测定一氧化氮(NO)含量。
将RAW 264.7细胞以每孔2.0×106个细胞的密度播种到6孔板中,随后的步骤按上述方法进行。收集细胞,提取RNA。所有样品的RNA完整性通过琼脂糖凝胶电泳和NanoDrop进行评估。将质量良好的RNA样品进行cDNA的反转录。qPCR使用引物见下表。本研究中使用了两步扩增法。95℃下30秒预孵化,40个循环,95℃下5秒变性,60℃下30秒延伸,然后1个循环,95℃下5秒,60℃下60秒,95℃下1秒熔解。基因表达与看家基因β-肌动蛋白的表达进行归一化,结果反映了相对于对照样本的倍数增加。

3.R301消化道抗逆性试验
(1)胆盐耐受性试验
向MRS液体培养基中添加牛胆盐至指定浓度:0.1、0.2、0.3、0.4、0.5g/L,将乳酸菌接种于含有不同浓度牛胆盐MRS培养基中培养12h后观察各组中菌株的生长情况,进行平板涂布计算活菌数量和存活率。
存活率=A0/A1×100%
注:A0为lg(处理0h活菌数),A1为lg(处理12h活菌数)
空白组为不添加牛胆盐的正常MRS液体培养基,每个处理组做三个重复。
(2)pH耐受性试验
使用1M HCl调节MRS液体培养基pH至指定值:1.0、2.0、3.0、4.0,将乳酸菌接种于不同pH值的MRS液体培养基中培养12h后观察各组中菌株的生长情况,进行平板涂布计算活菌数量和存活率。
存活率=A0/A1×100%
注:A0为lg(处理0h活菌数),A1为lg(处理12h活菌数)
空白组为自然pH值的MRS液体培养基,每个处理组做三个重复。
(3)模拟消化道存活试验
乳酸菌过夜培养后3000rpm离心10min弃上清,使用PBS清洗菌体两次,加入新鲜MRS培养基5mL和人工胃液5mL重悬菌体,200rpm 37℃摇床培养2h,分别于培养0h和2h取1mL菌液用于活菌计数;培养结束后3000rpm离心10min弃上清,使用PBS清洗菌体两次,加入新鲜MRS培养基5mL和人工小肠液5mL重悬菌体,200rpm 37℃摇床培养7h,分别于0h,2h,5h和7h取1mL菌液用于活菌计数。每个处理组做三个重复。
功能活性鉴定结果如下:植物乳杆菌的EPS具有很强的体外抗氧化能力,对ABTS自由基、DPPH自由基以及羟基自由基清除率达到100%,也具有一定的还原力。在0~100μg/mL浓度范围内,对正常细胞IPEC-J2和RAW 264.7均无损伤。并且EPS可以缓解由LPS引起的巨噬细胞炎症反应。此外还对R301进行模拟消化道抗逆性试验,结果表明R301可以很好地在人工消化液中存活,具有较强的消化道抗逆性。具体结果详见图17-20。
最后应说明的是:以上实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的精神和范围。
Claims (10)
1.一种植物乳杆菌(Lactobacillus plantarum)R301,其特征在于,其保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCCNo.26768。
2.一种菌剂,其特征在于,包括权利要求1所述植物乳杆菌(Lactobacillusplantarum)R301。
3.一种组合物,其特征在于,包括权利要求1所述植物乳杆菌(Lactobacillusplantarum)R301或权利要求2所述菌剂。
4.一种植物乳杆菌培养基,其特征在于,每100ml植物乳杆菌培养基中包括以下原料:1-4g葡萄糖,1-4g小肽,0.2-0.6g磷酸氢二钾,0.5-0.6g无水乙酸钠,0.2-0.4g柠檬酸三胺,0.01-0.05g七水合硫酸镁,0.01-0.1g无水硫酸锰和0.1-0.5ml Tween-80;
优选的,余量为水;
优选的,所述植物乳杆菌培养基用于培养权利要求1所述植物乳杆菌(Lactobacillusplantarum)R301。
5.一种植物乳杆菌的培养方法,其特征在于,培养基初始pH值6.5-7.5,种子液接种量2-7%(V/V);
优选的,发酵温度20-35℃;
优选的,利用植物乳杆菌培养基进行发酵,每100ml植物乳杆菌培养基中包括0.01-0.05g七水合硫酸镁;
优选的,利用权利要求1所述植物乳杆菌(Lactobacillus plantarum)R301和/或权利要求4所述植物乳杆菌培养基。
6.一种胞外多糖的制备方法,其特征在于,包括利用权利要求1所述植物乳杆菌(Lactobacillus plantarum)R301、权利要求2所述菌剂、权利要求3所述组合物、权利要求4所述植物乳杆菌培养基、权利要求5所述植物乳杆菌的培养方法中的一种或几种得到发酵液,从发酵液中提取胞外多糖;
优选的,对所述发酵液除菌体、乙醇沉淀、除蛋白、经离子交换柱、琼脂糖凝胶柱洗脱,得到胞外多糖,进一步优选得到分子量为3.1×106Da的胞外多糖;
优选的,所述胞外多糖的单糖组成为甘露糖、鼠李糖、葡萄糖、半乳糖和阿拉伯糖,单糖组成的摩尔比例为5.7:35.8:27.5:15.8:15.2。
7.一种胞外多糖,其特征在于,其由权利要求6所述胞外多糖的制备方法制备而成。
8.权利要求1所述植物乳杆菌(Lactobacillus plantarum)R301、权利要求2所述菌剂、权利要求3所述组合物、权利要求4所述植物乳杆菌培养基、权利要求5所述植物乳杆菌的培养方法中的一种或几种在制备胞外多糖中的应用。
9.植物乳杆菌胞外多糖在抗氧、抗炎或抗逆中的应用;
优选的,所述植物乳杆菌胞外多糖为权利要求7所述胞外多糖。
10.植物乳杆菌胞外多糖在以下任一项中的应用:
1)ABTS自由基清除;
2)DPPH自由基清除;
3)羟基自由基清除;
4)减少NO的产生;
5)缓解NF-κB的应激反应;
6)降低促炎因子IL-6的表达水平;
优选的,所述植物乳杆菌胞外多糖为权利要求7所述胞外多糖。
