Abstract
本发明属于生物医药和分子生物学技术领域,具体涉及一种高效杀伤产肠毒素脆弱拟杆菌的细菌素及其应用。本发明提供的细菌素,不仅杀伤脆弱拟杆菌且具有极强的杀菌活性,敏感菌株的半数抑菌浓度远低于常使用的抗生素。通过表达纯化该细菌素或利用该株非产肠毒素脆弱拟杆菌分泌上清实现了对ETBF的有效杀伤。该细菌素可作为一种有前途的抗菌药物,用于治疗和预防ETBF引起的炎症性疾病和对抗ETBF介导的结直肠癌发展。因此具有良好的实际应用之价值。

技术领域
本发明属于生物医药和分子生物学技术领域,具体涉及一种高效杀伤产肠毒素脆弱拟杆菌的细菌素及其应用。
背景技术
本发明背景技术中公开的信息仅仅旨在增加对本发明的总体背景的理解,而不必然被视为承认或以任何形式暗示该信息构成已经成为本领域一般技术人员所公知的现有技术。
人体肠道菌群是目前已知最复杂的微生物群落之一,与宿主建立了稳定的长期联系,当人体肠道菌群失衡时,例如机会致病菌发生易位,肠道生理环境发生扰动,肠道稳态被打破,可导致诸如炎症性肠病、急性腹泻等疾病的发生,严重时可发展至结直肠癌。
脆弱拟杆菌(Bacteroides fragilis)为革兰氏阴性短杆菌,拟杆菌属,异养厌氧型,无芽孢,其是人体肠道共生菌的重要组成部分,主要存在于远端结肠。某些脆弱拟杆菌能够合成并分泌分子量约为20kDa的脆弱拟杆菌肠毒素(Bacteroides fragilis toxin,BFT)。根据能否编码分泌BFT可将脆弱拟杆菌分为产肠毒素脆弱拟杆菌(EnterotoxigenicBacteroides fragilis,ETBF)与非产肠毒素脆弱拟杆菌(Non-toxigenic Bacteroidesfragilis,NTBF)。BFT是ETBF引起细胞和组织损伤的主要毒力因子,先前研究发现BFT可引起畜禽和人类腹泻,后续研究认为其是炎症性肠病的危险因素,于患者的粪便以及活检标本中被广泛检出。近年来,越来越多的临床以及实验证据证明BFT与结直肠癌的发展相关,结直肠癌患者ETBF的检出率显著高于非结直肠癌患者。
一般认为ETBF可通过多方面因素促进结直肠癌发展。(1)ETBF分泌的BFT实质是一种金属蛋白酶,可切割E-cadherin的细胞外结构域,E-cadherin是粘附连接的主要结构成分,负责细胞粘附,一方面E-cadherin的失活导致肠上皮细胞间紧密连接的破坏从而增加肠道通透性,同时E-cadherin的破坏启动了β-catenin的核定位,从而诱导c-myc表达并导致持续的细胞增殖,不受控制的细胞增殖会促进结直肠癌发展。(2)ETBF定植多发性肠瘤模型小鼠时,Stat-3信号通路被选择性激活,随后发生结肠粘膜Th17反应,上调IL-17的表达,引发癌变。在该模型中单独阻断IL-17或阻断IL-17和IL-23受体结合可减少结肠肿瘤的形成,表明Stat3/Th17适应性免疫促进结直肠癌发展。(3)BFT还可通过激活NF-κB和MAPKs通路促进肠上皮细胞合成并分泌细胞因子加重肠道炎症,形成异常的免疫微环境,促进肿瘤发展。目前亦有研究发现,在多发性肠瘤小鼠疾病模型中,ETBF的定植时长和肿瘤体积呈正相关,ETBF被清除后,小鼠炎症指标有所改善。由此可见,因ETBF是结直肠癌发展的危险因素之一,如果能有效杀伤ETBF,将可能减轻炎症性肠病的症状,减缓结直肠癌的发展。目前在小鼠模型中常采用抗生素如头孢西丁处理ETBF,但抗生素的靶向范围较广,很大程度上会影响肠道菌群结构,由肠道菌群结构变化引起的后续反应尚不可知,且频繁施用抗生素会导致抗生素耐药。目前尚未有一类特异性强且效率高的杀伤ETBF的抗菌剂。
发明内容
针对现有技术的不足,本发明的目的在于提供一种高效杀伤产肠毒素脆弱拟杆菌的细菌素及其应用。经试验证明,该细菌素能够有效杀伤脆弱拟杆菌且具有极强的杀菌活性。基于上述研究成果,从而完成本发明。
为了实现上述技术目的,本发明提供的技术方案如下:
本发明的第一个方面,提供一种细菌素,所述细菌素的氨基酸序列选自:
a1)如SEQ ID NO.1所示的氨基酸序列;或,
a2)如SEQ ID NO.1所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的多肽。
进一步的,所述a2)中,所述细菌素选自下组中任意一个或多个位点发生突变:N7A、W11K、S18E、Q28A、K40Q、D73A,其氨基酸残基编号采用SEQ ID NO.1所示的编号。
本发明通过试验验证,上述发生突变后获得的细菌素(即细菌素突变体)其杀菌活性发生改变,除细菌素N7A突变体外,其余突变体均具有较强的杀菌活性。且相较于野生型细菌素,细菌素D73A突变体活性增强较为明显。
本发明的第二个方面,提供一种核酸分子,所述核酸分子能够编码上述细菌素。
本发明的第三个方面,提供一种重组表达载体,所述重组表达载体包含第二方面所述的核酸分子。
本发明的第四个方面,提供一种宿主细胞,所述宿主细胞含有本发明第三方面所述的重组表达载体或染色体整合有本发明第二方面所述的核酸分子或者能够表达本发明第一方面所述细菌素。
也因此,所述宿主细胞可以是天然存在的细菌(如拟杆菌属细菌),亦可为经人工改造获得的基因工程菌(如大肠杆菌基因工程菌)。
进一步的,本发明还同时保护上述宿主细胞的发酵物或其代谢产物。
本发明的第五个方面,提供一种制备本发明第一方面所述细菌素的方法,包括步骤:培养本发明第四方面所述的宿主细胞,从而表达出所述细菌素;和分离纯化所述的细菌素。
本发明的第六个方面,提供上述细菌素、宿主细胞或者宿主细胞的发酵物或其代谢产物在作为或制备抗菌剂中的应用。
具体的,所述抗菌剂表现出对脆弱拟杆菌(特别是产肠毒素脆弱拟杆菌ETBF)强烈的杀伤活性,因此可用于环境消杀等方面。特别地,感染ETBF可导致人和非人动物罹患炎症性疾病(如炎症性腹泻、结肠炎)以及肿瘤(如结直肠癌)。
因此,本发明的第七个方面,提供上述细菌素、宿主细胞或者宿主细胞的发酵物或其代谢产物在制备产肠毒素脆弱拟杆菌感染相关疾病的药物。
同时,需要说明的是,考虑到ETBF对人和非人动物均可导致患病,因此所述药物可以为人用药和动物用药,其中,所述动物用药包含药物饲料添加剂。
本发明的第八个方面,提供一种治疗产肠毒素脆弱拟杆菌感染相关疾病的方法,所述方法包括向受试者施用治疗有效量的上述细菌素或抗菌剂或药物。
上述一个或多个技术方案的有益技术效果:
上述技术方案提供的细菌素,不仅杀伤脆弱拟杆菌且具有极强的杀菌活性,敏感菌株的半数抑菌浓度远低于常使用的抗生素。通过表达纯化该细菌素或利用该株非产肠毒素脆弱拟杆菌分泌上清实现了对ETBF的有效杀伤。该细菌素可作为一种有前途的抗菌药物,用于治疗和预防ETBF引起的炎症性疾病和对抗ETBF介导的结直肠癌发展。因此具有良好的实际应用之价值。

本发明的一个典型具体实施方式中,提供一种细菌素,所述细菌素的氨基酸序列选自:
a1)如SEQ ID NO.1所示的氨基酸序列;或,
a2)如SEQ ID NO.1所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的多肽。
进一步的,所述a2)中,所述细菌素选自下组中任意一个或多个位点发生突变:N7A、W11K、S18E、Q28A、K40Q、D73A,其氨基酸残基编号采用SEQ ID NO.1所示的编号。
本发明通过试验验证,上述发生突变后获得的细菌素(即细菌素突变体)其杀菌活性发生改变,除细菌素N7A突变体外,其余突变体均具有较强的杀菌活性。且相较于野生型细菌素,细菌素D73A突变体活性增强较为明显。
需要说明的是,上述SEQ ID NO.1所示的氨基酸序列(MQVFIKNRYGWTITLEVSPTDTVENVKQKIQDKEGFPPDKIRLIYGGKQME DGRTLADYNVQKDSTILICIRDVDC)所构成的细菌素为不含信号肽的成熟细菌素,而其前体(含信号肽)氨基酸序列则如SEQ ID NO.2所示(MRFIKQVLLTITLCNIMLFALPSTVNAMQVFIKNRYGWTITLEVSPTDTVEN VKQKIQDKEGFPPDKIRLIYGGKQMEDGRTLADYNVQKDSTILICIRDVDC)。
本发明的又一具体实施方式中,提供一种核酸分子,所述核酸分子能够编码上述细菌素。
本发明的又一具体实施方式中,所述核酸分子具有如SEQ ID NO.3所示的核苷酸序列。
本发明的又一具体实施方式中,提供一种重组表达载体,所述重组表达载体包含所述核酸分子。
根据本发明,所述重组表达载体通过上述核酸分子有效地连接到表达载体上获得,所述表达载体为病毒载体、质粒、噬菌体、黏粒、或人工染色体中的任意一种或多种;病毒载体可包括腺病毒载体、逆转录病毒载体或腺伴随病毒载体,人工染色体包括细菌人工染色体(BAC)、噬菌体P1衍生的载体(PAC)、酵母人工染色体(YAC)或哺乳动物人工染色体(MAC)。本发明的又一具体实施方式中,所述表达载体为质粒,具体可以为pET-28a质粒。
本发明的又一具体实施方式中,提供一种宿主细胞,所述宿主细胞含有所述重组表达载体或染色体整合有所述核酸分子或者能够表达所述细菌素。
所述宿主细胞可以为原核细胞或真核细胞。
本发明的又一具体实施方式中,所述宿主细胞是细菌细胞或真菌细胞;
其中所述细菌细胞为拟杆菌属、埃希氏菌属、农杆菌属、芽孢杆菌属、链霉菌属、假单胞菌属或葡萄球菌属内的任一种;
本发明的又一具体实施方式中,所述细菌细胞为脆弱拟杆菌、大肠杆菌(如大肠杆菌BL21)、根癌农杆菌(如GV3101)、发根农杆菌、乳酸乳球菌、枯草芽孢杆菌、蜡状芽孢杆菌或荧光假单胞菌。
所述真菌细胞包括酵母菌。
也因此,所述宿主细胞可以是天然存在的细菌(如拟杆菌属细菌),亦可为经人工改造获得的基因工程菌(如大肠杆菌基因工程菌)。
进一步的,本发明还同时保护上述宿主细胞的发酵物或其代谢产物。
本发明中,术语“发酵物”用于指代发酵产品。相应的发酵物可以是从发酵培养宿主细胞的过程获得的液体,因此,也可称为发酵液;液体可以含有宿主细胞(胞体),但并不必然需要含有宿主细胞(胞体)。液体优选的含有由本发明的宿主细胞产生的代谢产物,所述代谢产物尤其包括上述细菌素。
以及,在本发明的实施方式中,包含菌体的发酵液或培养液经离心、过滤、沉降或所属领域中已知的其他手段将在发酵液或培养液中生长的菌体细胞与液体分离,去除菌体细胞时所剩余的液体为“上清液”,并且在本发明中,上清液内含有宿主细胞的胞外代谢产物。在本发明的实施方式中,抗菌剂也可以包含该上清液。
以及,在本发明的实施方式中,包含菌体的发酵液或培养液经离心、过滤、沉降或所属领域中已知的其他手段将在发酵液或培养液中生长的菌体细胞与液体分离获取菌体,菌体可进行破碎获取菌体破碎物,破碎方式可以为超声(比如冰浴超声破碎细胞)或者本领域已知的其他手段,或者,更进一步的,对该菌体破碎物离心收集上清液,该上清液记为无细胞提取物,并且在本发明中,该菌体破碎物或无细胞提取物内含有宿主细胞的胞内代谢产物。在本发明的实施方式中,抗菌剂也可以包含该菌体破碎物或无细胞提取物。
以及,在本发明的实施方式中,为方便存储、运输,提高菌株存活率等,所述抗菌剂也可以是固体,进一步优选为冻干粉剂。即针对上述宿主细胞或其发酵物或其代谢产物进一步进行冷冻干燥获得,所述冷冻干燥技术(包括真空冷冻干燥技术)可采用常规方法进行,在此不再赘述。
本发明的又一具体实施方式中,提供一种制备所述细菌素的方法,包括步骤:培养所述宿主细胞,从而表达出所述细菌素;和分离纯化所述细菌素。
本发明的又一具体实施方式中,提供上述细菌素、宿主细胞或者宿主细胞的发酵物或其代谢产物在作为或制备抗菌剂中的应用。

其中,所述产肠毒素脆弱拟杆菌感染相关疾病包括但不限于炎症性疾病(如炎症性腹泻、结肠炎)以及肿瘤(如结直肠癌)。
进一步的,所述药物还可以包括至少一种药物非活性成分。所述药物非活性成分可以是药学上可接受的载体。所述药学上可接受的载体可为缓冲剂、乳化剂、悬浮剂、稳定剂、防腐剂、赋形剂、填充剂、凝结剂与调和剂、界面活性剂、扩散剂或消泡剂。
所述可药用载体还可以为病毒、微囊、脂质体、外泌体、纳米颗粒或聚合物及其任意组合。所述可药用载体的递送载剂可为脂质体、外泌体、生物相容性聚合物、脂蛋白、脂多糖、人工病毒包膜、无机颗粒、以及细菌或病毒、噬菌体、黏粒或质粒载体等。在此不再赘述。
所述药物还可以以单独的组合物或与主要的活性成分不同的剂量形式单独给予其它预防和/或治疗性化合物。主要成分的部分剂量可以与其它治疗性化合物同时给药,而其它剂量可以单独给药。在治疗过程中,可以根据症状的严重程度、复发的频率和治疗方案的生理应答,调整本发明药物的剂量。
本发明的药物可通过已知的方式施用至体内。例如通过静脉全身递送或者局部注射递送到感兴趣组织中。可选地经由静脉内、经皮、鼻内、粘膜或其他递送方法进行施用。这样的施用可以经由单剂量或多剂量来进行。本领域技术人员理解的是,本发明中有待施用的实际剂量可以在很大程度上取决于多种因素而变化,如靶细胞、生物类型或其组织、待治疗受试者的一般状况、给药途径、给药方式等等。
同时,需要说明的是,考虑到ETBF对人和非人动物均可导致患病,因此所述药物可以为人用药和动物用药,其中,所述动物用药包含药物饲料添加剂。

所述受试者是指已经是治疗、观察或实验的对象的动物,优选指哺乳动物,最优选指人。所述“治疗有效量”是指包括本发明细菌素或药物的量,该量可引起研究者、兽医、医生或其他医疗人员所追求的组织系统、动物或人的生物学或医学响应,这包括减轻或部分减轻受治疗的疾病、综合征、病症或障碍的症状。必须认识到,本发明所述活性成分的最佳给药剂量和间隔是由其性质和诸如给药的形式、路径和部位以及所治疗的特定动物等外部条件决定的,而这一最佳给药剂量可用常规的技术确定。同时也必须认识到,最佳的疗程,即同时化合物在额定的时间内每日的剂量,可用本领域内公知的方法确定。
以下通过实施例对本发明做进一步解释说明,但不构成对本发明的限制。应理解这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中为注明具体条件的试验方法,通常按照常规条件进行。
实施例
1.实验方法
1.1细菌素(前体)的氨基酸序列
其中,加粗下划线部分所示的氨基酸序列即为信号肽。
1.2细菌素的核苷酸序列(含信号肽)

1.3细菌素的表达载体构建
利用Gibson assembly方法将去除信号肽的细菌素编码核苷酸克隆至pET-28a载体中,细菌素N端引入6HIS-SUMO标签便于纯化,克隆位点位于载体多克隆位点处。骨架质粒pET-28a为常见的大肠杆菌表达载体,完整的载体图谱可见Addgene(https://www.addgene.org/vector-database/2565/)。
1.4细菌素的表达纯化
细菌素的表达纯化如以下步骤。
1)种子液制备:将测序正确的质粒转化进E.coli BL21,涂布于补充有50μg/mL卡那霉素的LB琼脂平板。从平板上挑取单克隆接种于10mL左右小体积LB液体培养基中,37℃,200rpm过夜培养。
2)大体积接种诱导表达:将种子液以1:100的比例接种至1L补充有50μg/mL卡那霉素的LB液体培养基,37℃培养约4h,BL21生长至OD600约等于0.8。冷却培养基至22℃后加入终浓度为0.2mM的IPTG,诱导表达约15-18h。
3)亲和层析初步纯化目的细菌素:以4000rpm,4℃,20min收集菌液,弃上清,用30mL lysis buffer(20mM Tris 8.0,150mM NaCl)重悬菌液,加入300μL 1mM PMSF用于抑制内源蛋白酶活性。700Mpa高压破碎重悬菌液,破碎完全的菌液在17000g,4℃条件下离心50min,离心完成后,取破碎液上清进行镍柱亲和层析。破碎液上清循环挂柱3-4次后,加入50μL 2mg/mL的ULP1酶进行柱上酶切,室温柱上酶切约3-4h后,每L LB收集3-6mL流穿液,因细菌素所带的HIS-SUMO标签已被ULP1酶切,因此流穿液中即包含目的细菌素。
4)凝胶排阻层析(分子筛)再次纯化目的细菌素:利用3kDa蛋白超滤管将流穿液浓缩至1mL。浓缩液于14000rpm,4℃,离心5min用以去除可能存在的蛋白沉淀,取上清进行凝胶排阻层析。凝胶排阻层析所使用的分离柱为Superdex 75gel-filtrationchromatography。根据纯化峰型判断细菌素的均一性,根据出峰位置判断细菌素的聚集状态,该细菌素性质较为均一,为单体。收集不同出峰位置的细菌素进行SDS-PAGE检测其纯度。
1.5半数抑菌浓度的测定
利用细菌素测定待测ETBF菌株的半数抑菌浓度如以下步骤。
1)制备种子液:划线ETBF于新鲜BHIS(在BHI培养基基础上补充有1g/L半胱氨酸,5mg/L血红素)琼脂平板。挑取单克隆于1mL液体BHIS培养基,过夜培养。
2)半数抑菌浓度测定:将种子液以1:100的比例接种至10mL BHIS液体培养基中,37℃厌氧生长约4-5h至对数生长期,此时OD600约等于0.6。将菌液以1:200接种至200μL补充有不同浓度梯度的抗生素或纯化细菌素的BHIS培养基中,每一浓度梯度重复四次。培养约14h-18h后,测定待测菌的OD600。与不添加抗生素或纯化细菌素组OD600值对比,OD600为其一半对应的抗生素浓度或细菌素浓度即为半数抑菌浓度。
1.6琼脂斑点法检测ETBF对于细菌素的敏感性
利用琼脂斑点法检测ETBF对于细菌素的敏感性如以下步骤。
1)制备种子液:划线ETBF于新鲜BHIS琼脂平板。挑取单克隆于1mL液体BHIS培养基,过夜培养。
2)琼脂斑点法检测ETBF对于细菌素的敏感性:将种子液以1:100的比例接种至5mLBHIS液体培养基中,37℃厌氧生长约4-5h至对数生长期,此时OD600约等于0.6。取100μg纯化的细菌素涂布于新鲜BHIS琼脂平板,待彻底吹干后,取2μL 10倍梯度稀释的菌液点在涂布有细菌素的平板上,同时点相同体积的菌液于无细菌素的平板以做对照。
1.7非产肠毒素脆弱拟杆菌分泌上清抑制ETBF生长实验
利用非产肠毒素脆弱拟杆菌分泌上清抑制ETBF生长实验如以下步骤。
1)制备种子液:划线NTBF,NTBF△Bacteriocin,ETBF于新鲜BHIS琼脂平板。挑取单克隆于1mL液体BHIS培养基,过夜培养。
2)浓缩NTBF分泌上清:将种子液以1:100的比例接种至10mL BHIS液体培养基中,37℃厌氧生长约4-5h至对数生长期,此时OD600约等于0.6。取NTBF,NTBF△Bacteriocin菌液,9000g,4℃,离心10min,取上清,0.22μm滤膜过滤除菌。利用3kDa蛋白超滤管将过滤后的10mL上清浓缩至200μL。取5-20μL浓缩上清点在新鲜BHIS琼脂平板上,彻底吹干后待用。
3)抑制ETBF生长能力测定:在厌氧箱中,将50μL处于对数生长期的加入5mL 0.8%BHIS软琼脂中,混匀后均匀倒在已补充浓缩上清的BHIS琼脂平板上,生长约18-24h后,观察抑菌圈大小。
1.8细菌素理性突变
利用Quick Change mutagenesis方法对野生型细菌素进行定点突变,野生型细菌素如1.1所示,所述突变体及其对应的氨基酸序列(已去除信号肽)分别为

利用Quick Change mutagenesis对野生型细菌素进行理性突变的步骤为
1.利用突变引物,以1.3中所述含有野生型细菌素的表达载体为模板,进行PCR扩增,胶回收目的条带。
2.利用DpnI酶处理胶回收产物,使得可能混入回收产物的PCR模板被酶切完全。
3.取适量胶回收产物直接转化E.coli BL21,挑取单克隆,提质粒测序验证突变体是否构建成功,保存阳性克隆用于突变体表达。
1.9细菌素突变体活性检测
构建好细菌素突变体表达载体后,以1.4所述方法纯化突变体。以1.5及1.6所述方法检测突变体活性。
2.实验结果
2.1细菌素的表达纯化
此细菌素大小为8.8kDa,其分子筛出峰位置约为15mL,为单体,分子筛峰型较锐利而单一,纯化的细菌素状态较好,其SDS-PAGE胶图如图1所示。
2.2ETBF半数抑菌浓度的测定
该细菌素对的待测ETBF半数抑菌浓度约为0.5μg/mL,而常用的抗生素如氯霉素(Chloramphenicol,Cam),其半数抑菌浓度<3.125μg/mL,根据不同抑菌剂分子量计算,该细菌素的抑菌能力远强于常用的抗生素如氯霉素、红霉素及四环素。结果如图2所示。
2.3琼脂斑点法检测ETBF对于细菌素的敏感性
利用琼脂斑点法检测ETBF对于该细菌素的敏感性,GS084为实验室保存的一株对该细菌素具有抗性的NTBF,ETBF-1,ETBF-2为实验室保存的两株ETBF。在补充有细菌素的平板上,两株ETBF均无法正常生长,结果如图3所示。
2.4非产肠毒素脆弱拟杆菌分泌上清抑制ETBF生长实验
利用编码该细菌素的非产肠毒素脆弱拟杆菌分泌上清抑制ETBF生长,我们首先在非产肠毒素脆弱拟杆菌基因组上敲除了该细菌素的编码基因,以便衡量该细菌素在杀伤ETBF时的贡献。ETBF抑菌圈与浓缩上清点样体积成正比,敲除了该细菌素编码基因的浓缩上清无法观测到ETBF抑菌圈,结果如图4所示。
2.5细菌素突变体活性检测
对细菌素各突变体进行纯化,利用纯化的细菌素处理ETBF,检测各个突变体对于ETBF的半数抑菌浓度以及抑制ETBF生长情况。结果表明,多个细菌素突变体活性有明显改变,除细菌素N7A突变体外,其余突变体具有较强的杀菌活性。相较于野生型细菌素,细菌素D73A突变体活性增强较为明显。
通过对细菌素进行理性设计,最终获得多个仍保有活性的突变体,包括具有增强活性的D73A突变体。结果如图5、6所示。
本发明未尽事宜为公知技术。

Claims (11)
1.一种细菌素,其特征在于,所述细菌素在SEQ ID NO.1所示的氨基酸序列基础上发生下组中任意一个位点的突变:S18E、Q28A、K40Q、D73A。
2.一种核酸分子,其特征在于,所述核酸分子能够编码权利要求1所述细菌素。
3.一种重组表达载体,其特征在于,所述重组表达载体包含权利要求2所述核酸分子。
4.如权利要求3所述的重组表达载体,其特征在于,所述重组表达载体通过权利要求2所述核酸分子有效地连接到表达载体上获得,所述表达载体为病毒载体、质粒、噬菌体、黏粒、或人工染色体中的任意一种或多种。
5.如权利要求4所述的重组表达载体,其特征在于,所述表达载体为质粒,具体为pET-28a质粒。
6.一种宿主细胞,其特征在于,所述宿主细胞含有权利要求3-5任一项所述重组表达载体或染色体整合有权利要求2所述核酸分子或者能够表达权利要求1所述细菌素。
7.如权利要求6所述的宿主细胞,其特征在于,所述宿主细胞为经人工改造获得的基因工程菌。
8.一种制备权利要求1所述细菌素的方法,其特征在于,包括步骤:培养权利要求6或7所述宿主细胞,从而表达出所述细菌素;和分离纯化所述细菌素。
9.权利要求1所述细菌素、权利要求6或7所述宿主细胞在制备抗菌剂中的应用,所述抗菌剂表现出对产肠毒素脆弱拟杆菌的杀伤活性;所述抗菌剂用于环境消杀。
10.权利要求1所述细菌素、权利要求6或7所述宿主细胞在制备治疗产肠毒素脆弱拟杆菌感染引起的疾病的药物中的应用。
11.如权利要求10所述的应用,所述产肠毒素脆弱拟杆菌感染引起的疾病包括炎症性腹泻、结肠炎。
