And (3) detecting the content of fibroblast growth factors: the effect of the tightening composition on the fibroblast growth factor content of the female mouse vagina was verified.
Female Kunming mice (8-9 months retired species) were purchased from Peking Vitolith laboratory animal technologies Inc.
Mouse Fibroblast Growth Factor (FGF) ELISA assay kit (cat# XZK 20221160) was purchased from Shanghai Xuanzekang BioCo.
Experimental samples: the compacting compositions of examples 1-3 and comparative examples 1-4.
The experimental method comprises the following steps: after 24 mice were acclimatized for 7d, the mice were randomly divided into 8 groups of 3 mice each, with a control group of 1 and an experimental group of 7. Vaginal administration of mice was started after 7 d: the medicine is administrated once daily, the dose is 0.2mL each time, 0.2mL of experimental sample (without bubbles) is sucked by a 0.25mL syringe (without needle core) and slowly pushed into the vagina of a female mouse, then the female mouse is put back into a cage, the medicine is continuously administrated for 7d, and the administration of the control group is 0.2mL of 1, 3-propanediol.
And (3) detecting the content of the fibroblast growth factor: mice were sacrificed at 8d, vaginal tissue was dissected, blotted dry with filter paper after glass fat, and PBS phosphate buffer saline (0.01 mol/L) was added with a mass ratio of tissue to buffer of 1:9, preparing a tissue homogenate mixture of PBS. Grinding the tissue homogenate mixture by a homogenizer, centrifuging by a centrifuge, taking supernatant, detecting the content of the mouse fibroblast growth factor (pg/mL) by using the supernatant of the mouse fibroblast growth factor ELISA detection kit, and taking an average value of experimental results as shown in table 1.

The experimental method comprises the following steps: 130 test persons were equally divided into 13 groups of 10 persons each, and the tightening liquid 14d of each application example and comparative application example was continuously used once a day.

by combining examples 1 to 3, comparative examples 1 to 4 and Table 1, it is possible to obtain a compact composition comprising, as functional substances, extracts of lime peel, cornel fruit, pomegranate peel and shizandra berry, which has excellent fibroblast growth promoting effect and vagina compacting effect. The applicant specifically analyzes that the compound of the lime peel extract, the dogwood fruit extract, the pomegranate peel extract and the shizandra berry extract has specificity, has unexpected promotion effect on the generation of fibroblast growth factors, and the increase of the fibroblast growth factors promotes the proliferation and migration of the fibroblasts, so that a large number of the fibroblasts are concentrated on the inner wall of the vagina, secrete synthetic elastic fibers, repair the damaged elastic fibers in the vagina, increase the content of the elastic fibers and collagen in the inner wall of the vagina, recover the elasticity of the inner wall of the vagina, improve the vaginal relaxation, realize the tightening of the vagina, and further achieve the effect of good tightening and maintaining the vagina by being assisted with the effects of the four plant extracts.
By combining application examples 5 to 6, application example 3 and Table 2, it can be obtained that the tightening liquid added with the lime peel extract, the dogwood fruit extract, the pomegranate peel extract, the shizandra berry extract, the evodia fruit extract and the myrtle fruit extract has better effect of tightening and maintaining vagina. The specific analysis of the inventor proves that the plant extracts have good antibacterial and nourishing effects, and the evodia rutaecarpa extract and myrtle fruit extract can enhance the matching effect of the plant extracts, so that the nourishing effect of the tightening composition on the inner wall of the vagina is further improved, and the ecological balance in the vagina is further maintained.
By combining application examples 6, 7-9 and Table 2, it can be obtained that the tightening liquid containing lime peel extract, dogwood fruit extract, pomegranate peel extract, shizandra berry extract, evodia rutaecarpa fruit extract, myrtle fruit extract and aminomethylpropanol has good effect of regulating the pH value of vaginal secretion. The inventor specifically analyzes that the aminomethyl propanol is added into the tightening liquid as a neutralizing agent, has good effect of regulating the pH value of the tightening liquid, so that the pH value of the tightening liquid is acidic, and is more suitable for being used in the intravaginal environment, while the lime peel extract, the dogwood fruit extract, the pomegranate peel extract, the shizandra berry extract, the evodia fruit extract and the myrtle fruit extract are added into the tightening liquid as plant extracts, especially the evodia fruit extract and the myrtle fruit extract enhance the matching effect of the plant extracts, and the aminomethyl propanol is added, so that the tightening liquid has the best combination effect, and the unexpected outstanding effect of regulating the pH value of vaginal secretion is reflected, thereby further regulating the intravaginal environment, further maintaining the ecological balance of the intravaginal environment, and further improving the tightening and maintaining effect of the tightening liquid on the vagina.

The tightening composition is characterized by comprising a solvent and a plant extract, wherein the plant extract accounts for 61.24% -82.46% of the tightening composition in mass percent, and the plant extract at least comprises a lime peel extract, a dogwood fruit extract, a pomegranate peel extract and a shizandra berry extract.

The tightening composition according to claim 1, wherein the lime peel extract accounts for 10% -20% of the tightening composition in mass percent; the cornus officinalis fruit extract accounts for 40% -46% of the tightening composition; the pomegranate peel extract accounts for 10% -14% of the tightening composition; the schisandra chinensis extract accounts for 1% -2% of the tightening composition.
3. The tightening composition of claim 1, wherein the plant extract further comprises evodia rutaecarpa extract, myrtle extract.
4. A compacting composition according to claim 3, characterised in that it comprises, in mass percent:
extract of lime peel: 10% -20%;
fructus Corni extract: 40% -46%;
pomegranate peel extract: 10% -14%;
schisandra chinensis extract: 1% -2%;
evodia rutaecarpa fruit extract: 0.2% -0.4%;
myrtle fruit extract: 0.04% -0.06%;
the balance being solvent.
5. A method of preparing the tightening composition of claim 3 or 4, characterized by the following: mixing fructus Citri sinensis peel extract, corni fructus extract, pericarpium Granati extract, fructus Schisandrae extract, fructus evodiae extract, fructus Rhodomyrti extract, and solvent, and stirring to obtain compact composition.
6. A tightening liquid containing the tightening composition according to any one of claims 1 to 4, characterized by comprising the following components in mass percent: 1% -2% of carbomer, 10% -20% of butanediol, 1% -2% of aminomethylpropanol, 20% -30% of a tightening composition, 0.2% -0.5% of phenoxyethanol and the balance of water.
7. A process for preparing a tightening fluid according to claim 6, comprising the steps of:
step one: mixing carbomer and water, homogenizing, heating to 80-85 ℃, and uniformly stirring to obtain a first content; mixing and stirring the butanediol and the compact composition uniformly to obtain a second content;
step two: stirring and cooling the first content to 40-45 ℃, and slowly pouring the second content into the first content in a stirring state;
step three: adding aminomethyl propanol and phenoxyethanol, and stirring to obtain tightening solution.
8. The process for preparing a tightening liquid according to claim 7, wherein in the first step, the first content is obtained by homogenizing for 5-8 min at 2000-2200 r/min and stirring for 10-15 min at 200-220 r/min; stirring for 2-5 min at 200-220 r/min to obtain a second content;
stirring for 10-15 min at 200-220 r/min;
and thirdly, stirring for 10-15 min at 200-220 r/min.
