Abstract
本发明公开了一种凝结芽孢杆菌AP15及其在肠道调节与尿酸、胆固醇代谢的应用。本发明所提供的凝结芽孢杆菌具体为凝结芽孢杆菌AP15(Bacillus coagulans AP15),它在中国微生物菌种保藏管理委员会普通微生物中心的保藏编号为CGMCC No.26711。本发明的凝结芽孢杆菌AP15(Bacillus coagulans AP15)CGMCC No.26711具有良好的降尿酸、降胆固醇功能,同时能够提高肠道中益生菌的丰度,抑制肠道中有害细菌的生长,调节肠道菌群,促进肠道中嘌呤、尿酸和胆固醇的分解代谢,调控尿酸、胆固醇代谢。同时,本发明的凝结芽孢杆菌可制成菌剂用于预防或治疗高尿酸血症、痛风、高胆固醇,是一种绿色、安全的菌剂。

技术领域
本发明属于生物技术领域,尤其涉及一种凝结芽孢杆菌AP15及其在肠道调节与尿酸、胆固醇代谢的应用。
背景技术
在“健康中国”战略大背景下,人们的价值观念已经从“以治病为中心”转变为“以人民健康为中心”。而随着经济的飞速发展,人们的生活方式发生了巨大的改变。其中,愈来愈丰盛的饮食使得人们对高蛋白、高脂肪的摄入不断增加,从而提高了许多疾病发生的几率,如:高尿酸血症。长期的高尿酸会引起痛风、高血压、心血管疾病及糖尿病等相关代谢障碍及疾病。根据2017年中国痛风现状报告白皮书报道:当时中国高尿酸血症患者已达1.7亿,其中痛风患者8000多万人,并且这个数字以每年9.7%的速度快速增长。痛风已成为我国仅次于糖尿病的第二大代谢性疾病,严重威胁着人们的健康。市场上常用的治疗高尿酸血症的方法主要是服用合成药物,包括别嘌呤醇、非布司他等,但这类药物长期服用会损伤肠胃、肾脏,引起红斑、光过敏等诸多不良影响。
长期高脂肪的摄入还会导致高胆固醇血症,高胆固醇血症患者通常没有明显的症状,多数情况下是在检查其它疾病或体检时才被发现,也有一些患者是在中风或心脏病发作的危险阶段才被发现。高胆固醇血症可以引起一些严重危害人体健康的疾病,主要有以下几种典型的疾病:(1)动脉粥样硬化:胆固醇的异常升高是动脉粥样硬化最主要的危险因素之一。(2)冠状动脉疾病:动脉粥样硬化是引起冠状动脉疾病的主要诱因之一,该疾病的特点是胆固醇的积累和在动脉壁形成的纤维斑块使作用于心肌供血的动脉缩小,进而限制血液的流动使满足心脏供血的氧气严重不足。(3)心肌梗死:心肌梗死是指当血液和氧气供应在一个或多个心脏动脉中部分或完全受阻,导致心脏细胞损伤或死亡的一种疾病。研究表明大约四分之一的心肌梗死患者中是高胆固醇血症患者。(4)缺血性中风:通常是由于动脉被血块或动脉粥样硬化斑块堵住,使大脑中的小血管中脱落引发的疾病。目前为止,药物治疗仍旧是治疗高胆固醇血症的主要方式。治疗过量胆固醇的药物主要为存在五种类型,他汀类、胆汁酸螯合剂、贝特类药物、烟酸和抑制胆固醇吸收的药物,其中他汀类药物是使用最为广泛和最为有效的。他汀类药物的副作用主要表现为胃肠道的不舒服、肌肉疼痛和晕眩。当同时服用两种或以上他丁类药物时,上述症状会更加严重。也有报道表明他汀类药物还会溶解横纹肌、提高血清转氨酶的含量、诱发原发性心肌病。还有临床试验表明,长期使用他汀类药物还极易诱发II型糖尿病。因此急需开发一种新的替代药物治疗的降胆固醇的方法。
发明内容
本发明的目的是提供一种凝结芽孢杆菌AP15及其在肠道调节与尿酸、胆固醇代谢的应用。我们从南极企鹅肠道中筛选得到的凝结芽孢杆菌AP15具有良好的降尿酸、降胆固醇功能,同时能够抑制肠道中有害细菌的生长,调节肠道菌群。
为了实现本发明的上述目的,采用了以下技术方案:
一种凝结芽孢杆菌AP15,该凝结芽孢杆菌保藏于中国普通微生物菌种保藏管理中心(简称:CGMCC,北京市朝阳区北辰西路1号院3号),其保藏编号为CGMCC No.26711。
上述的凝结芽孢杆菌的培养物。
上述的凝结芽孢杆菌或凝结芽孢杆菌的培养物在制备菌剂中的应用。
进一步地,上述凝结芽孢杆菌或其培养物用于调节肠道菌群和/或预防或治疗高尿酸血症、痛风和/或高胆固醇血症。
进一步地,上述凝结芽孢杆菌有效活菌剂量不低于108CFU。

术语“培养物”是指经人工接种和培养后,长有微生物群体的液体或固体产物(培养容器内的所有物质,发酵产物)的统称。即通过将微生物进行生长和/或扩增而获得的产物,其可以是微生物的生物学纯培养物,也可以含有一定量的培养基、代谢物或培养过程中产生的其他成分。

本发明所提供的凝结芽孢杆菌AP15能够显著降低降尿酸水平、胆固醇水平,显著缓解高尿酸血症、高胆固醇血症,使血尿酸水平、胆固醇水平基本恢复正常,显著减缓高尿酸引发的炎症反应,提高免疫力,缓解高尿酸血症、高胆固醇血症带来的不良影响,同时,还可使肠道微生物菌群中典型益生菌的丰度提高,改善肠道菌群结构,是一种安全、有效的益生菌。

为了更好地理解本发明,下面结合实施例和附图对本发明做进一步的详细说明,但本领域技术人员了解,下述实施例不是对本发明保护范围的限制,任何在本发明基础上做出的改变和变化,都在本发明的保护范围之内。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1降尿酸益生菌的筛选及鉴定
(一)实验材料
样品
本专利的凝结芽孢杆菌(Bacillus coagulans)筛选自南极西摩海滩企鹅肠道样品,为产乳酸芽孢杆菌属,保藏于中国微生物菌种保藏中心,保藏号为CGMCC No.26711。
培养基
尿酸固体培养基:取硫酸镁0.5g/L,氯化钠0.1g/L,三水合磷酸氢二钾0.5g/L,磷酸二氢钾0.5g/L,尿酸2g/L,琼脂20g/L,再利用10mol/L NaOH溶液调节pH到7;121℃高温高压蒸汽灭菌20min,倒板,存于4℃冰箱。
MRS培养基:取蛋白胨10g/L,牛肉膏8g/L,酵母膏4g/L,葡萄糖20g/L,磷酸氢二钾2g/L,柠檬酸三氨2g/L,三水合乙酸钠5g/L,七水合硫酸镁0.2g/L,四水合硫酸锰0.05g/L,吐温80 1g/L,再利用10mol/L NaOH溶液调节pH到6.2;121℃高温高压蒸汽灭菌20min,倒板,存于4℃冰箱中备用。
LB培养基:取酵母膏5g/L,蛋白胨10g/L,氯化钠10g/L,去离子水1000mL,若需固体培养基则再加入20g/L琼脂,再利用10mol/L NaOH溶液调节pH到7;121℃高温高压蒸汽灭菌20min,4℃冰箱中备用。
MRS-CHOL培养基:取胆固醇1.0g,吐温20g,牛胆盐3.0g水浴加热使胆固醇溶解,加入MRS培养基。
(二)菌种筛选
取3g样品于50mL塑料离心管中,加入20mL无菌水后,置于震荡仪上充分震荡。震荡完成后,使用高速冷冻离心机,于4℃,8000r/min离心5分钟后,取100μL涂布于尿酸固体培养基中,置于37℃恒温培养箱中培养24h。将长出的单菌落划线于MRS平板,37℃培养,进行多次分离纯化直至长出形态一致的单菌落,命名为AP15。
(三)菌种鉴定
16S rDNA基因序列系统进化分析
16S rDNA基因PCR扩增
以基因组DNA为模板PCR扩增16S rDNA基因,扩增引物采用27F(5'-AGAGTTTGATCCTGGCTCAG-3')(如SEQ ID NO:2)和1492R(5'-TACGACTTAACCCCAATCGC-3')(如SEQ ID NO:3)(均由南京金斯瑞生物科技有限公司合成)。PCR反应体系(20μL):引物各1μL,Prime STAR酶10μL,DNA模板8μL,ddH2O补足至20μL。PCR反应程序为:95℃预变性30s;95℃变性10s,55℃退火15s,72℃延伸90s,32个循环,最后72℃5min,扩增的PCR产物在1%琼脂糖凝胶上进行电泳检测,回收无杂带样品。
(四)序列比对与系统进化分析
将上述无杂带样品送至生工生物工程(上海)股份有限公司进行测序。将得到的测序结果的序列拼接,在NCBI数据库(https://blast.ncbi.nlm.nih.gov/Blast.cgi)中进行比对,根据细菌16S rRNA基因序列相似性,利用MEGA 7.0软件进行系统进化分析。凝结芽孢杆菌AP15的16S rDNA序列(如SEQ ID NO:1)如下:

菌株AP15的测序结果与NCBI数据库中已知标准菌株比对表明,该菌株与凝结芽孢杆菌的同源性最高,达100%,AP15菌株系统发育进化树见图1。
(五)全基因组测序与功能分析
用50mL MRS液体培养基以37℃、200rpm隔夜培养AP15菌株至OD600nm=1.0,以8000rpm离心10min,去除上清液,将菌体送上海美吉基因科技有限公司(美吉基因)进行基因组测序及分析。
实施例2体外实验的功效验证
(一)平板对峙实验
企鹅样品涂布于尿酸平板培养后的图片,如图2。如图2所示,企鹅样品中存在可降解尿酸的菌株。
从图中可以看到有明显的白色菌落,同时部分菌落旁还形成了尿酸降解圈。将上述平板中的菌落分别挑取到新的尿酸平板上,划线分离纯化得到单菌落。共得到12株可降解尿酸的菌株。图3为部分菌株在尿酸平板上的生长情况,结果显示分离出来的单株菌也存在尿酸降解能力,部分菌株的菌落旁还存在降解圈。图4为AP15菌株对三种金黄色葡萄球菌的抑菌圈实验结果,结果显示本发明AP15菌株能够抑制金黄色葡萄球菌生长。
(二)生长代谢曲线与尿酸代谢曲线
1.菌株的活化
从冰箱中找到保存的甘油管,用接种环划线到MRS平板上,置于37℃恒温培养36h至有单菌落生成;挑取单菌落接种到7mL液体MRS培养基中,置于37℃摇床培养20h至平台期。
2.接种
取高压蒸汽灭菌后的50mL离心管,每根离心管中加入14.25mL LB液体培养基,600μl 20mmol/l的尿酸标准样品,150μl隔夜培养的菌液,置于37℃摇床培养,每株菌做3个平行组。
3.菌体浓度的测定
从接种为0h开始,每隔2h取200μl菌液于96孔板中,置于酶标仪测定OD595。减去空白培养基的OD595后,按照OD595从0到1对应的菌体浓度为108CFU到109CFU计算测定时刻的菌体浓度。测定到24h时结束。
4.尿酸含量的测定
从接种为0h开始,每隔2h取100μl菌液,采用尿酸含量检测试剂盒测定菌液中的尿酸含量。测定到24h时结束,结果如图5所示。结果显示,经过24h的培养,AP15能将尿酸的浓度从800μmol/L代谢至400μmol/L以下,菌株AP15前0到4h处于快速生长期,尿酸含量先升后降,4h时降低了接近250μmol/L,降解比例达到了31.25%;6到20h属于减速期,尿酸含量回升后又稍微下降;20到24h处于稳定期,尿酸含量在此阶段从20h的560μmol/L降至400μmol/L,总体降解比例达到了50%。
(三)高剂量菌剂的4h内尿酸降解效果
1.菌种活化
从冰箱中找到保存的甘油管,用接种环划线到LB平板上,置于37℃恒温培养36h至有单菌落生成;挑取单菌落接种到7mL液体LB培养基中,置于37℃摇床培养20h至平台期。
2.接种培养
取高压蒸汽灭菌后的50mL离心管,每根离心管中加入6.4mL LB液体培养基,700μl隔夜活化的菌液,置于37℃摇床培养,每株菌做3个平行组。
3.菌体收集
隔夜培养后,从每管中取200μl菌液测定OD595,然后向每管中加入新鲜的LB培养基使OD595值稀释至0.5左右。每根管中倒出7mL于新的离心管内,置于高速冷冻离心中8000r/min,离心5分钟。离心结束后倒去上清,收集菌体;在向离心管内加入10mL无菌的PBS缓冲液,置于震荡机上震荡,使菌体分散均匀后,再次离心,8000r/min,离心5分钟,结束后倒去上清;用PBS洗涤两次后,收集下层菌体。
4.定量测定
向含有菌体的每根离心管中加入14.4mL LB液体培养基,600μl 20mmol/l的尿酸标准样品,置于37℃摇床培养,每株菌做3个平行组。
从接种为0h开始,每隔1h取200μl菌液于96孔板中,置于酶标仪测定OD595。减去空白培养基的OD595后,按照OD595从0到1对应的菌体浓度为108CFU到109CFU计算测定时刻的菌体浓度。测定到4h时结束。
从接种为0h开始,每隔1h取100μl菌液,采用尿酸含量检测试剂盒测定菌液中的尿酸含量。测定到4h时结束,结果如图6所示。由图6可得,AP15菌株的尿酸浓度从615μmol/L降低至180μmol/L,降解比例达到了70%,在1h测定时浓度下降较快,2h时尿酸浓度回升,之后一直下降。这说明提高初始接种量可以提高尿酸的降解比例,凝结芽孢杆菌AP15可以显著降低培养基中尿酸含量。
(四)胆固醇降解试验
1.胆固醇降解菌株24h生长和代谢曲线分析
取筛选出的菌的单菌落及实验室现有的菌株接种到15ml液体MRS培养基中,置于37℃摇床培养12h,将培养基全部倒入100ml MRS液体培养基中,于37℃摇床培养12h。
向灭菌后的50ml离心管中定量加入14.85ml MRS-CHOL液体培养基,150μl隔夜培养的菌液,置于37℃摇床培养。
从接种为0h开始,每隔2h取200μl菌液于96孔板中,置于酶标仪测定OD595。测定到24h时结束。
从接种为0h开始,每隔2h取10μl菌液,使用邻苯二甲醛法测定菌液中的胆固醇含量。测定到24h时结束。菌株24h内的生长和胆固醇代谢曲线如图7所示,菌AP15在0h~2h时处于迟缓期,此时菌生长较慢,胆固醇降解速率最大;2h~8h时菌进入生长期,生长速率较大,此时胆固醇降解速率较大;8h后菌生长速率与胆固醇降解速率同时开始下降,胆固醇降解率达到了77%。
2.胆固醇降解菌株大剂量接种4h生长和代谢曲线分析
取筛选出的菌的单菌落及实验室现有的菌株接种到15ml液体MRS培养基中,置于37℃摇床培养12h,将培养基全部倒入100ml MRS液体培养基中,于37℃摇床培养12h。
活化完成后,使用酶标仪测定菌液的OD595,向每管中加入MRS液体培养基培养基使OD595值稀释至0.5左右。倒出7ml于新的离心管内,置于高速冷冻离心中8000r/min,离心5分钟。离心结束后倒去上清,收集菌体;在向离心管内加入10ml无菌的PBS缓冲液,置于震荡机上震荡,使菌体分散均匀后,再次离心,8000r/min,离心5分钟,结束后倒去上清;用PBS洗涤两次后,收集下层菌体。
向含有菌体的离心管中加入15ml MRS-CHOL液体培养基,置于37℃摇床培养。
从接种为0h开始,每隔1h取200μl菌液于96孔板中,置于酶标仪测定OD595。测定到4h时结束。
从接种为0h开始,每隔1h取10μl菌液,使用邻苯二甲醛法测定菌液的胆固醇含量。测定到4h时结束。菌株4h内胆固醇代谢曲线如图8所示,高密度接种后菌株AP15后在4h时胆固醇代谢量有所提高。
实施例3小鼠动物实验
(一)小鼠养殖及分组
选用48只SPF级ICR雄性小鼠(体重大约在16-20g),每笼6只,两笼为一组,编号为A、B、C、D组,分别对应对照组、模型组、益生菌组、治疗组,具体处理如下:

每天12:00-14:00,将隔夜培养的菌液(此时已培养12-14h,根据经验值在对数生长期末期或平台期前期)测OD595,获得15mL OD值不小于1.0的菌液(适当稀释或浓缩),需结合菌种计数结果调整(按109CFU/只给药,综合菌液损失和存活率,确保成功给要在108CFU/只以上);将准备好的菌液离心,并用无菌脱脂牛奶1:1(v/v)重悬,每只小鼠灌胃300μL;将隔夜菌液以1:10(v/v)接种于15mL MRS液体培养基,37℃、200rpm摇菌。每天晚上21:00-22:00,测量中午接种的菌液的OD595,以1:100(v/v)比例接种于100mL MRS液体培养基,37℃摇菌过夜;同时,将活化菌液划线培养,验证是否污染。每隔3天,中午更换小鼠垫料,晚上收集一次小鼠粪便,并称量各组小鼠体重,记录鼠粮和饮用水消耗并补充。第二天中午和晚上观察小鼠的活动情况。
养殖15天后对每组小鼠体长及肢端情况进行测量和拍照记录,取各组小鼠眼球血、肾、肝、小肠、大肠,每组一只小鼠组织固定于4%多聚甲醛固定液中,常温固定一天以上。
(二)小鼠血清尿酸含量变化
将小鼠眼球血于4℃、3000rpm离心15分钟,吸取上清为血清。用尿酸含量检测试剂盒(Solarbio)测定不同处理组小鼠血清中尿酸浓度,对比不同组间小鼠血清中尿酸浓度差异,结果如图9所示。由图9可知,高尿酸模型组尿酸含量显著高于对照组的正常水平,而AP15益生菌组尿酸水平与对照组相近,处于正常水平;AP15治疗组与高尿酸模型组对比,尿酸含量显著下降,且低于正常水平,说明AP15菌株具有明显降低小鼠血清中尿酸浓度的功效。
实施例4小鼠肠道菌群结构的影响
刮取小鼠小肠黏膜层样品以及小鼠粪便浸出液,液氮研磨后用CTAB法提取样品中的总DNA,将其送测序公司执行高通量测序,并将测序结果与数据库进行比对,进行序列分析和物种注释,珊瑚共生菌多样性组成根据OTU注释结果(结果如图10所示)得到,并分析各组样品中菌群在纲、科、属等分类水平上的分布,获得小鼠肠道共生菌群中各菌种的丰度变化。
根据16S rDNA高通量测序获得的微生物多样性分析结果,AP15在肠道微生物中并未检出,说明两株益生菌可能不容易在小鼠肠道定殖。
组间差异显著性分析结果来看,与A组的空白相比,C组中由于AP15的作用导致益生菌组中Lachnospiraceae科与Eubacterium brachy(纤维素降解菌和产生短链脂肪酸(SCFA)的菌属)丰度的显著提高,这两种微生物都被报道与短链脂肪酸形成有关,说明凝结芽孢杆菌AP15的作用下,小鼠肠道微生物菌群中典型益生菌的丰度提高,肠道菌群结构得以改善。
实施例5小鼠代谢组分析
(一)两组间代谢差异火山图
如图11A所示,A比B的代谢产物中,下调的居多,说明高尿酸模型中尿酸的添加显著提高宿主的代谢水平。其中,尿酸含量没有显著差异。下调最大的产物为:4-O-(Indole-3-acetyl)-D-glucopyranose,lactucin(莴苣苦素),有报道称该物质具有较强的细胞毒作用,具有抗癌活性。如图11B所示,C比D的代谢产物中,下调的居多,差异最多的产物是LysoPE,说明高尿酸模型中尿酸的添加显著提高宿主的代谢水平。如图11C所示,C比A下调的居多,差异最大的代谢产物是surfactin。如图11D所示,B比D上调居多,B与D相比,上调与下调的代谢产物数量基本相当,但差异最大的三种代谢产物分别是surfactin A,surfactin和Docosapentaenoic acid(二十二碳五烯酸,DPA);上调差异最大的是Diosmetin 7-O-beta-D-glucuronopyranoside(香叶木素-7-O-葡萄糖苷,抗氧化活性)。
如图11E、图11F所示,经过T-Test统计分析,ABC三组的尿酸/尿酸盐相对丰度无显著差异,说明大部分尿酸可能经由肠道吸收进入血液,进入直肠部分的含量相对稳定;其中C组的低值较低,说明益生菌的加入可能通过尿酸代谢降低了尿酸含量在肠道中的基础值;而D组的尿酸的丰度相对降低,进一步证实了益生菌对尿酸的降解降低了肠道中的尿酸水平。如图11G所示,AP15的处理中,surfactin代谢产物较其他组显著提高,该产物可能是AP15用于调节菌群结构的主要产物之一。
(二)小鼠免疫指标
取新鲜冷冻小鼠肝、肾组织分别称重,每份样本中加入1mL无菌的新配制的PBS缓冲液,用MY-10手持式研磨仪(上海净信,中国)在冰上均匀研碎各种组织。2500rpm低速离心10min收集上清液。用检测试剂盒测定小鼠血清、肾脏、肝脏中IL-1β含量以及血清中肌酐(CRE)含量测定,结果如图12所示。
由图12可知,高尿酸引发小鼠炎症,引起分泌型免疫球蛋白sIgA和细胞因子IL-1β显著升高;而使用凝结芽孢杆菌AP15后相应指标显著下降,说明凝结芽孢杆菌AP15能显著减缓高尿酸引发的炎症反应,提高免疫力。
(三)组织切片影响
将解剖的小鼠肝、肾组织用PBS缓冲液冲洗干净表面,取肝脏0.8cm×0.8cm,肾脏取一半,使用4%的多聚甲醛溶液常温固定一天以上,将固定后的样本送至兰州大学医学院制作生理切片,并且使用HE染色。切片制作完成后,在显微镜(OLYMPUSBX53,日本)下观察不同组小鼠肝脏、肾脏的受损情况以及尿酸在肾小管处的积累情况,再加标尺拍照记录。由图13可知,对照组和益生菌组的组织差异不大,未出现严重的炎症反应和细胞破坏,说明枯草芽孢杆菌AP15不会对小鼠产生有害影响。
实施例6大鼠动物实验
选用40只雄性SD大鼠,共4组,每组10只,编号为A、B、C、D组,分别对应对照组、模型组、益生菌组、治疗组,具体处理如下:

1.大鼠尿酸水平变化情况
于给药第0、3、6、9、12、15天,中途取血制血清,用尿酸含量检测试剂盒(Solarbio)测定不同处理组大鼠血清中尿酸浓度,对比不同时间不同组间大鼠血清中尿酸浓度差异变化,结果如图14所示。如图14,高尿酸模型组的尿酸水平显著高于对照组,说明高尿酸模型建立成功。而治疗组尿酸水平与对照组和益生菌组基本一致,说明使用AP15能够显著缓解高尿酸血症,使血尿酸水平基本恢复正常水平。
2.大鼠肌酐水平变化情况
于给药第0、3、6、9、12、15天,中途取血制血清,每组5只,用检测试剂盒(南京建成,中国)测定小鼠血清中肌酐(CRE)含量测定,结果如图15所示。由图15可知,对照组和益生菌组的肌酐含量相对稳定,无太大波动,说明益生菌对大鼠肾小球的滤过作用没有产生不良影响。而高尿酸模型组和AP15治疗组的肌酐波动较大,且含量较高,说明高尿酸使大鼠的肾小管滤过作用受到较大影响,大鼠肾功能受损,而在此情况下添加AP15益生菌也难以修复高尿酸对肾脏的损伤。
3.大鼠体重、进食量、饮水量变化情况
将实验大鼠进行分组,每3天检查各组大鼠的体重、平均饮水和食物摄入量。验证菌株AP15对大鼠体重、进食和饮水方面的影响。
由图16分析可见,0-15天内,高尿酸模型大鼠的体重、进食量和饮水量均远远低于对照组,且为四组中最低;单独使用AP15菌株的大鼠体重、进食量和饮水量与对照组基本一致,说明AP15菌株不会对大鼠本身产生不良影响;而同时进行药饮和AP15灌胃的治疗组大鼠的体重、进食量和饮水量都比高尿酸模型组有显著提升,说明在有高尿酸血症的情况下,食用AP15菌株可以缓解高尿酸血症给大鼠带来的不良影响,表明AP15菌株具有较好的治疗效果。
综上,本发明所提供的凝结芽孢杆菌AP15能够显著降低降尿酸水平、胆固醇水平,显著缓解高尿酸血症、高胆固醇血症,使血尿酸水平、胆固醇水平基本恢复正常,显著减缓高尿酸引发的炎症反应,提高免疫力,缓解高尿酸血症带来的不良影响,同时,还可使肠道微生物菌群中典型益生菌的丰度提高,改善肠道菌群结构,是一种安全、有效的益生菌。
虽然以上描述了本发明的具体实施方式,但是熟悉本技术领域的技术人员应当理解,我们所描述的具体的实施例只是说明性的,而不是用于对本发明的范围的限定,熟悉本领域的技术人员在依照本发明的精神所作的等效的修饰以及变化,都应当涵盖在本发明的权利要求所保护的范围内。

1.一种凝结芽孢杆菌,其特征在于,所述凝结芽孢杆菌为凝结芽孢杆菌AP15(Bacilluscoagulans AP15),保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.26711。
2.如权利要求1所述的凝结芽孢杆菌的培养物。
3.如权利要求1所述的凝结芽孢杆菌或权利要求2所述的凝结芽孢杆菌的培养物在制备菌剂中的应用。
4.如权利要求3所述的应用,其特征在于,所述凝结芽孢杆菌或其培养物用于调节肠道菌群和/或预防或治疗高尿酸血症、痛风和/或高胆固醇血症。
5.如权利要求4所述的应用,其特征在于,所述凝结芽孢杆菌有效活菌剂量不低于108CFU。
6.一种菌剂,其特征在于,所述菌剂包括权利要求1所述的凝结芽孢杆菌或权利要求2所述的凝结芽孢杆菌的培养物。
7.如权利要求6所述的菌剂,其特征在于,所述凝结芽孢杆菌有效活菌剂量不低于108CFU。
