技术领域
本发明涉及一种利用水溶性乳香胶的用于改善黄褐斑以及老年斑等的皮肤美白用组合物。
背景技术
伴随着现代社会的生活水准的提升,人们不仅关注健康,对美容的关注度也在不断提升。其中,人们正在积极开发用于改善因为紫外线以及大气污染等造成的皮肤损伤和老化所诱发的如黄褐斑(melasma)以及雀斑(freckle)等色素沉积型皮肤疾病以及整体肤色的产品。
存在于表皮的基底层上的黑色素(melanin)是决定肤色的主要的色素,可以通过防止紫外线所造成的的损伤并去除活性氧而起到保护皮肤的作用。此外,在局部生成过量的黑色素的情况下,可能会导致如黄褐斑、雀斑、黑斑以及皮肤癌等皮肤疾病。此外,当持续且过量生成的黑色素滞留在真皮上时,会导致肤色变黑或面色发暗的问题,从而导致对美白需求的增加(Pigment Cell Res.2007Feb;20(1):2-13.;Curr Biol.2011Nov 22;21(22):1906-11.;Ann Dermatol Venereol.2012Nov;139Suppl 3:S73-7)。
黑色素是广泛存在与生物体内的分类高分子天然色素,在人体中是在存在于表皮基底层上的黑素细胞(melanocyte)内的黑色素体(melanosome)中合成并转移到周边角质细胞,从而影响人体的肤色。在所生成的黑色素过少的情况下,会诱发如白癜风等皮肤病变,而在与此相反的所生成的黑色素过多的情况下,可能会诱发包括如黑皮病(melanoderma)、炎症后黑皮病(postinflammatory melanoderma)以及日光性黑斑(solarlentigo)等后天性色素沉积过度症在内的皮肤疾病。
合成黑色素的一系列过程被统称为黑素原生成路径(melanogenesis)。黑色素的生成是以氨基酸中的一种即酪氨酸(tyrosine)作为基质,在借助于酪氨酸酶(tyrosinase)、酪氨酸酶相关蛋白-1(TRP-1,tyrosinase relatedprotein-1)以及酪氨酸酶相关蛋白-2(TRP-2,tyrosinase relatedprotein-2)经由多巴(DOPA,3,4-dihydroxyphenylalanine)转换成多巴醌(DOPA quinone)之后经过非酶促反应以及自氧化过程,接下来通过与氨基酸或蛋白的聚合反应而合成出黑色素(Korean J.FOODSCI.TECHNOL.,2000.32(3):736;Journal of Life Science,2013.23(12):1445;PigmentCell Res.1999.12(1):4-12.)。
最近除了酪氨酸酶、酪氨酸酶相关蛋白-1(TRP-1)以及酪氨酸酶相关蛋白-2(TRP-2)之外,还正在积极开展利用如小眼畸形相关转录因子(MITF,microphtalmia associatedtranscription factor)等蛋白对黑色素的合成进行抑制的研究活动。主要的细胞内信号传递路径为环磷酸腺苷/蛋白激酶A(cAMP/PKA,cyclic monophosphate/protein kinaseA)路径,其中环磷酸腺苷(cAMP)是一种经由蛋白激酶A(PKA)以及环磷酸腺苷反应元件结合蛋白1(CREB1,cAMP responsive element binding protein 1)促进小眼畸形相关转录因子(MITF)的表达,而小眼畸形相关转录因子(MITF)是黑色素合成过程中的重要的转录调节因子,可以促进酪氨酸酶、酪氨酸酶相关蛋白-1(TRP-1)以及酪氨酸酶相关蛋白-2(TRP-2)的转录(Pharmazie.2015Oct;70(10):646-9.;Int J Mol Sci.2015Oct 13;16(10):24219-42;Curr Pharm Biotechnol.2015;16(12):1111-9.)。
因为在生成黑色素的第一个步骤中酪氨酸酶会起到最为重要的作用,因此对美白剂的研究方向主要是集中在为了对酪氨酸酶的活性进行调节而开发出酪氨酸酶活性阻碍物质或对酪氨酸酶的基质的拮抗物质(Kor.J.Pharmacogn.,44(3):220,2013)。
目前作为美白剂使用最为广泛的物质包括熊果苷(arbutin)以及曲酸(kojicacid)。熊果苷是酪氨酸的拮抗剂,而曲酸可以通过对酪氨酸酶的活性部位进行鳌合而对从酪氨酸到多巴以及多巴醌的过程进行抑制(Korean J.Plant Res.,26(5):526,2013;J.Korean Soc.Food Sci.Nutr.,41(2):156,2012)。虽然已经得知熊果苷以及曲酸具有强力的美白效果,但是因为其具有如皮肤刺激等副作用,因此正在积极开展从副作用相对较少的天然物质寻找具有美白活性的物质的研究活动。
除此之外,收敛作用是可以防止皮肤老化并维持皮肤健康的功能中的一种。收敛作用是指皮肤蛋白与高分子类黄酮形成交联而使得皮肤收缩的现象。收敛作用可以起到使得如皮肤以及粘膜血管等收缩、对细胞间隙以及淋巴间隙的黏液分泌进行抑制、通过在皮肤以及粘膜的表面形成难溶性薄膜而为局部提供保护以及通过使得组织变得致密而维持弹性的作用,还可以起到降低细胞膜透过性的作用。如上所述,收敛效果可以通过收缩过度扩张的毛孔或汗孔而为皮肤赋予弹性以及柔韧性。因为具有收敛活性的物质同时具有与蛋白结合的性质,因为通常可以通过对与血红蛋白(hemoglobin)结合的程度进行测定而判断收敛效果的程度(Journal of the Korean Society of Fashion&Beauty 2007,5(3):99-104)。
本发明公开一种水溶性乳香胶的皮肤美白活性以及皮肤收敛活性。
发明内容
本发明的目的在于提供一种利用水溶性乳香胶的皮肤美白用组合物。
此外,本发明的目的在于提供一种利用水溶性乳香胶的皮肤收敛改善用组合物。
本发明的其他目的或具体目的将在下述内容中进行公开。
本发明如下述实施例以及试验例所示,是通过确认水溶性乳香胶不会对人类成纤维细胞即CCD-986sk细胞呈现出特殊的细胞毒性、可以在人类成纤维细胞即CCD-986sk中抑制酪氨酸酶基因的表达并与血红蛋白具有较高的结合力而完成。
考虑到如上所述的情况,本发明的一方面涉及一种作为有效成分包含水溶性乳香胶的皮肤美白用组合物,而另一方面涉及一种作为有效成分包含水溶性乳香胶的皮肤收敛改善用组合物。
在本说明书中,“水溶性乳香胶”是指通过对乳香胶进行微粒(平均粒径在1~999μm的范围内)至纳米粒子(平均粒径为1~999nm)化并悬浮到作为溶剂的水中的悬浮液(suspension)(即在作为溶剂的水中不会发生凝聚·沉淀而维持分散状态)。如上所述的水溶性乳香胶可以通过在对乳香胶进行粉碎而进行微粒化乃至纳米粒子化之后再向其中添加水并搅拌混合的方式获得。尤其是,通过在对乳香胶进行粉碎而进行微粒化之后再向其中添加水并混合,接下来再次对其混合物(或混合液)进行粉碎而对所述混合物中的乳香胶的粒子进行纳米粒子化的方式获得为宜。此时,为了通过使得分散的粒子稳定化而防止其发生凝聚·沉淀,可以在对微粒化乃至纳米粒子化的乳香胶与水进行混合前后以及粉碎前后添加适量的悬浮剂(suspending agents)。作为悬浮剂可以使用本行业所公知的任意的悬浮剂,例如可以使用如阿拉伯树胶(arbic gum)、黄芪胶(tragacanth gum)、琼脂(agar)、刺梧桐树胶(karaya gum)、刺槐豆胶(locust bean gum)、瓜尔豆胶(guar gum)、黄原胶(xanthan gum)、印度树胶(ghatti gum)以及果胶(pectin)等水溶性聚合物,环糊精(cyclodextrin)以及甘油或其脂肪酸酯衍生物,如甲基纤维素或羟甲基纤维素等改性纤维素(modified cellulose)以及各种表面活性剂等。如上所述的悬浮剂可以单独使用或对两种以上进行混合使用,而且可以在适合于悬浮的适量范围内,具体来讲可以在与100重量份的乳香胶相比的20至200重量份的范围内使用。与水溶性乳香胶的制造相关的更具体的内容,可以参阅下述实施例或参阅在发明名称为“通过纳米粒子化提升分散性以及水溶性的乳香胶水溶液的制造方法”的韩国注册专利第10-1865712号中公开的方法等。其中,乳香胶是指从乳香黄连木(mastic tree,Pistacia lentiscus)获得的树脂(resin)。
此外,本说明书中的“有效成分”是指可单独呈现出目标活性,或者与其自身没有活性的载体一起呈现出活性的成分。
本发明的组合物可以根据产品以及剂型等在可呈现出皮肤美白活性以及皮肤收敛活性等的范围内包含任意量(有效量)的有效成分,通常来讲在以组合物的整体重量为基准时,可以在0.001重量%至15重量%的范围内决定其有效量。其中,“有效量”是指对于其适用对象即哺乳动物,较佳地对于人体在医疗专家以及皮肤专家等所给出的给药期间内利用本发明的组合物进行给药时,可以呈现出如皮肤美白效果以及皮肤收敛效果等所预期的医疗·皮肤效果的包含于本发明的组合物中的有效成分的量。所述有效量可以在相关从业人员的一般的能力范围内通过试验决定。
本发明的组合物除了有效成分之外,可以为了提升·补强皮肤美白效果以及皮肤收敛改善效果,或为了通过附加如皮肤皱纹改善活性、皮肤过敏反应抑制活性以及皮肤保护活性(如因为紫外线而导致的皮肤损伤抑制以及皮肤保湿等)等类似活性而提升服用、摄入或使用的便利性,追加包含在本行业中已经对其安全性进行验证且已知具有相应活性的公知的任意化合物或天然提取物。
如上所述的化合物或提取物,包括各个国家的药典(韩国为“大韩民国药典”)、各个国家的健康功能食品公典(韩国为食药处公布的“健康功能食品标准及规格”)以及各个国家的功能性化妆品公典(韩国为食药处公布的“功能性化妆品标准及试验方法”)等法定文件中记载的化合物或提取物、根据用于对医药品的制造·销售做出规定的各个国家的法律(韩国为“药事法”)获得项目许可的化合物或提取物、根据用于对健康功能食品的制造·销售做出规定的各个国家的法律(韩国为“与健康功能食品相关的法律”)认定其功能性的化合物或提取物以及根据用于对功能性化妆品的制造·销售做出规定的各个国家的法律(韩国为“化妆品法”)认定其功能性的化合物或提取物。
作为如上所述的成分的实例,可以以在根据韩国“化妆品法”的化妆品公典(韩国食药处公告的“功能性化妆品标准及试验方法”)中认定为皮肤皱纹改善成分的如视黄醇、棕榈酸视黄酯、腺苷以及聚氧乙烯醚酰胺等;在相同的韩国化妆品公典中认定为紫外线保护成分的如甲酚曲唑、甲酚曲唑三硅氧烷、二没食子酰三油酸酯、亚苄基丙二酸盐聚硅氧烷以及二乙基己基丁酰胺基三嗪酮等;在相同的化妆品公典中认定为皮肤美白成分的熊果苷、烟酰胺、抗坏血酸葡萄糖苷、α-红没药醇以及油溶性甘草提取物(Oil SolubleLicorice(Glycyrrhiza)Extract)等;根据韩国的“与健康功能食品相关的法律”认定为紫外线保护成分的如松树皮提取物等复合物、磷脂酰丝氨酸、凹唇姜提取粉末以及红参与山茱萸等的复合提取物等;根据韩国的“与健康功能食品相关的法律”的绿茶提取物、竹叶提取物、甜瓜提取物、覆盆子提取物、蜂蜡醇、泛醇、辅酶Q10、番茄提取物、葡萄籽提取物、法国海岸松树皮提取物以及红参浓缩液等;根据韩国的“与健康功能食品相关的法律”认定为过敏皮肤状态改善功能性成分的沙克乳酸杆菌益生菌-65(L.sakei Probio 65)、含γ-亚麻酸的油脂、源自于果蔬的乳酸菌即植物乳杆菌CJLP133(L.plantarum CJLP133)以及益生菌ATP等为例。如上所述的化合物或天然提取物,可以在本发明的组合物中与其有效成分一起包含一种以上。
本发明的皮肤组合物在其具体的形态上可以是化妆品组合物。
在本发明的组合物为化妆品组合物时,所述化妆品组合物可以在其用途层面以及法律层面被归类为任意产品,具体来讲可以是具有如皮肤美白等用途的功能性化妆品以及非功能性一般化妆品等。在产品形态层面也可以被归类为任意产品形态,具体来讲可以被制剂化成如溶液、悬浮液、乳浊液、糊剂、凝胶、霜剂、乳液、粉剂、香皂、含表面活性剂的洁面剂、油剂、粉底霜、乳浊液粉底霜、蜡粉底霜以及喷雾剂等。具体的产品形态,可以是如柔肤水、滋养液、滋养霜、按摩霜、精华液、眼霜、洁面霜、洁面泡沫、洁面水、面膜、喷雾或粉底等剂型。
本发明的化妆品组合物除了其有效成分之外,还可以包含在化妆品组合物中通常使用的成分,如稳定剂、增溶剂、表面活性剂、维生素、色素以及香料等通常的佐剂以及载体。
在本发明的剂型为糊剂、霜剂或凝胶的情况下,作为载体成分可以使用如动物油、植物油、蜡、石蜡、淀粉、黄蓍胶、纤维素衍生物、聚乙二醇、有机硅、膨润土、二氧化硅、滑石或氧化锌等。
在本发明的剂型为粉剂或喷雾剂的情况下,作为载体成分可以使用如乳糖、滑石、二氧化硅、氢氧化铝、硅酸钙或聚酰胺粉末等,尤其是在喷雾剂的情况下,还可以追加包含如氯氟烃、丙烷/丁烷或二甲醚等推进剂。
在本发明的剂型为溶液或乳浊液的情况下,作为载体成分可以使用如溶剂、溶剂化剂或乳浊化剂,例如可以使用如水、乙醇、异丙醇、碳酸乙酯、乙酸乙酯、苯甲醇、苯甲酸苄酯、丙二醇、1,3-丁基乙二醇油、甘油脂肪族酯、聚乙二醇或山梨糖醇酐的脂肪酸酯等。
在本发明的剂型为悬浊液的情况下,作为载体成分可以使用如水、乙醇或丙二醇等液态稀释剂;如乙氧基化异硬脂醇、聚氧乙烯山梨糖醇酯以及聚氧乙烯山梨糖醇酐酯等悬浊剂;微晶纤维素、偏氢氧化铝、膨润土或黄蓍胶等。
在本发明的剂型为含表面活性剂的洁面剂的情况下,作为载体成分可以使用如脂肪醇硫酸盐、脂肪醇醚硫酸盐、磺基琥珀酸单酯、羟乙基磺酸盐、咪唑啉衍生物、牛磺酸甲酯、肌氨酸盐、脂肪酸酰胺醚硫酸盐、烷基酰胺甜菜碱、脂肪醇、脂肪酸甘油酯、脂肪酸二乙醇酰胺、植物油、亚油酸衍生物或乙氧基化甘油脂肪酸酯等。
本发明的化妆品组合物除了包含呈现出皮肤美白活性的有效成分之外,可以按照本行业通常使用的化妆品组合物的制造方法进行制造。
本发明的组合物在其具体的形态上可以是食品组合物。
本发明的食品组合物可以制造成任意形态,例如可以制造成如茶、果汁、碳酸饮料以及离子饮料等饮料类;如牛奶以及酸奶等加工乳品类;如口香糖、打糕、韩国传统糕点、面包、饼干以及面条等食品类;如片剂、胶囊、丸剂、颗粒剂、液剂、粉剂、片剂、糊剂、糖浆剂、凝胶剂、果冻剂以及棒剂等健康功能食品制剂类等。
此外,本发明的食品组合物在法律层面·功能层面的分类上可以归类为符合制造·流通时的实施法规的任意的产品分类。例如可以是根据韩国“与健康功能食品相关的法律”的健康功能食品、根据韩国“食品卫生法”的食品公典(食药处公告的“食品标准及规格”)上的各个食品类型的饼干类、豆类、茶类、饮料类以及特殊用途食品等。
在本发明的食品组合物中,除其有效成分之外还可以包含食品添加剂。食品添加剂可以理解为是在通常对食品进行制造、加工或保存时添加到食品中进行混合或浸润的物质,因为会与食品一起天天且长期摄入,因此需要保障其安全性。在根据用于对食品的制造·流通做出规定的各个国家的法律(韩国为“食品安全法”)的食品添加剂公典中,在成分层面或从能层面上对可保障其安全性的食品添加剂做出了限定性的规定。在韩国食品添加剂公典(食药处公告的“食品添加剂标准及规格”)中,将食品添加剂从成分层面上区分规定为化学合成品、天然添加剂以及混合制剂类,而如上所述的食品添加剂在功能层面上区分为甜味剂、调味剂、防腐剂、乳化剂、酸味剂以及增稠剂等。
甜味剂用于为食品赋予适当的甜味,在本发明的组合物中可以使用天然或合成的甜味剂。较佳地使用天然甜味剂为宜,而天然甜味剂可以以如固态玉米糖浆、蜂蜜、蔗糖、果糖、乳糖以及麦芽糖等糖类甜味剂为例。
调味剂可以为了提升香味或香气而使用,可以使用天然或合成的调味剂。较佳地使用天然调味剂为宜。在使用天然调味剂的情况下,还可以同时达成强化营养的目的。天然调味剂可以从如苹果、柠檬、柑橘、葡萄、草莓以及桃子等中获得,也可以从如绿茶叶、玉竹、竹叶、桂皮、菊花叶以及茉莉等中获得。此外,也可以使用从人参(红参)、竹笋、芦荟以及银杏等中获得的调味剂。天然调味剂可以是液态的浓缩液或固态的提取物。在一部分情况下可以使用合成调味剂,而作为合成调味剂可以使用如酯类、醇类、醛类以及萜类等。
作为防腐剂可以使用如山梨酸钙、山梨酸钠、山梨酸钾、苯甲酸钙、苯甲酸钠、苯甲酸钾以及乙二胺四乙酸(EDTA)等,作为乳化剂可以使用如阿拉伯树胶、羧甲基纤维素、黄原胶以及果胶等,而作为酸味剂可以使用如柠檬酸、苹果酸、富马酸、己二酸、磷酸、葡萄糖酸、酒石酸、抗坏血酸以及醋酸等。酸味剂除了改善味道的目的之外,还可以作为抑制微生物增殖的目的添加到食品组合物中而调节为适当的酸度。
作为增稠剂,可以使用悬浊剂、沉降剂、凝胶形成剂以及膨化剂等。
本发明的食品组合物除了如上所述的食品添加剂之外,还可以以补充·强化功能性以及营养性的目的包含本行业所公知且作为食品添加剂的稳定性得到保障的生理活性物质或矿物质类。
作为如上所述的生理活性物质,可以以包含于绿茶等中的儿茶素类、维生素B1、维生素C、维生素E以及维生素B12等维生素类和生育酚以及二苯甲酰硫胺等为例,而作为矿物质类,可以以如柠檬酸钙等钙制剂、如硬脂酸镁等镁制剂、如柠檬酸铁等铁制剂、氯化铬、碘化钾、硒、锗、钒以及锌等为例。
在本发明的食品组合物中,可以根据产品类型包含可达成其添加目的的适量的如上所述的食品添加剂。
关于可以包含于本发明的食品组合物中的其他食品添加剂,可以参阅各个国家的食品公典或食品添加剂公典。
本发明的皮肤美白用组合物在其具体的形态上可以是药剂学组合物。
在本发明的皮肤美白用组合物为药剂学组合物时,其目标疾病可以是皮肤色素沉积过度症,而作为如上所述的色素沉积过度症的具体实例,可以包含如雀斑、老年斑、肝斑、黄褐斑、褐斑或黑斑、日光性色素斑、蓝色黑皮病、使用如钙拮抗剂等药物之后的色素沉积过度、因为组织硬化疗法而导致的色素沉积过度后遗症、怀孕性褐斑、服用口服避孕药的女性的黑皮病、因为包括擦伤以及烫伤在内的伤口或皮肤炎而导致的炎症后色素沉积过度、光毒性反应或其他类似的小型固定色素性病变等,但是并不限定于此。
本发明的药剂学组合物除了有效成分之外还可以包含药剂学上容许的载体,并通过本行业所公知的通常的方法,根据给药路径制造成口服剂型或非口服剂型。其中,给药路径可以是包括局部路径、口服路径、静脉内路径、肌内路径以及通过粘膜组织的直接吸收在内的任意的适当的路径,也可以组合使用两种以上的路径。作为两种以上的路径组合的实例,对两种以上的剂型的药物浸渍那个组合的情况包括如第一次通过静脉内路径对某一种药物进行给药,而第二次通过局部路径对其他药物进行给药的情况。
药剂学上容许的载体,根据给药路径或剂型属于本行业所公知的内容,具体来讲可以参阅包含“大韩民国药典”在内的各个国家的药典。
在本发明的药剂学组合物被制造成口服剂型时,可以与适当的载体一起通过本行业公知的方法制造成如粉剂、颗粒剂、片剂、丸剂、糖片剂、胶囊剂、液剂、凝胶剂、糖浆剂、悬浮剂以及薄片剂等。此时作为适当的载体,可以以如乳糖、葡萄糖、蔗糖、葡萄糖、山梨糖醇、甘露糖醇以及木糖醇等糖类;如玉米淀粉、土豆淀粉以及小麦淀粉等淀粉类;如纤维素、甲基纤维素、乙基纤维素、羧甲基纤维素钠、羟丙基甲基纤维素等纤维素类;聚乙烯吡咯烷酮、水、羟基苯甲酸甲酯、羟基苯甲酸丙酯、硬脂酸镁、矿物油、麦芽、明胶、滑石粉、多元醇、植物油、乙醇以及甘油等为例。在进行制剂化时,可以根据需要包含适当的结合剂、润滑剂、崩解剂、着色剂以及稀释剂等。作为适当的结合剂可以以如淀粉、硅酸镁铝、淀粉铁酸盐、明胶、甲基纤维素、羧甲基纤维素钠、聚乙烯吡咯烷酮、葡萄糖、玉米甜味剂、海藻酸钠、聚乙二醇以及蜡等为例,作为润滑剂可以以如油酸钠、硬脂酸钠、硬脂酸镁、苯甲酸钠、醋酸钠、氯化钠、二氧化硅、滑石、硬脂酸及其镁盐和钙盐以及聚乙二醇等为例,而作为崩解剂可以以如淀粉、甲基纤维素、琼脂(agar)、膨润土、黄原胶、淀粉、海藻酸或其钠盐等为例。此外,作为稀释剂可以以如乳糖、葡萄糖、蔗糖、甘露醇、山梨糖醇、纤维素以及甘氨酸等为例。
在将本发明的药剂学组合物制造成非口服剂型时,可以与适当的载体一起通过本行业公知的方法制剂化成如注射剂、经皮给药剂、鼻腔吸入剂以及栓剂等形态。在制剂化成注射剂时,作为适当的载体可以使用如等渗水溶液或悬浊液,具体来讲可以使用如含有三乙醇胺的磷酸盐缓冲盐液(PBS,phosphate buffered saline)或注射用灭菌水、5%葡萄糖等等渗溶液。在制剂化成经皮给药剂时,可以制剂化成如软膏剂、霜剂、乳液剂、凝胶剂、外用液剂、糊剂、搽剂以及气雾剂等形态。在制剂化成鼻腔吸入剂时,可以使用如二氯氟甲烷、三氯氟甲烷、二氯四氟乙烷以及二氧化碳等适当的推进剂制剂化成气溶胶喷雾的形态,而在制剂化成栓剂时,作为其载体可以使用如Witepsol、Tween 61、聚乙二醇、可可脂、月桂酸脂、聚氧乙烯脱水山梨醇脂肪酸酯类、聚氧乙烯硬脂酸酯类以及脱水山梨糖醇脂肪酸酯等。
关于药剂学组合物的具体的制剂化,属于本行业所公知的内容,例如可以参阅文献[Remington's Pharmaceutical Sciences(19thed.,1995)]等。所述文献被视为本说明书的一部分。
本发明的药剂学组合物的较佳的给药量,根据患者的状态、体重、性别、年龄、患者的严重程度以及给药路径,可以是在每天0.001mg/kg~10g/kg的范围,较佳地可以是在0.001mg/kg~1g/kg的范围。给药可以每天一次或多次执行。所述给药量在任何方面都不应该解释为是为了对本发明的范围做出限定。
如上所述,根据本发明可以提供一种利用水溶性乳香胶的皮肤美白以及皮肤收敛改善用组合物。
本发明的组合物可以被产品化成如化妆品、食品以及药品等。

具体实施方式
在接下来的内容中,将参阅本发明实施例进行说明。但是,本发明的范围并不因为如上所述的实施例而受到限定。
<实施例>水溶性乳香胶的皮肤美白活性以及皮肤收敛活性评估
<实施例1>水溶性乳香胶的制备
通过对乳香胶粉末进行肉眼检查而对颜色进行确认并检查是否有异物。接下来,投入所筛选出的乳香胶粉末20重量%、纯净水75.7重量%、4重量%的环糊精以及添加剂(印度树胶)0.3重量%,并使用均质混合器(KNS Company株式会社)在30~50℃下以2,000~3,000rpm进行30~60分钟的分散,从而使其粒子大小达到约20~50μm。使用珠磨机(DNTECH株式会社)在20~40℃下对所述乳香胶的分散液进行3~6小时的研磨,从而使其最终分散粒子达到平均600~1,000nm。接下来,通过利用40~60μm的外壳过滤器进行过滤而获得悬浊液状态的乳香胶水溶液。在接下来的试验中,直接使用了所述水溶性乳香胶。
<实施例2>细胞毒性试验
首先,购买了由ATCC(Manassas,VA,USA)进行继代培养并冷冻在冷冻管(cryovial)中的人类成纤维细胞即CCD-986sk。在对其进行加热融化之后,在投入青霉素(100U/ml)、链霉素(100ug/ml)以及10%的热灭活血清的达尔伯克改良伊格尔培养基(DMEM,Dulbecco's Modified Eagle's Medium)中以95%的空气以及5%的CO2和37℃的条件进行培养。在CCD-986sk细胞被附着在培养皿中且细胞生长之后,利用0.25%的胰蛋白酶/10mM的乙二胺四乙酸(EDTA)收集细胞,并在按照1:3的比例分配之后维持在10%的达尔伯克改良伊格尔培养基(DMEM)中对细胞进行培养。
在试验中所使用的试剂即CCK-8是从(株)Dongin Biotech(Seoul,Korea)购买并使用。在将CCD-986sk细胞在96孔板(well plates)中以1×103cells/well的浓度分别利用100μl进行处理之后培养24小时。在细胞附着在底部之后,利用最多2mg/mL的实施例样本进行24小时的处理。接下来以10μl的CCK-8试剂以及100μl的达尔伯克改良伊格尔培养基(DMEM)的条件下培养4小时之后,在540nm下利用分光光度计(spectrophotometer)对其吸光度进行测定。
其结果如图1所示,可以确认实施例的样本直至0.5mg/ml并没有呈现出毒性,表明其可以安全使用。
<实施例3>酪氨酸酶基因表达测定
按照与所述实施例2相同的方法,利用0.1mg/ml的实施例的样本以及作为阳性对照组的抗坏血酸(Ascorbic acid;AA)对人类成纤维细胞进行处理之后,利用Appliedbiosystems公司的“QuantStudio 3”,按照制造企业的标准通过实时聚合酶链反应(PCR)对已知的黑色素调节基因即酪氨酸酶(Tyrosinase;TYR)的表达量进行了确认。此时,以不同条件下的三次独立试验结果为基础,记录了平均±标准偏差数据。酪氨酸酶(TYR)引物序列如下述表1所示。

其结果如图2所示,可以确认直至利用0.3mg/ml的实施例的样本对CCD-986sk细胞进行处理,与阳性对照组相比可以明显阻碍酪氨酸酶(TYR)的表达。
<试验例2>皮肤收敛活性试验
按照不同的浓度分别制造出500μl的实施例的样本,并在与磷酸盐缓冲液(PBS)混合的0.1%血红蛋白溶液等量混合之后进行1分钟的涡旋混合(Voltexing),然后在室温下放置5分钟。接下来,在96孔板(well plates)分注100μl,并在405nm下利用分光光度计(spectrophotometer)对其吸光度进行测定。
其结果如图3所示,实施例样本的皮肤收敛活性以随着浓度增加。

1.一种皮肤美白用组合物,作为有效成分包含水溶性乳香胶。
2.一种皮肤收敛改善用组合物,作为有效成分包含水溶性乳香胶。
3.根据权利要求2所述的组合物,其特征在于:
所述水溶性乳香胶是通过对乳香胶进行微粒(平均粒径在1~999μm的范围内)至纳米粒子(平均粒径为1~999nm)化并悬浮到作为溶剂的水中的悬浮液。
4.根据权利要求2或权利要求3所述的皮肤美白用组合物,其特征在于:
