Abstract
本发明公开了CIRP作为靶点在制备防治表皮异常增殖性皮肤病的药物中的应用,涉及生物医药技术领域,通过研究发现CIRP与表皮异常增殖性皮肤病如皮肤鳞癌的发生发展呈明显正相关,使用CIRP特异性siRNA或shRNA敲低皮肤鳞癌细胞系A431和SCL‑1中CIRP的表达,可抑制两种皮肤鳞癌细胞的增殖能力,并抑制ERK1/2信号通路的激活;通过构建皮肤鳞癌细胞裸鼠皮下移植瘤模型,发现使用CIRP shRNA敲低皮肤鳞癌细胞中CIRP的表达后,皮肤鳞癌细胞移植瘤的生长速度被明显抑制,并且移植瘤组织中Ki‑67和p‑ERK1/2蛋白的表达水平也显著降低。本发明针对siRNA或shRNA敲低CIRP的载体相比于其他治疗药物,干预靶点更为明确,除直接对异常增殖的细胞进行抑制,还可针对表皮异常增殖性皮肤病中ERK1/2信号通路起到治疗作用。

技术领域
本发明涉及生物医药技术领域,具体涉及CIRP作为靶点在制备防治表皮异常增殖性皮肤病的药物中的应用。
背景技术
表皮异常增殖性皮肤病,包括良性增殖性疾病如银屑病、恶性增殖性疾病如皮肤鳞状细胞癌(简称皮肤鳞癌),其发病机理无论是从病原学,免疫学,遗传学以及环境因子(诸如感染、外伤、压力等)的角度来说目前尚未清晰。其中,皮肤鳞癌是一种发生于表皮或附属器角质形成细胞的恶性增殖性皮肤病,是最常见的皮肤恶性肿瘤之一,其病因和发病机制尚未明晰、恶性程度较高,具有侵袭性生长和远处转移等特征。手术为目前皮肤鳞癌的主要治疗手段,但由于手术会影响功能和美观,其应用尚有限制,并且部分患者术后亦可局部复发。此外,目前针对皮肤鳞癌的靶向及免疫治疗药物价格昂贵,可能的副作用有待确定。因此,皮肤鳞癌治疗非常棘手,进一步寻找疗效更好、安全性更高的治疗靶点,是突破该病治疗瓶颈的关键所在。
冷诱导RNA结合蛋白(cold-inducible RNA-binding protein,CIRP)是富含甘氨酸的RNA结合蛋白家族的成员,最早是Nishiyama等(Nishiyama H, Itoh K, Kaneko Y, etal. A glycine-rich RNA-binding protein mediating cold-inducible suppressionof mammalian cell growth [J]. J Cell Biol, 1997, 137(4): 899-908.)于20世纪90年代,首次从小鼠睾丸的cDNA文库中分离得到,其位于第19号染色体p13.3位点。CIRP蛋白由172个氨基酸残基组成,分子量为18 kDa,氨基端含有在不同物种中高度保守的RNA识别基序,包含两个核糖核蛋白结构域,羧基端含有富含精氨酸和甘氨酸的结构域。CIRP作为一种RNA结合蛋白,广泛参与细胞的多种生理和病理过程,主要包括抑制细胞凋亡、发挥促炎效应、参与端粒酶活性调控及生物生殖发育和昼夜节律调节等。近年来,研究报道CIRP在多种急慢性炎症性疾病及多种肿瘤中发挥重要作用,但在异常增殖性皮肤病治疗方面尚未见报道。因此,实现CIRP作为药物靶点防治异常增殖性皮肤病是本领域技术人员亟需解决的技术问题。
发明内容
本发明针对上述问题,提供了CIRP作为靶点在制备防治表皮异常增殖性皮肤病的药物中的应用。解决了现有技术中存在的问题,实现了CIRP作为药物靶点实现表皮异常增殖性皮肤病的有效防治。
为了实现上述目的,本发明所采用的技术方案如下:

与现有技术相比,本发明的有益效果:
本发明通过研究发现CIRP与表皮异常增殖性皮肤病如皮肤鳞癌的发生发展呈明显相关,使用CIRP特异性siRNA或shRNA敲低皮肤鳞癌细胞系A431和SCL-1中CIRP的表达,可抑制两种皮肤鳞癌细胞的增殖能力,并抑制ERK1/2信号通路的激活;通过构建皮肤鳞癌细胞裸鼠皮下移植瘤模型,发现使用CIRP shRNA敲低皮肤鳞癌细胞中CIRP的表达后,皮肤鳞癌细胞移植瘤的生长速度被明显抑制,并且移植瘤组织中Ki-67和p-ERK1/2蛋白的表达水平也显著降低。因此,本发明针对siRNA或shRNA敲低CIRP的载体相比于其他治疗药物,干预靶点更为明确,除直接对异常增殖的细胞进行抑制,还可针对表皮异常增殖性皮肤病中ERK1/2信号通路起到治疗作用。

具体实施方式
为了使本发明的目的及优点更加清楚明白,以下结合实施例对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明。
实施例1
本实施例发现在皮肤鳞癌患者癌组织中CIRP表达升高,在皮肤鳞癌细胞系中CIRP表达较角质形成细胞系中升高;这些提示CIRP与恶性表皮增殖性皮肤病皮肤鳞癌的发展呈明显正相关,下面对本发明进一步详细地说明。
1.皮肤鳞癌组织和正常皮肤组织的免疫组织化学染色
具体方法如下:
(1)组织固定及石蜡切片制作
a)获取新鲜的组织样本后,用生理盐水彻底冲洗,去除血液和污染物,随后将组织块置于4%多聚甲醛中,固定24 h。
b)将组织从固定液中取出,在通风橱内将目的部位组织修剪平整,将修剪好的组织依次置入连续梯度的乙醇中进行脱水。具体浓度及放置时间如下:75%酒精4 h,85%酒精2h,90%酒精2 h,95%酒精1 h,无水乙醇(I)30 min,无水乙醇(II)30 min。
c)使用二甲苯透明组织,将组织置于二甲苯(I)和二甲苯(II)中各5-10 min。
d)将透明完全的组织依次置于液体石蜡中进行浸蜡,具体步骤为:65℃熔化石蜡(I)1 h,65℃熔化石蜡(II)1 h,65℃熔化石蜡(III)1 h。
e)将浸好蜡的组织于包埋机内进行包埋。先将熔化的石蜡加入包埋框中,待石蜡凝固之前将组织按照包埋面的要求放入包埋框内并进行标记。待蜡凝固后将蜡块从包埋框中取出并修整蜡块。
f)将修整好的蜡块,置于石蜡切片机中进行切片,厚4 μm。使用镊子将切好的切片放入摊片机40℃温水中使其完全伸展,将切片贴附于载玻片上,60℃烤片。
(2)IHC染色步骤
a)石蜡切片脱蜡:将制备好的石蜡切片依次置于二甲苯(I)和二甲苯(II)中各浸泡20 min,后依次置于无水乙醇(I)、无水乙醇(II)、95%酒精、80%酒精和60%酒精中各5min,最后在去离子水中浸洗1 min×3次。
b)抗原修复:将1×EDTA抗原修复液加入耐高温抗原修复盒中,在微波炉内用大火加热至沸腾,然后将切片放入其中,保证液体完全浸没,用保鲜膜将抗原修复盒包裹完全并用牙签扎孔,中火煮沸15 min后取出抗原修复盒,去除保鲜膜,室温冷却40 min后,使用去离子水浸洗1 min×3次。需注意在修复及后续滴加液体的各个步骤中,必须防止干片,以免造成假阴性和背景染色。
c)阻断内源性过氧化物酶:使用滤纸将组织周围的多余水分完全擦干,用组化笔在组织周围画圈,滴加新鲜配制的3%过氧化氢溶液均匀覆盖组织,置于湿盒中室温孵育10min,以阻断内源性过氧化物酶,然后用去离子水浸洗1 min×3次。
d)封闭:将切片甩干,置于湿盒内,滴加3% BSA封闭液均匀覆盖组织,室温封闭1h。
e)孵育一抗:轻轻甩除封闭液,滴加一定比例的用免疫染色一抗稀释液稀释好的一抗工作液覆盖组织,将切片置于湿盒中4℃孵育过夜。
f)孵育二抗:将切片置于TBST中洗涤5次,每次1 min。切片稍甩干后在圈内滴加用3% BSA稀释好的HRP标记的羊抗兔二抗覆盖组织,置于湿盒中室温孵育30 min。
g)DAB显色:切片置于TBS中洗涤5次,每次1 min。切片稍甩干后在圈内滴加新鲜配制的DAB显色液(A液:B液=20:1,现配现用),显微镜下观察显色反应,显色2 min后,用自来水终止反应。
h)复染细胞核:用苏木素复染2 min,再置于1×TBS溶液中10 min进行返蓝。
i)脱水封片:将切片依次置于60%酒精、80%酒精、95%酒精、无水乙醇(I)、无水乙醇(II)、二甲苯(I)和二甲苯(II)中各5 min使组织脱水透明,将切片从二甲苯中取出,稍晾干后滴加一滴中性树胶,盖上盖玻片,该过程需尽量防止气泡产生,使封片完全。
(3)IHC染色切片镜检
使用正置光学显微镜对切片进行观察和图像采集。
(4)IHC染色结果的判定
所有切片由3名病理医师根据染色强度及阳性细胞数量进行评估,每张切片随机选取5个高倍视野(10×40倍),评估染色强度并计算阳性细胞百分率。染色强度可分为4级,无着色为0分,淡黄色为1分,棕黄色为2分,棕褐色为3分。阳性细胞表现为细胞核和(或)细胞质呈淡黄色、棕黄色或棕褐色染色。阳性细胞百分率≤5%为0分,6%-25%为1分,26%-50%为2分,51%-75%为3分,>75%为4分。每个视野染色强度评分与阳性细胞百分率评分相乘则为该视野总评分,最终得分为5个视野得分的平均值。0分为阴性,1-4分为弱阳性,5-8分为中度阳性,9-12分为强阳性。以此作为判断CIRP表达水平的评价指标。
检测结果见附图1,结果表明在皮肤鳞癌癌组织中CIRP的表达较正常组织明显升高。以上结果表明CIRP与皮肤鳞癌的发展呈正相关。
2.皮肤鳞癌细胞系和角质形成细胞系中CIRP的RT-qPCR检测
检测方法如下:
(1)细胞总RNA的提取
该实验所用耗材及试剂均为RNase-free产品。
a)弃去6孔板中的培养基,用PBS缓冲液洗涤细胞2次;
b)加入800 μL TRIzol后,室温静置5 min,用移液器吹打数下,收集至1.5 mL EP管中,轻柔颠倒10次,室温静置5 min;
c)加入160 μL三氯甲烷(占TRIzol体积1/5),剧烈震荡约15 s,使其呈乳白状,室温静置3 min,4℃ 12000 rpm离心15 min,离心后分为三层,上层为无色透明的水层(RNA主要存在于其中),中间为一白色薄层,底层是呈粉红色的有机层;
d)小心地将上层无色透明的上清液(约TRIzol体积的1/2)转移至另一个1.5mL EP管中,加入等体积的异丙醇,上下颠倒10次充分混匀,室温静置10min;
e)4℃ 12000 rpm离心15 min,可见管底或管侧有胶状沉淀,小心弃去上清;
f)加入事先预冷的与TRIzol等体积的75%乙醇洗涤RNA沉淀,用移液器轻柔吹打数次,4℃ 7500 rpm离心5 min;
g)弃去上清,EP管倒扣,室温放置干燥约10 min使残余乙醇挥发,可见底部白色沉淀物变为半透明,加入约10-30 μL DEPC水,用移液器吹打使RNA充分溶解;
h)RNA纯度和浓度的测定:以DEPC处理水作为空白对照,260nm波长测定RNA浓度值。OD260/OD280在1.8-2.0之间时,认为RNA纯度较高。使用DEPC处理水调整RNA浓度,进行RNA反转录或将RNA产物放于-80℃冰箱保存备用。
(2)反转录反应
a)取RNase free的0.2 mL PCR管,按照表1所示反转录体系,在冰上配制反转录反应液,配制过程中要注意轻柔混匀;
b)加样完成后,混匀并短暂离心,放入PCR仪中进行反转录,按照表2所示反应条件进行反转录反应;
c)反应结束,取出反转录产物保存于-20℃备用。
(3)实时荧光定量PCR(real-time quantitative polymerase chain reaction,RT-qPCR)
a)引物序列:人CIRP及GAPDH基因的引物序列来自于上海生工生物工程股份有限公司,具体序列见表3。
b)取RNase free的0.1 mL PCR八连管,按照表4所示,在冰上配制反应体系;
c)加好样后,混匀并短暂离心;
d)将八连管放置于实时荧光定量PCR仪中,按照表5所示设置反应条件;
e)反应结束后,查看RT-qPCR的扩增曲线及熔解曲线,并记录各个基因的Ct值。用2-ΔΔCt的方法对Ct值进行分析,计算并比较目的基因的相对表达量。
CIRP在皮肤鳞癌细胞系和角质形成细胞系中的mRNA水平的RT-qPCR检测结果见附图2,在皮肤鳞癌细胞系中,CIRP mRNA相对表达量明显升高。
3.皮肤鳞癌细胞系和角质形成细胞系中CIRP蛋白水平的Western blotting检测
具体方法如下:
(1)细胞总蛋白的提取
a)每10 mL RIPA裂解液中加入蛋白酶抑制剂和磷酸酶抑制剂各1片,充分溶解、混匀后置于冰上备用;
b)弃去6孔板中的培养基,用PBS洗涤细胞两次,每孔加入120 μL裂解液,冰上裂解10 min;
c)在冰上充分刮细胞,将其收集至1.5 mL EP管中;
d)4℃ 12000 rpm离心20 min,将上清小心吸取至另一EP管中,记录上清体积;
(2)BCA法检测蛋白浓度
a)准备标准品:用去离子水将试剂盒中浓度为2 μg/μL的BSA溶液配制成浓度梯度的BSA标准品(2 μg/μL,1 μg/μL,0.5 μg/μL,0.25 μg/μL和0 μg/μL),用于建立标准曲线;
b)配制BCA工作液:根据BCA试剂盒说明书,按A液与B液=50:1的比例配制成工作液,混匀后向96孔板标准品孔和样品孔中加入工作液98 μL/孔,每个样品孔均设两个复孔;
c)加样:分别向标准品孔和样品孔加入标准品或待测蛋白样品2 μL/孔,37℃孵箱中孵育30 min;
d)上机检测:用酶标仪测定562 nm波长处的吸光度值;
e)计算蛋白浓度:根据标准品的浓度和吸光度值得出标准曲线和公式,利用公式计算出待测蛋白样品浓度;
(3)Western blotting所需蛋白样品准备:各蛋白样品分别加入1/4体积的5×loading buffer,金属浴100℃加热10 min进行蛋白变性,-20℃保存备用。
(4)Western blotting步骤
a)制胶
将规格适宜、干燥洁净的制胶玻璃板组装到制胶架上,配制下层分离胶后充分混匀,立刻将胶灌至梳齿底部下方0.5-1 cm处;立即配制浓缩胶,将浓缩胶灌至短玻璃的顶部。注意需缓慢平稳地灌胶,防止浓缩胶混入分离胶中;最后平稳地插入制胶梳,防止产生气泡,等待浓缩胶凝固,约30 min。
b)上样
提前从冰箱取出待测蛋白样品和预染蛋白marker,置于冰上备用;待浓缩胶凝固后将胶板组装至电泳槽,倒入适量的1×电泳缓冲液;小心拔出制胶梳,按预先设计的加样顺序加入蛋白样品20 μg/孔,在相应位置加入预染蛋白marker 5 µL/孔起指示作用。
c)SDS-PAGE电泳
接通电泳仪,200 V恒压电泳50 min,当指示剂溴酚蓝至分离胶底部时,停止电泳。
d)转膜(湿转法)
在转膜盘中倒入适量1×快速转膜液,将转膜夹、海绵垫和滤纸置于其中,裁剪适当大小的PVDF膜,在甲醇中活化1 min后转移至转膜液中,操作时需戴手套,防止膜被污染;撬开含有胶的玻璃板,切取所需的胶;在转膜液中,按照转膜夹黑面-海绵垫1块-滤纸2层-凝胶-PVDF膜-滤纸2层-海绵垫1块-转膜夹红面的顺序安装转膜“三明治”,安装时注意驱赶气泡;将组装好的转膜夹固定于转膜槽中,注意不要放反,加满转膜液,恒流400 mA转膜30min,转膜时间可根据蛋白分子量大小适当调整。
e)封闭
转膜结束后,将膜取出,蛋白面朝上放入快速封闭液中,若有两条带,则以“背靠背”原则放置,于室温慢速摇床上封闭10 min。
f)抗体孵育
孵育一抗:将膜根据目的蛋白的位置进行裁剪,孵育于相应的一抗中,于4℃冰箱内慢速摇床上孵育过夜;
洗膜:回收一抗,使用1×TBST缓冲液在室温水平快速摇床上洗涤3次,每次10min;
孵育二抗:加入相应的二抗,在室温慢速摇床上孵育1h;
洗膜:使用1×TBST缓冲液在室温水平快速摇床上洗涤3次,每次10 min。
g)ECL化学发光
提前打开成像仪进行预热,将ECL化学发光检测试剂盒中A液和B液按照1:1比例配制成发光液,混匀待用;使用滤纸将膜上的多余液体吸去,将发光液逐滴均匀覆盖于膜上;使用化学发光成像系统进行发光,保存图像,借助Image J软件对条带进行分析。
CIRP在皮肤鳞癌细胞系和角质形成细胞系中的蛋白水平的Western blotting检测结果见附图2,在皮肤鳞癌细胞系中,CIRP蛋白表达量明显升高。
4.siRNA干扰A431和SCL-1细胞中CIRP的表达
(1)针对CIRP的siRNA为si-CIRP-1和si-CIRP-2,si-NC为阴性对照,其序列见表6。
(2)siRNA转染过程及转染效率验证
a)铺板:细胞计数后接种至6孔板,在37℃、5% CO2细胞培养箱中常规培养,待细胞融合度达到50%左右时进行转染;
b)取6个1.5 mL无菌RNase-free EP管,在其中加入无血清培养基100 μL,并在其中3个EP管中分别添加1.25 μL的si-NC、si-CIRP-1和si-CIRP-2,在另外3个EP管中分别加入2.5 μL RNAiMAX转染试剂,用移液器轻柔混匀,室温静置5 min;
c)将各组siRNA培养基混合物分别滴加至RNAiMAX转染试剂培养基混合物中,用移液器轻柔混匀,室温静置15 min;
d)在加入相应的转染混合物之前将6孔板取出,吸弃旧培养基,加入1.5 mL新鲜培养基,待上述混合物静置15 min后分别滴加到相应孔中,轻柔地摇晃6孔板以混匀,做好标记后将6孔板放入37℃、5% CO2细胞培养箱中常规培养;
e)转染后48 h,提取细胞总RNA及总蛋白,通过RT-qPCR和Western blotting实验检测细胞中CIRP mRNA和蛋白的表达情况,验证转染效率。
CIRP siRNA在皮肤鳞癌细胞A431和SCL-1中敲低CIRP后CIRP mRNA和蛋白表达的情况的RT-qPCR和Western blotting检测,结果见附图3,CIRP siRNA明显抑制了两种细胞中CIRP的mRNA和蛋白表达。
5.CCK-8实验检测细胞增殖
(1)siRNA转染细胞后24 h,消化、离心、重悬si-NC转染组、si-CIRP-1转染组和si-CIRP-2转染组细胞并进行细胞计数;
(2)将各组细胞接种于96孔板,同时设空白对照组,在细胞周边孔内加入适量PBS防止边缘效应导致液体蒸发,每组设置5个复孔,共4个时间点,常规培养;
(3)以铺板时间为起点,在6 h、24 h、48 h和72 h后分别向96孔板中加入10 μLCCK-8溶液,再放回培养箱继续孵育2 h,用酶标仪检测450 nm波长处的各孔吸光度值;
(4)以时间为横坐标,OD值为纵坐标,绘制细胞生长曲线。
CIRP siRNA明显抑制皮肤鳞癌细胞系A431和SCL-1细胞的增殖能力,见附图4。
6.EdU实验检测细胞增殖
(1)将细胞接种于24孔板中,在37℃、5% CO2细胞培养箱中常规培养,待细胞融合度达到50%左右时转染siRNA,转染后继续常规培养;
(2)转染后48 h,按照试剂盒说明书配制2 × EdU工作液(20 μM),即按照完全培养基:EdU(10 mM)=500: 1的比例稀释EdU(10 mM)得到2 × EdU工作液(20 μM),放入培养箱中预热。随后将24孔板中旧培养基更换为新鲜培养基,再加入等体积的预热2×EdU工作液(20μM),使得最终作用于细胞的EdU终浓度为10μM,继续于37℃、5% CO2细胞培养箱中孵育2 h;
(3)EdU标记完成后,取出孔板,去除培养基,加入200 μL 4%的多聚甲醛,室温固定15 min;
(4)去除多聚甲醛,每孔用200 μL洗涤液洗涤细胞3次,每次5 min;
(5)去除洗涤液,每孔加入200 μL通透液,室温孵育10 min;
(6)去除通透液,每孔用200 μL洗涤液洗涤细胞2次,每次5 min;
(7)配制Click反应液,去除洗涤液后每孔加入100 μL Click反应液,轻轻摇晃培养板以确保反应混合物可以均匀覆盖样品,室温避光孵育30 min;
(8)反应结束后,去除Click反应液,用200μL洗涤液洗涤细胞3次,每次5min;
(9)细胞核染色:去除洗涤液后,每孔加入1×Hoechst 33342溶液200μL,室温避光孵育10min;
(10)去除1×Hoechst 33342溶液,用200μL洗涤液洗涤细胞3次,每次5min,再加入少量PBS以保持细胞湿润;
(11)尽快于倒置荧光显微镜下进行荧光检测并拍照。Azide 555的最大激发波长是555nm,最大发射波长是565nm;Hoechst 33342为蓝色荧光,最大激发波长为346nm,最大发射波长为460nm。
结果见附图5,CIRP siRNA明显抑制皮肤鳞癌细胞系A431和SCL-1细胞的增殖能力。
7.siRNA干扰A431和SCL-1细胞中CIRP的表达后ERK1/2信号通路相关蛋白的Western blotting检测
siRNA转染及Western blotting的具体方法同前。
在A431和SCL-1细胞应用CIRP siRNA敲低CIRP后48小时,细胞中ERK1/2信号通路的激活被明显抑制,见附图6。
8.shRNA干扰A431和SCL-1细胞中CIRP的表达
(1)CIRP shRNA慢病毒载体序列及阴性对照序列见表7。
(2)shRNA慢病毒转染过程及转染效率验证
a)转染前24 h,将A431和SCL-1细胞接种至6孔板,每孔加入2 mL完全培养基常规培养;
b)转染当日取出细胞进行观察,确保转染时细胞的融合度为40%-60%,且细胞状态良好;
c)将慢病毒取出置于冰上融化,混匀后吸取适量的慢病毒原液至加有2 mL完全培养基的离心管中进行稀释,再加入终浓度为5 μg/mL的polybrene以提高转染效率,混匀备用;
d)吸弃待转染细胞中的旧培养基,加入前述稀释好的病毒液,常规培养细胞;
e)转染6 h后观察细胞状态,若细胞状态良好,表明慢病毒对细胞没有明显毒性,则可继续培养,24 h后更换为新鲜完全培养基;
f)转染48 h后,观察荧光表达情况以初步判断转染效率,根据细胞生长情况进行及时换液和传代;
g)转染72 h后,使用嘌呤霉素(终浓度为5 μg/mL)筛选稳定表达的细胞株,筛选过程中需根据细胞的生长情况进行及时换液和传代(期间需继续添加嘌呤霉素以维持抗性),以保证细胞良好的生长状态;
h)继续培养扩增稳定表达的细胞株,并提取细胞总RNA和总蛋白,利用RT-qPCR和Western blotting实验验证转染效率。
CIRP shRNA在皮肤鳞癌细胞A431和SCL-1中敲低CIRP后CIRP mRNA和蛋白表达的情况的RT-qPCR和Western blotting检测结果见附图7,CIRP shRNA明显抑制了两种细胞中CIRP的mRNA和蛋白表达。
9.克隆形成实验检测细胞增殖
(1)取对数生长期的sh-NC和sh-CIRP细胞稳转株,常规消化收集细胞接种于6孔板中,接种后轻轻摇匀,将细胞放入培养箱中继续培养2 w,期间每3-4 d更换新鲜培养基;
(2)染色:去除培养基,用PBS洗涤细胞2次,每孔加入1 mL 4%多聚甲醛,室温固定细胞15 min,去除多聚甲醛,用PBS洗涤细胞1次,再加入800μL结晶紫染液,室温染色15min;
(3)染色结束后去除结晶紫溶液,用流水缓慢洗掉染色液,室温自然干燥;
(4)将6孔板置于光线充足处拍照,计算克隆形成数目。
结果见附图8,CIRP shRNA明显抑制皮肤鳞癌细胞系A431和SCL-1细胞的克隆形成能力。
10.裸鼠成瘤实验
(1)培养稳定转染sh-NC和sh-CIRP慢病毒的A431和SCL-1细胞株,收集对数期生长的细胞,常规消化后,1000 rpm离心5 min,弃上清;
(2)PBS洗两次,再离心,弃上清,用PBS将细胞重悬为单细胞悬液,并调整细胞浓度为5×107个/mL;
(3)4 w龄Balb/c雌性裸鼠在SPF级别动物房饲养10 d,无菌条件下消毒裸鼠右侧腰背部局部皮肤,用1 mL注射器分别吸取0.1 mL(5×l06个细胞)A431-sh-NC、A431-sh-CIRP、SCL-1-sh-NC和SCL-1-sh-CIRP细胞悬液,分别注射到各雌性Balb/c裸鼠右侧腰背部皮下,每组6只;
(4)接种后的动物在SPF级动物房中饲养,观察裸鼠的饮食及一般生活状态、开始成瘤的时间、成瘤速度,每3 d用游标卡尺测量移植瘤的长短径,按体积(mm3)=1/2(长径×短径2)计算肿瘤体积,绘制移植瘤体积变化曲线;
(5)接种后第15 d,用颈椎脱臼处死裸鼠,拍照后将移植瘤体完整剥离,拍照、称重;
(6)取一部分瘤组织放入4%多聚甲醛中固定,固定结束后,进一步用石蜡包埋切片,进行IHC染色检测,IHC染色方法同前。
裸鼠皮下移植瘤实验结果见附图9-10,使用shRNA敲低CIRP后,可明显抑制肿瘤的生长,同时增殖标志物Ki-67和p-ERK1/2蛋白的表达也被显著抑制。
皮肤鳞癌是表皮异常增殖性皮肤病中最主要的代表性疾病之一,由上述实施例结果可知,CIRP与皮肤鳞癌的发展呈正相关,使用siRNA或shRNA抑制CIRP的表达,能够显著抑制细胞的增殖和肿瘤生长,并且可通过抑制ERK1/2信号通路的激活来达到抑制皮肤鳞癌发展的作用。
鉴于此,CIRP可作为在制备防治表皮异常增殖性皮肤病的药物中的靶点,可以在基因水平和/或蛋白水平以CIRP作为药物靶点,如通过阻碍CIRP表达或转录的DNA或RNA,或者是阻碍或抑制CIRP作用于ERK1/2信号通路的药物;同时相应的针对siRNA或shRNA敲低CIRP的载体或CIRP的抑制剂制剂均可在制备防治表皮异常增殖性皮肤病的药物中进行应用。
以上所述仅为本发明的较佳实施例,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

1.CIRP作为靶点在制备防治表皮异常增殖性皮肤病的药物中的应用。
2.根据权利要求1所述的应用,其特征在于:所述的药物是在基因水平和/或蛋白水平以CIRP作为药物靶点。
3.根据权利要求1所述的应用,其特征在于:所述的药物是针对siRNA或shRNA敲低CIRP的载体。
4.根据权利要求1所述的应用,其特征在于:所述的药物是阻碍CIRP表达或转录的DNA或RNA。
5.根据权利要求1 所述的应用,其特征在于:所述的药物是阻碍或抑制CIRP激活ERK1/2信号通路的药物。
6.根据权利要求3 所述的应用,其特征在于:所述siRNA或shRNA敲低CIRP的载体在制备防治表皮异常增殖性皮肤病的药物中应用。
