The invention relates to the field of anti-aging preparations, in particular to a preparation method of a human anti-aging cell substance.
Background
Currently, with age, the bones, soft tissues and skin of the body change, while facial aging is the most prominent, the degree of aging is progressively more accentuated, and consumers have a progressively more enhanced pursuit of younger beauty. The aging behavior mainly includes: skin dryness/clarity, cigarette paper wrinkles, partial loss of skin elasticity, sagging, etc.; the proportion of anti-aging skin care products and medicines in the market is also increasing year by year.
Chinese patent publication No. CN114306203A discloses a preparation method and application of biological polypeptide and stem cell exosomes, wherein the stem cell exosomes effectively improve microcirculation of blood at focus, reconstruct capillary network, reduce restenosis of occluded blood vessels, are important raw material substances for repairing damaged tissues, and can moisten skin and endow the skin with smooth feeling; the biological polypeptide has various biological activities of scavenging free radicals, resisting oxidation, resisting aging and the like, can promote the proliferation of skin cells, can provide nutrition for skin, delay skin aging, has small irritation and lower side effect of medicines.
However, the technical scheme has the following defects:
the application mode of the scheme is oral administration, and most of nutrient substances of the prepared biological polypeptide dry powder are metabolized by human bodies, and cannot directly act on skin to resist aging, so that the effect efficiency is low and the utilization rate is low.
Disclosure of Invention
The invention aims at solving the problems in the background technology, and provides a preparation method of a human anti-aging cell substance, which is convenient to use, quick in effect on skin and high in utilization rate.
The technical scheme of the invention is that the preparation method of the human anti-aging cell substance comprises the following steps:
s1, soaking and cleaning the fish skin and the sea cucumber in saline water, and mashing the fish skin and the sea cucumber into a mixed paste after cleaning;
s2, adding protease into the mixed paste, performing enzymolysis in a constant-temperature oscillator, sealing the mixed paste after the enzymolysis is performed for a preset time, and then concentrating and removing impurities to obtain a mixed polypeptide solution;
s3, dissolving sodium ascorbyl phosphate into a citrate buffer solution, wherein the concentration of the citrate buffer solution is 0.1mol/L, and obtaining a solution component a with the ph value of 6.5-7.2;
s4, mixing the stem cell extract, lactic acid, glycerol, trehalose and triethanolamine, heating and stirring for 20-30min at 55-60 ℃ at a stirring speed of 100-120rpm to obtain a solution component b;
s5, stirring and mixing isopropyl myristate, soybean lecithin and sodium hyaluronate, gradually adding deionized water in the stirring process, and stirring for 10-15min at 60-70 ℃ at a stirring speed of 70-80rpm to obtain a solution component c;
s6, stirring and mixing the solution component a, the solution component b and the solution component c, performing ultrasonic treatment for 8-10min, and filtering to obtain a pasty semi-finished product;
s7, vibrating and stirring the pasty semi-finished product and the mixed polypeptide solution, uniformly mixing, and heating at 100-110 ℃ for 20-25min to obtain the pasty finished product.
Preferably, the following raw materials in parts by weight are adopted: 10-15 parts of fish skin, 20-30 parts of sea cucumber, 50-60 parts of brine, 10-12 parts of protease, 5-8 parts of sodium ascorbyl phosphate, 10-14 parts of citrate buffer solution, 30-45 parts of stem cell extract, 3-5 parts of lactic acid, 6-8 parts of glycerol, 7-10 parts of trehalose, 2-4 parts of triethanolamine, 5-8 parts of isopropyl myristate, 7-12 parts of soybean lecithin, 8-14 parts of sodium hyaluronate and 20-25 parts of deionized water.
Preferably, the sea cucumber is 30-35 standard sea cucumber, and the fish skin is tilapia skin, cod skin or eel skin.
Preferably, in step S2, the mixed paste is subjected to enzymolysis by using protease for 30-40min at 30-40deg.C for 50-55min.
Preferably, the protease enzymolysis process is divided into two times, 60% of protease is added in the first enzymolysis process, the enzymolysis time is 15-20min, and the enzymolysis temperature is 30-35 ℃; the remaining 40% of protease is added in the second enzymolysis process, the enzymolysis time is 15-20min, and the enzymolysis temperature is 35-40 ℃.
Preferably, the stem cell extract comprises an sheep umbilical cord blood stem cell extract and an apple stem cell extract, and the sheep umbilical cord blood stem cell extract and the apple stem cell extract are prepared according to the following steps of 6:4, proportioning.
Preferably, in step S6, the mixture of the solution component a, the solution component b and the solution component c after the ultrasonic treatment is subjected to filtration treatment through a 100-mesh filtration membrane.
Preferably, the method further comprises the step S8 of packaging the prepared pasty semi-finished product into a sealing box.
Compared with the prior art, the invention has the following beneficial technical effects:
the invention adopts various biological components, and the pasty finished product is finally prepared by multiple steps of processing of various raw materials, can be smeared on the face and the neck of a user in a smearing way, is convenient to use, has high utilization rate of nutrient substances in the finished product, and has quick response. Example 1
As shown in fig. 1-3, the preparation method of the anti-aging cell substance for the human body provided in this embodiment adopts the following raw materials in parts by weight: 10 parts of fish skin, 20 parts of sea cucumber, 50 parts of salt water, 10 parts of protease, 5 parts of sodium ascorbyl phosphate, 10 parts of citrate buffer solution, 45 parts of stem cell extract, 5 parts of lactic acid, 8 parts of glycerin, 10 parts of trehalose, 4 parts of triethanolamine, 8 parts of isopropyl myristate, 12 parts of soybean lecithin, 14 parts of sodium hyaluronate and 25 parts of deionized water, and the preparation method comprises the following steps:
s1, soaking and cleaning fish skin and sea cucumbers in brine, mashing the fish skin and the sea cucumbers into mixed paste after cleaning, wherein 30 standard sea cucumbers are selected as the sea cucumbers, and the fish skin is selected from tilapia skin, cod skin or eel skin, is rich in collagen and is beneficial to wrinkle resistance;
s2, adding protease into the mixed paste, performing enzymolysis in a constant-temperature oscillator, sealing the mixed paste after the enzymolysis is performed for a preset time, and then concentrating and removing impurities to obtain a mixed polypeptide solution; the enzymolysis time of the mixed paste by using protease is 30min, and the sealing time is 50min; the protease enzymolysis process is divided into two times, 60% of protease is added in the first enzymolysis process, the enzymolysis time is 15min, and the enzymolysis temperature is 30 ℃; the remaining 40% of protease is added in the second enzymolysis process, the enzymolysis time is 15min, the enzymolysis temperature is 35 ℃, and the mixed paste can be fully and effectively subjected to enzymolysis through double enzymolysis treatment;
s3, dissolving sodium ascorbyl phosphate into a citrate buffer solution, wherein the concentration of the citrate buffer solution is 0.1mol/L, so as to obtain a solution component a with a ph value of 6.5;
s4, mixing the stem cell extract, lactic acid, glycerol, trehalose and triethanolamine, heating and stirring for 30min at 60 ℃ at a stirring speed of 120rpm to obtain a solution component b; the stem cell extract comprises an sheep umbilical cord blood stem cell extract and an apple stem cell extract, and the sheep umbilical cord blood stem cell extract and the apple stem cell extract are prepared according to the following steps of 6:4, proportioning the materials in proportion;
s5, stirring and mixing isopropyl myristate, soybean lecithin and sodium hyaluronate, gradually adding deionized water in the stirring process, and stirring for 10min at 60 ℃ after the stirring is finished, wherein the stirring speed is 70rpm, so as to obtain a solution component c;
s6, stirring and mixing the solution component a, the solution component b and the solution component c, performing ultrasonic treatment for 10min, and filtering the mixture of the solution component a, the solution component b and the solution component c after ultrasonic treatment through a 100-mesh filter membrane to obtain a pasty semi-finished product after filtration;
s7, vibrating and stirring the pasty semi-finished product and the mixed polypeptide solution, uniformly mixing, and heating at 100 ℃ for 20min to obtain a pasty finished product;
s8, packaging the prepared pasty semi-finished product into a sealing box.
Example two
The preparation method of the anti-aging cell substance for human body provided in this embodiment is different from the first embodiment in that:
the raw materials in parts by weight are as follows: 12 parts of fish skin, 24 parts of sea cucumber, 55 parts of brine, 11 parts of protease, 6 parts of sodium ascorbyl phosphate, 12 parts of citrate buffer solution, 35 parts of stem cell extract, 4 parts of lactic acid, 7 parts of glycerin, 9 parts of trehalose, 3 parts of triethanolamine, 6 parts of isopropyl myristate, 10 parts of soybean lecithin, 10 parts of sodium hyaluronate and 22 parts of deionized water;
s1, soaking and cleaning fish skin and sea cucumbers in brine, and mashing the fish skin and the sea cucumbers into mixed paste after cleaning, wherein 32 standard sea cucumbers are selected as sea cucumbers;
s2, adding protease into the mixed paste, performing enzymolysis in a constant-temperature oscillator, sealing the mixed paste after the enzymolysis is performed for a preset time, and then concentrating and removing impurities to obtain a mixed polypeptide solution; the enzymolysis time of the mixed paste by using protease is 35min, and the sealing time is 50-55min; the protease enzymolysis process is divided into two times, 60% of protease is added in the first enzymolysis process, the enzymolysis time is 17min, and the enzymolysis temperature is 32 ℃; adding the rest 40% of protease in the second enzymolysis process, wherein the enzymolysis time is 18min, and the enzymolysis temperature is 38 ℃;
s3, dissolving sodium ascorbyl phosphate into a citrate buffer solution, wherein the concentration of the citrate buffer solution is 0.1mol/L, so as to obtain a solution component a with a ph value of 6.9;
s4, mixing the stem cell extract, lactic acid, glycerol, trehalose and triethanolamine, heating and stirring for 25min at 57 ℃ with the stirring speed of 112rpm to obtain a solution component b;
s5, stirring and mixing isopropyl myristate, soybean lecithin and sodium hyaluronate, gradually adding deionized water in the stirring process, and stirring for 12min at 64 ℃ after the stirring is finished, wherein the stirring speed is 74rpm, so as to obtain a solution component c;
s6, stirring and mixing the solution component a, the solution component b and the solution component c, performing ultrasonic treatment for 9min, and filtering the mixture of the solution component a, the solution component b and the solution component c after ultrasonic treatment through a 100-mesh filter membrane to obtain a pasty semi-finished product after filtration;
and S7, vibrating and stirring the pasty semi-finished product and the mixed polypeptide solution, uniformly mixing, and heating at 106 ℃ for 22min to obtain a pasty finished product.
Example III
The preparation method of the anti-aging cell substance for human body provided in this embodiment is different from the first embodiment and the second embodiment in that:
the raw materials in parts by weight are as follows: 15 parts of fish skin, 30 parts of sea cucumber, 60 parts of brine, 12 parts of protease, 8 parts of sodium ascorbyl phosphate, 14 parts of citrate buffer solution, 30 parts of stem cell extract, 3 parts of lactic acid, 6 parts of glycerin, 7 parts of trehalose, 2 parts of triethanolamine, 5 parts of isopropyl myristate, 7 parts of soybean lecithin, 8 parts of sodium hyaluronate and 20 parts of deionized water;
s1, soaking and cleaning fish skin and sea cucumbers in brine, and mashing the fish skin and the sea cucumbers into mixed paste after cleaning, wherein the sea cucumbers adopt 35 standard sea cucumbers;
s2, adding protease into the mixed paste, performing enzymolysis in a constant-temperature oscillator, sealing the mixed paste after the enzymolysis is performed for a preset time, and then concentrating and removing impurities to obtain a mixed polypeptide solution; the enzymolysis time of the mixed paste by using protease is 40min, and the sealing time is 50-55min; the protease enzymolysis process is divided into two times, 60% of protease is added in the first enzymolysis process, the enzymolysis time is 20min, and the enzymolysis temperature is 35 ℃; adding the rest 40% of protease in the second enzymolysis process, wherein the enzymolysis time is 20min, and the enzymolysis temperature is 40 ℃;
s3, dissolving sodium ascorbyl phosphate into a citrate buffer solution, wherein the concentration of the citrate buffer solution is 0.1mol/L, so as to obtain a solution component a with a ph value of 7.2;
s4, mixing the stem cell extract, lactic acid, glycerol, trehalose and triethanolamine, heating and stirring for 20min at 55 ℃ with the stirring speed of 100rpm to obtain a solution component b;
s5, stirring and mixing isopropyl myristate, soybean lecithin and sodium hyaluronate, gradually adding deionized water in the stirring process, and stirring for 15min at 70 ℃ after the stirring is finished, wherein the stirring speed is 80rpm, so as to obtain a solution component c;
s6, stirring and mixing the solution component a, the solution component b and the solution component c, performing ultrasonic treatment for 8min, and filtering the mixture of the solution component a, the solution component b and the solution component c after ultrasonic treatment through a 100-mesh filter membrane to obtain a pasty semi-finished product after filtration;
and S7, vibrating and stirring the pasty semi-finished product and the mixed polypeptide solution, uniformly mixing, and heating at 110 ℃ for 25min to obtain a pasty finished product.
For the three embodiments, three users are selected for experiments, the three users firstly clean the face and the neck with clean water, then respectively smear the pasty finished products prepared by the methods, the situations of the corners of eyes, the lower eyelids and the neck of the three users are basically the same, the first user smears the corresponding pasty finished products on the corners of eyes, the lower eyelids and the neck, and the other two users perform the same operation.
Comparative example
The control experiment is carried out by selecting the user with the same condition of the canthus, the lower eyelid and the neck of the other three users in the three embodiments, the user does not use the product to smear the canthus, the lower eyelid and the neck, only clean water is used for cleaning the face and the neck, normal cleaning treatment is carried out, the moisture loss condition, the skin elasticity condition and the luster condition of the canthus, the lower eyelid and the neck of the user are respectively detected, and the average value of all the detection at the three parts is respectively taken as the average moisture loss value, the average elasticity value and the average glossiness value.
The detection value immediately after the product is cleaned or immediately after the product is smeared on the eyes, the lower eyelid and the neck of a user is taken as an initial value, the detection value after 12 hours is taken as a final value, the initial value is subtracted from the final value, and then the initial value is divided by the final value, so that the 12-hour moisture loss rate, the 12-hour skin elasticity change rate and the 12-hour skin glossiness change rate are obtained through calculation, and a comparison chart shown in figure 4 is obtained. By comparing the parameters in the graphs, each indication of the comparison example is worse than the indication of the three examples after the user adopts the product, so that the method is fully demonstrated to have the efficacy of resisting human body aging.
The invention adopts various biological components, and the pasty finished product is finally prepared by multiple steps of processing of various raw materials, can be smeared on the face and the neck of a user in a smearing way, is convenient to use, has high utilization rate of nutrient substances in the finished product, and has quick response. Claims (8)
1. The preparation method of the human anti-aging cell substance is characterized by comprising the following steps:
s1, soaking and cleaning the fish skin and the sea cucumber in saline water, and mashing the fish skin and the sea cucumber into a mixed paste after cleaning;
s2, adding protease into the mixed paste, performing enzymolysis in a constant-temperature oscillator, sealing the mixed paste after the enzymolysis is performed for a preset time, and then concentrating and removing impurities to obtain a mixed polypeptide solution;
s3, dissolving sodium ascorbyl phosphate into a citrate buffer solution, wherein the concentration of the citrate buffer solution is 0.1mol/L, and obtaining a solution component a with the ph value of 6.5-7.2;
s4, mixing the stem cell extract, lactic acid, glycerol, trehalose and triethanolamine, heating and stirring for 20-30min at 55-60 ℃ at a stirring speed of 100-120rpm to obtain a solution component b;
s5, stirring and mixing isopropyl myristate, soybean lecithin and sodium hyaluronate, gradually adding deionized water in the stirring process, and stirring for 10-15min at 60-70 ℃ at a stirring speed of 70-80rpm to obtain a solution component c;
s6, stirring and mixing the solution component a, the solution component b and the solution component c, performing ultrasonic treatment for 8-10min, and filtering to obtain a pasty semi-finished product;
s7, vibrating and stirring the pasty semi-finished product and the mixed polypeptide solution, uniformly mixing, and heating at 100-110 ℃ for 20-25min to obtain the pasty finished product.

The method for preparing a human anti-aging cell substance according to claim 1, wherein the following raw materials are used in parts by weight: 10-15 parts of fish skin, 20-30 parts of sea cucumber, 50-60 parts of brine, 10-12 parts of protease, 5-8 parts of sodium ascorbyl phosphate, 10-14 parts of citrate buffer solution, 30-45 parts of stem cell extract, 3-5 parts of lactic acid, 6-8 parts of glycerol, 7-10 parts of trehalose, 2-4 parts of triethanolamine, 5-8 parts of isopropyl myristate, 7-12 parts of soybean lecithin, 8-14 parts of sodium hyaluronate and 20-25 parts of deionized water.
3. The method for preparing anti-aging cell substance for human body according to claim 1, wherein sea cucumber with 30-35 standard sea cucumber is selected as sea cucumber, and tilapia skin, cod skin or eel skin is selected as fish skin.
4. The method for preparing anti-aging cell substance according to claim 1, wherein in step S2, the mixed paste is enzymatically hydrolyzed with protease for 30-40min at 30-40 ℃ for 50-55min.

The method for preparing anti-aging cell substances for human body according to claim 4, wherein the enzymolysis process of protease is divided into two times, 60% protease is added in the first enzymolysis process, the enzymolysis time is 15-20min, and the enzymolysis temperature is 30-35 ℃; the remaining 40% of protease is added in the second enzymolysis process, the enzymolysis time is 15-20min, and the enzymolysis temperature is 35-40 ℃.

The method for preparing human anti-aging cell substances according to claim 1, wherein the stem cell extract comprises an sheep umbilical cord blood stem cell extract and an apple stem cell extract, and the sheep umbilical cord blood stem cell extract and the apple stem cell extract are prepared according to the following steps of 6:4, proportioning.
7. The method for preparing a human anti-aging cell substance according to claim 1, wherein in step S6, the mixture of the solution component a, the solution component b and the solution component c after the ultrasonic treatment is filtered through a 100-mesh filter membrane.
8. The method for preparing anti-aging cell substance according to claim 1, further comprising step S8 of packing the prepared pasty semi-finished product into a sealed box.
