本申请是申请号为201680053548.X的发明名称为“重组Orf病毒载体”的中国专利申请的分案申请,原申请是2016年07月15日提交的PCT国际申请PCT/EP2016/066926于2018年03月15日进入中国国家阶段的申请。
技术领域
本发明涉及重组Orf病毒载体、包含所述重组Orf病毒载体的细胞、包含根据本发明的重组Orf病毒载体和/或根据本发明的细胞的组合物、根据本发明的重组Orf病毒载体用于表达外源基因的用途和编码Orf病毒载体启动子的核酸分子。
背景技术
在生物技术中使用病毒载体以将遗传物质引入靶细胞。引入的遗传物质常常编码用于产生重组蛋白的外源基因。由于这个原因,病毒载体是重要的平台技术,特别是用于生产越来越多地使用的重组疫苗,和感染性疾病的传统预防,以及用于开发新的、创新的治疗概念,例如治疗性肿瘤免疫。
Orf病毒(ORFV)属于痘病毒科并且具有许多特征,使得其对于生产重组疫苗而言引人关注并且与其他技术相比其更优选。其是副痘病毒(Parapockenviren)属的原型并且属于痘病毒(Poxviridae)科。ORFV是具有令人想起羊毛球的形态的有包膜的、复杂的dsDNA病毒,并且平均尺寸为约260nm×160nm。它们具有含有高GC含量且尺寸为约130kbp至150kbp的线性DNA基因组,其中心区域在两侧由ITR区域(inverted terminal repeat,反向末端重复)界定并以将两个DNA单链彼此共价连接的发夹结构结束。在基因组的中心区域中存在主要对于病毒复制和形态发生必需且在痘病毒之间高度保守的占优势的基因。相比之下,在ITR区域中存在所谓的非保守毒力基因,其显著地决定宿主范围、致病性和免疫调节,并且由此表征病毒。
在痘病毒中,病毒复制限于胞质并开始于病毒与宿主细胞表面的结合。在融合之后,所谓的病毒核(蛋白质核)释放到包含病毒基因组、早期病毒mRNA和病毒转录因子(Transkriptionsfaktoren,TF)的胞质中。随后,在痘病毒特异性早期启动子的控制下,在合成病毒mRNA的地方起始早期病毒基因表达。在核结构分解之后,病毒DNA释放到胞质中。与仅在病毒TF的控制下发生的“早期”基因转录相比,后面的“中期”和“晚期”基因转录取决于细胞TF的帮助。结果,与“中期”和“晚期”基因的表达相比,痘病毒中早期基因的表达既不需要病毒DNA的复制也不需要病毒产生。
ORFV的特征在于非常窄的天然宿主范围,包括绵羊和山羊。感染经由允许病毒侵入的皮肤损伤而发生。随后亲皮性病毒(dermatropen Virus)的复制限于引发传染性皮炎或传染性深脓疱(Ecthyma contagiosum)的再生角化细胞。这种通常轻度发展的自限性感染表现为局部受限的皮肤或黏膜损伤,由此由多形核粒细胞的大量浸润引起的脓疱形成主要出现在口和乳房处。约4至6周后损伤愈合而不形成疤痕。尽管有强烈的免疫应答,但不形成持久的免疫保护,使得即使在数周后再感染仍是可能的,尽管临床症状和病毒产生显著减少。对于ORFV未观察到全身传播或病毒血症;实验性静脉内注射高剂量的感染性病毒之后也未观察到全身传播或病毒血症。
ORFV被认为是动物传染病的病原体并且在极少数情况下也可经由受伤的皮肤传递给人。在感染之后,可以观察到限于局部的模块化的肿胀,所述肿胀主要限于手指和手,偶尔存在淋巴结肿胀和发热。通常,所述进展是无害的,没有并发症并且在三至八周内完全治愈而没有临床后果。在免疫功能低下的患者中,还观察到更严重的感染进展,然而,其在用抗病毒药例如西多福韦(Cidofovir)或咪喹莫特(Imiquimod)治疗之后完全治愈。
ORFV在生产重组疫苗方面是令人感兴趣的。与天花病毒相比,ORFV的特征在于非常窄的自然宿主向性(包括绵羊和山羊)。因此,几乎可以排除如可以在牛痘和腺病毒的最常见的病毒载体中观察到的由天然感染引起的人中针对载体的抑制性“免疫前作用”。此外,异常弱的和短寿命的ORFV特异性载体免疫允许用针对另外的病原体的基于ORFV的疫苗非常有效的加强剂和/或更新疫苗接种或免疫。
在许可性和非许可性宿主中施用ORFV导致强烈的免疫刺激反应,其特征在于明显的先天免疫机制的诱导以及干扰素、细胞因子和趋化因子的释放。免疫后不久,树突细胞在注射部位累积,接下来通过激活T细胞和B细胞,诱导特异性适应性免疫应答。与来自主要诱导体液强调的免疫应答的灭活病毒和活载体的疫苗相比,用重组ORFV免疫后的细胞和体液免疫应答的平衡诱导是显著的优势。同时,它使得佐剂的使用变得多余,这可导致不意图的副作用和炎性反应。此外,其他优点可能实现在不使用抗生素和不在母鸡卵中产生的情况下在永久细胞系中标准化生产重组疫苗(提高蛋白质的数量和抗生素不耐性)。
在兽医领域批准的减毒ORFV疫苗的名称为D1701,与相同的ORFV毒株一致。这种以商品名Baypamun(Bayer)或Zylexis(Pfizer)已知的呈灭活形式的疫苗最初是通过从绵羊中分离野生型病毒和作为在牛肾培养细胞中进行多次传代的结果的随后适应而获得的。接下来在Vero细胞(非洲绿猴肾细胞)中进一步适应,这导致获得ORFV载体D1701-V。即使在受到免疫抑制的绵羊中,D1701-V被进一步减毒并且仅导致渐近感染。
迄今为止,重组ORFV载体使用VEGF基因座作为插入点。根据目前的知识,这导致以下假定的优势:即通过消除处于痘病毒特异性“早期”启动子控制下且被认为是毒力因子的vegf-e基因,发生载体的进一步减毒。同时,生产了几种基于ORFV的重组疫苗并在动物模型中对其检查。目前,针对多种传染性疾病使用重组ORFV疫苗,例如针对奥耶斯基氏病(Aujeszky′sche Krankheit)、狂犬病、博纳病(Borna’sche Erkrankung)、流感或典型猪瘟(classical swine fever)。
可以在过去几年中通过建立许多针对不同的病毒感染性疾病的疫苗来证实重组Orf病毒的良好的免疫刺激和预防可用效果。关于包含ORFV的重组痘病毒用于生产重组蛋白的用途的综述可见于Rziha等(2000),Generation of recombinant parapoxviruses:non-esssential genes suitable for insertion and expression of foreign genes,Journal or Biotechnology,第83卷,第137-145页,以及Büttner和Rziha(2002),Parapoxviruses:From the Lesion to the Viral Genome,J.Vet.Med.B,第49卷,第7-16页中。
发明内容
目前的重组ORFV载体的限制性使用可以特别通过长期的选择程序解释。特别是多价疫苗的生产迄今为止仍是不可能的。此外,已知的重组ORFV载体不具有精确可控的外源基因表达。
在这个背景下,构成本发明基础的问题是提供新的重组ORFV载体,通过所述载体可以减少或避免已知的ORFV载体和载体系统的缺点。
该问题通过提供重组Orf病毒(ORFV)载体来解决,所述重组Orf病毒载体包含以下:
(1)至少一个编码和表达外源基因的核苷酸序列,和
(2)至少一个控制所述核苷酸序列的表达的启动子,
其特征在于
所述核苷酸序列被插入到位于ORFV基因组中的以下区域中的插入位点(Insertionslokusse,IL)1、2和3中的至少一个中:
根据本发明,Orf病毒(ORFV)是指落入羊副痘病毒(Parapoxvirus ovis)种类下的所有病毒和病毒株,特别是毒株D1701。
根据本发明,重组ORFV载体是指配置成向生物细胞中转运外源基因和/或在生物细胞中表达外源基因的基于ORFV基因组的载体。
根据本发明,外源基因是指不来源于ORFV基因组的基因或开放阅读框(ORF)。
根据本发明,启动子是指这样的核酸部分:其允许本发明的ORFV载体中的外源基因的调控表达。优选地,其是指ORF启动子,即存在于野生型ORFV基因组中的启动子或由此衍生的启动子,或人工启动子,例如痘病毒启动子、CMV启动子等。
在重组ORFV载体中,ORFV基因组中的插入位点IL1、2和3的位置可以根据本发明以几种方式确定:通过ORFV基因组中的限制性片段、ORFV基因或开放阅读框(ORF)或核苷酸位置。
IL1、2和3定位的传统描述通过限制性图谱和通过插入区域所在的限制性片段发生。对于毒株D1701,ORFV的限制性图谱示例性地示于Cottone等(1998),Analysis ofgenomic rearrangement and subsequent gene deletion of the attenuated Orfvirus strain D1701,Virus Research,第56卷,第53-67页中。该出版物的内容是本公开内容的组成部分。在此,根据本发明,例如关于IL 1,标示HindIII片段C、KpnI片段G、BamHI片段C/G、EcoRI片段B意指该插入位点从HindIII片段C延伸至EcoRI片段B。IL2从HindIII片段I-J延伸至EcoRI片段A/E。IL3从HindIII片段G/D延伸至EcoRI片段D。
对于IL1标示006、007(dUTPase)、008(G1Li-Ank)、009(G2L)意指插入位点从基因或ORF 006延伸至基因或ORF 009。括号中的标示是指在目前已知的范围内的编码产物或编码的酶活性。
根据本发明,IL1位于开始于核苷酸400至600(500±100)且终止于核苷酸1800至3000(2,400±600)的区域中。
根据上表,对于IL1所述的内容适用于IL2和IL3。
核苷酸(nt)的指示位置由发明人根据不同ORFV毒株的多次测定确定。对毒株D1701或由其得到的变体的检查导致以下特别优选的位置:
IL1:nt 496-nt 2,750;nt 496-nt 1,912和nt 511-nt 2,750
IL2:nt 5,2112-nt 7,736
IL3:nt 15,656-nt 17,849。
本发明人已经意识到,在所指示的新发现的插入位点中,可以将外源基因稳定地整合到ORFV基因组中。这是出人意料的。迄今为止,假定基因组中的插入位点的区域是病毒复制所需的并且不适合于外源基因的表达。
通过选择插入位点,可以控制基因表达,这已被证明是根据本发明的ORFV载体的显著优势。例如,IL2中外源基因的表达与目前使用的VEGF基因座中的表达相比降低至二分之一。此外,通过选择启动子,可以以靶向方式影响外源基因表达的强度和时间点。
本发明的基本问题由此得到完全解决。
在根据本发明优选的重组ORFV载体的一个实施方案中,ORFV是毒株D1701。
该手段的优点在于使用这样的载体:其为减毒的且仅在宿主中引起渐近感染。根据本发明,涵盖D1701的所有变体(包括D1701-B和D1701-V)。
根据按照本发明优选的重组ORFV载体的另一个实施方案,启动子是ORFV启动子,进一步优选早期ORFV启动子,所述早期ORFV启动子优选地包含选自以下的核苷酸序列:SEQID No.1(P1)、SEQ ID No.2(P2)、SEQ ID No.3(VEGF)、SEQ ID No.4(优化的“早期”)、SEQID No.5(7.5kDa启动子)和SEQ ID No.6(共有“早期”)。
该手段的优点在于使用这样的启动子,其允许高的外源基因表达水平和表达的靶向控制。启动子P1和P2是本发明人新开发的。其余的启动子来源于牛痘病毒并且描述于Davidson和Moss(1989),Structure of Vaccinia virus late promoters,J.Mol.Biol,第210卷,第771-784页,以及Yang等(2011),Genome-wide analysis of the 5’and 3’endsof Vaccinia Virus early mRNAs delineates regulatory sequences of annotatedand anomalous transcripts,J.Virology,第85卷,第12期,第5897-5909页,Broyles(2003),Vaccinia virus transcription,J.gene.Virol.,第84卷,第9期,第2293-2303页的其他连接中。根据本发明人的发现,与P1相比,P2引起显著增强的表达强度。这是出人意料地,因为启动子P1 100%对应于来自牛痘病毒的共有序列,然而,P2并不如此。“最佳”牛痘病毒启动子(天花)在ORFV(副痘)中的低表达是矛盾和出人意料的。还出人意料的是,P2启动子导致强表达。
根据按照本发明的重组ORFV载体的另一个优选的实施方案,启动子相对于编码外源基因的核苷酸序列位于上游nt 28±10至nt -13±10的位置处。
该手段的优点在于启动子位于这样的位置:其允许外源基因的高表达强度和可控表达。
根据按照本发明的重组ORFV载体的另一个优选的实施方案,至少在IL1、2或3中的一个中,插入有多于一个,优选2、3、4或更多个编码和表达外源基因的核苷酸序列。
该手段的优点在于通过根据本发明的一个重组ORFV载体,可以表达多种外源基因。该实施方案特别适合于生产同时针对许多抗原结构的多价疫苗。在每个插入位点中,可以表达多种外源基因,优选2、3、4或更多种。
在根据本发明优选的配置中,重组ORFV载体包含另外的编码和表达外源基因的核苷酸序列,所述核苷酸序列在优选早期ORFV启动子的控制下并且被插入到在ORFV基因组中位于vegf E基因的插入位点中。
该手段的优点在于采用现有技术中已经描述和良好表征的插入位点。使用vegf基因座可以用于基因表达的靶向控制。在vegf基因座中,与新的表达基因座中的任一个(例如IL 2)相比,可以提高外源基因的表达。
根据按照本发明优选的一个实施方案,重组ORFV载体的外源基因选自以下抗原的组:
-病毒抗原,优选地
狂犬病毒抗原,包括糖蛋白(RabG);
甲型流感病毒抗原,包括核蛋白(NP)、血凝素(HA)、神经氨酸酶(NA);
-肿瘤抗原,优选病毒性肿瘤抗原,包括HPV选择性病毒肿瘤抗原;
-肿瘤相关抗原,包括病毒性肿瘤相关抗原,包括HPV选择性病毒性肿瘤相关抗原;
-寄生虫抗原,优选疟原虫抗原;
-细胞因子。
该手段的优点在于通过根据本发明的重组ORFV病毒,特别重要的抗原,特别是用于疫苗生产的抗原是可表达的。
本发明的另一个主题涉及包含根据本发明的ORFV载体的生物细胞,优选哺乳动物细胞,进一步优选Vero细胞。
本发明的另一个主题涉及包含本发明的ORFV载体和/或本发明的细胞的组合物,优选药物组合物。药物组合物可以优选为疫苗,进一步优选为多价疫苗。
根据本发明的ORFV载体的特性、优点、特征和进一步发展以对应的方式应用于本发明的细胞和本发明的组合物。
本发明的另一个主题涉及根据本发明的重组ORFV载体用于表达至少一种外源基因,进一步优选表达至少一种包含一种外源基因产物的疫苗(单价疫苗),进一步优选包含至少两种外源基因产物的疫苗(多价疫苗)的用途。
在此背景下,“至少”意指涵盖2、3、4、5、6、7、8、9等种外源基因。
根据本发明的重组ORFV载体的特征、优点、特性和进一步发展以对应的方式应用于本发明的用途。
本发明的另一个主题涉及编码ORFV启动子,优选包含选自核苷酸序列SEQ-IDNo.1(P1)和SEQ-ID No.2(P2)的核苷酸序列的早期ORFV启动子的核酸分子。
根据本发明的核酸分子编码新的ORFV启动子,其特别适合于在重组ORFV载体中表达外源基因。所述启动子引起非常强的早期基因表达。

具体实施方式
实施方案
1.ORFV基因组
ORFV基因组由线性双链DNA组成且长度为约138kB、GC含量为约64%且包含130-132个基因。ORFV基因组的结构与其他痘病毒的结构类似。其由中心区域与在痘病毒科中包括高度保守性的必需基因组成。在ORFV基因组中,存在88个在所有的脊椎动物痘病毒亚科(Chordopoxvirinae)中保守的基因。病毒基因定位在末端区域中,其对于体外生长是非必需的,然而与病毒的致病性和向性相关。
与其他Orf病毒相比,经适应以在细胞培养物中复制的D1701病毒显示出反向末端重复(inverted terminal repeat,ITR)的显著扩大。这些改变不仅引起包括vegf-e基因的数个基因的缺失而且引起其复制。在牛BK-KL3A细胞中扩增的D1701-B对在Vero细胞中生长的适应导致病毒基因组(现在称为D1701-V)中的三个额外的插入位点IL1、IL2和IL3。它们示于图1中。
2.痘病毒启动子
痘病毒包含“早期”、“中期”和“晚期”启动子。这些不同的启动子包括几个特征化的序列特征,以下以VACV为例进一步解释。VACV的“早期”启动子由长度分别为16或15个核苷酸的关键区域组成,其通过11个核苷酸的间隔区与长度为7个核苷酸的起始区间隔开。该关键区域相当富含腺嘌呤,而间隔区相当富含胸腺嘧啶。转录的起始总是发生在嘌呤上,除极少数例外。关键区域中的核苷酸替换可对启动子活性具有显著的负面影响,甚至完全丧失活性也是可能的。早期VACV 7.5kDa启动子的替换分析显示出优化的关键区域,此外,其成功地获得了表1所示的“早期”痘病毒启动子的共有序列。“中期”启动子由约14个核苷酸长的富含AT的核心序列,随后是10-11个核苷酸的间隔区,其后是短的起始区组成。“晚期”启动子的结构由约20个核苷酸的上游富含AT的区域组成,所述区域通过包含高度保守的序列-1TAAAT+4的约6个核苷酸的间隔区与转录起始位置分开。

3.重组ORFV载体的产生
本发明人已在寻找用于产生重组多价ORFV载体的新策略。在ORFV适应Vero培养细胞期间,病毒基因组中出现了几个缺失。检查缺失区域是否适合外源基因的整合(图1A)。
为此,与另外的质粒一起,设计包含IL2区的同源部分的转移质粒pDel2(图2)。将外源基因克隆到质粒中是通过使用多个MCS(multipler Klonierungsstellen,多克隆位点)来实现的。此外,质粒以允许同时整合多个外源基因的方式被设计,所述外源基因各自在人工早期ORFV启动子的控制下并且受到痘病毒特异性T5NT早期转录终止基序的限制(图2)。
设计新的人工早期ORFV启动子P1和P2的核苷酸序列。
在第一个实验中,应当检查IL2基因座是否适合外源基因的稳定整合。为此目的,在启动子P2的控制下将mCherry荧光标记基因克隆到pDel2转移质粒中。随后将所述质粒转染到感染了D1701-VrV的Vero细胞中,在通过荧光显微术鉴定红色有光泽的细胞之后目视选择新的重组病毒并培养通过几次噬斑纯化(Plaquereinigungen)获得的同源重组体D1701-V-D2-Cherry(图3A,a)。
通过特异性PCR分析和Southern印迹杂交确保mCherry基因正确整合到D1701-VrV的IL2基因座中,可以通过荧光和Western印迹分析但也可以通过流式细胞术来证实正确表达(图3A,b)。
在此背景下,可以在感染后早期检测到强表达。通过重组体的多次体外传代,可以证实外源基因在ORFV基因组中的整合是稳定的。
通过所产生的其中AcGFP基因整合到vegf-e基因中且mCherry基因整合到IL2基因座中的重组体D1701-V-GFP-D2-Cherry,也可以证实,两个荧光基因同时在不同的插入位点中早期表达(图3B)。
此外,应该检查是否同时有第二外源基因可以被稳定地整合到IL2基因座中。为此目的,除了受P2控制的mCherry基因以外,可将在P1启动子控制下的AcGFP基因克隆到pDel2转移质粒中。类似于先前描述的D1701-V-P2-Cherry选择,实现了同源重组体D1701-V-D1-GFP-D2-Cherry的选择和纯化。
此外,通过PCR和Southern印迹杂交分析证实两个外源基因都正确整合到IL2基因座中。通过荧光显微术和流式细胞术进行表达的检测(图3C)。
在表达分析中,将启动子P1和P2的强度相互比较并与启动子Pvegf进行比较。可以表明,启动子P2诱导最强的基因表达,而启动子P1包括最弱的基因表达(图4A+4C)。这是出人意料的,因为P1 100%对应于来自牛痘病毒中的共有序列,而P2并不如此。
也可以证实,在单个启动子的控制下第二调控外源基因的整合对第一外源基因的表达强度没有影响。第二基因是否整合到相同或不同的插入位点中是不相关的。在将受P2控制的mCherry基因插入到VEGF基因座中之后,可以与将受P2调控的mCherry基因整合到IL2基因座中的重组体相比来分析插入区域的影响。可以表明,VEGF基因座中的基因表达是IL2基因座中的基因表达的约2倍强(图4B+4D)。
总之,可以表明,Orf病毒载体D1701-V非常适合于生产多价重组体。可以将几种外源基因稳定地整合到病毒基因组中,例如通过新发现的插入位点IL1、2和3,或通过已知的插入位点VEGF。外源基因表达的强度既取决于启动子,也取决于插入位点。在将受P2控制的外源基因整合到VEGF基因座中之后实现了最强的基因表达。
本发明人已经产生了另外多种载体,其可以与不同的标记外源基因、插入区域和启动子的性质和群集区分开(表2)。

该表给出了在得到本发明的工作期间产生的荧光重组ORFV载体的概述。对于每个重组体在表中指示了插入区域(基因座)和用于控制外源基因表达的启动子(Pvegf、P1、P2)。此外,指示了外源基因表达的强度(非常强=++++至弱=+)。
本发明为开发新的基于重组ORFV的疫苗创造了多种选择。可以产生同时表达多种抗原的重组体。这在组合疫苗的通用疫苗或针对多种肿瘤抗原的治疗性肿瘤疫苗的生产中可成为显著的优点。此外,抗原和细胞因子的同时插入可以以靶向方式影响免疫应答。
Claims (14)
1.重组Orf病毒(ORFV)载体,其包含以下:
(1)至少一个编码并表达外源基因的核苷酸序列,和
(2)至少一个控制所述核苷酸序列的表达的启动子,
其特征在于,
所述核苷酸序列位于插入位点(IL)1、2和3中的至少一个中,所述插入位点(IL)1、2和3位于ORFV基因组中的以下区域中:
2.根据权利要求1所述的重组ORFV载体,其中ORFV是毒株D1701。
3.根据权利要求1或2所述的重组ORFV载体,其特征在于,ORFV启动子是早期ORF启动子,其优选地包含选自以下的核苷酸序列:SEQ ID No.1(P1)、SEQ ID No.2(P2)、SEQ IDNo.3(VEGF)、SEQ ID No.4(优化的“早期”)、SEQ ID No.5(7.5kDa启动子)和SEQ ID No.6(共有“早期”)。
4.根据前述权利要求中任一项所述的重组ORFV载体,其特征在于,所述启动子相对于编码所述外源基因的所述核苷酸序列位于上游nt 28±10至nt-13±10的位置。
5.根据前述权利要求中任一项所述的重组ORFV载体,其特征在于,至少在所述IL1、2或3中的一个中,插入有多于一个,优选2、3、4或更多个编码并表达外源基因的核苷酸序列。
6.根据前述权利要求中任一项所述的重组ORFV载体,其特征在于,其包含另外的编码并表达外源基因的核苷酸序列,所述核苷酸序列在优选早期ORFV启动子的控制下并且被插入到在所述ORFV基因组中位于vegf E基因的插入位点中。
7.根据前述权利要求中任一项所述的重组ORFV载体,其特征在于,所述外源基因选自以下抗原的组:
-病毒抗原,优选地
狂犬病毒抗原,包括糖蛋白(RabG);
甲型流感病毒抗原,包括核蛋白(NP)、血凝素(HA)、神经氨酸酶(NA);
-肿瘤抗原,优选病毒性肿瘤抗原,包括HPV选择性病毒性肿瘤抗原;
-肿瘤相关抗原,包括病毒性肿瘤相关抗原,包括HPV选择性病毒肿瘤相关抗原;
-寄生虫抗原,优选疟原虫抗原;
-细胞因子。
8.包含根据前述权利要求中任一项所述的重组ORFV载体的细胞,优选哺乳动物细胞,进一步优选Vero细胞。
9.包含根据权利要求1至7中任一项所述的重组ORFV载体和/或根据权利要求8所述的细胞的组合物,优选药物组合物。
10.根据权利要求9所述的组合物,其特征在于,所述药物组合物为疫苗,优选多价疫苗。
11.根据前述权利要求中任一项所述的重组ORFV载体用于表达至少一种外源基因的用途。
12.根据权利要求11所述的用途,用于生产包含至少一种外源基因产物的疫苗(单价疫苗),优选包含至少两种外源基因产物的疫苗(多价疫苗)。
13.编码ORFV启动子的核酸分子,其包含选自SEQ ID No.1(P1)和SEQ ID No.2(P2)的核苷酸序列。
14.根据权利要求13所述的核酸分子,其特征在于,所述ORFV启动子是早期ORFV启动子。
