本发明属于多功能菌粮及功能食品制备技术领域。本发明提供了一株锦带花桑黄(Sanghuangporus weigelae)S‑LWZ20210623‑1a CGMCC No.40572。该菌株已于2023年4月12日保藏在中国微生物菌种保藏管理委员会普通微生物中心,该中心简称为CGMCC,该菌株保藏号为CGMCC No.40572,该中心地址为北京市朝阳区北辰西路1号院3号。本发明提供菌株能将大米发酵后的产物中粗多糖含量提升1.34倍、总酚类含量提升3.05倍、可溶性蛋白含量提升14.88倍。
Description
技术领域
本发明属于多功能菌粮制备技术领域。
背景技术
大米作为日常生活中最常食用的谷物之一,可以为人类提供蛋白质、脂肪酸、维生素、矿物质等营养物质。随着其精制化,追求口感而对大米过度加工,导致其营养物质的大量流失。大米中富含多酚类物质,主要以游离态和结合态两种形式存在,且结合态多酚远高于游离态多酚,其中绝大部分酚类物质因以结合态形式与细胞壁中的多糖、蛋白质和脂质等物质相结合而不易被人体消化吸收。此外,大米中的蛋白质往往缺少赖氨酸、蛋氨酸、苏氨酸和色氨酸,同时使得其他氨基酸不能被充分利用,从而使蛋白质的消化率和谷物的营养价值被降低,大米营养强化已成为我国大米加工业亟待解决问题。
锦带花桑黄孔菌Sanghunagporus weigelae,隶属于担子菌门Basidiomycota、蘑菇纲Agaricomycetes、锈革孔菌目Hymenochaetales、锈革孔菌科Hymenochaetaceae、桑黄孔菌属Sanghuangporus,广泛分布于我国贵州、湖北、湖南、浙江等地,主要寄主为多种阔叶树的活立木或倒木。锦带花桑黄是重要的药用资源,具有抑肿瘤、抗氧化等多种功效。桑黄多糖是桑黄发挥药理作用最主要的成分之一,它是一类有多个酮糖或醛糖经糖苷键连接成的高分子聚合物,广泛存在于子实体、菌丝体及发酵液中。研究表明,桑黄多糖具有抗氧化、抗肿瘤、抑菌、消炎、免疫调节等活性。黄酮类化合物是一种高含量的次生代谢物,主要存在于桑黄的菌丝和子实体中。人体所需的黄酮类化合物无法通过自身合成,只能从食物中获取,黄酮的生理功能对人类的健康有很大的作用。近年来,随着人们对人体健康的认识日益提高,利用食用菌作为原料进行黄酮提取的研究,不仅越来越受到重视,且逐渐成为当今营养学领域的一个重要课题。
发明内容
有鉴于此,本发明提供了一株锦带花桑黄(Sanghuangporus weigelae)S-LWZ20210623-1a CGMCC No.40572。
本发明还提供了一种组合物,包括锦带花桑黄(Sanghuangporus weigelae)S-LWZ20210623-1a CGMCC No.40572。该菌株已于2023年4月12日保藏在中国微生物菌种保藏管理委员会普通微生物中心,该中心简称为CGMCC,该菌株保藏号为CGMCC No.40572,该中心地址为北京市朝阳区北辰西路1号院3号。本发明还提供了一种组合物,包括锦带花桑黄(Sanghuangporus weigelae)S-LWZ20210623-1a CGMCC No.40572。
本发明最后提供了锦带花桑黄(Sanghuangporus weigelae)S-LWZ20210623-1a在制备大米菌粮中的应用。
本发明至少包括以下有益效果:
第一、本发明中锦带花桑黄接种大米发酵产物粗多糖含量显著提升;
第二、本发明中锦带花桑黄接种大米发酵产物,黄酮类化合物含量显著升高,如具有多种生物活性,如抗氧化、抗耳毒性、抗炎、抗动脉粥样硬化、抗高血压的橙皮素、木樨草素;
第三、本发明中锦带花桑黄接种大米发酵产物赖氨酸、苏氨酸、色氨酸、蛋氨酸含量均有不同程度的升高,使得大米中缺乏的人体必需氨基酸得到补充,蛋白质的营养价值提高。
具体实施方式
实施例1
1、菌株的获得、分离、鉴定
菌株于2021年6月23日采集自江西庐山国家级自然保护区,采用组织分离法从川榛(Corylus heterophylla)活树的枝条上生长的新鲜子实体中获得,命名为菌株S-LWZ20210623-1a。组织分离法时选取新鲜没有发霉或蛀虫的子实体,使用75%的酒精进行表面消毒,选取菌肉部分约0.5cm3厘米大小的组织,接入到马铃薯琼脂培养基(PDA)培养基表面,置于26℃下恒温培养10-15天。经纯化后,在26℃下培养10-15天。
采用裂解法、使用真菌基因组快速提取试剂盒提取S-LWZ20210623-1a的DNA,通过PCR扩增获得其ITS序列,其序列信息如序列表中SEQ ID NO.1所示,通过与NCBI数据库BLAST对比及参考桑黄属DNA条形码库构建系统发育树,结合形态学分析,确定该菌株鉴定为锦带花桑黄(Sanghuangporus weigelae)。
锦带花桑黄(Sanghuangporus weigelae)S-LWZ20210623-1a,已于2023年4月12日保藏在中国微生物菌种保藏管理委员会普通微生物中心,该中心简称为CGMCC,该菌株保藏号为CGMCC No.40572,该中心地址为北京市朝阳区北辰西路1号院3号。
2、母种培养基制备与菌种活化
母种培养基(PDA)制备配方及方法如下:马铃薯煮沸液300g/L、葡萄糖20g/L、蛋白胨5g/L、琼脂20g/L、磷酸二氢钾1g/L、七水硫酸镁0.5g/L,pH自然,搅匀后倒入锥形瓶中,121℃灭菌20min,温度降低到室温时,后加入0.1%的硫酸链霉素和氨苄青霉素溶液。
将步骤(1)获得的于低温保藏的锦带花桑黄(Sanghuangporus weigelae)S-LWZ20210623-1a转置于上述的母种培养基(PDA)培养基进行传代培养,置于培养箱中26℃培养10天,即得到活化好的锦带花桑黄母种。
3、大米菌粮制备:
固体培养基的制备:将大米清洗干净,置于蒸馏水浸泡24h,取出沥干表面水分,分装到15×30×5C的聚丙烯菌袋中,再加入大米重量40%的蒸馏水,用3.8cm海绵盖、4×125封口膜和孔径为0.22μm的微孔滤膜依次叠加封口,121℃灭菌2.5h后冷却至室温,得到大米固体培养基。
接种培养:将步骤(2)中已活化的菌株S-LWZ20210623-1a母钟接种到上述的大米固体培养基,用无菌打孔器制造出直径为6mm的菌块,接种到PDA培养基中,随即放入26℃的恒温培养箱中培养45天。
菌粮制备:菌丝长满整个培养基后,40℃下烘干至恒重,用研磨机将大米磨粉并过90目筛,即得所述大米功能菌粮,-80℃保存备用。
对比例1
对照组大米原粮的制备:将大米清洗干净,置于蒸馏水浸泡24h至籽粒变软,取出沥干表面水分,分装到15×30×5C的聚丙烯菌袋中,再加入大米重量40%的蒸馏水,用3.8cm海绵盖、4×125封口膜和孔径为0.22μm的微孔滤膜依次叠加封口,121℃灭菌2.5h后冷却至室温,得到大米固体培养基。将获得的固体培养基40℃下烘干至恒重,用研磨机将大米磨粉并过90目筛,即得对照组原粮,-80℃保存备用。
实验例1

按下列方法测定:

粗多糖含量的测定采用苯酚-硫酸法(左琦,杨海锋,邢增涛,韩烨,顾赛红.食用菌总糖含量测定方法的研究.食用菌学报,2021,15(4):57-61)
2、锦带花桑黄接种大米固态发酵产物中总酚含量的测定:
样品中总酚含量的测定,采用福林酚比色法(王伟科,陆娜,闫静,付立忠,宋吉玲,袁卫东,周祖法.生长年限对段木栽培鲍姆桑黄子实体营养、活性成分及抗氧化活性的影响.菌物学报,2021,40(3):668-680)
3、锦带花桑黄接种大米固态发酵产物中可溶性蛋白含量的测定:采用BCA蛋白法含量测试盒测定

检测实验例锦带花桑黄接种大米发酵产物中的粗多糖、总酚、可溶性蛋白的含量,具体数据见表1:

由上表可以看出,实施例的三个重复相比于对比例,粗多糖、总酚、可溶性蛋白含量明显高于对比例,说明接种锦带花桑黄后,对原粮中粗多糖、总酚、可溶性蛋白含量有显著的提升的作用。
非靶向检测锦带花桑黄接种大米发酵产物中的活性物质含量检测表明,黄酮类中的橙皮素(Hesperetin)具有多种生物活性,如抗氧化、抗耳毒性、抗炎、抗动脉粥样硬化、抗高血压等;木樨草素(Luteolin)具有抗氧化、抗炎和抗肿瘤等多种药理作用。
此外,锦带花桑黄能显著提升原粮中异黄酮类化合物染料木苷(Genistin)含量,异黄酮是一类具有广泛生物活性的植物雌激素,能清除生物体内的自由基,减缓或消除机体对外界刺激因素,如致癌物的吸收,已在抗衰老、抗癌、预防心血管疾病等方面成功开发。
较对比例而言,接种锦带花桑黄的发酵产物中的倍半萜类化合物石竹烯(β-Caryophyllene)含量显著提升,该化合物属于双环倍半萜烯类,具有抗炎、抗氧化、抗焦虑的药理活性;此外,圆柚酮(Nootkatone)、辣椒醇(Capsidiol)也均有显著的提升。
较对比例而言,接种锦带花桑黄的发酵产物中植物甾醇(β-Sitosterol)等具有显著的提升,而植物甾醇具有降胆固醇的作用,也进一步增强了菌粮的功能价值。
较对比例而言,接种锦带花桑黄的发酵产物中的赖氨酸类和色氨酸类均有显著提升,有效的补充了大米中缺少赖氨酸、蛋氨酸、苏氨酸和色氨酸,使得其他氨基酸进一步被充分利用,更重要的是强化了大米赖氨酸含量,赖氨酸是谷类蛋白的第一限制氨基酸,从而提升了蛋白质的消化率及原粮的营养价值。
所述赖氨酸类包括:β-丙氨酰-L-亮氨酸(β-Alanyl-L-lysine);N-乙酰-L-赖氨酸(N-Alpha-acetyllysine);N6-(L-1,3-二羧丙基)-L-赖氨酸(N6-(L-1,3-Dicarboxypropyl)-L-lysine)、蛋氨酸(L-Methionine)、苏氨酸(L-Threonine)。
所述色氨酸类包括:N-乙酰基-D-色氨酸(N-Acetyl-D-tryptophan)、色氨酸(Tryptophan)。。
