Abstract
本发明公开了一种嗜酸乳杆菌LA‑G80、两歧双歧杆菌BB‑G90的组合及在护肝方面的应用,涉及微生物的应用技术领域,该益生菌组合物对急性酒精性胃粘膜损伤具有保护作用,同时对于化学性肝损伤有保护作用,且该益生菌组合物对调整优化体内菌群结构有积极作用,对提高免疫力也有帮助。

本发明涉及微生物的应用技术领域,特别涉及一种嗜酸乳杆菌LA-G80、两歧双歧杆菌BB-G90的益生菌组合物及在护肝方面的应用。
背景技术
医学研究表明,体内酒精只有不足10%可经由尿液、呼吸、汗液及唾液等直接排出体外,90%以上的乙醇均需在肝脏进行代谢。肝脏对乙醇的代谢过程主要是氧化分解的过程。而这个过程,往往使得肝细胞受损。而保护肝细胞因类似化学性物质受损成为居多人事的诉求。
发明内容
有鉴于此,本发明目的在于提供一种嗜酸乳杆菌LA-G80、两歧双歧杆菌BB-G90的益生菌组合物及在护肝方面的应用,该益生菌组合物对化学性肝损伤的保护作用,同时该益生菌组合物对优化调整体内菌群结构,提高免疫力有积极作用。
为了实现上述发明目的,本发明提供以下技术方案 :
一种益生菌组合物,主要由嗜酸乳杆菌LA-G80和两歧双歧杆菌BB-G90复合而成,所述嗜酸乳杆菌LA-G80的分类名为嗜酸乳杆菌LA-G80(Lactobacillus acidophilus LA-G80),保藏地址为中国典型培养物保藏中心,保藏编号为:CCTCC NO:M2013337,保藏时间为2013年7月16日;所述的两歧双歧杆菌BB-G90的分类名为两歧双歧杆菌BB-G90(Bifidobacterium bifidum BB-G90),保藏地点为:中国典型培养物保藏中心,保藏编号为:CCTCC NO:M2013194,保藏时间为:2013年5月9日。

嗜酸乳杆菌LA-G80,保藏编号CCTCC NO:M2013337,典藏时间,2013年7月16日;保藏单位为:中国典型培养物保藏中心,地址:中国武汉,武汉大学。
嗜酸乳杆菌LA-G80的培养方法为:在一定培养基的条件下,进行相对厌氧培养。培养温度为35-39℃。优选37-38℃。其中三角瓶采用静置培养的方式,添加质量体积分数为0~0.6%的碳酸钙在培养基中,培养过程不控制pH值。发酵罐采用通入非氧气不活泼气体(如纯度在99%以上的氮气等等),并保持罐压为0.02-0.1MPa。优选0.03-0.08MPa罐压条件下培养。发酵起始pH值控制在6.8±0.2范围内,后期发酵过程的pH值控制在5.2±0.2范围内,偏酸时采用碱液(如20%~25%的氢氧化钠或氨水中的一种或二种)流加控制,使得pH值控制在5.2±0.2范围内。在菌体繁殖生长的稳定期初期,结束发酵。
嗜酸乳杆菌LA-G80的发酵罐用基础培养基:葡萄糖,2~6%;牛肉浸粉,0.5~1%;蛋白胨,0.6~1%;酵母膏,0.5~1.5%;无水乙酸钠,0.5%;磷酸氢二钾,0.2%;柠檬酸氢二铵,0.2%;吐温80,0.1%;七水硫酸镁,0.025~0.05%;一水硫酸锰,0.005~0.01%,L半胱氨酸盐酸盐0.03~0.08%,碳酸钙,0.3~0.8%,其余为水,以上均按质量分数计。
发酵结束后,将发酵液降温,然后收集菌体,通常可采用离心的方法获得菌体。
菌体的基础保护剂为20%脱脂乳粉+5%蔗糖。在考虑规避过敏性物料的情况下,采用海藻糖,菊粉、低聚果糖、蔗糖等非乳基及过敏性原料的组合物作为菌体保护剂。
将保护剂与菌体混合均匀,得到乳化液。
将乳化液放入低温冷冻真空干燥设备(冻干机)中,进行真空低温冷冻干燥。获得菌饼。
菌饼经过粉碎后,获得菌粉。菌粉的细度可通过调整粉碎机的筛网孔大小获得。

两歧双歧杆菌BB-G90的基础培养基:乳糖,2~5%;葡萄糖0~2%;牛肉浸粉,0.6~1.2%;蛋白胨,0.6~1.2%;酵母膏,0.5~1.5%;大豆蛋白栋,0.1~0.5%;无水乙酸钠,0.5%;磷酸氢二钾,0.2%;柠檬酸氢二铵,0.2%;L-半胱氨酸盐酸盐,0.06~0.12%,吐温80,0.1%;七水硫酸镁,0.04~0.08%;一水硫酸锰,0.01~0.03%,碳酸钙,0~0.5%;其余为水,以上均按质量分数计。

实验表明:本发明提供的嗜酸乳杆菌LA-G80、两歧双歧杆菌BB-G90复合益生菌组合物能明显降低中毒小鼠血清谷丙转氨酶GPT、谷草转氨酶GOT,明显减轻肝脏病变。对急性酒精性胃粘膜损伤具有保护作用。同时对长期慢性饮酒的肠道菌群有优化的作用。对饮酒者人体的粪便pH值有下降作用。有利于减少肠道内毒素进入肝脏,从而有利肝脏健康。
总结:该益生菌组合物对急性酒精性胃粘膜损伤具有保护作用,同时对于化学性肝损伤有保护作用,且该益生菌组合物对调整优化体内菌群结构有积极作用。

以下公开本发明的一些实施例,本领域技术人员可以根据本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。
实施例1:制备嗜酸乳杆菌LA-G80菌粉和两歧双歧杆菌BB-G90菌粉
(一)制备嗜酸乳杆菌LA-G80菌粉,获取来源:广西巴马地区健康老人。
嗜酸乳杆菌LA-G80,保藏编号CCTCC NO:M2013337,典藏时间,2013年7月16日;
嗜酸乳杆菌LA-G80的培养方法为:在一定培养基的条件下,进行厌氧培养。培养温度为37.5±0.5℃。其中三角瓶采用静置培养的方式,添加0.5%的碳酸钙在培养基中,培养过程不控制pH值。发酵罐采用通入氮气,并保持罐压为0.05±0.02MPa罐压条件下培养。发酵起始pH值为6.86,后期发酵过程的pH值控制在5.2±0.2范围内,偏酸时采用碱液(质量分数25%的氢氧化钠)流加控制,使得pH值控制在5.2±0.2范围内。在菌体繁殖生长的稳定期初,结束发酵。
培养基:葡萄糖,4%;牛肉浸粉,1.0%;蛋白胨,1.0%;酵母膏,1.0%;无水乙酸钠,0.5%;碳酸钙,0.5%;磷酸氢二钾,0.2%;柠檬酸氢二铵,0.2%;吐温80,0.1%;七水硫酸镁,0.06%;一水硫酸锰,0.02%;L-半胱氨酸盐酸盐,0.05%;其余为水,以上均按质量体积分数计。
培养基经115℃,15分钟灭菌,降温后调pH值为6.86,接种后在37-38℃条件下发酵培养。
发酵结束后,将发酵液降温至8℃,然后采用加速沉降技术收集菌体。
菌体的基础保护剂为20%wt脱脂乳粉+5%wt蔗糖+75%wt水。
将保护剂与菌体按照质量比5:1混合均匀,得到乳化液。
将乳化液放入低温冷冻真空干燥设备(冻干机)中,进行真空低温-80℃冷冻干燥,获得菌饼。
菌饼经过粉碎后,获得菌粉,菌粉的细度可通过调整粉碎机的筛网孔大小获得,本例实施时为过40目筛。
(二)制备两歧双歧杆菌BB-G90菌粉,获取来源:内蒙古健康成人。
两歧双歧杆菌BB-G90,保藏编号CCTCC NO:M2013194。典藏时间,2013年5月9日;
两歧双歧杆菌BB-G90的培养方法为:在一定培养基的条件下,进行厌氧培养。培养温度为37.5±0.5℃。其中三角瓶采用静置培养的方式,添加0.2%的碳酸钙在培养基中,培养过程不控制pH值。发酵罐采用通入氮气,并保持罐压为0.05-±0.03MPa罐压条件下培养。发酵起始pH值为7.02,后期发酵过程的pH值控制在5.5±0.2范围内,偏酸时采用碱液(25%的氢氧化钠)流加控制,使得pH值控制在5.5±0.2范围内。在菌体繁殖生长的对数期末,结束发酵。
种子培养基:乳糖,4%;牛肉浸粉,1.0%;蛋白胨,1%;酵母膏,1.0%;无水乙酸钠,0.5%;大豆蛋白栋,0.25%;碳酸钙,0.25%;磷酸氢二钾,0.2%;柠檬酸氢二铵,0.2%;吐温80,0.1%;L-半胱氨酸盐酸盐,0.1%;七水硫酸镁,0.06%;一水硫酸锰,0.02%;其余为水,以上均按质量体积分数计。
培养基经115℃,15分钟灭菌,降温后调pH值为7.02,接种后在37-38℃条件下发酵培养。
发酵结束后,将发酵液降温至8℃,然后收集菌体。
菌体的基础保护剂为20%wt脱脂乳粉+5%wt蔗糖+75%wt水。
将保护剂与菌体混合均匀,得到乳化液。
将乳化液放入低温冷冻真空干燥设备(冻干机)进行真空低温冷冻干燥。获得菌饼。
菌饼经过粉碎后,获得菌粉。菌粉的细度可通过调整粉碎机的筛网孔大小获得。本例实施时为过40目筛。
实施例2:制备嗜酸乳杆菌LA-G80和两歧双歧杆菌BB-G90组合物:
将以上制得的嗜酸乳杆菌LA-G80菌粉、两歧双歧杆菌BB-G90菌粉,各自用食品级原料抗性糊精进行标准化,使活菌数控制在2.5*1010 CFU/g以上。
将以上制得的嗜酸乳杆菌LA-G80菌粉、两歧双歧杆菌BB-G90菌粉活菌比例按照优选的1:1混合,并加入添加剂,混合均匀,总活菌数控制在5*109 CFU/g以上,具体为5.6*109CFU/g。
添加剂按质量分数计为脱脂奶粉25%、乳糖20%、低聚果糖15%、菊粉10%,低聚木糖6%、果胶2%作为复配的辅料与22%的菌粉混合后作为混粉制备成胶囊剂,胶囊的净含量为0.2~0.25g/粒。
实施例3:对化学性肝损伤的保护作用。
试验物品:实施例2中嗜酸乳杆菌LA-G80和两歧双歧杆菌BB-G90等复配的混粉。
试验组别:人体日推荐量0.42g/60kg(相当于100亿CFU),设 低、中、高 3个 剂量组,分别相当于人体推荐量的5倍、10倍和30倍,即0.035、0.07、0.21g/kg,另设正常对照组给予蒸馏水和四氯化碳肝损伤模型组。
试验方法:50只小白鼠分为5组,每组10只经口给予不同剂量复合益生菌粉 ,各剂量组给以样品,模型组和对照组给以蒸馏水,连续喂养 30天。第30天,各剂量组和模型组以四氯化碳染毒,口服四氯化碳24mg/kg,正常对照组除外。第31天摘眼球取血/血清,测定如下生化指标:谷丙转氨酶GPT,谷草转氨酶GOT,白/球细胞比例A/G。并取肝脏进行病理组织学检查。

由上表可见,样品各剂量组与对照相比,均无显著差异。说明建模成立。
测定谷丙转氨酶GPT,谷草转氨酶GOT,白/球细胞比例A/G等生化指标:具体如下表。剂量组,尤其是复合高剂量组明显降低血清谷丙转氨酶GPT、谷草转氨酶GOT。

正常对照组的谷丙转氨酶、谷草转氨酶含量明显低于CCl4模型组,存在显著性差异,说明模型成立。样品复合高剂量组与CCl4 模型组比较,谷丙转氨酶、谷草转氨酶明显降低,均存在显著性差异。
在肝功能检查时,白球比在正常的肝功能情况下,白蛋白要高于球蛋白,白球比正常范围在1.5至2.5之间波动。
当出现肝功能损害时,可能会出现白球比降低或者是白球比倒置,尤其是在肝功能衰竭,或者是肝硬化时,白蛋白产生减少,可以导致白球比值降低。
白球比例偏高可能是由于白蛋白增多、球蛋白减少,或者是两者都有变化造成的,
白蛋白是由肝脏合成的,对球蛋白有胶体保护作用,能够增强人体的免疫力和抵抗力,一般情况下,白蛋白越多,说明人体越健康。球蛋白是由机体的免疫器官产生的,球蛋白偏高说明免疫系统亢进,偏低说明免疫能力不足。
血清白蛋白反映肝脏的合成功能,慢性乙型肝炎、肝硬化和肝功能衰竭的患者可显示血清白蛋白的下降。随着肝损害加重,白蛋白/球蛋白比值会逐渐下降。
血清白蛋白、球蛋白和白球比是反应肝功能的重要指标。且这三者要综合来看。某个数值,或某几个数值异常,均可能属于异常情况。从上述试验数据,综合来看,随着复合剂量的增加,对谷丙转氨酶、谷草转氨酶含量趋向于正常化是正向相关的。并且白/球细胞比例也是逐步趋向于正常化。
对肝脏进行病理组织学检查,结果如下表:

正常对照组的病变积分明显低于CCl4模型组,存在显著性差异,说明模型成立。样品复合高剂量组与CCl4 模型对照组相比,病理积分明显下降,存在显著差异性。
结论:实验复合益生菌组合物能明显降低中毒小鼠血清谷丙转氨酶GPT、谷草转氨酶GOT,明显减轻肝脏病变。说明上述实验复合益生菌组合物对保护肝脏有一定的作用。
从实验数据来看,同剂量的条件下,复合菌株对化学性肝损伤的效果要更加优良。分析认为,可能是以下的原因造成。
一方面,共同的,双歧杆菌和嗜酸乳杆菌均可调节人体免疫力,从而减少甚至抑制炎症的发生。通过调整菌群,竞争性抑制肠杆菌科的有害菌在肠道内的增长。益生菌产生的短链脂肪酸可为肠道细胞提供适当的营养,修复肠道黏膜,预防细胞调亡并改善肠道上皮细胞的稳定性状态。从而维持肠道稳定的通透性。这些功能缓解了病原体对肠道的侵扰。这些益生菌可以限制革兰氏阴性细菌的增长和杀伤有害细菌,防止病原体的转移。控制有害菌群细菌数量可以减少细菌来源的有毒物质进入肝脏。如乙醇、苯酚等,这些有毒物质可以引起肝脏损伤。从而减少及缓解酒精性肝损伤等现象的发生。这些益生菌在肠道内代谢产生大量乳酸、细菌素和过氧化氢,缓解和减少一些毒素产生高亲和力的自由基,进而导致的细胞脂类、蛋白和 DNA发生过氧化反应,而损伤肝细胞。
另一方面,嗜酸乳杆菌在人体整个胃肠道中,可以释放有利于双歧杆菌等其他益生菌生长的物质,增加大肠内益生菌的数量、以及增强它们的活力。也可以间接促进超氧化物歧化酶(SOD)和较强的谷胱甘肽过氧化物酶活力。减少相关细胞的过氧化,从而影响细胞及人体健康。
嗜酸乳杆菌具有抑制有害细菌生长的能力,能促进肠道内有益菌的增值和提高相关活力,能杀死有害菌,不仅能调节体内菌群失调,还能减少体内毒素的生成,从而减轻肝脏解毒负担,这样就是保护了肝脏。
另外,一些毒素会影响正常的代谢过程,产生高亲和力的自由基,进而导致细胞脂类、蛋白和 DNA发生过氧化反应,导致肝细胞损伤。从而可能诱导炎症反应,然后通过激活相关细胞使其释放肿瘤坏死因子(TNF-a),炎症和免疫反应促进促炎因子的产生,会导致肝损伤病人肝细胞死亡。
还有嗜酸乳酸杆菌菌株可以减轻相关的肝损伤,其机制可能是通过抑制NK细胞和特定细胞的活化以及促进TNF-a的表达并作用于TNFR2通路,从而对肝脏炎症起保护作用。
同时,乳杆菌与双歧杆菌是人体内两大主要的益生菌群。通常条件下,双歧杆菌的数量级别要超过乳酸杆菌类的。
肝脏细胞的活动特别需要维生素B族,以维持酶代谢的正常。维生素B能协助肝脏解毒并能够加快酒精在人体内的代谢,帮助酒精排出体外。 两歧双歧杆菌生成维生素B1能力较强。维生素B1在舒缓头痛、提振精神、减轻压力、防治高血压、防治肝病、强化肠胃等方面有着积极的作用。
因此,我们认为,在双歧杆菌和嗜酸乳杆菌共同作用下,有着相同的作用模式的前提下,又各自有着个性化的模式。在这个多重因素的作用下,达到了一定的叠加效应。从而表现出更加优良的效果。
实施例4:调整体内菌群的作用。
一、检测方法:测定体内常规益生菌群的变化
雄性大鼠50只,体重160~200g,随机平均分为5组(每组10只)。对照组:自由摄入标准颗粒饲料,自由饮水。慢性酒精组:进食同对照组,每日饮水以25%酒精替代,持续3月。慢性酒精恢复组,进食同对照组,每日饮水以25%酒精替代持续3个月后改为饮水3个月。急性酒精组:进食同对照组,每日给予25%酒精灌胃,连续3d,每日乙醇入量为8g/ kg 体重,分2次灌入。益生菌恢复组,进食同对照组,每日饮水以25%酒精替代持续3个月后改为饲喂含嗜酸乳杆菌LA-G80和两歧双歧杆菌BB-G90的复合菌粉及饮用水(添加益生菌0.5亿CFU每天每只)3个月。所有动物采用单笼饲养在清洁级动物房,处死大鼠前均禁食不禁饮12h。
二、肠粘膜菌群标本收集
颈椎脱白法处死大鼠后固定于解剖板上,常规消毒开腹,无菌纱布覆盖切口。距回盲瓣10cm处切下回肠段约5cm置于无菌平皿中,纵行剖开肠管,生理盐水反复冲洗,玻片轻轻刮取粘膜约0.1g,置于0.9ml无菌磷酸缓冲液中。
三、细菌培养计数
1、将所取标本分别以无菌生理盐水梯度稀释为1x10-2、1×10-3、1×10-4“稀释液,各取20ul接种于乳酸杆菌选择性培养基上。
2、37℃±1℃条件下,厌氧培养72小时。
3、以细菌计数仪计数菌落数,结果用菌落形成单位(cfu )表示: CFU / ml =菌落数×50×稀释倍数。
结果如下:(取平均值)

由上表可见,无论是慢性饮酒组、急性饮酒组,其体内的乳酸菌群都会发生明显变化。而采用减少饮酒的自然恢复和用益生菌恢复后,都得到了一定的效果。其中益生菌补充组体内益生菌检测的数据最多。
另外,将实施例2制得的复合菌粉按照每人每日服用,测得服用上述复配菌粉组合物后,志愿者的粪便pH值有所下降。在志愿者身体状态自然且正常,排便状况和规律基本正常的条件下,与服用前比较,粪便的pH值数据下降的幅度在0.2~0.6之间。这也可能意味着人员的肠道菌群获得优化。或者说补充的益生菌发生了相应的作用。我们认为可能意味着人体肠道内的有害菌如一些革兰氏阴性菌有所减少,而益生菌有所增加,从而使得益生菌产生的有机酸类的物质有所增加。如乙酸、丙酸、丁酸等等。理论上来说,这也有利肠道毒素的减少。有利减少肠道黏膜的通透性,使得内毒素从肠道进入门静脉,再到肝脏的几率降低,数量减少,从而减少肝脏的负担,间接的减少了对肝脏的损伤。
基于以上居多因素,可能是一些益生菌制品及本组合物既能到达预期试验效果的原因。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (7)
1.一种益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,所述益生菌组合物包括嗜酸乳杆菌LA-G80和两歧双歧杆菌BB-G90,所述嗜酸乳杆菌LA-G80的分类名为嗜酸乳杆菌LA-G80(Lactobacillus acidophilus LA-G80),保藏地址为中国典型培养物保藏中心,保藏编号为:CCTCC NO:M2013337,保藏时间为2013年7月23日;所述的两歧双歧杆菌BB-G90的分类名为两歧双歧杆菌BB-G90(Bifidobacterium bifidumBB-G90),保藏地点为:中国典型培养物保藏中心,保藏编号为:CCTCC NO:M2013194,保藏时间为:2013年5月9日;
所述益生菌组合物中嗜酸乳杆菌LA-G80、两歧双歧杆菌BB-G90两种菌粉活菌数比例为1:1。
2.根据权利要求1所述的益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,所述益生菌组合物中,总活菌数在5*109/g以上。
3.根据权利要求1所述的益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,将所述益生菌组合物制备成粉剂、颗粒剂、片剂、丸剂、液体制剂或胶囊剂。
4.根据权利要求3所述的益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,将所述益生菌组合物制备成胶囊剂,所述胶囊净含量为0.2~0.25g/粒。
5.根据权利要求1所述的益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,所述益生菌组合物中添加有辅料;所述辅料为脱脂奶粉、乳糖、低聚果糖、菊粉、低聚木糖、果胶中的一种或多种。
6.根据权利要求1所述的益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,所述益生菌组合物的制备步骤为:
(1)制备嗜酸乳杆菌LA-G80菌粉;
(2)制备两歧双歧杆菌BB-G90菌粉;
(3)将步骤(1)和步骤(2)得到的菌粉按活菌数量比混合后添加辅料按照常规方法制备成食品、药品或保健品;
所述步骤(1)中菌粉的制备过程包括菌种活化、种子培养、发酵培养、菌体收集、保护剂配置、乳化、真空低温冷冻干燥、粉碎;
所述步骤(2)中菌粉的制备过程包括菌种活化、种子培养、发酵培养、菌体收集、保护剂配置、乳化、真空低温冷冻干燥、粉碎。
7.根据权利要求6所述的益生菌组合物在制备护肝保健品或治疗化学性肝损伤药品中的应用,其特征在于,所述步骤(1)和步骤(2)中菌粉均为将发酵后收集到的菌体与基础保护剂按照质量比1:5混合后置于温度为-80℃中冻干后粉碎而成,所述菌体的基础保护剂为20%wt脱脂乳粉+5%wt蔗糖+75%wt水。
