Abstract
本发明公开了一种ENO1抗体及其在治疗肿瘤中的应用,所述抗体不仅具有良好的结合活性,而且能够显著抑制各种肿瘤细胞的自我更新能力、侵袭能力和耐药能力,能够有效抑制肿瘤的生长并部分抑制其复发,本发明提供的抗体可用于已报道在细胞表面表达ENO1的多种肿瘤的诊断、预后和治疗。

技术领域
本发明属于生物医药技术领域,具体地,本发明涉及一种ENO1抗体及其在治疗肿瘤中的应用。
背景技术
α-烯醇化酶(烯醇化酶-1,ENO1)是一种多功能酶,属于烯醇化酶家族,基因定位于1号染色体,其蛋白相对分子质量为48kDa,广泛分布于多种组织中,主要存在于细胞质中,也存在于细胞膜和细胞核中,根据其细胞定位在多种生理及病理过程中发挥重要功能。ENO1功能与其在细胞内的定位有关,大多ENO1存在于细胞质中,作为糖酵解过程的限速酶调节能量代谢,可通过调节肿瘤细胞的代谢过程促进肿瘤的发生发展。位于细胞表面的ENO1可以作为纤溶酶原受体调节细胞外的纤溶活性,进而可激活纤维蛋白溶酶并加速细胞外基质降解从而促进肿瘤细胞的侵袭转移。越来越多的研究发现ENO1在细胞内外表达量的变化与很多疾病有关,尤其是与肿瘤的发生发展过程密切相关。
除了作为糖酵解酶的作用外,ENO1还表达在大多数肿瘤细胞的表面,其与尿激酶型纤溶酶原激活剂受体(UPAR)、整合素和某些细胞骨架蛋白形成多蛋白复合体,这些蛋白与肿瘤细胞的黏附、迁移和增殖有关。在肿瘤中,ENO1可以调节血管内和细胞周围的纤溶活性,促进细胞迁移和肿瘤转移。多项研究已经证实ENO1在多种恶性肿瘤组织中表达均显著上调,包括肝癌、肺癌、食管癌、胃癌、前列腺癌、结肠癌、肾癌、皮肤癌、神经胶质瘤、骨肉瘤、鼻咽癌、口腔癌、甲状腺癌、胰腺癌、宫颈癌、乳腺癌、卵巢癌、子宫癌等,ENO1可通过不同的作用途径,在肿瘤增殖、凋亡、血管生成、侵袭、转移和放化疗抗性中发挥重要作用,从而参与肿瘤的发生和发展中。
因此,ENO1有望成为肿瘤靶向治疗的理想分子靶点,筛选出能够用于有效治疗肿瘤的靶向ENO1的抗体具有重要意义。
发明内容
有鉴于此,本发明的目的在于提供一种ENO1抗体及其在治疗肿瘤中的应用。
为了实现上述目的,本发明提供了如下技术方案:
本发明的第一方面提供了一种抗ENO1抗体或其抗原结合片段。
进一步,所述抗体或其抗原结合片段包含重链可变区和轻链可变区;
所述重链可变区包含HCDR1、HCDR2、HCDR3;所述轻链可变区包含LCDR1、LCDR2、LCDR3;
所述HCDR1、HCDR2、HCDR3的氨基酸序列为如SEQ ID NO:8所示的重链可变区中的CDR1、CDR2、CDR3;所述LCDR1、LCDR2、LCDR3的氨基酸序列为如SEQ ID NO:16所示的轻链可变区中的CDR1、CDR2、CDR3;

在本发明中,所述HCDR1、HCDR2、HCDR3对应的氨基酸序列并不局限于如SEQ IDNO:1、SEQ ID NO:2、SEQ ID NO:3所示的氨基酸序列,所述LCDR1、LCDR2、LCDR3对应的氨基酸序列并不局限于如SEQ ID NO:9、SEQ ID NO:10、SEQ ID NO:11所示的氨基酸序列,采用任何CDR定义方案对如SEQ ID NO:8所示的重链可变区中的CDR1、CDR2、CDR3进行定义、对如SEQ ID NO:16所示的轻链可变区中的CDR1、CDR2、CDR3进行定义得到的HCDR1、HCDR2、HCDR3、LCDR1、LCDR2、LCDR3对应的序列均在本发明的保护范围内。

在本发明中,本发明第一方面提供的抗体或其抗原结合片段的功能性变体同样包含在本发明的保护范围内,其中,所述“功能性变体”是指与亲本抗体相比具有明显或显著序列同一性或相似性的抗体、多肽或蛋白质,所述功能性变体保留了亲本抗体的生物活性。功能性变体涵盖例如本文中所述抗体(亲本抗体)的以下变体,其与亲本抗体相比在类似程度上、在相同程度上或在更高程度上保留识别靶细胞的能力。参考亲本抗体,功能性变体可例如与亲本抗体在氨基酸序列方面具有至少约30%、50%、75%、80%、90%、98%或更高的同一性。

在一些实施方案中,可用于本发明的宿主细胞包括原核细胞和真核细胞。常用的原核宿主细胞的例子包括大肠杆菌、枯草杆菌等。用于表达抗体的宿主细胞包括大肠杆菌、酵母细胞、昆虫细胞、COS细胞、CHO细胞等。较佳地,该宿主细胞是真核细胞,更佳地是哺乳动物细胞,最佳的是HEK-293T细胞、CHO细胞或哺乳动物来源的免疫细胞。
在一些实施方案中,所述宿主细胞通过如下方法制备得到:将本发明第二方面或第三方面所述的核酸分子或表达载体引入到宿主细胞中,所述引入的方法包括但不限于:物理方法、化学方法、生物方法。所述物理方法包括磷酸钙沉淀、脂质转染法、粒子轰击、微注射、电穿孔;所述化学方法包括胶体分散系统、基于脂质的系统;所述胶体分散系统包括大分子复合物、纳米胶囊、微球、珠;所述基于脂质的系统包括水包油乳剂、胶束、混合胶束、脂质体;所述生物方法包括DNA载体、RNA载体、慢病毒载体、痘病毒载体、单纯疱疹病毒载体、腺病毒载体、腺相关病毒载体。
在一些实施方案中,所述引入的方法可以通过各种合适的方式将前面所述的核酸分子或所述的载体引入到宿主细胞中,并不局限于本发明中所列举出的方法,例如磷酸钙转染、DEAE-葡聚糖介导的转染、显微注射、电穿孔、TALEN方法、ZFN方法、非病毒载体介导的转染(例如脂质体)或病毒载体介导的转染(例如慢病毒感染,逆转录病毒感染,腺病毒感染),以及其他用于转移入细胞的物理、化学或生物学手段,如转座子技术,CRISPR-Cas9等技术。

在一些实施方案中,合适的药物组合物的施用形式包括适用于肠胃外施用的形式,例如通过注射或输注,例如通过快速注射或连续输液、静脉内、可吸入或皮下形式。在产品用于注射或输注的情况下,其可采用在油性或水性媒介物中的悬浮液、溶液或乳液的形式并且其可含有配制试剂,诸如悬浮剂、防腐剂、稳定剂和/或分散剂。可替代地,根据本发明提供的抗体或其抗原结合片段可呈干燥形式,用于在使用之前用合适的无菌液体重构。还可制备适于在注射前溶解或悬浮于液体媒介物中的固体形式。
在一些实施方案中,本发明所述药物组合物的适合给药剂量根据制剂化方法、给药方式、患者/受试者的年龄、体重、性别、病态、饮食、给药时间、给药途径、排泄速度及反应灵敏性之类的因素而可以进行多种处方,熟练的医生通常能够容易地决定处方及处方对所希望的治疗有效的给药剂量。
本发明的第九方面提供了如下任一种方法:
(1)一种生产本发明第一方面所述的抗体或其抗原结合片段的方法,所述方法包括如下步骤:
①培养本发明第五方面所述的经工程改造的宿主细胞,从而获得含有所述抗体或其抗原结合片段培养物;
②从步骤①得到的培养物中分离或回收所述抗体或其抗原结合片段;

进一步,所述方法包括如下步骤:采用本发明第一方面所述的抗体或其抗原结合片段、本发明第六方面所述的试剂或本发明第八方面所述的试剂盒对受试者来源的待测样品进行检测,通过抗原抗体反应来检测待测样品中ENO1的存在情况,以诊断和/或辅助诊断受试者是否患有表达ENO1的疾病。
本发明还提供了一种治疗表达ENO1的疾病的方法。
进一步,所述方法包括如下步骤:给有需要的受试者施用有效量的本发明第一方面所述抗体或其抗原结合片段、本发明第五方面所述的经工程改造的宿主细胞、本发明第七方面所述的抗体药物偶联物和/或本发明第八方面所述的药物组合物。

下面结合具体实施例,进一步阐述本发明,仅用于解释本发明,而不能理解为对本发明的限制。本领域的普通技术人员可以理解为:在不脱离本发明的原理和宗旨的情况下可以对这些实施例进行多种变化、修改、替换和变型,本发明的范围由权利要求及其等同物限定。下述实施例中所使用的实验方法如无特殊说明,均为常规方法;下述实施例中所用的试剂、生物材料等,如无特殊说明,均可从商业途径得到。
实施例1抗ENO1单克隆抗体的筛选
1、ENO1蛋白的表达和纯化
(1)ENO1蛋白的表达:用接种环直接取冻存的大肠杆菌ENO-1/Rosetta,在LB培养基平板表面划线,于37℃培养16小时,摇床37℃、300rpm。挑取一个单菌落,转到3mL LB培养基中,37℃,280rpm,培养3-5小时,测得OD600=0.6-0.7。
(2)ENO1蛋白的纯化:Ni金属螯合亲和柱的挂Ni、预平衡处理,变性蛋白加入适宜体积Ni-NTA His-Bind Resins(3-5mg/mL柱容量,保守地可按2mg/mL估算,按蛋白含量估算),室温颠倒地结合1h,装柱,收集流穿液备检。取部分蛋白,确定洗涤和洗脱的pH值。采用0、20、50、500mM洗脱一次,直到无蛋白洗脱。所有洗脱液洗脱后立即调节pH至8.0,收集所有流穿液SDS-PAGE分析。
2、ENO1 mAb的制备
(1)动物免疫
将ENO1蛋白(100μg/mL)与弗氏完全佐剂等体积混合,然后经背部皮下多点注射雌性BALB/c小鼠;间隔三周后进行二次免疫,将ENO1蛋白(200μg/mL)与弗氏不完全佐剂等体积混匀,在BALB/c小鼠背部皮下多点注射;三周后进行三次免疫,与第二次免疫方案相同,三周后进行四次免疫,与第三次免疫方案相同,采集BALB/c小鼠的血液,应用间接ELISA法,测定血清中抗体滴度。冲击免疫为中(500μg/mL)ENO1静脉注射。
(2)BALB-C小鼠胸腺细胞(滋养细胞)
将小鼠从75%酒精中取出,用眼科剪剪开小鼠腹部的皮肤,暴露腹部取脾脏,取出完整脾脏,剪去其周围的脂肪或组织,拍照。将胸腺在100目筛网上用无齿镊轻轻研磨,用培养基冲洗细胞过筛网到大平皿中,获得胸腺细胞悬液,将细胞悬液转移至50mL离心管中。离心、洗涤:1200rpm,5min,用移液管吸弃上清,加入20mL ImEm培养液轻轻吹匀,重悬。
(3)免疫后小鼠脾细胞
用眼科剪剪开小鼠腹部的皮肤,暴露腹部取脾脏,用另一眼科剪剪开小鼠腹膜,取出完整脾脏,剪去其周围的脂肪或组织。将脾脏在100目筛网上用无齿镊轻轻摩擦,用培养基冲洗细胞过筛网到大平皿中,获得脾脏细胞悬液,将细胞悬液转移至50mL离心管中,取20μL细胞悬液进行细胞计数。
(4)SP2/0细胞培养
用移液管弃去DmEm-H培养液,加入10mL ImDm培养液浸泡,轻轻拍打培养瓶使细胞脱落,收集细胞悬液至50mL离心管,取20μL细胞计数,离心、洗涤:1200rpm,5min,用移液管吸弃上清,加入20mL培养液轻轻吹匀,重悬。
(5)脾细胞与SP2/0细胞混合
SP2/0的细胞悬液转移至已弃上清的脾细胞离心管中轻轻吹匀,重悬脾细胞与SP2/0混合:1200rpm,5min,离心,用移液管吸弃上清,将50mL离心管于掌中轻轻摩擦至底部细胞充分混合均匀,准备45℃温水于保温瓶中,用前加入些许冰块,调整温度至38℃,将50mL离心管至于温水中,缓缓加入1mL PEG,边加边轻轻摇匀,随后缓缓加入10mL ImDm培养液,合计5min。离心:1200rpm,5min,移液管吸弃上清,加入胸腺细胞悬液,轻轻混合均匀,1200rpm,5min,离心,用移液管吸弃上清,加入24mL ImDm培养液,重悬。将混合细胞与培养基(2mL细胞悬液+1mL HAT+2mL ImDm培养液+10mL HyClone高级进口血清+25mL 2.5%甲基纤维素),上下颠倒50mL离心管20min,至管内完全均匀。
(6)阳性克隆的筛选与克隆化
待克隆化的阳性融合细胞,可先转入24孔板扩大培养后再克隆,也可边扩大边从96孔板取样直接克隆。用吸管将细胞群吹打分散,制成悬液,按前述方法精确计数活细胞数。从96孔板直接取样克隆,计数取样只用1滴悬液,用另一支滴管加无血清培养液9滴作10倍稀释,计数。用HT培养液将细胞稀释至每毫升5、30和50个细胞各40mL。每种杂交瘤细胞用一块备好的含饲养细胞的96孔板,每个稀释度32孔,每孔加细胞悬液0.1mL,每孔应含0.5、3和5个细胞。由于计数和加样的误差,往往偏高或偏低。如从96孔板直接取细胞克隆时,应在克隆完后立即把孔中和稀释管中剩余细胞全部转入24孔中扩大培养,以备失败时重新克隆。在37℃,5%-7%培养箱中培养7天左右,镜检,标出所有板孔的细胞群落数。采样并进行抗体检测。将单克隆阳性孔细胞先转入24孔,后在培养瓶中扩大培养,然后冻存。共获得4株杂交瘤细胞株,分别命名为ENO1 mAb(17G12、12A11、21A3和23G5),随后,进行杂交瘤细胞的冻存。
(7)杂交瘤细胞的培养
杂交瘤细胞以含有12% FBS、2mmol/L L-谷氨酰胺、100U/mL青霉素及100μg/mL链霉素的DMEM-H完全培养液,在37℃、5% CO2,大批量培养杂交瘤细胞收集上清时,一般稳定传代15-20次后,需重新复苏一批新细胞;一般使用1:2比例传代,于传代后24h换液,48h后长满再传代。
(8)无血清杂交瘤上清的收集
杂交瘤扩大培养至T175瓶中,每次约30瓶;传代后第2天换液,48h后长到约75%-90%满后,移去完全培养基,改换无血清培养基约20mL/瓶;次日每天用1mol/L的无菌NaOH调节液体的PH至7.0-7.5左右,并添加新鲜无血清培养基2-3mL;待贴壁细胞逐渐减少,细胞活度下降至40%-50%时,3000rpm,10min离心,收集无血清杂交瘤上清。
(9)抗体纯化
将预装柱上面盖子旋开,用蠕动泵滴PBS赶走气泡,PBS平衡大约20个柱体积。上完样后取1mL的流穿于1.5mL的EP管中,用PBS洗杂,调流速至1mL/min(1mL柱子),洗杂至少5个柱体积,即5mL PBS,测OD280;管中加入75μL的1M Tris-HCl(pH 9.0)中和buffer。用0.1MGlycine-HCl(pH 2.7)洗脱buffer洗脱柱子,调流速1mL/min,接洗脱下来的液体,立即混合,使洗脱下来的抗体立即恢复PH值至7.0左右,每管测OD280。当OD值小于0.1时则可以停止接液。将洗脱下来的抗体混合,将混合后蛋白取10μL留样;将浓缩好的抗体放入20倍体积的预先预冷的PBS中4℃,搅拌透析,每次透析4h,透析3次;透析好的抗体离心,12000rpm,离心15min。0.22μm滤器过滤除菌,分装,以避免反复冻融。将收集到的ENO1 mAb进行抗体浓度测定及SDS-PAGE电泳分析。
(10)抗体亚类的检测
用PBS将捕获抗体稀释为1μg/mL,100μL/孔加入96孔酶标板,4℃孵育16h;弃去包被抗体,加入含1% BSA的PBS封闭液,300μL/孔,37℃封闭酶标板4h;弃去封闭液,用含0.05% Tween-20的PBS洗板三次,300μL/孔,1min/次×5次;按设计加入待测样品,100μL/孔,37℃孵育1.5h;分别加入HRP标记的各种类型二抗(HRP标记的羊抗鼠IgM、IgG1、IgG2a、IgG2b、IgG3、IgA、κorλ,1:2000稀释),100μL/孔,37℃孵育1h;PBS再洗涤2次;加入TMB显色,100μL/孔,37℃孵育30min;加入2M H2SO4,50μL/孔。酶标仪测定四种抗体(17G12、12A11、21A3、23G5)亚型,其对应的亚型为(IgG2b、IgG1、IgG1、IgG2b)。
(11)非竞争性ELISA检测抗ENO1单抗对重组ENO1的亲和力
采用非竞争性ELISA法(Beatty法)测定各单抗的亲和常数Ka。首先优化ENO1抗原包被浓度梯度和抗体系列稀释的梯度浓度,最终确定正式试验条件为:以不同浓度梯度的ENO1抗原(0.593、0.395、0.2633、0.1755、0.117、0.078、0.000g/mL)包被,采用四种单抗稀释浓度(32000、16000、8000、4000、2000、1000、500、250、125、62.5、31.2、15.6、7.8、3.9、1.95、0.98、0.5、0.25、0.12、0.06、0.03、0.0125、0ng/mL)。此条件下针对未标记的ENO1单抗进行测定,绘制抗体OD值为纵坐标,抗体浓度对数为横坐标的反应曲线,可以在各个ENO1包被浓度下均能得到较好的典型S型反应曲线。
3、抗ENO1单抗23G5测序
采用培养23G5杂交瘤细胞总RNA提取试剂盒进行RNA的抽提,提取的RNA溶液于-70℃保存。对提取的RNA进行反转录,末端修复,然后进行接头连接和PCR扩增纯化得到NGS文库,文库库检合格后用于上机测序。对构建好的文库进行高通量测序,每个样品测6G数据。二代高通量测序仪器测序得到的原始图像数据经base calling转化为序列数据,称之为raw data或raw reads,结果以FASTQ文件格式存储,包含reads的序列以及碱基的测序质量。对raw data进行数据过滤,从而获得clean data用于后续分析。使用Trinity-v2.11.0软件对Clean Reads进行无参组装。使用igblast进行比对分析,小鼠的免疫数据库来自IMGT。然后对fmt7文件进行整理和过滤。使用Change-O的子模MakeDb.py,ParseDb.py及ConvertDb.py进行分析。对数据进行分析整理得到单克隆抗体23G5轻链和重链的序列,并根据序列得到可变区氨基酸序列。
4、实验结果
ENO1蛋白的表达和纯化的SDS-PAGE实验证明筛选最佳的IPTG诱导浓度比例1:6000和诱导时间3h(见图1),0、500mM咪唑洗脱,循环上样3h(见图2)。抗体纯化的SDS-PAGE电泳结果见图3,经SDS-PAGE验证显示,ENO1 mAb纯化效果较好,无明显杂带,符合抗体重链和轻链大小。抗体亚类的检测结果见下表2。4个待检测抗体(23G5>12A11>21A3>17G12)对重组ENO1蛋白的亲和力结果见图4,结果显示,23G5对应的解离常数KD=3.729×10-9,21A3对应的解离常数KD=1.952×10-8,12A11对应的解离常数KD=6.034×10-9,17G12未作出“S”曲线,抗ENO1单抗对重组ENO1蛋白的亲和力:23G5>12A11>21A3>17G12。

实施例2抗ENO1单抗23G5对肝癌干细胞及肝癌治疗的体内外效果研究
1、单抗23G5对肝癌干细胞自我更新能力的影响
肿瘤干细胞的自我更新能力是以成球为主,本实施例研究了单抗23G5对肝癌干细胞自我更新能力的影响,将培养48h的肝癌Bel7402-V3亲本细胞消化后,以纯化的单抗23G51mg/mL为实验组,将细胞与其在37℃孵育2h,取500个细胞接种于sphere培养基中,14天后观察两组细胞成球数量及大小。未经单抗23G5处理的细胞对照组。
实验结果显示单抗23G5对Bel7402-V3细胞的成球抑制率达32.4%,能够显著抑制肝癌干细胞的自我更新能力,P<0.05,有统计学差异(见图5)。结果提示单抗23G5可能通过作用于肝癌干细胞而发挥了其抑制成球的作用。
2、单抗23G5对肝癌干细胞侵袭能力的影响
侵袭能力作为肿瘤干细胞的另一重要特征,是肿瘤转移、复发的重要相关因素。本实施例研究了单抗23G5对肝癌干细胞侵袭能力的影响,单抗23G5 1mg/mL为实验组,将细胞与其在37℃孵育2h后加入铺好Matrigel的Transwell上室中,下室加入完全培养基,培养24h后取出固定后染色进行观察。未经单抗23G5处理的细胞对照组。
实验结果显示经单抗23G5处理后的肝癌细胞的侵袭能力明显减弱,其对肝癌细胞侵袭的抑制率达30.7%,P<0.05,有统计学差异(见图6)。
3、单抗23G5对肝癌干细胞耐药能力的影响
鉴于耐药能力是肿瘤干细胞的一个重要特征,本实施例研究了单抗23G5对肝癌干细胞耐药能力的影响,将Bel7402-V3细胞以4000个/孔的数量接种96孔板,并加入抗体23G5(600μg/mL)于各孔中,培养24h后移去含抗体的培养基,每组细胞均用含0μg/mL、0.15μg/mL、0.3μg/mL、0.45μg/mL、0.6μg/mL、0.75μg/mL和0.9μg/mL共7种不同浓度顺铂的培养基培养细胞,48h后换含顺铂的培养基,5天后测定OD450值。未经单抗23G5处理的细胞对照组。
实验结果显示经单抗23G5处理后的肝癌细胞耐药能力明显下降,其IC50值为0.14μg/mL,而对照组的IC50值为0.27μg/mL,经单抗23G5处理的细胞的耐药能力明显低于对照组(见图7),结果表明单抗23G5能够显著降低肿瘤细胞耐药性。
4、单抗23G5对裸鼠人肝癌细胞移植瘤的体内治疗实验
将Bel7402-V3的sphere细胞接种至裸鼠体内,进行单抗23G5的体内治疗实验。实验分为PBS组(0.2mL)、低剂量抗体组(2.5mg/kg)、中剂量抗体组(5mg/kg)、高剂量抗体组(10mg/kg)、化疗组(顺铂0.6mg/kg)、化疗联合中剂量抗体组,共6组。每只小鼠每次腹腔注射0.2mL。接种细胞后开始治疗,每周治疗2次,一个月后结束治疗。
实验结果显示随着抗体23G5的剂量增加,其对移植瘤的抑制率也逐渐升高。在治疗结束一个月时,低、中、高剂量23G5抗体对于移植瘤体积的抑制率分别达到了8.4%,53.1%和67.4%。此外,从第28天到第31天的过程中,化疗组的移植瘤生长曲线已超过高剂量组,表明化疗组小鼠的瘤体积增长速率已经超过高剂量抗体组(见图8),提示化疗组的小鼠已进入肿瘤的复发阶段,而高剂量抗体组和化疗联合抗体组却没有显示出该现象。因此,该实验结果表明,单独的化疗药治疗肿瘤容易造成肿瘤的复发,而采用本发明所述的单抗23G5联合化疗药的方法可以有效抑制肿瘤的生长并部分抑制其复发。
实施例3抗ENO1单抗23G5对胃癌干细胞及胃癌治疗的体内外效果研究
1、单抗23G5对胃癌干细胞自我更新能力的影响
肿瘤干细胞的自我更新能力是以成球为主,本实施例研究了单抗23G5对胃癌干细胞自我更新能力的影响,以纯化的单抗23G5 1mg/mL为实验组,将胃癌细胞SNU-5与其在37℃孵育2h,取500个细胞接种于SNU-5的sphere培养基中,7天后观察两组细胞成球数量及大小。未经单抗23G5处理的细胞对照组。
实验结果显示单抗23G5对SNU-5的成球抑制率达30.6%(见图9),能够显著抑制胃癌干细胞的自我更新能力,P<0.001,有统计学差异。结果提示单抗23G5可能通过作用于胃癌干细胞而发挥了其抑制成球的作用。
2、单抗23G5对胃癌干细胞侵袭能力的影响
侵袭能力作为肿瘤干细胞的另一重要特征,是肿瘤转移、复发的重要相关因素。本实施例研究了单抗23G5对胃癌干细胞侵袭能力的影响,单抗23G5 1mg/mL为实验组,将细胞与其在37℃孵育2h后加入铺好Matrigel的Transwell上室中,下室加入完全培养基,培养24h后取出固定后染色进行观察。未经单抗23G5处理的细胞对照组。

3、单抗23G5对胃癌干细胞耐药能力的影响
鉴于耐药能力是肿瘤干细胞的一个重要特征,本实施例研究了单抗23G5对胃癌干细胞耐药能力的影响,以2000个/孔的数量接种96孔板,并加入抗体23G5(400μg/mL)于各孔中,培养24h后移去含抗体的培养基,每组细胞均用含0μg/mL、0.0625μg/mL、0.125μg/mL、0.25μg/mL、0.5μg/mL、1μg/mL和2μg/mL、4μg/mL和8μg/mL共8种不同浓度顺铂的培养基培养细胞,48h后换含顺铂的培养基,5天后测定OD450值。未经单抗23G5处理的细胞对照组。
实验结果显示经单抗23G5处理后的胃癌细胞的耐药能力明显下降,其IC50值为0.30μg/mL,而对照组的IC50值为0.92μg/mL,经单抗23G5处理的细胞生物耐药能力明显低于对照组(见图11),结果表明单抗23G5能够显著降低肿瘤细胞耐药性。
4、单抗23G5对裸鼠人胃癌细胞移植瘤的体内治疗实验
将SNU-5的sphere细胞接种至裸鼠体内,进行单抗23G5的体内治疗实验。实验分为PBS组(0.2mL)、低剂量抗体组(2.5mg/kg)、中剂量抗体组(5mg/kg)、高剂量抗体组(10mg/kg)、化疗组(顺铂0.6mg/kg)、化疗联合高剂量抗体组,共6组。每只小鼠每次腹腔注射0.2mL。接种细胞后开始治疗,每周治疗2次,两个月后结束治疗。
实验结果显示随着抗体的剂量增加,其对移植瘤的抑制率也逐渐升高。在治疗结束两个月时,低、中、高剂量23G5抗体对于移植瘤体积的抑制率分别达到了8.2%,31.1%和38.2%。此外,化疗组的抑制率40.1%。单抗联合化疗药的方抑制率60.2%(见图12)。提示化疗组的小鼠已进入肿瘤的复发阶段,而高剂量抗体组和化疗联合抗体组却没有显示出该现象。因此,该实验结果表明,单独的化疗药治疗肿瘤容易造成肿瘤的复发,而单抗联合化疗药的方法可以有效抑制肿瘤的生长并部分抑制其复发。
实施例4抗ENO1单抗23G5对骨肉瘤干细胞及骨肉瘤治疗的体内外效果研究
1、单抗23G5对骨肉瘤干细胞自我更新能力的影响
肿瘤干细胞的自我更新能力是以成球为主,本实施例研究了单抗23G5对骨肉瘤干细胞自我更新能力的影响,以纯化的单抗23G5 1mg/mL为实验组,将细胞与其在37℃孵育2h,取500个细胞接种于MG-63的sphere培养基中,14天后观察两组细胞成球数量及大小。未经单抗23G5处理的细胞对照组。
实验结果显示单抗23G5对MG-63细胞的成球抑制率达37.3%(见图13),能够显著抑制骨肉瘤干细胞的自我更新能力,P<0.05,有统计学差异。结果提示单抗23G5可能通过作用于骨肉瘤干细胞而发挥了其抑制成球的作用。
2、单抗23G5对骨肉瘤干细胞侵袭能力的影响
侵袭能力作为肿瘤干细胞的另一重要特征,是肿瘤转移、复发的重要相关因素。本实施例研究了单抗23G5对骨肉瘤干细胞侵袭能力的影响,单抗23G5 1mg/mL为实验组,将细胞与其在37℃孵育2h后加入铺好Matrigel的Transwell上室中,下室加入完全培养基,培养24h后取出固定后染色进行观察。未经单抗23G5处理的细胞对照组。
实验结果表明经单抗23G5作用后的骨肉瘤干细胞的侵袭能力明显减弱,其对MG-63细胞侵袭的抑制率达23.2%(见图14),P<0.001,有统计学差异。
3、单抗23G5对裸鼠人骨肉瘤细胞移植瘤的体内治疗实验
将MG-63的sphere细胞接种至裸鼠体内,进行单抗23G5的体内治疗实验。实验分为PBS组(0.2mL)、低剂量抗体组(2.5mg/kg)、中剂量抗体组(5mg/kg)、高剂量抗体组(10mg/kg)、化疗组(顺铂0.6mg/kg)、化疗联合高剂量抗体组,共6组。每只小鼠每次腹腔注射0.2mL。接种细胞后开始治疗,每周治疗2次,一个月后结束治疗。
实验结果显示随着抗体的剂量增加,其对移植瘤的抑制率也逐渐升高。在治疗期中低、高剂量23G5抗体对于移植瘤体积的抑制率分别达到了24.6%和67.2%。此外,化疗组的抑制率35.6%。单抗联合化疗药的抑制率71.2%(见图15),实验结果表明,单独的化疗药治疗肿瘤容易造成肿瘤的复发,而单抗联合化疗药的方法可以有效抑制肿瘤的生长并部分抑制其复发。

35.根据权利要求34所述的抗体药物偶联物,其特征在于,所述偶联物包括放射性核素、细胞因子、细胞毒素、酶、金纳米颗粒/纳米棒、纳米磁粒、病毒外壳蛋白、VLP、或其组合。
36.根据权利要求35所述的抗体药物偶联物,其特征在于,所述放射性核素包括131I、32P、89Sr、90Y、223Ra、125I、103Pd。
37.根据权利要求35所述的抗体药物偶联物,其特征在于,所述细胞因子包括IL-2、IL-3、IL-4、IL-5、IL-6、IL-9、IL-10、IL-12、IL-13、IL-14、IFN-γ、TNF-β、TNF-α、G-CSF、M-CSF。
38.根据权利要求35所述的抗体药物偶联物,其特征在于,所述偶联物包括烷化剂、抗代谢物、抗肿瘤抗生素、有丝分裂抑制剂、染色质功能抑制剂、抗血管生成剂、抗雌激素、抗雄激素、免疫调节剂。

40.根据权利要求38所述的抗体药物偶联物,其特征在于,所述抗代谢物包括甲氨喋呤、5-氟脲嘧啶、氟苷、5-氟脱氧尿嘧啶、卡培他滨、阿糖胞苷、氟达拉滨、6-巯基嘌呤、2-氯脱氧腺苷、5-氮杂胞苷、2,2-二氟脱氧胞嘧啶核苷、克拉屈滨、脱氧柯福霉素、喷司他丁。
41.根据权利要求38所述的抗体药物偶联物,其特征在于,所述抗肿瘤抗生素包括柔红霉素、阿霉素、去甲氧正定霉素、光神霉素、丝裂霉素C、戊柔比星、盐酸米托蒽醌、博来霉素、更生霉素、光辉霉素、甲基苄肼。
42.根据权利要求38所述的抗体药物偶联物,其特征在于,所述有丝分裂抑制剂包括紫杉萜、长春碱、紫杉醇、长春新碱、长春酰胺、长春瑞滨。
43.根据权利要求38所述的抗体药物偶联物,其特征在于,所述染色质功能抑制剂包括依立替康、依托扑沙、托泊替康、磷酸依托扑沙、鬼臼噻吩甙。
44.根据权利要求38所述的抗体药物偶联物,其特征在于,所述抗血管生成剂包括普啉司他、坦诺司他、伊洛马司他、丙亚胺、马马司他、巴马司他、CGS-27023A、溴氯哌喹酮、COL-3、新伐司他、BMS-275291、沙立度胺。
45.根据权利要求38所述的抗体药物偶联物,其特征在于,所述抗雌激素包括托瑞米芬、雷洛昔芬、它莫西芬、阿纳托唑、来曲唑、屈洛昔芬、奥多昔芬、依西美坦。
46.根据权利要求38所述的抗体药物偶联物,其特征在于,所述抗雄激素包括尼鲁米特、比卡鲁胺、安体舒通、氟他米特、非那司提、醋酸环丙氯地孕酮、西咪替丁。
47.根据权利要求38所述的抗体药物偶联物,其特征在于,所述免疫调节剂包括白介素、肿瘤坏死因子、干扰素、蘑菇多糖、西佐糖、罗喹美克、匹多莫特、甲氧聚乙二醇琥珀酰胺腺甙脱氨酶、胸腺肽制剂。
48.根据权利要求35所述的抗体药物偶联物,其特征在于,所述细胞毒素包括MMAE、DM1、Ozogamicin、Dxd、SN-38、MMAF、PBD、DM2、Amanitin、DM4、PNU-159682、IR700、PE-38、PE24、PE-T20、PE-T20-KDEL、PE4E、PE40。
49.根据权利要求34所述的抗体药物偶联物,其特征在于,所述抗体药物偶联物用于治疗表达ENO1的肿瘤;
所述表达ENO1的肿瘤为肝癌、胃癌、骨肉瘤。
50.一种用于检测ENO1蛋白的试剂盒,其特征在于,所述试剂盒包含权利要求25-33中任一项所述的试剂。
51.一种药物组合物,其特征在于,所述药物组合物包含权利要求1-3中任一项所述的抗体或其抗原结合片段、权利要求21-24中任一项所述的经工程改造的宿主细胞和/或权利要求34-49中任一项所述的抗体药物偶联物。
52.根据权利要求51所述的药物组合物,其特征在于,所述药物组合物还包含一种或多种药学上可接受的载体和/或辅料。
53.根据权利要求52所述的药物组合物,其特征在于,所述药物组合物用于治疗表达ENO1的肿瘤;
所述表达ENO1的肿瘤为肝癌、胃癌、骨肉瘤。
54.一种生产权利要求1-3中任一项所述的抗体或其抗原结合片段的方法,其特征在于,所述方法包括如下步骤:
(1) 培养权利要求21-24中任一项所述的经工程改造的宿主细胞,从而获得含有所述抗体或其抗原结合片段培养物;
(2) 从步骤(1)得到的培养物中分离或回收所述抗体或其抗原结合片段;
(3) 纯化步骤(2)得到的抗体或其抗原结合片段得到权利要求1-3中任一项所述的抗体或其抗原结合片段。
55.一种非诊断目的地检测ENO1蛋白的方法,其特征在于,所述方法包括如下步骤:
(1) 获取待测样品;
(2) 将步骤(1)收集的样品与权利要求1-3中任一项所述的抗体或其抗原结合片段、权利要求25-33中任一项所述的试剂接触;
(3) 检测抗体-抗原复合物的存在。
56.一种权利要求21-24中任一项所述的经工程改造的宿主细胞的制备方法,其特征在于,所述方法包括如下步骤:将权利要求18-20中任一项所述的表达载体引入到宿主细胞中。
57.根据权利要求56所述的方法,其特征在于,所述引入的方法包括脂质转染法、微注射、电穿孔、DNA载体、RNA载体、慢病毒载体、痘病毒载体、单纯疱疹病毒载体、腺病毒载体、腺相关病毒载体。
58.根据权利要求56所述的方法,其特征在于,所述宿主细胞为真核细胞或原核细胞。
59.根据权利要求58所述的方法,其特征在于,所述宿主细胞为免疫细胞。
60.根据权利要求59所述的方法,其特征在于,所述免疫细胞包括T细胞、B细胞、NK细胞、iNKT细胞、CTL细胞、树突状细胞、髓样细胞、单核细胞、巨噬细胞或其任意组合。
61.权利要求1-3中任一项所述的抗体或其抗原结合片段在非诊断目的地检测ENO1蛋白中的应用。
62.权利要求1-3中任一项所述的抗体或其抗原结合片段在制备用于检测ENO1蛋白的试剂或试剂盒中的应用。
63.权利要求1-3中任一项所述的抗体或其抗原结合片段、权利要求25-33中任一项所述的试剂或权利要求50所述的试剂盒在制备用于诊断和/或辅助诊断表达ENO1的肿瘤的产品中的应用。
64.权利要求1-3中任一项所述的抗体或其抗原结合片段、权利要求4-14中任一项所述的嵌合抗原受体、权利要求15-17中任一项所述的核酸分子或权利要求18-20中任一项所述的表达载体在制备用于治疗表达ENO1的肿瘤的经工程改造的宿主细胞中的应用;
所述表达ENO1的肿瘤为肝癌、胃癌、骨肉瘤。
65.权利要求1-3中任一项所述的抗体或其抗原结合片段、权利要求4-14中任一项所述的嵌合抗原受体、权利要求15-17中任一项所述的核酸分子、权利要求18-20中任一项所述的表达载体或权利要求21-24中任一项所述的经工程改造的宿主细胞在制备用于治疗表达ENO1的肿瘤的抗体药物偶联物中的应用;
所述表达ENO1的肿瘤为肝癌、胃癌、骨肉瘤。
66.权利要求1-3中任一项所述的抗体或其抗原结合片段、权利要求4-14中任一项所述的嵌合抗原受体、权利要求15-17中任一项所述的核酸分子、权利要求18-20中任一项所述的表达载体、权利要求21-24中任一项所述的经工程改造的宿主细胞或权利要求34-49中任一项所述的抗体药物偶联物在制备用于治疗表达ENO1的肿瘤的药物中的应用;
所述表达ENO1的肿瘤为肝癌、胃癌、骨肉瘤。
