背景技术
在美国,胰腺导管腺癌(PDAC)是癌症死亡的第四主导原因,显 示出引起死亡的快速临床进程。一旦诊断后,PDAC的存活期中值为6 个月,5年存活率仅为3%(Li等,Lancet363:1049-1057(2004))。
由于化疗和放疗仅具有适度益处,并且手术只能在20%的患者中 进行,因此允许手术切除的早期检测为更长的存活期提供了最佳希望 (Yeo等,Ann Surg 222:580-588(1995);讨论588-592)。事实上,在 高危患者组(例如遗传性癌症综合征、慢性胰腺炎和新发糖尿病)中 检测PDAC或高等级前兆,代表了癌症诊断方法中关键的尚未满足的 需求(Brentnall等,Ann.Intern.Med.131:247-255(1999);Canto等,Clin.Gastroenterol.Hepatol.2:606-621(2004))。
血清CA-19-9是一种临床上使用的生物标志物;然而,它缺乏检 测早期PDAC所需的灵敏度(Goggins,J.Clin.Oncol.23:4524-4531 (2005))。此外,腹部横截面成像已被证明对在高危患者中检测早期 PDAC来说不可靠(Pelaez-Luna等,Am J Gastroenterol 102:2157-2163 (2007))。
因此,在本医学领域中,鉴定生物标志物以用于开发作为诊断试 剂的结合配体例如用于检测肿瘤前/早期侵入性病变的成像探针以及用 于治疗,具有高优先级。
发明内容
本公开的各个方面涉及下述认知,即临床上有用的基于抗体的药 剂例如抗体药物偶联物(ADC)的成功开发,受到所述药剂对其靶的 特异性和选择性限制。网蛋白-1以一种有用的生物标志物,可用于各 种不同的癌症,包括卵巢癌、食道癌和头颈鳞状细胞癌以及胰腺导管 腺癌。与抗体靶例如被本妥昔单抗瑞他汀(Brentuximab vedotin)靶向 的CD30和被阿杜曲妥珠单抗美坦新(Ado-trastuzumab Emtansine)靶 向的Her2形成对比,网蛋白-1是特别有用的靶,因为它在某些癌细胞 (例如胰腺导管腺癌细胞、卵巢癌细胞等)中专一地存在于细胞表面 上,从而提供了精妙的特异性和选择性。因此,在某些实施方式中,本公开涉及特异性结合到癌细胞表面上的网蛋白-1的抗体和抗原结合 片段及其使用方法。在某些实施方式中,本公开所描述的抗网蛋白-1 抗体与表达网蛋白-1的细胞的结合引起所述细胞死亡(例如引发凋 亡)。

具体实施方式
结合网蛋白-1的抗体
本公开提供了结合癌细胞表面上的网蛋白-1的抗体和抗原结合片 段。本公开的单克隆抗体可以是鼠类、人源化或嵌合的,或采取其他 形式。本公开的抗体以及本公开的抗体的生产和鉴定方法的详细描述 被提供在本文中。
网蛋白-1是一种高分子量蛋白(500kDa),其除了将细胞骨架锚 定到细胞质膜和核膜之外,还将中间纤维连接到微管和微丝(综述在 Sonnenberg,等,Exp Cell Res 313:2189-2203(2007)中)。
通常,在正常的胰腺导管细胞中网蛋白-1的水平低,但在具有某 些癌症的细胞(例如先兆胰腺上皮内瘤变(PanIN)、胰腺导管腺癌细 胞(PDAC)、卵巢癌细胞等)中它的表达被上调。网蛋白-1在正常成 纤维细胞中表现出独特的细胞质和核定位,然而在具有某些癌症的细 胞(例如PDAC)中观察到在细胞膜上的异常表达。在自体免疫病症副 肿瘤天疱疮和相关的淋巴组织增生性瘤变Castleman病中,也已观察到 网蛋白-1的改变的亚细胞定位(Aho等,J Invest Dermatol 113:422-423 (1999))。网蛋白-1在信号转导中也起重要作用。因此,在具有某些癌 症的细胞(例如先兆胰腺上皮内瘤变(PanIN)、胰腺导管腺癌细胞(PDAC)、卵巢癌细胞等)中,网蛋白-1可能对调控与癌发生相关的 细胞迁移、极性和能量代谢的信号转导途径有影响。因此,在某些实 施方式中,本公开提供了结合癌细胞表面上的网蛋白-1的抗体和抗原 结合片段。
在某些实施方式中,抗体,也被称为免疫球蛋白,是由两条各约 25kDa的轻(L)链和两条各约50kDa的重(H)链构成的四聚体糖基 化蛋白质。在抗体中可以存在两种类型的轻链,被称为λ和κ。取决于 重链的恒定结构域的氨基酸序列,免疫球蛋白可以被分配到5个主要 类别:A,D,E,G和M,它们中的几类可以被进一步分成亚类(同 种型),例如IgG1、IgG2、IgG3、IgG4、IgA1和IgA2。每条轻链通常 包括N-端可变(V)结构域(VL)和恒定(C)结构域(CL)。每条重 链通常包括N-端V结构域(VH)、3或4个C结构域(CH1-3)和铰 链区。最靠近VH的CH结构域被命名为CH1。VH和VL结构域由被称为 构架区的4个序列相对保守的区域(FR1、FR2、FR3和FR4)构成, 这些构架区为3个高变序列区域(互补决定区,CDR)形成支架。CDR 含有大多数负责抗体与抗原的特异性相互作用的残基。CDR被称为CDR1、CDR2和CDR3。因此,重链上的CDR组分被称为CDRH1、 CDRH2和CDRH3,而轻链上的CDR组分被称为CDRL1、CDRL2和 CDRL3。CDR通常是指Kabat CDR,如《免疫学重要的蛋白质的序列》 (Sequences of Proteins ofImmunological Interest,US Department of Health and Human Services(1991),Kabat等主编)中所述。表征抗原结 合位点的另一个标准是参照如Chothia所述的高变环。参见例如Chothia, D.等,(1992)J.Mol.Biol.227:799-817;和Tomlinson等,(1995)EMBO J.14:4628-4638。另一个标准是由Oxford Molecular的AbM抗体建模软 件所使用的AbM定义。总的来说,参见例如“抗体可变结构域的蛋白 质序列和结构分析”(Protein Sequence andStructure Analysis of Antibody Variable Domains),在《抗体工程实验室手册》(Antibody Engineering Lab Manual,Duebel,S和Kontermann,R.主编, Springer-Verlag,Heidelberg)中。参照Kabat CDR描述的实施方式可以 可替选地使用参照Chothia高变环或AbM定义的环或任何这些方法的 组合的类似描述的关系来进行。

本公开的抗网蛋白-1抗体可以任选地包含抗体恒定区或其部分。 例如,VL结构域可以在其C-端末端附连到轻链恒定结构域如Cκ或Cλ。 同样地,VH结构域或其部分可以附连到重链如IgA、IgD、IgE、IgG 和IgM以及任何同种型亚类的全部或一部分。抗体可以包括适合的恒 定区(参见例如Kabat等,《免疫学重要的蛋白质的序列》(Sequences of Proteinsof Immunological Interest,No.91-3242,National Institutes of HealthPublications,Bethesda,Md.(1991))。因此,在本公开范围内的 抗体可以包含与本领域中已知的恒定区组合的VH和VL结构域或其抗 原结合部分。在某些实施方式中,本公开的抗网蛋白-1抗体包含的重 链恒定区包含由SEQ ID NO:4、14、26、36、48、58、70或80表示 的序列。
在某些实施方式中,所述VH和/或VL结构域可以被恢复到种系序 列,例如使用常规的分子生物学技术将这些结构域的FR突变,以匹配 由种系细胞产生的FR。在其他实施方式中,所述FR序列仍然与共有 种系序列不同。
在某些实施方式中,抗网蛋白-1抗体或抗原结合片段可以包括或 不包括所述抗体的构架区,例如如SEQ ID NO:6、8、10、12、17、 19、21、23、28、30、32、34、39、41、43、45、50、52、54、56、 61、63、65、67、72、74、76、78、83或85所示的。在某些实施方式 中,抗网蛋白-1抗体是鼠类抗体。在某些实施方式中,抗网蛋白-1抗 体是嵌合或人源化抗体。
应该认识到,在某些实施方式中,本公开设想了所述抗体的重链 可变区和轻链可变区的氨基酸和核酸序列的变体(例如同源物)。“同 源性”是指两个多核苷酸或两个多肽组成部分之间的同一性百分比。 术语“实质性同源性”在指称核酸或其片段时,表明当与另一个核酸 (或其互补链)最适比对并进行适当的核苷酸插入或删除时,所述比 对的序列存在约90至100%的核苷酸序列同一性。例如,在某些实施 方式中,享有实质性同源性的核酸序列具有至少90%、至少91%、至 少92%、至少93%、至少94%、至少95%、至少96%、至少97%、至 少98%、至少99%的序列同一性。当指称多肽或其片段时,术语“实 质性同源性”表明当与另一个多肽最适比对并适当添加间隙、插入或 删除时,所述比对的序列存在约90至100%的核苷酸序列同一性。术 语“高度保守的”意味着至少80%的同一性,优选为至少90%的同一 性,更优选为超过97%的同一性。例如,在某些实施方式中,高度保 守的蛋白质享有至少85%、至少90%、至少91%、至少92%、至少93%、 至少94%、至少95%、至少96%、至少97%、至少98%、至少99%的 同一性。在某些情况下,高度保守可以是指100%的同一性。同一性可 以由本领域技术人员通过例如使用本领域技术人员已知的算法和计算机程序容易地确定。
在某些实施方式中,本公开的抗网蛋白-1抗体可以以高亲和力结 合网蛋白-1,例如以低于10-7M、10-8M、10-9M、10-10M、10-11M或 更低的Kd。例如,抗网蛋白-1抗体或其抗原结合片段可以以5pM至 500nM之间、例如50pM至100nM之间、例如500pM至50nM之间 的亲和力结合到网蛋白-1。本公开还包括与本文中描述的任何抗体竞争 与网蛋白-1的结合,并具有50nM或更低(例如20nM或更低、10nM 或更低、500pM或更低、50pM或更低或5pM或更低)的亲和力的抗 体或抗原结合片段。所述抗网蛋白-1抗体的亲和力和结合动力学可以 使用本领域中已知的任何方法、包括但不限于生物传感器技术(例如 OCTET或BIACORE)来测试。
当在本文中使用时,术语“抗体”通常是指免疫球蛋白。本文中 也提供了其维持或具有特异性结合能力的所有衍生物。抗体制备物可 以是单克隆或多克隆的。
当在本文中使用时,术语“抗体片段”或“抗原结合片段”是指 抗体的小于全长的任何衍生物。一般来说,抗原结合片段保留全长抗 体的至少显著部分的特异性结合能力。抗原结合片段的实例包括但不 限于Fab、Fab’、F(ab')2、scFv、Fv、dsFv、双体抗体、亲和体和Fd 片段。抗原结合片段可以通过任何适合的手段来生产。例如抗原结合 片段可以通过完整抗体的片段化酶法或化学法生产,或者它可以从编 码部分抗体序列的基因重组生产。可替选地,抗原结合片段可以完全 或部分合成生产。抗原结合片段可以任选地是单链抗体片段。可替选 地,片段可以包含通过例如二硫键连接在一起的多个链。抗原结合片 段也可以任选地是多分子复合物。有功能的抗原结合片段通常包含至 少约50个氨基酸,更通常地包含至少约200个氨基酸。
单链Fv(scFv)是仅由通过多肽连接物彼此共价连接的可变轻链 (VL)和可变重链(VH)构成的重组抗原结合片段。VL或VH任一 者可以是NH2-端结构域。所述多肽连接物可以具有可变的长度和组成, 只要所述两个可变结构域被桥接而没有严重的空间干扰即可。通常, 所述连接物主要包含甘氨酸和丝氨酸残基的区段,并且出于溶解性原 因间插有一些谷氨酸或赖氨酸残基。
双体抗体是二聚体scFv。双体抗体的组分通常具有比大多数scFv 更短的肽连接物,并且它们对缔合成二聚体表现出偏好性。
Fv片段是由通过非共价相互作用保持在一起的一个VH和一个 VL结构域构成的抗原结合片段。术语dsFv在本文中用于指称具有工 程化改造的分子间二硫键以使VH-VL对稳定的Fv。
F(ab')2片段是本质上与通过在pH 4.0-4.5下用胃蛋白酶消化而从 免疫球蛋白(通常是IgG)获得的片段等同的抗原结合片段。所述片段 可以重组生产。
Fab片段是本质上与通过将F(ab')2片段中联结两个重链片段的一 个或多个二硫桥还原而获得的片段等同的抗原结合片段。所述Fab’片 段可以重组生产。
Fab片段是本质上与通过用木瓜蛋白酶消化免疫球蛋白(通常是 IgG)而获得的片段等同的抗原结合片段。所述Fab片段可以重组生产。 所述Fab片段的重链区段是Fd片段。
亲和体是一种小的蛋白质,其包含起到抗原结合分子作用(例如 抗体模拟物)的三螺旋束。通常,亲和体长度约为58个氨基酸,并具 有大约6kDa的摩尔质量。具有独特结合性质的亲和体分子,通过将位 于参与母体蛋白结构域的结合活性的两个α-螺旋中的13个氨基酸进行 随机化来获得。结合所需靶蛋白的特异性亲和体分子可以从含有数十 亿个不同变体的池(文库)中,使用诸如噬菌体展示的方法来分离。
结合网蛋白-1的抗体的生产
大量方法可用于获得本公开的抗体或其抗原结合片段。例如,抗 体可以使用重组DNA方法来生产。单克隆抗体也可以按照已知方法, 通过杂交瘤的产生来生产(参见例如Kohler和Milstein,(1975)Nature, 256:495-499)。然后使用标准方法例如酶联免疫吸附测定法(ELISA) 和表面等离子体共振(例如OCTET或BIACORE)分析来筛选以这种 方式形成的杂交瘤,以鉴定产生与特定抗原特异性结合的抗体的一个 或多个杂交瘤。任何形式的特定抗原可用作免疫原,例如重组抗原、 天然存在的形式、其任何变体或片段及其抗原性肽(例如作为线性表 位或在支架内作为构象表位的本文中描述的任何表位)。制造抗体的 一种示例性方法包括筛选表达抗体或其片段(例如scFv)的蛋白质表 达文库,例如噬菌体或核糖体展示文库。噬菌体展示被描述在例如 Ladner等,美国专利号5,223,409;Smith(1985)Science 228:1315-1317; Clackson等,(1991)Nature,352:624-628;Marks等,(1991)J.Mol. Biol.,222:581-597;WO92/18619;WO 91/17271;WO 92/20791;WO 92/15679;WO 93/01288;WO 92/01047;WO 92/09690和WO 90/02809 中。
除了使用展示文库之外,可以使用所述特定抗原(例如网蛋白-1) 来免疫非人类动物例如啮齿动物如小鼠、仓鼠或大鼠。在一个实施方 式中,所述非人类动物是小鼠。
在另一个实施方式中,从非人类动物获得单克隆抗体,然后进行 修饰,例如使用本领域中已知的重组DNA技术制造成嵌合的。已描述 了各种不同的用于制造嵌合抗体的方法。参见例如Morrison等,Proc. Natl.Acad.Sci.U.S.A.81:6851,1985;Takeda等,Nature314:452,1985; Cabilly等,美国专利号4,816,567;Boss等,美国专利号4,816,397;Tanaguchi等,欧洲专利公布EP171496;欧洲专利公布0173494;英国 专利GB 2177096B。
抗体也可以通过本领域中已知的方法进行人源化。例如,具有所 需结合特异性的单克隆抗体可以被商业化人源化(Scotgene,Scotland; 以及Oxford Molecular,PaloAlto,Calif.)。完全人源化的抗体,例如在 转基因动物中表达的那些人源化抗体,在本发明的范围之内(参见例 如Green等,(1994)Nature Genetics 7,13;以及美国专利号5,545,806 和5,569,825)。

本发明的某些方面涉及用多核苷酸或载体转化的分离的细胞(例 如宿主细胞)。宿主细胞可以是原核或真核细胞。存在于所述宿主细 胞中的多核苷酸或载体可以被整合在所述宿主细胞的基因组中,或者 它可以维持在染色体外。所述宿主细胞可以是任何原核或真核细胞, 例如细菌、昆虫、真菌、植物、动物或人类细胞。在某些实施方式中, 真菌细胞是例如酵母属、特别是酿酒酵母(S.cerevisiae)种的细胞。 术语“原核的”包括可以用DNA或RNA分子转化或转染以表达抗体 或相应的免疫球蛋白链的所有细菌。原核宿主可以包括革兰氏阴性以 及革兰氏阳性细菌,例如大肠埃希氏杆菌(E.coli)、鼠伤寒沙门氏菌(S.typhimurium)、粘质沙雷氏菌(Serratia marcescens)和枯草芽孢 杆菌(Bacillussubtilis)。术语“真核的”包括酵母、高等植物、昆虫 和脊椎动物细胞,例如哺乳动物细胞如NSO和CHO细胞。取决于在 重组生产程序中使用的宿主,被所述多核苷酸编码的抗体或免疫球蛋 白链可以是糖基化的或者可以是非糖基化的。抗体或相应的免疫球蛋 白链也可以包括起始甲硫氨酸氨基酸残基。
在某些实施方式中,在载体已被并入到适合的宿主中之后,可以 将所述宿主维持在适合于所述核苷酸序列的高水平表达的条件下,并 且如果需要,可以随后进行免疫球蛋白轻链、重链、轻链/重链二聚体 或完整抗体、抗原结合片段或其他免疫球蛋白形式的收集和纯化;参 见Beychok,《免疫球蛋白合成的细胞》(Cells of Immunoglobulin Synthesis,Academic Press,N.Y.,(1979))。因此,多核苷酸或载体被引 入到所述细胞中,其进而生产所述抗体或抗原结合片段。此外,包含 上述宿主细胞的转基因动物、优选为哺乳动物,可用于所述抗体或抗 体片段的大规模生产。
所述转化的宿主细胞可以按照本领域中已知的技术在发酵罐中生 长并培养,以获得最适细胞生长。在表达后,可以按照本领域的标准 程序来纯化完整抗体、它们的二聚体、单独的轻链和重链、其他免疫 球蛋白形式或抗原结合片段,所述技术包括硫酸铵沉淀、亲和柱、柱 层析、凝胶电泳等;参见Scopes,《蛋白质纯化》(Protein Purification,Springer Verlag,N.Y.(1982))。然后可以从生长培养基、细胞裂解液或 细胞膜级分分离所述抗体或抗原结合片段。所述例如微生物表达的抗 体或抗原结合片段的分离和纯化可以通过任何常规手段,例如制备型 色谱分离和免疫分离例如涉及使用针对例如所述抗体的恒定区的单克 隆或多克隆抗体的那些分离。
本公开的多个方面涉及杂交瘤,其提供单克隆抗体的无限持续的 来源。当在本文中使用时,“杂交瘤细胞”是指从B成淋巴细胞与骨 髓瘤融合配偶体的融合衍生的永生化细胞,为了制备产生单克隆抗体 的细胞(例如杂交瘤细胞),选择抗体滴度已被确认的个体动物(例 如小鼠),并在最后一次免疫接种后2天至5天收获它的脾脏或淋巴 结,并将其中包含的抗体产生细胞与骨髓瘤细胞融合,以制备产生所 需单克隆抗体的杂交瘤。抗血清中的抗体滴度的测量可以例如通过将 后文中所描述的标记的蛋白质与抗血清反应,然后测量结合到所述抗 体的标记试剂的活性来进行。细胞融合可以按照已知方法来进行,例 如由Kochler和Milstein描述的方法(Nature 256:495(1975))。作为融 合促进剂,使用例如聚乙二醇(PEG)或仙台病毒(HVJ),优选地使 用PEG。
骨髓瘤细胞的实例包括NS-1、P3U1、SP2/0、AP-1等。待使用的 抗体产生细胞(脾细胞)的数目与杂交瘤细胞的数目的比例优选为约 1:1至约20:1。PEG(优选为PEG 1000-PEG6000)优选地以约10%至 约80%的浓度添加。细胞融合可以通过将两种细胞的混合物在约20℃ 至约40℃、优选地约30℃至约37℃下温浴约1分钟至10分钟,高效 地进行。
各种不同的方法可用于筛选产生抗体(例如针对本发明的肿瘤抗 原或自体抗体)的杂交瘤。例如,将所述杂交瘤的上清液添加到抗体 直接或与载体一起吸附到其上的固相(例如微孔板),然后添加用放 射性物质或酶标记的抗免疫球蛋白抗体(如果在细胞融合中使用小鼠 细胞,则使用抗小鼠免疫球蛋白抗体)或蛋白A,以检测针对结合到 固相的蛋白质的单克隆抗体。或者,将所述杂交瘤的上清液添加到吸 附有抗免疫球蛋白抗体或蛋白A的固相,然后添加用放射性物质或酶 标记的蛋白质,以检测针对结合到所述固相的蛋白质的单克隆抗体。
所述单克隆抗体的选择可以按照任何已知方法或其改良方法来进 行。通常,使用添加有HAT(次黄嘌呤、氨基蝶呤、胸腺嘧啶)的用 于动物细胞的培养基。可以使用任何选择和生长培养基,只要所述杂 交瘤可以生长即可。例如,可以使用含有1%至20%、优选地10%至 20%胎牛血清的RPMI 1640培养基、含有1%至10%胎牛血清的GIT培 养基、用于杂交瘤培养的无血清培养基(SFM-101,Nissui Seiyaku)等。 通常,所述培养在约5%CO2气体下,在20℃至40℃、优选地37℃下 进行约5天至3周,优选地1周至2周。杂交瘤培养物的上清液的抗 体滴度可以按照与上文对于抗血清中抗蛋白质抗体的抗体滴度所描述 的相同的方式来测量。
作为直接从杂交瘤培养物获得免疫球蛋白的可替选方案,可以使 用永生化杂交瘤细胞作为重排的重链和轻链基因座的来源,用于后续 的表达和/或遗传操作。重排的抗体基因可以从适合的mRNA反转录以 产生cDNA。如果需要,可以将所述重链恒定区与不同同种型的重链恒 定区交换或整个消除。可以连接所述可变区以编码单链Fv区。可以连 接多个Fv区以提供对于超过一种靶的结合活性,或者可以使用嵌合的 重链和轻链组合。可以使用任何适合的方法来克隆抗体可变区并产生 重组抗体。
在某些实施方式中,获得编码重链和/或轻链的可变区的适合的核 酸,并将其插入到可以转染到标准重组宿主细胞中的表达载体中。可 以使用各种不同的这些宿主细胞。在某些实施方式中,哺乳动物宿主 细胞对于高效加工和生产来说可以是有利的。可用于此目的的典型的 哺乳动物细胞系包括CHO细胞、293细胞或NSO细胞。所述抗体或抗 原结合片段的生产可以通过将修饰的重组宿主在适合于所述宿主细胞 生长和所述编码序列的表达的培养条件下培养来进行。所述抗体或抗 原结合片段可以通过从所述培养物分离它们来回收。所述表达系统可 以被设计成包括信号肽,以便得到的抗体被分泌到培养基中;然而,细胞内生产也是可能的。
本公开还包括至少编码本文中描述的抗体的免疫球蛋白链的可变 区的多核苷酸。在某些实施方式中,由所述多核苷酸编码的可变区包 含通过任一上述杂交瘤生产的抗体的可变区的VH和/或VL的至少一 个互补决定区(CDR)。

化合物(例如抗网蛋白-1抗体或包含抗网蛋白-1抗体和靶向剂的 抗体-药物偶联物)的有效量,也被称为治疗有效量,是足以改善与癌 症相关的至少一种不良反应(例如肿瘤生长、转移)的量。包含在药 物组合物中的治疗有效量在每种情况下取决于几种因素,例如待治疗 患者的类型、大小(size)和状况、目标给药方式、所述患者吸收目标 剂型的能力等。通常,包含在每一剂型中的活性剂的量提供约0.1至约 250mg/kg,优选地约0.1至约100mg/kg。本领域技术人员能够凭经验 确定适合的治疗有效量。

在某些情况下,本公开的化合物被制备在胶体分散系中。胶体分 散系包括基于脂类的系统,包括水包油乳液、胶束、混合胶束和脂质 体。在某些实施方式中,本公开的胶体系统是脂质体。脂质体是在体 内或体外可用作递送载体的人造膜容器。已显示,尺寸在0.2-4.0μm 范围内的大单层囊泡(LUV)可以包封大型大分子。
通过将脂质体偶联到特定配体例如单克隆抗体、糖、糖脂或蛋白 质,可以将所述脂质体靶向特定组织。可用于将脂质体靶向例如平滑 肌细胞的配体,包括但不限于:与平滑肌细胞特异性受体相互作用的 完整分子或其片段,例如与癌细胞的细胞表面标志物相互作用的抗体。 这些配体可以通过本领域技术人员公知的结合测定法容易地鉴定。在 其他实施方式中,通过将脂质体偶联到本领域中已知的抗体,可以将 它靶向到组织。

本公开的制剂在可药用溶液中给药,所述可药用溶液可以常规地 含有可药用浓度的盐、缓冲剂、防腐剂、相容性载体、佐剂和任选地 其他治疗性成分。
术语可药用载体意味着适合于给药到人类或其他脊椎动物的一种 或多种相容的固体或液体填充剂、稀释剂或包封物质。术语载体是指 所述活性成分与其组合以便于施用的天然或合成的有机或无机成分。 所述药物组合物的组分也可以与本公开的化合物以及彼此之间混合, 使得不存在实质性损害所需药物效率的相互作用。

所述化合物可以以自身(纯净)给药或采取可药用盐的形式给药。 当在医学中使用时,所述盐应该是可药用的,但不可药用的盐可以方 便地用于制备其可药用盐。这些盐包括但不限于从下述酸制备的盐: 盐酸,氢溴酸,硫酸,硝酸,磷酸,马来酸,乙酸,水杨酸,对甲苯 磺酸,酒石酸,柠檬酸,甲磺酸,甲酸,丙二酸,琥珀酸,萘-2-磺酸 和苯磺酸。这些盐也可以被制备成碱金属或碱土金属盐,例如羧酸基 团的钠盐、钾盐或钙盐。

治疗方法
本公开的多个方面涉及特异性结合到某些癌细胞表面上的网蛋白 -1的抗体的发现。在某些实施方式中,本公开所描述的抗网蛋白-1抗 体与某些癌细胞的结合引起所述细胞死亡(例如启动凋亡)。不希望 受到任何特定理论限制,本公开描述的抗体在某些实施方式中可用于 治疗以网蛋白-1的表面表达为特征的癌症。当在本文中使用时,“治 疗癌症”是指减少患者中癌细胞的数目,减缓患者中癌细胞的生长, 减少癌细胞在患者中的转移,并包括关于缓解癌症症状或提高患者寿 命的任何类型的响应。
以网蛋白-1的表面表达为特征的癌症的实例包括但不限于卵巢癌 细胞、食管癌细胞、头颈部鳞状细胞癌癌细胞或胰腺癌细胞(例如胰 腺导管腺癌(PDAC))。然而,应该认识到,其他癌症(例如肺癌、 膀胱癌、乳腺癌、食管癌、口腔癌、舌癌、牙龈癌、皮肤癌(例如黑素瘤、基底细胞癌、卡波斯肉瘤等)、肌肉癌、心脏癌、肝癌、支气 管癌、软骨癌、骨癌、胃癌、前列腺癌、睾丸癌、宫颈癌、子宫内膜 癌、子宫癌、结肠癌、结肠直肠癌、胃癌、肾癌、膀胱癌、淋巴瘤、 脾癌、胸腺癌、甲状腺癌、脑癌、神经元癌、间皮瘤、胆囊癌、眼癌 (例如角膜癌、葡萄膜癌、脉络膜癌、黄斑癌、玻璃体液癌等)、关 节癌(例如滑膜癌)、成胶质细胞瘤、白细胞癌(例如淋巴瘤、白血 病等)、遗传性非息肉性癌(HNPC)、结肠炎相关癌症等),可以使 用本公开描述的抗网蛋白-1抗体来治疗。癌症的其他实例是肉瘤(例 如骨肉瘤和卡波斯肉瘤)。
在某些方面,本公开提供了一种用于治疗癌症的方法,所述方法 包括向患有癌症的对象给药有效量的本公开所描述的抗体或组合物 (例如抗网蛋白-1抗体或包含抗网蛋白-1抗体的组合物)。在某些实 施方式中,所述对象是哺乳动物。在某些实施方式中,所述对象是人 类。
通常,本公开的抗体和药物组合物优选地含有可药用载体或赋形 剂,其适用于使所述化合物或混合物可以作为片剂、胶囊或丸剂口服 给药,或者可以肠胃外、静脉内、真皮内、肌肉内或皮下或透皮给药。
所述含有抗网蛋白-1抗体和/或其他化合物的药物组合物可以通 过用于药物给药的任何适合的途径给药。各种不同的给药途径是可用 的。所选的具体模式当然将取决于所选的具体药剂、待治疗的具体病 症和治疗效果所需的剂量。本公开的方法一般来说可以使用医学上可 接受的任何给药模式来实践,这意味着产生疗效而不引起临床上不可 接受的不良反应的任何方式。在本文中讨论了各种不同的给药模式。 对于在疗法中的使用来说,可以将有效量的所述抗网蛋白-1抗体和/或 其他治疗剂通过将所述药剂递送到所需表面的任何模式,例如粘膜、 系统递送,给药到对象。
本公开的药物组合物的给药可以通过专业技术人员已知的任何手 段来实现。给药途径包括但不限于口服、肠胃外、静脉内、肌肉内、 腹膜内、鼻内、舌下、鞘内、吸入、皮下、眼、阴道和直肠。系统途 径包括口服和肠胃外。几种类型的装置经常被用于通过吸入给药。这 些类型的装置包括定量吸入器(MDI)、呼吸驱动型MDI、干粉吸入 器(DPI)、与MDI组合的隔离室/保持室以及雾化器。
对于口服给药来说,可以通过将所述活性化合物与本领域中公知 的可药用载体合并,容易地配制所述化合物。这些载体能够使本公开 的化合物被配制成片剂、丸剂、糖衣丸、胶囊、液体、凝胶、糖浆、 浆液、悬液等,用于被待治疗的对象口服摄入。用于口服使用的药物 制剂可以作为固体赋形剂获得,任选地研磨得到的混合物,并且如有 需要,在添加适合的辅助剂后加工颗粒混合物,以获得片剂或糖衣丸 核心。适合的赋形剂具体来说是填充剂例如糖,包括乳糖、蔗糖、甘 露糖醇或山梨糖醇,纤维素制备物例如玉米淀粉、小麦淀粉、水稻淀 粉、马铃薯淀粉、明胶、黄芪树胶、甲基纤维素、羟丙基甲基纤维素、 羧甲基纤维素钠和/或聚乙烯吡咯烷酮(PVP)。如果需要,可以添加 崩解剂,例如交联聚乙烯吡咯烷酮、琼脂或藻酸或其盐例如藻酸钠。 任选地,所述口服制剂也可以在盐水或缓冲液中配制,用于中和内部 的酸性条件,或者可以不使用任何载体进行给药。
可以口服使用的药物制剂包括由明胶制成的推入配合胶囊,以及 由明胶和增塑剂例如甘油或山梨糖醇制成的密封软胶囊。所述推入配 合胶囊可以含有与填充剂例如乳糖、粘合剂例如淀粉和/或润滑剂例如 滑石或硬脂酸镁以及任选的稳定剂混合的活性成分。在软胶囊中,可 以将所述活性化合物溶解或悬浮在适合的液体例如脂肪油、液体石蜡或液体聚乙二醇中。此外,可以添加稳定剂。也可以使用被配制用于 口服给药的微球。这些微球在本领域中已被很好地定义。用于口服给 药的所有制剂都应该采取适合于这种给药的剂量。对于颊给药来说, 所述组合物可以采取以常规方式配制的片剂或锭剂的形式。
对于通过吸入的给药来说,按照本公开使用的化合物可以方便地 以来自于加压包或雾化器的气溶胶喷雾表现形式递送,并使用适合的 推进剂例如二氯二氟甲烷、三氯一氟甲烷、二氯四氟乙烷、二氧化碳 或其他适合的气体。在加压气溶胶的情况下,剂量单位可以通过提供 阀门来确定,以递送计量过的量。在吸入器或吹入器中使用的例如明 胶胶囊和药筒,可以被配制成含有所述化合物与适合的粉剂基料例如 乳糖或淀粉的粉末混合物。
所述化合物在希望系统性递送时,可以被配制成用于通过注射例 如通过快速浓注或连续输注进行肠胃外给药。用于注射的制剂可以以 单位剂型形式呈现,例如在安瓿中或多药剂容器中,并添加有防腐剂。 所述组合物可以采取诸如在油性或水性介质中的悬液、溶液或乳液的 形式,并且可以含有配制剂例如悬浮剂、稳定剂和/或分散剂。
用于肠胃外给药的药物制剂包括采取水溶性形式的所述活性化合 物的水性溶液。此外,所述活性化合物的悬液可以被制备成适合的油 性注射悬液。适合的亲脂性溶剂或介质包括脂肪油例如芝麻油或合成 的脂肪酸酯例如油酸乙酯或甘油三酯或脂质体。水性注射悬液可以含 有提高所述悬液的粘度的物质,例如羧甲基纤维素钠、山梨糖醇或右旋糖酐。任选地,所述悬液也可以含有适合的稳定剂或提高所述化合 物的溶解性的试剂以允许制备高度浓缩的溶液。
可选地,所述活性化合物可以采取粉末形式,用于在使用之前用 适合的介质例如无菌无热原水重构。
所述化合物也可以被配制成直肠或阴道组合物例如栓剂或保留灌 肠剂,其含有例如常规的栓剂基料例如可可脂或其他甘油酯。

本公开所描述的抗网蛋白-1抗体和组合物可以多次给药到对象 (例如患有癌症的对象)。在某些实施方式中,本公开的抗体或组合 物被递送到对象的次数在2至10次的范围内(例如2、3、4、5、6、7、 8、9或10次)。在某些实施方式中,异源核酸被递送到对象超过10次。
在某些实施方式中,本公开的抗体或组合物的药剂被不超过每日 历天(即24小时时长)一次地给药到对象。在某些实施方式中,本公 开的抗体或组合物的药剂被不超过每2、3、4、5、6或7个日历天一 次地给药到对象。在某些实施方式中,本公开的抗体或组合物的药剂 被不超过每日历周(例如7个日历天)一次地给药到对象。在某些实 施方式中,本公开的抗体或组合物的药剂被不超过每两周(即在两个 日历周时长内一次)地给药到对象。在某些实施方式中,本公开的抗 体或组合物的药剂被不超过每日历月一次(例如在30个日历天中一次) 地给药到对象。在某些实施方式中,本公开的抗体或组合物的药剂被 不超过每6个日历月一次地给药到对象。在某些实施方式中,本公开 的抗体或组合物的药剂被不超过每日历年(例如365天或在闰年中366 天)一次地给药到对象。
免疫测定法
在某些实施方式中,本公开涉及一种用于在原位或体外检测细胞 例如癌细胞表面上的网蛋白-1的方法。在某些实施方式中,本公开涉 及一种用于在从对象获得的样品中检测细胞表面上的网蛋白-1的方 法。所述样品可以从对象,例如通过从对象提取肿瘤或其一部分来获 得。在某些实施方式中,可以从所述肿瘤分离细胞。然而,在某些实 施方式中,可以在分离的肿瘤的背景下检查细胞。
在某些实施方式中,用于原位检测网蛋白-1的方法包括在网蛋白 -1抗体或抗原结合片段能够与所述对象中的细胞例如癌细胞上的网蛋 白-1的可接近的表位形成结合复合物的条件下,向对象递送偶联到标 记物(例如放射性标记物)的所述网蛋白-1抗体或抗原结合片段;以 及检测所述对象中的所述标记物(例如使用放射自显影或其他核医学 检测技术,包括单光子发射计算机断层扫描(SPECT)、正电子发射 断层扫描(PET)和闪烁扫描术)。
在某些实施方式中,用于在从对象获得的肿瘤样品中检测网蛋白 -1的方法包括(a)将所述样品与所述抗体或抗原结合片段在适合于所 述抗体或抗原结合片段与所述抗原的结合的条件下接触,如果所述抗 原存在于所述样品中,则由此形成结合复合物;并且(b)确定例如在 所述肿瘤的细胞表面处结合到所述抗原的所述抗体或抗原结合片段的 水平(例如确定所述结合复合物的水平)。

在任何所述检测、诊断和监测方法中,所述抗体或抗原结合片段 或抗原可以被直接或间接偶联到固相支持物表面。用于偶联到固相支 持物的方法是标准的,并且可以通过共价和非共价相互作用来实现。 偶联方法的非限制性实例包括吸附、交联、蛋白A/G-抗体相互作用和 链亲和素-生物素相互作用。其他偶联方法对于本领域普通技术人员来 说将是显而易见的。

作为另一个实施方式,本文中提供的抗体和抗原结合片段也可以 使用在用于评估在对象中的网蛋白-1表达的方法中,所述方法包括从 所述对象获得生物样品,其可以是血液样品、任何其他适合的体液样 品(例如淋巴液)或组织样品(例如胰腺组织、卵巢组织、来自于对 象头颈部的组织、乳腺组织、肺组织等)。所述程序可以包括将所述 样品(例如胰腺组织)或从其分离的蛋白质样品与抗体或抗原结合片 段在能够在抗体或抗原结合片段与抗原之间形成结合复合物的条件下 接触。然后可以通过本领域中已知的方法来确定这些结合复合物的水 平。

在某些实施方式中,在与反应区室(例如膜槽)中的所述抗体和 抗原结合片段接触之前,将所述抗原固定化在所述固相支持物例如 PVDF膜上。向所述反应区室添加检测试剂,以检测固定化的结合复合 物。在某些实施方式中,所述检测试剂包含针对所述抗原的可检测标 记的第二抗体。在某些实施方式中,所述检测试剂包含针对所述第一 抗体或抗原结合片段的可检测标记的第二抗体。正如本文中公开的, 所述可检测标记物可以是例如放射性同位素、荧光团、发光分子、酶、 生物素组成部分、表位标签或染料分子。在某些实施方式中,所述第 一抗体或抗原结合片段本身被可检测标记,并因此是检测试剂。适合 的可检测标记物在本文中有所描述,并且对于本领域普通技术人员来 说将是显而易见的。
因此,提供了适合于家庭或临床使用(护理服务现场)的诊断试 剂盒,其包含(a)可检测标记的和/或未标记的抗体或抗原结合片段作 为抗原结合试剂(例如网蛋白-1结合试剂);(b)检测试剂;和任选 地(c)使用所述试剂检测样品中的抗原的完整说明书。在某些实施方 式中,所述诊断试剂盒包括固定化在固相支持物上的所述抗体或抗原 结合片段和/或网蛋白-1。本文中描述的任何固相支持物都适合于并入 到所述诊断试剂盒中。在优选实施方式中,所述固相支持物是板孔的 反应区室的表面。通常,所述板孔在多孔板中,其具有选自6、12、24、 96、384和1536的孔数,但不限于此。在其他实施方式中,所述诊断 试剂盒提供了可检测标记的抗体或抗原结合片段。诊断试剂盒不限于 这些实施方式,并且试剂盒组成的其他变化对于本领域普通技术人员 来说将是显而易见的。

实施例
人类网蛋白-1在大肠埃希氏杆菌中的表达和纯化
将可随时表达的构建物即带有His标签的载体(15mg/L,SEQ ID NO:2)和带有GST标签的载体(30mg/L,SEQ ID NO:3)两者用 于产生单克隆抗体。为了评估表达,将所述质粒转化到大肠埃希氏杆 菌B菌株中。试验在4mL试管中进行,并检查下述变量:温度,表达 时间和异丙基β-D-1-硫代半乳吡喃糖苷(IPTG)的浓度。
通过离心收获细胞。将细胞沉淀物通过超声裂解,并使用镍柱通 过一步纯化获得靶蛋白。将级分合并和针对储存缓冲液透析。使用不 同的储存缓冲液以确定哪种缓冲液产生浓度超过0.4mg/mL的最稳定 的蛋白质。蛋白质使用标准流程通过SDS-PAGE和蛋白质印迹进行分 析,以获得分子量和纯度测量值。蛋白质的浓度使用Bradford蛋白质 测定法来确定,使用牛血清白蛋白(BSA)作为标准品。
抗人类网蛋白-1蛋白的单克隆开发
开发了一组特定的识别靶蛋白(在SEQ ID NO:1中下划线,如 SEQ ID NO:92所示)的抗人类网蛋白-1蛋白单克隆抗体。
首先,将5只BALB/c小鼠用GenScript’s MonoExpress免疫方案 免疫并观察两周。
使用电融合进行两次融合。使用这种方法的平均融合效率为1个 杂交瘤/5000个B细胞左右。杂交瘤克隆的预期得率为2x104,并将所 述融合的细胞铺于96孔板中。进行ELISA以筛选所述融合蛋白的阳性 克隆。然后通过ELISA将来自于所述阳性克隆的上清液针对所述靶蛋 白进行进一步筛选。使用带有10*His标签的蛋白质作为反向筛选。所 选的克隆对所述靶蛋白阳性并且对所述带有10*His标签的蛋白质阴 性。将所述阳性克隆在用人类重组Sec 8(网蛋白-1区段8)包被的24 孔板中扩增,并收集每个克隆的2mL上清液,然后将细胞冷冻保存。 表3示出了在包被有人类重组网蛋白-1区段8的板上生长的细胞系的OD450nm。

将来自于两个融合体的阳性原代克隆通过有限稀释法进行亚克 隆,以确保所述亚克隆源自于单一亲代细胞。将所述克隆生长三代。 将所述亚克隆通过ELISA进一步筛选。在所述ELISA结果的基础上, 将每个原代克隆的两个稳定的亚克隆细胞系冷冻保存。
克隆PAb1和PAb2表现出最高的特异性网蛋白结合潜力(图1)。 克隆PAb3、PAb4和PAb5也被证实具有杀死癌细胞的能力(图2)。
使用滚瓶以大约15mg/L的浓度生产所述抗体。所述抗体蛋白使 用蛋白A/G亲和柱层析进一步纯化,并在PBS缓冲液中透析以备储存。 对于质量控制来说,所述抗体经历通过SDS-PAGE进行的纯度试验, 通过OD280nm处的吸收值进行的浓度确定和通过ELISA进行的抗原反 应性试验。
对所选抗体进行标准的全长抗体测序。所述抗体经历总RNA提 取、RT-PCR和5’RACE和3’RACE PCR。将所述可变区和恒定区的 靶PCR片段进行凝胶纯化并克隆在测序载体中。对每条链的至少5个 独立的阳性克隆进行测序,以便推演共有序列。
PAb1和PAb2的单克隆抗体测序
PAb1和PAb2使用下述程序进行测序。使用试剂 (Ambion,目录号15596-026)和来自于/>试剂的技术手册的程 序,从通过GenScript回收的杂交瘤细胞分离总RNA。然后通过琼脂糖 凝胶电泳分析所述总RNA。使用同种型特异性反义引物或通用引物, 使用PrimeScriptTM第一链cDNA合成试剂盒,使用制造商的流程将所 述总RNA反转录成cDNA。扩增抗体的VH、VL、CH和CL片段。然后 使用标准的分子克隆程序将扩增的抗体片段分开克隆到标准的克隆载 体中。进行菌落PCR来鉴定具有正确尺寸的插入片段的克隆。为每个 抗体片段测序超过5个具有正确尺寸的插入片段的单菌落。
最后,将具有正确的VH、VL、CH、CL插入片段尺寸的5个单菌 落送去测序。发现5个不同克隆的VH、VL、CH、CL基因是几乎一致的。 在下面的序列部分中列出的PAb1和PAb2的共有序列,代表了PAb1 和PAb2抗体的序列。
使用PAb1的体外测定法
进行了体外结合测定。图3A示出了PAb1特异性结合到重组人类 网蛋白-1蛋白的C-端部分。数据表明,PAb1以高亲和性(例如Kd<1 nM)选择性结合到重组人类Sec8-His蛋白。图3B示出了PAb1对网

图5A-5D示出了在用PAb1处理后与诱导的通过凋亡进行的癌细 胞死亡相关的数据。图5A示出了通过流式细胞术进行的L3.6pl细胞 的荧光减一对照实验。图5B示出了在用IgG对照处理后72小时的 L3.6pl膜联蛋白V阳性细胞。图5C示出了在用PAb1处理72小时后的L3.6pl膜联蛋白V阳性细胞。数据表明PAb1处理的L3.6pl细胞与 IgG处理的对照细胞相比经历明显更多的凋亡(正如通过膜联蛋白V 所评估的)(图5D)。
在用PAb1或IgG对照处理后72小时,测量了癌细胞类型和健康 细胞类型的存活率。EC50通过逻辑非线性回归来计算,并报告为使细 胞存活性降低50%的mAb浓度(nM)。数据示出在表4中(下表)。

通过共聚焦显微术研究了PAb1处理对YapC癌细胞的微管蛋白各 向异性的影响。图6A示出了没有用PAb1处理的YapC细胞中的微管 蛋白染色。图6B示出了在一甲基澳瑞他汀E(MMAE)处理后10分 钟的微管蛋白染色;MMAE阻断微管蛋白聚合。图6C示出了在PAb1 处理后24小时的微管蛋白染色。数据表明与未处理的对照细胞相比, 在用PAb1处理的细胞中各向异性降低(图6D)。
也研究了YapC癌细胞中PAb1与微管蛋白的共定位。示出了在微 管蛋白染色(图7A)和PAb1染色(图7B)后YapC的代表性的共聚 焦显微术图像。数据表明,微管蛋白和PAb1共定位在染色细胞的表面 上(图7C;箭头)。在几个细胞上也观察到了PAb1染色的增加。
使用PAb1的体外测定法
将带有皮下YapC肿瘤的免疫受损小鼠给药PAb1或IgG对照。数 据表明,在11天后,用3mg/kg PAb1治疗的小鼠与用IgG对照治疗的 小鼠相比具有明显更小的肿瘤体积(图8A)。数据还表明,1mg/kg PAb1 小鼠在第14天引发肿瘤体积的显著减小。与用0.3mg/kgPAb1治疗的 小鼠相比,两种较高的PAb1剂量(1mg/kg和3mg/kg)引起明显更低 的肿瘤体积(图8A),指示了剂量依赖性效应。数据还表明,用PAb1 治疗的动物在整个治疗的持续时间内没有体重减轻(图8B)。还示出 了用3mg/kg PAb1治疗的小鼠在第0天、第14天和第25天的照片(图 8C-8E)。
