Abstract
本发明公开了一种核酸适配体。本发明提供了核酸适配体6.9及其截短序列6.9S及其在识别冠状病毒方面的应用,本发明还提供了一种检测冠状病毒的方法及试剂盒,本发明提供的核酸适配体具有稳定、操作简便、迅速等优点,能够特异性识别冠状病毒,具有高特异性和敏感性,有广泛的应用价值和广阔的市场前景。

技术领域
本发明属于生物医药领域,具体涉及一种核酸适配体。
背景技术
自从2019年12月首次报道新型冠状病毒(SARS-CoV-2)以来,疫情在全球范围内迅速蔓延。据世界卫生组织(WHO)统计,截止到2022年2月4日,已报道全世界有440,807,756例确诊病例,死亡5,978,096人,新型冠状病毒不仅在全世界夺走了无数的生命,面对疫情,全世界各国政府为了阻止新型冠状病毒迅速蔓延而采取的封锁措施更是给全世界经济带来了重创。
新型冠状病毒感染者会出现急性、严重呼吸道疾病,伴有发热、咳嗽、气短及呼吸困难,严重的病例还会出现肾功能衰竭和死亡,当前并无有效的治疗药物。新型冠状病毒检测方法目前也并不完善,需依赖特定仪器、特殊实验室和专业技术人员,且检测过程复杂,不能满足及时检测需求,且存在一定的漏检率。因此,开发出一种灵敏度高、经济的快速检测技术,对新型冠状病毒的早期诊断具有十分重要的意义。
发明内容
为弥补现有技术的不足,本发明提供了一种核酸适配体,能够特异性识别新型冠状病毒。
为实现上述目的,本发明采用如下技术方案:
本发明的第一方面提供了一种核酸适配体,所述核酸适配体的序列包括SEQ IDNO.1或2任一所示的核苷酸序列或与其具有至少60%、至少70%、至少80%、至少85%、至少90%、至少92%、至少94%、至少96%、至少98%或至少99%的同源性的DNA序列或其截短形式。
进一步,所述核酸适配体的序列包括SEQ ID NO.2所示的核苷酸序列。
进一步,所述核酸适配体的序列如SEQ ID NO.1或SEQ ID NO.2所示。
本发明的第二方面提供了一种核酸适配体,所述核酸适配体包括在严格条件下与本发明第一方面所示的DNA序列杂交的DNA序列。
本发明的第三方面提供了一种核酸适配体,所述核酸适配体包括由本发明第一方面所示的DNA序列转录的RNA序列。
本发明的第四方面提供了一种核酸适配体衍生物,所述核酸适配体衍生物由本发明第一方面、第二方面或第三方面所述的核酸适配体连接、改造、修饰而成。
进一步,与核酸适配体连接的物质包括荧光素标记、同位素标记、治疗性物质、酶标记、生物素标记的一种或多种。
进一步,所述物质选自生物素标记。

本发明提供了一种核酸适配体,所述核酸适配体的序列包括SEQ ID NO.1或2任一所示的核苷酸序列或与其具有至少60%、至少70%、至少80%、至少85%、至少90%、至少92%、至少94%、至少96%、至少98%或至少99%的同源性的DNA序列或其截短形式。
所述核酸适配体的序列包括SEQ ID NO.2所示的核苷酸序列。
所述核酸适配体的序列如SEQ ID NO.1或SEQ ID NO.2所示。
在本发明中,核酸适配体与适配体、适体可互换使用,是指具有例如约20~200个碱基长度这样的短序列的核酸分子,并且是能够特异性结合成为靶标的分子、物质的单链核酸分子。
在本发明中,特异性结合或异性结合于或特异性针对在本发明中可互换使用,泛指试剂(例如核酸试剂,例如核酸适配体)与靶分子(例如蛋白质或病毒)的结合,并且所述结合与非特异性相互作用(例如,非特异性相互作用可以是与参考分子或随机分子的结合)在可测量程度和/或统计学上不同。特异性结合可以例如通过与类似于靶标的对照分子,例如过量的未标记的靶标竞争来测量,在这种情况下,如果标记的靶标与候选试剂的结合被过量的未标记的靶标竞争性地抑制,那么表明了特异性结合。在本发明中特异性结合特指核酸适配体与冠状病毒的结合。对靶标的KD为至少约100nM、至少约90nM、至少约80nM、至少约70nM、至少约60nM、至少约50nM、至少约40nM、至少约30nM、至少约20nM、至少约15nM、至少约13nM、至少约12nM、至少约10.8nM、至少约10.6nM、至少约10.4nM、至少约10.2nM、至少约5nM、至少约1nM或更大的分子可表现出特异性结合。
在本发明的具体实施方案中,核酸适配体与冠状病毒以10.2nM的KD结合。
在本发明中,KD值泛指解离常数,通过适合于适体和靶标对的技术测量,例如通过使用配体结合测定,利用放射性或荧光测量、表面等离子共振(SPR)、生物膜干涉技术(BLI,例如系统)、SRU生物系统等温滴定量热学(ITC)或MicroscaleThermophoresis(MST)测量。在一些实施方案中,KD值使用标准的基于荧光的配体结合测定和饱和分析来测定。
“与…具有至少60%、至少70%、至少80%、至少85%、至少90%、至少92%、至少94%、至少96%、至少98%或至少99%的同源性的DNA序列”是指相对于参比序列包含一个至若干个核苷酸添加、缺失或置换的核酸序列。
本发明涉及的核酸适配体只要具有与冠状病毒特异性结合的功能,则亦可以是上述SEQ ID NO.1或SEQ ID NO.2所示的序列中的1个或多个核苷酸被置换、缺失或添加而得到的序列。该被置换、缺失或添加的核苷酸优选为1~3个,更优选为1或2个,进一步优选为1个。另外,只要不妨碍SEQ ID NO.1或SEQ ID NO.2的核酸适配体功能,则亦可使同序列与不同的碱基序列、核酸适配体、肽、蛋白质(酶、抗体)连接。
在1个或多个核苷酸被置换的情况下,该置换可利用通用碱基进行。术语“通用碱基”表示该技术领域中通常所认为的含义。即,该术语通常是指与标准DNA/RNA的各碱基几乎无区别地形成碱基对、被胞内酶识别的核苷酸碱基类似物(例如,Loakes等,1997,J.Mol.Bio.270:426-435)。作为通用碱基的非限定性例子,可举出C-苯基、C-萘基及其他芳香族衍生物、肌苷、唑甲酰胺(carbozamide)、以及硝基唑衍生物(3’-硝基吡咯、4-硝基吲哚、5-硝基吲哚、及6-硝基吲哚等)(Loakes,2001,Nucleic Acids Res.29:2437)。
核苷酸在本发明环境中指代构成核酸的单体。核苷酸由戊糖、含氮碱基和磷酸酯基团形成,并且通过磷酸二酯键结合。核苷酸——DNA和RNA的部分——区别于戊糖,其分别是脱氧核糖和核糖。含氮碱基进而分成嘌呤含氮碱基——为腺嘌呤(A)和鸟嘌呤(G),和嘧啶含氮碱基——为胸腺嘧啶(T)、胞嘧啶(C)和尿嘧啶(U)。胸腺嘧啶仅出现在DNA中,而尿嘧啶仅出现在RNA中,本发明的核苷酸与核苷酸序列、序列可互换使用。
本发明提供了一种核酸适配体,所述核酸适配体包括在严格条件下与上述所示的DNA序列杂交的DNA序列。
在本发明中,杂交是指其中两个单链多核苷酸非共价结合以形成稳定的双链多核苷酸的过程。杂交也可指三链杂交。所得(通常)双链多核苷酸为“杂合体”。
本发明提供了一种核酸适配体,所述核酸适配体包括由上述所示的DNA序列转录的RNA序列。
在本发明中,转录涉及这样的过程,其中DNA序列中的遗传密码被转录成RNA。随后,RNA可以被翻译成蛋白质。
在本发明中,核酸适配体可使用该技术领域中公知的体外筛选法进行筛选及获得。体外筛选法是从含有随机核苷酸序列的核酸分子的库(例如,DNA库)中选择针对靶分子、细胞具有亲和性的适配体分子、排除不具有亲和性的分子的方法。通过重复实施使用PCR法等仅扩增该选择出的适配体分子、进而基于亲和性进行选择这样的循环,能够将具有强结合能力的适配体分子浓缩。
在本发明的实施方案中,使用指数富集的配基系统进化技术,简称SELEX(Systematic Evolution of Ligands by EXponential Enrichment)技术筛选核酸适配体。SELEX应用大容量的随机寡核苷酸文库(ssDNA文库和RNA文库),结合PCR体外扩增技术,以指数级富集与靶分子特异结合的寡核苷酸,经过反复的体外筛选、扩增,最终获得的核酸适配体(aptamer)基于空间结构与靶分子呈高特异性和高亲和力的结合。
本发明提供了一种冠状病毒的核酸适配体衍生物,所述核酸适配体衍生物由上述的核酸适配体连接、改造、修饰而成。
连接的物质包括荧光素标记、同位素标记、治疗性物质、酶标记、生物素标记的一种或多种。
其中,荧光素标记包括但不限于荧光素、藻红蛋白(PE)、得克萨斯红(Texas red,TR)、罗丹明、游离的镧系元素盐、螯合的镧系元素盐、CyDye、BODIPY和ALEXA。
同位素标记包括但不限于2H(氘标D)、13C、15N、17O、18O、34S。
治疗性物质指具有药用、药理、身心和治疗作用的物质,包括但不限于纳米颗粒、RNA药物、siRNA、合成的或生物学生产的药物。
酶标记是指具有酶活性的标记。一个典型的、优选的实例是辣根过氧化物酶(HRP)。
在本发明具体的实施方案中,使用生物素进行标记。
能够被用于连接于适配体的连接分子的化学物质是本领域公知的,例如二硫键连接、氨基连接、共价连接等等。另外的连接和修饰能够在www.trilinkbiotech.com/products/oligo/oligo_modifictions.asp上找到。分子能够在适配体的5’和3’位置中的一者或二者连接于适配体,或者甚至能够分子间连接。
核酸适配体的改造的物质包括由所述的核酸适配体的核苷酸序列的骨架衍生出的硫代磷酸酯骨架或由所述的核酸适配体的核苷酸序列改造成的相应锁核酸或肽核酸。
在本发明中,硫代磷酸酯骨架具有本领域普通技术人员一般理解的含义,其是指RNA和DNA核酸适配体的磷酸二酯骨架的非桥键氧原子可以被一个或两个硫原子取代,分别产生具有硫代磷酸酯或二硫代磷酸酯键的硫代磷酸酯骨架。已知这样的硫代磷酸酯骨架具有增加的对其靶标的结合亲和力,以及对核酸酶降解的增强的抗性。
肽核酸或PNA在本发明中指代这样的寡核苷酸:其骨架由通过肽键结合的N-(2-氨基乙基)-甘氨酸重复单元构成,其中不同含氮碱基通过亚甲基键(-CH2-)和羰基(-(C=O)-)结合于主链。
锁核酸或LNA在本发明中指代修饰RNA核苷酸,其核糖部分被连接2’位的氧与4’位的碳的另一键修饰,将核糖锁定在3'-内构象。术语5’或5’链或3’或3’链在本发明环境中指代这样的寡核苷酸:其中3’或5’端的核苷酸分别颠倒。
所述修饰包括一个或多个修饰核苷酸,和/或一个或多个取代。
所述修饰核苷酸在独立地选自核糖位置、脱氧核糖位置、磷酸位置和碱基位置的一个或多个位置处包含化学取代或修饰。
在本发明中,修饰核苷酸泛指一种核苷酸,它虽然不是天然存在的,但是是天然存在的核苷酸的类似物或酯。当在寡核苷酸或核酸分子(例如核酸适配体)的背景下使用时,其一般意指寡核苷酸的四种构成核苷酸(即,A、G、T/U和C)中的至少一者是天然存在的核苷酸的类似物或酯。在一些实施方案中,修饰核苷酸可以使寡核苷酸对核酸酶具有抗性。在一些实施方案中,修饰核苷酸主要引起适体与它的蛋白质靶标的疏水性相互作用,产生高结合效率和稳定的共结晶复合物。在C-5位具有取代的嘧啶是修饰核苷酸的一个实例。修饰可包括主链修饰、甲基化、罕见的碱基配对组合(例如异碱基(isobase)异胞苷(isocytidine)和异胍(isoguanidine))等。修饰还可包括3'和5'修饰,例如加帽。其它修饰可包括一种或多种天然核苷酸经类似物取代、核苷酸间修饰,例如具有不带电键(例如膦酸甲酯、磷酸三酯、氨基磷酸酯、氨基甲酸酯等)的那些修饰和具有带电键(例如硫代磷酸酯、二硫代磷酸酯等)的修饰、具有嵌入剂(例如吖啶、补骨脂素等)的修饰、包含螯合剂(例如金属、放射性金属、硼、氧化金属等)的修饰、包含烷基化剂的修饰以及具有修饰键(例如α异头核酸等)的修饰。此外,核苷酸的糖上通常存在的任一个羟基均可以被膦酸基或磷酸基替代;用标准的保护基保护;或激活,以与另外的核苷酸或固体载体形成另外的键。5'和3'端OH基团可以磷酸化或经以下取代:胺、具有约1个至约20个碳原子的有机封端基团部分、在一些实施方案中在约10kDa至约80kDa范围内的聚乙二醇(PEG)聚合物、在一些实施方案中在约20kDa至约60kDa范围内的PEG聚合物或其它的亲水性或疏水性生物或合成聚合物。在一些实施方案中,修饰可以在嘧啶的C-5位。这些修饰可以通过直接在C-5位的酰胺键联产生或通过其它类型的键联产生。
本发明提供了上述的核酸适配体、上述核酸适配体衍生物、上述试纸条或上述试剂盒在抑制冠状病毒感染或制备治疗治疗冠状病毒感染引起的疾病的药物中的应用。
所述疾病包括但不限于新型冠状病毒引起的疾病、感冒、肺炎。
所述疾病选自新型冠状病毒引起的疾病。
在本发明中,新型冠状病毒引起的疾病是指由一种新出现的冠状病毒SARS-CoV-2引起的呼吸道感染。新型冠状病毒引起的疾病的临床综合征范围从轻度或不复杂的疾病,例如发烧、疲劳、咳嗽(有痰或无痰)、厌食、乏力、肌肉疼痛、喉咙痛、呼吸困难、鼻塞、头痛,或很少有的腹泻、恶心和呕吐,到需要住院和氧气支持或进入重症监护室并可能需要机械通风的严重疾病。
本发明的药物还包括药学上可接受的载体。
在本发明中,作为可以抑制冠状病毒感染或治疗冠状病毒感染的疾病的药物优选的包括核酸适配体衍生物,更优选的包括与上述核酸适配体连接的物质,最优选的包括连接的治疗性物质。
药学上可接受的是指在合理的医学判断的范围内,适合用于与人和动物的组织接触而没有过度的毒性、刺激、过敏反应或其他问题或并发症,与合理的益处/风险比相称的那些化合物、材料、组合物和/或剂型。
药学上可接受的载体包括但不限于赋形剂、粘合剂、崩解剂、乳化剂、流动添加调节剂、润滑剂或稳定剂。
其中,赋形剂包括但不限于单糖、二糖类、环糊精和多糖类这样的糖(具体没有限制,包括葡萄糖、蔗糖、乳糖、棉子糖、甘露醇、山梨糖醇、肌醇、糊精、麦芽糊精、淀粉和纤维素)、金属盐(例如磷酸钠或磷酸钙、硫酸钙、硫酸镁)、柠檬酸、酒石酸、甘氨酸、低/中/高分子量的聚乙二醇(PEG)、普兰尼克(Pluronic)或它们的组合。
粘合剂包括但不限于使用玉米、小麦、大米或马铃薯淀粉的淀粉糊、明胶、黄蓍胶、甲基纤维素、羟丙基甲基纤维素、羧甲基纤维素钠和/或聚乙烯吡咯烷酮。
崩解剂包括但不限于玉米、小麦、大米或马铃薯淀粉、羧甲基淀粉、交联聚乙烯吡咯烷酮、琼脂、海藻酸或海藻酸钠或它们的盐。
乳化剂包括但不限于山梨糖醇酐脂肪酸酯、甘油脂肪酸酯、蔗糖脂肪酸酯、丙二醇脂肪酸酯。
流动添加剂和润滑剂包括但不限于硅酸盐、滑石、硬脂酸盐或聚乙二醇。
稳定剂包括但不限于抗坏血酸和亚硫酸盐等抗氧化剂、海藻糖和葡萄糖等糖类。
本发明的药物的剂型为不会使有效成分失活的形式,只要是在给药后能够在生物体内发挥其药理作用的形式,就没有特别限定。一般是因给药方法和/或处方条件而异。
其中,作为适合于经口给药的剂型,包括但不限于固体制剂(包括片剂、丸剂、舌下片剂、胶囊剂、滴剂、锭剂)、颗粒剂、粉剂、散剂、液剂。此外,固体制剂可以根据需要采用实施该领域中公知的包衣的剂型,例如糖衣片、明胶包衣片、肠溶片、薄膜包衣片、双层片、多层片。
非经口给药可以细分为全身给药和局部给药,局部给药可以进一步细分为组织内给药、经皮给药、经粘膜给药和经直肠给药,药物也可以制成适合于各给药方法的剂型。作为适合于全身或组织内给药的剂型,包括但不限于作为液剂的注射剂。作为适合于经皮给药或经粘膜给药的剂型,包括但不限于液剂(包括涂布剂、滴眼剂、滴鼻剂、吸入剂)、悬浮剂(包括乳剂、霜剂)、粉剂(包括滴鼻剂、吸入剂)、糊剂、凝胶剂、软膏剂、硬膏剂。作为适合于经直肠给药的剂型,包括但不限于栓剂。
需要说明的是,上述各剂型的具体形状、大小只要在各剂型中均处于该领域公知的剂型范围内即可,没有特别限定。
本发明的药物还可以以脂质体递送系统的形式施用,例如小单层囊泡、大单层囊泡和多层囊泡。脂质体可以由多种磷脂例如胆固醇、硬脂胺或磷脂酰胆碱构成。
本发明的药物还可以与作为可靶向药物载体的可溶性聚合物偶联。此类聚合物可以包括聚乙烯吡咯烷酮,吡喃共聚物,聚羟基丙基甲基丙烯酰胺酚,聚羟基乙基天冬酰胺酚,或聚氧化乙烯聚赖氨酸,其由棕榈酰残基取代。此外,本发明的药物可以与在实现药物的控制释放中有用的生物可降解的聚合物类别偶联,例如聚乳酸、聚ε己内酯、聚羟基丁酸、聚原酸酯、聚缩醛、聚二氢吡喃、聚腈基丙烯酸酯和水凝胶的交联或两亲嵌段共聚物。虽然已显示且描述了本发明的至少一个实施方案,但应当理解可以对其作出许多改变和修饰,而不背离如附加权利要求中定义的本发明的原理和范围。
下面结合具体实施例进一步阐述此发明。应理解的是,在此描述的特定实施方式通过举例的方式来表示,并不作为对本发明的限制。在不偏离本发明范围的情况下,本发明的主要特征可以用于各种实施方式。
实施例1核酸适配体特异识别灭活新型冠状病毒颗粒
1.1实验材料
灭活新型冠状病毒颗粒:科兴疫苗公司提供。
1.2实验方法
1.2.1核酸适配体的合成
合成6.9、6.9s和无关对照序列-A20S,通过公司合成使6.9、6.9s和A20S的5’端标记上Bio,具体序列如表1所示。

1.2.2酶联免疫实验
1)将一定量的灭活新型冠状病毒颗粒(或灭活HINI病毒颗粒)融于pH 9.7的碳酸盐缓冲液中,按照100μl/孔加入到酶联条中,4℃包被过夜;
2)弃包被液,每孔加入100μl含2% BSA封闭液,室温封闭60min;
3)将一定浓度的Bio-6.9和无关对照序列Bio-A20S;或Bio-6.9和其截短序列6.9s溶解于合适体积的缓冲液中(50mM HEPES,100mM NaCl,2mM MgCl2,5mM KCl,1mM CaCl2),于100℃变性5min后立即置于冰上充分冷却;
4)将经过变性处理后的Bio-6.9和无关对照序列(Bio-A20S文库);或Bio-6.9和其截短序列6.9s加入酶联条中,使核酸适配体与包被的灭活病毒颗粒共同孵育37℃30min;
5)弃去孔内液体,每孔用200μl的洗涤液洗涤,重复洗涤3次,最后一次洗涤后要把孔内液体完全甩干;
6)每孔加入100μl按照1:100稀释好的HRP酶,室温孵育40min,弃去孔内液体,洗板5次,方法同上;
7)每孔加入100μl TMB显色底物,37℃避光显色,当有明显颜色变化时,加10μl终止液,酶联仪读数。
1.2.3Bli技术
将100nM Bio标记的适配体6.9溶解于缓冲液中(50mM HEPES,100mM NaCl,2mMMgCl2,5mM KCl,1mM CaCl2),与带有链霉亲和素蛋白的探针相结合,随后分别与10μg/ml灭活新型冠状病毒颗粒溶液(上样液溶解)结合,最后用上样液解离。整个上样、结合、解离过程均在分子相互作用仪中进行。
1.2.4平衡解离常数(KD值)的测定
灭活新型冠状病毒颗粒包被在微孔板中(1μg/孔),将生物素标记的适配体6.9配制成浓度为0.78nM,1.56nM,3.125nM,6.25nM,12.5nM,25nM和100nM,随后分别加入至包被了SARS-CoV2病毒颗粒的孔中结合,根据OD450nm吸光度值绘制曲线。
1.3实验结果
ELISA实验证明核酸适配体6.9特异识别灭活新型冠状病毒颗粒,而不结合其它灭活病毒颗粒(HINI灭活病毒颗粒)。而HINI的特异核酸适配体A20S能结合HINI病毒颗粒,而不与灭活新型冠状病毒颗粒(图1)。表明筛选得到的核酸适配体6.9具有结合特异性。
核酸适配体6.9及其截短序列均可特异结合灭活新型冠状病毒颗粒而不结合其他无关蛋白,表明筛选得到的核酸适配体6.9及其截短序列具有结合特异性(图2)。
Bli技术证实适配体6.9特异结合灭活新型冠状病毒颗粒(图3)。
通过适配体6.9与灭活新型冠状病毒颗粒结合的平衡解离常数(KD值)的测定的实验可以得其平衡解离常数KD值约为10.2nM(图4),相关数值如表2所示。

上述实施例的说明只是用于理解本发明的方法及其核心思想。应当指出,对于本领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也将落入本发明权利要求的保护范围内。
Claims (10)
1.一种核酸适配体,其特征在于,所述核酸适配体的序列包括SEQ ID NO.1或2任一所示的核苷酸序列或与其具有至少60%、至少70%、至少80%、至少85%、至少90%、至少92%、至少94%、至少96%、至少98%或至少99%的同源性的DNA序列;
优选的,所述核酸适配体的序列包括SEQ ID NO.2所示的核苷酸序列;
优选的,所述核酸适配体的序列如SEQ ID NO.1或SEQ ID NO.2所示。
2.一种核酸适配体,其特征在于,所述核酸适配体包括在严格条件下与权利要求1所示的DNA序列杂交的DNA序列。
3.一种核酸适配体,其特征在于,所述核酸适配体包括由权利要求1所示的DNA序列转录的RNA序列。
4.一种核酸适配体衍生物,其特征在于,所述核酸适配体衍生物由权利要求1-3任一项所述的核酸适配体连接、改造、修饰而成;
优选的,与核酸适配体连接的物质包括荧光素标记、同位素标记、治疗性物质、酶标记、生物素标记的一种或多种;
优选的,所述物质选自生物素标记;
优选的,核酸适配体的改造的物质包括由所述的核酸适配体的核苷酸序列的骨架衍生出的硫代磷酸酯骨架或由所述的核酸适配体的核苷酸序列改造成的相应锁核酸或肽核酸。
5.根据权利要求4所述的核酸适配体衍生物,其特征在于,所述修饰包括一个或多个修饰核苷酸,和/或一个或多个取代;
优选的,所述修饰核苷酸在独立地选自核糖位置、脱氧核糖位置、磷酸位置和碱基位置的一个或多个位置处包含化学取代或修饰;
优选的,所述修饰核苷酸包含一个或多个独立地选自以下的修饰:2'-位置糖修饰、2'-氨基(2'-NH2)修饰、2'-氟(2'-F)修饰、2'-O-甲基(2'-OMe)修饰、2'-O-(2-甲氧基乙基)(2'-O-MOE)修饰、5-位置修饰嘧啶、胞嘧啶环外胺处的修饰、经5'-溴尿嘧啶取代、经5'-溴脱氧尿苷取代、经5'-溴脱氧胞苷取代、骨架修饰、锁核酸(LNA)、甲基化、3'帽和5'帽;

6.一种检测冠状病毒的试纸条,其特征在于,所述试纸条包括权利要求1-3任一项所述的核酸适配体、权利要求4或5所述的核酸适配体衍生物。
7.一种检测冠状病毒的试剂盒,其特征在于,所述试剂盒包括权利要求1-3任一项所述的核酸适配体、权利要求4或5所述的核酸适配体衍生物或权利要求6所述的试纸条;
优选的,所述冠状病毒包括SARS冠状病毒、MERS冠状病毒;
优选的,所述冠状病毒选自SARS冠状病毒;
优选的,所述SARS冠状病毒包括SARS-CoV-2冠状病毒和SARS-CoV-1冠状病毒;
优选的,所述SARS冠状病毒选自SARS-CoV-2冠状病毒。
8.一种检测冠状病毒的方法,其特征在于,所述方法包括使样品与权利要求1-3任一项所述的核酸适配体、权利要求4或5所述的核酸适配体衍生物接触,从而检测所述样品中的冠状病毒;
优选的,所述冠状病毒包括SARS冠状病毒、MERS冠状病毒;
优选的,所述冠状病毒选自SARS冠状病毒;
优选的,所述SARS冠状病毒包括SARS-CoV-2冠状病毒和SARS-CoV-1冠状病毒;
优选的,所述SARS冠状病毒选自SARS-CoV-2冠状病毒。
9.权利要求1-3任一项所述的核酸适配体、权利要求4或5所述的核酸适配体衍生物或权利要求6所述的试纸条在检测冠状病毒或制备检测冠状病毒的产品中的应用;
优选的,所述冠状病毒包括SARS冠状病毒、MERS冠状病毒;
优选的,所述冠状病毒选自SARS冠状病毒;
