Abstract
本发明提供了用于药物递送的含修饰蛋白的双层膜囊泡及其制备方法和应用,所述含修饰蛋白的双层膜囊泡是将修饰蛋白编码基因与基础载体连接后转入宿主细胞,诱导产生含修饰蛋白的双层膜囊泡;所述修饰蛋白包括标签蛋白TAG、适配体结合蛋白ABP或双层膜囊泡结构蛋白ASP中的一种或多种的混合。本发明采用抗原嵌合方式对双层膜囊泡进行修饰,双层膜囊泡可以装载多肽药物,还可以通过结合的适配体把多肽药物、表达蛋白的核酸、敲低(miRNA,siRNA)、敲除(sgRNA)核酸、或其他一切形式的核酸装载入该结构,也可以通过药物受体捕捉小分子药物和纳米材料装载入双层膜囊泡。双层膜囊泡具有高效的药物富集能力,并且具有低免疫源性,可以通过其生物囊膜结构快速的与靶细胞融合,并把药物送至靶细胞。
Description
技术领域:
本发明属于生物医药和基因工程领域,尤其是涉及一种用于药物递送的含修饰蛋白的双层膜囊泡及其制备方法和应用。
背景技术:
双层膜囊泡能吞噬自身细胞质蛋白或细胞器,直径一般为80-900nm,平均500nm左右。双层膜囊泡包裹和吞噬的底物包括细胞器如线粒体、内吞体、过氧化物酶体,以及外来的入侵物质等。所以比起细胞中许多囊泡结构,双层膜囊泡能够包裹更多更大的底物。另外遗传证据表明双层膜能够将缺乏分泌氨基末端前导肽或信号序列的蛋白质分泌到胞外。双层膜囊泡(双层磷脂双分子层和囊泡状结构)的生理特性使它们具有很好的生物相容性。并且双层膜囊泡可以自由的穿过血脑屏障(BBB),低免疫原性和毒性使其可以很容易的被任何器官和组织中的细胞摄取,且不会引起机体的免疫反应,具有无毒、生物相容性好、组织和肿瘤靶向性和循环半衰期长等优点,是新型药物递送系统的理性候选者。
核酸适配体,也被称为核酸适体、适配子等,是从人工合成的DNA/RNA文库中筛选得到的能够高亲合性和高特异性地与各种靶标结合的单链寡核苷酸。最早是由Szostak和Gold两个小组几乎同时提出。1990年,Ellington和Szostak报道了能结合小分子有机染料的RNA片段,并将其命名为Aptamer。适配体(aptamer)是指利用指数富集的配基系统进化(Systematic Evolution of Ligands by Exponential Enrichment,SELEX)技术,从人工合成的寡核苷酸文库中筛选得到的具有亲和性高、特异性强的能够与靶分子特异性结合的短的单链DNA和RNA分子。
LC3是双层膜囊泡形成的关键分子,也是其组成成分。LC3是一种蛋白轻链,是自噬过程的标志物。它的功能主要参与了自噬小体的形成,LC3前体分子被ATG4B剪去C端的5肽,裂解形成胞质形式LC3-I。然后被APG7L/ATG7激活,转移至ATG3并偶联脂酰乙醇胺(PE)以形成膜结合形式LC3-II,它可以附着到双层膜囊泡(autophagosome)的膜上,是双层膜囊泡的结构蛋白。另外报道表明,外泌的双层膜囊泡能够通过LC3把RNA结合蛋白分泌到胞外。而双层膜囊泡在所有真核细胞中广泛存在,很容易通过饥饿诱导等方式大量诱导双层膜囊泡的产生;并且通过双层膜囊泡LC3的修饰改造,很容易进行靶标药物的装载。
目前外泌体作为一种新型的药物递送系统,与传统药物递送系统相比,显示出了更高的生物相容性和安全性、更低的免疫原性、较好地穿越生物屏障的能力和一定的靶向性,然而外泌体仍然存在着许多缺点限制着其临床转化,主要包括:(1)外泌体的产量极低;缺乏经济高效的外泌体提取流程;(2)质量严格受控于上游步骤;(3)缺乏高效的外泌体载药方式;对外泌体进行载药等破坏膜的操作时,存在改变膜蛋白方向的风险,这可能造成免疫系统的识别继而引发不良反应。
发明内容:
为解决现有技术中外泌体等药物递送系统所不能解决的问题,本发明突破现有的药物递送系统的局限,通过双层膜囊泡结构蛋白ASPs把目标蛋白和适配体结合蛋白(Aptamer-binding protein,ABP)或多肽捕捉蛋白(Peptide Catcher protein,PC)、ABP-适配体-底物蛋白复合体包装入双层膜囊泡,而后在细胞内生成大量的携带有药物蛋白、核酸或核酸蛋白复合体的修饰双层膜囊泡,并通过这些双层膜囊泡把药物蛋白、核酸或核酸蛋白复合体递送到靶标细胞中,实现高效药载系统的制备和药物递送。
本发明采用的技术方案是:
本发明提供一种用于药物递送的含修饰蛋白的双层膜囊泡,所述含修饰蛋白的双层膜囊泡是将修饰蛋白编码基因与基础载体连接后转入宿主细胞,诱导产生含修饰蛋白的双层膜囊泡;所述修饰蛋白包括标签蛋白TAG、适配体结合蛋白ABP或双层膜囊泡结构蛋白ASP中的一种或多种的混合;所述基础载体包括真核表达启动子、抗性基因和选择性标记基因;选择性标记基因包括但不限于嘌呤霉素(Puromycin)。
优选的,所述标签蛋白TAG用于亲和层析或免疫沉淀法捕捉双层膜囊泡,包括Flag标签、Myc标签、HA标签、HIS标签或GST标签中的一种或多种;所述适配体结合蛋白ABP包括MS2蛋白、COM蛋白、PP7蛋白、BoxB蛋白或PEG10蛋白中的一种或多种;所述双层膜囊泡结构蛋白ASP包括但不限于LC3同源蛋白或ATG9同源蛋白中的一种或多种,如蛋白轻链3Aa(LC3Aa)、蛋白轻链3Ab(LC3Ab)、蛋白轻链3B(LC3B)、蛋白轻链3C(LC3C)、γ氨基丁酸受体(GABARAP),γ氨基丁酸受体I型(GABARAPL1)、γ氨基丁酸受体2型(GABARAPL2),自噬相关蛋白9(ATG9)等蛋白中的一种或多种,以上ASP来源于但不限于物种人。所述ABP通过ASP结合于双层膜囊泡内膜上。
优选的,所述修饰蛋白为标签蛋白TAG、适配体结合蛋白ABP和双层膜囊泡结构蛋白ASP可表达连接成的融合蛋白;所述融合蛋白的线性顺序为以下之一:TAG-ABP-ASP、ABP-TAG-ASP、ABP-ASP-TAG、TAG-ASP-ABP、ASP-TAG-ABP或ASP-ABP-TAG。
本发明还提供一种所述含修饰蛋白的双层膜囊泡的制备方法,所述方法按包括以下步骤:(1)将修饰蛋白编码基因克隆至基础载体中,制得重组质粒;所述基础载体包含真核表达启动子、抗性基因、选择性标记基因;在启动子后面接入修饰蛋白编码基因;选择性标记基因包括但不限于嘌呤霉素(Puromycin);抗性基因包括:卡那霉素或氨苄青霉素等;所述启动子包括人巨细胞病毒启动子(CMV)、是猴空泡病毒40启动子(SV40)等;
(2)通过真核细胞转染试剂将步骤(1)重组质粒转入目的细胞中,筛选阳性克隆细胞;所述目的细胞包括但不限于293T细胞;
(3)将步骤(2)阳性克隆细胞饥饿诱导产生含修饰蛋白的双层膜囊泡。
优选的,步骤(1)所述基础载体包括pEGFPC3、PCDH-CMV等真核表达载体。
优选的,步骤(3)饥饿诱导按如下步骤进行:a)将阳性克隆细胞接种至含10%胎牛血清DMEM培养基的10cm培养皿,37℃,5%CO2培养箱培养至80%的汇合度,更换无血清DMEM培养基,37℃,5%CO2培养箱进行饥饿诱导培养4h后,每皿添加终浓度100μmol/L的氯喹(CQ,溶酶体融合抑制剂),继续贴壁培养12小时,去除培养液,加入适量的PBS缓冲液冲洗,彻底去除胎牛血清;按每10cm培养皿(约107细胞)加入10ml外泌体专用无血清DMEM培养基,37℃,5%CO2培养箱培养48小时至细胞融合率70%以上;于3000g离心15min,收集细胞上清,重复离心两次去除细胞或细胞碎片;取最后一次上清采用100000g离心15min,收集沉淀并用PBS复溶,获得双层膜囊泡溶液;;
b)抗体亲和层析:将结合有修饰蛋白抗体的磁珠加入到步骤1)中的双层膜囊泡溶液中,在4℃下3000rpm离心,去除溶液上清,收集沉淀,用PBS溶液重悬并重复离心6次,收集最后一次离心的沉淀;所述结合有修饰蛋白抗体的磁珠,是指结合有结合有TAG抗体、ABP抗体或ASP抗体的磁珠;
c)双层膜囊泡的洗脱:步骤2)获得的沉淀采用PH=5.0的Tris-HCL缓冲液洗脱蛋白,于3000g离心15min,收集上清,获得含修饰蛋白的双层膜囊泡。
本发明还提供一种所述含修饰蛋白的双层膜囊泡在递送药物中的应用,所述药物包括蛋白多肽、核酸、蛋白多肽-核酸复合体、小分子药物或纳米材料。所述核酸包括但不限于DNA和RNA,其中RNA包括但不限于mRNA、干扰RNA(shRNA,siRNA)或sgRNA、circular RNA(circRNA)、long noncoding RNA(lncRNA),根据不同的药物底物,可分为串联适配体表达质粒、mRNA表达质粒、shRNA\miRNA表达质粒、sgRNA表达质粒、circRNA和lncRNA等核酸表达质粒。
递送药物的方法包括以下步骤:
(1)将药物递送基因与修饰蛋白编码基因共同与基础载体连接后,制得重组质粒A;或者将药物递送基因与基础载体连接后,制得重组质粒B;所述药物递送基因中包括药物编码基因、适配体核酸序列、ABP序列、药物受体编码基因中的一种或多种;所述药物编码基因为蛋白多肽、核酸或蛋白多肽-核酸复合体对应的编码核酸序列;
(2)通过真核细胞转染试剂将步骤(1)重组质粒A转入宿主细胞,或者将重组质粒B转入含有修饰蛋白编码基因的细胞,筛选阳性克隆细胞;
(3)将步骤(2)阳性克隆细胞饥饿诱导产生含修饰蛋白的双层膜囊泡;
(4)所述药物为蛋白多肽、核酸或蛋白多肽-核酸复合体时,用所述双层膜囊泡侵染细胞,释放药物;
所述药物为小分子药物或纳米材料时,通过含修饰蛋白的双层膜囊泡中的药物受体捕捉小分子药物和纳米材料装载入双层膜囊泡,装载药物的双层膜囊泡侵染细胞,释放药物;
所述修饰蛋白为标签蛋白TAG、适配体结合蛋白ABP或双层膜囊泡结构蛋白ASP的中的一种或多种可操作连接的融合蛋白。
本发明还提供一种含修饰蛋白的双层膜囊泡进行递送药物的方法,所述药物包括蛋白多肽、核酸、蛋白多肽-核酸复合体、小分子药物或纳米材料;
递送药物的方法包括以下步骤:
(1)将药物递送基因与修饰蛋白编码基因共同与基础载体连接后,制得重组质粒A;或者将药物递送基因与基础载体连接后,制得重组质粒B;所述药物递送基因中包括药物编码基因、适配体核酸序列、ABP序列、药物受体编码基因中的一种或多种;所述药物编码基因为蛋白多肽、核酸或蛋白多肽-核酸复合体对应的编码核酸序列;
(2)通过真核细胞转染试剂将步骤(1)重组质粒A转入宿主细胞,或者将重组质粒B转入含有修饰蛋白编码基因的细胞,筛选阳性克隆细胞;
(3)将步骤(2)阳性克隆细胞饥饿诱导产生含修饰蛋白的双层膜囊泡;
(4)所述药物为蛋白多肽、核酸或蛋白多肽-核酸复合体时,用所述双层膜囊泡侵染细胞,释放药物;
所述药物为小分子药物或纳米材料时,通过含修饰蛋白的双层膜囊泡中的药物受体捕捉小分子药物和纳米材料装载入双层膜囊泡,装载药物的双层膜囊泡侵染细胞,释放药物;
所述修饰蛋白为标签蛋白TAG、适配体结合蛋白ABP或双层膜囊泡结构蛋白ASP的中的一种或多种可操作连接的融合蛋白;
所述含修饰蛋白的双层膜囊泡是将修饰蛋白编码基因与基础载体连接后转入宿主细胞,诱导产生含修饰蛋白的双层膜囊泡;所述基础载体包括真核表达启动子、抗性基因和选择性标记基因。
所述步骤(3)中,饥饿诱导可按照前述步骤进行,其中步骤b)抗体亲和层析中,磁珠可以是结合有修饰蛋白抗体的磁珠或者结合有药物蛋白抗体的磁珠。
所述含有修饰蛋白编码基因的细胞可以通过将修饰蛋白编码基因与基础载体连接后转入宿主细胞获得。
更进一步,所述含有修饰蛋白编码基因的细胞按以下方法获得:
(1-1)表达修饰蛋白编码基因的质粒构建:所述修饰蛋白包括标签蛋白TAG、适配体结合蛋白ABP或双层膜囊泡结构蛋白ASP中的一种或多种的混合;优选所述修饰蛋白为纯化标签TAG、适配体结合蛋白ABP和双层膜囊泡蛋白ASP可表达连接的重组蛋白TAG/ABP/ASP,所述重组蛋白TAG/ABP/ASP不同肽段的线性顺序为以下之一:TAG-ABP-ASP、ABP-TAG-ASP、ABP-ASP-TAG、TAG-ASP-ABP、ASP-TAG-ABP或ASP-ABP-TAG;所述ABP包括MS2、COM、PP7、BoxB或PEG10;所述ASP包括但不限于LC3同源蛋白或ATG9同源蛋白,如LC3Aa、LC3Ab、LC3B、LC3C、GABARAP,GABARAPL1、GABARAPL2,ATG9等,以上ASP来源于但不限于物种人;所述TAG用于亲和层析或免疫沉淀法捕捉双层膜囊泡,该TAG包括Flag、Myc、HA、HIS或GST;

(1-1-1):合成表达修饰蛋白的编码基因;优选TAG/ABP/ASP的融合基因;
(1-1-2):将上述基因克隆至基础载体中,制得重组质粒(优选TAG/ABP/ASP重组质粒);所述基础载体包括真核表达启动子、抗性基因、选择性标记基因;在启动子后面接入表达修饰蛋白的编码基因(优选TAG/ABP/ASP的融合基因),由该启动子启动修饰蛋白(优选TAG/ABP/ASP融合蛋白)的表达;选择性标记基因包括但不限于嘌呤霉素(Puromycin);
(1-2)含重组蛋白的稳转细胞系的构建,包括以下步骤:
(1-2-1):通过真核细胞转染试剂把重组质粒转入目的细胞中,目的细胞包括但不限于293T细胞;
(1-2-2):通过筛选药物筛选出稳定表达修饰蛋白(优选TAG/ABP/ASP融合蛋白)的细胞系。
所述药物递送基因与基础载体连接,制备重组质粒B,一般按照以下之一步骤构建:
一:串联适配体重组质粒B
基础载体包括真核表达启动子、抗性基因、选择性标记基因;选择性标记基因包括但不限于嘌呤霉素(Puromycin);在基础载体的启动子后面接入单个或多个相同或不同aptamer序列的融合核酸序列,制备重组质粒B,能够实现多个底物ABP的结合;
二:ABP-多肽融合重组质粒B
在基础载体的启动子后面接入融合不同ABP核酸序列的多肽基因(ABP核酸序列-多肽基因)制备重组质粒B,由该启动子启动ABP-多肽融合表达,所述ABP-多肽能够结合到前述修饰双层膜囊泡中富集的串联适配体上,实现单种或多种多肽药物的同时富集装配;
三:核酸重组质粒B
在基础载体的启动子后面接入单个或多个RNA\aptamer融合基因,制备重组质粒B,RNA包括但不限于siRNA\microRNA\sgRNA\circRNA\lncRNA,由该启动子启动RNA\aptamer的表达,实现同一种RNA或不同种RNA的囊泡化药物载体内的富集,囊泡化药物载体包括但不限于双层膜囊泡;
进一步,构建方法为:合成适配体1-适配体2-适配体3……适配体n,或者核酸1-适配体1-核酸2-适配体2-核酸3-适配体3……核酸n-适配体n、或者适配体1-核酸1-适配体2-核酸2-适配体3-核酸3……适配体n-核酸n的核酸序列,其中n为1以上的整数,适配体包括但不限于MS2A(MS2 apatamer);将上述核酸序列克隆至基础载体的真核启动子之后,制得表达质粒B。
所述的适配体1-适配体2-适配体3……适配体n代表一个以上的适配体序列串联得到的融合序列;
所述的核酸1-适配体1-核酸2-适配体2-核酸3-适配体3……核酸n-适配体n、或者适配体1-核酸1-适配体2-核酸2-适配体3-核酸3……适配体n-核酸n的序列中,核酸1、核酸2、……核酸n各自独立代表一种DNA或RNA药物的序列,分别和对应的适配体序列依次串联,得到融合序列。
本发明双层膜囊泡能够通过ABP结合对应的核酸适配体(如PP7SL)连接的核酸或核酸-蛋白复合体;所述核酸适配体携带任意表达目标蛋白的核酸序列,包括但不限于shRNA,sgRNA,mRNA等核酸;所述蛋白复合体包括但不限于Cas9,转录因子等。
本发明还提供一种用于递送药物的表达质粒,所述表达载体包括递送捕捉mRNA、非编码RNA、环状RNA等所有RNA的表达质粒Flag-MS2-LC3、递送环状RNA的表达质粒pcDNA3.1MS2A-WT Split GFP、递送串联多底物结合的表达质粒
U6-Ms2A-ComA-PP7A-CMV-Com-RFP-H2A-PP7-GFP。
本发明提供的双层膜囊泡可将药物递送至靶细胞或靶向动物,用于制备靶向药物。
与现有技术相比,本发明有益效果主要体现在:
本发明提供了一种用于药物递送的含修饰蛋白的双层膜囊泡,采用抗原嵌合方式对双层膜囊泡进行修饰,构建双层膜囊泡蛋白(ASPs)与多肽的重组蛋白,多肽包括但不限于多肽药物、适配体结合蛋白(Aptamer-binding protein,ABP)、药物受体等。因此,修饰双层膜囊泡除了直接装载多肽药物,还可以通过结合的适配体(apatamer)把多肽药物、表达蛋白的核酸(mRNA)、敲低(miRNA,siRNA)、敲除(sgRNA)核酸、或其他一切形式的核酸装载入该结构,也可以通过药物受体捕捉小分子药物和纳米材料装载入双层膜囊泡。由于双层膜囊泡由细胞本身产生,因此其具有高效的药物富集能力,并且具有低免疫源性,可以通过其生物囊膜结构快速的与靶细胞融合,并把药物送至靶细胞。

具体实施方式
下面结合附图和具体实施例来对本发明的技术方案方案做出说明,但本发明的保护范围不限于此。实施例中所用到的仪器、试剂、耗材如无特殊说明,均可从商业公司购买得到。
本发明实施例所用DMEM培养基购自GBICO,货号0030034DJ。胎牛血清购自GBICO。人肾上皮细胞293T购自ATCC。
实施例1:GFP荧光蛋白的递送(绿色荧光图片)
1.pEGFP-LC3真核表达质粒构建
LC3基因片段:从NCBI上下载人源的LC3的基因序列(NM_022818),利用Primer 5.0软件设计特异性引物(表1),PCR扩增获得LC3的基因片段(核苷酸序列如SEQ ID NO.1所示)。
按照分子克隆的基本方法构建重组质粒:将获得LC3基因片段以及pEGFP-C3质粒(购自Addgene)用相应的快切酶(EcoR I和BAMH I)于37℃进行双酶切;酶切充分后,参照DNA清洁回收试剂盒(购自Sigma,型号G1N70)的说明书进行清洁回收,并测定各自浓度;然后将纯化的LC3基因片段和pEGFP-C3质粒于16℃进行连接,获得连接产物;取出大肠杆菌DH5α感受态细胞在冰上放置2min,将连接产物加在感受态中,冰上放置25min,42℃热击90s,在冰上再放置2min,涂于具有30μg/ml卡那霉素的LB平板。采用表1的引物进行菌液PCR初步验证,将结果为阳性的菌液送杭州尚亚公司做进一步的测序验证。经测序结果显示插入的LC3基因片段序列完全正确,可用于后续研究。从测序正确的阳性克隆中提取pEGFP-LC3质粒。

2.pEGFP-LC3稳转细胞系的构建:
2.1人肾上皮细胞293T的培养
1)预先加热水浴锅,温度至37-40℃,并在离心管中准备好10ml的DMEM培养基(购自GBICO,0030034DJ);
2)从液氮罐或冰箱中取出人肾上皮细胞293T,迅速放进37℃预热的水浴锅中,镊子夹住冻存管晃动,使其受热均匀;
3)当步骤2)冻存管内完全融化时,注意用酒精擦拭冻存管,将液体倒入步骤1)含有10ml的DMEM培养基的离心管中;
4)步骤3)离心管800rpm离心5min,去除上清,得到沉淀;
5)用DMEM培养基悬浮步骤4)沉淀,接种至含有10%胎牛血清的5mL DMEM的T25细胞培养瓶中,置于37℃,5%CO2培养箱中进行常规培养,48小时后可观察到95%的细胞贴壁生长,细胞状态良好,说明细胞复苏成功。
2.2.pEGFP-LC3质粒转染
以Lipofectamine 3000转染试剂(购自Invitrogen)为例进行瞬时转染,具体如下:
1)将复苏后常规培养的293T细胞按照1-3x105接种到6孔板中,加入2-4ml的DMEM培养基,混匀,在37℃,5%CO2培养箱中过夜;
2)无菌状态下配置如下溶液:
用125μL的无血清DMEM培养基稀释5μg的pEGFP-LC3质粒,获得质粒溶液,记为a溶液;用125μL的无血清DMEM培养基稀释10μL的Lipofectamine3000转染试剂,获得转染试剂溶液,记为b溶液;
3)将125μL质粒溶液和125μL转染试剂溶液混合并摇匀,室温下放置30min,获得ab溶液;
4)步骤1)293T细胞培养至80%单层左右,用无血清DMEM培养基洗涤细胞2次,每孔加入1ml的无血DMEM清培养基,并将步骤3)混合后的250μL的ab溶液逐滴加入到每孔,按十字方向轻摇混匀,37℃,5%CO2培养箱培养24小时;
5)将步骤4)转染液倒出,换为含有10%胎牛血清的DMEM培养基,继续培养72h,获得转染细胞。
2.3稳定细胞系的建立
步骤2.2培养72h后的转染细胞,加入选择性新霉素(Neomycin)进行稳定细胞株筛选,预实验确定新霉素的最佳浓度,确定新霉素对所选细胞的最低作用浓度,具体操作如下:
1)步骤2.2的5)获得的转染细胞接种至添加有10%胎牛血清DMEM培养基的24孔板中,37℃,5%CO2培养箱过夜培养,待第二天长成25%单层为宜。
2)第二天将培养液换成含不同浓度(0,50,100,200,400,800,1000ug/ml)新霉素的10%胎牛血清DMEM培养基。
3)37℃培养15天,以绝大多数细胞死亡时新霉素浓度为准,结果为400-800ug/ml,筛选细胞时可适当提高浓度,确定新霉素适宜浓度为600ug/ml。
4)步骤2.2的5)获得的转染细胞按照体积比1:10的比例接种至含600ug/ml新霉素的10%胎牛血清DMEM培养基,37℃,5%CO2培养箱传代培养72h。然后有限稀释法挑选单克隆。有限稀释法:将细胞消化下来做连续的10倍稀释,每稀释一梯度都在9孔板中培养,生长一周左右再次挑取单克隆进行培养,如此反复3次。
5)Western blot检测蛋白的表达情况,挑取多个单克隆进行表达检测,筛选出表达量最高的克隆,如图2所示,选取1号细胞系,记为GFP-LC3细胞系,传代保存。
3、饥饿诱导双层膜囊泡产生
上述GFP-LC3细胞系接种至含10%胎牛血清DMEM培养基的10cm培养皿,37℃,5%CO2培养箱培养至80%的汇合度,更换无血清DMEM培养基,37℃,5%CO2培养箱进行饥饿诱导培养4h后,每皿添加终浓度100μmol/L的氯喹(CQ,溶酶体融合抑制剂),继续贴壁培养12小时,取活细胞样品进行荧光显微镜观察,荧光图见图3所示,此时细胞有大量带有绿色荧光的双层膜囊泡产生,包括自噬体或LC3依赖的外泌体。
4、双层膜囊泡分离
步骤3贴壁培养12小时后,去除培养液,加入适量的PBS缓冲液冲洗,彻底去除胎牛血清;按每10cm培养皿(约107细胞)加入10ml外泌体专用无血清DMEM培养基,37℃,5%CO2培养箱培养48小时至细胞融合率70%以上;于3000g离心15min,收集细胞上清,重复离心两次去除细胞或细胞碎片;取最后一次上清采用100000g离心15min,收集沉淀并用PBS复溶,获得双层膜囊泡溶液;通过透射电镜观察,结果见图4,发现大量双层膜囊泡被提纯出来。
5.将偶连GFP抗体的磁珠(ABclonal,AE079)加入到步骤4中的双层膜囊泡溶液中,在4℃下3000rpm离心,去除溶液上清,收集沉淀,用PBS溶液重悬并重复离心6次,收集最后一次离心的沉淀。
6.步骤5获得的沉淀采用PH=5.0的Tris-HCL缓冲液洗脱GFP-LC3蛋白,于3000g离心15min,收集上清,获得携带有GFP-LC3的双层膜囊泡。
7.将293T细胞接种至含10%胎牛血清的DMEM培养基的24孔板,在37℃,5%CO2培养箱培养至80%汇合度,每孔加入100ul步骤6获得的双层膜囊泡,继续培养48小时以后,用荧光显微镜观察,结果见图5所示,结果显示有73%的细胞含有绿色荧光,说明GFP蛋白被成功传递。
实施例2:GFP mRNA的递送
1、制备Flag-MS2-LC3质粒:
按照分子克隆的基本方法构建重组质粒:将实施例1获得的LC3片段(核苷酸序列如SEQ ID NO.1所示)以及lenti Flag-MS2载体(购自爱博泰克生物技术有限公司),分别用相应的快切酶EcoR I和KPN I于37℃进行双酶切;酶切充分后,参照DNA清洁回收试剂盒的说明书进行清洁回收,并测定各自浓度;然后将纯化的LC3片段和lenti Flag-MS2载体于16℃进行连接,获得Flag-MS2-LC3质粒;取出E coli.DH5α感受态细胞在冰上放置2min,将连接产物Flag-MS2-LC3质粒加在感受态细胞中,冰上放置25min,42℃热击90s,在冰上再放置2min,涂于具有80μg/ml氨苄抗性的LB平板上。用表2引物进行菌液PCR初步验证,将结果为阳性的菌液送杭州尚亚公司做进一步的测序验证。经测序结果显示插入的LC3基因片段序列完全正确,可用于后续研究。

2、Flag-MS2-LC3细胞系构建
同实施例1步骤2中GFP-LC3细胞系的方法进行细胞系构建。简单来说,Flag-MS2-LC3质粒上带有潮霉素(Hygro)抗性。该质粒转染293T细胞48小时后通过Hygro药物进行筛选,通过有限稀释获得单克隆细胞,即为Flag-MS2-LC3细胞系。
3、pCDH-GFP-MS2A载体稳转Flag-MS2-LC3细胞系
参考实施例1的步骤2.2质粒转染方法,将pCDH-GFP-MS2A质粒(购自豪普斯生物有限公司,MS2A基因核苷酸序列如SEQ ID NO.2所示)转染入Flag-MS2-LC3细胞系中,采用实施例1步骤3的方法饥饿诱导双层膜囊泡产生,并采用实施例1步骤4-步骤6方法分离,获得带有GFP-MS2A RNA的MS2-LC3的双层膜囊泡;采用实施例1步骤7的方法进行荧光检测,结果见图6所示,结果显示有68%的细胞含有绿色荧光,说明GFP mRNA被成功传递。
实施例3:GFP基因的敲除载体sgRNA-CAS9递送
1.构建MS2A-sgRNA-Cas9表达质粒
根据GFP基因设计并替换载体lentiCRISPR v2(购自Addgene)的sgRNA(表3),然后把MS2A基因(核苷酸序列如SEQ ID NO.2所示)插入到载体lentiCRISPR v2的替换后的sgRNA scafold RNA的两个stem loop中,并构建成U6-MS2A-sgRNA-CMV-Cas9载体,记为MS2A-sgRNA-Cas9表达质粒,序列见SEQ ID NO.3,其中1-181bp为启动子U6,2071-2212bp为MS2A-scaldfoldRNA,2213-2510bp为EF1a,2511-4117bp为CAS9-NLS-FLAG。
2.MS2A-sgRNA-Cas9自噬体的制备
采用实施例1步骤2.2的方法,将步骤1构建的U6-MS2A-sgRNA-CMV-Cas9载体转染至实施例2步骤2制备的Flag-MS2-LC3细胞系中,采用实施例1步骤3到步骤6的方法制备含MS2A-sgRNA-Cas9的自噬体(双层膜囊泡)。
3.GFP敲除效果验证
取实施例1构建的GFP-LC3细胞系接种至含10%胎牛血清的DMEM培养基,在37℃,5%CO2培养箱培养至汇合度75-80%。采用实施例1步骤2.2的方法,将步骤2获得的含MS2A-sgRNA-Cas9的自噬体转染至GFP-LC3细胞系。将转染细胞在含10%胎牛血清的DMEM培养基,在37℃,5%CO2培养箱培养48小时后,采用荧光显微镜进行荧光观察,结果见图7,外泌体转染后导致绿色荧光从88%降到6%,说明GFP基因的sgRNA敲除基因被成功传递,细胞中的GFP基因被成功大量敲除。

实施例4:GFP环状RNA(CircRNA)的递送
1.构建MS2A-CircRNA-Flag表达质粒
按照分子克隆的基本方法构建重组质粒:将MS2A基因(核苷酸序列如SEQ ID NO.2所示)和pcDNA3.1(+)ZKSCAN1 MCS-WT Split GFP载体(购自Addgene)采用内切酶ClaI和AgeI于37℃双酶切;酶切充分后,参照DNA清洁回收试剂盒的说明书进行清洁回收,并测定各自浓度;连接、转化、单克隆筛选方法参照实施例1。经测序结果显示插入的基因片段序列完全正确,命名为pcDNA3.1 MS2A-WT Split GFP可用于后续研究。
2.含GFP基因的CircRNA的自噬体
采用实施例2步骤2.2的方法,将上述pcDNA3.1 MS2-WT Split GFP转染至实施例2中制备的Flag-MS2-LC3细胞系中,采用实施例1步骤4-6的方法制备得到含GFP基因的CircRNA的自噬体。
3.采用实施例1步骤7的方法,将步骤2中得到的自噬体转染293T细胞,细胞培养48小时以后,置细胞于荧光显微镜下进行观察,孵育完之后用荧光显微镜观察,结果见图8所示。结果显示有72%细胞显示出绿色荧光。说明表达GFP的环状RNA被成功传递。
实施例5:多底物递送
1.构建U6-Ms2A-ComA-PP7A-CMV-Com-RFP-H2A-PP7-GFP表达质粒

如表4所示,合成串联适配体序列U6-MS2A-ComA-PP7A-CMV-COM-RFP-H2A-PP7-GFP,序列见SEQ ID NO.4,并构建到真核表达载体PCDNA3.1上,形成
pcDNA3.1-U6-MS2A-ComA-PP7A-CMV-COM-RFP-H2A-PP7-GFP,序列见SEQ ID NO.4,组成见表5。

采用实施例1步骤2.2的方法,将上述
pcDNA3.1-U6-MS2A-ComA-PP7A-CMV-COM-RFP-H2A-PP7-GFP转染至实施例2中制备的Flag-MS2-LC3细胞系中。采用实施例1步骤4到步骤6方法,制备含
U6-MS2A-ComA-PP7A-CMV-COM-RFP-H2A-PP7-GFP的外泌体。
3.采用实施例1步骤7的方法,将步骤2中得到的外泌体转染293T细胞,细胞培养48小时以后,置细胞于荧光显微镜下进行观察,孵育完之后用荧光显微镜观察,结果见图9所示,结果显示有92%的细胞同时有绿色和红色荧光。说明表达RFP和GFP蛋白的mRNA被成功传递至子代细胞中。
Claims (10)
1.一种用于药物递送的含修饰蛋白的双层膜囊泡,其特征在于所述含修饰蛋白的双层膜囊泡是将修饰蛋白编码基因与基础载体连接后转入宿主细胞,诱导产生含修饰蛋白的双层膜囊泡;所述修饰蛋白包括标签蛋白TAG、适配体结合蛋白ABP或双层膜囊泡结构蛋白ASP中的一种或多种的混合;所述基础载体包括真核表达启动子、抗性基因和选择性标记基因。
2.如权利要求1所述的用于药物递送的含修饰蛋白的双层膜囊泡,其特征在于所述标签蛋白TAG用于亲和层析或免疫沉淀法捕捉双层膜囊泡,包括Flag标签、Myc标签、HA标签、HIS标签或GST标签中的一种或多种;所述适配体结合蛋白ABP包括MS2蛋白、COM蛋白、PP7蛋白、BoxB蛋白或PEG10蛋白中的一种或多种;所述双层膜囊泡结构蛋白ASP包括LC3同源蛋白或ATG9同源蛋白中的一种或多种,所述ABP通过ASP结合于双层膜囊泡内膜上。
3.如权利要求1或2所述的含修饰蛋白的双层膜囊泡的制备方法,其特征在于所述方法包括以下步骤:(1)将修饰蛋白编码基因克隆至基础载体中,制得重组质粒;所述基础载体包含真核表达启动子、抗性基因、选择性标记基因;在启动子后面接入修饰蛋白编码基因;
(2)通过真核细胞转染试剂将步骤(1)重组质粒转入目的细胞中,筛选阳性克隆细胞;
(3)将步骤(2)阳性克隆细胞饥饿诱导产生含修饰蛋白的双层膜囊泡。
4.如权利要求1或2所述的所述含修饰蛋白的双层膜囊泡在递送药物中的应用,所述药物包括蛋白多肽、核酸、蛋白多肽-核酸复合体、小分子药物或纳米材料。
5.如权利要求4所述的应用,其特征在于所述递送药物的方法包括以下步骤:
(1)将药物递送基因与修饰蛋白编码基因共同与基础载体连接后,制得重组质粒A;或者将药物递送基因与基础载体连接后,制得重组质粒B;所述药物递送基因中包括药物编码基因、适配体核酸序列、ABP序列、药物受体编码基因中的一种或多种;所述药物编码基因为蛋白多肽、核酸或蛋白多肽-核酸复合体对应的编码核酸序列;
(2)通过真核细胞转染试剂将步骤(1)重组质粒A转入宿主细胞,或者将重组质粒B转入含有修饰蛋白编码基因的细胞,筛选阳性克隆细胞;
(3)将步骤(2)阳性克隆细胞饥饿诱导产生含修饰蛋白的双层膜囊泡;
(4)所述药物为蛋白多肽、核酸或蛋白多肽-核酸复合体时,用所述双层膜囊泡侵染细胞,释放药物;
所述药物为小分子药物或纳米材料时,通过含修饰蛋白的双层膜囊泡中的药物受体捕捉小分子药物和纳米材料装载入双层膜囊泡,装载药物的双层膜囊泡侵染细胞,释放药物。
6.如权利要求5所述的应用,其特征在于所述含有修饰蛋白编码基因的细胞通过将修饰蛋白编码基因与基础载体连接后转入宿主细胞获得。
7.如权利要求5所述的应用,其特征在于所述药物递送基因与基础载体连接,制备重组质粒B,按照以下之一步骤构建:
一:串联适配体重组质粒B
基础载体包括真核表达启动子、抗性基因、选择性标记基因;选择性标记基因包括但不限于嘌呤霉素;在基础载体的启动子后面接入单个或多个相同或不同aptamer序列的融合核酸序列,制备重组质粒B,能够实现多个底物ABP的结合;
二:ABP-多肽融合重组质粒B
在基础载体的启动子后面接入融合不同ABP核酸序列的多肽基因制备重组质粒B,由该启动子启动ABP-多肽融合表达,所述ABP-多肽能够结合到前述修饰双层膜囊泡中富集的串联适配体上,实现单种或多种多肽药物的同时富集装配;
三:核酸重组质粒B
在基础载体的启动子后面接入单个或多个RNA\aptamer融合基因,制备重组质粒B,RNA包括但不限于siRNA\microRNA\sgRNA\circRNA\lncRNA,由该启动子启动RNA\aptamer的表达,实现同一种RNA或不同种RNA的囊泡化药物载体内的富集,囊泡化药物载体包括但不限于双层膜囊泡。
8.如权利要求7所述的应用,其特征在于所述构建方法为:合成适配体1-适配体2-适配体3……适配体n,或者核酸1-适配体1-核酸2-适配体2-核酸3-适配体3……核酸n-适配体n、或者适配体1-核酸1-适配体2-核酸2-适配体3-核酸3……适配体n-核酸n的核酸序列,其中n为1以上的整数,适配体包括但不限于MS2A;将上述核酸序列克隆至基础载体的真核启动子之后,制得表达质粒B;
所述的适配体1-适配体2-适配体3……适配体n代表一个以上的适配体序列串联得到的融合序列;
所述的核酸1-适配体1-核酸2-适配体2-核酸3-适配体3……核酸n-适配体n、或者适配体1-核酸1-适配体2-核酸2-适配体3-核酸3……适配体n-核酸n的序列中,核酸1、核酸2、……核酸n各自独立代表一种DNA或RNA药物的序列,分别和对应的适配体序列依次串联,得到融合序列。
9.如权利要求1或2所述的所述含修饰蛋白的双层膜囊泡在制备靶向药物中的应用。
10.一种用于递送药物的表达质粒,其特征在于所述表达质粒包括递送捕捉RNA的表达载体Flag-MS2-LC3、递送环状RNA的表达质粒pcDNA3.1 MS2A-WT Split GFP、递送串联多底物结合的表达质粒U6-Ms2A-ComA-PP7A-CMV-Com-RFP-H2A-PP7-GFP。
