技术领域
本发明属于分子病毒学和免疫学领域,具体来说,本发明涉及可特异性结合人乳头瘤病毒(HPV6)L1蛋白抗体或其抗原结合片段、编码它们的核酸、载体、细胞,所述抗体或其抗原结合片段在检测HPV6 L1蛋白中的应用,以及使用他们检测或定量测定HPV6 L1蛋白的方法。
背景技术
人乳头状瘤病毒(human papilloma virus,HPV)是一种嗜上皮性、高度的组织特异性、无包膜DNA病毒。其DNA的晚期区(L)开放阅读框编码主要衣壳蛋白L1和次要衣壳蛋白L2;L2蛋白变异度较高,与病毒抗原的多态性有关;而L1蛋白保守性高,是主要的型特异性抗原,可以诱导机体产生中和抗体,常用作HPV疫苗的抗原。目前已鉴定超过100余型HPV,不同亚型的HPV有不同的组织偏好并引发从良性疣到上皮细胞肿瘤(包括宫颈、阴道、阴门、肛门和咽喉等部位)的各种疾病。
宫颈癌在全球女性癌症死亡率中位居第二位,且病发趋势呈年轻化。15种高风险(HR)HPV型可导致宫颈、肛门、阴茎、阴道、外阴和口咽癌。其中,HPV-16和HPV-18型是迄今最为常见的癌症起因,约占宫颈癌的70%,其余为其他HR-HPV型(31、33、35、39、45、51、52、56、58、59、68、73和82)引起。HPV-16约占HPV阳性口咽癌(OPCs)的95%。持续低风险基因型HPV-6和HPV-11导致大多数肛门生殖器疣和呼吸道乳头状瘤,但很少与癌症相关(HumanPapillomavirus in Cervical Cancer and Oropharyngeal Cancer:One Cause,TwoDiseases Tara A.Bermanand John T.Schiller,PhD2 Cancer2017;123:2219-29)。
宫颈癌疫苗是人类健康的需求。而L1蛋白可自我装配形成病毒样颗粒(VLP)(Structure of Small Virus-like Particles Assembled from the L1 Protein ofHuman Papillomavirus 16Chen,X.S.,R.L.Garcea,Mol.Cell.5(3):557-567,2000)似乎是乳头瘤病毒疫苗的优秀候选者。VLP形态上类似于真的病毒体且在施用于动物或人后能够诱导高效价的中和抗体。因为VLPs不含有潜在的致癌病毒基因组,他们在HPV疫苗开发中安全地替代了活病毒(综述参见Schiller和Hidesheim,J.Clin.Virol.19:67-74(2000))。
葛兰素公司生产的双价重组HPV疫苗(https://www.fda.gov/downloads/BiologicsBloodVaccines/Vaccines/ApprovedProducts/UCM186981.pdf)以及默克公司生产的4价重组HPV疫苗/>(https://www.fda.gov/vaccines-blood-biologics/vaccines/gardasil)和9价重组HPV疫苗/>重组疫苗(https://www.fda.gov/vaccines-blood-biologics/vaccines/gardasil-9)均为VLP疫苗。
鉴于宫颈癌的高发病率和高致死率,仍然需要研发更多低价、安全和高效的新型HPV VLP疫苗。HPV病毒样颗粒分子量大,结构复杂,其完整性和结构正确与否会直接影响免疫和保护效果,因此本领域仍然需要针对特定HPV型别、甚至是针对特定完整性和结构正确的HPV VLP的特异性中和抗体,可以灵敏且高度特异性检测HPV病毒样颗粒,以期对生产的疫苗及其保护效果进行更好的评价。
发明内容
本发明的目的是为了提供针对特定HPV型别、甚至是针对特定完整性和结构正确的HPV VLP的特异性抗体,提供抗HPV6 L1蛋白抗体或其抗原结合片段,该抗体及其抗原结合片段具有高特异性。
本发明的第一方面涉及一种抗HPV6 L1蛋白抗体或其抗原结合片段,包含轻链可变区或其部分和/或重链可变区或其部分,
其中,轻链可变区或其部分包含氨基酸序列为SEQ ID NO:5的轻链CDR1、氨基酸序列为SEQ ID NO:6的轻链CDR2和氨基酸序列为SEQ ID NO:7的轻链CDR3的一个或多个;且
重链可变区或其部分包含氨基酸序列为SEQ ID NO:8的重链CDR1、氨基酸序列为SEQ ID NO:9的重链CDR2和氨基酸序列为SEQ ID NO:10的重链CDR3的一个或多个。
在一个实施方案中,所述抗体或其抗原结合片段包含:与抗HPV6 L1蛋白抗体轻链可变区氨基酸序列SEQ ID NO:20具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列,和/或与抗HPV6 L1蛋白抗体重链可变区氨基酸序列SEQ ID NO:12具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列。
在一个实施方案中,所述抗体进一步包含轻链恒定区和重链恒定区。在一个优选的实施方案中,轻链恒定区为与抗HPV6 L1蛋白抗体轻链恒定区氨基酸序列SEQ ID NO:24具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列,和/或重链恒定区为与抗HPV6 L1蛋白抗体重链恒定区氨基酸序列SEQ ID NO:16具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列。
在一个实施方案中,所述抗体为单克隆抗体。
在一个实施方案中,所述抗体与HPV6 L1抗原特异性结合,与其他型别的HPV L1抗原无交叉反应;优选地,其他型别HPV为HPV11、16、18、31、33、35、39、45、51、52、56、58和59型。
在一个实施方案中,抗原结合片段的形式为Fv、Fab、Fab′、Fab′-SH、F(ab′)2、Fd片段、Fd'片段、单链抗体分子或单域抗体;其中,单链抗体分子优选为scFv、di-scFv、tri-scFv、双体抗体或scFab。
所述抗体可以为分离的抗体。
本发明的第二方面涉及一种核酸,其包含:编码第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段的核苷酸序列。所述核酸可以为分离的核酸。
本发明的第三方面涉及一种载体,其包含第二方面所述的核酸。
本发明的第四方面涉及一种细胞,其包含第二方面所述的核酸,和/或第三方面所述的载体。所述细胞可以为分离的细胞。
本发明的第五方面涉及第一方面所述的抗体或其抗原结合片段在检测HPV6 L1蛋白中的应用。
本发明的第六方面涉及一种用于检测样品中HPV6 L1蛋白存在的方法,包括:
a)将第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段包被到测定表面上;
b)使所述测定表面与待测样品接触一段时间,该时间段足以容许结合,然后清洗所述测定表面;
c)使所述测定表面与同报告基团缀合的第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段接触一段时间,该时间段足以容许缀合抗体的结合,然后清洗所述测定表面;并
d)检测报告基团信号。
本发明的第七方面涉及一种用于定量测定HPV6 L1蛋白的方法,包括:
a)将第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段包被到测定表面上;
b)使所述测定表面与待测样品接触一段时间,该时间段足以容许结合,然后清洗所述测定表面;
c)使所述测定表面与同报告基团缀合的第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段接触一段时间,该时间段足以容许缀合抗体的结合,然后清洗所述测定表面;并且
d)通过与标准曲线比较来量化所述HPV6 L1蛋白。
在一个实施方案中,标准曲线通过如下步骤建立:
a)将第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段包被到测定表面上;
b)使所述测定表面与一系列浓度的HPV6-L1蛋白标准品接触一段时间,该时间段足以容许结合,然后清洗所述测定表面;
c)使所述测定表面与同报告基团缀合的第一方面所述的抗HPV6 L1蛋白抗体或其抗原结合片段接触一段时间,该时间段足以容许缀合抗体的结合,然后清洗所述测定表面;并测定其吸光度;且
d)以标准品的浓度为纵坐标,其相应的吸光度为横坐标,绘制标准曲线。
其中步骤(a)和(c)中的第一方面所述的抗体或其抗原结合片段可以相同或不同。
在一个实施方案中,其中的报告基团是辣根过氧化物酶。
在本发明中,如无特殊说明,均采用本领域常规的手段和方式。
与现有技术相比,本发明筛选得到的抗HPV6 L1蛋白抗体及其抗原结合片段具有显著的技术效果,特别的,本发明的抗体及其抗原结合片段与HPV6 L1抗原结合力强,且具有高特异性,不识别HPV11、16、18、31、33、35、39、45、51、52、56、58和59型。其与50ng/mL的上述13型的混合物无交叉反应,适合作为ELISA定量中的检测抗体进行HPV疫苗的免疫原性评价。

定义
除非另有说明,本文使用的所有技术和科学术语具有本发明所属的技术领域的普通技术人员通常理解的含义。为了便利地理解本发明,引述以下术语的通常的含义。
术语“抗体”意指免疫球蛋白分子,是指表现所需生物学活性的抗体的任何形式。包括但不限于单克隆抗体(包括全长单克隆抗体)、多克隆抗体和多特异性抗体(例如双特异性抗体),甚至包括抗体片段。典型地,全长抗体结构优选包含4条多肽链,通常通过二硫键相互连接的2条重(H)链和2条轻(L)链。每条重链包含重链可变区和重链恒定区。每条轻链包含轻链可变区和轻链恒定区。在此典型全长抗体结构外,其结构还包括其他衍生形式。
术语“互补决定区”(CDR,例如CDR1、CDR2和CDR3)是指抗体可变区的这样一些氨基酸残基,其存在对于抗原结合来说是必需的。每个可变区通常具有3个被鉴别为CDR1、CDR2和CDR3的CDR区域。每个互补决定区可包含来自如Kabat所定义的“互补决定区”的氨基酸残基(Kabat et al.,Sequences of Proteins of Immulological Interest,5thEd.PublicHealth Service,National Institutes of Health,Bethesda,MD.1991))和/或来自“高变环”的那些残基(Chothia and Lesk;J Mol Biol 196:901-917(1987))。
每个重链可变区和轻链可变区通常包含3个CDR和最多达4个FR,所述CDR和FR从氨基末端至羧基末端以例如以下顺序排列:FR1、CDR1、FR2、CDR2、FR3、CDR3、FR4。
给定抗体的互补性决定区(CDR)可以使用Kabat体系标识(Kabat等:Sequences ofProteins of Immunological Interest,第5版,美国卫生和公众服务部,PHS,NIH,NIH出版编号91-3242,1991)。
“抗体的抗原结合片段”包含完整抗体分子的一部分,其保留母体抗体的至少某些结合特异性,通常包括至少部分母体抗体的抗原结合区或可变区(例如一个或多个CDR)。抗原结合片段的实例包括但不限于Fv、Fab、Fab′、Fab′-SH、F(ab′)2、Fd片段、Fd'片段、单链抗体分子(例如scFv,di-scFv或tri-scFv、双体抗体或scFab)、单域抗体。
“单克隆抗体”是指获自基本上同质的抗体群体的抗体,即,所述包含单一抗体的群体除了可能以极少量存在的可能突变(例如天然突变)之外是相同的。因此,所述术语“单克隆”表明所述抗体的性质,即不是不相关抗体的混合物。与通常包括针对不同决定簇(表位)的不同抗体的多克隆抗体制剂相反,单克隆抗体制剂的每个单克隆抗体均针对抗原上的单独一个决定簇。除了其特异性之外,单克隆抗体制剂的优点在于它们通常不会被其他抗体污染。所述术语“单克隆”不应被理解为需要通过任何特定的方法产生所述抗体。所述术语单克隆抗体具体地包括嵌合抗体、人源化抗体和人抗体。
抗体“特异性结合”目的抗原例如肿瘤相关的多肽抗原靶(本文中,HPV6 VLP蛋白),即以足够的亲和力结合所述抗原以使得所述抗体可用作治疗性试剂,靶向表达所述抗原的细胞或组织,并且与其他蛋白质无显著交叉反应或者与除了上文提到的抗原靶的同源体和变体(例如突变形式、剪接变体,或蛋白水解作用截短的形式)以外的蛋白质无显著交叉反应。
术语“表位”包括能够特异性结合至抗体或T细胞受体的任何蛋白质决定簇。表位决定簇通常由分子的化学活性表面基团(例如氨基酸或糖侧链,或其组合)组成,并且通常具有特定三维结构特征以及特定的电荷特征。
“分离的”抗体是已经被鉴别并且从表达它的细胞的组分中分离的抗体。所述细胞的污染组分是会干扰所述抗体的诊断或治疗用途的物质,可能包括酶、激素和其他蛋白质性质或非蛋白质性质的溶质。分离的天然抗体包括重组细胞内的原位抗体,因为所述抗体的天然环境中的至少一种组分是不存在的。然而,通常情况下,分离的抗体是通过至少一个纯化步骤进行制备。
术语“HPVL1蛋白”如本文所用,术语“HPV”和“人乳头状瘤病毒”是指乳头状瘤病毒科的无包膜双链DNA病毒。它们的基因组是圆形的,并且大小约为8千碱基对。大多数HPV编码八种主要蛋白,六种位于“早期”区域(E1-E2),并且两种位于“晚期”区域(L1(主要衣壳蛋白)和L2(次要衣壳蛋白))。已经鉴定了超过120种HPV类型,并且它们由数字标出(例如,HPV-16、HPV-18等)。
术语“HPV”或“HPV病毒”指乳头状瘤病毒科的乳头状瘤病毒,为无包膜DNA病毒,该病毒基因组为双链闭环DNA,大小约为8kb,通常可以分为三个区域:①早期区(E),含有编码E1、E2、E4~E7病毒复制,转录及转化有关的非结构蛋白的6个开放阅读框,以及E3和E8开放阅读框;②晚期区(L)含有编码主要衣壳蛋白L1和次要衣壳蛋白L2的阅读框;③长调控区(LCR)不编码任何蛋白,但具有复制的起源以及多个转录因子结合位点。
术语“HPVL1蛋白”及“HPVL2蛋白”指由HPV基因的晚期区(L)编码,在HPV感染周期中晚期合成的蛋白。L1蛋白质是主要的衣壳蛋白并且具有55-60kDa的分子量。L2蛋白质是次要的衣壳蛋白质。72个L1五聚体构成二十面体HPV病毒粒子的外壳,包裹闭环双链DNA微染色体。L2蛋白质位于L1蛋白质内侧(Structure of Small Virus-like ParticlesAssembled from the L1 Protein of Human Papillomavirus 16Chen,X.S.,R.L.Garcea,Mol.Cell.5(3):557-567,2000)。
术语“HPV VLP蛋白”指当重组表达L1蛋白时,L1蛋白可自我装配形成病毒样颗粒(VLP蛋白),为大约含有72个L1五聚体集合体,与病毒体外壳相似。VLP蛋白可以在接种动物中诱导中和抗体,保护实验动物免受感染性病毒的随后攻击。因此,VLP蛋白似乎是乳头瘤病毒疫苗的优秀候选者。(Structure of Small Virus-like Particles Assembled fromthe L1 Protein of Human Papillomavirus 16Chen,X.S.,R.L.Garcea,Mol.Cell.5(3):557-567,2000)。
术语"报告基团"一般指的是产生可检测的辐射,例如荧光或发光辐射,的发射的部分,包括但不限于酶或者是放射性同位素。当报告基团为酶诸如碱性磷酸酶、辣根过氧化物酶或β-D-半乳糖苷酶时,合适的底物在与酶起反应时产生颜色变化,使用分光光度计定量颜色强度的测量。或当报告基团为放射性同位素时,使用适当的γ或β射线检测仪来量化报告基团。报告基团的强度与测试样品中待测物质的量正相关。
以下将通过实施例对本发明进行详细描述。
实施例1:抗体库的构建和筛选分离
1.动物免疫与效价检测
1.1动物免疫
取昆虫细胞系统表达的HPV6 VLP蛋白(SEQ ID NO.1,蛋白序列号:P69898&P06416),分别与弗氏完全佐剂(Sigma公司,货号F5881)和弗氏不完全佐剂(Sigma公司,货号F5506)乳化制成弗氏完全佐剂免疫原和弗氏不完全佐剂免疫原,其中HPV6VLP(500ug/只)与弗氏完全佐剂、弗氏不完全佐剂的体积比例为1:1。
通过背部皮下多点注射的方式,用弗氏完全佐剂免疫原免疫日本大耳白兔(2-2.2kg,购自北京顺东养殖有限公司)。首次免疫后,每间隔2周不完全弗氏佐剂免疫原以相同的方法和剂量对动物进行加强免疫。第四次免疫后第4天经耳缘静脉采血,测定血清效价(步骤见实施例1.2)。以(抗血清吸光值-空白吸光值)/(免疫前阴性对照血清吸光值-空白吸光值)>2.1为血清效价阳性标准,按照该标准,血清效价达到1:25000的动物于末次免疫后一周,取脾及骨髓组织建库筛选兔单抗。
1.2血清学检测效价
取适量HPV6 VLP蛋白,用包被液(0.05M Na2CO3,0.05M NaHCO3,pH 9.6,经0.2μm无菌过滤)稀释成0.1μg/mL,然后用单道移液器在96孔板每孔中加入100μL,轻拍板子使样品混匀,用保鲜膜封严,4℃下包被过夜;用洗涤液(含0.05% Tween20的TBS,pH 7.2-7.4)按200μL/孔洗板1次,扣干酶标板;用封闭液(含2% BSA的洗涤液)按300μL/孔封闭酶标板,室温下封闭1小时;用洗涤液按300μL/孔洗板2次,扣干酶标板;将待检兔血清用样品稀释剂进行梯度稀释,将1:25000、1:125000两个稀释倍数的样品及样品稀释剂以100μL/孔加样,以100μL/孔加入辣根酶标记山羊抗兔IgG(H+L)检测抗体(IR公司,货号:111-035-008)至96孔板内,室温下作用2小时;用洗涤液按200μL/孔洗板3次,扣干酶标板;以200μL/孔加入显色液(用底物稀释液将底物储液做1000倍稀释,每升稀释后缓冲液加入320μL0.75% H2O2,混合均匀后使用)室温放置12分钟;以50μL/孔加入终止液(2M H2SO4)终止反应;用酶标仪进行检测:测定波长为450nm。
2.噬菌体抗体库制备与筛选
兔子的脾和骨髓组织用TriPure Isolation Reagent试剂(来源:Roche)进行RNA提取,用反转录试剂盒(来源:Invitrogen)进行反转录获得cDNA,PCR扩增兔抗体的轻链可变区序列和重链可变区序列(引物参考文献:Rader et al.,2000),轻链PCR引物见表1,重链PCR引物见表2,采用重叠延伸拼接PCR法将编码兔抗体的轻链和重链可变区序列拼接成编码scFv的核苷酸序列,轻重链可变区通过接头TCTAGTGGTGGCGGTGGTTCGGGCGGTGGTGGAGGTGGTAGTTCTAGATCTTCC(编码SSGGGGSGGGGGGSSRSS)(SEQ ID NO.2)进行连接,再通过限制性内切酶SfiI(Fermentas)酶切连接到噬菌体载体pComb3x(北京义翘神州科技股份有限公司)中,电转化X-Blue感受态(北京义翘神州科技股份有限公司)构建免疫兔的噬菌体展示scFv抗体库。

表2重链PCR引物
将HPV6 VLP蛋白包被在ELISA板上,按照噬菌体抗体淘选的流程,筛选获得抗HPV6VLP蛋白阳性抗体富集的噬菌体文库(参考文献:antibody phage display:Methods andProtocols,Philippa M.O’Brien,Humana Press)。
从富集的文库中挑取单克隆噬菌体进行表达,用ELISA方法检测与HPV6 VLP蛋白的结合,Elisa检测数据见表3,筛选获得与HPV6 VLP蛋白特异性结合的高结合力抗体克隆,送测序公司测序获得核苷酸序列,其中R003 scFv抗体的核苷酸序列为SEQ IDNO.3,对应氨基酸序列为SEQ ID NO.4。

实施例2抗体的构建、生产与纯化
采用PCR方法获得R003 scFv抗体的重链可变区的核苷酸序列(SEQ ID NO.11),对应氨基酸序列(SEQ ID NO.12),再通过ScaI和KpnI(来源:Fermentas)酶切插入带有重链信号肽核苷酸序列(SEQ ID NO.13),对应氨基酸序列(SEQ ID NO.14)和兔IgG1重链恒定区核苷酸序列(SEQ ID NO.15),对应氨基酸序列(SEQ ID NO.16)的pSTEP2载体中获得完整的重链核苷酸序列(SEQ ID NO.17)表达载体,对应完整的重链氨基酸序列(SEQ ID NO.18)。
采用PCR方法获得R003 scFv抗体的轻链可变区的核苷酸序列(SEQ ID NO.19),对应氨基酸序列(SEQ ID NO.20),再通过ScaI和BamHI酶切插入带有轻链信号肽核苷酸序列(SEQ ID NO.21),对应氨基酸序列(SEQ ID NO.22)和兔kappa轻链恒定区核苷酸序列(SEQID NO.23),对应氨基酸序列(SEQ ID NO.24)的pSTEP2载体中获得完整的轻链核苷酸序列(SEQ ID NO.25)表达载体,对应完整的轻链氨基酸序列(SEQ ID NO.26)。提质粒后转染HEK-293细胞进行培养表达7天,培养上清采用蛋白A纯化柱进行纯化获得高纯度抗体HPV6-R003。

实施例3抗体的生物鉴定
1.抗体特异性鉴定
1.1单克隆抗体HPV6-R003与其他型别的HPV VLP无交叉反应
应采用间接ELISA方法,将HPV6,HPV11,HPV16,HPV18,HPV31,HPV33,HPV35,HPV39,HPV45,HPV51,HPV52,HPV56,HPV58,HPV59完整VLP蛋白分别用pH为7.2的磷酸盐缓冲液稀释成2μg/mL,100μL/孔包被酶标板,将待检抗体稀释至10ng/mL加样,通过辣根过氧化物酶标记山羊抗兔Fc二抗显色(Jackson公司,货号111035046)。
各包被蛋白序列信息及HPV6-R003抗体特异性鉴定结果见表7,结果显示,单克隆抗体HPV6-R003具有良好的特异性,与HPV6 VLP特异性结合,与HPV16、11、18、31、33、35、39、45、51、52、56、58和59型无交叉反应。

1.2抗体配对检测
结合抗体特异性和中和活性的检测结果,选择特异性好且中和活性较强的HPV6-R003,采用双抗夹心法,建立定量检测HPV6VLP蛋白的方法。
将HPV6-R003用pH为7.2的磷酸盐缓冲液稀释至2ug/ml,100μL/孔包被酶标板;将50ng/mL的HPV6 VLP蛋白作为标准品,连续进行六次2倍梯度稀释后,分别按100ul/孔体积加入反应孔中,同时将样品稀释液(0.1% BSA,0.05% Tween20,20mM Tris,150mM NaCl,pH 7.2-7.4)作为空白对照样品加样检测;然后,再以100ul/孔体积加入1ug/mL辣根过氧化物酶标记的HPV6-R003,对HPV6 VLP蛋白进行检测;加入TMB底物显色,在酸的作用下终止反应,读取450nm的吸光度(OD值)。450nm处OD值与样品中的HPV6VLP蛋白呈正相关。通过已知含量的标准品建立标准曲线计算待检样本中HPV6VLP蛋白的含量。

以此绘制标准曲线图1。
利用上述检测方法对除HPV6以外的13种HPV型(HPV11,HPV16,HPV18,HPV31,HPV33,HPV35,HPV39,HPV45,HPV51,HPV52,HPV56,HPV58,HPV59完整VLP蛋白)混合物进行交叉反应检测,评价检测方法的特异性。
将除HPV6以外的其它13种HPV型混合物稀释到50ng/ml(混合物中各亚型HPV VLP分别占比1/13),按100ul/孔体积加样进行检测,同时将样品稀释液(0.1% BSA,0.05%Tween20,20mM Tris,150mM NaCl,pH 7.2-7.4)作为空白对照样品加样检测,各做3个复孔,分别读取450nm的吸光度(OD值)。
利用该方法检测除HPV6以外的其它13种HPV型混合物时,样品孔OD450平均值小于空白对照孔OD450平均值加3倍标准差,即该方法检测其它13种HPV型混合物时,无交叉反应信号,数据见表9。

13种HPV型混合物进行交叉反应检测表明,单克隆抗体HPV6-R003与混合物中的HPV11、16、18、31、33、35、39、45、51、52、56、58和59型无交叉反应。
免疫原HPV6 VLP蛋白序列和HPV6-R003抗体序列如表10所示。

以上详细描述了本发明的优选实施方式,但是,本发明并不限于此。在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,包括各个技术特征以任何其它的合适方式进行组合,这些简单变型和组合同样应当视为本发明所公开的内容,均属于本发明的保护范围。
Claims (15)
1.一种抗HPV6 L1蛋白抗体或其抗原结合片段,包含轻链可变区或其部分和/或重链可变区或其部分,
其中,轻链可变区或其部分包含氨基酸序列为SEQ ID NO:5的轻链CDR1、氨基酸序列为SEQ ID NO:6的轻链CDR2和氨基酸序列为SEQ ID NO:7的轻链CDR3的一个或多个;且
重链可变区或其部分包含氨基酸序列为SEQ ID NO:8的重链CDR1、氨基酸序列为SEQID NO:9的重链CDR2和氨基酸序列为SEQ ID NO:10的重链CDR3的一个或多个。
2.根据权利要求1所述的抗HPV6 L1蛋白抗体或其抗原结合片段,其中,所述抗体或其抗原结合片段包含:与抗HPV6 L1蛋白抗体轻链可变区氨基酸序列SEQ ID NO:20具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列,和/或
与抗HPV6 L1蛋白抗体重链可变区氨基酸序列SEQ ID NO:12具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列。
3.根据权利要求1或2所述的抗HPV6 L1蛋白抗体或其抗原结合片段,其中,所述抗体进一步包含轻链恒定区和重链恒定区,
优选地,轻链恒定区为与抗HPV6 L1蛋白抗体轻链恒定区氨基酸序列SEQ ID NO:24具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列,和/或
重链恒定区为与抗HPV6 L1蛋白抗体重链恒定区氨基酸序列SEQ ID NO:16具有至少90%,92%,95%,98%或100%序列同一性的氨基酸序列。
4.根据权利要求1或2所述的抗HPV6 L1蛋白抗体或其抗原结合片段,其中,所述抗体为单克隆抗体。
5.根据权利要求1或2所述的抗HPV6 L1蛋白抗体或其抗原结合片段,其中,所述抗体与HPV6 L1抗原特异性结合,与其他型别的HPV L1抗原无交叉反应。
6.根据权利要求5所述的抗HPV6 L1蛋白抗体或其抗原结合片段,其中,其他型别HPV为HPV11、16、18、31、33、35、39、45、51、52、56、58和59型。
7.根据权利要求1或2所述的抗HPV6 L1蛋白抗体或其抗原结合片段,其中,抗原结合片段的形式为Fv、Fab、Fab′、Fab′-SH、F(ab′)2、Fd片段、Fd'片段、单链抗体分子或单域抗体;其中,单链抗体分子优选为scFv、di-scFv、tri-scFv、双体抗体或scFab。
8.一种核酸,其包含:编码权利要求1-7中任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段的核苷酸序列。
9.一种载体,其包含权利要求8所述的核酸。
10.一种细胞,其包含权利要求8所述的核酸,和/或权利要求9所述的载体。
11.权利要求1-7之任一项所述的抗体或其抗原结合片段在检测HPV6 L1蛋白中的应用。
12.一种用于检测样品中HPV6 L1蛋白存在的方法,包括:
a)将权利要求1-7之任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段包被到测定表面上;
b)使所述测定表面与待测样品接触一段时间,该时间段足以容许结合,然后清洗所述测定表面;
c)使所述测定表面与同报告基团缀合的权利要求1-7之任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段接触一段时间,该时间段足以容许缀合抗体的结合,然后清洗所述测定表面;并
d)检测报告基团信号。
13.一种用于定量测定HPV6 L1蛋白的方法,包括:
a)将权利要求1-7之任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段包被到测定表面上;
b)使所述测定表面与待测样品接触一段时间,该时间段足以容许结合,然后清洗所述测定表面;
c)使所述测定表面与同报告基团缀合的权利要求1-7之任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段接触一段时间,该时间段足以容许缀合抗体的结合,然后清洗所述测定表面;并
d)通过与标准曲线比较来量化所述HPV6 L1蛋白。
14.根据权利要求13所述的方法,其中,所述标准曲线通过如下步骤建立:
a)将权利要求1-7之任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段包被到测定表面上;
b)使所述测定表面与一系列浓度的HPV6-L1蛋白标准品接触一段时间,该时间段足以容许结合,然后清洗所述测定表面;
c)使所述测定表面与同报告基团缀合的权利要求1-7之任一项所述的抗HPV6 L1蛋白抗体或其抗原结合片段接触一段时间,该时间段足以容许缀合抗体的结合,然后清洗所述测定表面;并测定其吸光度;且
d)以标准品的浓度为纵坐标,其相应的吸光度为横坐标,绘制标准曲线。
15.根据权利要求12-14之任一项所述的方法,其中,步骤(a)和(c)中的权利要求1-7之任一项所述的抗体相同或不同,优选地,所述报告基团是辣根过氧化物酶。
