痛风(Gout)是一种常见且复杂的关节炎类型,各个年龄段均可能罹患本病,男性发病率高于女性。痛风患者经常会在夜晚出现突然性的关节疼,发病急,关节部位出现疼痛、水肿、红肿和炎症,疼痛感慢慢减轻直至消失,持续几天或几周不等。痛风发作与体内尿酸浓度有关,痛风会在关节腔等处形成尿酸盐沉积,进而引发急性关节疼痛。痛风是由单钠尿酸盐沉积所致的晶体相关性关节病。痛风主要包括急性发作性关节炎、痛风石形成、痛风石性慢性关节炎、尿酸盐肾病、尿酸性尿路结石、关节残疾、肾功能衰竭等症状。痛风发病的诱因主要是食用了富含大量嘌呤的食物。
目前西医对本病的治疗主要采用非甾类抗炎药(NSAIDs)、秋水仙碱(colchicine)、糖皮质激素三类药物,但其不良反应大,患者耐受性差。
其以多组分、多靶点的特点被现代医药所使用和研究,并在治疗人类疾病中发挥重要。
综上所述,研发一种用于痛风的中药组合物及其制备方法和应用,是中药技术领域中急需解决的关键问题。
发明内容
针对现有技术所存在的上述缺点,本发明在于提供一种用于痛风的中药组合物及其制备方法和应用,本发明所制备的中药组合物,不仅能够有效缓解疼痛,而且还能够治疗痛风,具有较好的经济效益。
为实现上述目的,本发明提供了如下技术方案:
本发明第一方面:提供了一种用于痛风的中药组合物,包括以下质量份数的原料:豨莶草10-20份、赤苍藤12-14份、桂枝14-16份、川芎10-20份、侧柏叶12-14份、羌活10-12份、辣椒8-10份、蔓荆子6-8份。
本发明进一步的设置为:包括以下质量份数的原料:豨莶草15份、赤苍藤13份、桂枝15份、川芎15份、侧柏叶13份、羌活11份、辣椒9份、蔓荆子7份。
本发明的第二方面:还提供了一种用于痛风的中药组合物的制备方法,包括以下步骤:
(1)将豨莶草置于研磨机中,研磨成80-100目的粉末,再以乙醇为提取剂,按照豨莶草粉末和乙醇质量比为1:3的比例水浴回流浸提,过滤后,得到豨莶草提取物;
(2)将侧柏叶剪切成0.3-0.4cm的段块,按照料液比为1:8的比例加入乙酸乙酯浸泡2-3h,再超声处理,过滤后,获得侧柏叶提取物;
(3)称取桂枝、川芎、羌活、辣椒和蔓荆子,加水提取4-5次,每次时间为1-2h,合并提取液,过滤后,获得混合提取液;
(4)将豨莶草提取物、侧柏叶提取物和混合提取液于40-50℃的条件下混合,制得中药组合物。
本发明进一步的设置为:在步骤(1)中,所述的水浴温度为50-60℃,浸提时间为2-3h。
本发明进一步的设置为:在步骤(2)中,所述的超声处理是在80℃下超声波40Hz处理30-32min。
本发明进一步的设置为:在步骤(3)中,所述的提取的温度为120-125℃,每次提取的加水量为桂枝、川芎、羌活、辣椒和蔓荆子总重量的10-12倍。
本发明进一步的设置为:在步骤(4)中,所述的中药组合物中豨莶草提取物、侧柏叶提取物和混合提取液的混合比例为1:2:4。
本发明的第三方面:还提供了上述制备方法制备的中药组合物在治疗痛风药物中的应用。
以下是本发明中药制剂的药材来源:
豨莶草:出自《唐本草》;“豨莶,叶似酸浆而狭长,花黄白色,田野皆识之。”“猪膏莓,叶似苍耳,茎固有毛,生下湿地,所在皆有。”《蜀本草》:“《图经》云,(豨莶)高二尺许,子青黄,夏采叶用,所在下湿地有之。”“猪膏莓,叶似苍耳,两枝相对,茎叶俱有毛,黄白色,五月、六月采苗,日干之。”。用于风湿痹痛,肢体麻木;中风,手足不遂;痈肿疮毒,湿疹瘙痒;高血压。
赤苍藤(学名:Erythropalum scandens Bl.)为铁青树科(Olacaceae)赤苍藤属(Erythropalum)多年生常绿大型木质藤本植物。赤苍藤具有较高的食用、药用及园林绿化观赏价值。
肉桂(拉丁学名:Cinnamomum cassia Presl)是樟科,樟属中等大乔木,树皮灰褐色。在《神农本草经》中记载有:“味辛温,主百病,养精神,和颜色,利关节,补中益气。为诸药先聘通使,久服通神,轻身不老。面生光华,眉好常如童子。
川芎(学名:Ligusticum chuanxiong hort),栽培植物,是一种中药植物,常用于活血行气,祛风止痛,川芎辛温香燥,走而不守,既能行散,上行可达巅顶;又入血分,下行可达血海。活血祛瘀作用广泛,适宜瘀血阻滞各种病症;可治头风头痛、风湿痹痛等症。昔人谓川芎为血中之气药,殆言其寓辛散、解郁、通达、止痛等功能。
侧柏叶,中药名。为柏科植物侧柏PLatycladus orientalis(L.)Franco的干燥枝梢和叶。在《本草从新》中记载有:最清血分湿热。止吐衄崩淋。肠风尿血。血痢。一切血证。去风湿诸痹。历节风痛。涂汤火伤。生肌杀虫。炙罨冻疮。
羌活,中药名。别名羌青、护羌使者、胡王使者、羌滑、退风使者、黑药,伞形科、羌活属植物,多年生草本,高60-120厘米,根茎粗壮,伸长呈竹节状。根颈部有枯萎叶鞘。茎直立,圆柱形,中空,有纵直细条纹,带紫色。其性温,解表散寒、祛寒湿,用于外感风寒、头痛无汗、寒湿痹、上肢风湿疼痛。
辣椒(学名:Capsicum annuum L.)是双子叶植物纲、茄科、辣椒属一年或有限多年生草本植物。在《药性考》中记载有:温中散寒,除风发汗,去冷癖,行痰逐湿。
蔓荆子,中药名。别名:蔓荆实、荆子、万荆子、蔓青子、蔓荆、白背木耳、小刀豆藤、白背风、白背草。来源为马鞭草科植物单叶蔓荆或蔓荆的干燥成熟果实。味辛、苦,性微寒。归胃、膀胱、肝经。其主要功效是疏散风热,清利头目,除湿处关节。
有益效果
采用本发明提供的技术方案,与已知的公有技术相比,具有如下有益效果:
本发明以豨莶草、赤苍藤、桂枝、川芎、侧柏叶、羌活、辣椒以及蔓荆子为原料,从中提取活性成分,用于治疗痛风症状,所制备的中药组合物,不仅能够有效缓解疼痛,而且还能够治疗痛风,具有较好的经济效益,具有广泛的应用前景,值得推广。

为使本发明实施例的目的、技术方案和优点更加清楚,下面将对本发明实施例中的技术方案进行清楚、完整地描述。显然,所描述的实施例是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
下面结合实施例对本发明作进一步的描述。
实施例1:
本发明提供了一种用于痛风的中药组合物,包括以下质量份数的原料:豨莶草10份、赤苍藤12份、桂枝14份、川芎10份、侧柏叶12份、羌活10份、辣椒8份、蔓荆子6份。
此外,本发明还提供了一种用于痛风的中药组合物的制备方法,包括以下步骤:
(1)将豨莶草置于研磨机中,研磨成80目的粉末,再以乙醇为提取剂,按照豨莶草粉末和乙醇质量比为1:3的比例水浴回流浸提,过滤后,得到豨莶草提取物。
进一步的,水浴温度为50℃,浸提时间为2h。
(2)将侧柏叶剪切成0.3cm的段块,按照料液比为1:8的比例加入乙酸乙酯浸泡2h,再超声处理,过滤后,获得侧柏叶提取物。
进一步的,超声处理是在80℃下超声波40Hz处理30min。
(3)称取桂枝、川芎、羌活、辣椒和蔓荆子,加水提取4次,每次时间为1h,合并提取液,过滤后,获得混合提取液。
进一步的,提取的温度为120℃,每次提取的加水量为桂枝、川芎、羌活、辣椒和蔓荆子总重量的10倍。
(4)将豨莶草提取物、侧柏叶提取物和混合提取液于40℃的条件下混合,制得中药组合物。
进一步的,中药组合物中豨莶草提取物、侧柏叶提取物和混合提取液的混合比例为1:2:4。
实施例2:
本发明提供了一种用于痛风的中药组合物,包括以下质量份数的原料:豨莶草15份、赤苍藤13份、桂枝15份、川芎15份、侧柏叶13份、羌活11份、辣椒9份、蔓荆子7份。
此外,本发明还提供了一种用于痛风的中药组合物的制备方法,包括以下步骤:
(1)将豨莶草置于研磨机中,研磨成90目的粉末,再以乙醇为提取剂,按照豨莶草粉末和乙醇质量比为1:3的比例水浴回流浸提,过滤后,得到豨莶草提取物。
进一步的,水浴温度为55℃,浸提时间为3h。
(2)将侧柏叶剪切成0.4cm的段块,按照料液比为1:8的比例加入乙酸乙酯浸泡3h,再超声处理,过滤后,获得侧柏叶提取物。
进一步的,超声处理是在80℃下超声波40Hz处理31min。
(3)称取桂枝、川芎、羌活、辣椒和蔓荆子,加水提取5次,每次时间为2h,合并提取液,过滤后,获得混合提取液。
进一步的,提取的温度为122℃,每次提取的加水量为桂枝、川芎、羌活、辣椒和蔓荆子总重量的11倍。
(4)将豨莶草提取物、侧柏叶提取物和混合提取液于45℃的条件下混合,制得中药组合物。
进一步的,中药组合物中豨莶草提取物、侧柏叶提取物和混合提取液的混合比例为1:2:4。
实施例3:
本发明提供了一种用于痛风的中药组合物,包括以下质量份数的原料:豨莶草20份、赤苍藤14份、桂枝16份、川芎20份、侧柏叶14份、羌活12份、辣椒10份、蔓荆子8份。
此外,本发明还提供了一种用于痛风的中药组合物的制备方法,包括以下步骤:
(1)将豨莶草置于研磨机中,研磨成100目的粉末,再以乙醇为提取剂,按照豨莶草粉末和乙醇质量比为1:3的比例水浴回流浸提,过滤后,得到豨莶草提取物。
进一步的,水浴温度为60℃,浸提时间为3h。
(2)将侧柏叶剪切成0.4cm的段块,按照料液比为1:8的比例加入乙酸乙酯浸泡3h,再超声处理,过滤后,获得侧柏叶提取物。
进一步的,超声处理是在80℃下超声波40Hz处理32min。
(3)称取桂枝、川芎、羌活、辣椒和蔓荆子,加水提取5次,每次时间为2h,合并提取液,过滤后,获得混合提取液。
进一步的,提取的温度为125℃,每次提取的加水量为桂枝、川芎、羌活、辣椒和蔓荆子总重量的12倍。
(4)将豨莶草提取物、侧柏叶提取物和混合提取液于50℃的条件下混合,制得中药组合物。
进一步的,中药组合物中豨莶草提取物、侧柏叶提取物和混合提取液的混合比例为1:2:4。
性能检测:
1)分别取大鼠100只,体重在200±10g,雌雄各半,均分为10组,分别设生理盐水组(对照组)、实施例1组(分为高、中、低剂量)、实施例2组(分为高、中、低剂量)和实施例3组(分为高、中、低剂量)。各组分别在大鼠腹部外搽给药3次,间隔20min,末次给药后30min,每组腹腔注射0.7%冰醋酸10ml/kg,记录20min内各大鼠的扭体次数,实施例1组、实施例2组和实施例3组中药组合物的高、中、低剂量累积分别给药量0.5ml/只、1.0ml/只和1.5ml/只,生理盐水组(对照组)的累积给生理盐水量0.5ml/只,记录各组实验的数据于表1。

由表1可见,与对照组相比,实施例1组、实施例2组和实施例3组的大鼠扭体次数均明显低于对照组(p<0.05),随着给药剂量的增加,实施例1组、实施例2组和实施例3组的大鼠扭体次数会明显减少(p<0.05),同时,实施例1组、实施例2组和实施例3组的大鼠扭体次数之间的差异并不明显(p>0.05)。本发明所制备中药组合物,能够起到缓解疼痛的效果。
2)临床使用实施的药物治疗痛风患者例痛风患者90例,年龄32-66岁,平均(46.72±2.76)岁;病程3月-10年,平均(6.22±1.51)年;所治患者临床症状、体征、实验室检查及影像学检查均符合痛风的诊断。将患者随机分为实验1组、实验2组和实验3组。其中,实验1组的患者使用实施例1的中药组合物,实验2组的患者使用实施例2的中药组合物,实验3组的患者使用实施例3的中药组合物。3组患者在性别、年龄、发病时间等一般资料比较无明显差异,具有可比性,使用方法为:取3-4g中药组合物,均匀涂抹于关节患处,单次以病患关节的皮肤面积为单位,每日2次,连续使用一周,疗程判定标准:显效:指治疗后,患者关节部位疼痛、红肿的症状消失;有效:指治疗后,患者关节部位的疼痛感与红肿症状减轻;无效:指治疗后,患者的临床症状未改善或在加重。并统计总有效率(显效与有效占总人数的占比)记录各组实验的数据于表2。

由图1和表2可见,实验组(实验1组、实验2组和实验3组)的所提供的中药组合物能够有效的治疗痛风。

Claims (8)
1.一种用于痛风的中药组合物,其特征在于,包括以下质量份数的原料:豨莶草10-20份、赤苍藤12-14份、桂枝14-16份、川芎10-20份、侧柏叶12-14份、羌活10-12份、辣椒8-10份、蔓荆子6-8份。
2.根据权利要求1所述的一种用于痛风的中药组合物,其特征在于,包括以下质量份数的原料:豨莶草15份、赤苍藤13份、桂枝15份、川芎15份、侧柏叶13份、羌活11份、辣椒9份、蔓荆子7份。
3.一种用于痛风的中药组合物的制备方法,其特征在于,包括以下步骤:
(1)将豨莶草置于研磨机中,研磨成80-100目的粉末,再以乙醇为提取剂,按照豨莶草粉末和乙醇质量比为1:3的比例水浴回流浸提,过滤后,得到豨莶草提取物;
(2)将侧柏叶剪切成0.3-0.4cm的段块,按照料液比为1:8的比例加入乙酸乙酯浸泡2-3h,再超声处理,过滤后,获得侧柏叶提取物;

4.根据权利要求3所述的一种用于痛风的中药组合物的制备方法,其特征在于,在步骤(1)中,所述的水浴温度为50-60℃,浸提时间为2-3h。
5.根据权利要求3所述的一种用于痛风的中药组合物的制备方法,其特征在于,在步骤(2)中,所述的超声处理是在80℃下超声波40Hz处理30-32min。
6.根据权利要求3所述的一种用于痛风的中药组合物的制备方法,其特征在于,在步骤(3)中,所述的提取的温度为120-125℃,每次提取的加水量为桂枝、川芎、羌活、辣椒和蔓荆子总重量的10-12倍。
7.根据权利要求3所述的一种用于痛风的中药组合物的制备方法,其特征在于,在步骤(4)中,所述的中药组合物中豨莶草提取物、侧柏叶提取物和混合提取液的混合比例为1:2:4。
8.如权利3-7中任一项制备方法制备的中药组合物在治疗痛风药物中的应用。
