A microbial preparation for preventing and controlling root-knot nematodes and grubs comprises a core layer, an intermediate layer and a coating layer; the core layer consists of the following raw materials in parts by weight: 11 parts of paecilomyces lilacinus spore powder; 11 parts of beauveria bassiana spore powder; 22 parts of sunflower seed oil; 2 parts of bacterial cellulose; 0.6 parts of silk fibroin; 1.1 parts of xanthan gum; 2 parts of fatty acid sucrose ester; the middle layer is prepared from peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder in a weight ratio of 1:1:1:1, the composition is as follows; the coating layer consists of sodium alginate and gluten protein in a weight ratio of 1:0.02;

Comparative example 3 was substantially the same as the microbial preparation obtained in example 3, except that the core layer of the microbial preparation in comparative example 3 did not contain xanthan gum; the adhesion of the core layer prepared in the comparative example 3 to the mixed powder of the middle peach leaf powder, the mulberry leaf powder, the pharbitis seed powder and the lotus leaf powder is poor, the middle layer is easy to fall off and is not favorable for cladding by the cladding layer, and the microbial preparation with complete surface is not easy to prepare and obtain;
performance detection
Storage stability of microbial formulations
The microbial preparation prepared in the embodiment 3 and the microbial preparation prepared in the comparative examples 1-2 are stored for 12 months at 25 ℃, and the germination rate of spores of the microorganisms is detected by periodically taking the microbial preparation, wherein the germination rate of spores refers to the sum of the germination rates of spores of paecilomyces lilacinus and beauveria bassiana; the test results are shown in Table 1.

As can be seen from the data in Table 1, the germination rate of spores can still reach more than 90% after the microbial preparation prepared in the embodiment 3 of the invention is stored for 12 months; exhibit good storage stability; in contrast, the spore germination rate was reduced in both comparative example 1 and comparative example 2, as compared to example 3.
Detection of insecticidal performance of microbial preparation
100 nematodes (second-instar larvae) are uniformly dispersed into 10kg of soil (the soil is subjected to high-temperature insecticidal treatment and does not contain root-knot nematodes), the microbial preparation prepared in the embodiment 3 and the comparative examples 1-2 is applied to the soil, water is applied to the soil, and the water content of the soil is kept at 18-25%; after 30 days of application of the microbial formulation; mortality of nematodes was recorded and the results are reported in table 2.
Uniformly dispersing 100 grubs into 10kg of soil (the soil is subjected to high-temperature insecticidal treatment and does not contain grubs), applying the microbial preparation prepared in the embodiment 3 and the comparative examples 1-2 into the soil, and applying water to the soil to keep the water content of the soil to be 18-25%; after 30 days of application of the microbial formulation; mortality of nematodes and grubs was recorded and the results are reported in table 2.

As can be seen from table 2, the insecticidal effects of comparative example 1 and comparative example 2 on nematodes and grubs are inferior to those of example 3, and the difference between comparative example 1 and example 3 is obvious, which indicates that the intermediate layer peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder have certain insecticidal effects; in contrast, comparative example 2 was lower than example 3, and the analysis was probably due to the fact that the microbial preparation prepared in comparative example 2 was inferior in water absorption and low in spore germination rate in a shorter insecticidal time (30 days), so that the insecticidal effect was lowered.
Field test
The prevention and treatment effects of the microbial preparations prepared in the embodiment 3 and the comparative example 1 on peanut root knot nematode and grub diseases and the influence on the growth condition of the peanuts are explored;
selecting a land block for planting peanuts for years, which is easy to generate root-knot nematodes and grubs, of the peanuts planted for a long time as a test land block;
test crop: peanut; variety: flower cultivation 16; sowing: sowing in year 2020 and 2, harvesting: 8 months and 26 days 2020;
the cultivation method comprises the following steps: mulching;
test site: geng Dantun village (yellow river farm) in muu county in Henan province;
selecting land block soil types: wet soil; texture: sand soil;
basic nutrients: organic matter: 9.2g/kg, total nitrogen: 0.67g/kg, quick-acting phosphorus: 8.2mg/kg, quick-acting potassium: 62.6mg/kg;
irrigation and drainage conditions: the land form is flat, and the irrigation and drainage conditions are good;
fertilizing in a test field: 45kg of special fertilizer (20-10-10) is applied to the bottom of each mu, and additional fertilizer is not applied in the growing period.
Test treatment: the test plots are divided into a control group D1 and test groups S1 and S2; when land preparation is carried out on plots before peanut sowing in 5 months, only 45kg of special peanut fertilizer is applied to plots in a control group as base fertilizer, and the microbial preparations prepared in the embodiment 3 and the comparative example 1 of the invention (5 kg are applied to each mu of bottom) are sequentially applied to plots S1 and S2 in test groups except the base fertilizer; 3 groups were repeated for each treatment; planting peanuts in the land parcels of the control group and the test group, carrying out the same field management in the control group and the test group, sampling after the peanuts are sowed for 40 days, respectively counting and recording the density of the mouths of the nematodes and the grubs, calculating an average value (table 3), sampling again after the peanuts are sowed for 80 days, and counting and recording the density of the mouths of the nematodes and the grubs; and the average value (table 3) was calculated, and the growth of peanuts in each treated plot (shown in fig. 1) was recorded, and the results of the peanuts and the yield were statistically recorded at the time of harvesting the peanuts, and the results are shown in fig. 2 and table 3.

As can be seen from the data in table 3, the microbial preparation prepared in example 3 of the present invention shows excellent effects on prevention and control of peanut root-knot nematodes and grubs, and compared with the control group, peanut yield is also significantly improved.
Compared with example 3, the control effect of the comparative example 1 on the peanut root-knot nematodes and the grubs is reduced, and compared with example 3, the comparative example 1 does not contain an intermediate layer consisting of peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder, and the analysis reasons are probably that the intermediate layer peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder in the microbial agent have certain insect repellent effect and have synergistic insect resistance with the microbial agent; however, as the peanut growth time is prolonged, the control effect of comparative example 1 tends to decrease; the plant diseases and insect pests of the peanut root-knot nematode and the grubs rebound; the analysis reasons may be that the microbial agent provides ideal constant environment for the microbial agent at the initial stage of application, part of spores lose activity along with the extension of time, the germination rate of the spores is reduced, and the pest control effect of the microbial agent is reduced.
The above embodiments are merely preferred embodiments of the present invention, the protection scope of the present invention is not limited thereto, and any simple changes or equivalent alternatives of the technical solutions that can be obviously obtained by those skilled in the art within the technical scope of the present invention disclosed herein fall within the protection scope of the present invention.
Claims (10)
1. The microbial preparation for preventing and controlling the root-knot nematodes and the grubs is characterized by comprising a core layer, an intermediate layer and a coating layer; the core layer comprises paecilomyces lilacinus spore powder and beauveria bassiana spore powder; vegetable oil; bacterial cellulose; silk fibroin; xanthan gum; sucrose esters of fatty acids; the middle layer consists of peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder; the coating layer consists of sodium alginate and gluten protein.
2. The microbial preparation for preventing and controlling root-knot nematodes and grubs as claimed in claim 1, wherein the weight ratio of the raw materials in the core layer is: 10-14 parts of paecilomyces lilacinus spore powder and 10-14 parts of beauveria bassiana spore powder; 20-28 parts of vegetable oil; 1-5 parts of bacterial cellulose; 0.5-1 part of silk fibroin, 1-1.4 parts of xanthan gum and 1-1.5 parts of sucrose fatty acid ester.
3. The microbial preparation for preventing and controlling root-knot nematodes and grubs as claimed in claim 1, wherein the weight ratio of peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder in the middle layer is 1:1:1:1.
4. the microbial preparation for preventing and controlling root-knot nematodes and grubs according to claim 1, wherein the particle size of the peach leaf powder, the mulberry leaf powder, the pharbitis seed powder and the lotus leaf powder is 100-300 meshes.
5. The microbial preparation for preventing and controlling root-knot nematodes and grubs according to claim 1, wherein the weight ratio of sodium alginate to gluten protein in the coating is 1:0.01-0.05.
6. A method for preparing the microbial preparation for preventing and controlling root-knot nematodes and grubs according to claim 1, which is characterized by comprising the following steps:
(1) Preparing a core layer solution: dispersing paecilomyces lilacinus spore powder and beauveria bassiana spore powder in vegetable oil; adding sucrose fatty acid ester, stirring uniformly, sequentially adding bacterial cellulose, silk fibroin and xanthan gum, stirring uniformly, and obtaining emulsion; injecting the emulsion into a pill dropping machine, and dripping into condensed liquid to obtain a core layer;
(2) Firstly removing condensed liquid on the surface of a core layer, then placing the core layer in mixed powder of peach leaf powder, mulberry leaf powder, pharbitis seed powder and lotus leaf powder, rolling, spraying sodium alginate/gluten protein mixed solution on the surface, placing in a cross-linking agent solution, and cross-linking, solidifying and drying to obtain the microbial preparation.
7. The method for preparing a microbial preparation for preventing and controlling root-knot nematodes and grubs as claimed in claim 6, wherein said condensed liquid is methyl silicone oil.
8. The method for preparing a microbial preparation for controlling both root-knot nematodes and grubs according to claim 6, wherein the cross-linking agent solution is sodium chloride solution.
9. The method for preparing a microbial preparation for controlling both root-knot nematodes and grubs according to claim 6, wherein the mass concentration of sodium alginate in the sodium alginate/gluten protein mixed solution is 2.5wt%.
10. The microbial preparation for preventing and controlling root-knot nematodes and grubs according to any one of claims 1 to 5, for preventing and controlling root-knot nematodes and/or grubs.
