本发明中公开了一株植物解铁、促生浅绛红链霉菌HC7‑22,所述浅绛红链霉菌HC7‑22分类命名为Streptomycespurpurascens,于2022年6月10日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCCNo.25053。本发明同时提供上述浅绛红链霉菌HC7‑22在促进植物生长、减少植物缺铁症状中的应用。本发明分离鉴定得到一株浅绛红链霉菌HC7‑22,其产嗜铁素为42.4%,为高产嗜铁素菌株,对植物具有明显的解铁、促生作用;而且耐酸碱和盐碱能力强,具有很好的抗逆性,可广泛应用于在植物实际种植中,有效减少植物缺铁症状。

技术领域
本发明涉及微生物技术领域,尤其涉及一株植物解铁、促生浅绛红链霉菌HC7-22及其应用。
背景技术
铁元素是植物正常生长发育的必须元素,不仅参与了色素的合成,可充当反应中的催化剂,在植物进行代谢时起到了一定的作用。虽然地壳中的铁丰度很高,但大部分铁元素以不能被植物直接利用的形式存在,尤其在盐碱化土壤中,pH升高降低了三价铁的溶解性,植物出现缺铁现象更加严重。在全球范围,植物缺铁症状约占十分之四,成为了当今世界最为严重的缺素症之一。
嗜铁素产生菌是一类可以分泌产生嗜铁素类物质的微生物,可特异性鳌合三价铁离子,增加铁离子的溶解度,从而为植物提供有效铁。目前已报道的嗜铁素产生菌包括假单胞属(Pseudomonas)、芽孢杆菌属(Bacillus)、固氮菌属(Azotobacter)、链霉菌属(Streptomyces)、克雷伯氏菌属(Klebsiella),肠杆菌属(Enterobacter)、产碱杆菌属(Alcaligenes)、节细菌属(Arthrobacter)、黄杆菌属(Flavobacterium)、伯克霍尔德菌(Burkholderia)、慢生根瘤菌(Bradyrhizobium)、红球菌属(Rhodococcus)和沙雷氏菌属(Serratia)等,但是现有的这些菌株种类有限,在应用过程中存在抗逆性弱,活性不稳定等缺点。
发明内容
本发明的目的在于解决现有技术中存在的嗜铁素产生菌抗逆性弱,活性不稳定的问题,提供一株植物解铁、促生浅绛红链霉菌HC7-22及其应用。
为解决上述技术问题,本发明采用的技术方案如下:一株植物解铁、促生浅绛红链霉菌HC7-22,所述浅绛红链霉菌HC7-22于2022年6月10日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.25053。
优选地,所述浅绛红链霉菌HC7-22的16S rRNA基因序列如SEQ ID No.1所示。
本发明同时提供上述浅绛红链霉菌HC7-22在促进植物生长、减少植物缺铁症状中的应用。
本发明所具有的有益效果:本发明分离鉴定得到一株浅绛红链霉菌HC7-22,其产嗜铁素为42.4%,为高产嗜铁素菌株,对植物具有明显的解铁、促生作用;而且耐酸碱和盐碱能力强,具有很好的抗逆性,可广泛应用于在植物实际种植中,有效减少植物缺铁症状。

本发明中的浅绛红链霉菌HC7-22于2022年6月10日保藏于中国微生物菌种保藏管理委员会普通微生物中心;简称CGMCC;保藏单位地址为:北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所;保藏编号为CGMCC No.25053,保藏分类命名为浅绛红链霉菌(Streptomyces purpurascens)。
具体实施方式
下面结合附图和具体实施例对本发明作进一步的说明。
实施例1浅绛红链霉菌HC7-22的分离鉴定
1.土壤样品采集及菌株分离和筛选
从陕西省韩城市矿场(N°35.48,E°110.44)采集土壤样品。利用逐级稀释法分离土壤中的细菌,将形态差异明显的单个菌落在改良高氏一号培养基上进行纯化和培养。将分离得到的菌株接种于CAS固体培养基上,在28℃培养3-5天。结果如附图1所示,菌株周围出现黄色透明晕圈,说明具有产嗜铁素能力。量取菌落直径(d)和透明圈直径(D),计算D/d,初步筛选出产铁载体活性的菌株,编号为HC7-22,该菌株在周围形成明显的黄色透明圈,D/d为3.3。
2.菌株HC7-22形态特征鉴定
将菌株HC7-22在改良高氏一号培养基上划线培养,28℃培养24h,观察菌落形状、透明度及菌落颜色等菌落特征。结果如附图2所示,菌株HC7-22在高氏1号培养基上气丝较少,粉红色,基丝绛红色,扫描电镜观察菌丝直径约4um,表面不光滑。
3.菌株HC7-22生理生化鉴定
对菌株HC7-22的淀粉水解、硝酸盐还原、v-p反应、H2S产生以及明胶液化的生理生化指标进行测定。结果如附图3所示,其中,A为淀粉水解,B为硝酸盐还原,C为V-P反应,D为硫化氢产生,E为明胶液化,B、C、D、E的左侧均为CK,右侧为HC7-22。结果显示,菌株HC7-22的水解淀粉,硝酸盐还原、v-p反应和硫化氢产生检测中均表现为阳性,而在明胶液化测定为阴性。
4.菌株HC7-22分子鉴定
使用Ezup柱式细菌基因组DNA抽提试剂盒对菌株HC7-22的DNA进行提取。使用通用引物7F和1492R对菌株的16S rRNA序列进行PCR扩增,通用引物7F和1492R的序列如SEQ IDNo.2和SEQ ID No.3所示。对目的条带回收后送测序。使用Vector NTI Advance 11软件将测序结果进行校正和拼接,用BLASTn对拼接的序列进行同源序列检索,确定其分类地位。菌株HC7-22的16Sr RNA基因序列如SEQ ID No.1所示。同源序列检索结果显示,菌株SHC7-22与浅绛红链霉菌(Streptonmyces purpurascens)的同源性达到99.64%,如附图4所示16SrRNA序列构建的进化树与S.purpurascens LZ16-14聚到一支,置信率为96%。形态学、生理生化特性结合分子序列确定为浅绛红链霉菌(S.purpurascens)。
实施例2浅绛红链霉菌HC7-22产嗜铁素能力测定
利用CAS检测液对实施例1分离得到的菌株HC7-22的产嗜铁素能力进行定量测定,分光光度计测量处理后在OD630nm处的吸光值(As),另取空白培养基测量OD630nm处的值(Ar),依据嗜铁素含量=(Ar-As)/Ar,计算出菌株嗜铁素相对含量。结果显示HC7-22的产嗜铁素为42.4%,为高产嗜铁素菌株。
实施例3浅绛红链霉菌HC7-22对马铃薯的促生效果
1.将实施例1分离得到的浅绛红链霉菌HC7-22接种于小米培养基中,置于28℃,180r/min摇床上振荡48h。无菌水调节菌株悬浮液的OD600=1.0用于接种。
2.盆栽效果
选取大小一致的马铃薯种薯播种在装有已灭菌的基质和沙子1:1比例混合的花盆中,按照约1mg/L土的量施入FeCl3马铃薯破土发芽后,以每盆100mL的量灌溉菌液,空白组灌等量的小米培养基(CK),以接种市售促生菌肥(黑金泰,哈尔滨瑞泰绿能农业发展有限公司)的处理为阳性对照(CK+);第一次接菌20d后进行第二次接菌。最后马铃薯植株生长40d后,收获植株。冲洗干净自然晾干,称量鲜重,测量植株株高、茎粗。再将植株装入纸袋中置于105℃的烘箱中杀青一个小时,在60℃下烘干直至重量不在变化,称量植株干重。盆栽马铃薯的植株生长情况如附图5所示。盆栽马铃薯植株生长量结果如表1所示。
3.大田促生结果
在内蒙古农业大学农场露地条件下测定实施例1分离得到的浅绛红链霉菌HC7-22对马铃薯最终收获量的影响。将实例1分离得到的浅绛红链霉菌HC7-22菌液调整到OD600=1.0,与田间土壤混合后种植马铃薯种薯,每个处理种植6棵马铃薯,以接种小米培养基的处理作为空白对照(CK),以接种以接种市售促生菌肥(黑金泰,哈尔滨瑞泰绿能农业发展有限公司)的处理为阳性对照(CK+),每个处理施入一定量的FeCl3作为铁源,在种植30d后再接种一次,种植60d后收取马铃薯,测定每株的马铃薯重量,比较不同处理马铃薯的收获量。大田马铃薯重量结果如表1所示。

附图5和表1结果显示,用HC7-22处理的马铃薯植株生长量(盆栽)和收获马铃薯重量(大田)的生长指标均显著高于对照组,说明对马铃薯植物具有显著的促生效果。
4.植株养分测定与土壤理化指标测定
将步骤2中收获的的马铃薯叶片采用H2SO4-H2O2消煮法消煮,消煮液用于植物叶片全铁含量的测定。用电感耦合等离子体发射光谱法测定植株叶片的全铁含量。结果显示:空白对照中叶片铁含量为0.94g/kg,CK+的铁含量为0.97g/kg,相比对照提高了3.6%,经HC7-22处理后的马铃薯植株铁含量为1.05,相比对照增加了11.96%。
实施例4不同pH与含盐量对浅绛红链霉菌HC7-22培养特性的影响
pH:将实施例1分离得到的HC7-22菌株接种在用1mol/LNaOH和HCl分别调整为pH为1、2、3、4、5、6、7、8、9、10、11、12的小米发酵液中,接入种子液,在180r/min、28℃培养48h,测定在OD600时的吸光值。
NaCl浓度:将实施例1分离得到的HC7-22菌株接种在盐浓度为0、0.1、0.5、1.0、2.0、5.0、10.0、12.0、15.0%的发酵液中,在180r/min、28℃培养48h,测定在OD600nm时的吸光值。
如附图6所示:HC7-22菌株在酸性条件下生长缓慢,而在弱碱坏境中菌体浓度开始缓慢平稳生长,在pH12的培养基中仍能正常生长。HC7-22菌株在15%的NaCl浓度中仍能生长,耐酸碱和盐碱能力强,具有很好的抗逆性。

1.一株浅绛红链霉菌(Streptomyces purpurascens)HC7-22,其特征在于,所述浅绛红链霉菌(Streptomyces purpurascens)HC7-22于2022年6月10日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No. 25053。
2.如权利要求1所述的浅绛红链霉菌HC7-22在促进植物生长和减少植物缺铁症状中的应用,其特征在于,所述植物为马铃薯。
