Abstract
EN2蛋白的免疫原性片段肽或特异性识别其的抗体组合物。本发明涉及EN2蛋白的免疫原性片段肽或特异性识别其的抗体组合物。借助本发明,可以通过特异性识别该肽的方法对EN2蛋白进行量化。此外,与现有的EN2蛋白抗体相比,使用该肽制备的抗体具有显著更好的检测灵敏度,因此可以用作前列腺癌的诊断剂组合物。

本申请是申请号为201780074265.8、发明名称为“EN2蛋白的免疫原性片段肽或特异性识别其的抗体组合物”的中国专利申请的分案申请,该母案申请是2017年11月28日提交的PCT国际专利申请PCT/KR2017/013631进入中国国家阶段的申请。
技术领域
本发明涉及EN2蛋白的免疫原性片段肽或特异性识别其的抗体组合物。
背景技术
前列腺是位于膀胱下方和直肠前方的核桃大小的男性生殖器官,并且用于产生精液并储存其的一部分。前列腺的上部与从膀胱连接到尿道的膀胱颈相邻并固定到前耻骨前列腺韧带,其下部由泌尿生殖膈膜固定。大多数发生在前列腺中的癌症是在前列腺细胞中形成的腺癌(腺细胞癌)。可以根据肿瘤组织的分化程度和细胞的特征对癌症的类型进行分类。
前列腺癌是世界上最常见的泌尿生殖系统肿瘤之一。在美国,2016年约180,890人被新诊断为前列腺癌,占美国所有新肿瘤诊断的10.7%,前列腺癌是继乳腺癌和肺癌之后的是第三种最常发生的肿瘤类型。基于2009年至2013年的全球统计数据,每100,000名男性中有129.4名男性患有前列腺癌,而根据2016年的统计数据,26,120名前列腺癌患者死亡。此外,前列腺癌在50岁之前是一种罕见的疾病,但是在50岁之后迅速增加。最近,由于寿命期望的延长,韩国老年男性的数量急剧增加,因此需要持续管理以通过早期诊断预防前列腺癌的恶化(例如转移等)。
特别是,人前列腺癌似乎具有转移到骨的趋势,并且已知不可避免地从雄激素依赖状态发展到雄激素抗性状态,从而提高患者死亡率。此外,约25%进行过前列腺癌治疗的男性因疾病复发需要另外治疗,而前列腺癌目前是美国男性癌症相关死亡的第二大原因。因此,前列腺癌的早期诊断和治疗是必要的。
目前使用的直接前列腺癌诊断方法的实例包括对前列腺直接成像的方法或活组织检查诊断方法。在通过直接成像或活组织检查进行诊断的情况下,难以在初始阶段诊断前列腺癌的发病,因此迫切需要开发用于体外诊断的方法。
作为间接方法,存在可使用前列腺特异性抗原(prostate-specific antigen,PSA)测定在体外进行的诊断方法。然而,用于诊断的PSA不仅在恶性前列腺上皮中产生,而且在正常和良性组织中也产生,从而导致前列腺癌检测中的高假阳性率。此外,血清PSA水平的显著提高可用于诊断前列腺癌的有效标准方法,而PSA血清水平中约2至10ng/mL的微弱提高使其无法达到明确的前列腺癌诊断。在这样的微弱提高的情况下,血清PSA可源自非肿瘤性疾病,例如良性前列腺增生(benign prostatic hyperplasia,BPH)、前列腺炎或其他物理创伤,并且用于前列腺癌诊断的PSA分析具有与检测特异性相关的问题。
因此,使用新生物标志物诊断前列腺癌被认为是重要的,并且已经进行了对其的研究(韩国专利申请公开No.10-2009-0111307),但仍然不够。最近,已经提出了使用EN2(锯齿状蛋白-2(Engrailed-2))作为生物标志物来诊断前列腺癌的方法,并且公知EN2作为能够解决在常规前列腺癌诊断中使用的PSA(前列腺特异性抗原)测定的检测特异性问题的生物标志物。EN2蛋白在细胞中作为转录因子发挥作用并且仅在前列腺癌细胞中过表达,不期望地引起DNA转录调节病症。此外,当EN2在前列腺癌细胞中的表达提高时,尿液排泄的EN2蛋白的量也会提高,因此EN2适合在体外分析中使用。因此,美国专利No.8460882、日本专利申请公开No.2012-532621和美国专利No.8722643公开了识别EN2蛋白的诊断组合物,韩国专利申请公开No.10-2016-0077788公开了与EN2特异性结合的DNA适配体。
通常,术语“抗体”意指将外部物质(具有至少预定尺寸和条件的物质)(抗原)注射到生物体中,并且在该生物体中形成具有能够通过体液免疫应答特异性识别所述外部物质之位点(表位)的抗体,然后从血液中收集并与血清相分离。由此形成的抗体是具有多个能够识别抗原的位点的多克隆抗体。
在体液免疫应答中,抗原经由多种抗原呈递细胞(antigen-presenting cell,APC)通过表位呈递给B细胞并活化B细胞。在活化过程期间,B细胞分化为浆细胞,其能够通过以下大量产生仅单一抗体并离体分泌:在产生抗体的全部基因中只重新排列产生能够与抗原反应的抗体的基因并除去剩余的不必要的基因。
由于抗原通常具有多个表位,因此存在多个种类的分化的浆细胞,从而产生多种抗体。从多种浆细胞(即产生抗体的基因是不同的)分泌的整组多样化的抗体被称为“多克隆抗体”,只从一种浆细胞制成的单一抗体被称为“单克隆抗体”。
自1980年以来,使用抗体的诊断产品的市场迅速扩大,并且抗体高度灵敏,足以检测根据疾病或症状而特异性表达的非常少量的蛋白质,从而被用于开发高效诊断试剂盒和诊断方法。为此,必须开发对根据疾病或症状而表达的蛋白质(抗原)具有特异性和灵敏度的抗体。为了产生具有特异性和灵敏度的抗体,可通过考虑数个因素来使抗体的效率最大化。
首先,比较待产生的抗原的一级序列,从而选择与抗原的动物物种具有高度异质性的待免疫动物,由此使免疫应答最大化。例如,当使用人蛋白质的序列时,可选择与上述序列具有高度异质性的动物用于诱导免疫应答,从而获得高效价抗体。
其次,考虑到抗原质量可根据抗原序列翻译之后的修饰特征以及立体结构而变化的事实进行选择。特别是,必须在考虑到不使用全部蛋白质而是使用其一些肽形式作为抗原的情况下进行选择。
第三,制备抗体以适应其目的。多克隆抗体是有利的,因为它易于产生,由于其的多种表位而有利于抗原的检测,并且能够根据抗原从广泛的范围中选择待免疫动物,但其抗体特异性低并且不适合大量产生一致效价。此外,单克隆抗体具有在培养上清液中产生抗体的优点,因此能够大量产生具有一致效价的特异性的抗体,但是其缺点在于其需要长时间的产生,在能够进行免疫的动物的范围方面受到限制,并且不适用于例如免疫染色的实验。
因此,一般来说,通过产生多克隆抗体来确定抗原的特异性,并且当需要大量产生具有一致效价的抗体时制备单克隆抗体。
同时,本发明人已经研究了与前列腺癌的诊断有关的组合物,并且已经制备了基于能够更有效地诊断EN2蛋白的EN2蛋白片段或能够识别该片段的抗体用于诊断前列腺癌的组合物,从而最终实现了本发明。

本发明涉及包含以下SEQ ID NO:1、2或3的氨基酸序列的肽。
在本发明中,用作比较例的市售抗体是使用将包含以下SEQ ID NO:4或5的氨基酸序列的肽作为抗原制备的抗体。
这些肽在EN2中的位置示于图1中。
包含SEQ ID NO:1、2或3的氨基酸序列的肽是EN2蛋白的免疫原性片段,并且是具有至少一个可被EN2蛋白抗体识别的表位的EN2蛋白片段,其数量在前列腺癌患者体内增加或减少。
本发明涉及通过特异性识别上述肽来诊断EN2蛋白的存在或不存在的方法。在此,EN2蛋白可以是从哺乳动物内部提取到外部的任何实例,也可以是重组EN2蛋白。为了评价EN2蛋白的存在或不存在,可使用任何方法,只要它是用于确定蛋白质的存在或不存在或用于其量化过程的普通实验方法即可。
此外,本发明涉及特异性识别包含SEQ ID NO:1、2或3的氨基酸序列的每个肽的抗体组合物或包含该抗体组合物的诊断剂。诊断剂还可包含经标记的二抗、发色团、经抗体偶联的酶和能够与其底物或抗体结合的其他物质。
当诊断剂包含含有SEQ ID NO:1、2和3的氨基酸序列的三种类型的肽的所有相应抗体时,与使用包含任一种或两种抗体的诊断剂时相比,可更准确地诊断前列腺癌。例如,为了鉴别待诊断对象体内包含的EN2蛋白,从待诊断对象收集的样品可分成三组。对于一组,使用针对包含SEQ ID NO:1的氨基酸序列的肽的抗体确定EN2蛋白的存在和浓度,并且对于每个剩下的组,对针对包含SEQ ID NO:2的氨基酸序列的肽和针对包含SEQ ID NO:3的氨基酸序列的肽的相应抗体分别确定EN2蛋白的存在和浓度,并且可进行总共三次EN2蛋白检测实验。这样,当使用诊断剂检测EN2蛋白时,可提高诊断结果的准确性和可靠性。
此外,本发明的诊断剂可包含能够对待诊断对象体内所包含的EN2蛋白进行量化的重组EN2蛋白。
抗体是针对抗原位点的特定蛋白质分子。因此,抗体优选是与本发明的肽特异性结合的抗体,并且可包括所有多克隆抗体、单克隆抗体和重组抗体。
可使用本领域中公知的技术容易地产生抗体。因此,本发明涉及使用本发明的肽制备抗体的方法。
多克隆抗体可从通过向动物注射本发明的肽作为抗原获得的血清中获得。动物可以是任何动物宿主,例如山羊、兔、猪等。单克隆抗体可以使用杂交瘤方法(Kohler G.和Milstein C.)或噬菌体抗体文库方法(Clackson等,Marks等)制备,如本发明所属领域中广泛已知的。杂交瘤方法可使用免疫相关宿主动物(例如小鼠)的细胞和癌细胞系或骨髓瘤细胞系进行。然后,如本发明所属领域中广泛已知的,通过使用聚乙二醇的方法将两种细胞融合,在此之后可通过标准组织培养方法来使产生抗体的细胞增殖。然后,通过使用有限稀释技术的亚克隆获得均匀的细胞群,在此之后可在体外或体内使用标准技术大量培养能够产生对本发明的肽具有特异性的抗体的杂交瘤。噬菌体抗体文库方法可以以这样的方式进行:其中获得针对本发明的肽的抗体基因,并使其以融合蛋白的形式在噬菌体表面上表达,从而在体外制备抗体文库,在此之后从文库分离并且由此产生与本发明的肽结合的单克隆抗体。由此产生的抗体可通过离心、电泳、透析、离子交换色谱法、亲和色谱法等分离。
抗体可包含抗体分子的功能性片段以及具有两条全长轻链和两条全长重链的完整形式。抗体分子的功能性片段是至少具有抗原结合功能的片段,包括Fab、F(ab’)、F(ab’)2、F(ab)2、Fv等。
此外,本发明涉及如下所述的制备抗体的方法。
优选地,该方法包括(步骤1)通过使用交联剂将本发明的肽与载体蛋白连接来制备用于产生抗体的抗原;
(步骤2)将抗原与佐剂混合并乳化;
(步骤3)以7至20天的间隔向动物皮内注射包含乳化佐剂的抗原2至5次;
(步骤4)在抗原施用之后7至20天将血清从动物的全血分离;以及
(步骤5)从血清分离和纯化免疫球蛋白。
在如上制备抗体的方法中,动物可以是能够引起免疫应答的任何动物,并且优选是哺乳动物。步骤5中的免疫球蛋白可以是任何类型,但优选是免疫球蛋白G。
当使用肽1、2或3诱导免疫应答以制备本发明的抗体时,肽1、2或3使用交联剂与载体蛋白连接并由此可用作抗原。在此,载体蛋白可包括BSA、KLH、OVA等,其具有低抗原性并且仅负责携带功能,并且用于连接肽和蛋白质的交联剂可包括EDC、戊二醛(载体分子与肽的N端的连接)、琥珀酰亚胺酯(例如MBS、SMCC)、联苯胺(BDB)(与Tyr残基连接)、高碘酸盐(与碳水化合物基团的附接)、异硫氰酸盐/酯(用于用荧光染料标记抗体)等。因此,根据所使用接头的特性,可以以多种方式使用缓冲溶液。
此外,本发明涉及使用本发明的肽或特异性识别其的抗体诊断前列腺癌的方法。
诊断方法包括:
(步骤1)将体蛋白从待分析的样品分离;
(步骤2)通过使步骤1的体蛋白与本发明的抗体接触来形成抗原-抗体复合物;以及
(步骤3)定量检测和分析步骤2中形成的抗原-抗体复合物。
在诊断方法中,步骤1的样品可从待确定其前列腺癌发生或进展的对象中提取,并且其优选的实例包括哺乳动物(人或者其他动物)的组织、细胞等,其更优选的实例包括尿液、血液、血浆、血清和肝细胞。
可使用任何已知的方法分离步骤1中的体蛋白,并且可通过本领域技术人员已知的多种方法中的任一种测量体蛋白的量。
配置步骤2的抗原-抗体复合物,以使得在样品的体蛋白中包含的EN2蛋白(或本发明的肽1、2或3)与抗体彼此特异性结合。在复合物中,抗原意指EN2蛋白(或本发明的肽1、2或3)。
通过上述分析方法,可比较在对照的情况下形成的抗原-抗体复合物的量和在待确定其前列腺癌发生或进展的对象的情况下形成的抗原-抗体复合物的量,并且确定待确定其前列腺癌发生或进展的对象的EN2蛋白的表达水平,从而可直接诊断前列腺癌。
在步骤3中,可基于其中测量重组EN2蛋白浓度的标准值来确定待确定其前列腺癌发生或进展的对象样品的EN2的量。在此,最优选使用所有三种抗体而不是一种或两种抗体在体内检测EN2蛋白。详细地,取自样品的蛋白质可分成三组,并且相应抗体可与对应于相应组的蛋白质反应,由此可检测EN2。
待确定其前列腺癌发生或进展的对象的EN2蛋白的表达水平可通过测量其是否落在3.1至65.4nM的范围内来确定(Sci.Rep.2013;3:2059),该范围是前列腺癌患者尿液中的已知的EN2蛋白浓度范围。
可基于检测标记的信号幅度定量测量形成的抗原-抗体复合物的量。检测标记可选自:酶、荧光物质、配体、发光物质、微粒、氧化还原分子和放射性同位素,但是本发明不限于此。当将酶用作检测标记时,酶的实例包括但不限于:β-葡糖醛酸糖苷酶、β-D-葡糖苷酶、β-D-半乳糖苷酶、脲酶、过氧化物酶、碱性磷酸酶、乙酰胆碱酯酶、葡萄糖氧化酶、己糖激酶等。荧光物质的实例包括但不限于:荧光素、藻蓝蛋白、荧光胺等。配体的实例包括但不限于生物素衍生物等。发光物质的实例包括但不限于萤光素等。微粒的实例包括但不限于:胶体、金等。氧化还原分子的实例包括但不限于:醌、1,4-苯醌、氢醌等。放射性同位素的实例包括但不限于:3H、14C等。
诊断方法的实例可包括但不限于:western印迹、ELISA(酶联免疫吸附测定)、RIA(放射免疫测定)、放射免疫扩散、乌赫特朗尼免疫扩散(ouchterlony immunodiffusion)、火箭免疫电泳、免疫组织化学、免疫沉淀、补体结合测定、FACS(荧光激活细胞分选仪)、蛋白质芯片等。
可使用任何对象作为可应用本发明的肽或抗体的待诊断对象,只要可以在体内产生EN2蛋白即可,并且可以对所有哺乳动物(包括人)进行诊断。哺乳动物可以是任何类型的哺乳动物,例如人、狗、猫、兔、牛、山羊、猪、马等。
有益效果
本发明涉及EN2蛋白的免疫原性片段肽或特异性识别其的抗体组合物。在本发明中,通过特异性识别该肽的方法可以对EN2蛋白进行量化。此外,与现有的EN2蛋白抗体相比,使用该肽制备的抗体的检测灵敏度非常高,因此可以有效地用于诊断前列腺癌的诊断剂中。特别是当诊断出前列腺癌时,与血液中的PSA(前列腺特异性抗原)检测(其是传统的前列腺癌诊断方法)结果相比,可以产生高诊断效力。

具体实施方式
通过下列实施例将给出本发明的一些优选实施方案的更好的理解。然而,本发明不限于这些实施例,而是可以以其他形式来实施。提供这些实施例以彻底解释本发明并将本发明的精神充分地传达给本领域技术人员。
<实施例1.EN2肽选择、合成和抗原制备>
实施例1-1.选择作为抗原的肽的氨基酸序列并合成肽
使用专业抗体公司程序(抗原分析器肽工具(Antigen profiler peptide tool)-Thermo Fisher Scientific),将EN2的整个蛋白质序列切割成30个氨基酸长的肽,并且基于抗原指数,对每个肽的抗原性进行1至5的评分,从中选择评分为3.8或更高的区域。在此,抗原指数是核苷酸具有抗原性的概率的量度,其取决于将其翻译为蛋白质之后修饰导致的结构效应、立体结构等。
在由此选择的区域中,分布有许多疏水性残基的区域被排除在选择之外,因为在折叠的蛋白质结构中外部暴露被阻断,并且在基于上述标准选择的肽序列中评分最高的前10种肽中,选择了三种具有少量疏水性残基的肽。
要求上述三种肽以10mg,98%纯度合成(Anigene,Korea),并获得三种包含SEQ IDNO:1至3的氨基酸序列的肽。因此,在EN2蛋白中合成的肽序列的位置示于图1中。
实施例1-2.载体蛋白与肽的连接
实施例1-1中制备的三种肽具有小尺寸,并且当这些肽直接用作抗原时,难以诱导免疫应答,因此使用交联剂将每个肽与载体蛋白(KLH(匙孔血蓝蛋白))连接。
具体地,每个肽、载体蛋白和交联剂以1:1:1的重量比溶解,并在室温下充分反应2小时或更长时间。因此,交联剂是EDC(1-乙基-3-[3-二甲基氨基丙基]碳二亚胺盐酸盐),并且使用0.1M MES(2-[N-吗啉代]乙磺酸)(pH 4.5至5)作为结合缓冲液(该缓冲液的作用为诱导胺基水解)。
在反应终止之后,基于结合材料与未结合材料之间的尺寸差异(KLH400kD和肽3.5kD,基于结合材料与未结合材料之间的尺寸差异),使用Thermo Scientific Zeba SpinDesalting Column(#89891)将它们彼此分离。
然后,将由此与载体蛋白连接的肽用作用于抗体制备的抗原。
实施例1-3.制备用佐剂乳化的抗原
为了将抗原施用于待免疫动物,需要将具有高溶解度的抗原与佐剂偶联以在体内长时间驻留,并由此制备其中佐剂和抗原被乳化的组合物。
同样为了诱导有效的免疫应答,将实施例1-2中制备的抗原总共施用四次,并且通过混合包含灭活分枝杆菌的弗氏完全佐剂(Freund’s complete adjuvant,FCA)以使免疫应答最大化来制备用于第一次施用的抗原,并且通过混合不包含灭活分枝杆菌的弗氏不完全佐剂(Freund’s incomplete adjuvant,FIA)制备用于剩余的三次施用的抗原。
在此,为了有效地使水溶性抗原和疏水性佐剂乳化,通过探针超声将抗原和佐剂混合,并将温度维持在4℃,以免在混合期间向抗原施加热量。将抗原和佐剂以1:1的体积比混合。
<实施例2.使用抗原诱导免疫应答>
实施例2-1.选择待免疫动物
将大鼠(Wistar)用作动物以诱导对人EN2蛋白序列的免疫应答。考虑到个体特异性,针对每个肽序列在9个7周龄雌性个体中诱导免疫应答,因为免疫应答可根据个体而变化。
实施例2-2.免疫动物的实验时间表和注射方法
免疫诱导的总时间表遵循图2的程序,其中首先施用抗原并在2周后再次施用,在此之后以10天的间隔另外施用抗原两次,由此进行总共四次施用。在第二次施用之后通过尾静脉出血检查免疫应答的程度(测试出血)。将皮内注射用作施用方法,并且在单次施用时选择2至3个部位,并向每个个体注射总共200μL。
<实施例3.从免疫动物分离血清并纯化免疫球蛋白G(IgG)>
实施例3-1.从免疫动物分离血清
如图2中所示,处死在实施例2-2中施用抗原的免疫动物。使用吸入麻醉剂(异氟烷)通过腹腔静脉血液采集获得全血。将全血在37℃下维持1小时,然后以2000rpm离心20分钟,从离心的上清液中获得每个个体4至5mL的血清。
实施例3-2.分离免疫球蛋白G(IgG)
由于血清包含包括白蛋白在内的多种蛋白质,因此单独纯化免疫球蛋白G(IgG)以提高抗体的效价和特异性。
使用与和大鼠IgG特异性结合的蛋白G偶联的树脂(Protein GSepharose 4FastFlow-GE Healthcare)进行分离和纯化。将分离的血清用缓冲液(PBS)以1:1的体积比稀释,与填充的树脂结合,并用相当于珠体积2至3倍的体积的缓冲液洗涤,然后洗脱。
作为洗脱缓冲液,使用pH为2至3的缓冲液来破坏蛋白G和IgG的特异性结合。为了在洗脱之后立即将pH升至正常水平,使用Tris(pH 9)。
将血清洗脱成10个级分,并且从SDS-PAGE凝胶明显的是,从7至9级分获得IgG(图3a和图3b显示了肽3的结果,并且具体显示了在图3a中对通过血清白蛋白鉴定的血清样品进行分级,因此在图3b中确定了IgG的存在)。对于每个肽,如图3c中所示,通过从各肽的血清中除去其他蛋白质,发现最终的IgG具有高纯度(全部:全抗体,HC:抗体重链,LC:抗体轻链)。对每个最终获得的肽的抗体(IgG)进行量化,用PBS/0.2%叠氮化钠/20%甘油缓冲液稀释,将其等分并储存在-80℃下。在此,各肽序列和个体获得的IgG量示于下表1中。

<实施例4.表达和纯化重组人EN2蛋白>
为了测试制备的抗体的灵敏度和准确度,制备重组EN2蛋白。具体地,将pET28b/EN2质粒转化到大肠杆菌(Escherichia coli)(BL21/DE3)中,从而在0.1mM IPTG和37℃的条件下过表达EN2蛋白。通过用裂解缓冲液(20nM Tris-Cl(pH 8.0),300mM NaCl,20mM咪唑1×蛋白酶抑制剂混合物,1mg/mL溶菌酶)超声裂解过表达的大肠杆菌细胞并将其离心,从而仅获得水溶性蛋白质。水溶性蛋白质与EN2/His标签和Ni-NTA琼脂糖珠上的Ni亲和结合。此后,用洗脱缓冲液(20mM Tris-Cl(pH 8.0),300mM NaCl,300mM咪唑,1×蛋白酶抑制剂混合物)分离EN2蛋白,并在储存缓冲液(50mM Tris-HCl(pH 8.0),200mM KCl,0.1mM EDTA,1mM DTT,0.5mM PMSF,20%甘油)中通过透析(截留10K)除去咪唑,然后在280nm的吸光度下进行BCA(二喹啉甲酸)量化和蛋白质浓度量化。
<实验例1.评价前列腺癌细胞系(PC3细胞系)中的EN2蛋白检测能力>
用于实验的前列腺癌细胞系PC3购自ATCC(American TypeCulture Collection,Rockville,MD,USA),并用25mM添加有10%胎牛血清(FBS)和1%青霉素/链霉素(P/S)的含有HEPES的RPMI-1640(PC3)或RPMI-1640(LNCaP)培养基在5% CO2,37℃湿润培养箱中培养。
通过western印迹测定测量了该抗体(在实施例3中获得)在具有高EN2表达的前列腺癌细胞系PC3中的抗原检测能力。在此,比较和测试了本发明中制备的抗体和市售的EN2抗体(A公司、B公司)。
A公司:Thermo Fisher Scientific(PA5-14363)
B公司:Novus Biologicals(H00002020-M03)
具体地,将培养的PC3在含有蛋白酶抑制剂混合物的RIRA缓冲液中裂解,并以12000rpm离心20分钟以收集水溶性蛋白质,并通过BCA量化方法确定总蛋白质含量。
更具体地,将2×106个PC3细胞在100cm2培养皿中培养3天,然后用冷PBS(磷酸盐缓冲盐水)溶液洗涤两次,从而收集细胞,然后将其在含有蛋白酶抑制剂混合物的RIPA缓冲溶液中在冰中裂解30分钟。将细胞裂解物以13,000rpm离心20分钟,并通过BCA方法测量上清液中的蛋白质浓度,并使用4至15% SDS-PAGE分离裂解的蛋白质。将通过PAGE分离的蛋白质转移到PVDF膜,用5%脱脂乳/TBS-T(Tris-盐水+吐温20)以1:2000的比稀释每个EN2抗体,并在4℃下反应过夜,并且第二天用TBS-T洗涤三次,用5%脱脂乳/TBS-T以1:2000的比稀释辣根过氧化物酶(HRP)偶联的抗RAT抗体,并使其在室温下反应2小时。使用ECL(增强的化学发光)溶液处理用TBS-T洗涤三次的膜,因此在作为过氧化物酶与二抗结合的底物的鲁米诺被过氧化物酶氧化时发出的蓝光被光敏化为X射线胶片。
这样,在SDS-PAGE凝胶中从PC3细胞中提取的20μL全蛋白以40μg/孔、20μg/孔和10μg/孔加载到96孔板的每个孔中,然后进行western印迹测定,在此之后使用具有相同浓度的抗体(6.6nM)测量蛋白质检测能力。结果示于图4中。图4a显示了通过western印迹获得的带图像,并且图4b是显示结果的数值的图。
如图4中所示,与市售抗体(A公司、B公司)相比,本发明的抗体(在实施例3中使用肽1至3作为抗原制备)显示出非常高的检测EN2蛋白的能力(在10μg/孔的蛋白质处理条件下,使用肽1至3作为抗原制备的抗体显示出了比市售抗体高2至10倍的检测能力)。
<实验例2.评价前列腺癌细胞系(LNCap细胞系)中的EN2蛋白检测能力>
作为另一种已知具有高EN2表达的前列腺癌细胞系,使用LNCaP细胞,并且在与实验例1相同的条件下评价抗体的检测能力。结果示于图5中。图5a显示了通过western印迹获得的带图像,并且图5b是显示结果的数值的图。
如图5中所示,与市售抗体(A公司和B公司)相比,本发明的抗体(针对实施例3中制备的肽1至3)显示出优异的选择性抗原检测能力。因此,基于实验例1和2的结果,实施例3中制备的抗体显示出与EN2抗原的选择性结合,而不是细胞特异性结合。
<实验例3.评价使用重组EN2蛋白的灵敏度>
代替前列腺癌细胞蛋白,将实施例4中制备的人EN2重组蛋白以最大50ng/20μL(2.5ng/μL)(60nM)的浓度至最小5ng/20μL(0.25ng/μL)(6nM)的浓度稀释,并在与实验例1相同的条件下测量抗体的灵敏度。结果示于图6中。图6a显示了通过western印迹获得的带图像,并且图6b是显示结果的数值的图。
如图6中所示,3种类型的肽的所有抗体都表现出最低为5ng/20μL(0.25ng/μL)(6nM)的EN2蛋白检测灵敏度。这表明本发明中制备的抗体具有能够检测实际患者尿液中EN2蛋白浓度的抗体灵敏度(前列腺癌患者尿液中的EN2蛋白浓度范围:3.1至65.4nM:Sci.Rep.2013;3:2059)。
<实验例4.评价尿液中的EN2蛋白灵敏度>
考虑到来自本发明中待诊断对象的样品是前列腺癌患者的尿液,测试了是否可以检测到分泌的尿液中的EN2蛋白。具体地,将重组EN2蛋白添加到来自正常人的尿液,其浓度被确定为前列腺癌患者的尿液的典型浓度,并立即用作样品。
在此,重组蛋白的浓度和整个实验过程与实验例3中的相同。结果示于图7中。图7a显示了通过western印迹获得的带图像,并且图7b是显示结果的数值的图。
如图7中所示,尽管尿液中包含的有机/无机物质有影响,但识别三种肽的所有相应抗体都能够检测到最小浓度为0.25ng/μL(6nM)的抗原。
<实验例5.通过免疫荧光染色测试评价在细胞系(LNCap)中的检测能力>
基于抗原-抗体特异性检测前列腺癌细胞系(LNCap)的胞内EN2蛋白。使用实施例3中制备的三种类型的抗体作为一抗,并且使用抗大鼠IgG/FITC作为二抗,并且通过共聚焦激光显微术分析与分布在胞质中的抗原的结合能力。其荧光染色图像示于图8中。图8a显示了肽1的结果,图8b显示了肽2的结果,并且图8c显示了肽3的结果。
如图8中所示,可以确定所有三种类型的抗体都与分布在细胞中的EN2蛋白结合,由此即使在非变性条件下进行抗原检测,也可以容易地使用本发明的抗体。
<实验例6.评价使用重组EN2蛋白的灵敏度(Kd:解离常数)>
将重组EN2蛋白在4℃下附着于96孔板过夜,并使用含有2%脱脂乳的TBST溶液在37℃下进行封闭。在此,在将重组EN2蛋白以250ng/孔固定并且将抗体进行连续稀释的条件下进行实验(图9),并且在将抗体固定和将抗原以0.5至50ng/mL稀释的条件下进行实验(图10)。
此后,用一抗处理每个孔,并且用以1/10000稀释的HRP缀合的抗大鼠IgG作为二抗。反应之后,使用TMB(3,3',5,5'-四甲基联苯胺)溶液进行显色反应,并使用1N硫酸终止反应。
使用ELISA读数仪对所得值进行量化,并使用Prism程序的关联动力学方法计算Kd值。结果示于图9中,并且每个抗体检测到的EN2含量示于图10中。
如图9中所示,解离常数Kd是占据抗体结合位点的1/2所需的抗原量,对抗原具有高亲和力的抗体具有小的解离常数。抗体的Kd值如下:肽1:4.087×10-13;肽2:5.825×10-12;和肽3:7.739×10-13。此外,肽3的抗体显示出最高值,并且与Kd值为10-7至10-10的正常天然抗体相比,显色的抗体具有10-12至10-13的Kd值,因此表现出高效价,由此证实制备了对重组EN2蛋白抗原具有高结合能力的抗体(在根据本发明的实施例3中制备)。
参考图10和下表2,通过ELISA,所有三种抗体都能够检测到最小浓度为0.5ng/mL的EN2抗原。

因此,这些结果显示本发明的抗体被认为是优异的抗体,因为它不仅可以用于定性测试以确定癌症的存在,而且还可以用于定量测试以通过对尿液中的EN2检测与癌症进展之间的相关性进行量化来评价癌症的进展。
<实验例7.评价在前列腺癌患者尿液样品中的效力>
为了评价用于实际测试的前列腺癌患者尿液样品中的EN-2检测能力,将3个前列腺癌患者样品(p1、p2、p3)以10000g离心10分钟并分离上清液,进行western印迹。将15μL的尿液样品与5μL的4×样品缓冲液混合,并在100℃下煮沸5分钟,并进行western印迹。在此,使用的抗体浓度为3.3nM,并且使用来自健康男性的尿液作为阴性对照(H.C.)。结果示于图13中。如实验例6中通过ELISA获得的前列腺癌患者的PSA表达结果示于图12中。
参考图13,可以得出结论,本发明的所有三种类型的抗体在实际患者样品中表现出高EN-2检测能力。
