As some preferred embodiments of the present invention, the solvent is prepared as follows:
(1) Adding the extractive solution of swertia japonica Makino, the extractive solution of green tea, the leaf oil of Rosmarinus officinalis, the essential oil of herba Menthae, glycerol, 1, 2-hexanediol and 1, 3-butanediol into ultrapure water, adding the balance of ultrapure water, and stirring;
(2) Filtering the mixed solution with a 0.45-micron filter membrane, and subpackaging to obtain the solution of the essence spray. .
As some preferred embodiments of the present invention, the preparation method of the lyophilized powder is as follows:
s1, dissolving trehalose in ultrapure water to obtain a phase a;
s2, filtering and sterilizing the SOD superoxide dismutase stock solution and the recombinant collagen stock solution by using a 0.22-micron microporous filter membrane respectively, adding the sterilized SOD superoxide dismutase stock solution and the recombinant collagen stock solution into the phase a, and uniformly mixing;

Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more apparent, the following description of the present invention is given for clarity and completeness.
In this embodiment, both the SOD superoxide dismutase and the recombinant collagen are obtained by gene recombination technology and microbial fermentation. The recombinant collagen is prepared by adopting a method disclosed in Chinese patent 201911384531.4 and an invention patent named as 'a recombinant humanized III-type collagen with a functional structure and an expression method thereof'. The dry and wet master bottles are commercially available products and are used for subpackaging the solution and the dry powder and mixing the solution and the dry powder in a dry and wet mode when in use.
Example 1
The composition of the essence spray for promoting hair growth and repairing hair roots of this example was as follows (% represents mass percent):
the preparation method of the solvent comprises the following steps:
(1) Adding the extractive solution of swertia japonica Makino, the extractive solution of green tea, the leaf oil of Rosmarinus officinalis, the essential oil of herba Menthae, glycerol, 1, 2-hexanediol and 1, 3-butanediol into ultrapure water, adding the balance of ultrapure water, and stirring;
(2) Filtering the mixed solution with a 0.45-micron filter membrane, and subpackaging into wet bottle devices of dry and wet mother bottles, wherein each bottle contains 20mL of the mixed solution to obtain the solution of the essence spray.
The preparation of the freeze-dried powder comprises the following steps:
(1) Dissolving 0.4g of trehalose in 1mL of ultrapure water to obtain a phase a;
(2) Filtering 0.2mL of SOD superoxide dismutase solution and 2mL of recombinant collagen solution with 0.22 μm microporous membrane respectively for sterilization, adding into phase a, and mixing;
(3) Placing the obtained mixed solution in a dry bottle device of a dry and wet primary bottle and fixedly placing the mixed solution in a vacuum freeze dryer, and freeze-drying to obtain freeze-dried powder;
(4) And (5) assembling the dry and wet primary and secondary bottles to obtain the essence spray for promoting hair growth and repairing hair roots.
Example 2
The composition of the essence spray for promoting hair growth and repairing hair roots of this example was as follows (% represents mass percent):
the preparation method is the same as example 1.
Example 3
The composition of the essential spray for promoting hair growth and repairing hair roots of this example was as follows (% represents mass percent):
the preparation method is the same as example 1.
Comparative example 1
The essence spray for promoting hair growth and repairing hair roots was prepared in the following manner: the method comprises the following steps of adding 5g of glycerol, 3g of 1, 3-butanediol, 1g of 1, 2-hexanediol, 3g of a swertia japonica extract, 0.5g of a green tea extract, 0.3g of rosemary leaf oil, 0.1g of mint essential oil, 0.01g of SOD superoxide dismutase, 0.1g of recombinant collagen and 2g of trehalose into the mixture, and finally adding ultrapure water to the final volume of 100mL. Filtering the obtained solution for sterilization, and subpackaging in spray bottles with 20 mL/bottle to obtain the final product.
Comparative example 2
The difference is that only SOD superoxide dismutase is used as a single component in the freeze-dried powder as in example 1.
Comparative example 3
The difference is that only a single component of recombinant collagen is present in the lyophilized powder as in example 1.
Comparative example 4
The difference is that only the vehicle had no lyophilised components as in example 1.
Comparative example 5
The difference from example 1 is that the recombinant collagen was replaced with equal mass of hydrolyzed keratin in the lyophilized powder.
Comparative example 6
The difference from example 1 is that the aqueous solution of swertia japonica Makino is not added.
Comparative example 7
The difference from example 1 is that the green tea extract was not added to the vehicle.
Investigation example 1 stability test
Electrophoresis analysis is carried out on the essence spray obtained in the comparative example 1 and the essence spray obtained in the example 1 in 12 months according to the compound essence spray obtained in the application example 1 in 12 months, the stability result is shown in figures 1 and 2, and the stability of the essence spray obtained in the example 1 is obviously superior to that of the comparative example 1.
Examination example 2 evaluation of safety
(1) Skin irritation test
The skin irritation test and evaluation were performed as specified in 2015 edition "technical standards for cosmetic safety", specifically: 4 white rabbits are taken, and 24 hours before the test, the hairs on two sides of the spine of the experimental animal back are cut off, the epidermis cannot be injured, and the hair removing range is about 3cm multiplied by 3cm on the left and the right respectively. About 0.5mL of the essence spray prepared in example 1 was applied directly to the skin, covered with two layers of gauze (2.5 cm. Times.2.5 cm), and fixed with a non-irritating adhesive tape and bandage. The other skin served as control. The application time is 2h by adopting a sealing test. After the test is finished, the residual test substance is removed by warm water or a non-irritant solvent. Skin reactions at the smeared sites were observed at 1, 24, 48 and 72h after removal of the test substances, skin reaction scores were made according to table 1, comprehensive evaluations were made with the mean value of the scores of the test animals, and skin irritation intensity was judged according to table 2 based on the highest mean value of the scores at each observation time point of 24, 48 and 72 h.

According to the skin irritation response scoring results of the product in table 3, the product is judged to be non-irritating to the skin.
(2) Skin allergy test
The Test and evaluation were carried out according to the skin allergy Test-topical closed skin Test (BT) specified in the cosmetic safety specification on the 2015 edition, in particular: 15 healthy guinea pigs were divided into 2 groups, a control group (5), and an experimental group (10). About 24 hours before the test, the left side of the back of the guinea pig is unhaired and removedThe wool range is 4cm 2 -6cm 2 . And in the induction contact stage, about 0.2ml of the test object is coated on the skin of a hair removal area of the experimental animal, covered by two layers of gauze and a layer of cellophane, and sealed and fixed by a non-irritant adhesive tape for 6 hours. During the challenge period, 14-28 days after the last induction, approximately 0.2ml of the test substance was applied to the right 2cm x 2cm depilated area of the guinea pig back (24 h before challenge), covered with two layers of gauze and a layer of cellophane, and fixed with a non-irritating adhesive plaster for 6h. Skin reactions were observed 24h and 48h after challenge contact and scored according to table 4. The experiment was performed with sterile water as a negative control, water alone as a control at the induction contact, and the test substance at the challenge contact.

According to the scoring result of the skin allergy test-local closed coating test of the product in the table 5, the product is judged to have no skin allergy.
Examination example 3 rat Hair regeneration experiment
54 SD rats (200 g +/-20 g) with male and female halves; the hair is removed in the back area of the rat by shaving, and the skin of the shaved area is uniformly coated with depilatory cream (Veet) with the area of 3 x 3cm, 1 time every two days and 10 days later, a depilatory area is formed at the administration position, namely the molding is successful. 54 rats which are successfully modeled are selected and divided into 9 groups, and each group is half a female and half a male. 8 groups of rats were administered the essence sprays of examples 1 to 3 and comparative examples 2 to 7, respectively, and group 9 was administered physiological saline treatment as a blank control group; the administration mode is that essence spray is sprayed on the formed depilation region, 1mL is sprayed each time, and the spraying is carried out once a day; the administration was continued for 30 days. The amount of new hair is observed and recorded, and the evaluation standard is shown in a table 6; the length of 10 hairs of each group of animals was measured by pulling out the hair from the administration area, and the average length of the hair of each group of rats was calculated, and the calculation results are shown in Table 7.

The essence spray prepared by the embodiment of the invention can effectively promote the growth of the hair of the rat and nourish the hair root. Compared with the comparative examples 2 to 7, the SOD superoxide dismutase compounded recombinant collagen can better promote the hair growth, stabilize hair roots and achieve better hair growth and hair care effects by combining the swertia japonica extract and the green tea extract.
Examination example 4 Effect of essence spray on SOD and MDA contents and Col I and Col III expressions in rat tissues
SOD and MDA contents and Col I and Col III expression levels in rat skin tissues in the investigation example 3 were analyzed. Compared with the detection results of comparative examples 2 to 4 (see table 8), the components of the essence spray prepared by the embodiment of the invention are synergistic, so that the expression of type I collagen and type III collagen in rat skin tissues can be effectively promoted, the proliferation and differentiation of hair follicle stem cells are stimulated, and damaged cells are rapidly repaired; SOD activity in tissues is obviously improved, MDA is obviously reduced, free radicals in rats are effectively removed, skin oxidative damage is protected, the activity of damaged cells is recovered, and then skin blood microcirculation is improved, and hair follicle development is promoted. The result shows that the SOD superoxide dismutase compounded recombinant collagen combined with the swertia japonica extract and the green tea extract can better repair and nourish hair roots and promote hair growth, and the hair growth and hair care effects of synergistic effect of 1+1 & gt 2 are achieved.

Investigation example 5 human body efficacy test
The essence sprays prepared in example 1 and comparative examples 2 to 4 were individually distributed to 50 test persons for testing. The use method of the essence spray is operated once a day by a tester, the tester scores the use effect of the essence spray according to the tester, each score is 10, and the higher the score is, the better the effect is. The results of the usage effect scoring are shown in table 9.

As can be seen from the data in table 9, the efficacy of the essence spray of example 1 was highly evaluated by the testers, and the total score was 25 minutes or more, and the results show that the components of the essence spray for promoting hair growth and repairing hair roots prepared by the present invention are synergistic, effectively remove in vivo toxins and free radicals, repair damaged hair follicle cells, promote hair growth, repair and nourish hair roots, and maintain healthy scalp microecology.
Claims (10)
1. The essence spray for promoting hair growth and repairing hair roots is characterized by comprising a solvent and freeze-dried powder:
the menstruum comprises the following components in percentage by mass: 3 to 10 percent of glycerin, 0.1 to 10 percent of 1, 3-butanediol, 0.5 to 5 percent of 1, 2-hexanediol, 2 to 4 percent of swertia japonica extract, 0.1 to 2 percent of green tea extract, 0.3 to 0.5 percent of rosemary leaf oil, 0.1 to 0.3 percent of mint essential oil and the balance of ultrapure water;
the freeze-dried powder comprises the following components in percentage by mass and volume with a solvent: SOD superoxide dismutase (SOD) at a concentration of 1mg to 5mg/20mL, recombinant collagen at a concentration of 10mg to 50mg/20mL, and trehalose at a concentration of 0.2g to 1g/20mL.
2. The essence spray for promoting hair growth and repairing hair roots according to claim 1, wherein the molecular weight of the SOD superoxide dismutase is 10-30kDa, and the specific activity of the SOD superoxide dismutase is 10000-30000U/mg.
3. The essence spray for promoting hair growth and repairing hair roots according to claim 1, wherein the molecular weight of the recombinant collagen is 25kDa to 90kDa.
4. The essence spray for treating alopecia and repairing hair roots of claim 1, wherein the SOD superoxide dismutase and the recombinant collagen are obtained by gene recombination technology and microbial fermentation.
5. The essence spray for promoting hair growth and repairing hair roots according to claim 1, which is composed of a solvent and a lyophilized powder:
the menstruum comprises the following components in percentage by mass: 5% of glycerin, 3% of 1, 3-butanediol, 1% of 1, 2-hexanediol, 3% of a Japanese swertia herb extract, 0.5% of a green tea extract, 0.3% of rosemary leaf oil, 0.1% of mint essential oil and the balance of ultrapure water;
the freeze-dried powder comprises the following components in percentage by mass and volume with a solvent: SOD superoxide dismutase (SOD) at the concentration of 2mg-4mg/20 mL, recombinant collagen at the concentration of 10mg-20mg/20 mL and trehalose at the concentration of 0.4g/20mL.
6. The preparation method of the essence spray for treating alopecia and repairing hair roots according to any one of claims 1 to 5, which specifically comprises the steps of preparing a solvent and preparing a freeze-dried powder.
7. A method for preparing the essence spray for promoting hair growth and repairing hair roots according to claim 6, which comprises:
(1) Adding swertia japonica Makino extract, green tea extract, rosewood leaf oil, mint essential oil, glycerol, 1, 2-hexanediol and 1, 3-butanediol into ultrapure water, supplementing the balance of ultrapure water and stirring uniformly;
(2) Filtering the mixed solution with a 0.45-micron filter membrane, and subpackaging to obtain the solution of the essence spray.
8. The method for preparing essence spray for promoting hair growth and repairing hair roots according to claim 6, wherein the method for preparing the freeze-dried powder comprises the following steps:
dissolving trehalose in ultrapure water to obtain a phase a;
filtering SOD superoxide dismutase stock solution and recombinant collagen stock solution with 0.22 μm microporous membrane respectively for sterilization, adding into phase a, and mixing;
and S3, placing the obtained mixed solution in a container, and freeze-drying to obtain freeze-dried powder.
9. The method for preparing an essence spray for promoting hair growth and repairing hair roots according to claim 6, further comprising the step of dispensing and combining the solvent and the lyophilized powder in dry and wet mother-son bottles.
