The present comparative example provides a scalp care essence for preventing hair loss and growing hair, which is different from example 1 only in that it does not contain the polygonum multiflorum extract, and the reduced amount thereof is distributed to the mass of hinokitiol, the swertia japonica extract, the biota orientalis extract, and the ginseng extract according to the mass ratio, and the other conditions are maintained.

The preparation method comprises the following steps:
(1) mixing and stirring half of the added amount of deionized water, sodium benzoate and EDTA disodium, and heating to 70 ℃ to obtain a material body A; heating hinokitiol to 70 ℃, and then adding half of the added amount of 1, 3-butanediol to obtain a material body B;
(2) mixing and stirring the material body B and the material body A, and cooling to 30 ℃ to obtain a material body C;
(3) mixing and stirring phenoxyethanol, the rest of 1, 3-butanediol, the rest of polyglycerol-10 laurate and the rest of deionized water to obtain a material body D;
(4) and mixing and stirring the material D and the material C, and then mixing and stirring the mixture with the swertia japonica extract, the biota orientalis extract, the polygonum multiflorum extract, the ginseng extract and the essence at 25 ℃ to obtain the anti-hair loss and hair-growing scalp care essence containing hinokitiol.

The comparison example provides an anti-hair loss and hair-growing scalp care essence, and the formula composition of the anti-hair loss and hair-growing scalp care essence is different from that of example 1 only in that the mass percentage content of polyglycerol-10 laurate is 0.05%, the reduction amount is distributed on the mass of deionized water, and other conditions are kept unchanged. Test example 1:
determination of autophagy activity of human hair papilla cells:
human dermal papilla cells cultured in the dermal papilla cell growth medium were seeded in a 96-well plate in an amount of 10,000cells/well and cultured for 24 h. Then, human dermal papilla cells were cultured in a dermal papilla cell basal medium for 48 hours with and without a hinokitiol solution (3.125. mu.g/mL, solvent 1, 3-butanediol) with 10. mu. mol/L chloroquinone as a positive control. Fluorescent reagents were prepared by adding 10. mu.L of Hoechst 33342 nuclear stain and 10. mu.L of Cyto-ID Green Detection Reagent 2 to 1mL of 10 × assay buffer and 9mL of purified water, respectively. After washing with 1 × assay buffer, 100 μ L of fluorescent reagent was added to each well and incubated in an incubator for 30 min. The fluorescent reagent was recovered, washed twice with 200. mu.L of 1 × assay buffer, and 100. mu.L of 1 × assay buffer was added. Hoechst 33342 (excitation: 360/40, emission: 460/40) and Cyto-ID Green Detection Reagent 2 (excitation: 485/20, emission: 528/20) were measured with a fluorescence plate analyzer. Experiments were performed at n-3 and mean and standard deviation were calculated.
As a result, as shown in FIG. 1, hinokitiol at a concentration of 3.125. mu.g/mL significantly enhanced the fluorescence intensity of Cyto-ID corrected with H33342, i.e., hinokitiol significantly increased the autophagy activity of human dermal papilla cells.
Test example 2:
determination of mitochondrial autophagy activity of human hair papilla cells:
human dermal papilla cells cultured in a dermal papilla cell growth medium were seeded in an amount of 200,000cells/well onto 6-well plates and cultured for 24 h. Then, human dermal papilla cells were cultured in a dermal papilla cell basal medium for 48 hours with and without a hinokitiol solution (3.125. mu.g/mL, solvent 1, 3-butanediol) with 500nmol/L of Rapamicin as a positive control. Cells were harvested with trypsin EDTA solution and 10. mu.L of mitochondrial dye at 100. mu. mol/L was added to 10mL of serum-free medium to prepare fluorescent reagent 2. The collected cells were washed with serum-free medium, 1mL of fluorescent reagent 2 was added, respectively, and placed in an incubator for 30 min. Fluorescent reagent 2 was recovered, washed twice with 200. mu.L serum-free medium, and then 100. mu.L serum-free medium was added. The percentage of cells stained with mitochondrial autophagy dye was measured with a Image Cytometer Tali Image Cytometer.
As shown in FIG. 2, hinokitiol at a concentration of 3.125. mu.g/mL can significantly increase the mitophagy dye-positive cell rate, i.e., hinokitiol can significantly increase the mitophagy activity.
Test example 3:
testing the blood flow volume of a human body:
after applying the oil to the ring finger of a non-dominant hand (generally, the default is left hand), video of blood flow was recorded by using a blood flow meter, and then 0.2g of a solution of swertia japonica extract (1, 3-butanediol having an active content of 1.5% and a solvent of 49.25%) was applied to the back of the hand of the same hand, and video was recorded after the sample was absorbed. And judging whether the effect of promoting blood circulation is effective or not according to the video animation result.
As a result, as shown in the static chart of FIG. 3, the capillaries smeared with the swertia japonica Makino extract liquid became more clearly visible due to the increase in the blood flow volume and the increase in the blood flow rate. In addition, the video of the test results also showed an increase in the blood flow rate of the capillaries after application of the swertia japonica extract.
Test example 4:
and (3) testing the system stability:
because the hinokitiol in the anti-hair loss and hair-growing scalp care essence is slightly soluble in water and is difficult to apply to an alcohol-free formula, the hinokitiol is solubilized into the formula creatively through the mild and non-irritant polyglycerol-10 laurate.
The anti-alopecia and hair-growing scalp care essences prepared in examples 1 to 3 and comparative examples 1 to 13 were left at 25 ℃, 40 ℃, 50 ℃ and 5 ℃ for 30 days, and the appearance of the samples was visually observed, and the test results are shown in table 1 (o indicates that the state is intact, the uniform and stable liquid state is maintained, and x indicates that delamination and precipitation occur under any of the test conditions).

As can be seen from the data in Table 1, the anti-hair-loss and hair-growing scalp care essence disclosed by the invention solubilizes hinokitiol into a formula through the mild and non-irritant polyglycerol-10 laurate, so that the stability is good, and the shampoo is mild and non-irritant.
Test example 5:
anti-alopecia hair-growing clinical test:
130 volunteers (39 30-40 years old, 39 40-50 years old and 52 50-60 years old) were selected and randomly divided into 13 groups, 10 persons (3 30-40 years old, 3 40-50 years old and 4 persons 50-60 years old) were selected, and the anti-hair loss and hair-growing scalp care essence liquids prepared in examples 1-3 and comparative examples 1-10 were respectively tried on each group.
Two selected areas of the same area (2cm x 2cm) of the subject were shaved and the bare scalp was applied with the test sample on one side and without any hair loss prevention hair growth product treatment on the other side as a blank control. The scalp condition before and after shaving was photographed by using a microscope for 30 days after applying the hair cream 1 time per day, 200. mu.L each time, continuously for 30 days. The average diameter, average density and average length of the grown hair were observed and counted by image analysis. And the subject is allowed to carry out subjective evaluation and scoring (comprehensive use feeling, actual feeling of the hair loss prevention and hair growth effect and mild feeling) on the product, the scoring range is 1-5 points, then the average value is taken, the score is 1-2 is poor, the score is 2-3 is general, the score is 3-4 is good, and the score is 4-5 is good. The results are shown in table 2:

From the results of table 2, it can be seen that: compared with the comparative example, the anti-hair-loss and hair-nourishing scalp care essence has unexpected synergistic effect on the anti-hair-loss and hair-nourishing effect of five specific functional components, namely hinokitiol, the swertia japonica extract, the biota orientalis extract, the polygonum multiflorum extract and the ginseng extract. Meanwhile, the user feels good in comprehensive use, actual feeling of the hair loss prevention and hair growth effect and mildness.
The applicant states that the invention is described by the above embodiments to describe the hair loss prevention and hair growth promotion scalp care essence, the preparation method and the application thereof, but the invention is not limited to the above embodiments, that is, the invention is not limited to the above embodiments. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
The preferred embodiments of the present invention have been described in detail, however, the present invention is not limited to the specific details of the above embodiments, and various simple modifications may be made to the technical solution of the present invention within the technical idea of the present invention, and these simple modifications are within the protective scope of the present invention.
It should be noted that the various technical features described in the above embodiments can be combined in any suitable manner without contradiction, and the invention is not described in any way for the possible combinations in order to avoid unnecessary repetition.

Claims (10)

1. The anti-hair-loss and hair-growing scalp care essence containing hinokitiol is characterized by comprising the following preparation raw materials: hinokitiol, swertia japonica extract, cacumen Platycladi extract, Polygoni Multiflori radix extract, Ginseng radix extract, and polyglycerol-10 laurate, while not containing ethanol.
2. The anti-hair-loss scalp care essence containing hinokitiol according to claim 1, wherein the anti-hair-loss scalp care essence is prepared from the following raw materials by 100% of the total mass of the raw materials: 0.01-1% of hinokitiol, 1-5% of swertia japonica extract, 1-5% of biota orientalis extract, 1-5% of polygonum multiflorum extract, 1-5% of ginseng extract and 0.1-2% of polyglycerol-10 laurate.
3. The anti-hair-loss and hair-growing scalp care essence containing hinokitiol according to claim 1 or 2, wherein the anti-hair-loss and hair-growing scalp care essence is prepared from a raw material further comprising any one or a combination of at least two of deionized water, a humectant, a preservative, a chelating agent or an essence.
4. The anti-hair-loss scalp care essence containing hinokitiol according to claim 3, wherein the moisturizer comprises 1, 3-butanediol.
5. The anti-alopecia, hair-growing scalp care essence containing hinokitiol according to claim 3, wherein the preservative comprises sodium benzoate and/or phenoxyethanol.
6. The anti-hair-loss scalp care essence containing hinokitiol according to claim 3, wherein the chelating agent comprises disodium EDTA.
7. The method for preparing anti-hair loss and hair-growing scalp care essence containing hinokitiol according to any one of claims 1 to 6, comprising the steps of:
(1) mixing and stirring part of deionized water, a water-soluble preservative and a chelating agent, and heating to obtain a material body A; mixing hinokitiol with a part of polyglycerol-10 laurate, stirring, heating, and adding a part of humectant to obtain a material body B;
(2) mixing and stirring the material body B and the material body A, and cooling to obtain a material body C;
(3) mixing and stirring the fat-soluble preservative, the rest of the humectant, the rest of the polyglycerol-10 laurate and the rest of the deionized water to obtain a material body D;
(4) and mixing and stirring the material D and the material C, and then mixing and stirring with the swertia japonica extract, the biota orientalis extract, the polygonum multiflorum extract and the ginseng extract to obtain the anti-hair loss and hair-growing scalp care essence containing hinokitiol.
8. The method of claim 7, wherein the fraction is 10-90% of the total;
preferably, the heating in step (1) is heating to 60-80 ℃;
preferably, the temperature reduction in the step (2) is to reduce the temperature to 10-40 ℃.
9. The method according to claim 7 or 8, wherein the step (3) further comprises mixing and stirring the essence;
preferably, the mixing and stirring of step (3) is carried out at 15-35 ℃.
10. Use of hinokitiol-containing anti-hair loss scalp care essence according to any one of claims 1 to 6 for the preparation of an anti-hair loss hair-growing product;
preferably, the anti-hair loss and hair-nourishing product comprises shampoo, shampoo cream or hair conditioner.
