背景技术
铁缺乏在人群中很常见。世界卫生组织的食品法典(CODEX Alimentarius)规定,富含植物性食物的膳食(如主要在印度消费的那些)的铁营养参考值(NRV)为22mg。然而,超过六分之一的全球人口已知患有营养性铁缺乏。贫血是铁缺乏的常见结果。
铁强化的食物是向铁缺乏人群提供铁的一种方式。这在如印度等国家尤其需要,因为这些国家的平均铁摄入量仅为推荐膳食摄入量的50-80%。此外,在印度,这种铁只能作为非血红素铁获得,因为该国大部分人口是素食者。
用于强化食品和饮料的铁的常见来源包括硫酸亚铁、乳酸亚铁、葡萄糖酸亚铁、甘氨酸亚铁和柠檬酸亚铁。优选可溶性形式的铁,因为不可溶性或微可溶性的铁来源(如单质铁和一些铁盐)显示出较差的生物利用度。
茶是一种流行、便宜的在全世界消费的饮料。因此,铁化合物强化茶会是一种极好的提供铁的方式。但是,可溶性铁源强化茶有一个众所周知的问题,因为存在于茶中的多酚与铁化合物络合会形成不可溶性铁-多酚复合物,其颜色较深。这些复合物在制备茶饮料的过程中形成,并且生产的茶液更深且与非强化的茶颜色不同。铁强化茶所引起的这种感官变化是消费者所不能接受的,因此之前一直在寻求解决这一问题的办法。
WO10116379涉及与用铁盐强化茶相关的问题,并指出为了这种铁强化的目的,向茶中添加任何常用的铁盐会导致茶多酚与这种铁盐的复合,导致茶的变色。因此,WO10116379公开了一种富铁的干茶,其具有99.5%-99.88%的常规茶、0.1%-0.3%的铁盐和0.02%-0.2%的淀粉基质,其中铁盐选自双甘氨酸硫酸亚铁和三甘氨酸铁。需要指出的是,WO10116379试图使用淀粉基质解决茶变色的问题。它没有使用或公开任何颜色校正组分。
IN01402CH2005要求提供一种用于食品/食品补充剂的强化剂的制备方法,其特征在于强化剂包括颜色相容的金属氨基酸螯合物或颜色相容的金属尿素复合物,其中将强化剂(如制备的有色金属氨基酸螯合物和/或有色金属尿素复合物)溶解在含有所需量的螯合剂的溶液中,然后可选地加入酸,得到无色溶液,该溶液可干燥并与食品/食品补充剂混合,或喷洒在食品/食品补充剂上并干燥,以得到强化的食品/食品补充剂而其颜色不发生任何变化。
IN01402CH2005指出,“金属”应理解为人体所需的宏观或微观营养素所需的金属,而“氨基酸”应理解为包括尿素或硫脲(被视为碳酸酰胺)在内的任何氨基酸。
IN01402CH2005列出了金属氨基酸螯合物的至少11种来源,可选自包括以下:钙、钴、铬、铜、铁、镁、锰、钼、钾、硒、锌等。它还列出了超过40种不同的螯合剂化合物,可以选自包括:羟基亚乙基-1,1-二膦酸、聚磷酸铵、骨磷酸盐、柠檬酸钙、磷酸二氢钙、乙二胺四乙酸二钠钙、葡萄糖酸钙、聚磷酸钙、硫酸钙、柠檬酸、磷酸氢二钾、乙二胺四乙酸二钠、焦磷酸二钠、DL-酒石酸、葡萄糖δ-内酯、柠檬酸异丙酯混合物、L(+)酒石酸、氧化硬脂精(Oxystearin)、三磷酸五钠、磷酸、柠檬酸二氢钾、磷酸二氢钾、聚磷酸酯钾、L(+)酒石酸钠钾、二乙酸钠、柠檬酸二氢钠、磷酸二氢钠、葡萄糖酸钠、L(+)酒石酸钠、偏磷酸钠、聚磷酸钠、硫代硫酸钠、山梨醇、山梨醇糖浆、柠檬酸硬脂酯、焦磷酸四钠、柠檬酸三乙酯、柠檬酸三钾、磷酸三钾、柠檬酸三钠、磷酸三钠或其组合。
IN01402CH2005还提到氨基酸螯合剂如“甘氨酸亚铁”是迄今为止发现的最具生物可利用性、毒性最小,与食品成分反应呈惰性,味道平淡的螯合剂,符合所期望的感官特性。
然而,如下文进一步所述,甘氨酸亚铁对与茶的反应不是惰性的,而且令人惊讶的是,添加了甘氨酸亚铁的茶基饮料的颜色会发生不期望的变化。当茶加热到实际沸腾时,情况尤其如此。
本发明的产品特别适用于诸如印度的人群,如前所述,铁仅作为非血红素铁可用于该国大部分素食人群。同样值得注意的是,印度的茶的冲泡有特定的特征,包括将茶加热到实际沸腾。这种冲泡对不溶性铁-多酚复合物的形成特别有利,因此这种强化茶中铁源或多种铁源特别容易引起颜色变化和变暗。因此,一些颜色校正剂必须特别有效地防止颜色变化和变暗。
发明内容
本发明人现发现,使用甘氨酸亚铁(甘氨酸亚铁)作为铁源来提供强化的茶基饮料是可能的。虽然甘氨酸亚铁通常会引起感官问题(包括冲泡产品的不期望的变暗,特别是当茶冲泡涉及将茶加热到实际沸腾),但本发明人现发现,如果甘氨酸亚铁与Na2H2EDTA组合使用,那么冲泡产品就不会出现与使用甘氨酸亚铁强化相关的变暗或其他颜色变化。
甘氨酸亚铁和Na2H2EDTA可以在制茶过程中掺入。它们也可适用于叶基饮料产品、即饮形式、水可溶性茶粉或颗粒或液体茶饮料。
获得的茶液具有良好的颜色和透明度,其用铁强化,并且没有与铁强化的茶相关的不良的视觉和感官属性,例如变暗和颜色变化。
因此,本发明提供一种产品,其包含:
a)茶组分,
b)甘氨酸亚铁;和
c)Na2H2EDTA。
优选地,所述产品为饮料前体。换言之,所述产品包含制备茶基饮料的茶组分。茶组分优选为待冲泡的茶叶、或待溶解的茶粉或待溶解的茶粒。

具体实施方式
产品
本发明涉及包含茶组分的铁强化的产品。本发明的产品涵盖即饮茶(包括瓶装茶基饮料如冰茶)、叶基饮料产品、即饮形式、水可溶性茶粉或颗粒或液体茶饮料。
本发明特别涉及饮料前体产品-换言之,茶以干燥的形式提供,并由消费者加水的产品。因此,茶组分可以是待冲泡的茶、或待溶解的茶粉、或待溶解的茶粒或其组合。
茶组分
为实现本发明目的,“茶”是指来自Camellia sinensis var.sinensis和/或Camellia sinensis var.assamica的材料。术语“叶茶”指的是来自茶植物的叶和/或茎的材料,处于未浸泡的状态(即没有经过溶剂提取的材料)。换句话说,术语“叶茶”指茶制造的最终产品(有时被称为“制茶”)。
如本文所用,术语“红茶”基本上指发酵茶,其中“发酵”指当某些内源性酶和底物聚集在一起时,茶所经历的氧化和水解过程。在所述的发酵过程中,叶和/或茎中的无色儿茶素被转化为黄色/橙色到深褐色的多酚类物质的复杂混合物。例如,红茶叶茶可以通过以下步骤从新鲜的茶材料中制造出来:萎凋、浸渍、发酵和干燥。在“Tea:Cultivation toconsumption”(由K.C.Wilson&M.N.Clifford编辑,1992出版)的第14章中可以找到红茶生产更详细的描述。
因此,本发明的茶组分可以源自植物茶树(Camellia sinensis)。茶组分可以为绿茶或红茶,优选地,茶组分为红茶。茶组分可以为叶茶或茶提取物或茶粉。优选地,茶组分为叶茶。更优选地,茶组分为红茶叶茶。
为了提供茶基饮料预期的感官属性、所需的多酚水平或两者均有,产品可包含按干重计95-99.9wt%的茶组分,更优选97-99.5wt%,还更优选98-99wt%,最优选约98.5wt%。
每份产品可以包含1g-10g的茶组分,更优选每份1.25g-5g,还更优选每份1.5g-4g,最优选每份约2g的茶组分。
如本文所用,术语“份”是指单份产品所需的量。就即食产品而言,一份通常是200至300ml的瓶或罐。对于饮料前体产品(换言之,茶以干燥的形式提供,并由消费者加水的产品),一份是制作饮料所需的饮料前体产品的量。
铁源
本发明的产品用铁强化。具体地,用甘氨酸亚铁强化。
甘氨酸亚铁的CAS号为20150-34-9,分子式为C4H8FeN2O4,其结构如下:
甘氨酸亚铁(FeBisglycinate)的同义词包括:铁二甘氨酸(iron bisglycinate);甘氨酸亚铁(ferrous bisglycinate)和双甘氨酸铁(II)盐(Bisglycine iron(II)salt)。
如上所述,归功于本发明,产品能够提供高水平的铁强化,且不会出现变暗或颜色变化的问题。产品包含按干重计0.03-3wt%的甘氨酸亚铁,更优选按干重计0.1-1wt%,还更优选按干重计0.2-0.5wt%,最优选按干重计约0.4wt%的甘氨酸亚铁。
每份产品可以包含1-30mg甘氨酸亚铁,更优选每份2.5-20mg,还更优选每份5-15mg,最优选每份约10mg的甘氨酸亚铁。
世界卫生组织的食品法典(CODEX Alimentarius)规定,富含植物性食物的膳食(如主要在印度消费的那些)的铁营养参考值(NRV)为22mg。由于颜色校正剂的存在,产品可以包含高水平的铁而不会出现变暗或颜色变化。因此,每份产品可包含前述铁的NRV的1-50%,优选2-35%,更优选3-20%,还更优选4-10%,最优选每份为铁的NRV的约5%。
每份产品可包含0.05mg-10mg铁,优选0.1mg-7.5mg,更优选0.25mg-5mg,还更优选0.5mg-2mg,最优选每份产品包含约1mg的铁。
为了避免疑义,当铁本身被表述为质量的量时,指的是产品部分中铁本身的量,而不是铁来源。例如1mg的铁是指1mg的铁离子,而不是指1mg的甘氨酸亚铁。
颜色校正剂
本发明的产品采用颜色校正组分,以防止在茶基产品中提供甘氨酸亚铁时引起的颜色变化。本发明为此目的采用Na2H2EDTA。
Na2H2EDTA的CAS号为6381-92-6,分子式为C10H14N2Na2O8,其结构如下:
Na2H2EDTA的同义词包括:二氢乙二胺四乙酸二钠;乙二胺四乙酸二钠盐;乙二胺四乙酸二钠二水合物;EDTA二钠盐;EDTA-Na2;依地酸(Edathamil);依地酸二钠盐二水合物和西奎斯特林(Sequestrene)Na2。
产品可以包含按干重计0.07 -7wt%的Na2H2EDTA,更优选按干重计0.2-5wt%,还更优选按干重计0.5-2wt%,最优选按干重计约1wt%的Na2H2EDTA。
每份产品可以包含5-100mg的Na2H2EDTA,更优选每份10-60mg,还更优选每份15-45mg,最优选每份约35mg。
颜色校正剂:铁源的摩尔比
产品可以包含Na2H2EDTA:甘氨酸亚铁的摩尔比为0.5:1至4:1,优选0.75:1至3.5:1,更优选1:1至3:1,还更优选1.5:1至3:1,最优选约2:1。
如前所述,本发明发现,使用所要求的铁源和颜色校正剂的组合,能够校正与茶基饮料的铁强化有关的变暗和颜色变化。
暗度和整体颜色校正系数
茶基饮料的颜色和暗度可以用CIE 1976L*a*b*颜色空间的坐标来表达。CIE L*a*b*值可以根据ISO/CIE联合标准(ISO 11664-4:2008(CE);CIE S 014-4/E:2007)通过色度测量法进行测量。颜色用三个值来表示:
-L*代表从黑色(0)到白色(100)的亮度,
-a*从绿色(-)到红色(+),和
-b*从蓝色(-)到黄色(+)。
样品可以相互比较,以确定这些值的差异(ΔL*、Δa*、Δb*),从这些值中还可以计算出总色差Delta E*(ΔE*)。
可以理解的是,在本发明的上下文中,ΔL*和ΔE*值是特别有意义的,因为它们可以分别用于确定正常的、未强化的茶基饮料的暗度和颜色,并将其与含有各种铁源和潜在颜色校正剂的茶基饮料进行比较。
本发明旨在提供一种提供铁强化茶基饮料的产品,其利用Na2H2EDTA来校正甘氨酸亚铁引起的暗度和整体颜色变化,从而使暗度和整体颜色尽可能地接近非强化茶基饮料。这可以分别表示为“暗度校正系数”(DCF)和“整体颜色校正系数”(OCCF)。
DCF的计算方法如下:
DCF=L* 样品-L* 空白
-其中L* 样品是由根据本发明的产品得到的强化和颜色校正的茶基饮料的L*值;以及
-其中L* 空白是仅有茶组分而没有铁源或颜色校正剂的同一茶产品的L*值。
相应地,DCF值优选-5至10,更优选-4至7.5,还更优选-3至5,仍更优选-2至4,最优选-0.5至3。
OCCF的计算方法如下:
OCCF=E* 样品-E* 空白
-其中E* 样品是由根据本发明的产品得到的强化和颜色校正的茶基饮料的E*值;以及
-其中E* 空白是仅有茶组分而没有铁源或颜色校正剂的同一茶产品的E*值。
相应地,DCF值优选0至15,更优选0至10,还更优选0至9,仍更优选1至8,最优选3至7。
优选地,根据40℃的茶基饮料计算DCF和OCCF。
如本文所用,术语“包含”涵盖术语“基本上由…组成”和“由…组成”。除非另有说明,本文包含的所有百分比和比例均按重量计算。应该注意的是,在指定任何数值或数量的范围时,任何特定的上限数值或数量都可以与任何特定的下限数值或数量相关联。
除了在实施例和比较例中,描述中表示材料数量、反应条件、材料的物理特性和/或用途的所有数字都应理解为前面有“约”。

实施例
评估甘氨酸亚铁对两种不同茶饮料颜色的影响。也测试各种化合物校正这些颜色变化的能力。
材料
测试两种茶饮料:
-实施例1-立顿黄牌(Lipton Yellow label)茶包,源自波兰的(购自波兰当地超市)
-实施例2-立顿黄牌(Lipton Yellow label)散茶,源自KSA(沙特阿拉伯王国),散茶(购自KSA当地超市)
测试的铁源为:
-甘氨酸亚铁(CAS号20150-34-9);含有23.2%wt的铁(供应者:Dr.Paul LohmannGmbH&Co.KGaA,Hauptstrasse 2,31860Emmerthal,Germany)
测试的潜在颜色校正剂为:
-焦磷酸二钠(二盐基NaPP)(CAS号:7758-16-9)

-抗坏血酸(CAS号:50-81-7)
ο供应者:Huijbregts Groep B.V.
-柠檬酸*H2O(CAS号:77-92-9)

样品制备
茶饮料:
茶饮料制备如下:
实施例1:
-将来自波兰的立顿黄牌的1个茶包放在瓷杯中。
-加入200ml沸水,将茶包浸泡2分钟((水接触到茶时,计时器开始计时),以提供茶液。
-将茶包从茶液中取出,让其滴落5秒钟,然后移除。
-在茶液中加入6.5g糖,然后用于如下所述的分析。
实施例2:
-在金属壶中放入11g来自KSA的立顿黄牌,加入580ml沸水,拟准备约4份饮料。
-在壶上盖上盖子,将壶放在炉子上,其温度足以使其温和地沸腾。
-在不搅拌情况下,将茶煮沸持续3分钟(加入茶后水开始沸腾时开始计时),以提供茶液。
-将茶液用筛子过滤,提供4x 130ml份量的茶液。
-在一份茶液中加入6g糖,然后用于如下所述的分析。
测试组合:
测试样品是通过将铁源和/或潜在颜色校正剂与干茶一起按表1和2所示的量加入按照上述的茶规程来制备的。
在使用潜在颜色校正剂的情况下,其添加量使得提供潜在颜色校正剂:铁(来自铁源)的摩尔比约为3:1至约2:1,如表中所示。
比色分析
将表1和2中所示的每种茶样品的20ml等分试样转移至石英比色皿中,并如下进行CIE L*a*b*分析。
使用Hunterlab Ultrascan-VIS分光光度计(波长:360-780nm)测量样品的颜色。这一工作测量透射颜色。
传感器使用直径为6英寸(152.4mm)并涂覆有SpectraflectTM的塑料积分球,以漫射从灯中发出的光线。光照亮并透射穿过样品。透镜放置于距垂直于样品表面8°角。透镜收集透射光并将其引导至衍射光栅,以将光分成其组成波长,由双二极管阵列测量并转换成数据。
位于传感器中间的传输室用于测量液体的透射颜色。在进行标准化和测量时,关闭传输室门。传输单元支架容纳10mm传输单元中的等分试样。安装时,将传输单元支架放入传输室的中心,即传输室的最宽部分。
传输室提供光学透明的玻璃比色槽,其固定路径长度为10mm。其尺寸为55mm x57mm(宽x高)。用于测量的最小样品体积为20mL。在全传输模式测量(TTRAN)下进行测量。将池放置在分光光度计内部的传输室前部的球体开口处。
测量
分光光度计由EasyMatch QC软件控制,该软件对可见光谱的透射值进行整合,得出三色刺激的X、Y和Z值。这些值模拟1931年2°标准观察者或1964年CIE 10°标准观察者(CIE XYZ)所定义的人类观察者的颜色匹配反应函数。

计算ΔE*和ΔL*
利用通过Hunterlab软件计算得到的L*a*b*值计算ΔE*和ΔL*的值。
计算Δa*和Δb*:
Δa*=a* 样品-a* 空白
Δb*=b* 样品-b* 空白
计算ΔL*:
ΔL*=L* 样品-L* 空白
ΔL*表示亮度和暗度的差异(+ΔL*表示样品比“空白”样品更亮,-ΔL*表示样品比“空白”样品更暗)
计算ΔE*:
L*(ΔL*)、a*(Δa*)和b*(Δb*)的Δ表示样品和“空白”样品在L*、a*和b*中彼此之间的差异大小。ΔL*、Δa*和Δb*可以为正(+)或为负(-)。但是总色差(Delta E*(ΔE*))始终为正。
比色分析结果
比色分析结果提供在表1和2的最后列。
-ΔE*显示相对于各自“空白”样品的在颜色上的变化。
-ΔL*显示与“空白”样品相比,茶是否显示变暗(负数)或变亮(正数)。
表1表明,单独的甘氨酸亚铁(A.Ex1)会导致明显的颜色变化(26.7)和变暗(-27.2)。此外,这种颜色变化和变暗并没有被二盐基NaPP、抗坏血酸和柠檬酸校正(B.Ex1、C.Ex1、D.Ex1)。然而,可以看到,当使用Na2H2EDTA时(1.Ex1),其作为极好的颜色校正剂,显著减少颜色变化(3.5)和变暗(2.2),与没有铁源的茶(空白)几乎相同。
表2进一步表明,Na2H2EDTA对以更苛刻的冲泡过程制备的不同茶产品同样有效。可以看出,单独的甘氨酸亚铁(A.Ex2)会引起显著的颜色变化(56.1)和变暗(-29.4)。此外,这种颜色变化和变暗并没有被二盐基NaPP、抗坏血酸和柠檬酸校正(B.Ex2、C.Ex2、D.Ex2)。然而,可以看到,当使用Na2H2EDTA时(1.Ex2,2.Ex2),其作为极好的颜色校正剂,显著减少颜色变化(6.2,2.4)和变暗(2.7,-0.3),与没有铁源的茶(空白)几乎相同。
因此,本发明人确定甘氨酸亚铁和Na2H2EDTA的组合提供不会发生颜色变化或变暗的铁强化的茶基饮料的独特能力。值得注意的是,使用Na2H2EDTA能够防止甘氨酸亚铁引起的变色或变暗,即使在泡茶时将茶加热至实际沸腾。
Claims (9)
1.产品,其包含:
a)茶组分,
b)甘氨酸亚铁;和
c)Na2H2EDTA。
2.权利要求1所述的产品,其中所述产品为饮料前体。
3.权利要求1或权利要求2所述的产品,其中所述茶组分源自植物茶树(Camelliasinensis)。
4.权利要求1-3中任一项所述的产品,其中所述茶组分为红茶。
5.权利要求1-4中任一项所述的产品,其中所述茶组分为叶茶。
6.权利要求1-5中任一项所述的产品,其中所述产品包含按干重计95-99.9wt%的所述茶组分。
7.权利要求1-6中任一项所述的产品,其中所述产品包含按干重计0.03-3wt%的甘氨酸亚铁。
8.权利要求1-7中任一项所述的产品,其中所述产品包含按干重计0.07-7wt%的Na2H2EDTA。
9.权利要求1-8中任一项所述的产品,其中所述产品包含Na2H2EDTA:来自甘氨酸亚铁的Fe的摩尔比为0.5:1至4:1。
