In some embodiments, the humectant is selected from glycerin, butylene glycol, 1, 3-propanediol, 1, 2-pentanediol, octaethylene glycol, or a combination thereof. The humectant can further improve the moisture retention of deep skin and realize long-acting moisture retention.
In some embodiments, the preservative is selected from phenoxyethanol, p-hydroxyacetophenone, 1, 2-pentanediol, 1, 2-hexanediol, caprylyl glycol, ethylhexylglycerin, benzoic acid, sodium benzoate, caprylhydroxamic acid, or combinations thereof. The preservative can be well compatible with other substances, and the shelf life of the anti-inflammatory and anti-aging essence can be prolonged and the shelf life of the essence can be prolonged by adding the preservative.
In some embodiments, the thickening agent is selected from carbomers, acrylic polymers crosslinked with C10-C30 alkanol acrylate, xanthan gum, or combinations thereof.
According to the embodiment of the application, the thickening agent has important effects on thickening, stability and rheological property of the anti-inflammatory and anti-aging essence lotion; by using the thickener in the above weight ratio, it is possible to give it a good consistency, as well as excellent spreadability and spread.
In some embodiments, the pH adjusting agent is selected from arginine, tromethamine, sodium hydroxide aqueous emulsion, or combinations thereof; and 5% of sodium hydroxide water emulsion can be selected. The pH regulator can regulate pH of the anti-inflammatory and anti-aging essence lotion, so that the essence lotion can better act on skin and avoid irritation. In addition, tromethamine acts on skin and is beneficial to healing and repairing damaged tissues.
In some embodiments, the vegetable fat is selected from jojoba oil, sunflower seed oil, castor oil, coconut oil, grape seed oil, shea butter, meadowfoam seed oil, macadamia nut oil, olive oil, palm oil, squalane, cocoa butter, jojoba fat, glycolipids, octyldodecanol, caprylic capric triglyceride, coco-caprylic capric triglyceride, oleic acid, linoleic acid, linolenic acid polyglycerides, and dioctyldodecanol dilinoleate, or combinations thereof.
According to the embodiment of the present application, the vegetable oil or fat has excellent safety, moisturizing property, fluidity and ductility, and ensures the moisturizing effect. When skin cracks, the grape seed oil, the coconut oil and the sunflower seed oil also have an antioxidant effect, can accelerate the skin crack repairing effect, and can realize wrinkle resistance and inflammation resistance from the moistening and repairing directions. The content of vegetable oil and fat in the anti-inflammatory and antiaging essence lotion is 0.1%, 0.15%, 0.2%, 0.21%, 0.22%, 0.23%, 0.25%, 0.28%, 0.3%, 0.4%, 0.5%, 0.6%, 0.7%, 0.8%, 0.9%, 1.0% by weight
In some embodiments, the emulsifier is selected from oleyl polyether-3 phosphate, oleyl polyether-3, oleyl polyether-5, or a combination thereof. The emulsifier can provide excellent emulsifying effect, can be quickly emulsified when meeting water, is easy to wash, can be dispersed in water to form an oil-in-water emulsion with strong fluidity, is fresh and comfortable after makeup removal, and has no greasy feeling.
In some embodiments, the metal chelating agent is selected from disodium ethylenediaminetetraacetate, EDTA, hydroxyethane diphosphonic acid, or combinations thereof. The metal chelating agent can remove trace impurities in each component and provide the action effect, and EDTA is ethylenediamine tetraacetic acid.
In some embodiments, the water is selected from deionized water, a yeast secondary fermentation filtrate, or a combination thereof. The deionized water can avoid introducing impurities; the secondary split yeast fermentation filtrate contains micromolecular polypeptides such as amino acid organic acid cytoplasm and the like, and is rich in vitamin B group; can resist and repair the barrier function of the damaged skin to strengthen the skin from the source, achieves the effect of brightening, and can strengthen the metabolism of the cuticle, arouse the elasticity and the vitality of the skin and enhance the anti-aging effect.
Preparation method of anti-inflammatory and anti-aging essence
In a third aspect, the present application provides a method for preparing the anti-inflammatory and anti-aging essence of the second aspect, comprising:
preparing an aqueous phase solution: mixing a humectant, a preservative, a thickener, and a metal chelator;
mixing at least one of aqueous phase solution, lactobacillus fermentation lysate, yeast fermentation lysate, hyaluronic acid, hyaluronate, hyaluronic acid derivative, tocopherol acetate, vegetable oil and fat, and emulsifier, homogenizing, and emulsifying to obtain antiinflammatory and antiaging essence lotion.
In some embodiments, preparing the aqueous phase solution comprises: the humectant, the preservative, the thickener, and the metal chelator are mixed under heating to mix the solutions uniformly.
In some embodiments, the humectant, preservative, thickener, and metal chelator are mixed under heating to 50-80 ℃ to mix the solutions evenly.
In one embodiment, the anti-inflammatory and anti-aging essence is obtained by mixing at least one of an aqueous phase solution, a lactobacillus fermentation lysate, a yeast dehiscence lysate, hyaluronic acid, a hyaluronate salt, a hyaluronic acid derivative, tocopherol acetate, vegetable oil and an emulsifier, homogenizing and emulsifying, and adding a pH regulator.
Anti-inflammatory and anti-aging cosmetic or pharmaceutical composition
In a fourth aspect, the present application provides an anti-inflammatory anti-ageing cosmetic or pharmaceutical composition comprising: an anti-inflammatory and anti-ageing composition according to the first aspect and a cosmetically acceptable adjuvant.
The cosmetic or pharmaceutical composition of the present application may be added with various desired cosmetic or pharmaceutically acceptable excipients according to various formulation forms of the composition. For example, when the composition of the present application is prepared as a liquid, the excipient may be selected from water, physiological saline, and the like; when the composition of the present invention is prepared into a gel, the excipient may be selected from cellulose derivatives, starch, gelatin, agar, polysaccharides, and the like; when the composition of the present application is prepared as a cream, the excipient may be selected from glycerin, vaseline, paraffin, etc.; when the composition of the present application is prepared as a foam, the excipient may be selected from hydroxypropyl methylcellulose, sodium lauryl sulfate polyethylene glycol, sodium fatty alcohol-polyoxyethylene ether sulfonate, and the like; when the compositions of the present application are prepared as a patch, the excipient may be selected from cellulose derivatives, starch, gelatin, agar, polysaccharides, and the like.
According to the embodiment of the application, the cosmetic or pharmaceutical composition is suitable for cleaning, washing and protecting skin products such as aqua products, emulsified products, oil products and the like, and can also be used in beauty products such as essences, facial masks and the like, such as ointments, foundation make-up and the like.

The present disclosure is more particularly described in the following examples that are intended as illustrations only, since various modifications and changes within the scope of the present disclosure will be apparent to those skilled in the art. Unless otherwise indicated, all parts, percentages, and ratios reported in the following examples are on a weight basis, and all reagents used in the examples are commercially available or synthesized according to conventional methods and can be used directly without further treatment, and the equipment used in the examples is commercially available.
Examples 1 to 4
The anti-inflammatory and anti-aging essence disclosed by the embodiment of the application comprises the following components in percentage by mass:
lactobacillus fermentation lysate, 1% -5%;
(ii) a secondary split yeast fermentation product lysate, 5% -15%;
1% -3% of at least one of hyaluronic acid, hyaluronate and hyaluronic acid derivative;
0.05 to 0.3 percent of tocopherol acetate;
0.1% -0.3% of bisabolol;
0.5 to 8 percent of humectant;
0.5 to 8 percent of preservative;
0.1 to 8 percent of thickening agent;
0.1 to 3 percent of pH regulator
0.1% -5% of metal chelating agent;

0.2 to 8 percent of emulsifier;
93.3 to 99.85 percent of water.
The contents of the components of the examples are shown in Table 1.
The preparation method of the anti-inflammatory and anti-aging essence comprises the following steps:
preparing an aqueous phase solution: mixing a humectant, a preservative, a thickener, and a metal chelator;
mixing at least one of aqueous phase solution, lactobacillus fermentation lysate, schizosaccharomyces fermentation lysate, hyaluronic acid, hyaluronate, hyaluronic acid derivative, tocopherol acetate, vegetable oil and fat, and emulsifier, homogenizing and emulsifying, and adding pH regulator to obtain the anti-inflammatory and anti-aging essence.
Preparing a hyaluronic acid mixed solution: mixing sodium hyaluronate, a sodium hyaluronate cross-linked polymer (weight average molecular weight of 1000000), hydrolyzed hyaluronic acid and sodium acetylated hyaluronate according to a weight ratio of 1. The sodium hyaluronate has a number average molecular weight of 5000-10000, and also has a number average molecular weight of 800000-100000, and a mass ratio of 1:1; the hyaluronic acid comprises a number average molecular weight of 5000-10000, and further comprises a hyaluronic acid with a number average molecular weight of 800000-100000, in a mass ratio of 1:1; the weight average molecular weight of the acetylated hyaluronic acid is 1000000; all raw materials are from Furuida.
The lactobacillus fermentation lysate was from korean SK Bioland, the schizosaccharomyces fermentation lysate was from korean SK Bioland, the bisabolol was from BASF, and the tocopherol acetate was from BASF.
The other materials may be from any commercially available cosmetic raw materials company.

Test section
1) And (3) safety testing:
the experimental steps are as follows: 30 volunteers, 6 of each volunteer, were divided into two groups, and the anti-inflammatory and anti-aging essence lotion samples of examples 1-5 were tested, each tester performed a spot-patch control experiment on the inner sides of the left and right arms, and the inner side of the left hand was coated with 1.0g of the product of the present invention each time, and the inner side of the right hand was coated with 1.0g of blank base material twice a day, and the skin reaction phenomena after 24h, 48h, and 72h were recorded.
The experimental results are as follows: after the anti-inflammatory and anti-aging essence lotion samples of examples 1 to 4 were coated for 24 hours, 48 hours and 72 hours, no allergic phenomena such as skin redness or erythema appeared, which indicates that the anti-inflammatory and anti-aging essence lotions of the anti-inflammatory and anti-aging essence lotion samples of examples 1 to 4 are safe and have no adverse reaction phenomenon.
2) Stability test
The anti-wrinkle compositions of examples 1-5 were subjected to stability tests, including heat, cold, alternating and light tests. Wherein, the heat resistance test conditions are as follows: keeping the temperature at 45 +/-1 ℃ and keeping out of the sun; the cold resistance test conditions are as follows: keeping the temperature at minus 20 +/-1 ℃ and keeping out of the sun; the alternating test conditions were: keeping the temperature at minus 20 ℃ for 3.5 days, changing the temperature at 45 ℃ for 3.5 days, and keeping out of the sun; the illumination test conditions were: the test chamber (1.47X 106 Lux. Hr) was illuminated, and the test results are shown in Table 3.

As can be seen from table 3, the anti-inflammatory and anti-aging essence disclosed in the embodiments of the present application has excellent stability, compatibility and stability.
3) Anti-wrinkle test
1. Dividing volunteers into 8 groups at random, wherein each group has 30 individuals, the age is 35-45 years, and the position and area of each group of test area are the same;
2. each group of volunteers continuously used the samples of examples 1-4 and the samples of practical examples 1-5 for 4 weeks, respectively, the using time and the using amount of each group are consistent, and other functional cosmetics are not used during the testing period;
3. volunteers self-evaluate the aging improvement and skin firmness. The wrinkles are reduced to 5 points full with naked eyes and are not changed to 0 point; the skin firmness is improved better, and the skin can be seen to be firmer to a full score of 5 points without changing to 0 point. As shown in table 4.

In addition, the skin of the forehead and the wrinkle part of the face of the volunteer is coated by using the examples and comparative examples of the application, and the anti-aging effect is detected by adopting an Antera 3D skin tester to test the roughness and the wrinkle roughness. In agreement with the scores of the above-mentioned table.
The skin using examples and comparative examples was subjected to a non-invasive imaging operation using an optical coherence tomography skin scanner to obtain pictures 1-2.
The non-invasive imaging of collagen of example 1, as shown in fig. 1, is the initial state of the skin; fig. 2 shows the state of the anti-inflammatory and anti-aging essence of the present application after 28 days. As can be seen from the figure, example 1 has the effect of promoting collagen production, and can achieve anti-aging.
4) Effect on photoaging 3D full-thickness skin model
The effects of the anti-inflammatory and anti-aging essence lotions of the examples and comparative examples on the photoaging 3D full-thickness skin model were tested.
An experimental instrument: an ultra-clean bench, a carbon dioxide incubator, UVA and UVB irradiators, a microplate reader (BIO-TEK), and a cryomicrotome (Leica).
Experiment reagents and consumables: 3D full-thickness skin model (self-made in laboratory), ELISA detection kit with different indexes, MTT kit and the like.
The experimental procedure was as follows:
(1) 3D full-layer model construction: constructing a 3D skin model by using keratinocytes and fibroblasts;
(2) Molding and dosing: SSUV irradiation treatment (UVA: 30J/cm2; UVB:50 mJ/cm) was performed every day when the model was shipped, i.e., 0 day 2 ) Then respectively coating a certain amount of samples on the surfaces of corresponding models, coating model culture solution only by contrast of the models, coating 6 composite holes in each composition once a day, and coating the composite holes once a day for a total action time of 4 days;
(3) Collecting and detecting: after the action of the sample is finished, collecting culture solution, taking 3 models of Shanghai enzyme-linked biology company in each group, carrying out the determination of model activity by using an MTT kit, carrying out tissue fixation on the remaining 3 models, and carrying out IHC (IHC staining) to determine the content of collagen at the true epidermal junction;
(4) And (3) analysis: and (5) performing statistical analysis by using a Test method.
The test results are shown in the following table.

The above results indicate that the samples of the examples improve the tissue viability of the 3D full-thickness skin model, promote the expression of collagen at the junction of the true epidermis, and improve the tissue viability, indicating that at least one of the lactobacillus fermentation lysate, the yeast fermentation lysate, the hyaluronic acid, the hyaluronate, the hyaluronic acid derivative, the tocopherol acetate, and the bisabolol produce an obvious synergistic effect.
5) Influence on human skin morphology
In this example, the volunteers were tested for wrinkle change in the right and left canthus, cheek moisture and TEWL change after 2 weeks and 4 weeks using the sunny slope of examples and comparative examples to evaluate the anti-inflammatory and anti-aging essence for its efficacy of anti-wrinkle, increasing skin moisture content and improving skin barrier function, with subjects on the wrinkle grade of 1-5 as the subjects.
The following tests were performed on volunteers before, 2 weeks and 4 weeks after using the product, respectively, using the half-face control test method:
1) Taking pictures of external canthus on left and right sides of a volunteer by using primos, and calculating parameters including wrinkle number, wrinkle area, wrinkle depth and the like by using software;
2) Testing the moisture content of the skin of the cheeks on the left and right sides by using a Corneometer;
3) The left and right cheeks were tested for their rate of percutaneous water loss (TEWL values) using a Tewameter.
After face cleaning in the morning and evening, a proper amount of test sample is taken after the lotion is used and is respectively and uniformly applied to the left face and the right face (including the periphery of eyes), the original skin care habit is kept unchanged in the whole test process, and the test sample is only used for replacing the face cream used by each subject.

The test procedure was as follows:
1) The tested areas were lateral external canthus, forehead and cheek. Before the test, the testee uniformly cleans the face with facial cleanser, and then wipes the face clean with a non-scrap absorbent paper towel;
2) Sitting still for 20 minutes in a room meeting the standard, water and beverages can not be drunk, the tested area is exposed and kept relaxed, and the tested area is prevented from being touched;
3) The left and right canthus are randomly divided into a product smearing area and a blank control area to ensure that the balance is achieved statistically;
4) During testing, the same facial fixing facility and fixing mode are adopted, and according to primos use instructions, pictures of wrinkles of left and right external canthus of a subject before use, after 2 weeks and 4 weeks of use are collected, and a Corneometer and a Tewameter are used for testing the moisture value of the cheek and the TEWL value of the volunteer on two sides respectively.
Number of subjects: the group was 30, with a mean age of 45.5 ± 8.5 years. The test results were averaged as follows.

The number of wrinkles is used to characterize the number of canthus wrinkles, with higher numbers indicating more wrinkles and fewer wrinkles. The number of skin wrinkles gradually decreased after the use of the example group, and the decrease in the number of wrinkles was very significant by 4 weeks (p < 0.01). The comparative example group had no significant difference in wrinkle area after 4 weeks of use compared to before use (P > 0.05).
Wrinkle depth is used to characterize the depth of the canthus wrinkles, with higher values indicating deeper wrinkles and conversely lighter wrinkles. The skin wrinkle depth gradually decreased after the use of the samples of the example group, and the wrinkle depth decrease was very significant up to 4 weeks (p < 0.01). The control group had no significant difference in wrinkle depth after 4 weeks of use compared to before use (P > 0.05).
The wrinkle area index is used to characterize the total area of canthus wrinkles, with higher values indicating more wrinkles and fewer wrinkles. The wrinkle area of the skin gradually decreased after the use of the example group, and the wrinkle area decreased very significantly by 4 weeks (p < 0.01). The wrinkle area of the sample of the comparative example group was not significantly different after 4 weeks of use compared to before use (P > 0.05). Compared with the comparative example group, the difference of the example group after 4 weeks of use is significant (p < 0.01), which indicates that the anti-inflammatory and anti-aging essence of the example has significant efficacy of reducing skin wrinkles.

And the water content variation rate is used for representing the water content of the stratum corneum of the skin, the water content of the blank control group is subtracted from the measured water content, and the obtained difference is divided by the water content of the blank control group to obtain the water content variation rate. If the moisture is reduced, it is negative, and the moisture is increased, it is positive. The higher the value the higher the skin moisture content, whereas the lower the skin moisture content. The samples of the example group showed a gradual increase in skin moisture content after application, with a very significant increase by 4 weeks (p < 0.01). The comparative example group samples showed no significant difference in skin moisture content after 4 weeks of use compared to the pre-use (P > 0.05). After 4 weeks of use, the samples of the example group were very significantly different (p < 0.01) from the samples of the comparative example group, indicating that the example group had the efficacy of increasing the moisture content of the skin.
TEWL is used to characterize the barrier function of the skin cutin, the higher the number the poorer the barrier function of the skin and vice versa. The TEWL values gradually decreased after use for the samples of the example group, with a significant decrease by 4 weeks (p < 0.05). The TEWL value after 4 weeks of use of the control group was not significantly different from that before use (P > 0.05). After 4 weeks of use, the difference between the example group and the comparative example group was significant (p < 0.05), indicating that the example group had efficacy in repairing skin barrier function.
While the invention has been described with reference to specific embodiments, the scope of the invention is not limited thereto, and those skilled in the art can easily conceive various equivalent modifications or substitutions within the technical scope of the invention. Therefore, the protection scope of the present application shall be subject to the protection scope of the claims.

Claims (10)

1. An anti-inflammatory and anti-aging composition comprises the following components in parts by weight:
lactobacillus fermentation lysate, 10 parts;
10-150 parts of a schizosaccharomyces cerevisiae fermentation product lysate;
2-30 parts of at least one of hyaluronic acid, hyaluronate and hyaluronic acid derivatives;
0.1-3 parts of tocopherol acetate;
0.2-3 parts of bisabolol.
2. The anti-inflammatory and anti-aging composition according to claim 1, which comprises the following components in parts by weight:
lactobacillus fermentation lysate, 10 parts;
20-100 parts of a schizosaccharomyces cerevisiae fermentation product lysate;
5-20 parts of at least one of hyaluronic acid, hyaluronate and hyaluronic acid derivatives;
1-2 parts of tocopherol acetate;
1-2 parts of bisabolol.
3. The anti-inflammatory and anti-aging composition according to claim 1 or 2, which comprises the following components in parts by weight:
lactobacillus fermentation lysate, 10 parts;
split yeast fermentation lysate, 20-50;
5-10 parts of at least one of hyaluronic acid, hyaluronate and hyaluronic acid derivatives;
1-1.5 parts of tocopherol acetate;
1-1.5 parts of bisabolol.
4. The anti-inflammatory and anti-aging essence comprises the following components in percentage by mass:
lactobacillus fermentation lysate, 1% -5%;
(ii) a secondary split yeast fermentation product lysate, 5% -15%;
1% -3% of at least one of hyaluronic acid, hyaluronate and hyaluronic acid derivative;
0.05 to 0.3 percent of tocopherol acetate;
0.1% -0.3% of bisabolol;
0-8% of humectant;
0-8% of preservative;
0.1 to 8 percent of thickening agent;

5. The anti-inflammatory and anti-aging essence lotion as claimed in claim 4, which comprises the following components in percentage by mass:

the preservative is selected from phenoxyethanol, p-hydroxyacetophenone, 1, 2-pentanediol, 1, 2-hexanediol, caprylyl glycol, ethylhexyl glycerin, benzoic acid, sodium benzoate, caprylyl hydroxamic acid, or combinations thereof; and/or the presence of a gas in the atmosphere,
the thickening agent is selected from carbomer, acrylic acid cross-linked polymer with C10-C30 alkanol acrylate, xanthan gum or combination thereof; and/or the presence of a gas in the gas,
the pH regulator is selected from arginine, tromethamine, sodium hydroxide water emulsion or the combination thereof; the water-soluble polymer can be selected as a sodium hydroxide water emulsion with the mass fraction of 5%; and/or the presence of a gas in the gas,
the metal chelating agent is selected from disodium ethylenediaminetetraacetate, EDTA, hydroxyethane diphosphonic acid or a combination thereof.

The anti-inflammatory and anti-aging essence according to claim 4 or 5, wherein the vegetable oil and fat is selected from jojoba oil, sunflower seed oil, castor oil, coconut oil, grape seed oil, shea butter, meadowfoam seed oil, macadamia nut oil, olive oil, palm oil, squalane, cocoa butter, jojoba fat, glycolipids, octyldodecanol, caprylic/capric triglyceride, coco-caprylic/capric triglyceride, oleic acid, linoleic acid, linolenic acid polyglycerol esters and dioctyl dodecanol dimer linoleate or a combination thereof; and/or the presence of a gas in the gas,
the emulsifier is selected from oleyl polyether-3 phosphate, oleyl polyether-3, oleyl polyether-5 or a combination thereof; and/or the presence of a gas in the gas,
the water is selected from deionized water, yeast bifidus fermentation filtrate or combination thereof.
8. A method for preparing the anti-inflammatory and anti-aging essence lotion as claimed in any one of claims 4 to 7, comprising:
preparing an aqueous phase solution: mixing a humectant, a preservative, a thickener, and a metal chelator;
mixing at least one of aqueous phase solution, lactobacillus fermentation lysate, schizosaccharomyces fermentation lysate, hyaluronic acid, hyaluronate, hyaluronic acid derivative, tocopherol acetate, vegetable fat and oil, and emulsifier, homogenizing and emulsifying to obtain the anti-inflammatory and anti-aging essence.
9. The method of claim 8, wherein preparing the aqueous solution comprises: the humectant, the preservative, the thickener, and the metal chelator are mixed under heating to mix the solutions uniformly.
10. An anti-inflammatory anti-aging cosmetic or pharmaceutical composition comprising: an anti-inflammatory and anti-ageing composition as claimed in any one of claims 1 to 3, together with a cosmetically acceptable adjuvant.
