Abstract

背景技术
焦亡是一种细胞炎症程序性死亡,它是由Gasdermin蛋白介导产生的。适量的焦亡有利于个体的发展,例如我们在肿瘤细胞中诱导表达Gasdermin蛋白来促进肿瘤细胞焦亡,在败血症细胞中抑制焦亡从而减少炎症因子的释放等。Gasdermin蛋白是一种结构自抑蛋白,在受到特定刺激后,Gasdermin蛋白解除自抑结构,其N端会与质膜结合,在膜上寡聚打孔,引起质膜破损,细胞内容物外泄,胞内外渗透压改变,最后导致细胞溶胀死亡。在哺乳动物细胞中具有6个Gasdermin家族蛋白:Gasdermin A-E,PJVK。研究表明,Gasdermin蛋白与许多疾病有关,例如Gasdermin A与哮喘有关,Gasdermin B的高表达与胃癌、肝癌、结肠癌和宫颈癌中肿瘤的侵袭性有关等,因此Gasdermin蛋白是一个很好的药物作用靶点。但是在哺乳动物细胞中存在6种Gasdermin蛋白,很难对单一Gasdermin蛋白进行功能研究,因此建立一个单一蛋白表达的生物模型对于了解疾病的病理机制和药物筛选具有重要的意义。
目前研究Gasdermin蛋白功能主要在哺乳动物细胞系研究体系和蛋白体外研究体系中进行。使用哺乳动物细胞系的研究方法主要包括:在细胞中将目的蛋白敲低或者敲除,以消除内源性蛋白的影响;也可以通过瞬转或稳转的方式在哺乳动物细胞中引入含有目标蛋白的质粒,使目标蛋白在细胞内过表达。综合以上敲低、敲除或过表达蛋白对细胞性状的影响,可推断其蛋白功能。然而哺乳细胞中存在多个Gasdermin同源蛋蛋白,不管是敲除、敲低、或者过表达目标蛋白,都很难消除其他Gasdermin蛋白的影响。为消除其他同源蛋白的影响,可以采用蛋白体外纯化提取单一蛋白,即以外源的mRNA或DNA为蛋白质的合成模板,通过人为控制添加蛋白质合成所需的底物、能量、以及转录和翻译相关蛋白因子等物质,实现外源蛋白质的合成。但这种方法不仅需要体外表达纯化目的蛋白,还要依赖其他实验,比如体外脂质体泄露实验以模拟蛋白在膜上打孔的表型,需要纯化的蛋白量较大,并且技术难度也较大,实验周期也较长。
但是,由于哺乳动物细胞基因的敲除和敲低的技术难度大、周期长,结果假阳性高,很难进行基因的大通量筛选,对于焦亡的机制研究较困难;对于Gasdermin蛋白的过表达,由于哺乳动物细胞转染质粒的效率较低,筛选较复杂,成功率也较低;而体外合成蛋白研究,很难模拟在生理条件下该蛋白的作用和功能;在哺乳动物细胞中,存在多条程序性死亡通路,包括程序性凋亡、程序性焦亡、程序性坏死,多种通路间存在交叉,会互相干扰;并且在哺乳动物细胞中存在6个Gasdermin蛋白的同源蛋白,当试图研究单一蛋白作用时,在特定的研究条件下存在多个Gasdermin蛋白的表达,很难确定单一蛋白的功能和作用。

为解决上述技术问题,本发明借助分子克隆技术,构建了一个可研究单一GSDM蛋白的细胞体系,将GASDERMIN基因克隆到pRS416-MET25质粒中,得到一个新的prMET25-GASDERMIN质粒。已知在酿酒酵母细胞(Saccharomyces cerevisiae)内,不存在Gasdermin蛋白的同源蛋白,因此可以将prMET25-GASDERMIN质粒转入到酿酒酵母中,当培养基去掉甲硫氨酸时,可以在生理条件下诱导单一Gasdermin全长蛋白和N端蛋白的表达。其中N端蛋白也能够识别酵母的质膜,在酵母质膜上打孔导致酵母的死亡,与哺乳细胞中的表型相似,并且可以通过调节启动子诱导水平来调节蛋白量的表达,从而控制焦亡表型的程度。并且由于可单独表达N端蛋白诱导焦亡,跳过了上游的信号从而避免其他程序性死亡通路的影响。通过细胞生长实验可以方便直接地观察到细胞的生长状况、当细胞发生焦亡时,质膜破损,可以利用PI(Propidium Iodide,一种不透膜的染料)染色测定荧光强度可以实时测定膜破损情况以及通过Western blot技术检测Gasdermin的表达量,样品制备简单,实验结果易于观察和检测,尤其利于进行高通量药物筛选。并且酵母基因与人类基因具有至少60%的同源性,目前酵母已经拥有成熟且全面的基因缺失库,使得本发明对焦亡的机制研究更为方便。
本发明的第一个目的是提供一种用于研究人类Gasdermin蛋白功能的细胞模型,所述的细胞模型是以酵母菌为宿主,表达单一的Gasdermin全长蛋白或Gasdermin N端蛋白,并以MET25启动子启动蛋白表达。
进一步地,所述的酵母菌为酿酒酵母。本发明使用的细胞为酿酒酵母Saccharomyces cerevisiae,这种细胞中并不存在人类Gasdermin蛋白的同源蛋白,因此可以分别表达单一的Gasdermin全长蛋白和N端蛋白。
进一步地,所述的Gasdermin蛋白为Gasdermin A、Gasdermin B、Gasdermin C、Gasdermin D或Gasdermin E。

将编码Gasdermin蛋白或Gasdermin N端蛋白的基因构建至pRS416-MET25质粒上,得到重组表达载体;将重组表达载体导入至酵母细胞中,得到所述的细胞模型。
本发明的第三个目的是提供所述的细胞模型在Gasdermin蛋白功能研究中的应用。
本发明的第四个目的是提供所述的细胞模型在筛选以Gasdermin蛋白为靶点的药物中的应用。
本发明的有益效果是:
本发明借助分子克隆技术,构建了一个可研究单一GSDM蛋白的细胞体系,将GASDERMIN基因克隆到pRS416-MET25质粒中,得到一个新的prMET25-GASDERMIN质粒。已知在酿酒酵母细胞(Saccharomyces cerevisiae)内,不存在Gasdermin蛋白的同源蛋白,因此将prMET25-GASDERMIN质粒转入到酿酒酵母中,当培养基去掉甲硫氨酸时,可以在生理条件下诱导单一Gasdermin全长蛋白和N端蛋白的表达,其中N端蛋白也能够识别酵母的质膜,在酵母质膜上打孔导致酵母的死亡,与哺乳细胞中的表型相似,并且可以通过调节启动子诱导水平来调节蛋白量的表达,从而控制焦亡表型的程度。并且由于可单独表达N端蛋白诱导焦亡,跳过了上游的信号从而避免其他程序性死亡通路的影响。通过菌落形成(colonyforming)实验可以方便直接的观察到细胞的生长状况;当细胞发生焦亡时,质膜破损,可以利用PI(Propidium Iodide,一种不透膜的染料)染色测定荧光强度从而实时监测膜破损情况;以及通过Western blot技术检测Gasdermin的表达量。样品制备简单,实验结果易于观察和检测,尤其利于进行高通量药物筛选。并且酵母基因与人类基因具有至少60%的同源性,目前酵母已经拥有成熟且全面的基因缺失库,使得本发明对焦亡的机制研究更为方便。

下面结合具体实施例对本发明作进一步说明,以使本领域的技术人员可以更好地理解本发明并能予以实施,但所举实施例不作为对本发明的限定。
下述实施例中所用的方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1:酵母体系的建立
1.蛋白表达启动子的选择
酵母内存在多种可诱导表达的启动子,本体系已经尝试PGK1启动子(持续高表达启动子)、GAL1启动子(培养基中添加半乳糖可诱导蛋白表达)、MET25启动子(培养基中去掉甲硫氨酸诱导蛋白表达)。
(1)采用MET25启动子构建的菌株进行生长实验:将分别转染了空载质粒(pRS416-MET25)、全长质粒(prMET25-GSDMA、prMET25-GSDMB、prMET25-GSDMC、prMET25-GSDMD、prMET25-GSDME)和N端质粒(prMET25-GSDMAN、prMET25-GSDMBN、prMET25-GSDMCN、prMET25-GSDMDN、prMET25-GSDMEN)的BY4741菌株。过夜培养后取6OD600,于6mL C-Ura培养基孵育孵育1h后,取1OD600菌液作为对照,再取4OD600于4mL C-Ura-Met培养基中诱导目的蛋白表达,分别于1h、3h、6h、9h、22h取1OD600 1300rpm离心后,弃上清,沉淀加入100μL灭菌水后重悬,五倍梯度稀释点在CSH-URA固体培养基上,观察其生长状况。结果如图1所示。
(2)以上述(1)中构建的菌株进行PI荧光强度实验:将转染质粒的菌株过夜培养后取6OD600,于6mL C-Ura培养基孵育孵育1h后,再取4OD600于C-Ura-Met培养基中诱导目的蛋白表达,分别于1h、3h、6h、9h、22h取1OD600,用灭菌PBS定容至1mL,取200μL置于底部透明四周黑色的平底96孔板中,加入2μL 20mg/mL的PI溶液,混合均匀后于室温孵育10min后,用酶标仪于535nm/617nm处测定荧光强度。(定量数据均来自3次独立实验,均以WT+Prs416-MET25作为对照,使用双侧检验法将诱导表达目标蛋白的菌株与空载质粒菌株比较,***P<0.005、0.005≤**P<0.01、0.01≤*P<0.05表示有统计学意义,P>0.05表示无差异ns,误差线为标准差。),结果如图2所示。
(3)采用GAL1启动子构建的菌株进行生长实验:将分别转染了空载质粒(pRS416-GAL1)、GSDM全长质粒(prGAL1-GSDMA、prGAL1-GSDMB、prGAL1-GSDMD、prGAL1-GSDME)和N端质粒(prGAL1-GSDMAN、prGAL1-GSDMBN、prGAL1-GSDMDN、prGAL1-GSDMEN)的BY4741菌株。过夜培养后取6OD600,于6mL C-Ura-Glucose培养基孵育1h后,取1OD600菌液作为对照,再取4OD600于4mL C-Ura-Glucose+Galactose培养基中诱导目的蛋白表达,分别于3h、6h、22h取1OD6001300rpm离心后,弃上清,沉淀加入100μL灭菌水后重悬,五倍梯度稀释点在CSH-URA固体培养基上,观察其生长状况。结果如图3所示。
PGK1启动子为持续高表达启动子,使目的蛋白过量表达导致细胞死亡,未能得到单克隆;MET25启动子,除GSDMCN外,其余GSDMN均出现明显的诱导死亡的现象(图1);采用PI染料(一种不透膜的染料)测定荧光强度以实时检测膜打孔情况(图2),在GAL1启动子的诱导条件下,细胞的死亡表型相对于MET25启动子有所减弱(图3)。实验结果表明MET25启动子为最为合适的诱导体系。
2.蛋白标签位置的确定
为检测蛋白的表达情况,我们以MET25启动子体系构建带标签的质粒,然后进行如下实验:
(1)将分别转染了空载质粒(pRS416-MET25)、GSDMB、GSDMD、GSDME全长蛋白N端带有HA-tag的质粒和N端带有HA-tag的GSDMBN、GSDMDN、GSDMEN质粒的BY4741菌株。过夜培养后取6OD600,于6mL C-Ura培养基孵育1h后,取1OD600菌液作为对照,再取4OD600于4mL C-Ura-Met培养基中诱导目的蛋白表达(A)分别于3h、6h、22h取1OD600 1300rpm离心后,弃上清,沉淀加入100μL灭菌水后重悬,五倍梯度稀释点在CSH-URA固体培养基上,观察其生长状况。(B)于相同时间点取2OD菌液以Pgk1为对照,测定蛋白表达情况。结果如图4所示,GSDM蛋白N端HA-tag的细胞焦亡表型受到抑制并不是通过抑制蛋白表达。
(2)将分别转染了空载质粒(pRS416-MET25)、C端带有HA-tag的GSDMA、GSDMB。GSDMD、GSDME全长质粒和C端带有HA-tag的GSDMAN、GSDMBN、GSDMDN、GSDMEN质粒的BY4741菌株。过夜培养后取6OD600,于6mL C-Ura培养基孵育1h后,取1OD600菌液作为对照,再取4OD600于4mL C-Ura-Met培养基中诱导目的蛋白表达,(A)分别于3h、6h、22h取1OD600 1300rpm离心后,弃上清,沉淀加入100μL灭菌水后重悬,五倍梯度稀释点在CSH-URA固体培养基上,观察其生长状况。(B)于相同时间点取2OD菌液以Pgk1为对照,测定蛋白表达情况。结果如图5所示,GSDM蛋白C端HA-tag的焦亡表型并未受到抑制并且蛋白正常表达。
上述结果表明蛋白N端带标签不会影响蛋白的表达但会严重抑制死亡表型(图4),而蛋白C端带标签不会影响蛋白的表达并且对细胞死亡表型抑制较弱(图5),有可能是由于Gasdermin蛋白是由N端序列识别质膜,因此当N端序列前有tag会影响蛋白的膜识别,因此C端tag更合适。
由此我们成功构建了一个酵母体系,可异源表达人源的Gasdermin蛋白,并且该蛋白在酵母中也能识别质膜诱导细胞焦亡。在去掉甲硫氨酸的诱导条件下,表达适量的Gasdermin蛋白N端蛋白,能跳过上游的炎症小体和caspase酶的信号,直接启动焦亡。通过在蛋白C末端加上标签,可在不影响蛋白致死功能的情况下,进行蛋白表达水平的监测。
实施例2:酵母体系应用
1.机制研究
①细胞代谢状态对Gasdermin蛋白的影响
在构建酵母体系时,我们发现在使用GAL1启动子的时候,与MET25启动子相比细胞焦亡表型受到抑制,MET25启动子与GAL1启动子的诱导过程中只有培养基中的糖原不同,由此我们猜测细胞的代谢状态可能会对Gasdermin蛋白产生一定的影响。因此我们在MET25启动子诱导体系下使用了不同浓度的葡萄糖,观察不同浓度葡萄糖是否对Gasdermin蛋白产生影响.生长实验过程如下:将分别转染了空载质粒(pRS416-MET25)、全长质粒(prMET25-GSDMA)和N端质粒(prMET25-GSDMAN)的BY4741菌株过夜培养后取6OD600,于6mL C-Ura-Glucose培养基孵育1h后,取1OD600菌液作为对照,再4份取4OD600分别置于4mL C-Ura-Met(0% Glucose)、4mL C-Ura-Met(1% Glucose)、4mL C-Ura-Met(2%Glucose)、4mL C-Ura-Met(5% Glucose)培养基中诱导目的蛋白表达,分别于3h、6h、22h取1OD600 13000rpm离心后,弃上清,沉淀加入100μL灭菌水后重悬,五倍梯度稀释点在CSH-URA固体培养基上,观察其生长状况。结果如图6所示,显示高浓度葡萄糖促进细胞死亡,低浓度葡萄糖抑制死亡。
同时使用PI染料检测膜打孔情况。PI荧光强度实验:将分别将转染了N端质粒(prMET25-GSDMAN)的BY4741菌株过夜培养后取6OD600于6mL C-Ura-Glucose培养基孵育1h后,取1OD作为对照,再4份取4OD600分别置于4mL C-Ura-Met(0% Glucose)、4mL C-Ura-Met(1% Glucose)、4mL C-Ura-Met(2% Glucose)、4mL C-Ura-Met(5% Glucose)培养基中诱导6h后,分别取1OD600菌液用灭菌PBS定容至1mL,取200μL置于底部透明四周黑色的平底96孔板中,加入2μL 20mg/mL的PI溶液,混合均匀后于室温孵育10min后,用酶标仪于535nm/617nm处测定荧光强度。(以上定量数据均来自3次独立实验,将对应菌株孵育1h平均值当做1标准化处理,使用双侧检验法将诱导表达目标蛋白的菌株与对应菌株孵育1h相比较,***P<0.005、0.005≤**P<0.01、0.01≤*P<0.05表示有统计学意义,P>0.05表示无差异ns,误差线为标准差。),结果如图7所示,高浓度葡萄糖促进GSDMAN在膜上的寡聚和打孔。
我们发现在高浓度葡萄糖的条件下促进焦亡表型,而低浓度抑制焦亡。我们猜测不同浓度的葡萄糖可能诱导改变细胞的活性氧(ROS)水平,因此我们检测了细胞内活性氧水平。并且检测了蛋白表达情况。
细胞内活性氧水平检测方法如下:将分别将转染了N端质粒(prMET25-GSDMAN)的BY4741菌株过夜培养后取6OD600于6mL C-Ura-Glucose培养基孵育1h后,取4份取4OD600分别置于4mL C-Ura-Met(0% Glucose)、4mL C-Ura-Met(1% Glucose)、4mL C-Ura-Met(2%Glucose)、4mL C-Ura-Met(5% Glucose)培养基中诱导6h后,分别取两份1OD600菌液,13000rpm离心后,弃上清,沉淀加入1mL 10μmol/mL的DHE染液重悬,其中一份于30℃摇床中孵育200min后取200μL置于底部透明四周黑色的平底96孔板中,用酶标仪于500nm/610nm处测定荧光强度。(以上定量数据均来自3次独立实验,将对应菌株孵育1h平均值当做1标准化处理,使用双侧检验法将诱导表达目标蛋白的菌株与对应菌株孵育1h相比较,***P<0.005、0.005≤**P<0.01、0.01≤*P<0.05表示有统计学意义,P>0.05表示无差异ns,误差线为标准差。)另一份加入孵育10min后加入5μL 10mmol/mL的FB-28染液再次孵育10min于倒置荧光显微镜成像。结果如图8所示,高浓度葡萄糖条件下提高细胞内活性氧水平。
蛋白表达情况检测方法如下:将转染了C端带tag的GSDMAN的质粒(prMET25-GSDMAN-HA)的BY4741菌株过夜培养后取6OD600于6mL C-Ura-Glucose培养基孵育1h后,取4份取4OD600分别置于4mL C-Ura-Met(0% Glucose)、4mL C-Ura-Met(1% Glucose)、4mL C-Ura-Met(2% Glucose)、4mL C-Ura-Met(5% Glucose)培养基中诱导6h后,取2OD菌液以Pgk1为对照,测定蛋白表达情况。结果如图9所示,高浓度葡萄糖可以增加GSDMAN蛋白的水平。
为考察葡萄糖对MET25启动子转录水平的影响,我们构建了prMET25-GFP质粒,在相同条件下诱导GFP蛋白的表达,结果如图10所示,实验结果发现不同葡萄糖浓度并不会改变细胞内活性氧水平,也对MET25启动子驱动的GFP蛋白的表达水平并没有影响。
以上实验结果表明,高浓度葡萄糖可以提高表达了GSDMAN蛋白的细胞内活性氧水平,并可能通过增加蛋白的稳定性促进其在膜上的聚集,提高焦亡水平;而低浓度葡萄糖降低表达了GSDMAN蛋白的细胞内活性氧水平,并可能通过降低蛋白的稳定性抑制Gasdermin蛋白在膜上的聚集,降低焦亡水平。由于高浓度葡萄糖对表达了prMET25-GFP质粒的细胞内活性氧水平没有影响,可以推断GSDMAN蛋白也可以促进细胞内活性氧的产生。以上只列举了Gasdermin A蛋白结果,其余Gasdermin蛋白结果与Gasdermin A类似。
②膜修复
GSDMDN靶向质膜和细胞器膜,并在膜上打孔。但是质膜的损伤并不一定会导致细胞死亡。据报导,细胞外环境中的Ca2+离子会触发膜修复程序,包括受损膜的内吞或以外泌体的形式脱落,后一种机制依赖于转运所需的内质体分选复合物的组成部分。ESCRT(endosomal sorting complexes required for transport),它包括ESCRT-0、-Ⅰ,-II、-III、VPS4,其中最为重要的ESCRT-III和VPS4,在哺乳动物中具有12个亚基:CHMP1A、CHMP1B、CHMP2A、CHMP2B、CHMP3、CHMP4A、CHMP4B、CHMP4C、CHMP5、CHMP6、CHMP7和IST1。而ESCRT最初是在酵母中被发现的,是在酵母系统中极为保守的膜修复系统,与哺乳细胞高度同源。并且酵母已经具有很完整的基因缺失库,因此我们将我们的质粒转入到亚基缺失菌株中,探索膜修复所必须的亚基,实验过程如下:将分别转染了空载质粒(pRS416-MET25)的BY4741菌株、N端质粒(prMET25-GSDMDN)的BY4741菌株和敲除菌株过夜培养后取6OD600,于6mL C-Ura培养基孵育1h后,取1OD600菌液作为对照,再4份取4OD600分别置于4mL C-Ura-Met培养基中诱导目的蛋白表达,分别于6h、22h取1OD600 13000rpm离心后,弃上清,沉淀加入100μL灭菌水后重悬,五倍梯度稀释点在CSH-URA固体培养基上,观察其生长状况。于6h取1OD600菌液用灭菌PBS定容至1mL,取200μL置于底部透明四周黑色的平底96孔板中,加入2μL20mg/mL的PI溶液,混合均匀后于室温孵育10min后,用酶标仪于535nm/617nm处测定荧光强度。(以上定量数据均来自3次独立实验,使用双侧检验法将诱导表达目标蛋白的菌株与敲除菌株相比较,***P<0.005、0.005≤**P<0.01、0.01≤*P<0.05表示有统计学意义,P>0.05表示无差异ns,误差线为标准差。)结果如图10所示,图10只展示了GSDMDN的实验结果,表明GSDMDN诱导焦亡中,VPS60、DID2、VPS24、VPS2、VPS20、SNF7、VPS4敲除菌株死亡表型更明显。
结果表明VPS60(CHMP5)、DID2(CHMP1A/B)、VPS24(CHMP3)、VPS2(CHMP2A/B)、VPS20(CHMP6)、SNF7(CHMP4A/B/C)、VPS4(VPS4)为膜修复所必须亚基。
2.药物筛选
基于我们发现的活性氧具有稳定Gasdermin蛋白,促进Gasdermin N端蛋白在膜上的作用,我们可以筛选一系列抗氧化药物,判断该药物是否对单一蛋白引起的焦亡具有抑制作用,以判断筛选的化合物是具有特异性还是广谱抑制作用。我们选取了黄酮类化合物,以木犀草素为例,在诱导表达GSDMBN的培养基中加入适量木犀草素后,可有效降低细胞内的活性氧的水平,从而抑制细胞焦亡,实验过程如下:将转染了N端质粒(prMET25-GSDMBN)的BY4741菌株过夜培养后取6OD600于6mL C-Ura培养基孵育1h后,分别取4份4OD600置于4mL含有0、10、20、40μmol木犀草素的C-Ura-Met培养基中诱导6h后,取1OD600菌液用灭菌PBS定容至1mL,取200μL置于底部透明四周黑色的平底96孔板中,加入2μL20mg/mL的PI溶液,混合均匀后于室温孵育10min后,用酶标仪于535nm/617nm处测定荧光强度。取1OD600菌液,13000rpm离心后,弃上清,沉淀加入1mL 10μmol/mL的DHE染液重悬,30℃摇床中孵育200min后取200μL置于底部透明四周黑色的平底96孔板中,用酶标仪于500nm/610nm处测定荧光强度。(以上定量数据均来自3次独立实验,使用双侧检验法将未加入木犀草素的菌株加入不同浓度木犀草素的菌株相比,***P<0.005、0.005≤**P<0.01、0.01≤*P<0.05表示有统计学意义,P>0.05表示无差异ns,误差线为标准差。)结果如图11所示。
以上为本系统目前的发现与应用,后续可以利用此平台进行多种机制研究实验,例如,构建点突变体筛选GSDM蛋白诱导焦亡的重要氨基酸位点;构建GSDM截断型突变体筛选最小致死片段;考察钙离子/钾离子流对ESCRT复合物膜修复的影响;以及高通量药物筛选等。

1.一种用于研究人类Gasdermin蛋白功能的细胞模型,其特征在于,所述的细胞模型是以酵母菌为宿主,表达单一的Gasdermin全长蛋白或Gasdermin N端蛋白,并以MET25启动子启动蛋白表达。
2.根据权利要求1所述的细胞模型,其特征在于,所述的Gasdermin蛋白为GasderminA、Gasdermin B、Gasdermin C、Gasdermin D或Gasdermin E。

4.根据权利要求2所述的细胞模型,其特征在于,Gasdermin A的N端蛋白的氨基酸序列如SEQ ID NO.6所示;Gasdermin B的N端蛋白的氨基酸序列如SEQ ID NO.7所示;GasderminC的N端蛋白的氨基酸序列如SEQ ID NO.8所示;Gasdermin D的N端蛋白的氨基酸序列如SEQID NO.9所示;Gasdermin E的N端蛋白的氨基酸序列如SEQ ID NO.10所示。
5.根据权利要求1所述的细胞模型,其特征在于,所述的酵母菌为酿酒酵母。
6.根据权利要求1所述的细胞模型,其特征在于,所述的细胞模型是以酵母质粒载体pRS系列、YCp系列或YEp系列质粒为表达载体。
7.根据权利要求1所述的细胞模型,其特征在于,Gasdermin蛋白或Gasdermin N端蛋白的C端设有HA-tag标签。
8.一种权利要求1~7任一项所述的细胞模型得构建方法,其特征在于,包括如下步骤:
将编码Gasdermin蛋白或Gasdermin N端蛋白的基因构建至pRS416-MET25质粒上,得到重组表达载体;将重组表达载体导入至酵母细胞中,得到所述的细胞模型。
9.一种权利要求1~7任一项所述的细胞模型在Gasdermin蛋白功能研究中的应用。
10.一种权利要求1~7任一项所述的细胞模型在筛选以Gasdermin蛋白为靶点的药物中的应用。
