Principle of testing
Hyaluronidase is a specific lyase of hyaluronic acid, is a participant in allergic reactions, and has a strong correlation with histamine release from mast cells, and the like. Whether the test sample has the effect of relieving can be judged by using the hyaluronidase inhibition rate, and the higher the hyaluronidase inhibition rate is, the stronger the effect of relieving without the substance is reflected, and otherwise, the weaker the effect is.
According to the test method, the hyaluronidase inhibition test results of the samples are compared with those of the negative control, and if the inhibition rate of the test sample is higher than that of the negative control and has significant difference (p is less than 0.05), the test sample can be considered to have certain relieving efficacy.
The test indexes are shown in Table 1
TABLE 1 test indexes
Test materials and methods:
the instrument equipment comprises:
BSA224S analytical balance
L6s ultraviolet spectrophotometer
Reagent:
(ii) a hyaluronidase enzyme(s),
the sodium hyaluronate of BR is prepared by the steps of,
BR dipotassium glycyrrhizinate with the purity not less than 98 percent
The test method comprises the following steps:
(1) Treatment of controls and test samples
Example samples;
positive control (dipotassium glycyrrhizinate, purity > 98%): diluting with water to reach a positive control concentration of 3%;
negative control: pure water.
(2) Procedure for testing
Setting up sample group, sample background group, solvent group and solvent background group, wherein each group needs 3 parallels, adding different reagent solutions into four groups, shaking, standing at room temperature for 30min for color development, and measuring absorbance value at 528nm with ultraviolet spectrophotometer.
(3) Formula for calculation
In the formula: a-is the absorbance of the reaction solution without the sample;
b-is the absorbance of the reaction solution without sample and enzyme;
c-is the absorbance of the reaction solution containing the sample and the enzyme;
d-is the absorbance of the reaction solution containing the sample and no enzyme.
(4) Data analysis
The statistical analysis software was SPSS, and the comparison between hyaluronidase inhibition for test samples, positive controls and negative controls was performed using independent sample t-tests. The above statistical analyses were all two-tailed tests with significance level α =0.05.P >0.05, indicating no significant difference between the two groups; p <0.05, indicating significant differences between the two groups.
The test results are shown in table 2:
TABLE 2 results of the soothing efficacy test
Note: hyaluronidase inhibition was maintained at three decimal places, p <0.05, indicating significant differences between samples and positive controls.
As can be seen from table 2, the children's aqueous gel containing an oat composition having soothing effect according to the present invention has an inhibitory effect on hyaluronidase, and example 3 is superior to examples 1 and 2, it can be seen that soothing effect increases with the increase of the addition amount of the oat composition, and when used in combination with bisabolol and ginger root extract, the oat composition and the bisabolol ginger root extract can be reduced in the use amount of each, and the reduction of the addition amount of the raw materials can reduce the production cost, so that example 3 is preferred in the present invention. The specific production process comprises the following steps:
pumping water and carbomer into the stirring kettle, stirring, dissolving, mixing, adding the rest materials one by one, and stirring, dissolving and mixing the above materials before adding each material.
