Abstract
The invention discloses an acne-removing composition and application thereof, wherein the effective components comprise high molecular hyaluronic acid or salt thereof, hydrolyzed hyaluronic acid or salt thereof and spearmint hydrosol. According to the acne-removing composition, the protective film is formed on the surface of the skin through the high-molecular hyaluronic acid or the salt thereof, so that the pollution of external pollutants to the skin is isolated, meanwhile, the problem of skin acne is relieved through the spearmint hydrosol, and the small-molecular hydrolyzed hyaluronic acid or the salt thereof can penetrate into the skin to quickly replenish water to an affected part, reduce inflammatory reaction and play a role in removing acnes.

Technical Field
The invention relates to an acne-removing composition and application thereof, belonging to the technical field of biomedicine and personal care.
Background
Acne, commonly known as whelk, is a chronic inflammatory pilosebaceous disease and is one of the most common diseases of dermatology. The main causes of acne in the skin are the following: 1. the sebaceous gland is exuberant in secretion to cause excessive grease, large or blocked pores and thickened cuticle, the grease gradually bulges towards the surface of the cortex, so that the grease blocks the pores, the bulged vertexes are oxidized into black, bacteria are stirred together with the grease in the pores, propionibacterium acnes are propagated in a large amount under the condition of oxygen deficiency, inflammatory bacteria are invaded to become pustules and nodules, and finally the whole hair follicle turns red and inflamed; 2. skin dryness is also a cause of acne, and due to insufficient skin moisture and excessive dryness, excessive oil is secreted to moisten the skin; 3. endocrine disorders.
Alleviating or treating acne typically begins with: 1. improving the moisturizing effect of the skin; 2. regulating oil balance of skin, and inhibiting excessive sebum secretion; 3. inhibiting the proliferation of propionibacterium acnes; 4. anti-inflammatory; 5. regulating endocrine secretion.
At the present stage, the commonly used raw materials are various kinds of tartaric acid raw materials, so that the tartaric acid raw materials have great irritation to the skin and bring troubles to a plurality of easily sensitive skins troubled by acne. And the existing acne removing product has single efficacy, cannot comprehensively care acne and has not ideal acne removing effect.

The invention aims to provide an acne-removing composition, which is prepared by matching pure natural spearmint hydrosol with hyaluronic acid or salt thereof, has low stimulation and difficult sensitization, and can effectively improve the problem of skin acne from inside to outside while isolating external pollution.

Further, in the acne-removing composition, the content of the polymer hyaluronic acid or the salt thereof is preferably 0.05-1.5%. The content of the hydrolyzed hyaluronic acid or a salt thereof is preferably 0.05 to 0.5%. The content of the spearmint hydrosol is preferably 5 to 60%, preferably 5 to 50%, more preferably 5 to 30%, and still more preferably 10 to 30%.
Further, in the acne-removing composition, the molecular weight of the high molecular hyaluronic acid or the salt thereof is 1000-1800kDa, preferably 1000-1500 kDa.
Further, in the acne-removing composition, the molecular weight of the hydrolyzed hyaluronic acid or the salt thereof is 1kDa-10kDa, preferably 5kDa-10 kDa.
Further, in the above acne-removing composition, the salt of the high molecular hyaluronic acid or hydrolyzed hyaluronic acid may be selected from one or more of sodium salt, potassium salt, calcium salt and zinc salt.
Further, in the acne-removing composition, the spearmint hydrosol is a natural raw material extracted by a distillation method, and the spearmint hydrosol is an aqueous phase part in an extracting solution obtained by extracting the spearmint by the distillation method. The spearmint plant is treated by a distillation method to obtain two products of spearmint essential oil and spearmint hydrosol, wherein the spearmint essential oil is an oil-soluble component, and the spearmint hydrosol is mainly a water-soluble component.

The invention has the following beneficial effects:
1. according to the acne-removing composition provided by the invention, the high-molecular hyaluronic acid or the salt thereof can effectively form a layer of protective film on the surface of the skin to isolate the pollution of external pollutants to the skin, so that a good relieving and repairing environment is provided for acne; the small-molecule hydrolyzed hyaluronic acid or the salt thereof is compounded with the spearmint hydrosol, so that the propagation of propionibacterium acnes can be effectively inhibited, the skin inflammation can be deeply relieved, and the problem of skin acne can be fundamentally relieved;
2. according to the acne-removing composition provided by the invention, through compounding of the hyaluronic acid or the salt thereof with high and low molecular weights, the spearmint hydrosol can be effectively delivered to the skin, meanwhile, the sealing and protecting effects on the small-molecular spearmint hydrosol can be realized, the loss of the small-molecular active ingredients of the spearmint hydrosol is prevented, and the acne-removing composition can better play a role in relieving acne.

The present invention is further described below in conjunction with specific examples, which are intended to be illustrative and explanatory only and are not restrictive of the invention.
Unless defined otherwise, technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art. Although methods and materials similar or equivalent to those described herein can be used in experimental or practical applications, the materials and methods are described below. In case of conflict, the present specification, including definitions, will control, and the materials, methods, and examples are illustrative only and not intended to be limiting. The present invention is further illustrated by the following examples, which are not intended to limit the scope of the invention.
The invention provides an acne-removing composition, and the effective components of the composition comprise high-molecular hyaluronic acid or salt thereof, hydrolyzed hyaluronic acid or salt thereof and spearmint hydrosol. According to the invention, through the compounding of the three components, a skin surface moisturizing and water locking layer mainly comprising film-forming high-molecular hyaluronic acid or salt thereof, a deep water replenishing and repairing layer mainly comprising hydrolyzed hyaluronic acid or salt thereof and spearmint hydrosol playing a role in relieving acne in the middle are formed in the space.
Hyaluronic Acid (HA) is a high-molecular polysaccharide naturally existing in human interstitial tissue, is widely applied to the fields of medicines, skin care products, foods and the like, HAs various different molecular weights, HAs different effects and expressions of HA with each molecular weight, HAs excellent film forming and water locking effects, and can permeate skin to deeply moisturize the skin.
In a specific embodiment, the content of the polymeric hyaluronic acid or a salt thereof is 0.01 to 2% by mass, for example, 0.01%, 0.05%, 0.1%, 0.5%, 1%, 1.5%, 2%, preferably 0.05 to 1.5%. The content of the hydrolyzed hyaluronic acid or a salt thereof may be, for example, 0.01%, 0.05%, 0.1%, 0.15%, 0.5%, 1%, 1.06%, 1.5%, 1.55%, 2%, and preferably 0.05 to 0.5% by mass.
In a specific embodiment, the high molecular weight hyaluronic acid or salt thereof has a molecular weight of 1000-1800kDa, such as 1000kDa, 1100kDa, 1200kDa, 1300kDa, 1400kDa, 1500kDa, 1600kDa, 1700kDa, 1800kDa, preferably 1000-1500 kDa. The molecular weight of the hydrolyzed hyaluronic acid or a salt thereof is 1kDa to 10kDa, and may be, for example, 1kDa, 2kDa, 3kDa, 4kDa, 5kDa, 6kDa, 7kDa, 8kDa, 9kDa, 10kDa, and preferably 5kDa to 10 kDa.
The spearmint hydrosol is a natural extract, and a condensed water solution obtained by distilling and extracting spearmint plants is the spearmint hydrosol. The spearmint plant is treated by a distillation method to obtain two products of spearmint essential oil and spearmint hydrosol, wherein the spearmint essential oil is an oil-soluble component, and the spearmint hydrosol is mainly a water-soluble component, so the spearmint essential oil and the spearmint hydrosol are two completely different products.
In a specific embodiment, the amount of the exposed portion of the spearmint is 3 to 60%, for example, 3%, 5%, 10%, 15%, 20%, 25%, 30%, 35%, 40%, 45%, 50%, 55%, 60%, preferably 5 to 50%, more preferably 5 to 30%, and still more preferably 10 to 30%.
The composition also comprises a preservative, wherein the preservative is selected from one or more of pentanediol, hexanediol, butanediol and ethylhexyl glycerol, and the mass percentage of the preservative in the composition is 0.1-5%.
The composition further comprises a solvent, preferably water, which serves to dissolve and adjust the content of the ingredients.
The following examples are given to illustrate the present invention, and the experimental methods used in the following examples are all conventional methods unless otherwise specified. Materials, reagents and the like used in the following examples are commercially available unless otherwise specified. Wherein the high molecular weight sodium hyaluronate and the hydrolyzed sodium hyaluronate are both from Huaxi Biotechnology corporation.
Examples and comparative examples

Test example 1 in vitro bacteriostatic test
(1) Preparation of the bacterial suspension
Respectively inoculating standard strains of Staphylococcus aureus ATCC6538 and Propionibacterium acnes ATCC11827, culturing Staphylococcus aureus under conventional culture conditions for 24h, culturing Propionibacterium acnes in an anaerobic incubator at 36 deg.C for 48h, adding appropriate amount of sterile sodium chloride solution to elute the culture, sucking the bacterial suspension out of a sterile test tube, adding 0.85% sterile sodium chloride solution, and making into 106cfu/mLAnd (4) bacterial suspension.
(2) Determination of the bacteriostatic Rate
And (3) determining the bacteriostasis rate by a suspension quantitative method according to the evaluation method of the antibacterial and bacteriostasis effects of the QB/T2738-2012 daily chemical products.
In a biological safety cabinet, respectively taking 5mL of the acne-removing composition prepared in each example and comparative example, placing the acne-removing composition into a sterile test tube, keeping the temperature at 20 ℃ for 5min, adding 0.1mL of staphylococcus aureus liquid, uniformly mixing, starting timing, acting for 10min, 1h and 3h, respectively taking 0.1mL of the mixed liquid, adding the mixed liquid into 5mL of PBS, standing for 10min, diluting properly, then pouring the mixed liquid into a flat plate, pouring the mixed liquid with a corresponding culture medium, culturing at 35 ℃ for 48h, then counting viable bacterial colonies, and simultaneously using the PBS to replace the acne-removing composition as a blank control. The experiment was repeated 3 times and the average was taken. Simultaneously, the experiment for inhibiting the propionibacterium acnes is carried out, and the method is the same except that the propionibacterium acnes is cultured by an anaerobic incubator. From the results of examples 1, 2, 3, 4, 5 and 10, it can be seen that the bacteriostatic effect of the composition increases with the increase of the amount of the spearmint hydrolat, but the results of examples 2 and 5 are not much different, which indicates that the bacteriostatic effect of the composition is not much affected by further increasing the amount of the spearmint hydrolat after increasing to a certain amount, and the amount of the spearmint hydrolat is preferably 5 to 60%, more preferably 5 to 50%, more preferably 5 to 30%, and most preferably 10 to 30% in view of cost.
