The invention discloses an ointment for treating bacterial skin diseases and a preparation method thereof, wherein zinc glycolate, urea, glyceryl stearate, triethanolamine and carbomer are added with a proper amount of water to be heated and ground until the mixture is pasty to form a component A; taking cetearyl alcohol, parabens or gallate compounds, grinding in a mortar, and dissolving with ethanol until the mixture is pasty to form a component B; heating component A to appropriate temperature, slowly adding essence and component B into component A, stirring while adding in one direction, mixing, stirring, and cooling to room temperature to obtain the final product. The ointment prepared by the invention is mainly used for treating bacterial skin diseases, and can also be used for treating skin diseases such as dermatitis, eczema and the like caused by bacteria.

The invention belongs to the technical field of medicines and preparation thereof, and particularly relates to an ointment for treating bacterial skin diseases and a preparation method thereof.
Background
The bacterial skin diseases refer to skin lesions caused by bacterial infection and bacterial toxin invasion, the skin lesions are common at exposed parts such as heads, faces, necks and hands, clinically show red, swollen, hot, painful and purulent parts, have certain transmissibility, and the transmission route is contact transmission, the main pathogenesis of the bacterial skin diseases is allergic reaction (allergic to bacterial products) or toxic reaction (damage caused by bacterial toxins), and the bacterial skin diseases can be divided into acute and chronic diseases, most of the bacterial skin diseases, especially the coccal skin diseases have acute attack and short course of disease; only a few of the chronic diseases, such as skin tuberculosis, leprosy and the like, have the treatment principles of diminishing inflammation, sterilizing, drying and astringing, and preventing the inflammation from spreading.
Zinc glycolate is an organic zinc-containing compound with an antibacterial effect, the main bactericidal active component is divalent zinc ions, and due to the oxidation-reduction property of the zinc ions, when the zinc ions are combined with bacterial cell membranes, the zinc glycolate reacts with organic matters in the bacterial cell membranes to destroy the structure of membrane proteins and lose the activity of the membrane proteins, so that the aim of sterilization is fulfilled.
Urea is often used as an ingredient of an ointment to increase the moisture of the skin, and at the same time, urea also dissolves proteins, increases the water content of the stratum corneum, increases the hydration of the stratum corneum, makes the skin soft, and has antibacterial, antipruritic, and granulation-promoting effects.
Methyl paraben is commonly used as a sterilization preservative for organic synthesis, food, cosmetics and medicines, and has a phenolic hydroxyl structure and strong antibacterial performance, and the antibacterial action mechanism is as follows: destroying the cell membrane of the microorganism, denaturing the protein in the cell, and inhibiting the activities of the respiring enzyme system and the electron transfer enzyme system of the microorganism cell.
The invention aims to achieve good antibacterial treatment effect through the synergistic and addition effects generated by the combination of zinc glycolate serving as a main drug and methyl p-hydroxybenzoate serving as an auxiliary material and an antibacterial compound, and simultaneously reduce the irritation effect on skin caused by long-term administration.
Disclosure of Invention
The invention provides a novel ointment and a preparation method thereof, aiming at overcoming the defects of the existing ointment for treating bacterial skin diseases, and aiming at preparing a compound ointment by combining a plurality of antibacterial agents so as to improve the healing effect, shorten the healing time and reduce the irritation to the skin.
The technical scheme adopted by the invention is as follows:
a preparation method of an ointment for treating bacterial skin diseases comprises the following steps:
s1: heating zinc glycolate, urea, glyceryl stearate, triethanolamine and carbomer with appropriate amount of water, and grinding until paste is formed to form component A;
s2: taking cetearyl alcohol, parabens or gallate compounds, grinding in a mortar, and dissolving with ethanol until the mixture is pasty to form a component B;
s3: heating component A to appropriate temperature, slowly adding essence and component B into component A, stirring while adding in one direction, mixing, stirring, and cooling to room temperature to obtain the final product.
Furthermore, the paraben compound is selected from methyl paraben, ethyl paraben and propyl paraben, and the gallic acid ester is selected from methyl gallate, ethyl gallate and propyl gallate.
The preparation method comprises the following steps of: 100-200 parts of zinc glycolate, 30-70 parts of glycerol, 40-60 parts of cetostearyl alcohol, 100-150 parts of urea, 80-120 parts of glyceryl stearate, 30-70 parts of triethanolamine, 30-70 parts of carbomer, 60-100 parts of p-hydroxybenzoate or gallate compound, 80-120 parts of ethanol and 10-50 parts of essence.

EXAMPLE 3 study of irritation of drug on mouse skin

(4) the zinc glycolate with a single component, the methyl p-hydroxybenzoate with a single component, the ointment prepared by the invention and the physiological saline are acted on the tested part of the mouse under the same condition;
(5) each time lasts for 0.5h, after 0.5h, the medicine is washed out by warm water, once a day, the medicine is applied for one week, the observation is continuously carried out for 7 days, and the skin irritation scores of the mice are respectively observed on 1 day, 3 days and 7 days.
(6) The results are shown in Table 7, the skin irritation response score criteria are shown in Table 8, and the skin irritation intensity evaluation criteria are shown in Table 9.

As can be seen from the results of example 3, the ointment prepared according to the present invention (average stimulation response score: 1.6, which is mild irritation) has lower skin irritation and better therapeutic effect than the single component zinc glycolate (average stimulation response score: 3.8, which is moderate irritation) and the single component methylparaben (average stimulation response score: 4.2, which is moderate irritation).
In conclusion, the bacterial ointment and the preparation method thereof have certain treatment effect on bacterial skin diseases and eczema, and compared with the antibacterial single component of zinc glycolate and the antibacterial single component of methyl p-hydroxybenzoate, the antibacterial ointment prepared by the invention has the advantages of shorter treatment time, lower skin irritation, stronger antibacterial performance and obvious synergy.
